Mucopolysaccharidosis IIIB (MPS IIIB) is a lysosomal storage disorder characterized by severe behavioural disturbances and progressive loss of cognitive and motor function. There is no effective treatment, but behavioural testing is a valuable tool to assess neurodegeneration and the effect of novel therapies in mouse models of disease. Several groups have evaluated behaviour in this model, but the data are inconsistent, often conflicting with patient natural history. We hypothesize that this discrepancy could be due to differences in open field habituation and home cage behaviour. Eight-month-old wild-type and MPS IIIB mice were tested in a 1-h open field test, performed 1.5 h after lights on, and a 24-h home cage behaviour test performed after 24 h of acclimatization. In the 1-h test, MPS IIIB mice were hyperactive, with increased rapid exploratory behaviour and reduced immobility time. No differences in anxiety were seen. Over the course of the test, differences became more pronounced with maximal effects at 1 h. The 24-hour home cage test was less reliable. There was evidence of increased hyperactivity in MPS IIIB mice, however, immobility was also increased, suggesting a level of inconsistency in this test. Performance of open field analysis within 1-2 h after lights on is probably critical to achieving maximal success as MPS IIIB mice have a peak in activity around this time. The open field test effectively identifies hyperactive behaviour in MPS IIIB mice and is a significant tool for evaluating effects of therapy on neurodegeneration.  相似文献

In three separate experiments groups of rats were chronically administered neuroleptics in a variety of ways (chronic injections, subcutaneous implants, and decanoate injections) and examined for oral movements (OMs) in two different tests: in an open cage using a human observer, or in a plexiglas tube enclosure, where OMs were monitored both by a human observer and computerized video analysis system. These two testing methods showed different effects of neuroleptic administration. In the open cage, OMs tended to be enhanced during chronic neuroleptic exposure and to rapidly subside upon drug withdrawal. The enhanced OMs were especially present just after drug injections, when activity levels were low. In the observation tube environment, however, OMs tended to be low soon after drug treatments, and elevated upon withdrawal. Thus, the type of behavioral test used determines how neuroleptic-induced increases in oral activity should be interpreted.  相似文献

The goal of this work was to establish the effects of irradiation of the head with gamma-rays on selected forms of innate behavior, as well as on the habituation process to new environment in the open field test in rats. During a 5-day control period behavioral parameters reflecting motoric and explorative activities, as well as anxiety (6 parameters in total) were followed daily in 20 male Sprague-Dawley rats in an open field. Fourteen animals were repeatedly tested after irradiation of the head with a single dose of 10 Gy of gamma-rays. The results showed statistically significant depression of motoric and explorative activities during the first 3 days after irradiation in comparison with the control period. After irradiation the level of anxiety was elevated. Habituation to a new environment was observed in non-irradiated, but not in irradiated animals. These results suggest that the applied dose of ionizing radiation influenced the brain centers involved in control of innate behavioral functions connected with response to a new environment in rats.  相似文献

The intermediate filament protein nestin is expressed by neural stem cells, but also by some astrocytes in the neurogenic niche of the hippocampus in the adult rodent brain. We recently reported that nestin-deficient (Nes?/?) mice showed increased adult hippocampal neurogenesis, reduced Notch signaling from Nes?/? astrocytes to the neural stem cells, and impaired long-term memory. Here we assessed learning and memory of Nes?/? mice in a home cage set up using the IntelliCage system, in which the mice learn in which cage corner a nose poke earns access to drinking water. Nes?/? and wildtype mice showed comparable place learning assessed as the incorrect corner visit ratio and the incorrect nose poke ratio. However, during reversal place learning, a more challenging task, Nes?/? mice, compared to wildtype mice, showed improved learning over time demonstrated by the incorrect visit ratio and improved memory extinction over time assessed as nose pokes per visit to the previous drinking corner. In addition, Nes?/? mice showed increased explorative activity as judged by the increased total numbers of corner visits and nose pokes. We conclude that Nes?/? mice exhibit improved reversal place learning and memory extinction, a finding which together with the previous results supports the concept of the dual role of hippocampal neurogenesis in cognitive functions.

