Summary The first of two major steps in the infection process in roots ofParasponia rigida (Ulmaceae) following inoculation byRhizobium strain RP501 involves the invasion ofRhizobium into the intercellular space system of the root cortex. The earliest sign of root nodule initiation is the presence of clumps of multicellular root hairs (MCRH), a response apparently unique amongRhizobium-root associations. At the same time or shortly after MCRH are first visible, cell divisions are initiated in the outer root cortex of the host plant, always subjacent to the MCRH. No infection threads were observed in root hairs or cortical cells in early stages. Rhizobial entry through the epidermis and into the root cortex was shown to occur via intercellular invasion at the bases of MCRH. The second major step in the infection process is the actual infectionper se of host cells by the rhizobia and formation of typical intracellular infection threads with host cell accommodation. This infection step is probably the beginning of the truly symbiotic relationship in these nodules. Rhizobial invasion and infection are accompanied by host cortical cell divisions which result in a callus-like mass of cortical cells. In addition to infection thread formation in some of these host cortical cells, another type of rhizobial proliferation was observed in which large accumulations of rhizobia in intercellular spaces are associated with host cell wall distortion, deposition of electron-dense material in the walls, and occasional deleterious effects on host cell cytoplasm.  相似文献

The germ tubes ofColletotrichum lagenarium showed negative phototropism to UV-blue (300–520 nm) and far-red (>700 nm) regions with maximum in the near ultraviolet (NUV) region, while monochromatic radiations of 575–700 nm (yellow-red region) induced positive phototropism with maximum in the red region. Green light (520–575 nm) was ineffective. Negative phototropism-inducing wavelength regions inhibited germ tube growth and positive phototropism-inducing wavelength regions promoted it significantly.Bipolaris oryzae did not show any phototropic response and light did not affect the germ tube growth. These results indicate that the lens effect, in combination with the light growth reaction and light growth inhibition, is the mechanism of the phototropism of germ tubes ofC. lagenarium. NUV radiation, which induced negative phototropism ofC. lagenarium, promoted appressorium formation, while red light, which induced positive phototropism, suppressed it significantly. In the case ofB. oryzae, light did not affect the infection structure formation. These results indicate that negative phototropism of germ tubes ofC. lagenarium favors the infection process by facilitating the contact of the tips of germ tubes with the host surface, while positive phototropism has the opposite effect.  相似文献

Summary A morphological analysis of the initiation and development of root nodules ofElaeagnus angustifolia andMyrica cerifera inoculated with pure-culturedFrankia strains DDB 011610 or DDB 020110 was undertaken. From ultrastructural observations it was determined that both of theseFrankia strains can infectElaeagnus by an intercellular penetration mechanism andMyrica by the root hair infection mechanism. This indicates that both of these strains have the ability to infect host plant roots by either of two mechanisms. The reverse, thatElaeagnus orMyrica could be infected by both mechanisms, was not observed. The infection and nodule development processes of these two plants in combination with these strains were similar to observations made in previous studies (Miller andBaker 1985,Torrey andCallaham 1979). However, one exception was identified in the development of the prenodule ofMyrica when infected with strain 011610, in that endophytic hyphae developed vesicles within the cells of the prenodule. A preparation of bean rust (Uromyces phaseoli) germ tube walls, consisting of short, filamentous particles, was labeled with fluorescein iso-thiocyanate. Freeze sections of host and non-host tissue were incubated in the labeled preparation. Maximum staining was observed in host plant tissue (Phaseolus vulgaris), in which bean rust regularly forms haustoria. In tissue of the non-host plantsVigna sinensis andPhaseolus lunatus, where fewer haustoria were formed, staining was only weak. However, no staining was observed in the non-host tissue ofPhaseolus aureus, Helianthus annuus, Brassica oleracea andHordeum vulgare in which the infection hypha did not form haustoria. This would appear to indicate that formation of haustoria is induced by a specific attachment of the hyphal wall to the host wall. A total of 86 adult bluethroats (Luscinia svecica) from nine different localities, covering the full length of the Fennoscandian mountain range, were screened for blood parasites of the three genera Haemoproteus, Plasmodium and Leucocytozoon using a recently developed polymerase chain reaction method. The overall occurrence of infection was 59.3%. Prevalence of Leucocytozoon spp. (47.7%), Plasmodium spp. (23.3%) and Haemoproteus spp. (1.2%) was detected. Of the infected birds, 15.1% carried mixed infections. Five different mitochondrial DNA-lineages of Leucocytozoon spp., eight lineages of Plasmodium spp. and one lineage of Haemoproteus spp. were found. Due to large sequence divergence these corresponded to at least five different species, but with the possibility of all 14 being independent evolutionary units with the potential of evolving different effects on the host. Of the lineages of Leucocytozoon spp., the most common was found throughout the range. The occurrence of the second most common lineage of Leucocytozoon spp. showed significant variation in prevalence between sites. The data also showed molecular evidence of one lineage of Leucocytozoon sp. existing in more than one species of avian host, thus challenging the use of host taxon as a taxonomic character when distinguishing between different species leucocytozoids.Communicated by F. The structure and musculature of the mouth tube of the lernaeopodidIsobranchia appendiculata Heegaard is described in detail. Situated on the median eminence, the mouth tube appears to be lodged inside a median longitudinal groove on the ventral wall of the cephalothorax. The components of the mouth tube, the labrum and the labium are loosely held together along their lateral edges. The thin, transparent walls of both the labrum and the labium are reinforced by chitinous rods. The musculature of the mouth tube consists of two pairs ofretractores oris, four pairs ofcompressores labri and two pairs oflevatores labii. The mandibles are operated by a pair of antagonistic muscles, theroto adductor mandibulae androto abductor mandibulae. Like other lernaeopodids,I. appendiculata resorts to a browsing type of feeding.  相似文献

