The skeleton of Australopithecus afarensis (A.L. 288-1, better known as "Lucy") is by far the most complete record of locomotor morphology of early hominids currently available. Even though researchers agree that the postcranial skeleton of Lucy shows morphological features indicative of bipedality, only a few studies have investigated Lucy's bipedal locomotion itself. Lucy's energy expenditure during locomotion has been the topic of much speculation, but has not been investigated, except for several estimates derived from experimental data collected on other animals. To gain further insights into how Lucy may have walked, we generated a full three-dimensional (3D) reconstruction and forward-dynamic simulation of upright bipedal locomotion of this ancient human ancestor. Laser-scanned 3D bone geometries were combined with state-of-the-art neuromusculoskeletal modeling and simulation techniques from computational biomechanics. A detailed full 3D neuromusculoskeletal model was developed that encompassed all major bones, joints (10), and muscles (52) of the lower extremity. A model of muscle force and heat production was used to actuate the musculoskeletal system, and to estimate total energy expenditure during locomotion. Neural activation profiles for each of the 52 muscles that produced a single step of locomotion, while at the same time minimizing the energy consumed per meter traveled, were searched through numerical optimization. The numerical optimization resulted in smooth locomotor kinematics, and the predicted energy expenditure was appropriate for upright bipedal walking in an individual of Lucy's body size.  相似文献

Olfactory receptors (ORs) are G protein-coupled receptors that detect odorants in the olfactory epithelium, and comprise the largest gene family in the genome. Identification of OR ligands typically requires OR surface expression in heterologous cells; however, ORs rarely traffic to the cell surface when exogenously expressed. Therefore, most ORs are orphan receptors with no known ligands. To date, studies have utilized non-cleavable rhodopsin (Rho) tags and/or chaperones (i.e. Receptor Transporting Protein, RTP1S, Ric8b and Gαolf) to improve surface expression. However, even with these tools, many ORs still fail to reach the cell surface. We used a test set of fifteen ORs to examine the effect of a cleavable leucine-rich signal peptide sequence (Lucy tag) on OR surface expression in HEK293T cells. We report here that the addition of the Lucy tag to the N-terminus increases the number of ORs reaching the cell surface to 7 of the 15 ORs (as compared to 3/15 without Rho or Lucy tags). Moreover, when ORs tagged with both Lucy and Rho were co-expressed with previously reported chaperones (RTP1S, Ric8b and Gαolf), we observed surface expression for all 15 receptors examined. In fact, two-thirds of Lucy-tagged ORs are able to reach the cell surface synergistically with chaperones even when the Rho tag is removed (10/15 ORs), allowing for the potential assessment of OR function with only an 8-amino acid Flag tag on the mature protein. As expected for a signal peptide, the Lucy tag was cleaved from the mature protein and did not alter OR-ligand binding and signaling. Our studies demonstrate that widespread surface expression of ORs can be achieved in HEK293T cells, providing promise for future large-scale deorphanization studies.  相似文献

Native species can have a range of responses to nonnative introductions, from negative to positive, and understanding how and why native species respond differently to nonnatives remains an important management challenge. Based on differences and similarities in ecology and behavior, we predicted how abundance and diet of two native warblers, Lucy’s warbler (Oreothlypis luciae) and yellow warbler (Setophaga petechia), would differ in habitats with different amounts of nonnative tamarisk trees and the three nonnative insects obligately dependent on tamarisk (Tamarix spp.). Specifically, we predicted that Lucy’s warblers would have similar densities across sites, yellow warbler densities would be inversely related to tamarisk cover, and both warblers, being generalist insectivores, would incorporate tamarisk biocontrol insects in their diet. Based on point counts and fecal samples at six sites along the Virgin River in the southwestern United States, we found that yellow warblers decreased in abundance with increasing tamarisk cover, while Lucy’s warbler abundance did not and that diet of the two warblers did not differ, with both species exhibiting strong selection for the nonnative tamarisk weevil (Coniatus splendidulus) and weak to no selection for the nonnative tamarisk leafhopper (Opsius stactogalus). Both warblers showed negative selection for the tamarisk beetle (Diorhabda carinulata) and its larvae, even when those insects were 10–100 times more abundant during outbreaks. Although both warblers exploited the novel food resources offered by tamarisk, with those insects contributing half or more of total prey biomass, Lucy’s warblers were better able to maintain densities in tamarisk habitats. We hypothesize this was due to the Lucy’s warbler’s ability to exploit a broader array of habitats surrounding tamarisk sites and its cavity nesting habit that buffers its nests from the higher temperatures and lower humidity of tamarisk-dominated habitat. Our results suggest that predictions based on detailed knowledge of the form and function of native and nonnative species can be used to predict native bird response to nonnatives.  相似文献

