实验研究壳聚糖季铵盐对凡纳滨对虾生长性能及非特异性免疫能力的影响。在饲料中分别添加0、0.05%、0.10%、0.15%和0.20%的壳聚糖季铵盐, 制成5组等氮等能饲料。将900尾[初体质量(3.820.34) g]健康的凡纳滨对虾随机分成5组(45尾4平行), 养殖时间56d。结果表明:饲料中添加壳聚糖季铵盐显著影响凡纳滨对虾的生长, 0.15%实验组凡纳滨对虾的增重率和特定生长率最佳(P0.05)。饲料中添加壳聚糖季铵盐0.1%、0.15%和0.20%能显著提高凡纳滨对虾血清溶菌酶和碱性磷酸酶及酚氧化酶的活性(P0.05)。饲料中添加壳聚糖季铵盐可显著提高凡纳滨对虾抗副溶血弧菌感染的能力(P0.05), 0.15%组的保护效果最好, 其相对免疫保护率为33.24%。壳聚糖季铵盐能显著提高凡纳滨对虾的生长性能和抗病能力, 本实验条件下适宜的添加量为0.15%。  相似文献

This paper describes the purification of Penicillin G Amidase (EC 3.5.1.11) using quaternary ammonium salts with the aim of increasing the activity of immobilised enzymes prepared from the purified solutions. Two different quaternary ammonium salts were tested with different solutions of the enzyme. It was concluded that the quaternary ammonium salts used selectively precipitated the non-enzymatic protein leaving in solution practically all the enzyme resulting in a high yield of purification. Optimal conditions for purification using the two types of quaternary ammonium salts were determined. Immobilisation studies were performed from various purified enzyme solutions, using different amounts of a quaternary ammonium salt. The immobilised enzymes so obtained showed a much higher activity than the immobilised enzyme obtained from non-purified enzyme solutions.  相似文献

The biological activity of lysosomotropic n-alkyl N,N-dimethylglycinates (DMG-n) was compared with that of a quaternary ammonium salt IM (methochloride of DMG-12). The activity of the glycinates appeared to be carbon chain length dependent and was similar at pH 6 and pH 8. Nutritional auxotrophy and respiratory deficiencies have no influence on DMG-n sensitivity. Both IM and DMG-n inhibit plasma membrane H+-ATPase activity while mitochondrial ATPase is relatively non-sensitive to glycinates. No cross-resistance to IM and DMG-n was observed.  相似文献

A comparison of the use of the quaternary ammonium salts, cetyltrimethylammonium bromide (CTAB) and the commercial mixture Cetavlon, for the isolation of xanthan gum from fermentations of Xanthomonas campestris indicated that the former was the more efficient complexating agent. Although in both cases more than the stoichiometric requirement was necessary to achieve quantitative recovery of the polysaccharide, CTAB left only 1·7% material in the supernatant from the precipitation of xanthan gum compared to 15% left by Cetavlon. This is congruent with the view that the efficiency of quaternary ammonium salts increases with increased paraffin chain length.An assessment of the use of Cetavlon for the isolation of xanthan gum in a recycle procedure showed that an 11·5% loss of precipitant per cycle occurred. In the procedure, the xanthan gum was precipitated as the purified K+ salt from a dispersion of its quaternary ammonium complex in 2-propanol. Concentration of the 2-propanol wash permitted recovery of the quaternary ammonium salt.  相似文献

The complexes formed between quaternary ammonium cations and polyanionic glycosaminoglycans can be partitioned between partially miscible aqueous inorganic salt and alcohol phases. Small changes in salt concentration can completely shift the complex from one phase to the other. The effect of the phase composition variables: the type of inorganic salt, the type of quaternary ammonium salt, and the alcohol used, were systematically investigated. The sharp transition from solubility in the upper non-aqueous phase to solubility in the lower, aqueous phase was found to be strongly affected by the type of inorganic salt. This transition occurred at higher salt concentrations when NaCl, KCl, or LiCl were used than when CaCl2 or MgCl2 were used. Differences in behavior among glycosaminoglycans were larger for NaCl than for CaCl2. The complex is stabilized to dissociation by salt by increasing hydrophobicity of the non-aqueous phase. However, aggregation of the complex into an insoluble form is also favored by an increasingly hydrophobic environment. The most consistent partition was observed with 1- and 2-butanol. The partition isotherm of chondroitin 4-sulfate was investigated at constant salt concentration. It was found that the partition coefficient varies with the concentration of chondroitin 4-sulfate, although the magnitude of this effect could be diminished by increasing the quaternary ammonium salt concentration.  相似文献

