In C4 plants, photorespiration is decreased relative to C3 plants. However, it remains unclear how much photorespiratory capacity C4 leaf tissues actually have. We thoroughly investigated the quantitative distribution of photorespiratory organelles and the immunogold localization of the P protein of glycine decarboxylase (GDC) in mesophyll (M) and bundle sheath (BS) cells of various C4 grass species. Specific differences occurred in the proportions of mitochondria and peroxisomes in the BS cells (relative to the M cells) in photosynthetic tissues surrounding a vein: lower in the NADP-malic enzyme (NADP-ME) species having poorly formed grana in the BS chloroplasts, and higher in the NAD-malic enzyme (NAD-ME) and phosphoenolpyruvate carboxykinase (PCK) species having well developed grana. In all C4 species, GDC was localized mainly in the BS mitochondria. When the total amounts of GDC in the BS mitochondria per unit leaf width were estimated from the immunogold labeling density and the quantity of mitochondria, the BSs of NADP-ME species contained less GDC than those of NAD-ME or PCK species. This trend was also verified by immunoblot analysis of leaf soluble protein. There was a high positive correlation between the degree of granal development (granal index) in the BS chloroplasts and the total amount of GDC in the BS mitochondria. The classical definition of mesophyll conductance (g m) represents an apparent parameter (g m,app) as it places (photo)respired CO2 at the same compartment where the carboxylation by Rubisco takes place. Recently, Tholen and co-workers developed a framework, in which g m better describes a physical diffusional parameter (g m,dif). They partitioned mesophyll resistance (r m,dif = 1/g m,dif) into two components, cell wall and plasmalemma resistance (r wp) and chloroplast resistance (r ch), and showed that g m,app is sensitive to the ratio of photorespiratory (F) and respiratory (R d) CO2 release to net CO2 uptake (A): g m,app = g m,dif/[1?+?ω(F?+?R d)/A], where ω is the fraction of r ch in r m,dif. We herein extend the framework further by considering various scenarios for the intracellular arrangement of chloroplasts and mitochondria. We show that the formula of Tholen et al. implies either that mitochondria, where (photo)respired CO2 is released, locate between the plasmalemma and the chloroplast continuum or that CO2 in the cytosol is completely mixed. However, the model of Tholen et al. is still valid if ω is replaced by ω(1?σ), where σ is the fraction of (photo)respired CO2 that experiences r ch (in addition to r wp and stomatal resistance) if this CO2 is to escape from being refixed. Therefore, responses of g m,app to (F?+?R d)/A lie somewhere between no sensitivity in the classical method (σ =1) and high sensitivity in the model of Tholen et al. (σ =0).  相似文献

Physical, chemical, and regulatory properties of glycolate oxidase (GO) isolated from the leaves of C4 and C3 plants (Zea mays L., cv. Voronezhskaya 76 and Glycine max (L.) Merr., cv. Pripyat’, respectively) were studied. The homogenous preparations were obtained by multistage enzyme purification from soybean leaves and maize mesophyll and bundle sheath. The glycolate oxidase (GO) preparations obtained consisted of two types of subunits, 37 and 44 kD. The GO isolated from C3 plant leaves had many in common with that extracted from C4 plant bundle sheath as regards physical, chemical, and catalytic properties. The primary function of GO in both plant types is metabolism of glycolate, which is a product of ribulosebisphosphate oxalacetic acid oxidation and is used by plants for biosynthesis of hydrocarbons and amino acids.  相似文献

