Samples of each instar of the 3-host tick, Haemaphysalis (Kaiseriana) longicornis were collected from grazing Bos taurus cattle to investigate their survival on hosts with previous exposure to the tick and on previously unexposed ‘naive’ hosts. The ticks were allocated to categories based on their size to observe at what stage of development mortality occurred. Analysis of the age structure of the ticks showed that small but variable proportions of larvae, nymphs and females matured on previously exposed hosts and larger proportions on the naive hosts. This suggests that cattle acquire and express different levels of resistance against each instar of the tick. The technique used has potential for estimating levels of resistance of hosts to 3-host ticks under field conditions.  相似文献

We conducted laboratory and field experiments to elucidate the life history of Ixodiphagus hookeri, a parasitoid of the ixodid tick Amblyomma variegatum in Western Kenya. Ixodiphagus hookeri females oviposited in unfed host nymphs as well as engorged nymphs, but rarely in engorged larvae. While I. hookeri developed to adults in engorged nymphs, the eggs laid in unfed nymphs disappeared within 2 days after oviposition. As temperature increased, development time of I. hookeri from oviposition to adult emergence in engorged nymphs decreased from 46 days at 23 °C to 35 days at 28 °C, and their immature survival in engorged nymphs decreased from 67% at 23 °C to 22% at 28 °C. No parasitoid adult emerged from hosts at 30 °C. Individual hosts parasitized by single females produced 42–53 adult wasps, 73% of which were females. As a typical pro-ovigenic species, I. hookeri females had an average of 84 mature eggs at emergence and lived only for a few days. When laboratory-reared, unfed nymphs of A. variegatum were attached to cattle for 4–9 days in subsistence farmers’ fields in Western Kenya, 25% of the engorged nymphs and 4% of the unfed nymphs on cattle were parasitized by I. hookeri, demonstrating that I. hookeri females search for and oviposit in A. variegatum nymphs on cattle. Unlike other strains of I. hookeri that overwinter as eggs in unfed nymphs, I. hookeri could continuously reproduce throughout the year in Western Kenya.  相似文献

Three experiments were carried out in which Theileria parva was irradiated in its tick vector, Rhipicephalus appendiculatus. In the first experiment, infected unfed adult ticks were irradiated at doubling doses from 4 to 32 krad. Some of the ticks were then fed for 4, 5, 6, 7 and 8 days on rabbits, and the parasites in their salivary glands examined. Five male and 5 female ticks from each irradiation dose were put onto each of a pair of susceptible cattle, whose reactions were recorded. Increasing doses of irradiation resulted in progressive destruction of the parasites. All cattle receiving ticks irradiated at doses up to and including 16 krad died of East Coast fever (ECF), and one of the cattle receiving ticks irradiated at 32 krad died.In the second experiment, recently engorged nymphs were irradiated at 1, 2 or 4 krad, and moulting nymphs at doses of 2, 4, 8, 12, 16 or 32 krad. The salivary glands of the resultant adult ticks were examined after the ticks had fed for 4, 5, 6, 7 or 8 days on rabbits. Engorged nymphs irradiated at 4 krad failed to moult, whilst moulting nymphs irradiated at 32 krad moulted but failed to attach to rabbits. Doses of irradiation survived by the ticks had no apparent morphological effect on the parasites they contained.In the third experiment, infected unfed adult ticks were irradiated at 0, 5, 10, 15, 20 25 30, 35, 40, 45, 50 or 60 krad. The ticks were fed on rabbits for 5, 6 or 7 days. Some of them were then examined morphologically, whilst others were ground in MEM/BPA and aliquots of the supernatant used to inoculate groups of 5 cattle. The reaction of these cattle, together with the morphological examination of the parasites, suggested that increasing doses of irradiation destroyed increasing numbers of parasites.  相似文献

