Mature-green tomato fruit (Lycopersicon esculentum Mill.) were treated asymmetrically with 2 millimolar silver thiosulfate (STS) through a cut portion of the peduncle while still attached to the plant. One-half of the fruit received silver and remained green while the other half ripened normally and was silver-free (less than 0.01 parts per billion). Harvested mature-green fruit were also treated with STS through the cut pedicel. Green tissue from silver-treated fruit had levels of 1-aminocyclopropane-1-carboxylic acid (ACC, the immediate ethylene precursor) slightly less or similar to that of turning or red-ripe tissue from the same fruit, and similar to that of mature-green tissue from control fruit. Ethylene production was higher in green tissue from silver-treated fruit than from either red tissue from the same fruit, or mature-green tissue from control fruit. By inhibiting ACC synthesis with aminoethoxyvinyl glycine, and by applying ACC ± silver to excised disks of pericarp tissue from control or silver-treated tomatoes, we showed that short-term silver treatment did not affect the biological conversion of ACC to ethylene, while long-term treatment stimulated both the conversion of ACC to ethylene and the synthesis of ACC.  相似文献

Water relations and growth of tomato fruit pericarp tissue   总被引:2,自引:0,他引:2  
The water relations of young tomato fruit pericarp tissue were examined and related to tissue expansion. The relationship between bulk turgor pressure and tissue expansion (as change in fresh mass or length of tissue) was determined in slices of pericarp cut from young, growing fruit by incubation in different osmotic concentrations of polyethylene glycol 6000 or mannitol. The bulk turgor of this tissue was low (about 0.2 MPa), even in fruit from plants that were otherwise fully turgid, whether measured psychrometrically or by length change in osmotic solutions. The rate of tissue growth at maximum turgor was less than that at moderate turgor unless calcium was added to the incubation medium. However, added calcium also decreased the rate of growth at lower turgor pressures. Yield turgor was < 0.1 MPa, but it was increased by the addition of calcium ions. Electrolyte leakage from tissue was greatest at maximum turgor pressure but was decreased by the addition of calcium ions or osmoticum. Tissue growth was unaffected by a range of plant growth regulators (IAA, abscisic acid, benzyladenine and GA3) but was inhibited, particularly at high turgor, by low concentrations of malic or citric acid. The low turgor pressure of pericarp tissue could be due to the presence of apoplastic solutes within the pericarp, and evidence for this is discussed. Thus, fruit tissue may be able to maintain optimal expansion rates only at moderate turgor and low calcium concentration.  相似文献

Background and AimsIn angiosperms, many species disperse their seeds autonomously by rapid movement of the pericarp. The fruits of these species often have long rod- or long plate-shaped pericarps, which are suitable for ejecting seeds during fruit dehiscence by bending or coiling. However, here we show that fruit with a completely different shape can also rely on pericarp movement to disperse seeds explosively, as in Orixa japonica.MethodsFruit morphology was observed by hard tissue sectioning, scanning electron microscopy and micro-computed tomography, and the seed dispersal process was analysed using a high-speed camera. Comparisons were made of the geometric characteristics of pericarps before and after fruit dehiscence, and the mechanical process of pericarp movement was simulated with the aid of the finite element model.Key ResultsDuring fruit dehydration, the water drop-shaped endocarp of O. japonica with sandwich structure produced two-way bending deformation and cracking, and its width increased more than three-fold before opening. Meanwhile the same shaped exocarp with uniform structure could only produce small passive deformation under relatively large external forces. The endocarp forced the exocarp to open by hygroscopic movement before seed launching, and the exocarp provided the acceleration for seed launching through a reaction force.ConclusionsTwo layers of water drop-shaped pericarp in O. japonica form a structure similar to a slingshot, which launches the seed at high speed during fruit dehiscence. The results suggest that plants with explosive seed dispersal appear to have a wide variety of fruit morphology, and through a combination of different external shapes and internal structures, they are able to move rapidly using many sophisticated mechanisms.  相似文献

