Idli, a traditional cereal/legume-based naturally fermented steamed product with soft and spongy texture is highly popular and widely consumed in India. The inherent viable bacterial populations of mesophilic aerobes and lactics in idli batter increased in their numbers with time at 35 °C, reaching numbers in the range of 13 to 15 log10 CFU g–1. Simultaneously, the pH level decreased from 6.2 to 4.4. Strains of Bacillus cereus F 4810, Escherichia coli D 21 and Staphylococcus aureus FRI 722 (foodborne pathogens) introduced into the idli batter at an initial level of 4.3 log10 CFU g–1 was able to survive and grow well in an initial period of 6 h. However, the strain of S. aureus showed a constant increase in its numbers reaching 9.3 log10 CFU g–1 in 12 h. The addition of plantaricin LP84, a bacteriocin produced by Lactobacillus plantarum NCIM 2084 to idli batter at 1% (v/w) level was able to retard the growth of the inoculated cultures during fermentation. Two aspects were established from this study, (i) that foodborne pathogens occurring as contaminants in idli batter can survive and grow under conditions of natural fermentation and (ii) the efficacy of a lactic bacteriocin as a potential food biopreservative.  相似文献

Ersho is a clear, yellow liquid that accumulates on the surface of fermenting teff-flour batter and is collected to serve as an inoculum for the next fermentation. The pH of ersho samples was about 3.5 and titratable acidity ranged between 3.1% and 5.7%. The mean aerobic mesophilic counts from four households varied between 6.9×106 and 1.3×108 c.f.u./ml and the aerobic bacterial flora consisted of Bacillus spp. Mean yeast counts ranged between 5.2×105 and 1.8×106 c.f.u./ml and comprised, in order of abundance, Candida milleri, Rhodotorula mucilaginosa, Kluyveromyces marxianus, Pichia naganishii and Debaromyces hansenii. Candida milleri was the most dominant isolate in all samples. About 90% of the teff flour samples had aerobic mesophilic counts 105 c.f.u./g and Gram-positive bacteria constituted about 71% of the total isolates. About 80% of samples had Enterobacteriaceae counts of 104 c.f.u./g.M. Microbial dehalogenation of tetrachloroethene (PCE) and cis-dichloroethene (cis-DCE) was studied in cultures from a continuous stirred tank reactor initially inoculated with aquifer material from a PCE-contaminated site. Cultures amended with hydrogen and acetate readily dechlorinated PCE and cis-DCE; however, this transformation was incomplete and resulted in the accumulation of chlorinated intermediates and only small amounts of ethene within 60 days of incubation. Conversely, microbial PCE and cis-DCE dechlorination in cultures with benzoate and acetate resulted in the complete transformation to ethene within 30 days. Community fingerprinting by denaturing gradient gel electrophoresis (DGGE) revealed the predominance of phylotypes closely affiliated with Desulfitobacterium, Dehalococcoides, and Syntrophus species. The Dehalococcoides culture VZ, obtained from small whitish colonies in cis-DCE dechlorinating agarose cultures, revealed an irregular cell diameter between 200 and 500 nm, and a spherical or biconcave disk-shaped morphology. These organisms were identified as responsible for the dechlorination of cis-DCE to ethene in the PCE-dechlorinating consortia, operating together with the Desulfitobacterium as PCE-to-cis-DCE dehalogenating bacterium and with a Syntrophus species as potential hydrogen-producing partner in cultures with benzoate. When grown in batch culture for a prolonged period (up to 11 months), the cells ofIsochrysis aff.galbana (Isochrysis Tahitian strain T. Iso) lost their capacity to divide and became enlarged, with a reduction in the content of photosynthetic pigment and photosynthetic capacity. Upon returning to fresh medium, the cells of a 11-month-old culture regained their capacity to divide within 3 days under light conditions. The resulting cells possessed similar size, photosynthetic pigment and photosynthetic capacity as those of logarithmic growth phase. The results suggest that the aged cultures ofIsochrysis can be used as starter cultures for the production of live food for aquaculture animals.  相似文献

