Images of chlorophyll fluorescence were used to demonstrate patchy stomatal closure at low humidities in leaves of well-watered Xanthium strumarium plants. The pattern and extent of patchy stomatal closure were shown to be different for the two surfaces of amphistomatous leaves by taking images of leaves with CO2 available to only one leaf was exposed to low humidity, patchiness was more extensive on that surface. Gas-exchange experiments were also conducted to determine the apparent photosynthetic capacity of the mesophyll (photosynthesis rate at constant ci when it was supplied with CO2 through both surfaces or through each surface alone. These experiments showed declines in the apparent photosynthetic capacity of the mesophyll at low humidities that were consistent with patchy stomatal closure on one or both surfaces. The results suggest that patchy stomatal closure can be a factor in the steady-state reponses of stomata to humidity. In amphistomatous leaves this is further complicated by the fact that patches on one epidermis may not coincide with those of the other surface.  相似文献

Gas exchange and abscisic acid content of Digitalis lanata EHRH. have been examined at different levels of plant water stress. Net photosynthesis, transpiration and conductance of attached leaves declined rapidly at first, then more slowly following the withholding of irrigation. The intercellular partial pressure of CO2 decreased slightly. The concentration of 2-cis(S)ABA increased about eight-fold in the leaves of non-irrigated plants as compared with well-watered controls. A close linear correlation was found between the ABA content of the leaves and their conductance on a leaf area basis. In contrast, the plot of net assimilation versus ABA concentration was curvilinear, leading to an increased efficiency of water use during stress. After rewatering, photosynthesis reached control values earlier than transpiration, leaf conductance and ABA content. (±)-Abscisic acid (ABA) at 10-5 M was added to the transpiration stream of leaves of 16 species (C3 and C4, monocotyledons and dicotyledons). Stomatal responses followed one of three patterns: i) stomata that were wide and insensitive to CO2 initially, closed partially and became sensitive to CO2; ii) for stomata that were sensitive to CO2 before the application of ABA, the range of highest sensitivity to CO2 shifted from high to low intercellular partial pressures of CO2, for instance in leaves of Zea mays from 170–350 to 70–140 bar; iii) when stomata responded strongly to ABA, their conductance was reduced to a small fraction of the initial conductance, and sensitivity to CO2 was lost. The photosynthetic apparatus was affected by applications of ABA to various degrees, from no response at all (in agreement with several previous reports on the absence of effects of ABA on photosynthesis) through a temporary decrease of its activity to a lasting reduction. Saturation curves of photosynthesis with respect to the partial pressure of CO2 in the intercellular spaces indicated that application of ABA could produce three phenomena: i) a reduction of the initial slope of the saturation curve (which indicates a diminished carboxylation efficiency); ii) a reduction of the level of the CO2-saturated rate of assimilation (which indicates a reduction of the ribulose-1,5-bisphosphate regeneration capacity); and iii) an increase of the CO2 compensation point. Photosynthesis of isolated mesophyll cells was not affected by ABA treatments. Responses of the stomatal and photosynthetic apparatus were usually synchronous and often proportional to each other, with the result that the partial pressure of CO2 in the intercellular spaces frequently remained constant in spite of large changes in conductance and assimilation rate. Guard cells and the photosynthetic apparatus were able to recover from effects of ABA applications while the ABA supply continued. Recovery was usually partial, in the case of the photosynthetic apparatus occasionally complete. Summary The gas exchange characteristics of photosynthetic tissues of leaves and stems of Eriogonum inflatum are described. Inflated stems were found to contain extraordinarily high internal CO2 concentrations (to 14000 bar), but fixation of this internal CO2 was 6–10 times slower than fixation of atmospheric CO2 by these stems. Although the pool of CO2 is a trivial source of CO2 for stem photosynthesis, it may result in higher water-use efficiency of stem tissues. Leaf and stem photosynthetic activities were compared by means of CO2 fixation in CO2 response curves, light and temperature response curves in IRGA systems, and by means of O2 exchange at CO2 saturation in a leaf disc O2 electrode system. On an area basis leaves contain about twice the chlorophyll and nitrogen as stems, and are capable of up to 4-times the absolute CO2 and O2 exchange rates. However, the stem shape is such that lighting of the shaded side leads to a substantial increase in overall stem photosynthesis on a projected area basis, to about half the leaf rate in air. Stem conductance is lower than leaf conductance under most conditions and is less sensitive to high temperature or high VPD. Under most conditions, the ratio C i /C a is lower in stems than in leaves and stems show greater water-use efficiency (higher ratio assimilation/transpiration) as a function of VPD. This potential advantage of stem photosynthesis in a water limited environment may be offset by the higher VPD conditions in the hotter, drier part of the year when stems are active after leaves have senesced. Restrictions to photosynthesis can limit plant growth at high temperature in a variety of ways. In addition to increasing photorespiration, moderately high temperatures (35–42 °C) can cause direct injury to the photosynthetic apparatus. Both carbon metabolism and thylakoid reactions have been suggested as the primary site of injury at these temperatures. In the present study this issue was addressed by first characterizing leaf temperature dynamics in Pima cotton (Gossypium barbadense) grown under irrigation in the US desert south‐west. It was found that cotton leaves repeatedly reached temperatures above 40 °C and could fluctuate as much as 8 or 10 °C in a matter of seconds. Laboratory studies revealed a maximum photosynthetic rate at 30–33 °C that declined by 22% at 45 °C. The majority of the inhibition persisted upon return to 30 °C. The mechanism of this limitation was assessed by measuring the response of photosynthesis to CO2 in the laboratory. The first time a cotton leaf (grown at 30 °C) was exposed to 45 °C, photosynthetic electron transport was stimulated (at high CO2) because of an increased flux through the photorespiratory pathway. However, upon cooling back to 30 °C, photosynthetic electron transport was inhibited and fell substantially below the level measured before the heat treatment. In the field, the response of assimilation (A) to various internal levels of CO2 (Ci) revealed that photosynthesis was limited by ribulose‐1,5‐bisphosphate (RuBP) regeneration at normal levels of CO2 (presumably because of limitations in thylakoid reactions needed to support RuBP regeneration). There was no evidence of a ribulose‐1,5‐bisphosphate carboxylase/oxygenase (Rubisco) limitation at air levels of CO2 and at no point on any of 30 ACi curves measured on leaves at temperatures from 28 to 39 °C was RuBP regeneration capacity measured to be in substantial excess of the capacity of Rubisco to use RuBP. It is therefore concluded that photosynthesis in field‐grown Pima cotton leaves is functionally limited by photosynthetic electron transport and RuBP regeneration capacity, not Rubisco activity.  相似文献