A colony of Amblyomma neumanni was started with engorged females collected from cattle in the province of Salta (24° 51S, 65° 33W), Argentina. The larvae and nymphs were fed on rabbits and the adults on calves. The non-parasitic stages were maintained in darkness at 27 ± 1°C and 83–86% RH. The life cycle (pre-feeding period not tested) had a mean duration of 205.7 days. The mean time (days) for the different phases of the cycle were as follows: feeding of females 8.8, pre-oviposition 23.8, oviposition 41.4, minimum egg incubation 76.1, feeding of larvae 8.5, pre-moult to nymphs 16.4, feeding of nymphs 7.9 and pre-moult to adults 22.8. The mean recovery rates of larvae, nymphs and females were 83.8, 85.6 and 89.3%, respectively. The nymphs moulting to females were heavier (8.1 ± 2.34 mg) than those moulting to males (6.0 ± 2.34 mg; p < 0.01), but their range of engorgement weight showed overlap (2.3–16.2 versus 2.2–12.8 mg, respectively). Two gynandromorphs were detected between the nymphs. D. 1984: Survival of Rhipicephalus appendiculatus (Acarina: Ixodidae) and persistence of Theileria parva (Apicomplexa: Theileriidae) in the field. International Journal for Parasitology14; 483–489. Two paddocks with populations of the African brown ear tick (Rhipicephalus appendicuiatus), one of which carried virulent Theileria parva (the causative organism of East Coast fever [ECF] of cattle), were left unstocked for periods of 338 and 354 days. Groups of 1–3, ECF-susceptible cattle were then introduced eight times during the following year to assess the tick and disease challenge, The ticks were also monitored continuously on the ground. The test cattle developed fatal ECF from adult ticks which had fasted for up to 554 days. A non-pathogenic, antigenicaliy distinct, Theileria species was also detected which was still transmitted by adult ticks after 600 days. The mean survival time of unfed larvae was 175 days (max. 240 days); unfed nymphs 270 and 450 (max. 540 days); unfed adults 420 days (max. 730 days), with females in a clear majority after 270 days.  相似文献

In population biology, loop analysis is a method of decomposing a life cycle graph into life history pathways so as to compare the relative contributions of pathways to the population growth rate across species and populations. We apply loop analysis to the transmission graph of five pathogens known to infect the black-legged tick, Ixodes scapularis. In this context loops represent repeating chains of transmission that could maintain the pathogen. They hence represent completions of the life cycle, in much the same way as loops in a life cycle graph do for plants and animals. The loop analysis suggests the five pathogens fall into two distinct groups. Borellia burgdorferi, Babesia microti and Anaplasma phagocytophilum rely almost exclusively on a single loop representing transmission to susceptible larvae feeding on vertebrate hosts that were infected by nymphs. Borellia miyamotoi, in contrast, circulates among a separate set of host types and utilizes loops that are a mix of vertical transmission and horizontal transmission. For B. miyamotoi the main loop is from vertebrate hosts to susceptible nymphs, where the vertebrate hosts were infected by larvae that were infected from birth. The results for Powassan virus are similar to B. miyamotoi. The predicted impacts of the known variation in tick phenology between populations of I. scapularis in the Midwest and Northeast of the United States are hence markedly different for the two groups. All of these pathogens benefit, though, from synchronous activity of larvae and nymphs.  相似文献

A rearing method for Ixodes (Pholeoixodes) hexagonus, the hedgehog tick, was established which enabled the life cycle of immature stages to be studied under laboratory conditions. Larvae were fed on Swiss mice and nymphs on the cars of New Zealand rabbits. The feeding time of the larvae and nymphs on both hosts was 4–17 days. Larvae moulted to nymphs 15–21 days after detachment from mice. The premoulting period was 13–26 days for newly emerged males and 15–27 days for females. Engorged nymphs which developed into males weighed less (5.64±0.91 mg) than those that developed into females (6.019±88mg). The sex ratio (male: female) under laboratory conditions was 1:1.13.  相似文献

Previously unexposed ‘naive’ cattle were infested with between 500 and 20,000 larvae of the tick Boophilus microplus, to observe whether density dependent mortality of the tick occurs on cattle which have not acquired resistance. Similar proportions of larvae matured at different densities, which contrasts with previous findings of increased mortality at higher densities on cattle with acquired resistance to B. microplus. Major differences were observed between the proportions of larvae maturing in different experiments.  相似文献

The development of Borrelia theileri infections in the tick Boophilus microplus was studied during all stages of the tick developmental cycle. Light microscopical examination of hemolymph and ovary smears from ovipositing females allowed identification and separation of infected and uninfected ticks. A Borreli-free tick colony was established. Small numbers of spirochetes were present in larvae, with numbers increasing through the nymphal and adult tick stages. Borreliae occurred in hemolymph, hypodermis, midgut, Malpighian tubules, ovary, Gené's organ, and the central ganglion of engorging and ovipositing females and their eggs. The ovary, central ganglion, and hemolymph seemed to be preferred sites for the spirochete, with extensive multiplication occurring in hemocytes. No measurable effect of spirochete multiplication upon feeding and reproductive performance of ticks could be detected. Infections in cattle caused fever of short duration which coincided with the presence of spirochetes in blood smears. Morphology and size of blood and tick forms were consistent with those of B. theileri reported by other authors. B. theileri is important because infections of invertebrate and vertebrate hosts may interfere with the interpretation of data in various experimental designs, and because it is probably endemic in populations of one or more tick species and their hosts throughout the world.  相似文献