The controlled fermentation of turnip slices using Lactobacillus plantarum or Leuconostoc mesenteroides as starter cultures led to earlier acid production and earlier and more pronounced inhibition of Enterobacteriaceae than with uninoculated (natural) fermentation. Unlike the natural fermentation, the controlled fermentations did not show a yeast secondary fermentation and also had a better colour. Due to its ability to produce higher amounts of acid, the use of Lact. plantarum is more desirable than of Leuc. mesenteroides.  相似文献

【目的】菌糠的营养素含量齐全,但纤维素含量过高是阻碍其饲料化利用的主要因素。故本研究筛选适合于发酵杏鲍菇菌糠的微生物菌株,以改善其饲用品质。【方法】首先,本研究采用纤维素-刚果红、苯胺蓝和MRS-Ca (De Man, Rogosa, Sharpe-Ca)筛选培养基,结合纤维素、木质素酶活力及抑菌活性的测定,从EM (effective microorganisms)原液发酵的杏鲍菇菌糠中分离筛选具有较强纤维素、木质素降解能力及抑菌能力的细菌/真菌。通过细菌16S rRNA和真菌18S rDNA基因序列分析确定菌株所属种属。其次,将筛选出的菌株菌液等体积混合制成复合菌剂用于固态发酵杏鲍菇菌糠。测定不同发酵时长菌糠营养成分含量以确定最佳发酵时间,并与相同工艺条件下EM原液发酵的杏鲍菇菌糠进行饲用品质比较。【结果】筛选并鉴定得到纤维素酶活性较高的特基拉芽孢杆菌(Bacillus tequilensis)菌株P11、发酵毕赤酵母(Pichia fermentans)菌株R8和马克斯克鲁维应变酵母(Kluyveromyces marxianus)菌株MU5;木质素酶活性较高的解淀粉芽孢杆菌(Bacillus amyloliquefaciens subsp.plantarum)菌株MU7;抑菌活性较高的类肠膜魏斯氏菌(Weissella paramesenteroides)菌株R4和乳酸片球菌(Pediococcus acidilactici)菌株R9。使用以上菌株复合发酵杏鲍菇菌糠7 d后,各项指标达到稳定。与EM原液发酵的杏鲍菇菌糠相比,复合菌剂发酵杏鲍菇菌糠的NDF和ADF分别显著降低了19.6%和21.44%(P0.05);CP (crude protein)、CA (crude ash)和EE (ether extract)含量分别显著提高了10.44%、5.26%和123.53%(P0.05)。【结论】本研究筛选得到的芽孢杆菌、酵母菌和乳酸菌优势菌株复合后用于发酵杏鲍菇菌糠可以很好地改善其饲用品质,效果优于生产中常用市售EM原液。  相似文献

Starter cultures consisting ofStreptococcus diacetilactis, Strept. cremoris, Lactobacillus brevis andSaccharomyces cerevisiae were tested singly and in mixtures for ability to ferment milk to producenono with organoleptically acceptable qualities. Only mixed cultures containing eitherStrept. diacetilactis orStrept. cremoris andL. brevis were suitable. Presence of yeast adversely affected either acid formation or diacetyl production.Nono containingStrept. diacetilactis was acceptable, even in the presence ofSacch. cerevisiae, because of the high diacetyl production. The incidence and diversity of enterococci in retail food samples of meat, dairy and vegetable origin was investigated. Enterococci were present, at concentrations of 10(1) to 10(4) CFU/g. Fifty selected isolates from food samples grouped in two separate clusters by RAPD analysis. Cluster G1 (72% of the isolates) contained the E. faecium CECT 410T type strain, and also showed a high degree of genetic diversity. Cluster G2 (28% of the isolates) contained the E. faecalis CECT 481T type strain and was genetically more homogeneous. Virulence traits (haemolysin, gelatinase or DNAse activities, or the presence of structural genes cylL, ace, asal and esp) were not detected. All isolates were sensitive to the antibiotics ampicillin, penicillin, gentamicin, streptomycin and chloramphenicol. A high pecentage of isolates were resistant to erythromycin and rifampicin. Many isolates showed intermediate sensitivity to several antibiotics (tetracycline, ciprofloxacin, levofloxacin, or quinupristin/dalfopristin). Vancomycin and teicoplanin resistance was detected in one strain, but vanA, vanB, vanC1, vanC2 or vanC3 genes were not detected. Many of the isolates showed functional properties of food or health relevance. Production of antimicrobial substances was detected in 17 of the isolates, and 14 of them carried structural genes for enterocins A, B and/or P.  相似文献