The response of photosynthetic carbon assimilation and chlorophyll fluorescence quenching to changes in intercellular CO2 partial pressure (Ci), O2 partial pressure, and leaf temperature (15-35°C) in triazine-resistant and -susceptible biotypes of Brassica napus were examined to determine the effects of the changes in the resistant biotype on the overall process of photosynthesis in intact leaves. Three categories of photosynthetic regulation were observed. The first category of photosynthetic response, ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco)-limited photosynthesis, was observed at 15, 25, and 35°C leaf temperatures with low Ci. When the carbon assimilation rate was Rubisco-limited, there was little difference between the resistant and susceptible biotypes, and Rubisco activity parameters were similar between the two biotypes. A second category, called feedback-limited photosynthesis, was evident at 15 and 25°C above 300 microbars Ci. The third category, photosynthetic electron transport-limited photosynthesis, was evident at 25 and 35°C at moderate to high CO2. At low temperature, when the response curves of carbon assimilation to Ci indicated little or no electron transport limitation, the carbon assimilation rate was similar in the resistant and susceptible biotypes. With increasing temperature, more electron transport-limited carbon assimilation was observed, and a greater difference between resistant and susceptible biotypes was observed. These observations reveal the increasing importance of photosynthetic electron transport in controlling the overall rate of photosynthesis in the resistant biotype as temperature increases. Photochemical quenching of chlorophyll fluorescence (qP) in the resistant biotype never exceeded 60%, and triazine resistance effects were more evident when the susceptible biotype had greater than 60% qP, but not when it had less than 60% qP.  相似文献

