Summary In crosses between T. tauschii (D t) accesions, their polymorphic gliadin forms were inherited as blocks of gliadin components -Gli-D t1, Gli-D t2 — as single Mendelian characters. From the progeny of four tri-parental crosses (test-crosses), HMW glutenin subunits derived from T. tauschii (Glu-D t1) segregated as alleles of the Glu-D1 locus in bread wheat. In three of the tri-parental crosses, a small proportion (2.5%) of the progeny with atypical segregation patterns, were identified through somatic chromosome counts, to be aneuploids (1.9% hypoploids and 0.6% hyperploids). Chromosomal mapping studies revealed that the synteny of genes for HMW glutenin subunits and gliadins in T. tauschii are conserved in the D genome homologue (chromosome 1D) of T. aestivum. The map distance between the Glu-D1/-D t1 and Gli-D1/-D t1 loci was calculated to be 63.5 cM, while a linkage to the centromere of 7.7–9.7 cM was estimated for the Glu-D1/-D t1 locus.  相似文献

Summary A triple (1AL.1RS/1BL.1RS/1DL.1RS) and three double (1AL.1RS/1BL.1RS, 1AL.1RS/1DL.1RS, 1BL.1RS/1DL.1RS) wheat-rye 1RS translocation stocks were isolated from a segregating population using the Gli-1, Tri-1 and Sec-1 seed proteins as genetic markers. These stocks carried 42 chromosomes and formed the expected multivalents (frequency of 14–25%) at metaphase 1. They gave floret fertility ranging from 40–60%. These stocks were subsequently used to determine the genetic control of low-molecular-weight (LMW) glutenin subunits in Chinese Spring and Gabo by means of two-step one-dimensional SDS-PAGE. All of the B subunits and most of the C subunits of glutenin were shown to be controlled by genes on the short arms of group-1 chromosomes in these wheats. The other C subunits were not controlled by group-1 chromosomes. The triple translocation line served as a suitable third parent in producing test-cross seeds for studying the inheritance of the LMW glutenin subunits and gliadins in wheat cultivars, e.g. Chinese Spring and Orca. The segregation patterns of the LMW glutenin subunits in these cultivars revealed that the subunits were inherited in clusters and that their controlling genes (Glu-3) were tightly linked with those controlling gliadins (Gli-1). The LMW glutenin patterns d, d and e in Orca segregated as alternatives to the patterns a, a and a in Chinese Spring controlled by Glu-A3, Glu-B3 and Glu-D3 loci on chromosome arms 1AS, 1BS and 1DS, respectively, thus indicating that these patterns were controlled by allelic genes at these loci.  相似文献

Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. Summary Analysis of intergeneric substitution lines in hexaploid wheats by a two-step electrophoretic method of protein separation revealed that low-molecular-weight (LMW) subunits of glutelin in Triticum longissimum, T. Umbelullatum, Elytrigia elongata (2 x) were controlled by chromosomes/chromosome arms 1S l , 1U, and 1ES, respectively. A LMW glutelin band in Secale montanum was detected but its chromosomal location could not be determined. Genes controlling gliadins and HMW subunits of glutelin were also located on chromosome 1S l in T. longissimum.The term glutelin refers to the polymeric prolamins of cereals, e.g., glutenins in wheat, HMW, and 75-k secalins in rye  相似文献

