The occurrence of entomopathogenic fungi was investigated in irrigated vegetable fields and citrus orchards soils, over a nine-month period (April-December 1999), using the Galleria bait method (GBM). Entomopathogenic fungi were found to occur in 33.6% of the soil samples studied, with positive samples yielding 70 fungal isolates, belonging to 20 species from 13 genera. Conidiobolus coronatus was the most frequent and abundant entomopathogenic species recovered, comprising 31.4% of the total number of isolates. Soil pH, soil moisture content and the geographical location had minor or no effect on the isolation of entomopathogenic fungi in the fields studied. On the other hand, organic matter content of soil, and vegetation type were found to significantly affect the occurrence of entomopathogenic fungi in soil habitats, with orchard fields yielding larger numbers of isolates than vegetable fields. Using Koch's postulates the pathogenicity of fungal isolates to Galleria larvae was found to range from 16-100% (mortality rate). Isolates of C. coronatus proved to be the most virulent isolates recovered. The effect of media and temperature on mycelial growth rate, conidial production and conidial germination of six entomopathogenic fungal species (C. coronatus, Entomophaga grylli, Erynia castrans, Hirsutella jonesii, Paecilomyces farinosus and Sporodiniella umbellata) was also studied. Mycelial growth rate, spore production and spore germination were significantly affected by media, temperature and isolates. In view of the present results, C. coronatus appears to be a good candidate for pest control in agricultural soils, as it has a wide tolerance to agricultural practices, has frequently been isolated from both vegetable and orchard fields, and is characterized by high mycelial growth rate, conidial production and conidial germination.  相似文献

In wheat, corn and sorghum grains twenty-six genera and seventy-seven species including sixteen ofAspergillus and twenty-one ofPenicillium were identified.In grain samples adjusted to various moisture levels (up to 18.8 % on a dry-weight basis) and stored for 4 months at 8, 20 and 30 C seed-borne fungi were monthly identified and counted, and the germinability of the grains was tested. In the control samples (moisture content 7.1–8.2 %) temperature had no effect on the fungal counts and on the precentage germination. With the rise of temperature and moisture content the fungal counts markedly increased and the germinability declined.The list of fungi dominant in one or more of the experiments, included eight species ofAspergillus, six ofPenicillium, two ofFusarium and one each ofAlternaria andCurvularia. The order of dominance of these fungi varied according to the type of grain, the length of storage and the treatment.A. niger was the dominant organism in the control samples of the three grains. It could grow successfully at high moisture contents but above 15 %, it was usually overgrown by other fungi. In wheatP. citrinum andA. sydowii, in cornP. citrinum andA. terreus, and in sorghumA. terreus andA. niger were the dominant fungi at moisture contents above 15 % after four months storage at 30 C. When stored at 8 CPenicillium species tended to predominate over other fungi in grains with high moisture contents.In surface-sterilized grains adjusted to 15 % moisture content, inoculated with six dominant fungi separately and incubated at 30 C,A. niger, P. citrinum andP. variabile caused severe deterioration in the three grains;A. ochraceus in wheat only andF. moniliforme in wheat and corn;C. spicifer was slightly injurious to the three grains.  相似文献

In sorghum seed stored at 22–25° C for 535 days, invasion by storage fungi and loss of germinability increased greatly with small increases in moisture content between 13.5 and 15.5 %. Seeds dried for 18 hours at 70° C, then exposed to a relative humidity of 75 %, had a lower equilibrium moisture content, but were more heavily invaded by storage fungi and lost germinability fsater, than those that had been conditioned to 20 % moisture before storage at 75 % relative humidity. The 10 samples of Grade No. 2 sorghum examined averaged about 13.0 % moisture, germinated an average of 59 %, yieldedAlternaria from 75 % of the surface-disinfected kernels andAspergillus glaucus from 4 %; judged by these criteria, the lots from which the samples came were in good condition for continued storage.  相似文献

