对单倍体的用途和来源作了简略的概括。回顾了通过离体诱导获得雌性单倍体的研究历程,并分析了这一方法的优势。离体诱导雌性单倍体的效果受到供体植株的基因型、供体植株的生理状态、胚囊发育时期、材料预处理、花器附属物、培养基、培养方式、培养条件等一系列因素的影响。对这些影响因子的有关研究进行了系统的总结。在雌性单倍体的个体发育方面,对一些有代表性的实验结果进行了总结。  相似文献

未授粉子房和胚珠培养离体诱导雌核发育的研究已近半个世纪,目前共有20科40余种种子植物的未授粉子房或胚珠离体培养诱导了雌核发育,但成功获得雌性单倍体的植物只有10科25种2变种。本文重点对离体雌核发育的胚胎学的研究资料进行综述,包括离体条件下胚珠中胚囊的发育状况、雌核发育的模式、雌性单倍体形成的途径以及自发胚乳的离体诱导并指出该领域研究中存在的问题,进一步在研究方法上提出了设想和建议。  相似文献

单倍体培养是快速获得菊科纯合系的重要途径。目前已进行单倍体研究的菊科植物共有13个种,其中9个已成功获得单倍体植株。菊科中诱导单倍体的途径有花药培养、小孢子培养、离体雌核培养、远源杂交和辐射花粉诱导单倍体。本文详细论述了不同外植体发育时期、预处理、培养基、培养条件等因素对单倍体植株诱导再生的影响。对菊科植物单倍体诱导的几种途径进行对比总结,指出研究中存在的问题并提出思路和建议。  相似文献

百合未授粉子房离体培养胚胎形成及植株再生   总被引:2,自引:0,他引:2  
未受精子房离体培养是诱导雌核产生单倍体的技术之一。以1个野生种和3个杂种系共7个百合(Lilium)基因型为实验材料, 探讨了基础培养基、花蕾取样时期和外源激素等因素对百合未授粉子房离体培养胚胎形成的影响。结果表明, CBM、MS和BDS三种基础培养基均可诱导百合未授粉子房胚胎形成, 但以BDS培养基诱导效果最佳; 开花前1天的花蕾较适于百合未授粉子房离体培养; 2 mg·L-1 2,4-D + 2 mg·L-1 6-BA和2 mg·L-1 2,4-D + 2 mg·L-1 KT两种外源激素配方均适用于百合未授粉子房离体培养诱导胚胎形成。在培养过程中, 大多数胚性胚珠中只含有1个胚胎, 位于珠孔端、合子端或极核处, 少数胚性胚珠中含有双胚胎。通过百合未授粉子房离体培养, 从5个基因型材料中共获得146株再生植株。采用根尖染色体计数法对其中的62株进行了倍性测定, 其中43株与母体植株染色体倍性不同。  相似文献

迄今,有关向日葵离体雌核发育的研究,仅蔡得田与周嫦(1983)、Gelebart与San(1987)由未传粉子房培养分别得到单倍体小苗和植株,而在未受精胚珠培养方面,阎华等对诱导胚状体形成的因素作了大规模研究,但未继续研究其植株再生问题。我们在上述基础上,经过三次重复实验,建立了向日葵未受精胚珠培养再生单倍体植株的技术流程,使其进一步用于育种及细胞工程成为可能。  相似文献

介绍被子植物未受精胚珠与子房离体培养诱导单倍体植株的研究进展。迄今已有9个科21种植物用这一方法诱导出单倍体植株,植物的基因型、外植体的发育程度、接种前的预处理、培养基和培养条件等均能影响诱导率的高低。胚胎学观察揭示大孢子与胚囊内的卵细胞、助细胞和反足细胞均有可能在培养中启动分裂,通过胚状体或愈伤组织形成单倍体植株。本项技术在植物单倍体育种中可发挥重要作用。  相似文献

通过不同培养基及培养条件的筛选,离体诱导12个品种的非洲菊未授粉胚珠的结果表明:MS中的大量元素 Heller中的微量元素 1/2铁盐 0.2mg·L~(-1)6-BA 0.1mg·L~(-1)IAA较适宜于非洲菊未授粉胚珠愈伤组织的诱导和芽的再生,8个品种中以品种‘E19’诱导愈伤组织的诱导率最高(为23.1%);5个品种可再生形成不定芽,再生率为4.8%-19.6%。用根尖染色体鉴定法鉴定再生的23个植株的倍性的结果显示,21.7%为二倍体,43.5%为单倍体,34.8%为混倍体.  相似文献

Gynogenesis was used to map eight loci to their centromeres in Xenopus laevis. Several loci remote from their centromeres were identified. This information may be useful in distinguishing gynogenetic diploid progeny produced by suppression of second polar bodies from gynogenetic diploid progeny homozygous at all loci produced by suppression of first cleavage of gynogenetic haploids.  相似文献

Flow cytometry was used to screen ploidy levels in 47 cultured in vitro sugar beet gynogenetic lines of various origin and age, obtained after plant regeneration from unfertilized ovules. When donor plants were diploid, the majority of regenerants were found to have cells with 1C, 2C and 4C relative DNA content (mainly haploid and diploid) and there were large differences in the rate of spontaneous in vitro chromosome doubling between individual homozygous lines. Six ovule-derived lines regenerated from fertile and sterile diploid donors of forty-five lines were solid diploids from the very early stages of their in vitro cultivation, and thus could not be classified as doubled haploids. In the case of tetraploid donor plants, the gynogenetic regenerants demonstrated 2x-ploidy level. The results obtained in chimeric plants with both haploid and diploid cells indicated the possibility to overcome mixoploidy by their re-cultivation through generative shoot tip culture. The flow cytometry method confirmed data obtained by conventional microscopic chromosome counting in dividing leaf cells and was found very useful for screening of a large number of regenerants and for characterizing the process of in vitro gynogenetic lines formation in sugar beet.  相似文献