Significant stimulation of the number of appressoria, penetration and colonization by conidia ofHelminthosporium carbonum occurred on decolorized maize leaves when exogenous carbohydrates and leaf leachates were added. Germination and germ tube length, however, did not exhibit appreciable differences on decolorized or non-decolorized maize leaves. Lower germination was recorded by leached conidia on decolorized leaves; while appressoria, penetration and colonization were absent. Addition of exogenous nutrients (sucrose>leaf leachates>yeast extract>glucose) enabled conidia to accomplish appressoria, penetration and colonization. Optimum levels for various nutrients observed were 2% (w/v) sucrose/glucose or 0.1% (w/v) yeast extract. Higher concentrations inhibited the infection stages of the pathogen. Depletion of host carbohydrates from green islands/infection sites adversely affect appressoria formation, penetration and colonization; and the loss of carbohydrates from the spore affects germination. Cytokinin-like activity at the infection site/green islands increased with the period of incubation of the host as compared to the surrounding tissue or tissue under water drops. The culture filtrate extracts ofH. carbonum recorded cytokinin-like activity which increased with growth of the fungus. TLC (thin layer chromatography) of cytokinin-like substances (tissue extract and culture filtrate) revealed major activity was confined to Rf zones 0.6 to 0.8 which co-chromatographed with zeatin and zeatin riboside. These substances increase at infection sites by virtue of which carbohydrates accumulate at these sites ensuring a continuous supply to the growing pathogen.  相似文献

The host range specificity ofAgrobacterium with five tea cultivars and an unrelated species (Artemisia parviflora) having extreme surface characteristics was evaluated in the present study. The degree ofAgrobacterium infection in the five cultivars of tea was affected by leaf wetness, micro-morphology and surface chemistry. Wettable leaf surfaces of TV1, Upasi-9 andKangra jat showed higher rate (75%) ofAgrobacterium infection compared to Upasi-10 and ST-449, whereas non-wettable leaves ofA. parviflora showed minimum (25%) infection. This indicated that the leaves with glabrous surface having lower 8 (larger surface area covered by water droplet), higher phenol and wax content were more suitable forAgrobacterium infection. Caffeine fraction of tea promotedAgrobacterium infection even in leaves poor in wax (Upasi-10), whereas caffeine-free wax inhibited bothAgrobacterium growth and infection. Thus, study suggests the importance of leaf surface features in influencing theAgrobacterium infection in tea leaf explants. Our study also provides a basis for the screening of a clone/cultivar of a particular species most suitable forAgrobacterium infection the first step inAgrobacterium-mediated genetic transformation.  相似文献

