Sublethal effects seen amongstRhipicephalus appendiculatus feeding on ivermectin-treated rabbits were diverse and dependent both on drug dose, pharmacokinetics and tick feeding patterns: changes in drug formulation, the time of infestation relative to treatment, and the tick instar used, profoundly influenced acaricidal activity. Death was a sequel to paralysis only if tick feeding was interrupted for sufficient time to produce irreversible dehydration. Concurrent pharmacokinetic investigations revealed that, for the larvae ofR. appendiculatus, the mean critical lethal dose of ivermectin imbibed over a 5-day engorgement period was 3500 g/kg. This quantity of ivermectin was achieved in the blood-meals of larvae feeding on rabbits treated subcutaneously with a single dose of Ivomec injection (MSD)*800 g/kg, provided infestation took place within 24 h of treatment. At lower drug doses, or if larval infestations were delayed for>24 h post-treatment, the quantity of circulating ivermectin (and thus imbibed by the tick larvae) fell below 3500 g/kg and an increasing percentage of larvae successfully engorged and detached. More than 90% of such larvae moulted to the nymphal stage. Nymphae and larvae exhibited similar susceptibility to ivermectin on treated rabbits which could be explained by similar feeding patterns. However, adult female and male ticks were markedly less susceptible and interpretation of ivermectin-induced effects was more complex.  相似文献

The effectiveness of a single treatment with either ivermectin or moxidectin was determined by administering a single subcutaneous injection of each endectocide at 200 g per kg body weight to cattle infested with all parasitic developmental stages (adults, nymphs, and larvae) of Boophilus microplus (Canestrini). The percentage reduction in the number of females that reached repletion following treatment (outright kill) was 94.8 and 91.1% for ivermectin and moxidectin, respectively. In addition, the reproductive capacity of the females that did survive to repletion was reduced by >99%, regardless of the endectocide. Based on these two factors, the therapeutic level of control obtained against ticks on the cattle at the time of treatment was 99.0 and 99.1% for ivermectin and moxidectin, respectively. Engorged females recovered from either group of treated cattle weighed 3-times less than untreated females, and the egg masses produced by treated females weighed 5–8-times less than egg masses produced by untreated females. Partitioning of data into three separate 7-d post-treatment intervals allowed for an estimation of the efficacy of each endectocide against each individual parasitic development stage (adult, nymph, and larva). Results indicated that both endectocides were 99.7% effective against ticks that were in either the adult or nymphal stage at the time of treatment. However, the level of control against ticks in the larval stage of development at treatment was significantly lower at 97.9 and 98.4% for ivermectin and moxidectin, respectively. Analysis of the persistent (residual) activity of the two endectocides indicated that neither material provided total protection against larval re-infestation for even 1-wk following treatment. Against larvae infested 1–4 wk following treatment, the level of control with moxidectin ranged from 92.4% (1 wk) to 19.5% (4 wk). These control levels were higher at each weekly interval than for ivermectin, which ranged from 82.4% (1 wk) to 0.0% (4 wk). The potential for the use of these injectable endectocide formulations in the US Boophilus Eradication Program is discussed.This paper reports the results of research only. Ivermectin administered orally to Spanish goats, Capra hircus (L.), or to white-tailed deer, Odocoileus virginianus (Zimmerman), was highly effective against lone star ticks, Amblyomma americanum (L.). For Spanish goats, daily oral doses of 20 micrograms/kg resulted in greater than or equal to 2 ppb ivermectin in the blood. This level was sufficient to cause greater than 95% reduction of estimated larvae from feeding ticks. A bioassay with horn flies, Haematobia irritans (L.), was developed to estimate oral intake of ivermectin. Probit analysis of dose-mortality data indicated that a 50% reduction in adult horn fly emergence can be expected when the manure from goats treated orally with ivermectin at 10, 20, 35, and 50 micrograms/kg/d was mixed with untreated cow manure at a rate of 0.345, 0.110, 0.100, and 0.092%, respectively. In studies with white-tailed deer, daily oral doses of 35 and 50 micrograms/kg/d provided 100% control of adult and about 90% control of nymphs that were placed on treated fawns. A single oral dose of 50 micrograms/kg gave greater than 90% control of adult and nymphal ticks attached to treated fawns at the time of drug administration and 70% control of ticks placed on treated deer three days thereafter. When ticks were placed on fawns treated with a single dose of ivermectin (50 micrograms/kg) the engorgement period was longer, ticks were lighter in weight, and females laid fewer eggs than ticks detaching from control fawns. A single oral dose of ivermectin at 20 micrograms/kg prevented about 60% of the adult and nymphal ticks attached at the time of drug administration from engorging, but did not affect other ticks placed on the animals after treatment.  相似文献