Leaves of C3 plants which exhibit a normal O2 inhibition of CO2 fixation at less than saturating light intensity were found to exhibit O2-insensitive photosynthesis at high light. This behavior was observed in Phaseolus vulgaris L., Xanthium strumarium L., and Scrophularia desertorum (Shaw.) Munz. O2-insensitive photosynthesis has been reported in nine other C3 species and usually occurred when the intercellular CO2 pressure was about double the normal pressure. A lack of O2 inhibition of photosynthesis was always accompanied by a failure of increased CO2 pressure to stimulate photosynthesis to the expected degree. O2-insensitive photosynthesis also occurred after plants had been water stressed. Under such conditions, however, photosynthesis became O2 and CO2 insensitive at physiological CO2 pressures. Postillumination CO2 exchange kinetics showed that O2 and CO2 insensitivity was not the result of elimination of photorespiration.

It is proposed that O2 and CO2 insensitivity occurs when the concentration of phosphate in the chloroplast stroma cannot be both high enough to allow photophosphorylation and low enough to allow starch and sucrose synthesis at the rates required by the rest of the photosynthetic component processes. Under these conditions, the energy diverted to photorespiration does not adversely affect the potential for CO2 assimilation.

Gas exchange and contents of photosynthetic intermediates of leaves of Arbutus unedo L. were determined with the aim of recognizing the mechanisms of inhibition that were responsible for the midday depression of photosynthesis following exposure to dry air, and the decline in photosynthetic capacity following application of abscisic acid (ABA). Rapidly killed (<0.1 s) leaf samples were taken when gas analysis showed reduced CO2 assimilation. Determination of the contents of 3-phosphoglyceric acid (PGA), ribulose 1,5-bisphosphate (RuBP), triose phosphates, fructose 1,6-bisphosphate and hexose phosphates in the samples showed that significant variation occurred only in the level of PGA. As a result, the ratio PGA/RuBP decreased with increasing inhibition of photosynthesis, particularly when application of ABA had been the cause. A comparison of metabolite patterns did not bring out qualitative differences that would have indicated that effects of ABA and of dry air had been caused by separate mechanisms. Glycerol induced a limitation on photosynthetic carbon assimilation by phosphate when supplied to leaves of barley (Hordeum vulgare L.) and spinach (Spinacia oleracea L.). This limitation by phosphate was evidenced by (i) reversibility of the inhibition of photosynthesis by glycerol by feeding orthophosphate (ii) a decrease in light-saturated rates of photosynthesis and saturation at a lower irradiance, (iii) the promotion of oscillations in photosynthetic CO2 assimilation and in chlorophyll fluorescence, (iv) decreases in the pools of hexose monophosphates and triose phosphates and increases in the ratio of glycerate-3-phosphate to triose phosphate, (v) decreased photochemical quenching of chlorophyll fluorescence, and increased non-photochemical quenching, specifically of the component which relaxed rapidly, indicating that thylakoid energisation had increased. In barley there was a massive accumulation of glycerol-3-phosphate and an increase in the period of the oscillations, but in spinach the accumulation of glycerol-3-phosphate was comparatively slight. Several component processes of photosynthesis were measured in osmotically stressed mesophyll cells of Xanthium strumarium L. The ribulose-1,5-bisphosphate regeneration capacity was reduced by water stress. Photophoshorylation was sensitive to water stress but photosynthetic electron transport was unaffected by water potentials down to-40 bar (-4 MPa). Soybean [Glycine max (L.) Merr. cv. Williams 82 and A3127] plants were grown in the field under long-term soil moisture deficit and irrigation to determine the effects of severe drought stress on the photosynthetic capacity of soybean leaves. Afternoon leaf water potentials, stomatal conductances, intercellular CO2 concentrations and CO2-assimilation rates for the two soil moisture treatments were compared during the pod elongation and seed enlargement stages of crop development. Leaf CO2-assimilation rates were measured with either ambient (340 l CO2 l–1) or CO2-enriched (1800 l CO2 l–1) air. Although seed yield and leaf area per plant were decreased an average of 48 and 31%, respectively, as a result of drought stress, leaf water potentials were reduced only an average of 0.27 MPa during the sampling period. Afternoon leaf CO2-assimilation rates measured with ambient air were decreased an average of 56 and 49% by soil moisture deficit for Williams 82 and A3127, respectively. The reductions in leaf photosynthesis of both cultivars were associated with similar decreases in leaf stomatal conductance and with small increases in leaf intercellular CO2 concentration. When the CO2-enriched air was used, similar afternoon leaf CO2-assimilation rates were found between the soil moisture treatments at each stage of crop development. The effect of long-term (7 days) and shortterm (up to 2 h) treatment of barley plants with jasmonic acid (JA) on the components contributing to stomatal and nonstomatal limitation of photosynthesis was studied. Net CO2 assimilation rate (A) responses to intercellular CO2 concentration (C i ), i.e., A/C i curves, were used to assess the photosynthetic ability. Long-term treatment of barley plants with JA led to a noticeable decrease in both the initial slope of the A/C i curves and the maximum A at saturating C i . The proportion of stomatal and nonstomatal factors in limitation of photosynthesis depended on the applied JA concentration. Short-term treatment with JA affected neither the stomatal conductivity for CO2 nor the rate of photosynthetic CO2 assimilation. We suggest that JA may affect photosynthesis indirectly, either as a stress-modulating substance, or through the alterations in gene expression.  相似文献