Zoospores of the dinophytePaulsenella cf.chaetoceratis, parasitizing the marine diatomStreptotheca thamesis, attach to the girdle region of the host and drive a peduncle into the cell interior. The peduncle consists of a non-cytoplasmic crook, a cytoplasmic feeding tube, and a presumably cellulosic sheath around the proximal part of the feeding tube. The crook seems to be used for attachment and penetration of the host. The mobile feeding tube induces shrinkage of the host vacuoles and takes up the complete host cytoplasm within less than 1 h. Phagocytosis depends on an intact host plasmalemma, which is not penetrated by the feeding tube. The trophic phase ends with retraction of the feeding tube. While the food is digested within a large vacuole, the trophont transforms into a thick-walled primary cyst. After about 12 h the primary cyst divides to form 3 or 4 secondary cysts. Finally, about 24 h after attacking the host, each secondary cyst releases two zoospores which may be again ready for infection within 1 h, without passing through any intermediate stage. Summary The behaviour ofRhizoctonia solani on the surface of groundnut seedling hypocotyls was studied at different stages of growth. Characteristic variation in the growth and production on infection cushions was observed. No infection cushions were observed as the seedling reaches the age of three weeks. Hypocotyl exudates were collected at various stages of growth and analysed for total phenols, carbohydrates and ninhydrin positive compounds. The exudates were also analysed for individual amino acids, sugars and organic acids. Gradual decline in the total quantities of various compounds was observed with age. Quantitative and qualitative change in the individual compounds was also evident and some of the compounds were not at all detected as age increases. The possible correlation between the exudation and behaviour of the fungus on host surface is discussed.  相似文献

Summary Motile dinospores ofPaulsenella attach to a host diatom frustule, form a feeding tube, drive it between epi- and hypocingulum, pierce the host plasmalemma and suck up host cytoplasm gradually. This mode of endocytosis (myzocytosis) implies that the host plasmalemma is not ingested and that the host cytoplasm within the food vacuole is bounded only by the vacuolar membrane. The feeding tube is formed by the emergence of a preformed microtubular basket consisting of plates of microtubules. At its entrance into the cell body the feeding tube channel is surrounded by an electron-dense ring. Similar sphincters enclose the two exits through which the two flagella emerge. These sphincters are composed of microfibrils which reveal a cross striation when the fixative does not contain calcium ions. The flagellar bases as well as the internal part of the feeding tube are surrounded by a common cavity which is in open connection also with the ampullae of the pusule. The light and electron microscopical observations do not support the assumption that food uptake is driven by a flow of the membrane of the feeding tube channel caused by an interaction with the microtubular basket (as postulated for food uptake inSuctoria) but rather by an hydrostatic gradient which might be caused by rhythmical ion pumping and be based on the existence of the common cavity and the sphincters. Myzocytosis is inhibited by cytochalasin B.—The fine structure of dinospores and trophonts, especially with respect to the cell covering, the amphiesma, and the en- and excystment, is described.  相似文献

The parasitic dinophyte, Paulsenella sp., is attracted chemotactically by its host, the centric diatom Streptotheca thamesis. Seemingly rather short-lived components of the diatom mucilage are involved in the process. These components are presumably secreted by the Golgi apparatus and pass the thecal slit between epi-and hypocingulum. Experiments with concanavalin A indicate that glucose and-or mannose are constituents of the effective component, which is obviously not produced by egg cells, sperm cells and auxozygotes, since these stages do not attract the parasite. Plasmolysis inhibits secretion of the effective component. The readiness of the parasite to attack a host cell (its appetence) is light dependent: it is low in the dark period and increases after the beginning of the light period in cultures with a 14 h light/10 h dark regime. Endogenous rhythms of the parasite do not seem to be involved. In contrast, the attractiveness of the host is not influenced by the illumination.  相似文献