Water relations of the tomato during fruit growth   总被引:10,自引:5,他引:5  
Fruit and stem water potentials of tomato plants were measured continuously for several days using automated psychrometers. A linear voltage displacement transducer was used to simultaneously measure diameter changes on an adjacent fruit. A strong correlation was observed between the water potential gradient of the fruit and stem, and changes in fruit diameter. Fruit diameter increased when the apoplasmic water potential gradient favoured solution flow into the fruit and fruit shrinkage occurred only when the water potential gradient was inverted. Based on our data and other published data (Ehret & Ho 1986; Lee 1989a) on phloem transport in tomato, we have concluded that low stem water potentials have an immediate and direct effect on phloem turgor; reducing the driving force for sap flow into the fruit. Since fruit water potential remained relatively constant, the diurnal variation in stem water potential was sufficient to account for the correlation with changes in fruit diameter. There are consequences with respect to predicting the accumulation of dry matter in tomato fruit.  相似文献

A method of measuring the water potential of stored potato tubers (Solanum tuberosum L.) was needed to investigate the relationship of bacterial soft rot in tubers to water potential. Pressure chamber measurements, while useful for tubers with functional stolons, cannot be made on stored tubers. Measurements could be made on excised tissue pieces in a hygrometer chamber and with hygrometers implanted into tubers. We report here our evaluation of these hygrometric methods using a comparison with the pressure chamber on tubers harvested with stolons intact.

In tubers of high water potential, measurements on excised tissue were as much as 0.5 megapascals lower than the pressure chamber, probably due to turgor-driven expansion of the sample when freed from constraints imposed by surrounding tissue. Good agreement (±0.05 megapascals) was found between the implanted hygrometer and the pressure chamber at potentials higher than −0.5 megapascals. At lower water potentials, both hygrometer measurements were higher than the pressure chamber. Respirational heating of the tissue contributed to the increase in the excised tissue samples, but not with the implanted hygrometers because of the hygrometer design. The osmotic pressure balanced the pressure chamber measurement of potential at −0.7 megapascals, but was too small to do so at lower potentials. At most, 25% of this discrepancy can be accounted for by dilution by apoplastic water. We believe that the pressure chamber measurement is too low at low water potentials and that the error is associated with air bubbles in the xylem. At low potentials air emerged from xylem vessels along with sap, and fewer xylem emitted sap as potentials decreased.

Prior temperature exposure affects subsequent chilling sensitivity   总被引:5,自引:0,他引:5  
The chilling sensitivity of small discs or segments of tissue excised from chillingsensitive species was significantly altered by prior temperature exposure subsequent to holding the tissue at chilling temperatures as measured by a number of physiological processes sensitive to chilling. This temperature conditioning was reversible by an additional temperature exposure before chilling, and mature-green and red-ripe tomato tissue exhibit similar chilling sensitivities. Exposing pericarp discs excised from tomato fruit (Lycopersicon esculentum Mill. cv. Castelmart), a chilling-sensitive species, to temperatures from 0 to 37°C for 6 h before chilling the discs at 2.5°C for 4 days significantly altered the rate of ion leakage from the discs, but had no effect on the rate of ion leakage before chilling and only a minimal effect on discs held at a non-chilling temperature of 12°C. Exposing chillingsensitive tissue to temperatures below that required to induce heat-shock proteins but above 20°C significantly increased chilling sensitivity as compared to tissue exposed to temperatures between 10 and 20°C. Rates of ion leakage after 4 days of chilling at 2.5°C were higher from fruit and vegetative tissue of chilling-sensitive species (Cucumis sativus L. cv. Poinsett 76, and Cucurbita pepo L. cv. Young Beauty) that were previously exposed for 6 h to 32°C than from similar tissue exposed to 12°C. Exposure to 32 and 12°C had no effect on the rate of ion leakage from fruit tissue of chilling tolerant species (Malus domestica Borkh. cv. Golden Delicious, Pyrus communis L. cv. Bartlett). Ethylene and CO2 production were higher and lycopene synthesis was lower in chilled tomato pericarp discs that were previously exposed for 6 h to 32°C than the values from tissue exposed to 12°C for 6 h before chilling. Increased chilling sensitivity induced by a 6 h exposure to 32°C could be reversed by subsequent exposure to 12°C for 6 h.  相似文献

