目的从云南传统发酵豆豉中分离和筛选得到高产γ-氨基丁酸(GABA)的乳酸菌,同时研究其分泌产生的GAD酶学特性,以期为GABA的自动化发酵及GAD的固定化生产提供参考依据。方法通过高效液相色谱分析,筛选到一株高产GABA的乳酸菌,并初步研究其GAD酶学特性。结果从云南传统发酵豆豉中筛选得到高产GA-BA的Lactobacillus plantarum YM-4-3(产量高达5.74 mmol/L,即0.592 g/L),其最适发酵条件为35℃,MSG含量为3%,初始pH 6.0,静置厌氧(5%CO2)发酵96 h;酶学特性研究结果表明,该菌株由来GAD是一种酸性酶,在酸性条件下稳定,最适反应pH为4.0,最适反应温度40℃,辅因子磷酸吡哆醛的最适浓度为200μmol/L。结论云南传统发酵豆豉可作为筛选具有较强GABA转化能力乳酸菌的资源库,该研究将有助于新型乳酸菌发酵豆豉的研发。  相似文献

以大肠杆菌(Escherichia coli)来源的谷氨酸脱羧酶(GadB)为研究对象,通过组合突变,获得了pH适用范围拓宽、催化活力提高和稳定性增强的组合突变体M2。与野生型GadB-WT相比,组合突变体M2的pH适用范围有效拓宽,在pH6.0时催化活力比GadB-WT提高113.43%。之后对含有M2突变体基因重组菌的发酵培养基和诱导条件进行优化,优化后单位培养基酶活力比未优化时提高了104.13%。在此基础上对M2的酶学性质进行测定,测得其最适pH为5.0,最适温度为37℃。通过稳定性测定M2的pH稳定性和热稳定性与野生型GadB-WT相比都有一定程度的增强。M2的动力学参数Km值为7.316μmol/L,kcat为13.387 s-1,kcat/Km为1.830 L/(s·μmol)。研究获得的组合突变体M2进一步丰富了催化合成γ-氨基丁酸的GadB突变体酶库,具有良好应用前景。  相似文献

目的:鉴定1株产γ氨基丁酸(γ-aminobutyric acid,GABA)的乳酸菌HS3,并研究了其谷氨酸脱羧酶(Glutamate decarboxylase,GAD)粗酶酶学性质。方法:根据形态培养特征、生理生化特征和16S rDNA序列比对及系统发育分析对菌株HS3进行了鉴定。采用菌体细胞破碎后的粗酶液,研究了温度、pH和金属离子对酶活的影响。结果:菌株HS3的形态培养和生理生化特征符合肠球菌属(Enterococcus)特征,其16S rDNA序列与Enterococcus faecium(EU717962)16S rDNA序列同源性达99%,鉴定菌株HS3为屎肠球菌(Enterococcus faecium),菌株HS3 GAD最适作用温度为40℃,最适作用pH4.5。酶的热稳定较好,50℃处理4h,在pH3.5~6.0酶活基本稳定。Ca2+对酶有激活作用,5mmol/L和50mmol/L浓度酶活分别提高了37.41%和17.43%。Ba2+和Zn2+在5mmol/L浓度时激活作用明显,而Mg2+在5mmol/L浓度激活作用较好。结论:菌株HS3的GAD活力较高,稳定性较好,为生物合成GABA提供了新的微生物菌种资源。  相似文献

γ-氨基丁酸是哺乳动物体内的一种抑制性神经递质,具有许多重要的生理功能。利用MRS培养基从自然生境中分离了1000余株细菌,经纸层析和HPLC检测,发现其中一株能转化谷氨酸钠生成GABA,产量为4.84g/L;转化产物通过HPLC-MS得到了证实。通过形态特征和生化特性鉴定,初步判断该菌为乳酸菌。  相似文献

基于Berthelot显色测定ω-氨基酸的反应 ,探讨了比色法快速测定谷氨酸脱羧酶活力的测定条件。结果表明 ,该方法灵敏度较高 ,重现性好 ,成本低 ,快捷 ,可替代氨基酸分析仪分析法。对乳酸菌谷氨酸脱羧酶提取液的适宜反应底物体系为 0.2mol LMacIlvaine ,pH4.7,内含 0.1mmol LPLP (5 磷酸吡哆醛 ) ,10mmol L底物L MSG (L 谷氨酸钠 )。 2 0 0 μL底物溶液和 1~ 1 0 0 μL酶液在 3 0℃反应 ,然后冰浴中加入 2 0 0 μ/L 0.  相似文献