The role of host size, movement, feeding status, color, and species in the visual host evaluation and recognition behavior of the tick parasitoid, Ixodiphagus hookeri Howard was investigated. Freshly emerged female parasitoids were subjected to a choice bioassay, where the test materials were placed in sealed vials and the vials placed in a Petri dish. When presented with A. variegatum live and mummified nymphs, females examined: larger nymphs significantly longer than smaller nymphs, fed nymphs significantly longer than unfed nymphs, dead and live nymphs for similar lengths of time, and grey live nymphs and yellow-brown and dark brown mummified nymphs for similar lengths of time. The total number of visits to vials containing these test materials were also not significantly different, except there were significantly more visits to yellow-brown mummies when compared to the number of visits to dark brown mummies. When presented with A. variegatum (host) and R. appendiculatus (nonhost) nymphs, the females examined A. variegatum nymphs significantly longer than R. appendiculatus nymphs. The total number of visits to vials containing A. variegatum nymphs were significantly more than the visits to the vials containing R. appendicualtus nymphs. Furthermore, females spent significantly more total examination time per visit on larger and fed A. variegatum nymphs when compared to smaller and unfed nymphs, respectively. Direct and indirect detection were significant when females were presented with fed versus unfed A. variegatum nymphs, grey nymphs versus yellow-brown mummies, and R. appendiculatus versus A. variegatum nymphs. Direct and indirect detection for the rest of the bioassays were not significantly different. Finally, The percentages of females contacting large fed A. variegatum nymphs first were significantly different from those of females contacting small unfed R. appendiculatus nymphs first. The firstcontact percentages for the rest of the bioassays were not significantly different.  相似文献

The dynamics of development of the population of taiga ticks Ixodes persulcatus Schulze, 1930 from Eastern Siberia was studied under experimental conditions. Ticks were reared at 23–25°C, relative humidity of 80–90%, and a light: darkness ratio of 16: 8 h. The mean time of egg incubation was 35.2 days; the feeding of females, larvae, and nymphs took 6.4, 3.5, and 4.5 days, respectively. The existence of two subpopulations, differing in the time of development of engorged larvae into nymphs and nymphs into adults, was experimentally demonstrated. Most ticks had short periods of larval and nymphal development that lasted 25 ± 2 and 40 ± 4 days, respectively. However, some ticks exhibited prolonged development time of 75.5 ± 2 and 95 ± 9 days, respectively. The total duration of the life cycle was 121 days in the former group and 226 days in the latter group.  相似文献

The effectiveness of a single treatment with either ivermectin or moxidectin was determined by administering a single subcutaneous injection of each endectocide at 200 g per kg body weight to cattle infested with all parasitic developmental stages (adults, nymphs, and larvae) of Boophilus microplus (Canestrini). The percentage reduction in the number of females that reached repletion following treatment (outright kill) was 94.8 and 91.1% for ivermectin and moxidectin, respectively. In addition, the reproductive capacity of the females that did survive to repletion was reduced by >99%, regardless of the endectocide. Based on these two factors, the therapeutic level of control obtained against ticks on the cattle at the time of treatment was 99.0 and 99.1% for ivermectin and moxidectin, respectively. Engorged females recovered from either group of treated cattle weighed 3-times less than untreated females, and the egg masses produced by treated females weighed 5–8-times less than egg masses produced by untreated females. Partitioning of data into three separate 7-d post-treatment intervals allowed for an estimation of the efficacy of each endectocide against each individual parasitic development stage (adult, nymph, and larva). Results indicated that both endectocides were 99.7% effective against ticks that were in either the adult or nymphal stage at the time of treatment. However, the level of control against ticks in the larval stage of development at treatment was significantly lower at 97.9 and 98.4% for ivermectin and moxidectin, respectively. Analysis of the persistent (residual) activity of the two endectocides indicated that neither material provided total protection against larval re-infestation for even 1-wk following treatment. Against larvae infested 1–4 wk following treatment, the level of control with moxidectin ranged from 92.4% (1 wk) to 19.5% (4 wk). These control levels were higher at each weekly interval than for ivermectin, which ranged from 82.4% (1 wk) to 0.0% (4 wk). The potential for the use of these injectable endectocide formulations in the US Boophilus Eradication Program is discussed.This paper reports the results of research only. The life cycle and behavior of Amblyomma rotundatum were evaluated under laboratory conditions. The experiment started with four engorged females collected from toads (Rhinella schneideri) naturally infested at the Pirapitinga Ecological Station in the state of Minas Gerais, Brazil. Developmental periods of free-living stages were assessed in an incubator at 27 ± 1 °C, >80 % RH and darkness. The complete life cycle, including pre-attachment periods for each parasitic stage, ranged from 126 to 228 days. The pre-attachment, feeding and molting periods increased as the life cycle progressed from larva to adult female. Oviposition lasted about 20 days, with the peak occurring on days 4 and 5. Longevity of nymphs and adult females was quite similar (approximately 250 and 240 days, respectively) and slightly longer than that of larvae. Lesions caused by tick feeding are discussed and a list of known hosts, including new host records for A. rotundatum, is offered.  相似文献