利用热偶湿度计(thermocouplepsychrometer)研究了野生型、GA-缺陷型和ABA-缺陷型番茄发育过程中果实种子的水分关系,发现除ABA-缺陷型种子胶囊和果肉水势变化特殊外,3种类型果实水分状况变化基本一致;在整个发育时期内.前期种子胶囊和果肉水分流向种子,中期种子水分流向种子胶囊和果肉,后期种子和果实间的水势达到平衡。鉴于种胚脱水是一种主动过程,种胚水势一直低于整个种子、种子胶囊和果肉。内源赤霉素可明显增加果实和种子的重量,但对增加种胚溶质的作用不大。由于内源脱落酸可以促使果实成熟和衰老,促进果实细胞解体,大大降低种子胶囊和果肉水势,因而抑制成熟种子在果实内萌发。  相似文献

Understanding how the fruit microclimate affects ascorbate (AsA) biosynthesis, oxidation and recycling is a great challenge in improving fruit nutritional quality. For this purpose, tomatoes at breaker stage were harvested and placed in controlled environment conditions at different temperatures (12, 17, 23, 27 and 31°C) and irradiance regimes (darkness or 150 µmol m-2 s-1). Fruit pericarp tissue was used to assay ascorbate, glutathione, enzymes related to oxidative stress and the AsA/glutathione cycle and follow the expression of genes coding for 5 enzymes of the AsA biosynthesis pathway (GME, VTC2, GPP, L-GalDH, GLDH). The AsA pool size in pericarp tissue was significantly higher under light at temperatures below 27°C. In addition, light promoted glutathione accumulation at low and high temperatures. At 12°C, increased AsA content was correlated with the enhanced expression of all genes of the biosynthesis pathway studied, combined with higher DHAR and MDHAR activities and increased enzymatic activities related to oxidative stress (CAT and APX). In contrast, at 31°C, MDHAR and GR activities were significantly reduced under light indicating that enzymes of the AsA/glutathione cycle may limit AsA recycling and pool size in fruit pericarp, despite enhanced expression of genes coding for AsA biosynthesis enzymes. In conclusion, this study confirms the important role of fruit microclimate in the regulation of fruit pericarp AsA content, as under oxidative conditions (12°C, light) total fruit pericarp AsA content increased up to 71%. Moreover, it reveals that light and temperature interact to regulate both AsA biosynthesis gene expression in tomato fruits and AsA oxidation and recycling.  相似文献

Regulation of tomato fruit growth by epidermal cell wall enzymes   总被引:12,自引:0,他引:12  
Water relations of tomato fruit and the epidermal and pericarp activities of the putative cell wall loosening and tightening enzymes Xyloglucan endotransglycosylase (XET) and peroxidase were investigated, to determine whether tomato fruit growth is principally regulated in the epidermis or pericarp. Analysis of the fruit water relations and observation of the pattern of expansion of tomato fruit slices in vitro , has shown that the pericarp exerts tissue pressure on the epidermis in tomato fruit, suggesting that the rate of growth of tomato fruit is determined by the physical properties of the epidermal cell walls. The epidermal activities of XET and peroxidase were assayed throughout fruit development. Temporal changes in these enzyme activities were found to correspond well with putative cell wall loosening and stiffening during fruit development. XET activity was found to be proportional to the relative expansion rate of the fruit until growth ceased, and a peroxidase activity weakly bound to the epidermal cell wall appeared shortly before cessation of fruit expansion. No equivalent peroxidase activity was detected in pericarp tissue of any age. It is therefore plausible that the expansion of tomato fruit is regulated by the combined action of these enzyme activities in the fruit epidermis.  相似文献