谷氨酸脱羧酶(glutamic acid decarboxylase,GAD,EC4.1.1.15)在生物体内广泛存在,其催化产物γ-氨基丁酸(γ-aminobutyric acid,GABA)是哺乳动物体内一种重要的抑制性神经递质。在对自身免疫性疾病以及糖尿病研究中,特别是1型糖尿病,GAD、GABA以及谷氨酸脱羧酶抗体(glutamic acid decarboxylase-antibody,GAD-Ab)等的水平作为病理分析、疾病诊断、免疫治疗的重要参数,历来备受研究者关注。本文就GAD及其催化产物GABA的研究进展进行了综述,为更好地研究自身免疫性疾病的发病机理,探索更加有效安全的治疗方法提供参考。  相似文献

富锌产γ-氨基丁酸乳酸菌的筛选及初步鉴定   总被引:1,自引:0,他引:1  
用含0.2%-0.25%Zn^2+的MRS培养基从发酵酸菜、酸奶中分离出6株具富锌能力细菌,经形态和生理生化特性鉴定,初步判断菌株为乳酸菌。采用纸层析检测MRSG培养基培养的6株菌上清液后,初步确定有4株能产γ-氨基丁酸。以此4菌株基因组为模板PCR扩增到540 bp的片段,经测序和序列分析,证明是谷氨酸脱羧酶基因高度保守区域,初步证明谷氨酸脱羧酶基因的存在。此4株菌发酵液经HPLC检测和分析后,证实2号菌株能转化谷氨酸钠生成GABA,产量为4.8 g/L,在应用方面具有很大的潜力。  相似文献

本文研究了用海藻酸钙包埋法制备含谷氨酸脱羧酶固定化细胞的方法以及研究了制备的条件和影响其制备的因素。该法具有包埋细胞活力回收高,方法简便等优点。比较研究了固定化细胞和自然细胞谷氨酸脱羧酶的一些生物化学性质。其中固定化细胞最适pH和pH稳定性增加,最适温度及热稳定性下降;表观米氏常数增大;二价金属离子Zn~(++)、Cu~(++)、Mg~(++)、Fe~(++),Sr~(++)程度不同的抑制酶活性,Ca~(++)激活固定化细胞酶活性,EDTA无抑制作用。对固定化细胞和自然细胞酶活力活化的研究中发现这两种细胞经蒸馏水保温处理后酶活性都上升,且自然细胞酶活的上升较固定化细胞大;而用底物溶液处理后,自然细胞无变化,固定化细胞酶活下降。  相似文献

Identification and characterization of lactic acid bacteria in ragi tape   总被引:4,自引:0,他引:4  
One hundred and eighteen lactic acid bacteria (LAB) were isolated from five different types of ragi tape, a traditional dry-starter of Balinese rice wine. The isolates could be classified into three groups based on the cell shape and capability to produce gas from glucose. Group I contained 66 homofermentative cocci, group II contained seven homofermentative rods, and group III contained 45 heterofermentative rods. Among these 118 isolates, 21 isolates representing these groups were selected and were first identified using phenotypic characters. The identification performed phenotypically was confirmed by sequencing of variable region 8 (V8) of the 16S rDNA. The comparative studies led to the identification of Pediococcus pentosaceus, Enterococcus faecium, Lactobacillus curvatus, Weissella confusa, and W. paramesenteroides from the ragi tape examined.  相似文献

AIMS: Considering the significant rise in the probiotic market in Columbia, and given the lack of reports concerning the microbial population and strain performance in products from different producers, this study aims at determining the number of viable starter bacteria and probiotics in bio-yoghurts available at the Columbian market, identifying the species and analysing the performance of the isolated strains in bile acid resistance, antagonistic activity against pathogens, and adherence capacity to human intestinal epithelial cells. METHODS AND RESULTS: Seven bio-yoghurts were analysed for the bacterial species present. Species identification was carried out using 16S rRNA gene targeted PCR. The cultured bacteria were tested for bile acid resistance, adherence to a human intestinal epithelial cell line, and antagonism against the pathogen Salmonella enterica serovar Typhimurium. A total of 17 different strains were identified. Based on plate counting, all bio-yoghurts have at least total viable cells of approximately 10(7) CFU ml(-1). Streptococcus thermophilus and Lactobacillus delbrueckii ssp. bulgaricus were the most frequently isolated bacteria. Viable Bifidobacterium was only recovered from one product. However, after PCR analysis, DNA of this genus was confirmed in five out of seven products. Major differences were found for S. typhimurium antagonism. The adherence capacity to Caco-2 cells was observed in 10 of the isolated strains. In general, low survival to simulated gastric juice was observed. CONCLUSIONS: Some of the isolated strains have probiotic potential, although not all of them were present in the advised amount to exert beneficial health effects. However, the full correct scientific name of the isolated bacteria and their viable counts were not included on the product label. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first report describing the identification and functionality of starter bacteria and probiotics present in dairy products on the Columbian market.  相似文献