Macrorhaphis acuta were bred in the laboratory (25°C) and fed on larvae ofAscotis selenaria reciprocaria. The incubation period was 8.4 days and the mean hatch was 94.4%. There were 5 nymphal instars which occupied 3.3, 5.0, 5.6, 6.6 and 12.0 days, respectively, from the lst to the 5th. Each nymph consumed an average of 22.8 host larvae to complete development. Adults had a mean longevity of 74.9 days and each consumed an average of 67.5 host larvae. Significant reduction was noted in pupation of 5th instar host larvae sucked by adult predators for periods ranging from 4 to 10 mn. The food requirements of 3rd, 4th, and 5th instar nymphs fed on 3rd instar host larvae were also investigated. The conversion ratio varied with the weight of food consumed during the instar and later instars were more efficient than earlier ones. The minimum life cycle of Dermacentor silvarum Olenev had a mean duration of 87.5 days (range 74–102 days) under laboratory conditions [(27±1 °C), 70% RH, 6 L: 18 D]. The mean time in (days) for the different stages of its cycle was as follows: incubation period of eggs was 15.3 days; prefeeding, feeding and premoulting periods of larvae and nymphs averaged 5.5, 4.0 and 7.3 days, and 5.2, 5.0 and 14.6 days, respectively; prefeeding, feeding, preoviposition and oviposition periods of female adults lasted for 7.8, 4.5, 4.3 and 14.0 days, respectively. There existed a highly significant correlation between engorged body weight of females and egg masses laid (r = 0.9877, p<0.001). The reproductive efficiency index (REI) and reproductive fitness index (RFI) in females were 11.09 and 9.58, respectively. No relationship between nymphal engorged body weight and resultant sexes was observed. Delayed feeding and non-oviposition (in June and July) existed in females, and low temperature (−10 °C) treatment for 45 days could terminate oviposition diapause. However, the egg masses laid by post-diapause females were significantly smaller than those laid by females engorged in March, April and May.  相似文献

Bothriocroton hydrosauri is a three-host ixodid tick that infests large reptiles in southeastern Australia, where its most common host is a large scincid lizard Tiliqua rugosa . Based on previous ecological and behavioural studies of this system, we propose a 'ripple' model of tick population dynamics, where only a few female ticks succeed in producing surviving offspring. These females then are the centres of ripples of their progeny spreading into the broader landscape. The model predicts higher relatedness among larvae than among nymphs or adults on a host, and significant spatial autocorrelation in larvae extending further than for the later life stages. The model also predicts that adult ticks are likely to encounter related partners and that this will generate inbreeding within the population. We tested those predictions using nine polymorphic microsatellite loci on a sample of 848 ticks (464 larvae, 140 nymphs and 244 adults) collected from 98 lizard hosts from near Bundey Bore Station in South Australia. Our data support the predictions and indicate that the dynamics of transmission among hosts play an important role in parasite population structure.  相似文献

Friesian cattle were immunized with two inoculations of anti-tick Bm86 (Tick-GARD) vaccine and were challenged 30 or 90 d later with Boophilus annulatus larvae derived from 1.2 g of eggs. No nymphs or adult ticks were found on the immunized cattle during four weeks after challenge. Repeated infestations (2 to 4) with larvae on three other calves during a period of 160 and 390 d after the immunization did not result in development of nymphal and adult stages. In control, non-immunized cattle infested with corresponding batches of larvae 1380 to 4653 replete adult female ticks were collected. Larvae issued from Babesia bovis-infected female ticks transmitted the infection to Bm86-immunized cattle, but the progeny of B. bigemina-infected females did not. Since B. bigemina is transmitted exclusively by nymphal stages of Bo. annulatus these results support the observation that immunity induced by Bm86 affects the larval stage of this tick.  相似文献

