The aqueous fraction of Fushimi sweet pepper increased the repair effect of the solvent control against UV-induced cyclobutane pyrimidine dimers in human fibroblast to 150%, but ordinary sweet pepper did not have a statistically significant effect. When Fushimi sweet pepper was boiled, the activity of the aqueous fraction was elevated to 209% of the control (p<0.05), while that of the grilled state was decreased to 125% of the control. The repair activity of a dialyzate (MW<12,000) of the aqueous fraction from Fushimi sweet pepper showed 191% of the control (p<0.05). The dialyzate was contained 1.9% in the weight of the fresh fruit body of Fushimi sweet pepper, and the activity can be stable in its boiling state, and it might be therefore considered to be the worthy source for expecting the DNA repair activity in human diet.  相似文献

Abstract

Field experiments were carried out at the Teaching and Research Farm of the Abubakar Tafawa Balewa University, Bauchi during the rainy season of 2005. The study was carried out with sole objective of evaluating the efficacy of six selected plant materials (sweetsop, red pepper, garlic, neem, mahogany and gmalina) against the major insect pests of cowpea variety, Dan Sokoto. The experiment was laid down in randomised complete block design with seven treatments. Each treatment was replicated three times. The results of the study showed that all the plant materials used were significantly (p < 0.05) better than control where no plant material was used in controlling the population of B. tabaci, E. dolichi, M. sjostedti and C. tomentosicollis at 1, 2 and 3 days after application of the treatments. Similarly the effects of these plant materials on the number of seeds/pod showed a significant (p < 0.05) difference between plots treated with plant materials and control. On the grain yield of the crop, all the plant materials showed significant effect except mahogany and the control which were statistically similar. Furthermore, the order of level of control indicated that sweet sop has (70.7%), garlic (69.3%), neem (61.0%), red pepper (54.0%), ash (30.9%), and mahogany (3.5%). The result of the present finding therefore recommends the use of sweet sop, garlic and neem as they were found to be the most promising in the control of major cowpea insect pests.  相似文献

Lack of reliable assays for DNA repair has largely prevented measurements of DNA repair from being included in human biomonitoring studies. Using newly developed modifications of the comet assay we tested whether a fruit‐ and antioxidant‐rich plant‐based intervention could affect base excision repair (BER) and nucleotide excision repair (NER) in a group of 102 male volunteers. BER and NER repair capacities were measured in lymphocytes before and after a dietary intervention lasting 8 weeks. The study had one control group, one group consuming three kiwifruits per day and one group consuming a variety of antioxidant‐rich fruits and plant products in addition to their normal diet. DNA strand breaks were reduced following consumption of both kiwifruits (13%, p = 0.05) and antioxidant‐rich plant products (20%, p = 0.02). Increased BER (55%, p = 0.01) and reduced NER (?39%, p < 0.01) were observed in the group consuming a wide variety of plant products. Reduced NER was also observed in the kiwifruit group (?38%, p = 0.05), but BER was not affected in this group. Here we have demonstrated that DNA repair is affected by diet and that modified versions of the comet assay can be used to assess activity of different DNA repair pathways in human biomonitoring studies. In previous studies, the relaxant effect of Tymus vulgaris has been demonstrated on guinea pig tracheal chains. Therefore, in the present study, the relaxant effects of n-hexane, dichloromethane, methanol and aqueous fractions of Tymus vulgaris on tracheal chains of guinea pigs were examined. The relaxant effects of four cumulative concentrations of each fraction (0.4, 0.8, 1.2 and 1.6 g%) in comparison to saline as negative control and four cumulative concentrations of theophylline (0.2, 0.4, 0.6 and 0.8 mM) were examined for their relaxant effects on precontracted tracheal chains of guinea pig by 60 mM KCl (group 1) and 10 ìì methacholine (group 2, n = 7 for each group). In group 1, all concentrations of the n-hexane fraction and theophylline and three last concentrations (0.8, 1.2 and 1.6 g%) of dichloromethane and two higher concentrations (1.2 and 1.6 g%) of methanol fractions showed significant relaxant effects compared to that of saline (p<0.05 to p<0.001). In group 2, all concentrations of theophylline, n-hexane and dichloromethane fractions and three concentrations (0.8, 1.2 and 1.6 g%) of methanol and two higher concentrations (1.2 and 1.6 g%) of aqueous fractions showed significant relaxant effects compared to that of saline (p<0.05 to p<0.001). In addition, with group 1, the relaxant effect of all concentrations of all fractions except the n-hexane fraction, were significantly less than those of theophylline (p<0.05 to p<0.001). The n-hexane fraction showed higher relaxant effect than theophylline. The relaxant effect of all concentrations of the n-hexane fraction and the three last concentrations (0.8, 1.2 and 1.6 g%) of dichloromethane and aqueous fractions were significantly greater in group 2 than in group 1 (p<0.05 to p<0.001). There were significant positive correlations between the relaxant effects and concentrations for theophylline and all fractions (except aqueous fraction in group 1) in both groups, but a negative correlation for the aqueous fraction in group 1 (p<0.05 to p<0.001). These results showed a potent relaxant effect for n-hexane and weaker relaxant effect for other fractions from Tymus vulgaris on tracheal chains of guinea pigs.  相似文献