Specific protein inhibitors of microbial serine proteinases were isolated from wheat (Triticum aestivum L.), rye (Secale cereale L.) and triticale using affinity chromatography on subtilisin-Sepharose 4B. The wheat inhibitor had an isoelectric point (pI) at pH 7.2, while the rye inhibitor consisted of two forms with pI values of 6.8 and 7.1. In triticale, two components were present with pIs 7.2 and 6.8. All the inhibitors had M r values of approx. 20 000. The isolated proteins were effective inhibitors of subtilisins Carlsberg and BPN, and of fungal proteinases (EC 3.4.21.14) from the genus Aspergillus, but they were completely inactive against trypsin (EC 3.4.21.4) chymotrypsin (EC 3.4.21.1) and pancreatic elastase (EC 3.4.21.36). The inhibitors formed complexes with subtilisin in a molar ratio of 1:1. 选取广西环江县喀斯特峰丛洼地:草丛(T)、灌丛(S)、原生林(PF)(中坡位)不同植被类型,原生林上、中、下不同坡位,按土壤发生层采集淋溶层(A层,0-10 cm)、过渡层(AB层,20-30 cm,草丛和灌丛;30-50 cm,原生林)、淀积层(B层,70-100cm)样品,研究土壤微生物量碳、氮(Soil microbial biomass carbon (SMBC)、soil microbial biomass nitrogen (SMBN))、微生物碳熵、氮熵(ratio of SMBC to soil organic carbon (qMBC)、ratio of SMBN to soil total nitrogen (qMBN))、土壤基础呼吸(soil basic respiration (SBR))以及代谢熵(microbial metabolic quotient(qCO2))的剖面分异特征及其影响因素.结果表明,植被、土层深度显著影响土壤微生物量及基础呼吸,随植被恢复,SMBC、SMBN、SBR由草丛、灌丛、原生林依次上升,并随土壤发生层位的加深逐渐减少,qCO2在3种植被类型间差异显著:T>PF>S;原生林A层SMBC,SMBN在各坡位间均显著高于AB层、B层,SBR在A层由下坡位至上坡位递减,而在AB和B层的上、下坡位间无显著差异,qCO2坡位间无显著差异(P>0.05);SMBC与SMBN之间存在显著正相关(r=0.825,P<0.01,n=45),且SMBC、SMBN、SBR分别与有机碳、全氮、碱解氮均呈显著正相关.因此,随植被恢复,土壤质量明显改善,且坡位对A层土壤的影响较AB层和B层更显著,对于维持土壤微生物调节的土壤养分循环功能,调控土壤氮素营养与土壤有机质同等重要,这为合理制订喀斯特生态恢复措施提供了理论依据.  相似文献

【目的】研究34株不同来源罗伊氏乳杆菌(Lactobacillus reuteri)基因多样性及生境适应性机制,了解该菌株在肠外生境及肠内生境中适应性的异同,为L. reuteri优良菌株的开发提供理论基础。【方法】基于二代测序平台对11株源于发酵食品(酸马奶、酸粥)的L. reuteri进行测序,并应用比较基因组学将其与发酵食品、酸面团、食草动物L. reuteri分离株基因组进行比较分析。【结果】分离自酸马奶、酸粥L. reuteri基因组大小平均为2.14 Mb,GC含量平均为38.77%,且同种来源分离株系统发育关系距离较近。泛-核心基因集分别包含7242个、969个基因家族,其中酸马奶分离株特异性基因最多为459个。功能注释结果显示不同来源菌株碳水化合物、氨基酸相关基因数量及种类差异较大,仅在发酵食品和食草动物株中发现抗生素耐药性基因。注释到的碳水化合物活性酶中仅出现在发酵食品与食草动物分离株的为GH3 (β-葡萄糖苷酶等)和GH43 (β-木糖苷酶等),特有的分别为AA3 (纤维二糖脱氢酶等)和GH66 (葡萄糖转移酶等)。【结论】不同来源L. reuteri具有广泛的基因...  相似文献