In peanut seeds adjusted to various levels of moisture content (8.5 %, 13,5 %, 17.5 % and 21 % on a dry-weight basis) and stored for six months at 5°, 15°, 28° and 45 °C, seed-borne fungi were monthly identified and counted using the dilution plate method, and the germinability of seeds was tested. The total count of seed-borne fungi (recovered at 28 °C) increased regularly and the germinability declined with the rise moisture content and with the lengthening of the storage period. At 5°, 15° and 28 °C,A. fumigatus was almost the most dominant fungal species followed by several fungi such asAspergillus flavus, A. niger, A. terreus, Penicillium funiculosum, P. notatum, Pyrenochaeta decipians andScopulariopsis brevicaulis. The degree of dominance of each fungal species depended upon the conditions of storage and the length of storage period.A. flavus gained its highest count in seeds adjusted to 13.5 % stored at 15 °C for 1 month.Penicillium predominated at 17.5 % and 21 %, and at 13.5 % and 17.5 % when the seeds were stored for long periods at 5° and 15 °C respectively.The total count of thermophilic fungi (recovered at 45 °C) significantly increased with the rise of moisture content and with the lengthening of the storage period between 1 and 2 months and regularly declined after 4 months.A. fumigatus was so extremely dominant that it was the main component of the total fungal flora. Several fungi truly thermophilic were isolated also. Pollen grains may become desiccated after independence from parent plants and remain viable in an inactive dry state during presentation and dispersal, until the conditions for rehydration and germination are prepared. But some pollen types do not tolerate the desiccation state and lose the germination power soon after release, and therefore, are difficult to store. In this study, moisture content, germinability, cytology and dehydrin and phenolics contents were surveyed in pistachio pollen at fresh and desiccated states. Mature pollen lost 54.1% of its initial moisture after 48 h desiccation along with severe decrease in germinability. Light microscopy results indicated that a low rate of pollen grains have vegetative cell rupture caused by desiccation, but a higher rate of grains were intact in appearance. Numerous amyloplasts persisted after desiccation as a sensitivity indicator. A 16 kDa dehydrin band was detected by western blot method with higher content in desiccated than fresh samples. High performance liquid chromatography (HPLC) analysis showed that the total content of phenolics increased slightly by desiccation. These results indicate the insufficiency of dehydrin and phenolics accumulation for achievement of desiccation tolerance. Furthermore, the severe loss of germinability in desiccated pistachio pollen may be the result of deficiency in some other protective mechanisms that need further investigations.  相似文献

The variation in seed moisture, mould content and seed germination of two species of melon,Citrullus lanatus L. andCitrullus vulgaris Shrad. stored in jute bags was monitored at intervals of 2 months for one year. Seed moisture decreased in the first 6 months of storage followed by a gradual increase in the rainy months. The field fungi predominated immediately after harvest and disappeared completely after 6 months of storage. The storage fungi increased in both species and abundance throughout the storage period.Citrullus lanatus maintained high germination percentage for the first 4 months,C. vulgaris for the first 2 months, thereafter seed germinability decreased with prolonged storage.  相似文献

In plant species that rely on mycorrhizal symbioses for germination and seedling establishment, seedling recruitment and temporal changes in abundance can be expected to depend on fungal community composition and local environmental conditions. However, disentangling the precise factors that determine recruitment success in species that critically rely on mycorrhizal fungi represents a major challenge. In this study, we used seed germination experiments, 454 amplicon pyrosequencing and assessment of soil conditions to investigate the factors driving changes in local abundance in 28 populations of the orchid Neottia ovata. Comparison of population sizes measured in 2003 and 2013 showed that nearly 60% of the studied populations had declined in size (average growth rate across all populations: ?0.01). Investigation of the mycorrhizal fungi in both the roots and soil revealed a total of 68 species of putatively mycorrhizal fungi, 21 of which occurred exclusively in roots, 25 that occurred solely in soil and 22 that were observed in both the soil and roots. Seed germination was limited and significantly and positively related to soil moisture content and soil pH, but not to fungal community composition. Large populations or populations with high population growth rates showed significantly higher germination than small populations or populations declining in size, but no significant relationships were found between population size or growth and mycorrhizal diversity. Overall, these results indicate that temporal changes in abundance were related to the ability of seeds to germinate, but at the same time they provided limited evidence that variation in fungal communities played an important role in determining population dynamics.  相似文献

Six lots of wheat from six farmers' bins in Manitoba were adjusted to a range of 5 moisture contents, held at 6 temperatures and sampled at 6 times during storage. After sampling, seeds were surface sterilized with mercuric chloride and subsamples plated on Czapek's agar, on filter paper moistened with water, or on filter paper moistened with a 7.5% sodium chloride solution. The microflora on the seed was determined, germination counts were made, and the seed was examined by government inspectors for its condition and grade. A total of 1192 samples were examined. In addition, 180 samples were subjected to a fat acidity test. Deterioration for each combination of temperature and moisture, the fungi involved, and the consequent effect on condition, grade and fatty acid content were observed. The effect of high moisture content (>20%) and low temperatures (3–10 ° C) on infection of seed by Penicillium and the consequent effect on germination, condition, grade and fatty acid value are stressed. The interrelationship among fungi, and among fungi and temperature, moisture, storage time, fat acidity values and germination are indicated by correlation coefficient matrices.Deceased July 25, 1982.  相似文献