The increasing CO2 concentration in Earths atmosphere is expected to cause a greater decline in the nutritional quality of C3 than C4 plants. As a compensatory response, herbivorous insects may increase their feeding disproportionately on C3 plants. These hypotheses were tested by growing the grasses Lolium multiflorum C3) and Bouteloua curtipendula C4) at ambient (370 ppm) and elevated (740 ppm) CO2 levels in open top chambers in the field, and comparing the growth and digestive efficiencies of the generalist grasshopper Melanoplus sanguinipes on each of the four plant × CO2 treatment combinations. As expected, the nutritional quality of the C3 grass declined to a greater extent than did that of the C4 grass at elevated CO2; protein levels declined in the C3 grass, while levels of carbohydrates (sugar, fructan and starch) increased. However, M. sanguinipes did not significantly increase its consumption rate to compensate for the lower nutritional quality of the C3 grass grown under elevated CO2. Instead, these grasshoppers appear to use post-ingestive mechanisms to maintain their growth rates on the C3 grass under elevated CO2. Consumption rates of the C3 and C4 grasses were also similar, demonstrating a lack of compensatory feeding on the C4 grass. We also examined the relative efficiencies of nutrient utilization from a C3 and C4 grass by M. sanguinipes to test the basis for the C4 plant avoidance hypothesis. Contrary to this hypothesis, neither protein nor sugar was digested with a lower efficiency from the C4 grass than from the C3 grass. A novel finding of this study is that fructan, a potentially large carbohydrate source in C3 grasses, is utilized by grasshoppers. Based on the higher nutrient levels in the C3 grass and the better growth performance of M. sanguinipes on this grass at both CO2 levels, we conclude that C3 grasses are likely to remain better host plants than C4 grasses in future CO2 conditions.  相似文献

It is plausible that the nutritional quality of C3 plants will decline more under elevated atmospheric CO2 than will the nutritional quality of C4 plants, causing herbivorous insects to increase their feeding on C3 plants relative to C4 plants. We tested this hypothesis with a C3 and C4 grass and two caterpillar species with different diet breadths. Lolium multiflorum (C3) and Bouteloua curtipendula (C4) were grown in outdoor open top chambers at ambient (370 ppm) or elevated (740 ppm) CO2. Bioassays compared the performance and digestive efficiencies of Pseudaletia unipuncta (a grass-specialist noctuid) and Spodoptera frugiperda (a generalist noctuid). As expected, the nutritional quality of L. multiflorum changed to a greater extent than did that of B. curtipendula when grown in elevated CO2; levels of protein (considered growth limiting) declined in the C3 grass, while levels of carbohydrates (sugar, starch and fructan) increased. However, neither insect species increased its feeding rate on the C3 grass to compensate for its lower nutritional quality when grown in an elevated CO2 atmosphere. Consumption rates of P. unipuncta and S. frugiperda were higher on the C3 grass than the C4 grass, the opposite of the result expected for a compensatory response to the lower nutritional quality of the C4 grass. Although our results do not support the hypothesis that grass-specialist insects compensate for lower nutritional quality by increasing their consumption rates more than do generalist insects, the performance of the specialist was greater than that of the generalist on each grass species and at both CO2 levels. Mechanisms other than compensatory feeding, such as increased nutrient assimilation efficiency, appear to determine the relative performance of these herbivores. Our results also provide further evidence against the hypothesis that C4 grasses would be avoided by insect herbivores because a large fraction of their nutrients is unavailable to herbivores. Instead, our results are consistent with the hypothesis that C4 grasses are poorer host plants primarily because of their lower nutrient levels, higher fiber levels, and greater toughness.  相似文献

Although most fire research in plant ecology focuses on vegetation responses to burning, shifts in plant community composition wrought by climate change can change wildland fuelbeds and affect fire behaviour such that the nature of fire in these systems is altered. Changes that introduce substantially different fuel types can alter the spatial extent of fire, with potential impacts on community succession and biodiversity. Montane grasslands of sub-Saharan Africa are threatened by climate change because species distributions can shift with climatically determined ranges. We studied the impact of patches of the temperate C3 grass Festuca costata in C4-dominated grassland at the transition between their subalpine ranges in South Africa’s Drakensberg. We used empirical data on fuel moisture and fuel load across F. costata-dominated patches in a C4-dominated matrix in fire spread models to predict the effect of larger, higher-moisture F. costata patches on the spatial extent of fire. Results indicate F. costata reduces fire spread and burn probability in F. costata patches, and the effect increases as live fuel moisture increases and patches get larger. However, as a native species, F. costata does not appear to have the extreme, fire-suppressing effect of non-native C3 grasses in other C4 grasslands. Instead, F. costata patches likely increase variability in the spatial extent of fire in this C4-dominated grassland, which likely translates to spatial variability on vegetation succession.  相似文献