The efficacy of an injectable microsphere formulation of ivermectin for control of the cattle tick, Boophilus annulatus (Say), was tested on 2 groups of 6 Hereford heifers held on separate 7-ha, tick-infested, buffel grass pastures. Cattle in one pasture were injected subcutaneously in the neck with a controlled-release microsphere formulation of ivermectin at the rate of 2.4 mg AI/kg body weight; the other group was injected with carrier only. Beginning 4 wk after injection and continuing throughout the remainder of the test (16 wk), no engorged ticks (> or = 5.5 mm) were found on any of the treated cattle, whereas large numbers of engorged ticks were found on the untreated controls. During this period, a few ticks were recovered from untreated sentinel animals placed in the treatment pasture during 7-8 wk after treatment, but none were recovered from animals exposed from 11-12 wk or 14-15 wk. Large numbers of B. annulatus ticks were found on untreated sentinel cattle placed in the control pasture during these same periods. Although the cattle, pastures, and tick habitat were approximately equal, the treated cattle gained an average of 77 kg compared with an average of 42 kg for the control group. This technology offers a possible alternative to the current official program of dipping and vacating pastures for eradication of Boophilus sp. infestations from the quarantine zone in southern Texas. Larger scale testing is needed to determine the potential of the injectable microsphere formulation and to optimize its use in eradication or control strategies.  相似文献

The efficacy of ivermectin, released intraruminally from a 28-day-delivery device was evaluated in two titration studies against induced infestations of adultRhipicephalus appendiculatus, R. evertsi andHyalomma truncatum on cattle. Cattle were given a sufficient number of devices to release ivermectin at approximately 20, 40, 60 or 80 g kg–1 day–1 at a steady-state rate 7–28 days after administration. Tick mortality was recorded, engorged female ticks were weighed and individually incubated, and reproductive data were recorded to determine a reproductive index for the species at various dose levels. Mortality of male and female ticks compared to that of controls was directly related to the daily dose of ivermectin, as was the number of ticks not engorging. Ticks fed on ivermectin-treated cattle had a smaller mass when engorged and laid smaller egg-masses, both absolutely and as a proportion of engorged mass.The index of reproduction ofR. appendiculatus was reduced by more than 99.9% at 20 g kg–1 day–1, and the reproductive indices ofR. evertsi andH. truncatum were reduced by more than 99.9% at dose rates of 40 g kg–1 day–1 and above.Practical implications of the application of sustained-release ivermectin for the control of multihost ticks and tick-borne diseases are discussed.  相似文献

Transgenic plants of lettuce cultivar (cv.) South Bay were produced by using Agrobacterium tumefaciens vectors containing the -glucuronidase (GUS) reporter gene and the NPT II gene for kanamycin resistance as a selectable marker. High frequency of transformation, based on kanamycin resistance and assays for GUS expression, was obtained with 24 to 72-h-old cotyledon explants cocultivated for 48 h with Agrobacterium tumefaciens. After the cocultivation period, the explants were placed in selection medium containing 50 or 100 mg l–1 of kanamycin, 100 mg l–1 cefotaxime and 500 mg l–1 carbenicillin for 10 days. Surviving explants were transferred every 14 days on shoot elongation medium. Progenies of R0 plants demonstrated linked monogenic segregation for kanamycin resistance and GUS activity.Florida Agricultural Experiment Station Journal Series R-02231. Two field experiments were conducted to evaluate the efficiency of Rusa deer in the development of the cattle tick Boophilus microplus in comparison with that of steers in the same pastures and under the same conditions of infestation. No difference was noted between a mixed steer/deer herd and a pure steer or pure deer herd in the infestation pattern of each host, suggesting that attachment to the alternative host is mechanical and not affected by the simultaneous presence of the primary host on the pasture. Deer are capable of producing engorged viable females, with weight and reproductive performances similar to or even better than females fed on steers. For moderate levels (1 million larvae per hectare) and high levels (32 million larvae per hectare) of pasture infestation, tick burdens on steers were not very different (e.g. average 1911 and 2681ticks per m2 skin, respectively, on day 24). This may be because of saturation of steer skin sites at the moderate larval dose. Deer harboured 2.7–33 times fewer ticks than steers and produce no engorged females at the moderate larval level and 32 times fewer engorged females than steers at the high larval level. Infestation of deer was dosedependent with averages of 12 and 399ticks per m2 skin on day 25 at the moderate and high larval levels, respectively. At a high infestation level of the environment, Rusa deer may contribute, but to a limited extent, to infestation of pastures and, consequently, of cattle. However, their role in sustaining a viable tick population requires further investigation.  相似文献