Sargassum muticum (Phaeophyceae, Fucales) has recently been introduced to Limfjorden (Denmark) where its closest relative is the indigenous Halidrys siliquosa. Previous studies have demonstrated large quantitative (canopy biomass) and qualitative (canopy persistence) differences in the habitat available to epibiota within the canopies of these two macroalgae. We therefore hypothesised that these algae would support different epibiota communities and tested this by sampling the epibiota of S. muticum and H. siliquosa on seven occasions throughout 1997 by enclosing entire thalli in mesh bags. We found 53 epibiota taxa and, with only one exception, they were all recorded on both host species. Species richness and abundance of epibiota exhibited clear seasonal variation on both host species, although epibiota biomass was seasonally constant on H. siliquosa but not on S. muticum. These patterns were consistent with the different life histories of the host species. There was a weakly negative correlation between thallus size and epibiota biomass for both host species. When taking species-specific seasonal variation in thallus size into consideration, S. muticum and H. siliquosa were found to support significantly different epibiota biomasses. Multivariate analyses showed that epibiota community structure was different, although highly overlapping, between the two species, whereas there was an almost parallel temporal development in epibiota community structure. We conclude that it is unlikely that the introduction of S. muticum to Limfjorden has caused major changes in local epibiota community structure. Summary Susceptibility toPhomopsis stalk disease ofPlantago lanceolata genotypes, sampled in three different populations with a variable degree of infection by the fungusPhomopsis subordinaria, was determined under greenhouse conditions. Susceptibility of the host varied within, but not among populations. No relationship between the intensity of the disease in the field and the mean susceptibility of the host genotypes sampled at those locations could be established. Host susceptibility appeared to be composed of the host genotypes sampled at those locations could be established. Host susceptibility appeared to be composed of different (uncorrelated) plant characteristics. Determining whether host genotypes are highly or slightly susceptible can only be achieved by field trials, where the plants are exposed to the whole set of disease inducing factors. The relevance of host susceptibility to the intensity of disease in the field is discussed in relation to the variation in pathogenicity of the fungus and the variation in environmental factors prevailing inP. lanceolata populations underP. subordinaria pathogen pressure. The influence of phosphorus limitedAsterionella on the zoospore production of its fungal parasiteRhizophydium planktonicum was measured, using laboratory cultures of host and parasite. At saturated phosphorus concentrations the host reached a specific growth rate of 0.95.d–1. Growing on these host cells, the mean parasite zoospore production was 26 spores per sporangium, and the mean development time of a sporangium was 45 hours. Growing on phosphorus limited hosts, the zoospore production decreased to less then 9 spores per sporangium, and the development time decreased to 40 hours. On phosphorus limited hosts, zoospores were produced at a slower rate. The algal growth rate was reduced to a greater extent than the fungal growth rate. Therefore, it could be concluded that phosphorus limitation ofAsterionella will facilitate the development of an epidemic of its parasiteRhizophidium, at least at high diatom densities, when possible differences in infectability of the algae play a minor role.  相似文献

Interaction between an entomopoxvirus (PsEPV) and a gregarious braconid endoparasitoid,Cotesia kariyai,inPseudaletia separatalarvae showed that infection of larvae with PsEPV was deleterious to the development and survival ofC. kariyai.The survival and development ofC. kariyaiin PsEPV-infectedP. separatalarvae depended on the length of time between parasitization and viral infection. No parasitoid larvae emerged from PsEPV-infected hosts when host larvae were exposed simultaneously to parasitization and PsEPV inoculation whereas more than 80% of the hosts produced parasitoids when PsEPV was administered 5 days postparasitization.C. kariyailarvae in PsEPV-infected hosts showed a retarded development, shrank, and died about 8 days after viral exposure. Virion-free plasma from PsEPV-infectedP. separatalarvae was toxic to the parasitoid larvae even up to a dilution level of 32 when it was injected intrahemocoelically into the host larvae. Development of parasitoids in hosts that were simultaneously parasitized and injected with the virion-free plasm never progressed beyond the egg stage. The parasitizedP. separatalarvae injected with the virion-free plasma did not pupate and died within 30 days after injection.  相似文献