Two biotypes of the bread-wheat cultivar Alpe were shown to possess contrasting alleles at each of the glutenin (Glu-B1, Glu-D1, Glu-B3 and Glu-D3) and gliadin (Gli-B1 and Gli-D1) loci on chromosomes 1B and 1D. Fourteen near-isogenic lines (NILs) were produced by crossing these biotypes and used to determine the genetic control of both low-molecular-weight (LMW) glutenin subunits and gliadins by means of one-dimensional or two-dimensional electrophoresis. Genes coding for the B, C and D groups of EMW subunits were found to be inherited in clusters tightly linked with those controlling gliadins. Southern-blot analysis of total genomic DNAs hybridized to a -gliadin-specific cDNA clone revealed that seven NILs lack both the Gli-D1 and Glu-D3 loci on chromosome 1D. Segregation data indicated that these null alleles are normally inherited. Comparison of the null NILs with those possessing allele b at the Glu-D3 locus showed one B subunit, seven C subunits and two D subunits, as fractionated by two-dimensional A-PAGExSDS-PAGE, to be encoded by this allele. Alleles b and k at Glu-B3 were found to code for two C subunits plus eight and six B subunits respectively, whereas alleles b and k at Gli-B1 each controlled the synthesis of two -gliadins, one and two -gliadins. The novel Gli-B5 locus coding for two -gliadins was shown to recombine with the Gli-B1 locus on chromosome 1B. The two-dimensional map of glutenin subunits showed -gliadins encoded at the Gli-A2 locus on chromosome 6A. Summary The zymogram phenotypes of glucose-phosphate isomerase (GPI), alcohol dehydrogenase-1 (ADH-1), glutamate oxaloacetate transaminase (GOT), superoxide dismutase (SOD), lipoxygenase (LPX), esterase (EST) and the banding patterns of gliadin and glutenin seed storage proteins were determined for Triticum aestivum cv. Chinese Spring (CS), Dasypyrum villosum, the octoploid amphiploid T. aestivum cv. Chinese Spring D. villosum (CS × v) (2n=8x=56; AABBDDVV), and for five CS-D. villosum disomic addition lines. The genes Gpi-V1, Adh-V1, Got-V2, and Sod-V2 coding for GPI-1, ADH-1, GOT-2, and SOD-2 isozymes were located in D. villosum on chromosome 1V, 4V, 6V, and 7V, respectively. Genes coding for gliadin- and glutenin-like subunits are located in D. villosum chromosomes 1V. There are no direct evidence for chromosomal location of genes coding for GOT-3, EST-1 and LPX-2 isozymes. The linkage between genes coding for glutenin-like proteins and GPI-1 isozymes in chromosome 1V is evidence of homoeology between chromosome 1V and the chromosomes of homoeologous group 1 in wheat.Research supported by the National Research Council (Italy) and National Science Foundation (USA). Behavioral and physiological responses to hypoxia were examined in three sympatric species of sharks: bonnethead shark Sphyrna tiburo, blacknose shark, Carcharhinus acronotus, and Florida smoothhound shark, Mustelus norrisi, using closed system respirometry. Sharks were exposed to normoxic and three levels of hypoxic conditions. Under normoxic conditions (5.5–6.4mg l–1), shark routine swimming speed averaged 25.5 and 31.0cm s–1 for obligate ram-ventilating S. tiburo and C. acronotus respectively, and 25.0cm s–1 for buccal-ventilating M. norrisi. Routine oxygen consumption averaged about 234.6 mg O2kg–1h–1 for S. tiburo, 437.2mg O2kg–1h–1 for C. acronotus, and 161.4mg O2 kg–1 h–1 for M. norrisi. For ram-ventilating sharks, mouth gape averaged 1.0cm whereas M. norrisi gillbeats averaged 56.0 beats min–1. Swimming speeds, mouth gape, and oxygen consumption rate of S. tiburo and C. acronotus increased to a maximum of 37–39cm s–1, 2.5–3.0cm and 496 and 599mg O2 kg–1 h–1 under hypoxic conditions (2.5–3.4mg l–1), respectively. M. norrisi decreased swimming speeds to 16cm s–1 and oxygen consumption rate remained similar. Results support the hypothesis that obligate ram-ventilating sharks respond to hypoxia by increasing swimming speed and mouth gape while buccal-ventilating smoothhound sharks reduce activity.  相似文献

