Soil is a reservoir for Aspergillus flavus and A. parasiticus, fungi that commonly colonize peanut seeds and produce carcinogenic aflatoxins. Densities of these fungi in soil vary greatly among fields and may influence the severity of peanut infection. This study examined the relationship between soil density of Aspergillus species and the incidence of peanut seed colonization under laboratory conditions. Viable peanut seeds were wounded and inoculated with 20 soils differing in composition and density of Aspergillus species and were then incubated for 14 days at 37 degrees C (seed water activity = 0.92). The effect of soil density of individual section Flavi species (A. flavus strains L and S, A. parasiticus, A. caelatus, and A. tamarii), section Nigri, and A. terreus on the incidence of seed colonization was best expressed as a function of exponential rise to maximum. Exponential curves often rose to maximum percentages of seed colonization by section Flavi species that were well below 100% despite high species densities in some soils. Competition primarily among section Flavi species may explain the reduced incidences of seed colonization. An average of two or fewer propagules of each Aspergillus species in the soil at the wound site was required for colonization of 20% of peanut seeds. Other fungal species were capable of invading peanut seeds only when soil densities of sections Flavi and Nigri species were low.  相似文献

Most aspergilli that produce aflatoxin are members of Aspergillus section Flavi, however isolates of several Aspergillus species not closely related to section Flavi also have been found to produce aflatoxin. Two of the species, Aspergillus ochraceoroseus and an undescribed Aspergillus species SRRC 1468, are morphologically similar to members of Aspergillus section Circumdati. The other species have Emericella teleomorphs (Em. astellata and an undescribed Emericella species SRRC 2520) and are morphologically distinctive in having ascospores with large flanges. All these aflatoxin-producing isolates were from tropical zones near oceans, and none of them grew on artificial media at 37 C. Aflatoxins and sterigmatocystin production were quantified by high-pressure liquid chromatography (HPLC) and confirmed by HPLC-mass spectrometry (LC-MS) detection. Phylogenetic analyses were conducted on these four species using A. parasiticus and Em. nidulans, (which produce aflatoxin and the aflatoxin precursor sterigmatocystin, respectively) for comparison. Two aflatoxin/sterigmatocystin biosynthesis genes and the beta tubulin gene were used in the analyses. Results showed that of the new aflatoxin-producers, Aspergillus SRRC 1468 forms a strongly supported clade with A. ochraceoroseus as does Emericella SRRC 2520 with Em. astellata SRRC 503 and 512.  相似文献

AIMS: The objectives of this study were: (i) to evaluate genetic relatedness among Aspergillus section Flavi strains isolated from soil and peanut seeds in Argentina; (ii) to determine if AFLP molecular markers could be useful to identify isolates up to species level, and to correlate these markers with the isolates' toxigenic potentials and/or vegetative compatibility group (VCG) affiliations. METHODS AND RESULTS: Amplified fragment length polymorphism (AFLPs) analysis was applied to compare 82 isolates of Aspergillus section Flavi. Cluster analysis showed a clear separation of A. flavus and A. parasiticus, and comparison of fingerprints revealed several specific markers for each group of isolates. AFLP analysis indicates that no genotypical differences can be established between aflatoxigenic and nonaflatoxigenic producers in both species analysed. In addition, candidate AFLP markers associated with a particular VCG were not found. CONCLUSIONS: There was a concordance between morphological identification and separation up to species level using molecular markers. The findings of specific bands for A. flavus and A. parasiticus may be useful for the design of specific PCR primers in order to differentiate these species and detect them in food. SIGNIFICANCE AND IMPACT OF THE STUDY: The present study provides new data on molecular characterization of Aspergillus section Flavi in Argentina.  相似文献

Three new aflatoxin-producing species belonging to Aspergillus section Flavi are described. They are Aspergillus mottae, A. sergii and A. transmontanensis. These species were isolated from Portuguese almonds and maize. An investigation examined morphology, extrolite production and DNA sequence data to characterize these isolates and describe the new species. Phylogenetic analysis showed that A. transmontanensis and A. sergii form a clade with A. parasiticus whereas A. mottae shares a most recent common ancestor with the combined A. flavus and A. parasiticus clade.  相似文献