A colony ofAmblyomma parvum was started with engorged females collected from cattle in the Province of Salta (25°01 S, 63°56 W), Argentina. The progeny of those ticks were fed on rabbits and the non-parasitic stages maintained at 27±1°C, 83–86% RH in darkness. The life cycle (prefeeding period not evaluated) had a mean duration of 99.6 days. The mean time (days) for the different phases of the cycle were as follows: feeding period of females, 8.0; pre-oviposition period, 5.7; oviposition period, 17.5; minimum incubation period of the eggs, 31.8; feeding period of larvae, 3.2; premoult period to nymphs, 10.9; feeding period of nymphs, 4.7; premoult period to adults, 17.8. The oviposition pattern was typical of an ixodid tick, including a linear relationship between weights of engorged females and the number of eggs laid (r=0.8659). The males increased 18% in weight after feeding on hosts (P<0.01). The mean recovery rates of larvae, nymphs and females were 28.2%, 95.3% and 90.7%, respectively. The nymphs moulting to females were heavier (6.8±0.69 mg) than those moulting to males (3.2±0.29 mg) (P<0.01). Although Amblyomma brasiliense Aragão 1908 has been reported as one of the most aggressive ticks to humans in Brazil, information about the biology of this tick species is virtually inexistent. This work reports data on the life cycle of A. brasiliense fed on rabbits and pigs and maintained in an incubator at 20°C, 90% RH and 12 h of light for off-host development. Tick yield of adult females fed on pigs and rabbits was 81.2% and 58.3%, respectively. Females fed on pigs had mean engorgement weight of 862.3 mg and egg mass of 208 mg, while females fed on rabbits had mean engorgement weight of 606.1 mg and egg mass of 160 mg; these values did not differ statistically between host species. Feeding period of female ticks fed on pigs (10 days) was significantly shorter than that on rabbits (17 days). Mean preoviposition period was slightly longer (35.9 days) for ticks fed on pigs than on rabbits (30 days). The minimum incubation period of eggs of ticks from both host species was similar and over 100 days. Egg production efficiency was low for females fed on both hosts (less than 30% and 20% for ticks from pigs and rabbits, respectively). More than 55% of larvae and 79% of nymphs fed on rabbits, set free inside the feeding chambers, engorged successfully. These ticks attained an engorgement weight of 1.3 and 18.2 mg, respectively, and fed for approximately 5 days. The minimum pre-molt period was 30 days for engorged larvae and over 44 days for nymphs. Molting success was low, less than 50% in the case of larvae and less than 20% for nymphs. Further studies are required to better determine the off-host requirements of this tick species.  相似文献

Isoxazolines are a novel class of parasiticides that are potent inhibitors of γ-aminobutyric acid (GABA)-gated chloride channels (GABACls) and l-glutamate-gated chloride channels (GluCls). In this study, the effects of the isoxazoline drug fluralaner on insect and acarid GABACl (RDL) and GluCl and its parasiticidal potency were investigated. We report the identification and cDNA cloning of Rhipicephalus (R.) microplus RDL and GluCl genes, and their functional expression in Xenopus laevis oocytes. The generation of six clonal HEK293 cell lines expressing Rhipicephalus microplus RDL and GluCl, Ctenocephalides felis RDL-A285 and RDL-S285, as well as Drosophila melanogaster RDLCl-A302 and RDL-S302, combined with the development of a membrane potential fluorescence dye assay allowed the comparison of ion channel inhibition by fluralaner with that of established insecticides addressing RDL and GluCl as targets. In these assays fluralaner was several orders of magnitude more potent than picrotoxinin and dieldrin, and performed 5-236 fold better than fipronil on the arthropod RDLs, while a rat GABACl remained unaffected. Comparative studies showed that R. microplus RDL is 52-fold more sensitive than R. microplus GluCl to fluralaner inhibition, confirming that the GABA-gated chloride channel is the primary target of this new parasiticide. In agreement with the superior RDL on-target activity, fluralaner outperformed dieldrin and fipronil in insecticidal screens on cat fleas (Ctenocephalides felis), yellow fever mosquito larvae (Aedes aegypti) and sheep blowfly larvae (Lucilia cuprina), as well as in acaricidal screens on cattle tick (R. microplus) adult females, brown dog tick (Rhipicephalus sanguineus) adult females and Ornithodoros moubata nymphs. These findings highlight the potential of fluralaner as a novel ectoparasiticide.  相似文献