Most mistletoe–host ecophysiological studies have paid attention to the influence of parasites on host performance. This paper explored the impact of varying hosts on the photosynthesis of a single mistletoe species. Here, we studied an obligate xylem‐tapping tropical mistletoe (Dendrophthoe curvata (Blume) Miquel) parasitizing four different hosts (Acacia auriculiformis A. Cunn. Ex Benth, Andira inermis (W. Wright) DC., Mangifera indica L. and Vitex pinnata L.) in a homo geneous tropical heath forest patch in Brunei Darussalam. We compared photosynthetic capacity and photosynthesis‐related characteristics of the mistletoe on four different hosts to evaluate the overall impact of hosts on the parasite. Results showed that the mistletoe–host patterns of CO2 assimilation rates, transpiration rates and water use efficiency varied significantly based on the host. In the D. curvata–Vitex pinnata association, the mistletoe exhibited significantly lower CO2 assimilation rates but showed no significant variations in transpiration rates and water use efficiency when compared to the host. In D. curvata–Andira inermis and D. curvata–Mangifera indica associations, the mistletoe showed significantly higher photosynthetic rates than the hosts, whereas in the D. curvata–Acacia auriculiformis association, there was no significant difference in photosynthetic rates between the counterparts. Host specificity also significantly influenced some mistletoe photosynthetic parameters such as light saturated photosynthesis, specific leaf area, leaf chlorophyll content, CO2 assimilation rates, stomatal conductance, transpiration rates and water use efficiency. Different tree hosts intrinsically offer different resources to their obligate mistletoe parasites based on their physiology and environmental parameters. We argue that host‐specific responses have driven these intra‐specific variations in mistletoe physiology. This study provides background for future investigation on potential host‐regulated mechanisms that drive functional changes in host‐dependent mistletoes.  相似文献

Castor bean (Ricinus communis L.) has a high photosynthetic capacity under high humidity and a pronounced sensitivity of photosynthesis to high water vapor pressure deficit (VPD). The sensitivity of photosynthesis to varying VPD was analyzed by measuring CO2 assimilation, stomatal conductance (gs), quantum yield of photosystem II (II), and nonphotochemical quenching of chlorophyll fluorescence (qN) under different VPD. Under both medium (1000) and high (1800 micromoles quanta per square meter per second) light intensities, CO2 assimilation decreased as the VPD between the leaf and the air around the leaf increased. The gs initially dropped rapidly with increasing VPD and then showed a slower decrease above a VPD of 10 to 20 millibars. Over a temperature range from 20 to 40°C, CO2 assimilation and gs were inhibited by high VPD (20 millibars). However, the rate of transpiration increased with increasing temperature at either low or high VPD due to an increase in gs. The relative inhibition of photosynthesis under photorespiring (atmospheric levels of CO2 and O2) versus nonphotorespiring (700 microbars CO2 and 2% O2) conditions was greater under high VPD (30 millibars) than under low VPD (3 millibars). Also, with increasing light intensity the relative inhibition of photosynthesis by O2 increased under high VPD, but decreased under low VPD. The effect of high VPD on photosynthesis under various conditions could not be totally accounted for by the decrease in the intercellular CO2 in the leaf (Ci) where Ci was estimated from gas exchange measurements. However, estimates of Ci from measurements of II and qN suggest that the decrease in photosynthesis and increase in photorespiration under high VPD can be totally accounted for by stomatal closure and a decrease in Ci. The results also suggest that nonuniform closure of stomata may occur in well-watered plants under high VPD, causing overestimates in the calculation of Ci from gas exchange measurements. Under low VPD, 30°C, high light, and saturating CO2, castor bean (C3 tropical shrub) has a rate of photosynthesis (61 micromoles CO2 per square meter per second) that is about 50% higher than that of tobacco (C3) or maize (C4) under the same conditions. The chlorophyll content, total soluble protein, and ribulose-1,5-bisphosphate carboxylase/oxygenase level on a leaf area basis were much higher in castor bean than in maize or tobacco, which accounts for its high rates of photosynthesis under low VPD.  相似文献