AIMS: Populations of Aspergillus section Flavi were studied from a commercial field of maize in Río Cuarto, Córdoba, Argentina. METHODS AND RESULTS: The Aspergillus species were isolated from soil, debris and insects during three periods: pre-planting, growing maize and post-harvest. The colony count from non-rhizospheric soil in the pre-planting period was higher than in growing maize and the post-harvest period. Debris samples analysed during all periods showed similar infection percentages for Aspergillus section Flavi. The samples of insects collected during the maize-growing period showed a lower percentage of Aspergillus isolates than the samples from soil and debris. Aflatoxigenic strains were present in lower levels in each component of the agroecosystem studied. All the strains that produced sclerotia were L strains. CONCLUSIONS: In this field agroecosystem, the only strains with a high probability for transfer to the storage agroecosystem were L strains with low toxigenic potential. SIGNIFICANCE AND IMPACT OF THE STUDY: Maize pre-harvest contamination with aflatoxigenic inoculum was not significant.  相似文献

AIMS: The effectiveness of the food-grade antioxidants butylated hydroxytoluene (BHT), trihydroxybutyrophenone (THB), propyl paraben (PP) and butylated hydroxyanisole (BHA) at 1, 10 and 20 mmol l(-1) concentrations on germination, growth, and aflatoxin B(1) (AFB(1)) production by Aspergillus section Flavi strains was determined. METHODS AND RESULTS: Assays on the lag phase of germination, germination percentage, germ tube elongation rate, lag phase, growth rate and AFB(1) production by three strains of Aspergillus flavus and three of Aspergillus parasiticus were carried out in vitro on peanut extract meal agar conditioned at different water activities (a(w): 0.982, 0.971, 0.955, 0.937). The antioxidants PP and BHA efficiently inhibited the germination of the two species tested at the doses 10 and 20 mmol(-1). The antioxidants PP and BHA at 1 mmol l(-1) and THB at 20 mmol l(-1) reduced the germ tube elongation rate most effectively, regardless of a(w) levels. An increase in the lag time and a reduction in the growth rate of 100% of the strains was observed, this was due to the action of BHT at the doses 10 and 20 mmol(-1) at 0.982, 0.971 and 0.955 a(w), although these treatments stimulated the AFB(1) accumulation in most of the fungi tested. The more effective antioxidants were PP and BHA, which increased the lag phase, reduced the growth rate and AFB(1) production in all of the strains at the four a(w) assayed. At concentrations 10 and 20 mmol l(-1), these antioxidants totally inhibited fungal development. CONCLUSIONS: The study shows that the antioxidants BHA and PP are effective fungal inhibitors to peanut Aspergillus section Flavi in wide range of water activity. SIGNIFICANCE AND IMPACT OF THE STUDY: The results suggest that phenolic antioxidants, BHA and PP, can be effective fungitoxicants on aflatoxigenic strains in peanut at industrial level.  相似文献

The Aspergillus parasiticus aflR gene, a gene that may be involved in the regulation of aflatoxin biosynthesis, encodes a putative zinc finger DNA-binding protein. PCR and sequencing were used to examine the presence of aflR homologs in other members of Aspergillus Section Flavi. The predicted amino acid sequences indicated that the same zinc finger domain, CTSCASSKVRCTKEKPACARCIERGLAC, was present in all of the Aspergillus sojae, Aspergillus flavus, and Aspergillus parasiticus isolates examined and in some of the Aspergillus oryzae isolates examined. Unique base substitutions and a specific base deletion were found in the 5' untranslated and zinc finger region; these differences provided distinct fingerprints. A. oryzae and A. flavus had the T-G-A-A-X-C fingerprint, whereas A. parasiticus and A sojae had the C-C-C-C-C-T fingerprint at the corresponding positions. Specific nucleotides at positions -90 (C or T) and -132 (G or A) further distinguished A. flavus from A. oryzae and A. parasiticus from A. sojae, respectively. A sojae ATCC 9362, which was previously designated A. oryzae NRRL 1988, was determined to be a A. sojae strain on the basis of the presence of the characteristic fingerprint, A-C-C-C-C-C-C-T. The DNAs of other members of Aspergillus Section Flavi, such as Aspergillus nomius and Aspergillus tamarii, and some isolates of A. oryzae appeared to exhibit low levels of similarity to the A. parasiticus aflR gene since low amounts of PCR products or no PCR products were obtained when DNAs from these strains were used.  相似文献