In industry Escherichia coli is the preferred host system for the heterologous biosynthesis of therapeutic proteins that do not need posttranslational modifications. In this report, the development of a robust high-cell-density fed-batch procedure for the efficient production of a therapeutic hormone is described. The strategy is to guide the process along a predefined profile of the total biomass that was derived from a given specific growth rate profile. This profile might have been built upon experience or derived from numerical process optimization. A surprisingly simple adaptive procedure correcting for deviations from the desired path was developed. In this way the batch-to-batch reproducibility can be drastically improved as compared to the process control strategies typically applied in industry. This applies not only to the biomass but, as the results clearly show, to the product titer also.  相似文献

Phagotrophic phytoflagellates in microbial food webs   总被引:5,自引:2,他引:5  
Phagotrophy by pigmented flagellates is known from the literature but has recently been rediscovered in the context of microbial food webs. Particle ingestion rates were found to be equivalent for pigmented and nonpigmented microflagellates in both field and laboratory studies. Ingestion rates of the chrysophytes Ochromonas danica, O. minuta, and Poterioochromonas malhamensis, the dinoflagellate Peridinium inconspicuum, and the cryptophytes Cryptomonas ovata and C. erosa were compared with those of two nonpigmented Monas species using 0.57 μm polystyrene beads as a food source. Ingestion rates were 0.31 to 3.17 beads/cell/h and filtration rates were 10−7 to 10−8 ml/cell/h with no detectable difference between pigmented and nonpigmented forms. Ingestion rates in unpigmented Monas species showed a linear increase with increasing particle concentration from 1.9 × 106 to 1.6 × 107 beads/ml. Light and DOC levels in the range of those encountered by phytoflagellates in the field also influenced laboratory measurements of bead ingestion by Poterioochromonas malhamensis. Ingestion rates decreased and photosynthesis increased over the natural PAR light range from 0 to 1800 microeinsteins/s/m2. At 40 microeinsteins/s/m2 maximum ingestion rates and high rates of photosynthesis occurred simultaneously. Ingestion rates decreased above 4 mgC/l supplied as glucose. DOC levels commonly occurring in Lake Oglethorpe range from 3.5 to 10.0 mgC/l. These studies suggest that mixotrophy, the trophic utilization of particulate food and dissolved organic matter as well as photosynthetically fixed organic matter, is a balanced process that can be regulated by environmental conditions. In field studies during a chrysophyte bloom, phytoflagellate grazing exceeded heterotrophic microflagellate grazing and constituted up to 55% of the bactivory of all microflagellates, ciliates, rotifers, and crustaceans combined. Neither bacterial abundance, light nor temperature were good predicters of grazing rates for the phagotrophic phytoflagellate association as a whole during this unstratified period. 【目的】研究15株分离自自然发酵食品的粪肠球菌(Enterococcus faecalis)和1株粪肠球菌模式株对15种(1种β-内酰胺类和14种非β-内酰胺类)抗生素的耐药性,并通过菌株耐药表型与基因型的关联分析找到粪肠球菌潜在的耐药基因。【方法】利用微量肉汤稀释法检测试验菌株对15种抗生素的耐药性,运用Scoary软件进行表型与基因型的关联分析。【结果】16株粪肠球菌对卡那霉素、万古霉素、利奈唑胺和红霉素100%敏感;对其余11种抗生素均表现出不同程度的耐药,其中对克林霉素为100%耐药。基因组关联分析发现了9个功能基因与5种抗生素(氯霉素、环丙沙星、甲氧苄啶、新霉素和四环素)存在显著相关关系,其中基因FAM000296和FAM005768与氯霉素、甲氧苄啶和环丙沙星的耐药性有关。进一步分析发现基因FAM000296和FAM005768同被注释为Sec G,但基因FAM005768与基因FAM000296相比在3′端丢失了21个碱基。【结论】分离自自然发酵食品的粪肠球菌对多种抗生素具有耐药性,其基因组中携带有潜在的耐药基因。因此,对分离自自然发酵食品的粪肠球菌需要进行全面的安全性评估后方可考虑应用。  相似文献