The complete set of 86 isolated-pentagon-rule (IPR) isomers of C92 has been described by the SAM1 quantum-chemical method, and their energetics checked by density functional theory at the B3LYP/6-31G* level. Although the lowest-energy cage is not identical in both approaches, it still exhibits D 2 symmetry in both cases. As energetics themselves cannot produce reliable relative stabilities at high temperatures, entropy terms are also computed and the relative-stability problem is treated entirely in terms of the Gibbs function. The lowest-energy structure is not the most populated isomer at higher temperatures – it is replaced by a D 3 structure. Further stability interchanges are possible at very high temperatures, when C 3 and C 1 structures are also important. Background  

The key enzymes of photosynthetic carbon assimilation in C4 plants have evolved independently several times from C3 isoforms that were present in the C3 ancestral species. The C4 isoform of phosphoenolpyruvate carboxylase (PEPC), the primary CO2-fixing enzyme of the C4 cycle, is specifically expressed at high levels in mesophyll cells of the leaves of C4 species. We are interested in understanding the molecular changes that are responsible for the evolution of this C4-characteristic PEPC expression pattern, and we are using the genus Flaveria (Asteraceae) as a model system. It is known that cis-regulatory sequences for mesophyll-specific expression of the ppcA1 gene of F. trinervia (C4) are located within a distal promoter region (DR).  相似文献

There are numerous studies describing how growth conditions influence the efficiency of C4 photosynthesis. However, it remains unclear how changes in the biochemical capacity versus leaf anatomy drives this acclimation. Therefore, the aim of this study was to determine how growth light and nitrogen availability influence leaf anatomy, biochemistry and the efficiency of the CO2 concentrating mechanism in Miscanthus × giganteus. There was an increase in the mesophyll cell wall surface area but not cell well thickness in the high-light (HL) compared to the low-light (LL) grown plants suggesting a higher mesophyll conductance in the HL plants, which also had greater photosynthetic capacity. Additionally, the HL plants had greater surface area and thickness of bundle-sheath cell walls compared to LL plants, suggesting limited differences in bundle-sheath CO2 conductance because the increased area was offset by thicker cell walls. The gas exchange estimates of phosphoenolpyruvate carboxylase (PEPc) activity were significantly less than the in vitro PEPc activity, suggesting limited substrate availability in the leaf due to low mesophyll CO2 conductance. Finally, leakiness was similar across all growth conditions and generally did not change under the different measurement light conditions. However, differences in the stable isotope composition of leaf material did not correlate with leakiness indicating that dry matter isotope measurements are not a good proxy for leakiness. Taken together, these data suggest that the CO2 concentrating mechanism in Miscanthus is robust under low-light and limited nitrogen growth conditions, and that the observed changes in leaf anatomy and biochemistry likely help to maintain this efficiency.  相似文献

The invasion of African grasses into Neotropical savannas has altered savanna composition, structure and function. The projected increase in atmospheric CO2 concentration has the potential to further alter the competitive relationship between native and invader grasses. The objective of this study was to quantify the responses of two populations of a widespread native C4 grass (Trachypogon plumosus) and two African C4 grass invaders (Hyparrhenia rufa and Melinis minutiflora) to high CO2 concentration interacting with two primary savanna stressors: drought and herbivory. Elevated CO2 increased the competitive potential of invader grasses in several ways. Germination and seedling size was promoted in introduced grasses. Under high CO2, the relative growth rate of young introduced grasses was twice that of native grass (0.58 g g−1 week−1 vs 0.25 g g−1 week−1). This initial growth advantage was maintained throughout the course of the study. Well-watered and unstressed African grasses also responded more to high CO2 than did the native grass (biomass increases of 21–47% compared with decreases of 13–51%). Observed higher water and nitrogen use efficiency of invader grasses may aid their establishment and competitive strength in unfertile sites, specially if the climate becomes drier. In addition, high CO2 promoted lower leaf N content more in the invader grasses. The more intensive land use, predicted to occur in this region, may interact with high CO2 to fincreasesavor the African grasses, as they generally recovered faster after simulated herbivory. The superiority of invader grasses under high CO2 suggests further in their competitive strength and a potential increased rate of displacement of the native savannas in the future by grasslands dominated by introduced African species.  相似文献

