A satisfactory synthetic medium has been developed for continuous growth of Fraxinus pennsylvanica Marsh. callus cultures. The medium contains modified Reinert and White (1956) inorganic nutrient solution with 5 mg/liter Fe as NaFe-EDTA and supplemented with myoinositol 10 mg/liter, pyridoxine HCl 0.1 mg/liter, 2,4-dichlorophenoxyacetic acid 0.04 mg/liter, kinetin 1 mg/liter, sucrose 20 g/liter, and agar 10 g/liter. myo-Inositol, pyridoxine and an auxin are essential. α-Naphthaleneacetic acid is an effective alternate auxin. Kinetin and to some extent gibberellic acid improve the yields. Thiamine has no effect.  相似文献

The potential for biological nitrification of an industrial waste containing 4,000 mg of ammonia N (NH4+-N) and 10,000 mg of fluoride per liter was investigated. Ammonium sulfate and sodium fluoride were tested in various combinations of 100 to 2,000 mg of NH4+-N per liter and 0 to 5,000 mg of F per liter in suspended-growth stirred-tank reactors containing enriched cultures of nitrifying bacteria from a municipal sewage treatment plant. The stirred-tank reactors were fed once per day at a constant hydraulic retention period and cell retention time of 10 days. Temperature was 23°C, and pH was 7.0 to 7.5. Clarified secondary effluent was used to make up feeds and to provide minor nutrients. Steady-state data, confirmed by mass balances, were obtained after five to six retention periods. In the absence of fluoride, nitrification efficiency was near 100% for up to 500 mg of NH4+-N per liter. The influence of fluoride was studied at a low ammonia concentration (100 mg/liter) and exerted no significant effect on nitrification at concentrations of up to 200 mg/liter. Maximum effect of fluoride was reached at 800 mg of F per liter, and no greater inhibition was observed for up to 5,000 mg of F per liter. At the highest concentrations studied, ion pairing of ammonium and fluoride may exert a significant effect on kinetic coefficients. Kinetic analyses showed maximum specific substrate removal rates (qmax) of NH4+-N to be about 2.3 mg of N per mg of volatile suspended solids per day in the absence of fluoride and 0.85 mg of N per mg of volatile suspended solids per day in the presence of fluoride. The form of inhibition due to the presence of fluoride was shown to be not competitive, conforming to a mixed inhibition model.  相似文献

For better operational control of the completely mixed oxygen activated sludge process (CMOAS), a study concerning the kinetics, performance, and operational stability of the Ramanathan-Gaudy model was conducted. Short-term experiments were conducted at various dilution rates (1/9, 1/6, 1/3, 1/1.5, and 1/1.0 hr?1) by using two recycle solids concentration values (5000 and 10,000 mg/liter). The influent substrate was an actual industrial organic wastewater (soft drink waste) and its concentration was maintained at 1000 mg/liter COD. The hydraulic recycle ratio, α, was maintained at 0.30. It was found that for CMOAS system with constant recycle cell concentration, a “steady state” with respect to reactor biological solids and effluent COD at different dilution rates could be attained. No appreciable dilute-out of reactor biological solids and substrate was observed up to the dilution rate of 1 hr?1 for both systems of different XR (5000 and 10,000 mg/liter). For the system of XR = 5000 mg/liter, except the dilution rate of hr?1, the effluent filtrate COD was lower than 100 mg/liter, the aerator biological solids concentration was about 1550 mg/liter, and the COD removal efficiency was higher than 90% for all dilution rates. For the system of XR = 10,000 mg/liter, the effluent filtrate COD was lower than 71 mg/liter, the aerator biological solids concentration was about 2750 mg/liter, and the COD removal efficiency was higher than 90% throughout all the dilution rates selection in the present study. The value of the Sludge Volume Index (SVI) was the range of 37.0 to 58.5 and provided good settleability of sludge. The sludge yield was 0.53 for the system of XR = 5000 mg/liter and 0.57 for the system of XR = 10,000 mg/liter. The carbohydrate and the protein content of the cells were 10.1–21.6% and 35.6–50.6%, respectively. For predicting the reactor biological solid and effluent COD of the CMOAS system by using the Ramanathan-Gaudy model, two sets of values for the biological kinetic constants should be considered since it provided the best fit of predicted values of the observed values. In the present study, μm = 0.4 hr?1, ks = 92 mg/liter for 1/3 ? D ? 1, and μm = 0.05 hr?1, ks = 11.1 mg/liter for 1/9 ? D < 1/3 were used to calculate the predicted values of reactor biological solid and effluent filtrate COD.  相似文献