Aims:  The antifungal effect of Pimpinella anisum (anise), Pëumus boldus (boldus), Mentha piperita (peppermint), Origanum vulgare (oregano) and Minthosthachys verticillata (peperina) essential oils against Aspergillus section Flavi (two isolates of Aspergillus parasiticus and two isolates of Aspergillus flavus ) was evaluated in maize meal extract agar at 0·982 and 0·955 water activities, at 25°C.
Methods and Results:  The percentage of germination, germ-tube elongation rate, growth rate and aflatoxin B1 (AFB1) accumulation at different essential oils concentrations were evaluated. Anise and boldus essential oils were the most inhibitory at 500 mg kg−1 to all growth parameters of the fungus. These essential oils inhibited the percentage of germination, germ-tube elongation rate and fungal growth. AFB1 accumulation was completely inhibited by anise, boldus and oregano essential oils. Peperina and peppermint essential oils inhibited AFB1 production by 85–90% in all concentrations assayed.
Conclusions:  Anise and boldus essential oils could be considered as effective fungitoxicans for Aspergillus section flavi .
Significance and Impact of the Study:  Our results suggest that these phytochemical compounds could be used alone or in conjunction with other substances to control the presence of aflatoxigenic fungi in stored maize.  相似文献

AIMS: Antagonist activity of Kluyveromyces spp. isolates on Aspergillus section Flavi was studied. METHODS AND RESULTS: The screening of isolates were made through studies of growth at different water activities and temperatures, index of dominance (I(D)), ecological similarity, antifungal activity and impact on aflatoxin B1 accumulation. High optical density was obtained at 25 and 30 degrees C and 48 h of incubation. Cell growth decreases with decrease in water activity. The predominant interaction was mutual intermingling at a(w) = 0.982 and 0.955, while at a(w) = 0.999 and 0.937 mutual inhibition for contact was exhibited. All isolates were catabolically identical to Aspergillus section Flavi and compete by nutritional source. At high water activities yeasts showed inhibitory activity on Aspergillus strains, inhibition percentages varied between 75 and 100%. The isolates Y9, Y14, Y16, Y22, Y25 and Y33 showed antifungal activity and inhibitory activity on aflatoxin B1 accumulation at all water activities assayed from all Aspergillus section Flavi strains. CONCLUSIONS: The data show that the isolates selected in a wide range of environmental conditions could exert their roll like biological control agents for Aspergillus section Flavi in storage maize ecosystem. SIGNIFICANCE AND IMPACT OF THE STUDY: Isolates of Kluyveromyces spp. may have practical value in the postharvest control of storage maize.  相似文献

Peanuts and other seed and grain crops are commonly contaminated with carcinogenic aflatoxins, secondary metabolites produced by Aspergillus flavus and A. parasiticus. Aflatoxin contamination of peanuts in the field can be reduced by 77–98% with biological control through the application of nontoxigenic strains of these species, which competitively exclude native aflatoxin-producing strains from developing peanuts. In this study, viable peanut seeds were artificially wounded and inoculated with field soil containing natural fungal populations that were supplemented with conidia of nontoxigenic A. flavus NRRL 21882 (niaD nitrate-nonutilizing mutant) and A. parasiticus NRRL 21369 (conidial color mutant). Increasing soil densities of applied nontoxigenic strains generally resulted in an increase in the incidence of seed colonization by applied nontoxigenic strains, a decrease in seed colonization by native A. flavus and A. parasiticus, and a decrease in aflatoxin concentration in seeds. Reduction of aflatoxins in peanut seeds depended on both the density and the aflatoxin-producing potential of native populations and on the fungal strain used for biological control. Wild-type strain A. flavus NRRL 21882 and its niaD mutant were equally effective in reducing aflatoxins in peanuts, indicating that nitrate-nonutilizing mutants, which are easily monitored in the field, can be used for evaluating the efficacy of biocontrol strains.  相似文献