A set of n-alkyl esters of N,N-dimethylglycine (DMG-n) and their methobromides (DMGM-n) was synthesized, and their activities on yeast Saccharomyces cerevisiae were compared. The compounds differ in the number of carbon atoms in the aliphatic chain. Aminoesters with 12 carbon atoms appeared to be most active. Unlike quaternary ammonium salts previously tested, the activities of the compounds were not pH-dependent; the minimal inhibitory concentrations (MIC) were identical at pH 8 and at pH 6. In contrast to quaternary ammonium salts, aminoesters showed similar effects on respiratory sufficient (rho+) and respiratory deficient (rho0) mutants. When tested on glucose stimulated proton extrusion, aminoesters applied at MIC increased external pH. Aminoesters inhibited the plasma membrane H+-ATPase, whereas they were less inhibitory on the mitochondrial ATPase. In order to further compare the aminoesters and their corresponding quaternary ammonium salts, derivatives of N,N-dimethylalanine (DMAL-n and DMALM-n, respectively) were synthesized. The quaternary ammonium salts appeared to have a higher inhibitory potency than aminoesters, especially at pH 8, and alanine derivatives inhibited growth at a lower concentration than glycine derivatives. Both alanine derivatives of the aminoester and the quaternary ammonium salt inhibited the plasma membrane H+- ATPase at lower concentrations than glycine derivatives, but the alanine aminoester was without a detectable effect on the mitochondrial ATPase.  相似文献

In an effort to improve the water solubility of camptothecin, 16 water soluble 10-substituted quaternary ammonium salt derivatives of camptothecin were prepared. Their antitumor activity was evaluated on cancer cells in vitro. All of these salts possess lower cytotoxicities than CPT in comparison. The camptothecin salts 16, 20 showed similar cytotoxic activity to topotecan. Especially the salts 21 showed similar cytotoxic activity to CPT in vitro.  相似文献

An epitope was found on the alpha2-nicotinic isoform of the neuronal nicotinic acetylcholine receptor that would likely form salt bridges with quaternary ammonium compounds and a cation-pi interaction with the pi-cloud of an aromatic ring. Chlorisondamine, a nicotinic antagonist, exerts a long-lasting, if not permanent, blockade of the ion channel gated by acetylcholine. Blocking of the ion channel prevents nicotine from exerting its rewarding effect on the CNS. Chlorisondamine contains two quaternary ammonium groups and a tetrachloroisoindoline ring. We propose that chlorisondamine interacts with an epitope on the alpha2 isoform of the rat neuronal nicotinic receptor (residues 388-402, GEREETEEEEEEEDE), where one or both of the quaternary ammonium groups of chlorisondamine form a salt bridge with dither a glutamic acid side chain or a phosphate group, whereas the tetrachlorobenzene portion of the tetrachloroisoindoline ring interacts with the guanidinium group of arginine in a cation-pi association: In this work, a new way of probing the interaction of a receptor epitope (alpha2) with organic molecules (chlorisondamine and hexachlorobenzene) was undertaken using matrix assisted laser desorption/ionization mass spectrometry.  相似文献

The water-soluble quaternary ammonium salt of chitosan was synthesized by reaction of chitosan with glycidyl trimethylammonium chloride. It was characterized by Fourier transform infrared and carbon-13 magnetic resonance spectra. Its microbiostatic effects against Staphylococcus aureus, Escherichia coli and Candid albicans were explored by microcalorimetry. The antimicrobial activity of the quaternized chitosan was stronger in alkaline condition than that in weak acidic condition.  相似文献

A derivative of chitooligosaccharide (COS) with quaternary ammonium functionality was synthesized and characterized by means of FT-IR and NMR spectroscopy. Its amtimicrobial activity was evaluated against Streptococcus mutans, which is a principal etiological agent of dental caries in humans. Introduction of quaternary ammonium group to COS has been easily accomplished by coupling of glycidyl trimethylammonium chloride (GTMAC) to COS in aqueous solution without an additional catalyst. The degree of substitution (%), as determined by (1)H NMR, of GTMAC to the COS increased up to 116% at 70 degrees C for 24h. The resulting COS-GTMAC exhibited the growth inhibition of above 80% against S. mutans after 5h, whereas the COS showed the growth inhibition of about 10%. It was found that antimicrobial activity of the COS could be considerably enhanced by the introduction of quaternary ammonium functionality.  相似文献

