PGA1 was incubated with rabbit renal cortical homogenates containing HSA (0–4.5%). The ability of this tissue to readily metabolize PGA1 progressively decreased with increasing HSA levels in the incubates The rate of disappearance of 3H-PGA1 was twice as rapid in rats treated with salicylic acid (S. A.) in comparison to control animals; since only 30% of the injected radioactivity was bound to the plasma of the S.A. treated rats, as compared to 90% bound to control plasma, an association may exist between the degree of binding of 3H-PGA1 and its rate of clearance. The studies indicate that PGA1 interaction with HSA decreases its metabolism in vitro, and slows down its clearance in vivo, implicating HSA as a possible factor in prostaglandins metabolism in vivo.  相似文献

Isolated cultures of mononuclear phagocytes from 12 humans (7 normal controls and 5 with cancer) produced prostaglandins (PGs) of the E series when stimulated with artificial immune complexes (HSA and anti-HSA). The amount of PGs produced by monocytes from breast cancer patients and macrophages from ascites fluid taken from patients with abdominal cancers was 4 fold that produced by blood monocytes from normal controls. Immune complex (IC) determinations from the serums of these humans (Raji cell method) showed that only patients in the advanced stages of the diseases, and none of the controls, had elevated levels of IC. In the presence of IC, there was noted a depression in the immune reactivity of mononuclear cell cultures, as determined by 3H thymidine uptake following stimulation with PHA, Con A and PWM. The depressed blastogenic response was not in all instances fully reversed by the addition of indomethacin or aspirin, indicating that PGs were not the only causative factor. It is concluded that mononuclear phagocytes elaborate immunosuppressive factors in response to stimulation by immune complexes. One of these factors is PGE. Further, this results in a homeostatic regulation which prevents damage from IC deposition and may be, in part, responsible for immunosuppression of cancer patients.  相似文献

To unravel the genetic basis for the pepsinogen A (PGA) protein polymorphism, we have isolated and characterized a number of PGA genes, distinguishable by polymorphic EcoRI fragments of 12.0, 15.0, and 16.6 kb. Using a HindIII or AvaII polymorphism, we can discriminate between different 15.0 (15.0 and 15.0*) and 12.0 (12.0s and 12.0l) genes, respectively. The coding sequences of a 15.0 and a 16.6 gene were determined, together with considerable stretches of the 5'- and 3'-flanking regions and introns. The genes were demonstrated to encode Pg5 and Pg4, respectively. Because substitutions in codons 43 and 207 appeared to be critical in the determination of the encoded proteins, we sequenced only these regions in the two 12.0 genes and the 15.0* gene. On the basis of these partial sequences, we assume that these genes encode Pg3. In the evolutionary model of the PGA gene cluster presented here, the 12.0 genes arose by an unequal, but homologous crossover. The results of sequence analysis of the second intron of the 12.0s, 12.0l, 15.0, and 16.6 genes suggest that the two 12.0 genes have arisen from two different crossover events.  相似文献

We have developed a method which can measure the menstrual prostaglandin (PG) activity in a single tampon specimen by bioassays. This method makes it possible to monitor the menstrual PG activity continuously during menstruation. Using this technique, we determined the menstrual PG patterns of two normal non-dysmenorrheic subjects, one subject on oral contraceptives (OC) and one subject with moderate to severe dysmenorrhea. Two to four cycles were studied per subject. We observed three mentrual patterns among the four subjects studied. Compared to the two normal controls, the subject on OC had a significantly lower menstrual fluid total and menstrual PG activity. The mean values +/- S.E. per menstrual period were 33.4 g +/- 1.5 vs 21.5 g +/- 2.0 and 28.6 microgram (PGF2alpha equivalent) +/- 1.5 vs 11.3 microgram +/- 4.2 respectively (control vs OC). The dysmenorrheic subject had a menstrual fluid total of 37.0 g +/- 1.9 similar to the two normal controls. Her menstrual PG activity (49.8 microgram +/- 7.7), however, was nearly two times higher than the normal controls. In one cycle studied, the dysmenorrheic subject was treated with a PG synthetase inhibitor, ibuprofen (Motrin). Remarkable relief was achieved. The alleviation of symptoms was accompanied by a concomitant marked reduction in the menstrual PG activity.  相似文献

An analytical procedure for the estimation of norethindrone (17β-hydrox-19-nor-17α-pregn-4-en-20-yn-3-one) in human plasma has been developed. After extraction and purification on a silver ion exchange column, norethindrone is converted to a methoxime-trimethylsilyl ether derivative and measured by a gas chromatograph-mass spectrometer-accelerating voltage alternator system. Norethindrone-13C20,21?7?2H is used as a carrier and internal standard. Results correlated well with those obtained by radiochromatography. Maximum plasma levels of norethindrone in chronically treated and naive subjects ranged from 17–38 ng/ml.  相似文献

Conclusions The results we have obtained on forearm muscles of normal human subjects are complementary to those on isolated frog muscle and they lead, to the same conclusions. We are satisfied that our31P TMR measurements of [P-metabolites] in normal subjects are reliable, and we are now extending these methods for use in cases of degenerative muscular disorders (Edwards et al., 1982). Chemical estimates of [ADP] and [AMP] are not directly relevant to metabolic studies, and so an important contribution of NMR to the study of muscular mechanisms is its ability to estimate the concentrations of these metabolites that are free to take part in metabolic reactions.  相似文献

Radioimmunoassay of 5alpha, 7alpha-dihydroxy-11-keto-tetranor-prosta-1,16-dioic acid, main urinary metabolite of prostaglandin F2alpha (PGF2alpha-MUM), was performed in normal subjects. Twenty-four hours secretion of PGF2alpha-MUM were 29.04 +/- 9.73 microgram in males and 18.22 +/- 5.88 microgram in females on an average. And an oral administration of aspirin resulted in the remarkable decrease of PGF2alpha-MUM in both sexes.  相似文献

Thirty-nine normal volunteers of both sexes were randomly assigned to receive 8 sessions of temperature biofeedback or autogenic training to increase finger temperature. Temperature biofeedback subjects produced significant elevations in finger temperature during training, whereas those who received autogenic training did not. Temperature feedback subjects had significantly higher heart rates and diastolic blood pressures during training compared to autogenic subjects. There were no significant changes or group differences in plasma catecholamine levels. These data do not support the hypothesis that feedback-induced vasodilation is accompanied by decreased sympathetic activation in normal populations, when only temperature biofeedback is employed.Supported by research grant No. HL-30604 from NHLBI. Lymphocytes from acute lymphocytic leukemia (ALL) subjects were converted by mitogens to blast-like cells whose microscopic appearance and rate of formation was indistinguishable from those in mitogen incubated control lymphocytes. In ALL lymphocytes, however, pokeweed mitogen (PWM) failed to stimulate GGT expression; the mean increase it caused in thymidine kinase (TK) activity and thymidine incorporation was normal, though there were appreciable individual variations. These variations were also apparent with concanavalin A (Con A) but, in most ALL cases, TK and thymidine incorporation rose to much higher levels than in Con-A-treated control lymphocytes. The results indicate that evaluation of the response to mitogens by quantitative biochemical criteria provides a sensitive method for revealing functional impairments in microscopically normal ALL lymphocytes.  相似文献