The amphibious leafless sedge Eleocharis retroflexa ssp. chaetaria expresses C4-like biochemical characteristics in both the terrestrial and submerged forms. Culms of the terrestrial form have Kranz anatomy, whereas those of the submerged form have Kranz-like anatomy combined with anatomical features of aquatic plant leaves. We examined the immunolocalization of C3 and C4 enzymes in culms of the two forms. In both forms, phosphoenolpyruvate carboxylase; pyruvate, Pi dikinase; and NAD-malic enzyme were compartmentalized between the mesophyll (M) and Kranz cells, but their levels were somewhat reduced in the submerged form. In the terrestrial form, ribulose-1,5-bisphosphate carboxylase/oxygenase (rubisco) occurred mainly in the Kranz cells, and weakly in the M chloroplasts. In the submerged form, the rubisco occurred at higher levels in the M cells than in the terrestrial form. In both forms, the C4 pattern of enzyme expression was clearer in the M cells adjacent to Kranz cells than in distant M cells. During the transition from terrestrial to submerged conditions, the enzyme expression pattern changed in submerged mature culms that had been formed in air before submergence, and matched that in culms newly developed underwater. It seems that effects of both environmental and developmental factors overlap in the C4 pattern expression in this plant.  相似文献

The effects of the diurnal variations in ambient temperature on some C3 and C4 enzymes in the Salsola dendroides and Suaeda altissima species of Chenopodiaceae family were studied during the intensive vegetation period. Activities of phosphoenolpyruvate carboxylase (PEPC) and cytosolic aspartate aminotransferase (AsAT) were shown to decrease in both species in the afternoon and evening. The activity of the mitochondrial AsAT decreased in S. altissima, remained relatively constant in S. dendroides during the day. The activity of alanine aminotransferase was high in the S. dendroides species in the morning and evening and decreased in the S. altissima species by the evening. Glucose-6-phosphate activated PEPC in both species throughout the day. The study of the redox status-regulated C3 enzymes showed temperature-related increases in NADP-glyceraldehyde 3-phosphate dehydrogenase activity in both plants, in fructose-2,6-bisphosphatase activity in the S. altissima species, and in NADP-MDH activity in the S. dendroides species in the afternoon.  相似文献

We describe the long-term effects of a CO2 exhalation, created more than 70 years ago, on a natural C4 dominated sub-tropical grassland in terms of ecosystem structure and functioning. We tested whether long-term CO2 enrichment changes the competitive balance between plants with C3 and C4 photosynthetic pathways and how CO2 enrichment has affected species composition, plant growth responses, leaf properties and soil nutrient, carbon and water dynamics. Long-term effects of elevated CO2 on plant community composition and system processes in this sub-tropical grassland indicate very subtle changes in ecosystem functioning and no changes in species composition and dominance which could be ascribed to elevated CO2 alone. Species compositional data and soil δ13C isotopic evidence suggest no detectable effect of CO2 enrichment on C3:C4 plant mixtures and individual species dominance. Contrary to many general predictions C3 grasses did not become more abundant and C3 shrubs and trees did not invade the site. No season length stimulation of plant growth was found even after 5 years of exposure to CO2 concentrations averaging 610 μmol mol−1. Leaf properties such as total N decreased in the C3 but not C4 grass under elevated CO2 while total non-structural carbohydrate accumulation was not affected. Elevated CO2 possibly lead to increased end-of-season soil water contents and this result agrees with earlier studies despite the topographic water gradient being a confounding problem at our research site. Long-term CO2 enrichment also had little effect on soil carbon storage with no detectable changes in soil organic matter found. There were indications that potential soil respiration and N mineralization rates could be higher in soils close to the CO2 source. The conservative response of this grassland suggests that many of the reported effects of elevated CO2 on similar ecosystems could be short duration experimental artefacts that disappear under long-term elevated CO2 conditions.  相似文献