The capacity of bank voles (Clethrionomys glareolus Schreb) for rapid spatial learning was assessed in cue-controlled open field using their species-specific habits (high fear). Animals were tested in open field after 3 days of pretraining. During testing, novel stimuli (black-and-white geometric figures arranged in symmetric or asymmetric modes for different vole groups) were attached to a curtain surrounding the open field, and a shelter (a familiar box from the home cage) was introduced into the field. Testing consisted of four 10-min trials, the last (probe) trial was performed without the shelter. Time spent in different fielld areas was recorded. The voles remembered previous location of the shelter and preferred to visit this area. It was possible only in the condition with asymmetric arrangement of visual stimuli when animals could use them for navigation. The findings conclude that bank vole have a good capacity for rapid spatial learning.  相似文献

Corticotropin-releasing factor was administered into the lateral cerebral ventricles of rats. Sixty minutes later, animals were tested in an open field conflict test or in their home cages for a variety of behaviors which have been shown to be related to the degree of responsiveness to novelty. CRF, in a dose related fashion, altered the frequency of those behaviors which are normally expressed in response to the novel environment. Specifically, CRF caused an increase in grooming and decreased in the amount of rearing, the number of approaches to a food pellet placed in the center of the open field, the amount of food eaten in both the open field and the home cage and a decrease in the mean amount of food eaten per approach to the food pedestal.  相似文献

The between‐laboratory effects on behavioral phenotypes and spatial learning performance of three strains of laboratory mice known for divergent behavioral phenotypes were evaluated in a fully balanced and synchronized study using a completely automated behavioral phenotyping device (IntelliCage). Activity pattern and spatial conditioning performance differed consistently between strains, i.e. exhibited no interaction with the between‐laboratory factor, whereas the gross laboratory effect showed up significantly in the majority of measures. It is argued that overall differences between laboratories may not realistically be preventable, as subtle differences in animal housing and treatment will not be controllable, in practice. However, consistency of strain (or treatment) effects appears to be far more important in behavioral and brain sciences than the absolute overall level of such measures. In this respect, basic behavioral and learning measures proved to be highly consistent in the IntelliCage, therefore providing a valid basis for meaningful research hypothesis testing. Also, potential heterogeneity of behavioral status because of environmental and social enrichment has no detectable negative effect on the consistency of strain effects. We suggest that the absence of human interference during behavioral testing is the most prominent advantage of the IntelliCage and suspect that this is likely responsible for the between‐laboratory consistency of findings, although we are aware that this ultimately needs direct testing.  相似文献

Effects of intracerebroventricular administration of neuropeptide Y on open field behaviour, behavioural habituation and corticosterone response to open field testing, and on home cage activity have been investigated in the rat. In the open field, NPY reduced activity in a dose-dependent manner. Behavioural habituation was not influenced. After 5 days of recovery, NPY-treated animals did not differ from non-treated in any of the measured parameters. Peripheral corticosterone levels were not significantly affected, although there was a strong tendency towards an increase. Injection of 2 nmol NPY did not produce any gross neurological deficits. At this dose, NPY greatly suppressed home cage activity. The effect lasted throughout the recording period of 22 h, abolishing the normal circadian variation in activity. After 5 days of recovery, the effect was no longer present. Our interpretation of these findings is, that NPY is a highly potent endogenous agent capable of producing certain important aspects of behavioural sedation in a reversible manner. Since NPY did not decrease the corticosterone response to a novel stimulus, its pattern of actions seems to differ from synthetic sedative drugs.  相似文献