A series of 4-oxo-4H-pyrido[1,2-a]pyrimidine derivatives, substituted at the 2-position with piperidines bearing quaternary ammonium salt side chains, were synthesized and evaluated for their ability to potentiate the activity of the fluoroquinolone levofloxacin (LVFX) and the beta-lactam aztreonam (AZT) in Pseudomonas aeruginosa. Attachment of the charged entity using an N-ethylcarbamoyloxy linker led to the discovery of the highly soluble compound 22 (D13-9001), which maintained good potency in vitro and displayed excellent activity in vivo in a rat pneumonia model of P. aeruginosa.  相似文献

Quaternary ammonium chitosan sulfates with diverse degrees of substitution (DS) ascribed to sulfate groups between 0.52 and 1.55 were synthesized by reacting quaternary ammonium chitosan with an uncommon sulfating agent (N(SO3Na)3) that was prepared from sodium bisulfite (NaHSO3) through reaction with sodium nitrite (NaNO2) in the aqueous system homogeneous. The structures of the derivatives were characterized by FTIR, 1H NMR and 13C NMR. The factors affecting DS of quaternary ammonium chitosan sulfates which included the molar ratio of NaNO2 to quaternary ammonium chitosan, sulfated temperature, sulfated time and pH of sulfated reaction solution were investigated in detail. Its anticoagulation activity in vitro was determined by an activated partial thromboplastin time (APTT) assay, a thrombin time (TT) assay and a prothrombin time (PT) assay. Results of anticoagulation assays showed quaternary ammonium chitosan sulfates significantly prolonged APTT and TT, but not PT, and demonstrated that the introduction of sulfate groups into the quaternary ammonium chitosan structure improved its anticoagulant activity obviously. The study showed its anticoagulant properties strongly depended on its DS, concentration and molecular weight.  相似文献

Biodegradation of two amphiphilic “soft” antimicrobially active derivatives of lauric (dodecanoic) acid, a quaternary ammonium salt and an amine oxide bearing an amide or ester group, was followed using microorganisms from activated sludge. Primary biodegradation was determined by ion-selective electrodes, total biodegradation as the chemical oxygen demand. Though organic ammonium salts quickly undergo primary biodegradation, the rest of the molecule is difficult to destroy. In contrast, amine oxides are easily biodegradable.  相似文献

In the present study, a fine ultrastructural localization of nicotinic acetylcholine receptor (nAChR) was attempted, using d-tubocurarine (d-TC), a quaternary ammonium compound binding to nAChR. The localization was based on the binding avidity of immunoglobulin G (IgG) for acetylcholine (ACh) and other quaternary ammonium compounds, such as d-TC. d-TC was applied to the frog neuromuscular preparation and caused a blockade of neuromuscular transmission. Then, d-TC was rendered insoluble in situ by silicotungstic acid (STA), a precipitating agent of soluble proteins and quaternary ammonium compounds. After tissue fixation, a normal rabbit serum was applied to the fine precipitate of the insoluble salt of d-TC silicotungstate (quaternary ammonium radical of d-TC) to form the immunochemical complex d-TC- rabbit IgG at ACh binding sites. The IgG of the complex was revealed by means of the conventional immunoperoxidase procedure used for ultrastructural localization. Under the electron microscope, fine diaminobenzidine (DAB) precipitates appeared as regular rod-like structures oriented to cytoplasmic side of the horizontal part (crest) of the postsynaptic membrane (between the junctional folds) which is known to be endowed with nAChR. The rod-like precipitates were not observed in the postsynaptic junctional folds which are devoid of nAChR. The distance separating the rods each other was rather constant (12 - 15 nm), while the length of the rods was variable and exceeded the usual length of nAChR. The present work indicates that the rod-like structures, already observed in association with sarcoplasmic side of the postsynaptic membrane, did correspond to the intramembranous and intracytoplasmic part of nAChR and related proteins. These cytochemical results confirm that d-TC binds to ACh binding sites in the pore of nAChR, and raise the question of DAB staining of cytoskeletal proteins related to the nAChR complex.  相似文献