We succeeded in purifying the fraction containing the major glycolipids in monogalactosyl diacylglycerol, digalactosyl diacylglycerol and sulfoquinovosyl diacylglycerol (SQDG) from dried vegetables. This glycolipids fraction was an inhibitor of DNA polymerase alpha (pol alpha) in vitro and also the proliferation of human cancer cells. In this study, eight common vegetables were investigated in terms of the glycolipids fraction, the amounts of major glycolipids, mammalian DNA polymerase inhibitory activity and antiproliferative activity toward human cancer cells. Green tea possessed the largest amount of glycolipids overall. Spinach contained the largest amount of SQDG, followed by parsley, green onion, chive, sweet pepper, green tea, carrot and garlic. Spinach had the strongest inhibitory effect on pol alpha activity and human cancer cell proliferation. A significant correlation was found between SQDG content and inhibition of DNA polymerase. Therefore, the inhibition of pol alpha activity by SQDG may lead to cell growth suppression. Of the six subspecies of spinach (Spinacia oleracea) tested, "Anna" had the largest amount of SQDG, strongest inhibitory activity toward DNA polymerase and greatest effect on human cancer cell proliferation. Based on these results, the glycolipids fraction from spinach is potentially a source of food material for a novel anticancer activity.  相似文献

Treating the leaves of sweet pepper (Capsicum annuum L. var. grossum) with an aqueous solution of cellulase resulted in a four-fold increase in the salicylic acid level compared to a control plant. The level of endogenous azelaic acid was also elevated by the cellulase treatment. Azelaic acid has recently been reported to act as a mobile "priming" agent to arm plants against pathogenic attack. Our results are consistent with this and that the cellulase treatment enhanced the ability of sweet pepper to withstand viral attack.  相似文献

Algal phycocyanins promote growth of human cells in culture   总被引:4,自引:0,他引:4
Summary The growth-promoting substances in a non-dialyzable extract of Synechococcus elongatus var. on RPMI 8226 cells (a human myeloma cell line) were separated by gel filtration and ion exchange chromatography. By gel filtration with Sepharose 4B, the dialyzate was separated into two fractions. One fraction was green-colored (P-1) and the other was blue-colored (P-2). The P-2 fraction had a higher growth-promoting activity than P-1. By ion exchange chromatography, the P-2 fraction was separated into two blue-colored fractions of phycocyanin and allophycocyanin. Both biliproteins promoted the growth of RPMI 8226 cells; however, allophycocyanin was more active than phycocyanin. Editor's statement This reporrt that phycobiliprotein from algae is capable of stimulating animal cell growth is unique, and raises the possiblity that related compounds such as biliverdin might also be similarly active. Some puzzling aspects, such as the lack of increase in specific activity during purification, as well as the possibility that the activity might be due to a contaminant, remain to be resolved.  相似文献