The dorsal skin of the leech Hirudo medicinalis was used for electrophysiological measurements performed in Ussing chambers. The leech skin is a tight epithelium (transepithelial resistance = 10.5±0.5 k· cm-2) with an initial short-circuit current of 29.0±2.9 A·cm-2. Removal of Na+ from the apical bath medium reduced short-circuit current about 55%. Ouabain (50mol·l-1) added to the basolateral solution, depressed the short-circuit current completely. The Na+ current saturated at a concentration of 90 mmol Na+·l-1 in the apical solution (K M=11.2±1.8 mmol·l-1). Amiloride (100 mol·l-1) on the apical side inhibited ca. 40% of the Na+ current and indicated the presence of Na+ channels. The dependence of Na+ current on the amiloride concentration followed Michaclis-Menten kinetics (K i=2.9±0.4 mol·l-1). The amiloride analogue benzamil had a higher affinity to the Na+ channel (K i=0.7±0.2 mol·l-1). Thus, Na+ channels in leech integument are less sensitive to amiloride than channels known from vertebrate epithelia. With 20 mmol Na+·l-1 in the mucosal solution the tissue showed an optimum amiloride-inhibitable current, and the amiloride-sensitive current under this condition was 86.8±2.3% of total short-circuit current. Higher Na+ concentrations lead to a decrease in amiloride-blockade short-circuit current. Sitmulation of the tissue with cyclic adenosine monophosphate (100 mol·l-1) and isobutylmethylxanthine (1 mmol·l-1) nearly doubled short-circuit current and increased amiloride-sensitive Na+ currents by 50%. By current fluctuation analysis we estimated single Na+ channel current (2.7±0.9 pA) and Na+ channel density (3.6±0.6 channels·m-2) under control conditions. After cyclic adenosine monophosphate stimulation Na+ channel density increased to 5.4±1.1 channels·m-2, whereas single Na+ channel current showed no significant change (1.9±0.2 pA). These data present a detailed investigation of an invertebrate epithelial Na+ channel, and show the similarities and differences to vertebrate Na+ channels. Whereas the channel properties are different from the classical vertebrate Na+ channel, the regulation by cyclic adenosine monophosphate seems similar. Degradation of polyaromatic hydrocarbons by Burkholderia cepacia 2A-12   总被引:1,自引:1,他引:0  
A new strain of bacterium degrading polyaromatic hydrocarbons (PAHs), Burkholderia cepacia 2A-12, was isolated from oil-contaminated soil. Of three PAHs, the isolated strain could utilize naphthalene (Nap) and phenanthrene (Phe) as a sole carbon source but not pyrene (Pyr). However, the strain could degrade Pyr when a cosubstrate such as yeast extract (YE) was supplemented. The PAH degradation rate of the strain was enhanced by the addition of other organic materials such as YE, peptone, glucose, and sucrose. YE was a particularly effective additive in stimulating cell growth as well as PAH degradation. When 1 g YE l–1, an optimum concentration, was supplemented into the basal salt medium (BSM) with 215 mg Phe l–1, the specific growth rate (0.30 h–1) and Phe-degrading rate (29.6 mol l–1 h–1) were enhanced approximately ten and three times more than those obtained in the BSM with 215 mg Phe l–1, respectively. Both cell growth and PAH degradation rates were increased with increasing Phe and Pyr concentrations, and B. cepacia 2A-12 had a tolerance against Phe and Pyr toxicity at the high concentration of 730–760 mg l–1. Through kinetic analysis, the maximum specific growth rate ( max) and PAH degrading rate ( max) for Phe were obtained as 0.39 h–1 and 300 mol l–1 h–1, respectively. Also, max and max for Pyr were 0.27 h–1 and 52 mol l–1 h–1, respectively. B. cepacia 2A-12 could simultaneously degrade crude oil as well as PAHs, indicating that this bacterium is very useful for the removal of oils and PAHs contaminants.  相似文献