Boza is a low-alcohol beverage produced from the fermentation of barley, oats, millet, maize, wheat or rice. The number of lactic acid bacteria isolated from three boza samples ranged from 9 × 106 to 5 × 107 CFU/mL. Carbohydrate fermentation reactions and PCR with species-specific primers classified the isolates as Lactobacillus paracasei subsp. paracasei, Lactobacillus pentosus, Lactobacillus plantarum, Lactobacillus brevis, Lactobacillus rhamnosus and Lactobacillus fermentum. No filamentous fungi were isolated. Yeasts were isolated from two of the three boza samples, with cell numbers ranging from 1.3 × 102 to 1.8 × 103 CFU/mL. Results obtained from sequencing of the D1/D2 rDNA region identified the yeasts as Candida diversa, Candida inconspicua, Candida pararugosa, Issatchenkia orientalis, Pichia fermentans, Pichia guillliermondii, Pichia norvegensis, Rhodotorula mucilaginosa and Torulaspora delbrueckii. C. inconspicua has been isolated from human sputum and tongue and is an opportunistic pathogen. R. mucilaginosa is also an opportunistic pathogen implicated in fungaemia, endocarditis and meningitis. P. norvegensis has been associated with septicaemia in humans. Saccharomyces cerevisiae, commonly associated with fermented beverages, has not been detected in any of the boza samples, despite enrichment.  相似文献

The pickle, a traditional fermented vegetable product, is popular in Sichuan Province of China. The objective of this study was to investigate the diversity of dominant lactic acid bacteria (LAB) in pickles by analyzing 36 samples collected from 6 different regions in Sichuan Province. The LAB counts in these samples varied from 3.90 to 8.40 log cfu ml-1. In total, 185 presumptive LAB with Gram-positive and catalase-negative properties were obtained from these samples using MRS agar, and those strains were identified at the species level by physiological tests, 16S rRNA gene sequencing and multiplex PCR assay. The results revealed that all isolates were accurately identified as Enterococcus thailandicus (2 strains), Lactobacillus alimentarius (16 strains), L. brevis (24 strains), L. paracasei (9 strains), L. plantarum (81 strains), L. pentosus (38 strains), L. sakei (8 strains), L. spicheri (1 strain), Leuconostoc lactis (1 strain) and Pediococcus ethanolidurans (5 strains). The predominant LAB in Sichuan pickle was L. plantarum, which were isolated from most samples. The results also demonstrated that different regions in Sichuan Province have complex compositions of LAB species, and such a rich resource of LAB strains provides raw data for further studies involving probiotic strain selection.  相似文献

A modified decarboxylase assay medium (DCA medium) was used for studying the production of biogenic amines by Leuconostoc oenos DSM 20252 and two strains of Lactobacillus buchneri (Lb14 and St2A). The DCA medium contained histidine, lysine, ornithine and tyrosine as precursors of the respective biogenic amines. Under the experimental conditions both strains of Lact. buchneri produced > 90% of the maximum amount of histamine within 24 h. Only tyramine was produced by Leuc. oenos DSM 20252. accountine for 88% of the maximum theoretical amount within 24 h.  相似文献

The lactic acid bacteria of kefir were isolated and characterized using phenotypical, biochemical, and genotypical methods. Polyphasic analyses of results permitted the identification of the microflora to the strain level. The genus Lactobacillus was represented by the species Lb. kefir and Lb. kefiranofaciens. Both subspecies of Lactococcus lactis (lactis and cremoris) were isolated. Leuconostoc mesenteroides subsp. cremoris was also found. The kefir studied contained few species of lactic acid bacteria but showed a high number of different strains. We found that the polyphasic analysis approach increases the confidence in strain determination. It helped confirm strain groupings and it showed that it could have an impact on the phylogeny of the strains.  相似文献

从酸菜汁中分离筛选到一株产共轭亚油酸(CLA)能力较高的乳酸菌。经鉴定,确定为植物乳杆菌Lactobacliius plantarum。微氧条件可提高CLA的产量,催化亚油酸(LA)生成CLA的酶受着LA的诱导。37℃对细胞生长和CLA生成最为有利。对数生长期为6~12h,18h后进入稳定期。在14~22h,CLA生成量快速增加,24h时达到最高值。该菌的培养物经萃取、甲酯化后,进行了气相色谱分离,生成的CLA产物为c9/t9,c11-CLA和t10,c12-CLA异构体的混合物。  相似文献

Five samples of Airag and 20 of Tarag (both in Mongolia) were collected from scattered households. One hundred strains of lactic acid bacteria (LAB) were isolated and identified from these samples according to phenotypic characterization and 16S rRNA gene sequence analysis. Eighty-five isolates belonged to the genus Lactobacillus, 15 being classified as coccoid LAB. All isolates belonged to 5 genera and 11 to different species and subspecies. Lactobacillus (Lb.) helveticus was predominant population in Airag samples, Lb. fermentum and Lb. helveticus were the major LAB microflora in Tarag.  相似文献