Despite the observed impact of water stress on photosynthesis, some of the most used models of CO2 assimilation in C3 and C4 functional types do not directly account for it. We discuss an extension of these models, which explicitly includes the metabolic and diffusive limitations due to water stress on photosynthesis. Functional relationships describing the photosynthetic processes and CO2 diffusion inside leaves are modified to account for leaf water status on the basis of experimental results available in the literature. Extensive comparison with data shows that the model is suitable to describe the reduction in CO2 assimilation rate with decreasing leaf water potentials in various species. A simultaneous analysis of photosynthesis, transpiration and soil moisture dynamics is then carried out to explore the actual impact of drought on different photosynthesis processes and on the overall plant activity. The model well reproduces measured CO2 assimilation rate as a function of soil moisture and could be useful to formulate hypotheses for detailed experiments as well as to simulate in detail transpiration and photosynthesis dynamics under water stress.  相似文献

为揭示油橄榄(Olea europaea L.)耐旱性与光合特性之间的关系,以筛选出的适宜于半干旱川西南地区种植的7个引进油橄榄品种为供试材料,采用盆栽模拟干旱胁迫的方法,研究持续干旱胁迫对其光合特性的影响。结果表明:(1)随着干旱胁迫程度加剧,7个油橄榄品种叶片相对含水量均显著降低,至干旱胁迫后期(25d),各品种叶片均出现大幅失水,其中品种‘科拉蒂’失水率最高(45.79%),而品种‘小苹果’失水率最低(25.52%),说明‘小苹果’叶片在干旱胁迫下较其他油橄榄品种具有更高保水能力。(2)随着干旱胁迫程度加剧,7个油橄榄品种叶片光合色素含量均不同程度降低,表明光合色素分解量大于合成量;干旱胁迫持续25d时,品种‘豆果’的叶绿素a和叶绿素b含量下降幅度最大(P0.05),品种‘皮削利’类胡萝卜素含量下降幅度最大(P0.05),而品种‘小苹果’叶绿素a含量下降幅度最小。(3)随着干旱胁迫的持续进行,各油橄榄品种叶片净光合速率(Pn)、蒸腾速率(Tr)、气孔导度(Gs)和胞间CO2浓度(Ci)均不同程度降低,而水分利用效率(WUE)则呈上升趋势;干旱胁迫期间,品种‘佛奥’的Pn、Tr和Ci以及‘皮削利’的Gs降幅均高于其他品种,而‘小苹果’的Pn、Gs和Ci降幅均为最小且WUE上升幅度最大。研究发现,在持续干旱胁迫条件下,油橄榄幼苗叶片均大幅失水,光合色素结构被破坏、色素分解、含量降低,同时气孔关闭蒸发减少,光合作用减弱,而供试油橄榄品种中‘小苹果’对干旱胁迫的适应性最强,适宜于在半干旱的川西地区种植。  相似文献