Typically, studies on indoor fungal growth in buildings focus on structures with known or suspected water damage, moisture, and/or indoor fungal growth problems. Reference information on types of culturable fungi and total fungal levels are generally not available for buildings without these problems. This study assessed 50 detached single-family homes in metropolitan Atlanta, Ga., to establish a baseline of “normal and typical” types and concentrations of airborne and dustborne fungi in urban homes which were predetermined not to have noteworthy moisture problems or indoor fungal growth. Each home was visually examined, and samples of indoor and outdoor air and of indoor settled dust were taken in winter and summer. The results showed that rankings by prevalence and abundance of the types of airborne and dustborne fungi did not differ from winter to summer, nor did these rankings differ when air samples taken indoors were compared with those taken outdoors. Water indicator fungi were essentially absent from both air and dust samples. The air and dust data sets were also examined specifically for the proportions of colonies from ecological groupings such as leaf surface fungi and soil fungi. In the analysis of dust for culturable fungal colonies, leaf surface fungi constituted a considerable portion (>20%) of the total colonies in at least 85% of the samples. Thus, replicate dust samples with less than 20% of colonies from leaf surface fungi are unlikely to be from buildings free of moisture or mold growth problems.  相似文献

AM真菌是一类广泛分布的土壤真菌,与宿主植物形成共生结构后,对于植物生长和植被恢复有多种有益的生理学和生态学作用。1999年11月至2000年10月,通过每月分别从0-10cm和10-20cm土层采集根际土样,对以色列荒漠地区丛枝霸王(Zygophyllum dumosum Boiss)根际AM真菌进行了系统的生态学研究。AM真菌的分布和定殖与季节变化和采样浓度密切相关。菌根真菌的最高定殖率并不伴随有最大的孢子密度,最高的定殖率发生在1999年11月,而最大的孢子密度则出现在2000年9月。10-20cm土层中的菌根真菌定殖率和孢子密度明显高于0-10cm土层。土壤温度与所有菌根结构的定殖率呈正相关,土壤有机质含量与泡囊和丛枝定殖率呈正相关,而土壤总可溶性氮对泡囊和丛枝定殖有显著正效应,对孢子密度有显著负效应。结果建议,孢子密度、泡囊和丛枝定殖程度可作为检测荒漠土壤状况的生态指标。研究应用于我国特别是西部荒漠地区的植被建立和恢复,可望发挥重任作用:(1)AM真菌能与绝大多数高奶系形成共生联合体,促进根系对土壤矿质营养和水分的吸收,提高植物对干旱、高温、盐碱、根部病害等的抗性,提高逆境条件下植物的成活率,深入研究荒漠生态系统中AM真菌动态分布,以及筛选优势AM菌种和人工接种,进行菌根化育苗,为植被建立和恢复提供优质苗木;(2)菌根的丛枝定殖时间短,主要发生在幼根,泡囊定殖时间长,主要发生在老根,而AM真菌的生长发育和繁殖所需的碳水化合物来自植物根系的分泌活动,所以,通过检测不同时期菌根各种结构的定殖程度和孢子的丰富度,可以获得宿主植物根系的生长状况,进而对土壤环境作出科学的评估。  相似文献

The fungal odor compounds 3-methyl-l-butanol, l-octen-3-ol and 3-octanone were monitored in nine experimental bins in Winnipeg, Manitoba containing a hard red spring wheat during the autumn, winter and summer seasons of 1984–85. Quality changes were associated with seed-borne microflora and moisture content in both ventilated and non-ventilated bins containing wheat of 15.6 and 18.2% initial moisture content. All three odor compounds occurred in considerably greater amounts in bulk wheat in non-ventilated than in ventilated bins, particularly in those with wheat having 18.2% moisture content. The presence of these compounds usually coincided with infection of the seeds by the fungi Alternaria alternata (Fr.) Keissler, Aspergillus repens DeBarry, A. versicolor (Vuill.) Tiraboschi, Penicillium crustosum Thom, P. oxalicum Currie and Thom, P. aurantiogriseum Dierckx, and P. citrinum Thom. High production of all three odor compounds in damp wheat stored in non-ventilated bins was associated with heavy fungal infection of the seeds and reduction in seed germinability. High initial moisture content of the harvested grain accelerated the production of all three fungal volatiles in non-ventilated bins.  相似文献

In bromeliads, nothing is known about the associations fungi form with seeds and seedling roots. We investigated whether fungal associations occur in the seeds and seedling roots of two epiphytic Aechmea species, and we explored whether substrate and fungal associations contribute to seed germination, and seedling survival and performance after the first month of growth. We found a total of 21 genera and 77 species of endophytic fungi in the seeds and seedlings for both Aechmea species by Illumina MiSeq sequencing. The fungal associations in seeds were found in the majority of corresponding seedlings, suggesting that fungi are transmitted vertically. Substrate quality modulated the germination and growth of seedlings, and beneficial endophytic fungi were not particularly crucial for germination but contributed positively to survival and growth. Overall, this study provides the first evidence of an endophytic fungal community in both the seeds and seedlings of two epiphytic bromeliads species that subsequently benefit plant growth.  相似文献