Late radiation effects were investigated in the mouse small intestine after a daily fractionated radiation treatment. Mice were given 14 X 3 Gy in 2 weeks over a partial abdominal irradiation field. There was evidence for late injury in the intestinal epithelium, the submucosa, and the subserosa. Late damage in the epithelium was shown histologically by a reduced crypt number and villus atrophy at 3 and 6 months but not at 24 h after the end of treatment. The reduction in crypt number was significant in the ileum at 3 and 6 months after irradiation: 100 +/- 4 and 98 +/- 5 (SEM) per circumference, respectively, versus 132 +/- 3 and 146 +/- 6 in age-matched controls (P less than 0.01, t test). The mitotic activity in the crypts of the irradiated animals was significantly increased at all investigated times, suggesting a prolonged but insufficient compensatory response to maintain the mucosal integrity. The repercussion on intestinal epithelial function was, at least in part, reflected by a progressively reduced body weight gain up to 5 g at 3 months after treatment. The ability of the surviving crypt stem cells to form microcolonies after irradiation, however, was not impaired. Evidence for injury in the submucosa was provided from macroscopic and histological examination. Macroscopically, at 6 months after treatment, narrowed and rigid bowel segments surrounded by fibrotic adhesions were observed, causing partial intestinal obstruction. In addition, sometimes focal areas of hemorrhage and infarction in small bowel segments were present. Histologically, diffuse and pronounced submucosal edema without increased fibrosis was seen, together with markedly dilated small blood vessels in focal areas of macroscopic intestinal infarction. The intestinal perfusion, as assessed by 86Rb extraction, was significantly but transiently reduced at 3 months after irradiation. These data suggest mainly late effects in the small intestine after this daily fractionated irradiation treatment. The reduced number of epithelial cells and the submucosal edema are possibly mediated by radiation injury in the intestinal microvasculature.  相似文献

Cyclic AMP and cyclic GMP levels were measured in the anterior and posterior pituitary, hypothalamus, pineal and cerebellum of female rats sacrificed during proestrus, metestrus and diestrus. In the first experiment rats were sacrificed by microwave irradiation between 0900 and 1100, between 1600 and 1800 and between 2100 and 2300. Cyclic AMP and cyclic GMP levels did not vary in any region tested as a function of the estrous cycle except for slightly elevated cyclic GMP levels in the posterior pituitary during proestrus. However the time of day at which the animals were sacrificed affected levels of cyclic AMP in the hypothalamus and cerebellum and levels of cyclic GMP in the cerebellum. In a second experiment female rats were all sacrificed between 2130 and 2330 during proestrus and diestrus. In this experiment rats were sacrificed either immediately upon removal from the home cage or after 10 min of immobilization stress, or after 10 min of open field activity. No differences in pituitary cyclic nucleotides were seen between proestrous and diestrous animals. However, stressed animals showed large cyclic AMP increases in the pituitary, and activity increased cyclic GMP levels in the cerebellum and pineal.  相似文献

In this paper I examine exploratory behavior and learning in two groups of common marmosets (Callithrix jacchus jacchus), both living under laboratory conditions. The main aim of my studies was to compare behavior occurring in an experimental testing situation with home-cage behavior, and to assess conditions under which exploration is enhanced. One group of marmosets was tested individually in an experimental room away from a larger animal holding room, and a second group was observed in their own home cages, surrounded by other marmosets. Marmosets in both groups were given the opportunity to learn simple tasks, such as pressing a lever for food or jumping on to a wire grid to open a door and see a neighbouring marmoset, and were provided with sets of novel or familiar objects to encourage play and exploration. In the isolated experimental cage, marmosets which learnt a lever-pressing task showed higher general activity levels than controls; in the home cages the mere addition of novel objects resulted in enhancements in exploratory behavior (measured in terms of contacts with the objects). Although the activities associated with learning tended to decline over time, the novel objects remained a constant interest throughout the experiment. The results have implications for maintaining marmosets in captivity, especially in situations when they are housed separately.  相似文献