The effect of leaf water potential () on net CO2 assimilation rate (A), stomatal conductance (g), transpiration (E) and water-use efficiency (WUE) was measured for three cultivars of cacao (Theobroma cacao L.) seedlings during three recurrent drought cycles. Net assimilation varied greatly at high water potentials, but as dropped below approximately -0.8 and -1.0 MPa, A was reduced to less than 1.5 mol CO2 m-2 s-1. The relation between g and A was highly significant and conformed to an asymptotic exponential model, with A approaching maximal values at stomatal conductances of 55–65 mmol H2O m-2 s-1. Net assimilation varied linearly (r=0.95) with transpiration, and the slope of the A-E relation (WUE) was approximately 3.0 mol CO2 mmol-1 H2O throughout the range of stomatal conductances observed. C i was insensitive to water stress, even though both g and A were strongly affected. Under the experimental conditions used here, mesophyll photosynthesis did not appear to control g through changes in C i. As stress intensified within each drying cycle, WUE of nonirrigated seedlings did not decline relative to that of controls even though CO2 and water vapor exchange rates underwent large displacements. Diffusion of inorganic carbon into isolated bundle sheath cells from a variety of C4 species was characterized by coupling inward diffusion of CO2 to photosynthetic carbon assimilation. The average permeability coefficient for CO2 (PCO2) for five representatives from the three decarboxylation types was approximately 20 micromoles per minute per milligram chlorophyll per millimolar, on a leaf chlorophyll basis. The average value for the NAD-ME species Panicum miliaceum (10 determinations) was 26 with a standard deviation of 6 micromoles per minute per milligram chlorophyll per millimolar, on a leaf chlorophyll basis. A PCO2 of at least 500 micromoles per minute per milligram chlorophyll per millimolar was determined for cells isolated from the C3 plant Xanthium strumarium. It is concluded that bundle sheath cells are one to two orders of magnitude less permeable to CO2 than C3 photosynthetic cells. These data also suggest that CO2 diffusion in bundle sheath cells may be made up of two components, one involving an apoplastic path and the other a symplastic (plasmodesmatal) path, each contributing approximately equally.  相似文献

Despite mounting evidence showing that C4 plants can accumulate more biomass at elevated CO2 partial pressure (p(CO2)), the underlying mechanisms of this response are still largely unclear. In this paper, we review the current state of knowledge regarding the response of C4 plants to elevated p(CO2) and discuss the likely mechanisms. We identify two main routes through which elevated p(CO2) can stimulate the growth of both well-watered and water-stressed C4 plants. First, through enhanced leaf CO2 assimilation rates due to increased intercellular p(CO2). Second, through reduced stomatal conductance and subsequently leaf transpiration rates. Reduced transpiration rates can stimulate leaf CO2 assimilation and growth rates by conserving soil water, improving shoot water relations and increasing leaf temperature. We argue that bundle sheath leakiness, direct CO2 fixation in the bundle sheath or the presence of C3-like photosynthesis in young C4 leaves are unlikely explanations for the high CO2-responsiveness of C4 photosynthesis. The interactions between elevated p(CO2), leaf temperature and shoot water relations on the growth and photosynthesis of C4 plants are identified as key areas needing urgent research.  相似文献

The response of photosynthetic CO2 assimilation to salinization in 19 year old Prunus salicina was evaluated under field conditions for a 3 year period. The observed decline in CO2 assimilation capacity was apparently related to increasing leaf chloride (Cl) content, and independent of changes in leaf carbohydrate status. The response of net CO2 assimilation (A) to leaf intercellular CO2 partial pressure (Ci) indicated that the reduction in the capacity for A with Cl was not the result of decreased stomatal conductance but a consequence of nonstomatal inhibition. The nonstomatal limitations to CO2 assimilation capacity, as determined by the response of A to Ci and biochemical assay, were related to a decline in the activity of ribulose 1,5-bisphosphate carboxylase (Rubpcase) and the pool size of triose phosphate, ribulose 1,5-bisphosphate (Rubp) and phosphoglycerate with increasing salinity. Lack of agreement between the initial slope of the A to Ci response curve and Rubpcase activity suggests the occurrence of heterogeneous stomatal apertures with the high salinity treatment (28 millimolar). Prolonged exposure to chloride salts appeared to increase the Rubp or Pi regeneration limitation, decrease Rubpcase activity and reduce leaf chlorophyll content. Observed changes in the biochemical components of CO2 fixation may, in turn, affect total leaf carbohydrates, which also declined with time and salinity. The reduction in Rubpcase activity was apparently a consequence of a reduced Rubpcase protein level rather than either a regulatory or inhibitory effect.  相似文献