Dibutyl phthalate, oleic acid and terpineol were used to extract paclitaxel in situ fromTaxus chinensis suspension cultures. Oleic acid/terpineol (1:1, v/v) added to the cultures gave a higher paclitaxel concentration, compared with either of them alone. Oleic acid/terpineol (1:1, v/v) incorporated into the cultures at 3:50 (v/v) 4 days after elicitation, which was carried out by adding 50 mg chitosan l–1, 60 M methyl jasmonate and 30 M Ag+ to 10-day-old cultures, resulted in the greatest paclitaxel production of 48 mg l–1 at day 10 after elicitation. This was double that of the culture by elicitation, and 7-fold higher than that of the culture by in situ extraction.  相似文献

1-Methylcyclopropene (1-MCP), formerly designated as Sis-X, has been shown to be an effective inhibitor of ethylene responses in carnation flowers in either the light or the dark. The binding appears to be to the receptor and to be permanent. A 6 h treatment at 2.5 nl l–1 is sufficient to protect against ethylene, and 0.5 nl l–1 is sufficient if exposure is for 24 h. As carnation flowers age, a little higher concentration appears to be needed. Most of the natural increase in ethylene production during senescence is prevented by treatment with 1-MCP. A closely related compound, methylenecyclopropane shows ethylene activity. A tritium labelled 1-MCP (60 mCi mmol–1) has been prepared. A higher specific activity is needed for more critical studies.  相似文献

A hexaploid wheat landrace collected from the Baluchistan province of Pakistan was found to possess a novel high-molecular-weight glutenin subunit (HMW-GS). The subunit has a very slow electrophoretic mobility as revealed by SDS-PAGE, and its molecular weight is comparable to that of the highest molecular weight glutenin subunit (2.2 encoded in the D-genome) reported so far in hexaploid wheat varieties and landraces of Japanese origin. Evidence obtained from (PCR) gene amplification studies using the primers specific for Glu-1 loci proved that the gene coding for this novel subunit belongs to the Glu-A1 locus located on the long arm of chromosome 1A. Digestion of the amplified gene (PCR product) with restriction enzymes indicated that the novel gene differs from prevailing Glu-A1 alleles (null, 1 and 2*) by an extra DNA fragment of approximately 600 base pairs. The results also indicated that the novel subunit is most probably a derivative of subunit 2* that has very likely incorporated the 600-bp fragment following a process of unequal crossing over. The present findings were further substantiated by reserved phase high performance liquid chromatography (RP-HPLC) analysis.  相似文献