The microbial communities of freshwater hot spring mats from Boekleung (Western Thailand) were studied. Temperatures ranged from over 50 up to 57°C. Green-, red-, and yellow colored mat layers were analyzed. In order to detect the major components of the microbial communities constituting the mat as well as the microorganisms showing significant metabolic activity, samples were analyzed using DNA- and RNA-based molecular techniques, respectively. Microbial community fingerprints, performed by denaturing gradient gel electrophoresis (DGGE), revealed clear differences among mat layers. Thermophilic phototrophic microorganisms, Cyanobacteria and Chloroflexi, constituted the major groups in these communities (on average 65 and 51% from DNA and RNA analyses, respectively). Other bacteria detected in the mat were Bacteroidetes, members of the Candidate Division OP10, Actinobacteria, and Planctomycetes. Differently colored mat layers showed characteristic bacterial communities and the major components of the metabolically active fraction of these communities have been identified.  相似文献

The volatile organic compound composition (VOCs) of printing press air wasfound to contain mostly ethanol, but also ethyl acetate, 1-propanol, 2-propanol,1-methoxy-2-propanol and 3-ethoxy-1-propanol. A pilot plant bioscrubber inoculatedwith a mixed microbial population was constructed on-site. The bioscrubber wasable to treat the polluted gas efficiently. It, however, suffered from strong wall growthand blockages in the column. The efficiencies of the pilot plant and a bioreactor iscompared. The yeasts Candida guilliermondii and Saccharomyces cerevisiae known to tolerate ethanol were selected instead of mixed population to avoid the wall growth and blockages in the bioreactor. The removal of the VOCs both individually and as a complex mixture was tested in a microcultivation system and in continuous chemostat cultures with and without cell recycling. The Candida yeast could use all the compounds as a carbon source while growth of S. cerevisiae was markedly slower on the methoxylated and ethoxylated propanols. Best total removal of the VOCs was 99% and achieved by C. guilliermondii. The only compound that was not totally removable in the chemostat experiment with C. guilliermondii was 1-methoxy-2-propanol. In laboratory scale the total and volumetric removal of VOCs by C. guilliermondii was more efficient compared to the pilot plant encouraging to scale up and applying the yeast bioreactor to real field conditions.  相似文献

Yeast strains with amylolytic activity were isolated from cassavatapé and its precursor,ragi. they were divided into two groups based on their characteristics: group 1, possessing high amylolytic activity and low ethanol yield; and group 2, possessing low amylolytic activity and high ethanol yield. The major strains of the group 1 were identified asEndomyces fibuliger, and those of group 2 were identified asPichia anomala. Based on 18S rDNA analysis, an isolate fromragi that had a high amylolytic activity was thought to be an undescribed species that was related to the basidiomycetous genera.  相似文献

Intra-population quinoline alkaloid profiles surveying quinine, quinidine, cinchonine and cinchonidine were determined for each of five populations of Cinchona ledgeriana grown as shoot-culture for 125 days. No significant difference in respect of mean alkaloid content between populations was detected. In contrast, there was considerable between-seedling variation in alkaloid content within each population. When nutrients were delivered to shoot-cultures in droplet form by means of an aerosol spray (as compared to the supply of nutrients direct from agar-or liquid-based reservoirs) alkaloid profile was greatly perturbed; most notable in this respect was a four-fold increase in the production of cinchonidine concomitant with a four-fold decrease in the production of cinchonine. These data are discussed with reference to the optimisation of quinoline alkaloid production by juvenile shoot-cultures of Cinchona ledgeriana.  相似文献

Summary Using 15 Chinese and Japanese cultivars of sweetpotato, Ipomoea batatas (L.) Lam., we succeeded in developing an efficient plant regeneration system from embryogenic suspension cultures. The embryogenic callus derived from shoot apices of the 15 cultivars was used to initiate embryogenic suspension cultures in Murashige and Skoog (MS) medium containing 9.05 μM 2,4-dichlorophenoxyacetic acid (2,4-D). Rapidly proliferating and well-dispersed embryogenic suspension cultures were established. Cell aggregates 0.7–1.1 mm in size from embryogenic suspension cultures were transferred to solid MS medium supplemented with 9.05 μM of 2,4-D and formed embryogenic callus with somatic embryos. The embryogenic callus with somatic embryos was further transferred to MS medium supplemented with 3.78 μM of abscisic acid, resulting in the germination of somatic embryos. Within 20 wk after the initiation, the frequencies of cell aggregates forming plantlets reached approximately 100% for the 15 tested cultivars. These plantlets, when transferred to soil, showed 100% survival. No morphological variations were observed.  相似文献