The effect of the component concentrations of a synthetic medium on acetone and butanol fermentation by Clostridium acetobutylicum ATCC 824 was investigated. Cell growth was dependent on the presence of Mg, Fe, and K in the medium. Mg and Mn had deleterious effects when in excess. Ammonium acetate in excess caused acid fermentation. The metabolism was composed of two phases: an acid phase and a solvent one. Low concentrations of glucose allowed the first phase only. The theoretical ratio of the conversion of glucose to solvents, which was 28 to 33%, was obtained with the following medium: MgSO4, 50 to 200 mg/liter; MnSO4, 0 to 20 mg/liter; KCl, 0.015 to 8 g/liter (an equivalent concentration of K+ was supplied in the form of KH2PO4 and K2HPO4); FeSO4, 1 to 50 mg/liter; ammonium acetate, 1.1 to 2.2 g/liter; para-aminobenzoic acid, 1 mg/liter; biotin, 0.01 mg/liter; glucose, 20 to 60 g/liter.  相似文献

Eighteen strains of obligately oligotrophic bacteria that grow in a medium containing 1 mg of organic carbon per liter and do not grow in a rich medium (5 g/liter of nutrient) were isolated as dominant organisms from the oligotrophic water of Lake Biwa. The growth properties of these, especially of five strains, were examined. The maximum cell yield ranged from 8.5×104/ml to 2.3×106/ml, and their doubling times ranged from 6.6/h to 11.8/h in LT10–4 medium (0.5 mg trypticase and 0.05 mg yeast extract in 1 liter of filtered and aged lake water). They also showed good growth in lake water medium without adding nutrients. The optimum concentrations for their growth were 5 mg/1, 5–50 mg/1, 50 mg/1, or 500 mg/1, depending on the strains. They utilized glutamate, glycine, serine, and glycolate, but not acetate, proline, or leucine. Several properties were examined. Their growth properties were very different from those of oligotrophs or oligocarbophiles isolated by other researchers.  相似文献

Sphagnum peat extracts or hydrolysates have been obtained and used as a culture medium for the production of Candida utilis biomass as single cell proteins. Acid hydrolysis of ground peat (4–60 mesh) in an autoclave operated under a set of conditions for acid strength (0.3-1.5 (v/v) H2SO4), holding time (1–4 hr), temperature (100–165°C), and weight ratio of dry peat to solution (3.3–16.7 g dry peat/100 g solution) yielded carbohydrate-rich extracts of different concentrations (1–34g/liter). The best yield (mg total carbohydrate/g dry peat) was obtained for a holding time of I hr and a temperature of 152°C. Low peat concentratio (4.1 g dry peat/100 g solution)resulted in high yield(280mg total carbohydrate/gdry peat) with a corresponding low carbohydrate content in hydrolysate (13 g/liter), while a lower yield with a higher carbohydrate content (34 g/liter)in hydrolysate were found when increasing peat concentration (16.7 g dry peat/100 g solution). Shake-fladk experiments using peat hydrolysates as the culture medium together with NH4OH (~4.8 g/liter) and K2HPO4(5 g/liter) as nitrogen and phosphate supplement, respectively, gave a maximum biomass concentration of 7.5 g/liter after 60 hr at 30°C and 200rpm. Batch cultivation in a fermentor under controlled conditions for aeration (4.2 liter/min), agitation (500rpm), temperature (30°C), and pH (5.0) produced a maximum biomass of 10 g/liter after 20 hr with a specific growth rate of 0.13 hr?1. For the continuous cultivation, a maximal biomass productivity of 1.24 g/gliter-he was obtained at a dilution rate of 0.125 hr ?1. Monod's equation's equation has been used for the estimation of the coefficients μMax, Ks, and Y. It was found that the yield coefficient Y is not constant during the progress of batch cultivation.  相似文献

Summary Shoot multiplication was induced in cowpea, cv. Georgia-21, from shoot tip explants. Shoot tips, 5 mm long, were isolated from in vitro-grown seedlings and cultured on MS medium containing N6-benzyladenine (BA) at 1, 2.5, or 5 mg/liter (4.4, 11.1, or 22.2 μM) or 6-furfurylaminopurine (kinetin) at 1, 2.5, or 5 mg/liter (4.6, 11.6, or 23.2 μM) combined with 2,4-dichlorophenoxyacetic acid (2,4-D) at 0.01, 0.1, or 0.5 mg/liter (0.05, 0.5, or 2.3 μM) or naphthaleneacetic acid (NAA) at 0.01, 0.1, or 0.5 mg/liter (0.05, 0.5, or 2.7 μM). Cultures were maintained at a 12-h photoperiod (40 μmol·m−2·s−1) and 23 ± 2° C. Treatments with BA induced greater shoot proliferation than those with kinetin. The highest number of shoots was produced on 5 mg (22.2 μM) BA per liter in combination with NAA or 2,4-D at 0.01 mg/liter (0.05 μM). Callus proliferated from the basal ends of shoot pieces in all treatments. The cultures also formed roots in the presence of kinetin, but not on BA-containing medium. To produce whole plants, the shoots were separated and rooted on 0.1 mg (0.5 μM) NAA per liter. Resulting plants grew normally under greenhouse conditions. Shoot tips provide an excellent explant source for cowpea micropropagation and can be used for callus induction.  相似文献