Effects of azadirachtin, a triterpenoid extracted from neem seed, Azadirachta indica A. Juss., were similar to those of insect growth regulators against the immature stages of the born fly, Haematobia irritans (L.), the stable fly, Stomoxys calcitrans (L.), and the house fly, Musca domestica L. When an ethanolic extract of ground seed was blended into cow manure, LC50 and LC90's for larval horn flies were 0.096 and 0.133 ppm azadirachtin, respectively. An emulsifiable concentrate (EC) had an LC50 for larval horn flies of 0.151 ppm and an LC90 of 0.268 ppm. For larval stable flies, the EC formulation had an LC50 of 7.7 ppm and an LC90 of 18.7 ppm azadirachtin in manure. Against larval house flies, the LC50 and LC90 were 10.5 and 20.2 ppm, respectively. When the EC formulation was administered orally to cattle at a rate of greater than or equal to 0.03 mg azadirachtin per kg of body weight per day or when ground neem seed was given as a daily supplement of greater than or equal to 10 mg seed per kg body weight, horn fly development in the manure was almost completely inhibited. In contrast, ground seed mixed in cattle feed at the rate of 100-400 mg seed per kg of body weight per day caused less than 50% inhibition of stable flies in the manure.  相似文献

Summary Laboratory experiments on sporangial germination and zoospore activity in Coelomomyces opifexi which utilises a suparlittoral environment are described. Sporangial germination depends upon (a) salinity of the medium used and (b) whether the sporangia were derived from living or deceased larvae. Sporangia from living larvae germinated almost instantaneously in distilled, tap, brackish pond and sea water with a salinity of 4.2 There was only partial germination at a salinity of 17, and none at all in 35 (full sea water). Sporangia from deceased larvae required a conditioning of 7 days or more under moisture at 23°C or 28°C before germination. Sporangia from living and moribund larvae became thick-walled and darker when exposed to a salinity of 8.5 or higher. These, likewise, required a conditioning period for germination. The biological and ecological significance of these observations are discussed.
Zusammenfassung Die in vitro-Versuche an das Keimen und die Zootätigkeit von der supralitteral lebenden spezies Coelomomyces opifexi sind hier beschrieben worden. Das sporenbeheltische Keimen ist abhängig von (a) der Salzhaltigkeit der Umgebung und (b) ob die Sporenbehalter von lebenden oder toten Puppen erhalten sind. Die von lebenden Puppen herstammenden Sporenbehalter keimen sofort in distilliertem, Leitings,- Brack- und 4.2 tigem Salzwasser, nur zum Teil in 17 tigem Salzwasser und gar nicht in Meereswasser (35). Die Wänder der von toten Puppen herstammende Sporenbehalter waren dichter und schwarzer und brauchten mindestens 7 Tage zum keimen in einer feuchten Umgebung von 23° bis zu 28°C. Wenn der Salzgehalt stieg über 8.5, so worden die Wänder beider Arten dichter und schwarzer und brauchten ebenso eine bedingte Periode zum keimen. Die biologische und ekologische Bedeutung dieser Beobachtungen sind diskutiert worden.

Laboratory radioisotope experiments were used to investigate the effects of phytoplankton seasonal succession on the selectivity and clearance rates of a suspension-feeding copepod in two Indiana lakes. Responses to particle size and quality were tested by allowing adult female Diaptomus birgei feeding in natural seston to select between a small (6×7 m) flagellate (Chlamydomonas reinhardii) and a large, poor quality food (heat-killed Carteria olivieri, 22×25 m). Short-term responses were tested in one lake by additional treatments in which copepods were acclimated for 5–6 h in filtered lake water (starved) or natural seston with added Chlamydomonas (enriched). Copepods from both lakes fed selectively on the small live flagellate during the spring bloom of edible phytoplankton but fed selectively on the larger, poor quality particle during the clear water phase when food was scarce. These results are interpreted as an interaction between the concentration-dependent selectivity for high quality foods predicted by optimal diet theory and a perceptual bias for large-sized particles. Selectivity for high-quality food was intermediate and clearance rates were depressed when total phytoplankton abundance was high but dominated by filamentous cyanobacteria. In each experiment copepods also responded to the short-term manipulations by exhibiting weaker discrimination against the poor quality particle in the starvation treatment and stronger discrimination in the enriched treatment. A two-way mixed model ANOVA revealed substantial short term (37%) and seasonal (53%) components to the total variance in selectivity. Clearance rates were also influenced by both phytoplankton succession and the short-term resource manipulations. As expected, clearance rates on the poor quality food were more sensitive to the abundance of alternative foods. These results show how the feeding behavior of a freshwater copepod is modulated by both seasonal and short-term variation in natural food.  相似文献