This paper deals with investigations on fungal infection, moisture content, germinability and deterioration of three seeds, viz., maize (starchy), groundnut (oily) and soybean (proteinaceous) in storage at the locality of Santiniketan, West Bengal, India, under natural condition for 1 year. The airspora of storage environment was trapped using culture plate method. Different species of Aspergillus (A. candidus, A. flavus, A. niger, A. terreus, and A. ruber) were dominant followed by Rhizopus, Penicillium, Curvularia, Fusarium, Alternaria, etc. Seed moisture was maximum in the rainy season followed by a gradual decrease during longer storage. A gradual decrease in field fungi with simultaneous increase in storage fungi accompanied by a reduction in germinability occurred in all seeds as storage proceeded. A gradual loss of carbohydrate (both soluble and insoluble) content in all the seeds were recorded. A loss of protein content was recorded followed by a small increase. Oil content decreased in prolonged storage with simultaneous increase in fatty acid. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Hydration offers an effective means for raising seed performance in many crop species. The objective of this study was to evaluate the effect of vermiculite hydration on germinability and several physiological activities related to vigor in artificially aged watermelon seeds differing in ploidy. Aging was achieved by incubating the seeds at 45°C and 79% relative humidity for 6 days, then the seeds were air-dried to their original moisture level (4.7%). Hydration was achieved by mixing the untreated and aged seeds with moist vermiculite No. 3 at 25°C for 24 h. The partially hydrated seeds were air-dried at 25°C for 36 h to 4.7% moisture level. Significant differences existed between unaged and aged seeds, with lower germination percentage and slower germination speed in the latter. Aging also increased lipid peroxidation and reduced the activity of peroxide-scavenging enzymes. The germinability of aged watermelon seed was restored partially by vermiculite hydration. The activities of protein synthesis and peroxide-scavenging enzymes in axis and cotyledon portions of the seeds were also increased by hydration treatment. The changes in germination and related physiological responses in relation to aging and hydration are similar in seeds differing in ploidy, despite differences in their germination performance, seed leakage, extent of lipid peroxidation and activities of peroxide-scavenging enzymes.  相似文献

To prevent indoor fungal growth, understanding the moisture relations of fungi is a key element. Indoor moisture is quantified by the relative humidity (RH). RH controls the water activity of the indoor materials that fungi grow on, a well‐studied parameter known to limit fungal growth. RH, however, also controls the amount of water present in these materials, the moisture content. The significance of the moisture content of these materials to indoor fungal growth is currently overlooked. In the work reported here, growth experiments with the indoor fungus Penicillium rubens on gypsum substrates were performed to test whether the moisture content influences growth on porous materials. Second, we report the development of a video microscopy method that for the first time quantified hyphal growth on a porous material. It is found that a higher moisture content leads to earlier colonization and higher hyphal extension rates. This is a fundamental step in unravelling the effect of RH on indoor fungal growth. The real‐time monitoring of colonization of gypsum provides a new view of growth on indoor surfaces.  相似文献

Precise knowledge of seed quality after harvest and during storageis of particular importance for seed producers. We analyseddifferent sunflower seed lots (Helianthus annuusL.) characterizedby extremes of germination ability. We used RNA analysis tostudy possible changes in gene expression in seeds unable togerminate. Total RNA content was very small in dry seeds showinga low germination ability. Capacity for total RNA synthesisat the onset of imbibition was also reduced in these seeds.In addition, correlations were found between these parametersand germination ability at 19 °C. We demonstrated a highcorrelation between the amount of total RNA in the dry seed,the capacity of RNA synthesis at the onset of imbibition andthe seed moisture content at the time of the harvest. The abilityof dry seed mRNAs to be translatedin vitrowas also reduced andseven polypeptides, from stored mRNAs, were characteristic ofthe cotyledons from high germinability seeds. During steeping and germination in micromalting equipment, the number of colonies of filamentous fungi increased from two to five times, colonies of yeasts from five to ten times, and bacteria from 50 to more than 100 times the numbers present in the grain before malting. Kiln drying according to a commercial schedule reduced the number of all types of microflora below the number present before kilning, but all were present in larger numbers in the kilned malt than in the original grain. In barley stored at room temperature and at a moisture content of 15 to 18%, members of the Aspergillus glaucus group increased with increasing time and increasing moisture content, and germination percentage of the seeds decreased. Stored free of storage fungi at room temperature, barley with a moisture content just over 15% retained a high germination percentage for 5 months, but at a moisture content of 16% the germination decreased to zero.  相似文献