The Swimming Pool Water Disinfectant Test Method of the Association of Official Analytical Chemists was used to determine the effect of the accepted level of 2 ppm of some commercial quaternary ammonium algicides on the germicidal activity of chlorine. Accurate determinations on the amounts of residual available chlorine in chlorine-quaternary mixtures could not be made by the usual chemical methods. This made it necessary to base all comparisons on the starting concentrations of available chlorine rather than the final concentration as specified in the method employed. No evidence was obtained to support the use of lower concentrations of residual available chlorine for disinfection in the presence of algicidal quaternaries than those commonly recognized as effective by the American Public Health Association. The rate of kill against the gram-positive test organism Streptococcus faecalis was faster in quaternary-chlorine mixtures than in the sodium hypochlorite control solutions. The practical significance of this result in the bench method identified cannot be ascertained in the absence of more sensitive and precise chemical procedures for determining concentrations of residual available chlorine in the presence of quaternaries or in actual swimming pool tests.  相似文献

AIMS: Enhancement of algicidal activity by immobilization of algicidal bacteria antagonistic to Stephanodiscus hantzschii. METHODS AND RESULTS: In laboratory studies, A diatom-lysing bacterium, Pseudomonas fluorescens HYK0210-SK09 showed strong algicidal activity against S. hantzschii, but a natural mesocosm study revealed that this bacterium failed to fully control natural blooms of Stephanodiscus at the low water temperatures that favour these blooms. Here, we sought to develop an effective immobilization strategy for enhancing the algicidal activity of HYK0210-SK09 in the natural setting. Bacterium HYK0210-SK09 was immobilized with various carriers including agar, alginate, polyurethane and cellulose sponge. The bacterial cells immobilized with cellulose sponge (CIS) induced more rapid and complete lysis of S. hantzschii than other carriers, and had a higher packing ability than polyurethane. Furthermore, CIS-immobilized cells showed higher lysis of S. hantzschii at the same concentrations as that of free cells (< or =1 x 10(7) cells ml(-1)), and had especially strong algicidal activity at the low temperatures (<10 degrees C). Based on these laboratory studies, we assessed the possible application of HYK0210-SK09 cells in the field by performing a mesocosm study during the winter season. The CIS-immobilized cells with species-specific activity towards the genera Stephanodiscus showed extremely high algicidal activity (up to 95%) against a bloom of Stephanodiscus hantzschii even at low water temperatures, because of high cell packing and subsequent cell protection against low temperatures and predators, whereas free cells showed negligible algicidal activities under these conditions. CONCLUSION: Immobilizing cells of HYK0210-SK09 in CIS foam, rather than in the other matrices tested, could achieve more efficient control of Stephanodiscus blooms and showed a significant algicidal activity on in vitro and in vivo blooms, even at low water temperature. SIGNIFICANCE AND IMPACT OF THE STUDY: Collectively, these results indicate that CIS of algicidal bacteria may form an important strategy for effective management of Stephanodiscus blooms at low water temperatures.  相似文献

In our studies of the health effects of internalized depleted uranium, we developed a simple and rapid light microscopic method to stain specifically intracellular uranium deposits. Using J774 cells, a mouse macrophage line, treated with uranyl nitrate and the pyridylazo dye 2-(5-bromo-2- pyridylazo)-5-diethylaminophenol, uranium uptake by the cells was followed. Specificity of the stain for uranium was accomplished by using masking agents to prevent the interaction of the stain with other metals. Prestaining wash consisting of a mixture of sodium citrate and ethylenediaminetetraacetic acid eliminated staining of metals other than uranium. The staining solution consisted of the pyridylazo dye in borate buffer along with a quaternary ammonium salt, ethylhexadecyldimethylammonium bromide, and the aforementioned sodium citrate/ethylene-diaminetetraacetic acid mixture. The buffer was essential for maintaining the pH within the optimum range of 8 to 12, and the quaternary ammonium salt prevented precipitation of the dye. Staining was conducted at room temperature and was complete in 30 min. Staining intensity correlated with both uranyl nitrate concentration and incubation time. Our method provides a simple procedure for detecting intracellular uranium deposits in macrophages.  相似文献