The objective of this study was to evaluate the ability of the Aspergillus flavus pAF28 DNA probe to produce DNA fingerprints for distinguishing among genotypes of Petromyces alliaceus (Aspergillus section Flavi), a fungus considered responsible for the ochratoxin A contamination that is occasionally observed in California fig orchards. P. alliaceus (14 isolates), Petromyces albertensis (one isolate), and seven species of Aspergillus section Circumdati (14 isolates) were analyzed by DNA fingerprinting using a repetitive sequence DNA probe pAF28 derived from A. flavus. The presence of hybridization bands with the DNA probe and with the P. alliaceus or P. albertensis genomic DNA indicates a close relationship between A. flavus and P. alliaceus. Twelve distinct DNA fingerprint groups or genotypes were identified among the 15 isolates of Petromyces. Conspecificity of P. alliaceus and P. albertensis is suggested based on DNA fingerprints. Species belonging to Aspergillus section Circumdati hybridized only slightly at the 7.0-kb region with the repetitive DNA probe, unlike the highly polymorphic hybridization patterns obtained from P. alliaceus and A. flavus, suggesting very little homology of the probe to Aspergillus section Circum dati genomic DNA. The pAF28 DNA probe offers a tool for typing and monitoring specific P. alliaceus clonal populations and for estimating the genotypic diversity of P. alliaceus in orchards, vineyards, or crop fields.  相似文献

Intensive studies of the mycoflora of Dixie Spanish peanuts grown under different combinations of tillage, organic matter, and previous crop revealed no consistent reduction in either Total Fungi,A spergillus spp.A. niger, A. flavus, Penicillium spp., orFusarium spp. for any of the 16 combinations studied. The level of fungous invasion in apparently sound peanut shells and seeds was consistently high, but the species composition of this flora changed as the season progressed.Penicillium was the most common genus, being isolated from over half the shells sampled. The level in seeds was lower, but in both shells and seeds there were morePenicillium spp. in fruits in the ground than in fruits from stacks.A spergillus spp. occurred at lower levels thanPenicillium, but they also decreased in fruits in the stack. Of particular interest is the fact that at all three collecting dates the amount ofAspergillus isolated from seeds was at least twice that from shells. The lower frequency of certain fungi in fruits from stacks is undoubtedly related to the lower moisture level in the drying fruits. Other fungi, especiallyFusarium spp., increase sharply in the dried tissues so that the overall level of invasion remains high. This may be of considerable practical importance since certain species ofFusarium are now known to produce toxins. Whether the increase in these fungi is due to their ability to grow under conditions of low moisture or is related to altered interspecific competition resulting from differences in the abilities of other species to survive the dry habitat, or both, is uncertain.
Zusammenfassung Eingehende Untersuchungen der Mykoflora der Dixie Spanish Erdnüsse unter verschiedenen Kombinationen von Ackern, organischem Material und vorherigem Anbau zeigten keine beständige Verminderung des gesamten Pilzgutes,Aspergillus spp.A. niger, A. flavus, Penicillium spp., oderFusarium spp., für keine der 16 Kombinationen. Der Grad der Pilzinvasion in anscheinend gesunden Erdnußhülsen und Samen war ständig hoch, aber die Artzusammensetzung dieser Flora wechselte mit der fortschreitenden Jahreszeit.Penicillium war die häufigste Gattung und war von der Hälfte der Hülsen isoliert. Der Grad in den Samen war niedriger, aber sowohl in Hülsen als auch in Samen war mehrPenicillium spp. in Früchten im Boden als in Früchten im Stapel.Aspergillus spp. kam zu einem niedrigeren Grade alsPenicillium vor, aber sie nahmen auch in Früchten im Stapel ab. Von besonderem Interesse ist die Tatsache, daß die Anzahl vonAspergillus spp., die von Samen isoliert worden sind, war mindestens das Zweifache von denen, die von Hülsen erhalten worden sind, an allen drei Sammeldaten. Die niedrigere Häufigkeit gewisser Pilze in Früchten vom Stapel ist zweifellos mit dem niedrigeren Feuchtigkeitsgrad der trockenen Früchte im Zusammenhang. Andere Pilze, insbesondereFusarium spp., nahmen im trockenen Gewebe stark zu, so daß der allgemeine Grad der Invasion hoch blieb. Dies mag von einer besonderen, praktischen Gewichtigkeit sein, da gewisse Arten vonFusarium bekannterweise Toxine produzieren. Ob die Zunahme dieser Pilze die Folge ihrer Fähigkeit ist, unter Bedingungen niedriger Feuchtigkeit zu wachsen oder sie mit dem veränderten interspezifischen Wettbewerb im Zusammenhang steht als Ergebnis der Unterschiede in der Fähigkeit anderer Pilze, in trockener Umgebung zu überleben oder beide, ist ungewiß.