Biological control of different species of pest with various species of generalist predators can potentially disrupt the control of pests through predator-predator interactions. We evaluate the impact of three species of generalist predatory mites on the biological control of green peach aphids, Myzus persicae (Sulzer) with the aphidophagous gall midge Aphidoletes aphidimyza (Rondani). The predatory mites tested were Neoseiulus cucumeris (Oudemans), Iphiseius degenerans (Berlese) and Amblyseius swirskii Athias–Henriot, which are all commonly used for pest control in greenhouse sweet pepper. All three species of predatory mites were found to feed on eggs of A. aphidimyza, even in the presence of abundant sweet pepper pollen, an alternative food source for the predatory mites. In a greenhouse experiment on sweet pepper, all three predators significantly reduced population densities of A. aphidimyza, but aphid densities only increased significantly in the presence of A. swirskii when compared to the treatment with A. aphidimyza only. This stronger effect of A. swirskii can be explained by the higher population densities that this predator reached on sweet pepper plants compared to the other two predator species. An additional experiment showed that female predatory midges do not avoid oviposition sites with the predator A. swirskii. On the contrary, they even deposited more eggs on plants with predatory mites than on plants without. Hence, this study shows that disruption of aphid control by predatory mites is a realistic scenario in sweet pepper, and needs to be considered when optimizing biological control strategies.  相似文献

The combined release of species of generalist predators can enhance multiple pest control when the predators feed on different prey, but, in theory, predators may be excluded through predation on each other. This study evaluated the co-occurrence of the generalist predators Macrolophus pygmaeus Rambur and Orius laevigatus (Fieber) and their control of two pests in a sweet pepper crop. Both predators consume pollen and nectar in sweet pepper flowers, prey on thrips and aphids, and O. laevigatus is an intraguild predator of M. pygmaeus. Observations in a commercial sweet pepper crop in a greenhouse with low densities of pests showed that the two predator species coexisted for 8 months. Moreover, their distributions in flowers suggested that they were neither attracted to each other, nor avoided or excluded each other. A greenhouse experiment showed that the predators together clearly controlled thrips and aphids better than each of them separately. Thrips control was significantly better in the presence of O. laevigatus and aphid control was significantly better in the presence of M. pygmaeus. Hence, combined inoculative releases of M. pygmaeus and O. laevigatus seem to be a good solution for controlling both thrips and aphids in greenhouse-grown sweet pepper. The predators are able to persist in one crop for a sufficiently long period and they complement each other in the control of both pests. This study also provides further evidence that intraguild predation does not necessarily have negative effects on biological control.  相似文献

The predatory mite Iphiseius degenerans (Berlese) is commercially available as a biological control agent of thrips and spider mites in greenhouse crops. Developmental duration and immature survival of I. degeneransreared on nine types of food (almond pollen, apple pollen, castor bean pollen, plum pollen, sweet pepper pollen, Tetranychus urticaeKoch, Frankliniella occidentalis(Pergande), Ephestia kuehniella Zeller eggs and Artemia franciscana Kellogg cysts) and on three substrates (Multicel, sweet pepper leaf, and bean leaf) were determined in the laboratory. All experiments were carried out at 25 °C. On Multicel, mean developmental times on pollen ranged from 6.0 to 7.1 days, with the lowest value recorded on almond pollen and the highest on sweet pepper pollen. When reared on castor bean pollen significantly longer developmental times were obtained on a sweet pepper leaf compared to a bean leaf or Multicel. Developmental duration when offered T. urticaeon Multicel ranged between 6.1 and 6.9 days, on a bean leaf development was completed in 8.0 days. The longest developmental times on Multicel were recorded on Ephestia eggs (7.0 days) and on decapsulated Artemia cysts (7.5 days). No development beyond the protonymphal stage occurred in the absence of food or when encapsulated Artemia cysts or thrips larvae were offered on Multicel. On a sweet pepper leaf and a bean leaf, respectively 6.7 and 10.0% of the eggs reached adulthood when thrips larvae were provided as food; developmental times recorded here, were 9.0 and 8.3 days. Overall, immature mortality occurred mainly in the protonymphal stage and ranged from 0.0 to 36.7%. In conclusion, I. degenerans is able to feed on a variety of natural and unnatural foods, but thrips larvae and sweet pepper pollen are unfavourable food for immature development. This could compromise the establishment of this biological control agent when used against thrips in sweet pepper crops.  相似文献