Summary Nine induced tetraploid females were artificially inseminated by UV-irradiated sperm collected from diploid males, in order to induce the gynogenetic development of their ova. Most of the resulting embryos were diploid (or minor aneuploids). Several gynogenetic tetraploids, likely to issue from unreduced ova, were also detected in these progenies. The same females fertilized by normal sperm of diploid males gave a majority of triploids and several pentaploids, while the fertilization by normal sperm of tetraploid males gave rise to a majority of tetraploids and one hexaploid. The same crosses, after the eggs had been heat-shocked to double the maternal genetic contribution, yielded about three-quarters pentaploids and one quarter haploids (normal sperm of diploids), or three-quarters hexaploids and one quarter diploids (normal sperm of tetraploids). These haploids and diploids are likely to result from androgenesis.  相似文献

Summary Hybridization of Petunia axillaris and P. parodii with Nicotiana tabacum was attempted using the method of in vitro pollination and fertilization. Seedlings were produced when the Petunia species and N. tabacum were used as the maternal parents; however, most of these had the identical somatic chromosome complement of the maternal parent. With crosses involving P. axillaris as the maternal parent, a low frequency of haploids was also produced. Due to the potential of haploids in basic and applied genetic research, additional experiments were carried out to determine whether in vitro pollination was necessary to stimulate haploid production and to more closely define the optimal time for ovule excision and culture. Four treatments were applied to accomplish these objectives. They were: placentas cultured prior to the time of anthesis, with and without pollination, and placentas cultured after the time of anthesis, with and without pollination. In vitro pollination had no effect on the frequency of haploids produced. Placenta attached ovules cultured prior to the time of anthesis produced significantly more haploids than those cultured after anthesis. The preanthesis treatment produced a frequency of 6.5 haploids per 100 ovaries cultured. Artificial mitotic gynogenesis, a chromosome set manipulation, is applied to provide the homozygous progeny with only maternal inheritance. Here, gynogenetic development was induced in the sterlet Acipenser ruthenus L. (Acipenseridae) by activation of the eggs originating from albino females with the UV-irradiated spermatozoa from wild-coloured males, followed by the heat shock applied to suppress the first mitotic division in the haploid zygotes. All experimentally obtained gynogenetic offspring possessed recessive albino coloration. Moreover, the genetic verification, based on three microsatellite DNA markers, confirmed the only maternal inheritance in the albino progeny. Cytogenetic screening enabled identification of the aneuploids, haploids, diploids, triploids, tetraploids and mosaic individuals among the gynogenetic larvae that hatched from the eggs subjected to the heat shock. Furthermore, 40% of the larvae from the haploid variants of the research that were not exposed to the temperature shock showed the diploid chromosome number. A variation of the ploidy level observed in the gynogenetic sterlets may be the consequence of the spontaneous polyploidisation that occurred in the haploid zygotes. Moreover, observation during embryogenesis showed varied stages of eggs development and the asynchronous cell cleavages that may have resulted in the chromosomal disturbances observed in the gynogenetic sterlets here.  相似文献

Summary In crosses between a homozygous rootless mutant line of Nicotiana tabacum used as female and other Nicotiana tabacum lines, androgenetic haploids can be directly selected by their ability to form plantlets with a normal rooting system, whereas hybrid plants are killed few weeks after sowing. These androgenetic plants have the nucleus of the male parent into the cytoplasm of the female parent. In crosses where the homozygous rootless mutant line is used as a pollen donor, gynogenetic haploids can also be directly selected. Haploids can therefore be derived from male sterile plants using this approach. A generalization of this system for direct cytoplasm transfer and for the screening of spontaneous haploids in dicotyledons is proposed.  相似文献

Half-smooth tongue sole (Cynoglossus semilaevis) is an important cultured marine fish as well as a promising model fish for the study of sex determination mechanisms. In the present study, a protocol for artificial gynogenesis of half-smooth tongue sole was developed in order to identify the sex determination mechanism and to generate all-female stock. The optimal UV-irradiation dose for genetically inactivating sea perch spermatozoa was determined to be ≥30 mJ/cm2. The optimal initiation time for cold shock of gynogenetic embryos was determined to be 5 min after fertilization, while the optimal temperature and treatment duration were determined to be 20–25 min at 5°C. Chromosomes from common diploids, gynogenetic haploids, and diploids were analyzed. WW chromosomes were discovered in some of the gynogenetic diploids. The microsatellite marker was applied to analyze gynogenetic diploid fry. Among the 30 gynogenetic diploid fry, 11 fry contained only one allele, while 19 contained two alleles, which had the same genotype as their mother. The female-specific DNA marker was observed in four individuals out of ten gynogenetic diploid fry. Ploidy analysis of 20 putative gynogenetic fry showed them all to be diploid. Thus, a protocol for the induction of artificial gynogenesis has been developed for the first time in half smooth tongue sole, and the sex determination mechanism in the tongue sole was determined to be female heterogametic with the ZW chromosome.  相似文献