Ethylene production, 1-aminocyclopropane-1-carboxylic acid (ACC) levels and ACC-synthase activity were compared in intact and wounded tomato fruits (Lycopersicon esculentum Mill.) at different ripening stages. Freshly cut and wounded pericarp discs produced relatively little ethylene and had low levels of ACC and of ACC-synthase activity. The rate of ethylene synthesis, the level of ACC and the activity of ACC synthase all increased manyfold within 2 h after wounding. The rate of wound-ethylene formation and the activity of wound-induced ACC synthase were positively correlated with the rate of ethylene production in the intact fruit. When pericarp discs were incubated overnight, wound ethylene synthesis subsided, but the activity of ACC synthase remained high, and ACC accumulated, especially in discs from ripe fruits. In freshly harvested tomato fruits, the level of ACC and the activity of ACC synthase were higher in the inside parts of the fruit than in the pericarp. When wounded pericarp tissue of green tomato fruits was treated with cycloheximide, the activity of ACC synthase declined with an apparent half life of 30–40 in. Gradients in water potential form the driving force for the movement of water for cell enlargement. In stems, they are oriented radially around the vascular system but should also be present along the stem. To test this possibility, growth, water potential, osmotic potential, and turgor were determined at intervals along the length of dark-grown soybean (Glycine max L. Merr., cv. Wayne) hypocotyls. Transpiration was negligible in the dark, humid conditions, so that all water uptake was for growth. Elongation occurred in the terminal 1.5 centimeters of the hypocotyl. Water potential was −3.5 bars in the elongating region but −0.5 bar in the mature region, both in intact plants and detached tissue. There was a gradual transition between these values that was related to the growth profile along the hypocotyl. Tissue osmotic potentials generally paralleled tissue water potentials, so that turgor was the same throughout the length of the hypocotyl. If the elongating zone was excised, growth ceased immediately. If the elongating zone was excised along with mature tissue, however, growth continued, which confirmed the presence of a water-potential gradient that caused longitudinal water movement from the mature zone to the elongating zone. When the plants were grown in vermiculite having low water potentials, tissue water potentials and osmotic potentials both decreased, so that water potential gradients and turgor remained undiminished. It is concluded that growth-induced water potentials reflect the local activity for cell enlargement and are supported by appropriate osmotic potentials.  相似文献

比较了福建省 1 0个主栽龙眼品种果实的果皮形态和结构 ,结果表明 :不同品种在果皮厚度、外果皮表面颜色、龟状纹、放射线、瘤状突、刺毛、外果皮皮孔、周皮层厚度、栓质层厚度和连续性、中果皮薄壁组织细胞排列、石细胞大小、含量、排列和分布 ,维管束发达状况、排列和分布 ,内果皮表皮细胞排列和角蜡质层厚度等方面均存在着明显差异。风梨味、东壁、油潭本、乌龙岭、红核子、蕉眼龙眼果皮厚 ,外果皮表面瘤状突和剌毛多 ,外果皮周皮层、栓质层厚且连续性好 ,中果皮石细胞 (团 )含量多且排列紧密 ,分布在中果皮外侧且在中果皮中所占比例大 ,维管束发达且排列有序 ,内果皮角蜡质层厚 ;这些品种果实耐贮运、抗病性强。而水涨、赤壳、福眼、普明庵龙眼果皮薄 ,外果皮周皮层薄、栓质层不发达 ,中果皮石细胞 (团 )含量少、分布分散 ,维管束不发达 ,薄壁组织细胞胞间隙大 ,皮孔间隙大、皮孔通道与中果皮组织细胞间隙相通 ;这些品种的果实不耐贮运、抗病性弱。讨论了龙眼外果皮表面主色为褐色和内果皮比外果皮更容易褐变的解剖学原因及龙眼果皮形态结构与果实耐贮运的关系。  相似文献