Grain protein concentration of durum wheat is often too low, particularly in low-N-input systems. The aim of our study was to test whether a durum wheat-winter pea intercrop can improve relative yield and durum wheat grain protein concentration in low-N-input systems. A 2-year field experiment was carried out in SW France with different fertilizer-N levels to compare wheat (Triticum turgidum L., cv. Nefer) and pea (winter pea, Pisum sativum L., cv. Lucy) grown as sole crops or intercrops in a row-substitutive design. Without N fertilization or when N was applied late (N available until pea flowering less than about 120 kg N ha?1), intercrops were up to 19% more efficient than sole crops for yield and up to 32% for accumulated N, but were less efficient with large fertilizer N applications. Wheat grain protein concentration was significantly higher in intercrops than in sole crops (14% on average) because more N was remobilized into wheat grain due to: i) fewer ears per square metre in intercrops and ii) a similar amount of available soil N as in sole crops due to the high pea N2 fixation rate in intercrops (88% compared to 58% in sole crops).  相似文献

The body height of Australopithecus afarensis A.L. 288-1 ("Lucy") has recently been estimated and calculated as between 1 m to 1.06 m; other estimates give ca. 1.20 m. In addition, it is often stated that her relative leg length was shorter than that of modern humans. Using relative leg-, femur- and tibia length it is shown that both statements together can not be true; either her body height must at least have been around 1.06 to 1.10 m to give "Lucy" human-like leg proportions, or, to achieve a shorter, more ape-like leg ratio, a body height of ca. 1.20 m must be assumed.  相似文献

Until recently, the hard X-ray, phase-sensitive imaging technique called grating interferometry was thought to provide information only in real space. However, by utilizing an alternative approach to data analysis we demonstrated that the angular resolved ultra-small angle X-ray scattering distribution can be retrieved from experimental data. Thus, reciprocal space information is accessible by grating interferometry in addition to real space. Naturally, the quality of the retrieved data strongly depends on the performance of the employed analysis procedure, which involves deconvolution of periodic and noisy data in this context. The aim of this article is to compare several deconvolution algorithms to retrieve the ultra-small angle X-ray scattering distribution in grating interferometry. We quantitatively compare the performance of three deconvolution procedures (i.e., Wiener, iterative Wiener and Lucy–Richardson) in case of realistically modeled, noisy and periodic input data. The simulations showed that the algorithm of Lucy–Richardson is the more reliable and more efficient as a function of the characteristics of the signals in the given context. The availability of a reliable data analysis procedure is essential for future developments in grating interferometry.  相似文献

Conclusions The present studies appear to confirm previous suggestions (Baker 1958) that some of the fixatives used for the study of membranes may not give an adequate preservation of the lipid components unless a sufficient concentration of calcium is present.These experiments are being continued, and we plan to investigate any changes that may occur during subsequent dehydration and embedding processes. It is hoped to preserve the helices so that they may be studied by thin-sectioning techniques, since a procedure that is satisfactory for these helices may also be of value in the examination of thin sections of biological membranes. Furthermore, as the helical structures appear to contain globular micelles of phospholipid, 40 to 50 Å in diameter (Lucy and Glauert 1964), continued investigation of these structures may also shed light on the formation, stability and function of globular micelles of lipid within natural membranes (Lucy 1964).  相似文献