Eretmocerus mundus is native to the Mediterranean region where it is often observed to enter greenhouses to parasitize B. tabaci on fruiting vegetables and other host crops. Fecundity on tomato and pepper was evaluated by placing newly emerged pairs (n = 15) of E. mundus on leaf discs infested with second instar B. tabaci, the preferred stage, maintained at 25 °C and changed daily until death of the female. All whitefly nymphs were observed for host feeding and inverted to count parasitoid eggs. Adult longevity was estimated at 7.3±0.8 d on tomato and 10.1±1.0 d on sweet pepper. Fecundity (number of hosts parasitized) was estimated 147.8±12.6 per female on tomato and 171.1±21.5 on pepper. Incidence of host feeding (number of hosts killed) was significantly greater on sweet pepper than on tomato, 15.6±1.9 vs. 10.4±1.3 nymphs per female, respectively. No significant differences were detected in the duration of life stages between sweet pepper and tomato. Preimaginal survivorship in clip cages was estimated at 69.5±11.9% on tomato and 76.6±10.5% on sweet pepper, with no statistical differences. Net reproductive rate (R o) was estimated at 63.8±8.2 and 51.0±4.4 on tomato and sweet pepper respectively. Generation time (T) was significantly greater on sweet pepper (19.3±0.5) than on tomato (17.9±0.4), but the estimate of intrinsic rate of increase (r m) was not statistically different at 0.216±0.005 and 0.219±0.004 respectively. These values are well above those reported for B. tabaci on any crop, indicating the potential of E. mundus to control this pest on solanaceous crops in the greenhouse.  相似文献

The effect of various diets on nymphal development and survival of two predaceous anthocorid bugs,Orius albidipennis (Reuter) andO. laevigatus (Fieber) was investigated in the laboratory. Five different diets were compared: eggs ofEphestia kuehniella Zeller; eggs ofE. kuehniella plus mixed flower pollen; only mixed flower pollen; pollen from sweet pepper (Capsicum annuum L. cv. Mazurka) flowers; and sweet pepper plants. A high percentage of predators successfully completed nymphal development on those diets containing lepidopterous eggs. When fed on sweet pepper pollen, the survival percentages were 65% forO. laevigatus and 38% forO. albidipennis. No nymphs of either species completed the nymphal stage on mixed flower pollen or on sweet pepper plants. Development was significantly faster on diets containing eggs ofE. kuehniella. Results are discussed in relation to the capability of the bugs to survive periods of prey scarcity and to the optimization of release strategies for these predators in the greenhouse.  相似文献

The development of gas-filled root porosity in response to temporary low oxygen supply was tested for a range of edible and ornamental crops: rice, maize, wheat, sugar beet, tomato, cucumber, sweet pepper, carnation, gerbera and rose. In a first experiment, the roots of tomato, maize and gerbera had a higher gas-filled root porosity, Ep (% v/v), when grown permanently in a non-aerated instead of aerated solution. The Ep of roots increased during two weeks when half the root system of a young plant was transferred to a non-aerated solution; in older plants this response was not seen. Carnation had a negligible gas-filled porosity in all treatments. In a second experiment, a comparison was made between high (20 kPa) and low (about 2 kPa) O2 partial pressure in a recirculating nutrient solution. Half of the root system was transferred to low O2 at various growth stages. In most species older plants did not increase Ep on exposure to low O2. For tomato, sweet pepper and rose, Ep was normally in the range 3–8% (v/v). Young plants of cucumber, wheat and sugar beet also had an Ep in that range, but in older plants values ranged from 1 to 3%. Transverse root sections examined by light microscopy showed, on average, 60% more intercellular spaces in the root cortex than the measurements of gas-filled porosity, probably because some gaps and spaces in the cortex were not gas-filled. This effect was most pronounced in tomato. A negative pressure in the cortex may be needed for gaps to be gas-filled. An exodermis may increase the effectiveness of gas spaces in the cortex by closing the gas channels and, by offering some resistance to water uptake, allowing a negative pressure head in the cortex which keeps gaps gas-filled. A redox dye method was developed to study the length of root which is effectively supplied with oxygen, as a function of Ep. Results indicated that for every percent Ep the root can remain aerated over at least 1 cm in a non-aerated medium under the conditions of the test.  相似文献