The effect of a decoction of pulverized aloe leaves (Aloe marlothii) mixed with tap water and administered orally through a gastric tube was determined against ticks on cattle. The decoction was administered to six randomly selected calves seven times at 3-day intervals while the remaining calves received an equivalent amount of tap water only. Five hours after first treatment the calves were infested with Boophilus decoloratus larvae hatched from one gram of eggs. Resultant engorged female ticks were collected, counted and weighed daily. Fertility estimates were determined for 30 engorged female ticks collected from each calf in both the treated and control groups of calves. The treatment had no significant effect on total or daily numbers of engorged females collected per group, nor on their biomasses. Fertility estimates showed the treatment group to yield a marginally higher, yet insignificant, egg laying response (ELR) and reproductive estimate (RE).  相似文献

The beef industry endures major economic losses from a complex of flies that feed on bovine blood and mucus. For cattle on pasture, the most important of these pests are horn flies (Haematobia irritans [L.] [Diptera: Muscidae]) and face flies (Musca autumnalis [Diptera: Muscidae] De Geer). Pasture dragging to spread manure pats has been promoted as a management tactic for these species because their larvae inhabit bovine manure pats, but the efficacy of this practice has not been empirically validated. Spreading pats might promote fly mortality through desiccation or overheating, but these processes are weather-dependent and warrant testing in disparate climates. We evaluated pasture dragging effects while monitoring for weather interactions throughout nine experiment rounds in summers of 2018 and 2020 in Pennsylvania, USA. The manure spreading treatments increased pat surface area up to 300% but failed to significantly reduce emergence of horn flies and face flies as compared to controls. In contrast, precipitation and temperature were significant predictors in fly emergence models. Surprisingly, face fly emergence was significantly elevated in dragged pats twice in 2020. These data call for a reevaluation of pasture dragging as a management technique for horn flies and face flies across a range of climates.  相似文献

The concentration of ivermectin in the serum of Hereford heifers treated with a single Ivomec SR bolus reached a maximum of 8.8 +/- 0.9 ppb at 2 wk post-treatment. The single bolus treatment resulted in <10% mortality of adult horn flies feeding on the blood of the treated animals over the 21-wk trial. Bioassays of the manure from treated cattle showed complete inhibition of development of immature horn flies through week 19 post-treatment. When the trial was repeated using two Ivomec SR boluses/heifer, the concentration of ivermectin in the serum of the treated cattle reached a maximum level of 31.2 +/- 3.9 ppb at week 13 post-treatment. During the first 17 wk of treatment, the use of two boluses/heifer resulted in 96.2 and 81.2% mortality of adult male and female horn flies feeding on the blood of treated animals, respectively. From these studies, we conclude that a single Ivomec SR bolus used as an anthelmintic treatment can be expected to provide significant control of immature horn flies developing in the manure, but not of adults feeding on the treated cattle.  相似文献

The present study in solid-liquid contactors, an attempt has been made to calculate the minimum/critical impeller speed required for complete suspension of solids. A new correlation, involving Reynolds number, modified Froude number, along with the agitation characteristics is proposed. Two llamas (Lama glama) were infested with approximately 20,000 Boophilus microplus larvae each and maintained under controlled conditions to collect the detached adult ticks. A total of 72 engorged and semi-engorged females were recovered with a mean weight of 163.0±70.6mg. Sixty-three (87.5%) of these ticks oviposited and 60 (95.2%) of the egg masses produced larvae. Six of the heaviest females laid 18,838 eggs from which 16,545 (87.8%) hatched. These results show that llamas are able to maintain a population of B. microplus under experimental conditions. Further studies are required to determine the role of llamas as alternative hosts for B. microplus under field conditions.  相似文献

An alkaline 5-phosphodiesterase (5-PDE) from barley (Hordeum distichum) malt sprouts was partially purified by thermal treatment and acetone precipitation to diminish phosphomonoesterase (PME) activity. 5-PDE was purified 40-fold to a specific activity of 30 U mg–1 protein with a final yield of about 32%. With synthetic substrate, the enzyme had an optimum pH of 8.9, maximum activity at 70 °C over 10 min, and a Km of 0.26 mM. The partially purified enzyme was activated by 10 mM Mg2+ up to 168% of the original activity, while Zn2+, Mn2+ and Cu2+ ions, chelating agent (EDTA) and NaN3 (1–10 mM), and 5-ribonucleotides (1–5 mM) were inhibitory. Final enzyme preparation was stable over 8 d at 4 °C), at 70 °C for up to 120 min and without loss of activity over 90 d at –18 °C.  相似文献