Aspergillus takadae is characterized by its heterothallic reproduction, pale yellow cleistothecia, broadly lenticular ascospores with two short equatorial crests and smooth convex surfaces, and broadly ellipsoidal to ovate conidia with a smooth wall. The validation of these novel species is supported further by the analyses of the β-tubulin, calmodulin, actin, and RPB2 sequences. In addition, the phylogenetic tree and DDBJ accession numbers of all the species of Aspergillus section Fumigati are presented. We report on the crossing of A. takadae species and the result of crossing A. takadae with a closely related species, A. spathulatus.  相似文献

West African Aspergillus flavus S isolates differed from North American isolates. Both produced aflatoxin B1. However, 40 and 100% of West African isolates also produced aflatoxin G1 in NH4 medium and urea medium, respectively. No North American S strain isolate produced aflatoxin G1. This geographical and physiological divergence may influence aflatoxin management.  相似文献

Examination by scanning electron microscopy and incubation on potato-dextrose agar medium showed that dry seeds ofRetama raetam were externally free of fungi. When planted in sandy loam soil, the seeds become colonized with eleven soil-borne fungal species. The fungi were isolated on cellulose agar, pectin agar and lignin agar media.Aspergillus flavus, A. niger, A. fumigatus, Penicillium capsolatum andFusarium oxysporum had broad occurrence and were recovered on all the three media. The production of hydrolytic enzymes by the isolated fungi depends on the substrate and species.Penicillium capsolatum, P. spinulosum andA. niger had wide enzymatic amplitude and they were able to produce cellulolytic, pectolytic and lignolytic activities on corresponding substrates as well as on seed-coat-containing media. The lignolytic activities of the isolated species exceptChaetomium bostrychodes andTrichoderma viride were enhanced by applying the seed-coat materials as C- source rather than lignin. SoakingR. raetam seeds in culture filtrates of most of the fungi grown on seed-coat-supplemented media induced a pronounced and distinct stimulating effect on seed germination. The most effective filtrates were those ofP. capsolatum, P. spinulosum andSporotrichum pulverulentum.  相似文献

AIMS: To examine sclerotium characteristics of two Aspergillus flavus and two A. parasiticus strains at different growth media and water stress. METHODS AND RESULTS: The effects of growth media and water activity (0.999, 0.971, 0.955 and 0.937) on characteristics of sclerotia production (number, size and volume) of four isolates of Aspergillus section Flavi were examined. There was total inhibition under the driest conditions (0.955 and 0.937). When an osmotic potential of 0.971 was generated in Czapek agar (CD) and maize meal extract agar with sucrose and sodium nitrate (MMEA S/N), an increase in sclerotial size and volume was observed. The amount of sclerotia produced by cultures at 0.999 a(w) value was higher on CD. CONCLUSIONS: The data show that the sclerotia characteristics of A. flavus and A. parasiticus have been influenced by water availability and growth media composition. SIGNIFICANCE AND IMPACT OF THE STUDY: The information obtained shows that if we know the nutritional and water stress requirements for sclerotia production, it could be possible to develop effective prevention strategies to inhibit the survival of these fungi in grain.  相似文献