Paraheliotropic (light-avoiding) leaf movements have been associated with high light intensity, high temperature, and drought. We investigated leaf elevation for intact plants, pulvinus bending for excised motor organs, and size change for protoplasts from motor tissue for two Phaseolus species: Phaseolus acutifolius A. Gray, native to hot, arid regions, and Phaseolus vulgaris L., the common bean. Leaf angles above horizontal were measured for central trifoliolate leaflets of intact plants at 24, 27, and 30[deg]C at 500 and 750 [mu]mol photons (400-700 nm) m-2 s-1 over a range of water potentials; equivalent angles were determined for excised motor organs under similar conditions. Diameters were measured for protoplasts from abaxial and adaxial motor tissue over a range of photon flux density values, temperatures, and water potentials. In general, higher photon flux density and temperature resulted in elevation of leaves, bending of excised pulvini, and equivalent changes in protoplast volume (swelling of abaxial protoplasts and shrinking of adaxial protoplasts). In intact plants, lower water potentials yielded greater paraheliotropism; abaxial protoplasts increased in size, whereas adaxial ones did not change. P. acutifolius typically exhibited greater paraheliotropism than did P. vulgaris under the same conditions, a set of physiological responses likely to be highly adaptive in its native arid habitat.  相似文献

Hardness as a basis of fruit choice in two sympatric primates   总被引:5,自引:0,他引:5  
Fruit color and size are significant determinants of food choice in mammals and birds, but hardness, an important physical property of fruit and seeds, has generally been overlooked as a determinant of food choice in studies of mammalian foraging behavior. Two methods were used to determine fruit hardness during a field study of two sympatric primates, the black spider monkey (Ateles paniscus) and the bearded saki monkey (Chiropotes satanas) in Surinam. We measured both puncture resistance of fruit pericarp and crushing resistance of seeds. Puncture resistance of the pericarp of some fruit opened by Chiropotes was as much as 15 times greater than that of all fruit successfully opened by Ateles. In contrast, crushing resistance of species of seeds masticated by Chiropotes was significantly lower than that of seeds swallowed by Ateles. These data demonstrate that hardness of both fruit pericarp and seed may play a significant role in food choice among sympatric vertebrates. Measurements of both puncture resistance of the pericarp and crushing resistance of the seed are necessary for understanding the significance of fruit hardness in these primates.  相似文献

Summary 3H-glucose was fed to excised Sultana grape berries via their pedicels for up to 5 hours. Autoradigraphy showed that the label was distributed throughout the fruit within 1 hour. Microautoradiography of tissue sections taken at a number of points showed that within the pedicel the walls of cortical cells had become heavily labelled, suggesting that the cortical cell walls offered a diffusion pathway for the solutes entering the vascular system from the external aqueous solution. Transport along the pedicel was confined to the central vascular tissue with little radioactivity occurring in the cortical cells. Within the pericarp, the vascular bundles and walls of nearby parenchyma cells had become heavily labelled, indicating that the labelled solute was present within the vicinity of cell walls. The general pattern of 3H-glucose accumulation by excised berries was similar to the deposition pattern of 24C-labelled photosynthate within attached fruit.  相似文献

Pod morphology and anatomy have been studied in 28 species and four varieties of Indian Indigofera. Pods of Indigofera spp. differ with respect to size, the relative thickness of the fruit wall, the number of sclerenchymatous layers in the endocarp, the presence/absence of hypodermis and trichomes, and the presence of separation tissue. Anatomically, the pericarp is broadly characterized into three types: type I (thin pericarp and three to five sclerenchymatous layers in the endocarp), type II (intermediary pericarp thickness and six to eight sclerenchymatous layers), and type III (thick pericarp and more than eight sclerenchymatous layers). The distribution of these types across the tribe is not congruent with the current phylogenetic analyses. Type III pericarp (present in the early diverging lineages of the tribe) represents the most primitive state, whereas type I and type II pericarps are derived. Fruits of Indigofera generally show normal explosive dehiscence as a means of dispersal of seeds, although some species show adaptations for dispersal by wind. In Indigofera, dehiscence is caused by a separation layer present at the dorsal and ventral sutures except in some species (Indigofera hochstetteri, Indigofera karnatakana, Indigofera glandulosa var. sykesii, and Indigofera trita var. scabra) in which no separation tissue is present; these species show delayed dehiscence or an indehiscent condition. The indehiscent pod type is considered to be apomorphic. Air volume measurement of 'Braeburn' apple fruit   总被引:2,自引:0,他引:2  
The radial disposition of air in the flesh of fruit of Malus domestica Borkh., cv 'Braeburn' was investigated using a gravimetric technique based on Archimedes' principle. Intercellular air volume was measured by weighing a small tissue sample under water before and after vacuum infiltration to remove the air. In a separate procedure, the volume of the same sample was measured by recording the buoyant upthrust experienced by it when fully immersed in water. The method underestimates tissue air volume due to a slight invasion of the intercellular air spaces around the edges of the sample when it is immersed in water. To correct for this error, an adjustment factor was made based upon an analysis of a series of measurements of air volume in samples of different dimensions. In 'Braeburn' there is a gradient of declining air content from just beneath the skin to the centre of the fruit with a sharp discontinuity at the core line. Cell shape and cell packing were observed in the surface layers of freshly excised and stained flesh samples using a dissecting microscope coupled to a video camera and a PC running proprietary software. Tissue organization changed with distance below the skin. It is speculated that reduced internal gas movement, due to the tightly packed tissue of 'Braeburn' and to the potential diffusion barrier at the core line between the cortex and the pith, may increase susceptibility of the flesh to disorders associated with tissue browning and breakdown.  相似文献