A resynthesis study was conducted to clarify the relationship between the root endophyte, Heteroconium chaetospira and the ericaceous plant, Rhododendron obtusum var. kaempferi. The host plant roots were recovered 2 months after inoculation, and the infection process and colonization pattern of the fungus were observed under a microscope. The hyphae of H. chaetospira developed structures resembling ericoid mycorrhizas, such as hyphal coils within the host epidermal cells. These structures were morphologically the same as previously reported ericoid mycorrhizal structures. The frequencies of hyphal coils within the epidermal cells of host roots ranged from 13 to 20%. H. chaetospira did not promote or reduce host plant growth. This is the first reported study that H. chaetospira is able to form structures resembling mycorrhizas within the roots of ericaceous plants.  相似文献

Recent work has suggested that the outcomes of host–symbiont interactions can shift between positive, neutral and negative depending on both biotic and abiotic conditions. Even organisms traditionally defined as parasites can have positive effects on hosts under some conditions. For a given host–parasite system, the effects of infection on host fitness can depend on host vigour, route of transmission and environmental conditions. We monitored sublethal microsporidian infections in populations of Gammarus pseudolimnaeus (Amphipoda: Gammaridae) from four cool water streams in southwestern Michigan, USA. Our objectives were to: (i) infer the mechanism of transmission (horizontal, vertical or mixed) from observed effects of infection on host fitness, (ii) determine if the magnitude of the effects on host fitness is a function of parasite load (infection intensity) compared with simple presence or absence of infection, and (iii) determine if there is variation in parasite effects on host fitness in isolated populations. PCR and DNA sequence analyses revealed that there were two microsporidia present among the four host populations: Dictyocoela sp. and Microsporidium sp. PCR screening of a subset of infected hosts showed that Dictyocoela sp. accounted for 90% of infections and was present in all four G. pseudolimnaeus populations, while Microsporidium sp. was found in two populations but was only relatively common in one. We found very low prevalence in males (∼5%), but high prevalence in females (range: 37–85%). Female fitness was positively associated with infection in two streams, resulting from either higher fecundity or more reproductive bouts. Infection had a negative effect on the number of reproductive bouts in a third population, and no effect on fecundity in a fourth population. Infection intensity explained additional variation in fecundity in one population; females with intermediate infection intensity had higher fecundity than females with either light or heavy infection intensity. Given the high prevalence of infection in females compared with males and the generally weak negative fitness effects coupled with some positive fitness effects, it is likely that both Dictyocoela sp. and Microsporidium sp. are primarily vertically transmitted, feminizing microsporidia. Our results suggest that microsporidian effects on G. pseudolimnaeus fitness were context-dependent and varied with host sex and local environment.  相似文献

Laboratory experiments were conducted to examine the effect of ryegrass infection by the endophytic fungusAcremonium loliiLatch, Christensen and Samuels onMicroctonus hyperodaeLoan, a parasitoid ofListronotus bonariensis(Kuschel). Progression of parasitoids through the larval instar stages was shown to depend on adequate nutrition of the weevil host. Compared to confinement on endophyte-free ryegrass, parasitized weevils held on nonpreferred diets comprising leaf segments from endophyte-infected ryegrass and switchgrass contained parasitoid larvae with retarded development. Similarly, development of parasitoid larvae was retarded in hosts feeding on artificial diet containing diterpenes and alkaloids ofA. loliiorigin. Several diterpenes incorporated into the diet reduced survival of the parasitoid larvae. Attack rate of parasitoids was reduced when the quality of potential host weevils was compromised by confinement on nonpreferredA. lolii-infected ryegrass or without food for 14 days.  相似文献

Staphylococcus epidermidis represents the most frequent pathogen involved in nosocomial infections and infections of indwelling medical devices. The strain-to-strain variation of the gene encoding the quorum-sensing pheromone of S. epidermidis as well as the correlation between specificity groups and origin from infection were determined. The pro-pheromone gene was highly conserved and showed infrequent, non-synonymous, single-nucleotide polymorphisms that led to conservative amino acid exchanges only. Importantly, one specificity group was significantly more frequent among strains isolated from infection. The finding that quorum-sensing specificity groups are linked to infection demonstrates the relevance of quorum-sensing for virulence in this critical human pathogen and contributes to the scientific basis needed for the development of quorum-sensing-targeting drugs.  相似文献