A colony ofAmblyomma parvum was started with engorged females collected from cattle in the Province of Salta (25°01 S, 63°56 W), Argentina. The progeny of those ticks were fed on rabbits and the non-parasitic stages maintained at 27±1°C, 83–86% RH in darkness. The life cycle (prefeeding period not evaluated) had a mean duration of 99.6 days. The mean time (days) for the different phases of the cycle were as follows: feeding period of females, 8.0; pre-oviposition period, 5.7; oviposition period, 17.5; minimum incubation period of the eggs, 31.8; feeding period of larvae, 3.2; premoult period to nymphs, 10.9; feeding period of nymphs, 4.7; premoult period to adults, 17.8. The oviposition pattern was typical of an ixodid tick, including a linear relationship between weights of engorged females and the number of eggs laid (r=0.8659). The males increased 18% in weight after feeding on hosts (P<0.01). The mean recovery rates of larvae, nymphs and females were 28.2%, 95.3% and 90.7%, respectively. The nymphs moulting to females were heavier (6.8±0.69 mg) than those moulting to males (3.2±0.29 mg) (P<0.01). A comparison of biological values ofA. parvum with American and non-AmericanAmblyomma species is presented.  相似文献

We tested the effects of the potent acaricides, avermectin B1a (AVM) and 22,23-dihydroavermectin B1 (ivermectin; IVM) when injected directly into partially fed and fully engorged female ticks. When injected into small ticks (Amblyomma hebraeum Koch), neither drug (up to 100 g/kg b.w.) inhibited subsequent engorgement nor affected oviposition latency, weight of total egg mass laid nor viability of laid eggs. At higher concentrations (1000 and 5000 g/kg b.w.), AVM and IVM were markedly toxic. When injected into engorged ticks, both drugs increased oviposition latency, and reduced fecundity at about 75–100 g/kg b.w. Vitellogenesis, as assessed by a spectrophotometric assay of the ovaries, was not inhibited. Also at 50–100 g/kg b.w., AVM and IVM caused paralysis of the abdominal dorso-ventral muscles and the leg muscles. Both drugs, at 7 days post-injection, proved detrimental to salivary gland function in both small and large ticks, but had little effect on salivary gland weight. At concentrations which did not inhibit oviposition (20–50 g/kg b.w.) many of the eggs dried out even though they were kept at high RH. We then demonstrated inAmblyomma americanum, Dermacentor andersoni andD. albipictus that removal of egg wax (by extraction with hexane) induced a marked increase in water permeability. IVM neither increased water permeability ofD. andersoni eggs nor diminished the amount of egg wax deposited on the surface of the eggs, when injected posteriorly through the alloscutum. However, injection of IVM, dimethylsulphoxide (vehicle for IVM) or distilled water through the articulation between the capitulum and scutum (anterior injection), did markedly reduce the wax coating and increased egg permeability. We suggest that anterior injection damages Gené's organ and thus causes the latter effects.  相似文献

The cold-hardiness of Dermacentor marginatus using laboratory-reared offspring of ticks collected in Germany was characterized. Investigations of unfed stages revealed that adult ticks suffered 50% mortality at –10°C after 4–5 months, but larvae and nymphs suffered mortality within few days, whereas –15°C was lethal for all stages within a very short period. Larval hatch and moulting of engorged larvae and nymphs did not occur at 10°C. Embryonic development of eggs with larval hatch was considerably reduced by exposure of eggs to 10°C. Engorged females did not lay eggs at 10°C, the oviposition capability, however, persisted over 6 months at 10°C, 5 months at 5°C, 3 months at 0°C and 2 months at –10°C without substantial decrease of the oviposition capacity or reduction of viable eggs. These results present evidence that unfed adult ticks are the ecoepidemiologically most effective stages, which are capable to tolerate long and extremely cold winters without substantial impairment of the population density. It is also considered that engorged females interrupt their oviposition at low and subzero temperatures delaying it for months and so contribute in bypassing winter conditions. None of the stages survived supercooling indicating that D. marginatus is freeze intolerant. Mean supercooling point (SCP) ranged between –26°C in eggs and –12, 6°C in engorged females. Compared with eggs, the SCP of the other stages was significantly higher. In conclusion, the SCP is considered to have no predictive value in the context with cold-hardiness.  相似文献