In vitro anthelmintic activity of crude extracts of the ripe fruits of Hedera helix was investigated on eggs and adult nematode parasites Haemonchus contortus. Aqueous extract of H. helix was also evaluated for in vivo anthelmintic activity at dose of 1.13 and 2.25 g/kg in sheep artificially infected with H. contortus. ED(50) for egg hatch inhibition was 0.12 and 0.17 mg/ml for aqueous and hydro-alcoholic extracts, respectively. There was no statistically significant difference in the activity of the two extract types (p>0.05). Hydro-alcoholic extract showed better in vitro activity against adult parasites compared to the aqueous extract. Significant faecal egg count reduction (FECR) was detected in groups treated with both doses of H. helix (p<0.05) on day 2 post-treatment. On day 7 post-treatment significant reduction was detected only for higher dose of H. helix (p<0.05) while on day 14 post-treatment there was no significant FECR in both groups treated with H. helix. The percentage of larvae recovered from culturing faeces obtained from groups of sheep treated with lower and higher doses of H. helix was 47.52% and 36.07%, respectively, which was significantly lower than (p<0.05) that recovered from the control group (60%). Significant (p<0.05), dose dependent total worm count reduction (WCR) was observed for groups of sheep treated with H. helix. Increasing the dose of H. helix improved the efficacy against the male than the female parasites. Treatment with both doses of H. helix helped the animals maintain their packed cell volume (PCV) unlike the untreated control group. The overall findings of the current study indicated that H. helix has a potential anthelmintic benefit and further in vitro and in vivo evaluation of the different parts and fractions is needed to make use of this plant for therapeutic purposes.  相似文献

A major constraint facing sweet pepper production is infestation by nematodes leading to reduced yields. Field studies were conducted during the 2012 cropping season at the Experimental Farms of the University for Development Studies, Nyankpala, Northern region, Ghana, to determine efficacy of various levels of moringa leaf powder for the control of root-knot nematodes in sweet pepper (Capsicum annuum L.) in the savanna ecology of Ghana. Treatments consisted of three levels of moringa leaf powder (40, 60 and 80?g/L) per plot and 0?g/L (control). The experiment was laid out in a randomised complete block design with each treatment replicated four times. The infestations of root-knot nematodes were significantly lower in the moringa leaf powder-treated plots than the control. Although significant differences were not observed in all the parameters evaluated among the moringa leaf powder treatments, sweet pepper plants treated with 80?g/L of moringa leaf powder per plot recorded the highest mean value of plant height, number of leaves, number of fruits per plant, fruit weight per plant total yield per plot and the thickest plant girth. Similarly, the sweet pepper plants treated with 80?g/L of moringa leaf powder had the lowest infection index (root gall) and nematode population. Application of moringa leaf powder at 40, 60 and 80?g/L increased sweet pepper yield and decreased nematode population confirming their potential in management of root-knot nematodes.  相似文献