Copper-silver (Cu-Ag) ionization has effectively controlled Legionella spp. in the hot water systems of numerous hospitals. However, it was ineffective at controlling Legionella in one Ohio hospital despite the confirmation of adequate total concentrations of copper and silver ions. The pH of the water at this hospital was found to be 8.5 to 9.0. The purpose of this study was to investigate the impact of pH and other water quality parameters, including alkalinity (HCO3), hardness (Ca2+ and Mg2+), and amount of dissolved organic carbon (DOC), on the control of Legionella by Cu-Ag ionization. Initial concentrations of Legionella and copper and silver ions used in batch experiments were 3 × 106 CFU/ml and 0.4 and 0.08 mg/liter, respectively. Changes in bicarbonate ion concentration (50, 100, and 150 mg/liter), water hardness (Ca2+ at 50 and 100 mg/liter; Mg2+ at 40 and 80 mg/liter), and level of DOC (0.5 and 2 mg/liter) had no significant impact on the efficacy of copper and silver ions in killing Legionella at a neutral pH. When the pH was elevated to 9 in these experiments, copper ions achieved only a 10-fold reduction in the number of Legionella organisms in 24 h, compared to a millionfold decrease at pH 7.0. Silver ions were able to achieve a millionfold reduction in 24 h at all ranges of water quality parameters tested. Precipitation of insoluble copper complexes was observed at a pH above 6.0. These results suggest that pH may be an important factor in the efficacy of copper-silver ionization in controlling Legionella in water systems.  相似文献

The glucose uptake of a species of Chlamydomonas was studied at various concentrations of d -glucose plus glucose-1-14C (0.003–10.0 mg/liter) and at various light levels (0–220 ft-c). The alga grows at 4 C either in the light or in the dark with added glucose, cellobiose, maltose, or fructose. Uptake of glucose could be described by the Michaelis-Menten equation, and both the maximum velocity of uptake and the half-saturation constant increased when the cells were exposed to glucose in the dark. However, the high value of the half-saturation constant (5 mg glucose/liter) compared with the low levels of glucose in nature (5–10 μg/liter) makes it unlikely that a transport system is effective under natural conditions. Even if a total of 10.0 mg/liter of glucose plus other organic compounds were available as substrate, the rate of photosynthesis would still be more than 10 times higher (at 220 ft-c) than the rate of organic substrate uptake. Light had no effect on the total uptake of glucose but did reduce the percentage of 14CO2 evolved from 61% of the total 14CO taken up in the dark to 0% at 220 ft-c. This decrease could be due to either preferential use of the 14CO2 in photosynthesis or of the photosynthate in respiration.  相似文献

The influence of Ca2+, Mg2+, Mn2+, and Fe2+ ions on lipid accumulation, fatty acid composition and arachidonic acid (ARA) production byMortierella sp. S-17 was investigated. A beneficial effect of Mn2+ in the concentration range of 2–500 mg/L on lipogenesis was observed. The other elements at about 1 g/L repressed lipid accumulation and ARA yield. The highest yield of ARA (723 mg per liter or 148 mg per gram of dry mycelium) after incubation of the fungus in a glucose medium in the presence of 2 mg Mn2+ per liter was obtained. A strong inhibitory effect of Fe2+ (above 40 mg/L) on ARA formation was observed.  相似文献

Mixed leukocyte cultures from normal donors were set up in standard tissue culture media. Blast cell transformation was measured 6 days later by 3H-thymidine uptake and assessed qualitatively with stained smears. Media RPMI 1629 and RPMI 1640 allowed much more intense blastogenesis than Medium 199, the difference being due to the low folic acid content of Medium 199 (0.01 mg/liter) compared with RPMI 1629 (10 mg/liter) and RPMI 1640 (1 mg/liter). Further experiments indicated that folic acid increased the multiplication rate of blast cells but did not promote the induction or initial transformation phases of the mixed leukocyte reaction. The effect of folic acid on the PHA response was entirely different: 3H-thymidine uptake in 3 day PHA cultures was decreased, and there was no apparent effect on the number or percentage of blast cells seen on the smears. The reason for this difference is at present unknown, but some possibilities are discussed.  相似文献