Using primary cultures of gill pavement cells from freshwater rainbow trout, a method is described for achieving confluent monolayers of the cells on glass coverslips. A continuous record of intracellular pH was obtained by loading the cells with the pH-sensitive flourescent dye 2,7-bis(2-carboxyethyl)-5(6)-carboxyfluorescein and mounting the coverslips in the flowthrough cuvette of a spectrofluorimeter. Experiments were performed in HEPES-buffered media nominally free of HCO3. Resting intracellular pH (7.43 at extracellular pH=7.70) was insensitive to the removal of Cl or the application of 4-acetamido-4-isothiocyanatostilbene-2,2-disulfonic acid (0.1 mmol·l–1), but fell by about 0.3 units when Na+ was removed or in the presence of amiloride (0.2 mmol·l–1). Exposure to elevated ammonia (ammonia prepulse; 30 mmol·l–1 as NH4Cl for 6–9 min) produced an increase in intracellular pH (to about 8.1) followed by a slow decay, and washout of the pulse caused intracellular pH to fall to about 6.5. Intracellular non-HCO 3 buffer capacity was about 13.4 slykes. Rapid recovery of intracellular pH from intracellular acidosis induced by ammonia prepulse was inhibited more than 80% in Na+-free conditions or in the presence of amiloride (0.2 mmol·l–1). Neither bafilomycin A1 (3 mol·l–1) nor Cl removal altered the intracellular pH recovery rate. The K m for Na+ of the intracellular pH recovery mechanism was 8.3 mmol·l–1, and the rate constant at V max was 0.008·s–1 (equivalent to 5.60 mmol H+·l–1 cell water·min–1), which was achieved at external Na+ levels from 25 to 140 mmol·l–1. Summary Variation in high molecular weight (HMW) glutenin subunit composition among 167 accessions of dicoccum wheat (Triticum turgidum L. var. dicoccum Schrank) of diverse origins was investigated using one-dimensional sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). A total of 20 alleles were identified, and 9 of them were found to be different from those previously detected by Payne and Lawrence (1983 b) in hexaploid wheat (Triticum aestivum L.). The newly discovered alleles enhance the genetic variability available to improve the industrial quality of wheats and some of them may facilitate basic research on the relationship of industrial quality with HMW glutenin subunit number. The novel variants include a GLU-A1 encoded subunit which has higher molecular mass than any other so far described in tetraploid and hexaploid wheats, and a null GLU-B1 allele. Dicoccums containing neither GLU-A1- nor GLU-B1-encoded subunits were also identified. A comparison of the mean number of HMW glutenin subunits contained in various primitive and modern domesticated wheats of different ploidy levels and the identification of wheats containing no HMW glutenin subunits suggest that the occurrence of null GLU-1 alleles in these species depends on chance rather on an inherent tendency on the part of modern polyploid wheats to suppress the activity of redundant GLU-1 genes.  相似文献

Isolated posterior gills (no. 7) of shore crabsCarcinus maenas acclimated to brackish water of a salinity of 10 S were bathed and perfused with 50% sea water (200 mmol·l-1 Na+), and the internal perfusate collected during subsequent periods of 5 min. During a single passage through the gill the pH of the perfusion medium decreased from ca. 8.1 to ca. 7.7, a result implying that the gill possesses structures which recognize unphysiologically high pH values in the haemolymph and regulates them down to physiological values of ca. 7.7. The calculated apparent proton fluxes from the epithelial cells into the haemolymph space amounted to 17.9 mol·g fw-1·h-1, a value of only 3.8% of net Na+ fluxes observed under comparable conditions. When 0.1 mmol·l-1 KCN, an inhibitor of mitochondrial cytochrome oxidase, or 5 mmol·l-1 ouabain, a specific inhibitor of Na+/K+-ATPase were applied in the internal perfusate, down-regulation of pH was no longer observed and the gill was completely depolarized, i.e. transepithelial potential differences dropped from-7.8 to 0 mV (haemolymph space negative to bath). Regulation of pH was completely inhibited by antagonists of carbonic anhydrase (0.1 mmol·l-1 acetazolamide or 0.01 mmol·l-1 ethoxyzolamide) applied in the perfusate. Inhibitors of Na+/H+ exchange, 0.1 mmol·l-1 amiloride applied in the external bathing medium or in the internal perfusate, and symmetrical 0.01 mmol·l-1 5-(N-ethyl-N-isopropyl)amiloride, as well as inhibitors of Cl-/HCO3 - exchange and Na+/HCO3 - cotransport, 0.5 mmol·l-1 4,4-diisothiocyanatostilbene-2,2-disulphonate or 0.3 mmol·l-1 4-acetamido-4-isothiocyanatostilbene 2,2-disulphonate applied on both sides of the gill, and inhibitors of H+-ATPase, 0.05 mmol·l-1 N-ethylmaleimide and 0.1 mmol·l-1 N,N-dicyclohexylcarbodiimide —applied on both sides of the gill — did not alter the acidification of the perfusate observed in controls. Using artificial salines buffered to pH 8.1 with 0.75 mmol·l-1 tris (hydroxymethyl) aminomethane instead of 2 mmol·l-1 HCO3 -, apparent proton fluxes were reduced to 11% of controls, a result suggesting that pH regulation by crab gills needs the presence of HCO3 -. The findings obtained suggest that pH regulation by crab gills depends on the oxidative metabolism of the intact branchial epithelium and that carbonic anhydrase plays a central role in this process. Na+/H+ exchange, anion exchange or cotransport and active proton secretion seem not to be involved. Summary An attempt to produce a set of addition lines of Aegilops sharonensis to the wheat variety Chinese Spring produced only one addition line. This was due to preferential transmission of one chromosome from Ae. sharonensis. This chromosome was studied in detail by established cytological methods of chromosome observation and by the newer techniques of C-banding and in situ hybridization of a cloned DNA sequence. The chromosome was found to be partially homologous to an Ae. sharonensis chromosome of similar behaviour in another wheat addition line. The incomplete homology of the two Ae. sharonensis chromosomes was due to the presence of a translocated segment of a wheat chromosome. — Substitution lines of the Ae. sharonensis chromosome for wheat homoeologous group 4 were produced and the Ae. sharonensis chromosome thereby designated 4 S l .  相似文献