The review is done to summarise the history of the discoveries of the many anatomical, agronomical, and physiological aspects of C4 photosynthesis (where the first chemical products of CO2 fixation in illuminated leaves are four-carbon dicarboxylic acids) and to document correctly the scientists at the University of Arizona and the University of California, Davis, who made these early discoveries. The findings were milestones in plant science that occurred shortly after the biochemical pathway of C3 photosynthesis in green algae (where the first chemical product is a three-carbon compound) was elucidated at the University of California, Berkeley, and earned a Nobel Prize in chemistry. These remarkable achievements were the result of ground-breaking pioneering research efforts carried out by many agronomists, plant physiologists and biochemists in several laboratories, particularly in the USA. Numerous reviews and books written in the past four decades on the history of C4 photosynthesis have focused on the biochemical aspects and give an unbalanced history of the multidisciplinary/multinstitutional nature of the achievements made by agronomists, who published much of their work in Crop Science. Most notable among the characteristics of the C4 species that differentiated them from the C3 ones are: (I) high optimum temperature and high irradiance saturation for maximum leaf photosynthetic rates; (II) apparent lack of CO2 release in a rapid stream of CO2-free air in illuminated leaves in varying temperatures and high irradiances; (III) a very low CO2 compensation point; (IV) lower mesophyll resistances to CO2 diffusion coupled with higher stomatal resistances, and, hence, higher instantaneous leaf water use efficiency; (V) the existence of the so-called “Kranz leaf anatomy” and the higher internal exposed mesophyll surface area per cell volume; and (VI) the ability to recycle respiratory CO2 by illuminated leaves.  相似文献

Recent research on the photosynthetic mechanisms of plant species in the Chenopodiaceae family revealed that three species, including Bienertia sinuspersici, can carry out C4 photosynthesis within individual photosynthetic cells, through the development of two cytoplasmic domains having dimorphic chloroplasts. These unusual single-cell C4 species grow in semi-arid saline conditions and have semi-terete succulent leaves. The effects of salinity on growth and photosynthesis of B. sinuspersici were studied. The results show that NaCl is not required for development of the single-cell C4 system. There is a large enhancement of growth in culture with 50–200 mM NaCl, while there is severe inhibition at 400 mM NaCl. With increasing salinity, the carbon isotope values (δ13C) of leaves increased from −17.3o/oo (C4-like) without NaCl to −14.6o/oo (C4) with 200 mM NaCl, possibly due to increased capture of CO2 from the C4 cycle by Rubisco and reduced leakiness. Compared to growth without NaCl, leaves of plants grown under saline conditions were much larger (~2 fold) and more succulent, and the leaf solute levels increased up to ~2000 mmol kg solvent−1. Photosynthesis on an incident leaf area basis (CO2 saturated rates, and carboxylation efficiency under limiting CO2) and stomatal conductance declined with increasing salinity. On a leaf area basis, there was some decline in Rubisco content with increasing salinity up to 200 mM NaCl, but there was a marked increase in the levels of pyruvate, Pi dikinase, and phosphoenolpyruvate carboxylase (possibly in response to sensitivity of these enzymes and C4 cycle function to increasing salinity). The decline in photosynthesis on a leaf area basis was compensated for on a per leaf basis, up to 200 mM NaCl, by the increase in leaf size. The influence of salinity on plant development and the C4 system in Bienertia is discussed.  相似文献

Sulfate assimilation and glutathione synthesis were traditionally believed to be differentially compartmentalised in C4 plants with the synthesis of cysteine and glutathione restricted to bundle sheath and mesophyll cells, respectively. Recent studies, however, showed that although ATP sulfurylase and adenosine 5′ phosphosulfate reductase, the key enzymes of sulfate assimilation, are localised exclusively in bundle sheath in maize and other C4 monocot species, this is not true for the dicot C4 species of Flaveria. On the other hand, enzymes of glutathione biosynthesis were demonstrated to be active in both types of maize cells. Therefore, in this review the recent findings on compartmentation of sulfate assimilation and glutathione metabolism in C4 plants will be summarised and the consequences for our understanding of sulfate metabolism and C4 photosynthesis will be discussed.  相似文献