Phanerochaete chrysosporium was grown in fermentors on NaOH-extracted maple, pine, and cedar barks at the optimum substrate concentration of 1% (w/v). The yields (mg protein/liter) on maple, pine, and cedar were 1500, 1200, and 880, respectively, which are probably due to the different lignin contents of the barks. Lignin is not utilized. The productivities at 30°C obtained for pine (4.07 × 10?2 g protein/liter hr) and cedar (2.63 × 10?2 g protein/liter hr) barks were greater than for maple (2.63 × 10?2 g protein/liter hr). The substrate (bark) was the limiting component of the fermentation. Over the 26–38°C temperature range protein productivity increased by a factor of three (1.55 × 10?2 vs. 4.61 × 10?2 g protein/liter hr) for maple bark. Low agitation rates resulted in an overproduction of cellulase and reduced levels of microbial protein.  相似文献

The effects of Cd, Cu, and Ni on pure cultures of thermophilic methanogenic bacteria were studied. The bacteria used wereMethanobacterium thermoautotrophicum and TAM, a thermophilic, acetate-decarboxylating, methanogenic bacterium. Much lower concentrations of heavy metals were needed to cause initial inhibition of TAM (1 mg/liter Cu and Cd; 5 mg/liter Ni) compared withM. thermoautotrophicum (10 mg/liter Cu and Cd; and 100 mg/liter Ni). No growth of TAM occurred at 5 mg/liter Cu and 25 mg/liter Ni, while the corresponding values forM. thermoautotrophicum were 50 mg/liter Cu and 200 mg/liter Ni. Cd (50 mg/liter) was totally inhibitory toM. thermoautotrophicum but allowed minimal growth of TAM. Ni stimulated both organisms at an optimal concentration of 5 mg/liter forM. thermoautotrophicum and 1 mg/liter for TAM. The toxicity of Cd and Cu was found to depend upon the presence of Ni in the medium.  相似文献

Summary Side shoots excised from underground dormant buds ofCynara scolymus L. were used as primary explants to establishin vitro cultures. A 3×3 factorial experiment with all possible combinations of three concentrations (0.5, 1.0, 2.0 mg/liter or 2.22, 4.44, 8.88 μM) ofN 6-benzyladenine (BA) and three concentrations (0, 0.1, 0.2 mg/liter or 0, 0.54, 1.07 μM) of 1-naphthaleneacetic acid (NAA) was used to determine the optimum growth regulator combination for shoot multiplication. The highest rate of axillary shoots was induced on Murashige and Skoog agar medium supplemented with 0 mg NAA/liter and 1.0 mg BA/liter (4.44 μM). Other cytokinins tested (kinetin, zeatin, and 2-isopentenyl-adenine were less effective than BA in inducing axillary shoot growth. Up to 60% of elongated microshoots rooted after 5 weeks on 1/2 MS agar medium supplemented with 2 mg/liter (11.42 μM) indole-3-acetic acid (IAA). Seventy percent of rooted plantlets were transferred successfully into soil. Plants are under evaluation for their genetic uniformity and clonal fidelity.  相似文献

A synergistic effect between silver and UV radiation has been observed that can appreciably enhance the effectiveness of UV radiation for inactivation of viruses. At a fluence of ca. 40 mJ/cm2, the synergistic effect between silver and UV was observed at silver concentrations as low as 10 μg/liter (P < 0.0615). At the same fluence, an MS-2 inactivation of ca. 3.5 logs (99.97%) was achieved at a silver concentration of 0.1 mg/liter, a significant improvement (P < 0.0001) over the ca. 1.8-log (98.42%) inactivation of MS-2 at ca. 40 mJ/cm2 in the absence of silver. Modified Chick-Watson kinetics were used to model the synergistic effect of silver and UV radiation. For an MS-2 inactivation of 4 logs (99.99%), the coefficient of dilution (n) was determined to be 0.31, which suggests that changes in fluence have a greater influence on inactivation than does a proportionate change in silver concentration.  相似文献