The effect of higher cell densities on the expression and segregational stability of a recombinant E. coli- B. subtilisshuttle plasmid coding for carboxymethylcellulase (CMCase) activity, was studied in E. coli DH5. Of the various feeding policies adopted for maximal expression and stability, exponential feeding resulted in the highest biomass of 15g dry cell weight (DCW) l–1 and plasmid stability of 45%. A CMCase activity of 11400 Uml–1 was achieved as compared to 230 Uml–1 during batch cultivation. Catabolites related to tricarboxylic acid cycle affected growth and carotenogenesis in Phaffia rhodozyma. Glutamate, glutamine, aspartate, asparagine and proline at 75 mM of N increased biomass from 2 g l–1 to 2.9–4.7 g l–1 but decreased carotenoid from 420 g g–1 yeast to 200–260 g g–1 yeast in strain 67-385. However, simple nitrogen sources did not decrease carotenoid formation. Tricarboxylic acid intermediates repressed carotenogenesis to a less degree than the corresponding amino acids. Carotenoid hyper-producing mutants were impaired in nitrogen utilization. These results indicated that nitrogen assimilation and the concentrations of tricarboxylic acid cycle intermediates are involved in regulation of carotenoid biosynthesis.  相似文献

Synopsis Arsenic persists in Chautauqua Lake, New York waters 13 years after cessation of herbicide (sodium arsenite) application and continues to cycle within the lake. Arsenic concentrations in lake water ranged from 22.4–114.81 g l–1, = 49.0 ag l–1. Well water samples generally contained less than 10 g l–1 arsenic. Arsenic concentrations in lake water exceeded U.S. Public Health Service recommended maximum concentrations (10 g l–1) and many samples exceeded the maximum permissible limit (50 g l–1). Fish accumulated arsenic from water but did not magnify it. Fish to water arsenic ratios ranged from 0.4–41.6. Black crappie (Pomoxis nigromaculatus) contained the highest arsenic concentrations (0.14–2.04 g g–1 ), X = 0.7 g g–1) while perch (Perca flavescens), muskellunge (Esox masquinongy) and largemouth bass (Micropterus salmoides) contained the lowest concentrations (0.02–0.13 g g–1). Arsenic concentrations in fish do not appear to pose a health hazard for human consumers.  相似文献

A 23 full factorial design was used to study the influence of different experimental variables, namely wort gravity, fermentation temperature and nutrient supplementation, on ethanol productivity from high gravity wort fermentation by Saccharomyces cerevisiae (lager strain), under pilot plant conditions. The highest ethanol productivity (0.69 g l–1 h–1) was obtained at 20°P [°P is the weight of extract (sugar) equivalent to the weight of sucrose in a 100 g solution at 20°C], 15°C, with the addition of 0.8% (w/v) yeast extract, 24 mg l–1 ergosterol and 0.24% (v/v) Tween 80.  相似文献