A “long‐backed” scenario of hominin vertebral evolution posits that early hominins possessed six lumbar vertebrae coupled with a high frequency of four sacral vertebrae (7:12‐13:6:4), a configuration acquired from a hominin‐panin last common ancestor (PLCA) having a vertebral formula of 7:13:6‐7:4. One founding line of evidence for this hypothesis is the recent assertion that the “Lucy” sacrum (A.L. 288‐1an, Australopithecus afarensis) consists of four sacral vertebrae and a partially‐fused first coccygeal vertebra (Co1), rather than five sacral vertebrae as in modern humans. This study reassesses the number of sacral vertebrae in Lucy by reexamining the distal end of A.L.288‐1an in the context of a comparative sample of modern human sacra and Co1 vertebrae, and the sacrum of A. sediba (MH2). Results demonstrate that, similar to S5 in modern humans and A. sediba, the last vertebra in A.L. 288‐1an exhibits inferiorly‐projecting (right side) cornua and a kidney‐shaped inferior body articular surface. This morphology is inconsistent with that of fused or isolated Co1 vertebrae in humans, which either lack cornua or possess only superiorly‐projecting cornua, and have more circularly‐shaped inferior body articular surfaces. The level at which the hiatus' apex is located is also more compatible with typical five‐element modern human sacra and A. sediba than if only four sacral vertebrae are present. Our observations suggest that A.L. 288‐1 possessed five sacral vertebrae as in modern humans; thus, sacral number in “Lucy” does not indicate a directional change in vertebral count that can provide information on the PLCA ancestral condition. New stature estimates are provided for A.L.288-1 (Australopithecus afarensis) based on (1) the relationship between femur length and stature in separate samples of human pygmies and pygmy chimpanzees and (2) model II regression alternatives to standard least-squares methods. Estimates from the two samples are very similar and converge on a value of approximately 3'6" for "Lucy." These results are compared to prior estimates and extended to other small-bodied hominids such as STS-14 and O.H.62. A new foot-to-stature ratio is also estimated for A.L.288-1, and its potential biomechanical significance for gait is evaluated in comparison to other groups.  相似文献

The effect of up to 20 mol% incorporation of alpha-tocopherol on acyl chain order and dynamics in liquid crystalline phosphatidylcholine (PC) membranes was studied as a function of acyl chain unsaturation by electron spin resonance (ESR) of 5-, 7-, 12- and 16-doxyl spin labelled stearic acids intercalated into the membrane. Order parameters S in the upper portion of the chain (positions 5 and 7) and correlation times tau C in the lower portion (positions 12 and 16) determined from the ESR spectra indicate that in general alpha-tocopherol restricts acyl chain motion within the membrane. The magnitude of the increases in order appears to be dependent upon phospholipid molecular area, being the greatest (up to 15%) in saturated dimyristoylphosphatidylcholine (14:0-14:0 PC) which possesses a relatively small area per molecule as opposed to much smaller increases (less than 3%) in unsaturated PC membranes of larger molecular area. This behavior is interpreted as incompatible with the hypothesis of Lucy and coworkers (A.T. Diplock and J.A. Lucy (1973) FEBS Lett. 29, 205-210), who proposed that membranes are structurally stabilized by interactions between the phytyl side chain of alpha-tocopherol and the polyunsaturated chains of phospholipids.  相似文献