A simple and rapid technique of measuring pyrophosphates in plasma, urine, and bone tissue is described, using the UDPG-pyrophosphorylase reaction and fluorimetrical determination of NADPH formed in a combined system of phosphorylation and reduction. This very specific method avoids the separation of Pi2 by column chromatography, and its very great sensitivity enables measurement on a final sample corresponding to 0.2 ml of plasma, with a precision of about 3%.The mean plasma PPi concentration (±SE of mean) was 3.53 ± 0.19 μmoles/liter (SE=0.93), or 0.207 ± 0.006 mg Pi/liter. The normal range for this population (99% confidence limit) is therefore between 1.10 and 5.90 μmoles/liter or 0.068 – 0.366 mg Pi/liter. Analysis of the variation of the duplicate measurements shows a very small divergence of 3.4% or ±0.12 μm.Normal 24 hr urinary excretion is 53.0 ± 6.8 μmoles with 99% confidence limits of 10.0 and 96.0 μmoles.The average PPi concentration in calvaria of 20 10-day-old rats is 3.685 ± 0.16 nmoles/mg fresh weight.  相似文献

Summary Protoplasts ofBrassica nigra (L.) Koch were isolated from stem peels of bolting racemes and cultured in 1.5 ml of VN1 liquid medium. The protoplasts in the liquid medium were plated on top of half strength MS medium supplemented with 400 mg/liter glutamine, 15 mg/liter glutathione, 50 mg/literl-serine, 0.25 mg/liter 6-benzylaminopurine, 0.5 mg/liter 2,4-dichlorophenoxyacetic acid, 1.5% sucrose, and 5% mannitol, pH, 5.7, solidified with 0.3% agarose. Ten percent of calli obtained from the protoplasts developed into plantlets within 4 wk after transfer onto 2N regeneration medium which contains MS salts plus 200 mg/liter casein hydrolysate, 0.625 mg/liter 6-benzylaminopurine, 0.625 mg/liter kinetin, 0.625 mg/liter 6-(γ,γ-dimethylallylamino)-purine, 0.625 mg/liter zeatin, 0.5 mg/liter 1-naphthaleneacetic acid, 1.5% sucrose, and 0.4% agarose. THis is the first report of plant regeneration fromB. nigra protoplasts.  相似文献

The mummichog, Fundulus heteroclitus (L.), an estuarine cyprinodontiformteleost, is used extensively and increasingly as a bioassayorganism in toxicological investigations and as an indicatorof marine water quality owing, in part, to its wide geographicrange, abundance throughout most of that range, and adaptabilityto laboratory conditions. Available data are summarized foracute toxicities to mummichog adults of 118 contaminants at22±2% salinity, 19.5±0.5°C, pH 7.8±0.2,and dissolved oxygen >4.0 mg/liter. Organochlorine insecticideswere the most toxic chemicals tested under these conditions;approximate concentrations of 15 organochlorine pesticide compoundsfatal to 50% in 96 hr ranged between 0.0001 mg/liter and 0.1mg/liter. Comparable data for other groups of chemicals were:11 organophosphorus insecticides, 0.001 and 100.0 mg/ liter;30 trace metals and metalloids, 0.01 and 10,000.0 mg/liter;34 synthetic detergents, 1.0 and 1,000.0 mg/liter; 10 chemicaloil dispersants, 1.0 and 10,000.0 mg/liter; 13 oildispersantmixtures, 100.0 and 10,000.0 mg/liter; 3 household soaps, 100.0and 10,000.0 mg/liter; and 2 crude oils, 1,000.0 and>10,000.0mg/liter. Parameters currently used to evaluate sublethal effectsof pollutants in mummichogs include bioaccumulation, histopathology,growth and early development, teratology, liver enzymes, bloodchemistry, fin regeneration, organ weight, and behavior; theseare illustrated with appropriate examples. Beta vulgaris cells were cultured in specially constructed fluidized bed reactors on Murashige and Skoog medium with 3% sucrose as a carbon source. Biomass density levels of approximately 17–18 g dry cell weight liter–1 for 5 liter fermenters and 9–10 g dry cell weight liter–1 for 50 liter reactor were observed. Pith-derived calluses of Nicotiana tabacum L. cv. Wisconsinno. 38 were inoculated on an RM-1964 medium containing variousconcentrations of a morphactin, chlorflurenol (CFl) and kinetin(KIN). An addition of KIN (0.1–2 mg/liter) alone was effectivefor shoot formation from the calluses, but a high dose (10 mg/liter)resulted in the inhibition of growth and in no differentiation.The inhibitory effect of a high dose of KIN was counteractedwith CFl. Three combinations of KIN and CFl; CFl (0.1 mg/liter)$KIN(2.0 mg/liter); CFl (0.1 mg/liter)$KIN (10.0 mg/liter) and CFl(1.0 mg/liter) $KIN (2.0 mg/liter), were successful for 100%shoot redifferentiation in inoculated calluses. An appropriatebalance between CFl and KIN seems to be involved in shoot formation.The present results can best be interpreted by assuming thatCFl acts as an auxin in cultured tissues.