Chronic stress is known to enhance mammals’ emotional reactivity and alters several of their cognitive functions, especially spatial learning. Few studies have investigated such effects in birds. We investigated the impact of a two-week stress on Japanese quail’s emotional reactivity and spatial learning. Quail is an avian model widely used in laboratory studies and for extrapolation of data to other poultry species. As sensitivity to chronic stress can be modulated by intrinsic factors, we tested juvenile female Japanese quail from three lines, two of them divergently selected on tonic immobility duration, an indicator of general fearfulness. The different emotional reactivity levels of quail belonging to these lines can be revealed by a large variety of tests. Half of the birds were submitted to repeated unpredictable aversive events for two weeks, whereas the other half were left undisturbed. After this procedure, two tests (open field and emergence tests) evaluated the emotional reactivity of treated and control quails. They were then trained in a T-maze for seven days and their spatial learning was tested. The chronic stress protocol had an impact on resting, preening and foraging in the home cage. As predicted, the emotional reactivity of treated quails, especially those selected for long tonic immobility duration, was higher. Our spatial learning data showed that the treatment enhanced acquisition but not memorization. However, intrinsic fearfulness did not seem to interact with the treatment in this test. According to an inverted U-shaped relationship between stress and cognition, chronic stress can improve the adaptability of birds to a stressful environment. We discussed the mechanisms possibly implied in the increase of emotional reactivity and spatial abilities.  相似文献

We examined the effects of radiation on decreases in osteoclast numbers after regional irradiation of rats with carbon ions and gamma rays. Male Wistar rats were subjected to hind-leg irradiation with carbon ions (290 MeV/u) or gamma rays at doses of 15, 22.5, or 30 Gy. The effects of carbon ions and gamma rays on osteoclasts were studied using histologic and morphometric methods. At doses of 15 Gy and 22.5 Gy, osteoclast numbers increased transiently until day 5 after irradiation and then decreased rapidly in both the carbon ion and gamma ray irradiation groups. The carbon ion group showed reduced osteoclast size compared with the gamma ray group. Carbon ion irradiation had a more marked effect on osteoclast activity, and suppressed maturation to a greater extent than gamma irradiation. These observations suggest that carbon ion irradiation induces differential modulation of osteoclast growth factor expression.  相似文献

Monoamine oxidase (MAO) plays an important role in the metabolism of neuro-transmitter biogenic amines. Its activity was determined in mouse brain and liver after exposure to different kinds of ionizing radiation and after pretreatment with a radioprotective agent. After a lethal dose of mixed neutron-gamma irradiation the MAO activity decreased in the brain and increased in the liver. In contrast, after a lethal dose of 60Co-gamma irradiation enzyme activity was considerably increased in the brain while in the liver it increased like after mixed neutron-gamma irradiation. AET (S2-aminoethyl-isothiuronium-Br X HBr), when administered in a radio-protective dose, inhibited MAO activity in the brain, while it increased in the liver. Even more marked changes of enzyme activity were observed in both brain and liver after AET pretreatment and mixed neutron-gamma irradiation. On the basis of the results it is suggested that different kinds of ionizing radiation lead to different types of lipid peroxidation in the lipid environment surrounding MAO, an event leading to altered enzyme activity. AET itself inhibited MAO in the brain and increased the activity in the liver but did not prevent the alterations caused by ionizing radiation in enzyme activity.  相似文献

Many studies using genetic mouse models are performed with animals on either one of the two closely related genetic backgrounds, C57BL/6J or C57BL/6N. These strains differ only in a few genetic loci, but have some phenotypic differences that also affect behavior. In order to determine the effects of chronic stress hormone exposure, which is relevant for the pathogenesis of psychiatric disorders, we investigated here the behavioral manifestations of long‐term increase in corticosterone levels. Thus, male mice from both sub‐strains were subcutaneously implanted with corticosterone (20 mg) or placebo pellets that released the hormone for a period of 21 days and resulted in significantly elevated plasma corticosterone levels. Corticosterone significantly increased food intake in B6N, but not in B6J mice. At various time points after pellet implantation, we performed tests relevant to activity and emotional behaviors. B6J mice displayed a generally higher activity in the home cage and the open field. Corticosterone decreased the activity. In B6N mice, corticosterone also decreased sucrose preference, worsened the coat state and increased forced swim immobility, while it had no effect in the B6J strain. Altogether, these results indicate that B6N mice are more sensitive to some of the effects of chronic corticosterone treatment than B6J mice.  相似文献