The aim of this work was to investigate whether parthenocarpicfruit growth could avoid flushing, i.e. an irregular yield pattern,in sweet pepper. Plants were grown in a greenhouse compartmentfrom April until August. Half of the plants were grown withouta fruit set treatment (control), whereas parthenocarpic fruitswere allowed to develop on the other plants by preventing self-pollinationand applying auxin to the stigma. For node positions 3 to 17,fruit set per node varied between 21 and 55% for control plants[coefficient of variation (CV) = 11%], whereas auxin-treatedplants showed much less variation in fruit set (41–57%;CV = 5%) and average fruit set was higher. In agreement withfruit set, fruit yield was also much more regular in the auxin-treatedplants. Fruit fresh yield varied between 0.2 and 1.0 kg m-2forcontrol plants (CV = 20%), and between 0.4 and 0.8 kg m-2forauxin-treated plants (CV = 9%). Results showed that developingseeds in sweet pepper fruits are the main cause of the abortionof new flowers, and irregular fruit set and yield. Parthenocarpicfruit growth resulted in flatter, 30% smaller fruits, becauseof a reduction in fruit growth rate; the duration of fruit growthwas 1 week longer than for fruits from control plants. Parthenocarpicfruits were hardly affected by blossom-end rot (BER) with only1% of fruits being affected compared to 31% in the control.Total dry mass production was the same for treated and controlplants; however, in auxin-treated plants, 50% of the total drymass was allocated to the fruits, compared to 58% in controlplants. To investigate the functions of recombinant human dentin phosphoprotein (rhDPP), we examined cell adhesion, viability and the odontoblastic differentiation activity of human dental pulp cells (hDPCs). Firstly, rhDPP was constructed using pBAD-HisA plasmid in Escherichia coli. Cell adhesion and viability of hDPCs by rhDPP was examined using a crystal violet assay and a MTT assay, ALP, mineralization activity and odontoblastic differentiation-related mRNA levels of hDPCs were measured to elucidate the odontoblastic differentiation effect of rhDPP on hDPCs. Initially, rhDPP significantly and dose-dependently increased hDPCs adhesion versus the untreated control (p?<?0.05). Cell viability was also significantly increased by rhDPP at 5?days (p?<?0.001). Furthermore, the odontoblastic differentiation effect of rhDPP was verified by measuring ALP activity, mineralization activity and the mRNA levels of odontoblastic differentiation markers. Taken together, rhDPP is expected to play an important role on hDPCs, thereby suggesting its potential use for tooth repair and regeneration.  相似文献

The antioxidant properties of hot-water extracts from a dried chili pepper were maintained through the postharvest ripening process at 10 °C for 18 months. In order to isolate the antioxidant from the ripe pepper, we fractionated hot-water extracts by size-exclusion gel chromatography. A certain fraction showed antioxidative activity via the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity assay. Structural analysis by reversed-phase high performance liquid chromatography (HPLC), LC-MS, and nuclear magnetic resonance (NMR) revealed that the antioxidant was a known compound, p-coumaryl alcohol. This study indicates that an effective antioxidant in chili pepper sustains its antioxidative effects during the postharvest ripening process.  相似文献

Abstract:  The influence of three solanaceous plants (tomato, sweet pepper and eggplant) on the functional response of the predatory bug Picromerus bidens to densities of fourth-instar larvae of the beet armyworm Spodoptera exigua was assessed. Logistic regression indicated a type II functional response on all host plants. Over all prey densities, P. bidens killed significantly fewer fourth instars of S. exigua on tomato than on sweet pepper or eggplant (1.96 ± 0.17 vs. 4.37 ± 0.19 and 3.90 ± 0.15 larvae per predator per 24 h respectively). A higher theoretical maximum predation rate was estimated on sweet pepper (11.1 prey larvae per day) and eggplant (7.4) than on tomato (5.4). The mean number of prey killed per day by P. bidens females ranged from 0.78 at a density of one prey on tomato to 8.45 at a density of 24 prey on sweet pepper. The data indicated that the estimates of handling time ( T h) and attack rate ( a ) were highly affected by host plant. Based on asymptotic 95% confidence intervals, a lower attack coefficient was found on tomato (0.02 h−1) than on sweet pepper or eggplant (0.07 and 0.11 h−1 respectively). On the other hand, handling times were significantly longer on tomato (4.42 h) and eggplant (3.23 h) than on sweet pepper (2.15 h). This laboratory study suggests that plant characteristics influence the ability of P. bidens to respond to changes in prey density.  相似文献