ABSTRACT: BACKGROUND: The current target of universal access to long-lasting, insecticide-treated nets (LLIN) is 80% coverage to reduce malaria deaths by 75% by 2015. So far, campaigns have been the main channel for large-scale delivery of LLINs, however the World Health Organization has recommended that equal priority should be given to delivery via routine antenatal care (ANC) and immunization systems (EPI) to target pregnant women and children from birth. These various channels of LLIN delivery are targeted to children of different ages. Since risk of mortality varies with child age and LLIN effectiveness declines with net age, it was hypothesized that the age at which a child receives a new LLIN, and therefore the delivery channel, is important in optimizing the health impact of a net. METHODS: A simple dynamic mathematical model was developed of delivery and impact of LLINs among children under five years of age and their household members, incorporating data on age-specific malaria death rates, net use by household structure, and net efficacy over time. RESULTS: The presented analysis finds that supplementing a universal mass campaign with extra ANC delivery would achieve a 1.4 times higher mortality reduction than campaign delivery alone, reflecting that children born in the years between campaigns would otherwise have access to old nets or no nets at an age of high risk. The relative advantage of supplementary ANC delivery is still present though smaller if malaria transmission levels are lower or if there is a strong mass effect achieved by mass campaigns. CONCLUSION: These results indicate that LLIN delivery policies must take into account the age of greatest malaria risk. The dynamics of an epidemic model with voluntary vaccinations are studied. Individual vaccination decisions are modelled using an economic/game-theoretic approach: agents in the model decide whether to vaccinate or not by weighing the cost and benefit of vaccination and choose the action that maximizes their net benefit. It is shown that, when vaccine efficacy is low, there are parameter values for which multiple steady-state equilibria and periodic equilibria coexist. When multiplicity of steady states is obtained, which one the population reaches in some cases depends entirely on agents' expectations concerning the future course of an epidemic and not on the initial conditions of the model. Studies by a number of workers using the Langmuir film balance have shown that when carbohydrates, such as sucrose or glycerol, are dissolved in a subphase on which a phospholipid is spread, film expansion occurs (Cadenhead & Demchak, 1969; Cadenhead & Bean, 1972; Maggio et al., 1976; Maggio & Lucy, 1978). Recently such effects have been observed again, particularly with the carbohydrates galactose and trehalose (Johnston et al., 1984). The origin of these film expansions was uncertain, and various suggestions have been made to explain them. One idea was that they might be due to interactions which these carbohydrates have with the water molecules close to the polar head groups of the lipids. Recent studies in our two laboratories, described here, show that the magnitude of the expansion effects is variable and that in general they arise from surfactant impurities in the sugars. These impurities are observed in carbohydrates which are reputedly of high grade; the amount of impurity present can vary from batch to batch, and sometimes they can be difficult to remove. Film balance techniques or subphase preparation can mask the detection of minor impurities. The presence of surfactant impurities in reputedly pure carbohydrates needs to be considered in other biochemical and biophysical studies of lipids and cell membranes.  相似文献

We show the applicability of 4Pi-confocal microscopy to three-dimensional imaging of the microtubule network in a fixed mouse fibroblast cell. Comparison with two-photon confocal resolution reveals a fourfold better axial resolution in the 4Pi-confocal case. By combining 4Pi-confocal microscopy with Richardson–Lucy image restoration a further resolution increase is achieved. Featuring a three-dimensional resolution in the range 100–150 nm, the 4Pi-confocal (restored) images are intrinsically more detailed than their confocal counterparts. Our images constitute what to our knowledge are the best-resolved three-dimensional images of entangled cellular microtubules obtained with light to date.  相似文献

Summary Intercellular bridges between developing germ cells were observed in human fetal ovaries at 10 to 20 weeks gestation. Bridges were frequently found between cells in early stages of degeneration, with similar regressive changes being present in the conjoined cells. In advanced stages of cellular degeneration, bridges were less frequently found and were generally distorted and partially disrupted. Similarity in appearance of adjacent degenerating cells was common, even in late stages of degeneration. These observations suggest that cellular interconnection may be responsible for synchronous degeneration of germ cells during oogenesis.The author thanks Mrs. Lucy A.