During caryopsis development the layers of the pericarp, integuments, and nucellus all contribute to the formation of the caryopsis coat. The coat consists of a layer of outer pericarp epidermal transfer cells, a collapsed and senescent layer of middle pericarp cells, and a discontinuous layer of inner pericarp epidermal transfer cells. The latter is not present across the placental pad. The integuments are present as a collapsed dense layer, the nucellus is discontinuous and cellular. The placental pad occurs at the ventral surface of the caryopsis, opposite the scutellum and coleorhiza. It consists of 15–20 collapsed cell layers, including the pigment strand and placental vascular bundle. From the inside several partially collapsed cell layers of the nucellar projection occur which contain transfer-cell walls. The middle dense layers, the pigment strand, consist of the middle pericarp remnant, plus the remains of the placental vascular bundle. The pericarp inner epidermis does not extend across the pad. The aleurone layer is a continuous uniseriate layer around the entire caryopsis except at the placental pad; here it is crushed and contains the remnant of a transfer-cell wall. The outer pericarp epidermis is a continuous layer of transfer cells across the pad. These cells contain membranous inclusions suggesting that they may be functional during germination.  相似文献

The larvae of Dacus (Leptoxyda) persicus (aak fruit fly) are key predispersal seed predators in Calotropis procera (Asclepiadaceae). Based on fruit characteristics, two morphs are distinguishable in C. procera viz., the soft-fruited morph (SF morph) and the hard-fruited morph (HF morph). The work reported here examined whether the fruit characteristics influenced the infestation by the aak fruit fly and, if so, what mechanism(s) were operative. Fruits in the SF morph were significantly more acceptable to the aak fruit fly than those of the HF morph irrespective of their size class and availability or fly population density. A general ranking of fruit acceptability for oviposition by the aak fruit fly within the fruit size class was: size class III ≥ size class II > size class I and IV. The negative relationship between fruit infestation and pericarp toughness, which is suggestive of trade-offs between the fly’s oviposition obligation and energy/time (predation risk) constraint, was found to correlate with the requirement of greater force to puncture the pericarp in the hard fruits. Lower penetrability of the pericarp in the hard fruits appeared to be primarily due to the thickness of pericarp and secondarily on account of the thickened walls of endocarpic–mesocarpic cells in the inner pericarpic layer. The present data point to the existence of two fruit morphs in C. procera differing in the acceptability of fruits for oviposition by the aak fruit fly primarily on account of toughness and internal structure of the pericarp.  相似文献

Rose achene imbibition is difficult. Comparison of sulphuric acid treated, Cross and Beavanś reagent (C. B. reagent) treated seeds with untreated seeds showed that, during imbibition, water is essentially found in the most external pericarp layer. Inner layers appear less permeable. It is possible to distinguish three layers in the pericarp by taking into account results from imbibition assays, X-ray radiographies and scanning electron microscopic observations. They can be interpreted as representing the epi-, meso- and endocarp.  相似文献