The photosynthetic pathway composition (C3:C4 mixture) of an ecosystem is an important controller of carbon exchanges and surface energy flux partitioning, and therefore represents a fundamental ecophysiological distinction. To assess photosynthetic mixtures at a tallgrass prairie pasture in Oklahoma, we collected nighttime above-canopy air samples along concentration and isotopic gradients throughout the 1999 and 2000 growing seasons. We analyzed these samples for their CO2 concentration and carbon isotopic composition and calculated C3:C4 proportions with a two-source mixing model. In 1999, the C4 percentage increased from 38% in spring (late April) to 86% in early fall (mid-September). The C4 percentages inferred from ecosystem respiration measurements in 2000 indicate a smaller shift, from 67% in spring (early May) to 77% in mid-summer (late July). We also sampled daytime CO2 concentration and carbon isotope gradients above the canopy to determine ecosystem discrimination against 13CO2 during net uptake. These discrimination values were always lower than corresponding nighttime ecosystem respiration isotopic signatures would suggest. After accounting for the isotopic disequilibria between respiration and photosynthesis resulting from seasonal variations in the C3:C4 mixture, we estimated canopy photosynthetic discrimination. The C4 percentage calculated from this approach agrees with the percentage determined from nighttime respiration for sampling periods in both growing seasons. Isotopic imbalances between photosynthesis and respiration are likely to be common in mixed C3:C4 ecosystems and must be considered when using daytime isotopic measurements to constrain ecosystem physiology. Given the global extent of such ecosystems, isotopic imbalances likely contribute to global variations in the carbon isotopic composition of atmospheric CO2.  相似文献

Cellular anatomy and expression of glycine decarboxylase (GDC) protein were studied during leaf development of the C3-C4 intermediate species Moricandia arvensis. Leaf anatomy was initially C3-like and the number and profile area of mitochondria in the bundle-sheath cells were the same as those in adjacent mesophyll cells. Between a leaf length of 6 and 12 mm there was a bundle-sheath-specific, 4-fold increase in the number of mitochondrial profiles, followed by a doubling of their individual profile areas as the leaves expanded further. Subunits of GDC were present in whole-leaf extracts before the anatomical development of bundle-sheath cells. Whereas the GDC H-protein content of leaves increased steadily throughout development, the increase in GDC P-protein was synchronous with the development of mitochondria in the bundle sheath. The P-protein was confined to bundle-sheath mitochondria throughout leaf development, and its content in individual mitochondria increased before the anatomical development of the bundle sheath. Anatomical and biochemical attributes of the C3-C4 character were present in the cotyledons and sepals but not in other photosynthetic organs/tissues. In leaves and cotyledons that developed in the dark, the expression of the P-protein and the organellar development were reduced but the bundle-sheath cell specificity was retained.  相似文献

Utilization of storage starch in the cells of cotyledon mesophyll and root meristem in the course of alfalfa (Medicago sativa L.) seed germination on the solutions of NaCl, Na2SO4, and mannitol at different concentrations and identical osmotic pressure was investigated using the method of transmission electron microscopy. Ultrastructural analysis showed changes in the number of starch grains and deceleration of chloroplast development depending on the osmotic component of salt influence. At low concentrations corresponding to osmotic pressure of 202.6 kPa, Na2SO4 did not affect the formation of the photosynthetic machinery and utilization of starch inclusions; mannitol contributed to the preservation of considerable reserve of starch without disturbing the development of chloroplasts; NaCl did not inhibit the development of the photosynthetic machinery and induced an increase in the number of starch grains presumably at the expense of newly produced starch. When the concentration of the investigated substances increased up to the values corresponding to the osmotic pressure of 607.8 kPa, NaCl did not suppress transformation of amyloplasts into chloroplasts and utilization of starch; Na2SO4 inhibited the development of chloroplasts and starch utilization; mannitol decelerated transformation of amyloplasts and inhibited mobilization of starch grains. The obtained results make it possible to propose a method of preliminary estimation of tolerance of dicotyledons to abiotic stresses based on the cytological analysis of utilization of starch grains and formation of photosynthetic compartments of chloroplasts in the mesophyll of cotyledons.  相似文献