The substituted pterocarpenes named aracarpene-1 (1) and aracarpene-2 (2) were isolated from wounded peanut seeds challenged by a strain of Aspergillus caelatus. The structures of these putative phytoalexins were determined by interpretation of NMR and MS data. The aracarpenes were investigated for their antifungal and antibacterial activities as well as antioxidant, anti-inflammatory, and cytotoxic activities in mammalian cells. Aracarpene-2 demonstrated high antibacterial properties against tested gram-positive and gram-negative bacteria, whereas aracarpene-1 displayed low antibacterial properties against the same bacteria. Both compounds had no antifungal activity against Aspergillus flavus. Together with peanut stilbenoids that are also produced in the challenged seeds, these compounds may represent a class of low-molecular weight peanut metabolites with a defensive role(s) against pathogenic microorganisms.  相似文献

AIMS: The effect of osmotic and matric potential stress on growth and sugar alcohols (polyols: glycerol, erythritol, arabitol and mannitol) and sugars (trehalose and glucose) accumulation in toxigenic and nontoxigenic colonies of Aspergillus flavus and A. parasiticus was evaluated. METHODS AND RESULTS: Growth of Aspergillus section Flavi with significant reductions at 20 and 30 degrees C was more sensitive to changes in matric potential, between 60 and 100% in the range of -7 to -14 MPa. No significant differences were found between toxigenic and nontoxigenic strains for both species. Total polyol accumulation in unamended maize meal agar medium (-0.75 MPa water potential) was higher at 30 than 20 degrees C. The major change in concentrations of endogenous sugars and total polyols was in matrically amended medium (with PEG 8000) at -7 and -10 MPa. Accumulation of glucose, arabitol, mannitol and erythritol content of A. flavus and A. parasiticus mycelial colonies was greater in normal unstressed maize meal agar medium (-0.75 Mpa) at 20 degrees C. This was modified by solute and matric stress. CONCLUSIONS: The data showed relative sensitivity to osmotic and matric potential, and temperature, and the impact on growth rates, polyol and sugar accumulation in mycelia of A. flavus and A. parasiticus. SIGNIFICANCE AND IMPACT OF THE STUDY: The matric potential effects on growth may be of particular importance for growth and survival in environments with low-matric potential stress. The tolerance of spoilage fungi such as Aspergillus section Flavi to such modifications could increase the potential for spoilage and mycotoxin production in such substrates. This knowledge is important for understanding the relative ecological fitness of these aflatoxigenic species and in the development of prevention strategies for their control.  相似文献

Major insights into sexual development and cryptic sexuality within filamentous fungi have been gained from investigations using Aspergillus species. Here, an overview is first given into sexual morphogenesis in the aspergilli, describing the different types of sexual structures formed and how their production is influenced by a variety of environmental and nutritional factors. It is argued that the formation of cleistothecia and accessory tissues, such as Hülle cells and sclerotia, should be viewed as two independent but co-ordinated developmental pathways. Next, a comprehensive survey of over 75 genes associated with sexual reproduction in the aspergilli is presented, including genes relating to mating and the development of cleistothecia, sclerotia and ascospores. Most of these genes have been identified from studies involving the homothallic Aspergillus nidulans, but an increasing number of studies have now in addition characterized 'sex-related' genes from the heterothallic species Aspergillus fumigatus and Aspergillus flavus. A schematic developmental genetic network is proposed showing the inter-relatedness between these genes. Finally, the discovery of sexual reproduction in certain Aspergillus species that were formerly considered to be strictly asexual is reviewed, and the importance of these findings for cryptic sexuality in the aspergilli as a whole is discussed.  相似文献