1. Although the home range is a fundamental ecological concept, there is considerable debate over how it is best measured. There is a substantial literature concerning the precision and accuracy of all commonly used home range estimation methods; however, there has been considerably less work concerning how estimates vary with sampling regime, and how this affects statistical inferences. 2. We propose a new procedure, based on a variance components analysis using generalized mixed effects models to examine how estimates vary with sampling regime. 3. To demonstrate the method we analyse data from one study of 32 individually marked roe deer and another study of 21 individually marked kestrels. We subsampled these data to simulate increasingly less intense sampling regimes, and compared the performance of two kernel density estimation (KDE) methods, of the minimum convex polygon (MCP) and of the bivariate ellipse methods. 4. Variation between individuals and study areas contributed most to the total variance in home range size. Contrary to recent concerns over reliability, both KDE methods were remarkably efficient, robust and unbiased: 10 fixes per month, if collected over a standardized number of days, were sufficient for accurate estimates of home range size. However, the commonly used 95% isopleth should be avoided; we recommend using isopleths between 90 and 50%. 5. Using the same number of fixes does not guarantee unbiased home range estimates: statistical inferences differ with the number of days sampled, even if using KDE methods. 6. The MCP method was highly inefficient and results were subject to considerable and unpredictable biases. The bivariate ellipse was not the most reliable method at low sample sizes. 7. We conclude that effort should be directed at marking more individuals monitored over long periods at the expense of the sampling rate per individual. Statistical results are reliable only if the whole sampling regime is standardized. We derive practical guidelines for field studies and data analysis.  相似文献

The effects of gamma-irradiation on glucose oxidase activity were studied inBacillus cereus T spores. Radiation was found to activate oxidase in native spores. This activation was greatly enhanced when irradiated spores were subsequently heat-activated. Although activation by heat alone was 1.6 times that by radiation alone, the combined treatment with both radiation and heating resulted in a 12-fold enhancement of oxidase activity over that obtainable by optimum heat activation. Activation by combined treatment of gamma-rays and heat depended on the presence of exchangeable calcium during heating but not during irradiation.  相似文献

BACKGROUND AND PURPOSE: Ivermectin is a common anthelmintic drug, widely used in laboratory rodents for treatment of pinworm and mite infestations. We evaluated the action of ivermectin on sensitive behavioral tasks in mice during treatment for mites within a barrier facility. METHODS: A total of 21 (5 males, 16 females) mice (129/SvEv) were used for measuring body weight, open field locomotor activity, and rotarod motor coordination. For acoustic startle and prepulse inhibition, 20 C57BL/6J and 29 AKR/J mice were studied. For the Morris water task, the same 20 C57BL/6J mice were studied. Ivermectin (0.08% sheep drench) was administered in the drinking water of the home cage for 8 weeks. Control groups received normal tap water in identical bottles. RESULTS: Ivermectin did not affect general health, body weight, motor coordination, swimming behavior, or spatial learning in several inbred strains of mice. However, it induced a small but significant effect on some sensitive behaviors. CONCLUSIONS: A cautious approach to initiating ivermectin treatment in mice should be used for sensitive behavioral experiments.  相似文献

The effects of UV-C (254 nm) and UV-B (280-320 nm) on chlorophyll fluorescence induction and ultraweak luminescence (UL) in detached leaves of Hibiscus rosa-sinensis L. were investigated. UL from leaves exposed to UV-B and UV-C radiation reached a maximum 72 h after irradiation. In both cases most of the light was of a wavelength over 600 nm. An increase in the percentage of long wavelength light with time was detected. UV radiation increased peroxidase activity, which also reached a maximum 72 h after irradiation. UV-B and UV-C both reduced variable chlorophyll fluorescence. No effect on the amount of chlorophyll or UV screening pigments was observed with the short-term irradiation used in this investigation.  相似文献
