香菇多糖是从香菇菌丝体或子实体中提取纯化得到的高分子葡聚糖,作为香菇主要的生物活性物质,具有提高免疫力、抗病毒、抗氧化和抗真菌等生物活性。本研究对幼菇期、菌褶期、采收期、成熟期和开伞期5个不同发育阶段香菇粗多糖含量、理化性质和体外免疫活性进行比较分析。结果表明,其粗多糖含量、理化性质和体外免疫活性具有明显差异。粗多糖得率呈现先增加后降低的趋势,菌褶期得率最高;粗多糖含量也呈现先增加后降低的趋势,成熟期含量最高;多糖的分子量因发育期不同而有较大差异,发育后期所得粗多糖分子量千万级以上组分的比例增加,分子量百万级和十万级的多糖组分比例降低;不同发育阶段香菇子实体多糖的单糖组成均包含果糖、半乳糖、葡萄糖和甘露糖4种单糖,且都以葡萄糖为主;5个阶段的多糖组分均有较好的体外免疫活性,其中幼菇期粗多糖在浓度500μg/mL时表现出最高的体外免疫活性。本研究探讨了香菇不同发育阶段子实体多糖的理化性质及体外免疫活性,为香菇采收期的确定及香菇多糖的制备与利用提供理论依据。  相似文献

A fluorescent technique has been developed for in situ staining of cellulose. The staining agent is a conjugate of cellulase and fluorescein isothiocyanatc (FITC). Application of this agent does not disturb intercellular or intracellular substances. The technique depends on the specific binding of the fluorescent labeled enzyme to its substrate. The stain has been tested on cell-free noncellulose polysaccharides similar to cellulose and does not stain them. The technique has been used to localize cellulose during the life cycle of Dictyostelium discoideum with results that correspond to previous work using other methods.  相似文献

Cistus creticus ssp. creticus is an indigenous shrub of the Mediterranean area. The glandular trichomes covering its leaf surfaces secrete a resin called “ladanum”, which among others contains a number of specific labdane-type diterpenes that exhibit antibacterial and antifungal action as well as in vitro and in vivo cytotoxic and cytostatic activity against human cancer cell lines. In view of the properties and possible future exploitation of these metabolites, it was deemed necessary to study the geranylgeranyl diphosphate synthase enzyme (GGDPS, EC 2.5.1.30), a short chain prenyltransferase responsible for the synthesis of the precursor molecule of all diterpenes. In this work, we present the cloning, functional characterisation and expression profile at the gene and protein levels of two differentially expressed C. creticus full-length cDNAs, CcGGDPS1 and CcGGDPS2. Heterologous yeast cell expression system showed that these cDNAs exhibited GGDPS enzyme activity. Gene and protein expression analyses suggest that this enzyme is developmentally and tissue-regulated showing maximum expression in trichomes and smallest leaves (0.5–1.0 cm). This work is the first attempt to study the terpenoid biosynthesis at the molecular level in C. creticus ssp. creticus.  相似文献

The objective of this study was to determine the effectiveness of a 42-kDa endochitinase coded by the ThEn42 gene from Trichoderma harzianum as a potential source of transgenic resistance to Rhizoctonia root rot of barley caused by Rhizoctonia solani AG8 and/or R. oryzae. The gene cThEn42 was codon optimized (GC content increased from 53.3 to 65.1%) and then synthesized to produce the modified cThEn42GC in Pichia pastoris for in vitro tests. Two expression vectors were constructed: one with the fungal signal peptide and the fungal activation peptide [FSP-FAP-cThEn(GC)] and the other with barley chitinase 26 signal peptide followed by the fungal signal and activation peptides [SP(HVChi26)-FSP-FAP-cThEn(GC)]. N-terminal sequencing showed that, of two proteins secreted into liquid medium, FSP was cleaved off faithfully in one protein and both FSP and FAP were cleaved from the other protein. Purified endochitinase provided strong in vitro inhibition of both R. solani AG8 and R. oryzae. The enzyme had an intermediate inhibitory activity against Gaeumannomyces graminis var. tritici, and no inhibitory activity against Fusarium graminearum, F. pseudograminearum, and F. culmorum.  相似文献

A commercial enzyme preparation from a selected strain of Penicillium funiculosum has been partially purified using a single stage chromatofocusing fractionation to produce an amylase-free mixture of hydrolytic enzymes. This mixture has been used to remove the non-starch polysaccharides from aqueous extracts of laboratory milled barley. The structure of the resulting purified α-glucan fraction has been examined by gel filtration before and after enzymic debranching and by iodine staining. The mild damage caused to the starch within the barley kernel releases a low molecular weight amylopectin molecule with no detectable amylose in the product. In this respect the product is different from that produced after severe, direct grinding of the purified barley starch where some amylose appears in the water soluble extract. Although the small amount of α-glucan is not of any quantitative industrial significance in itself, it does reflect the extent and type of physical damage which is taking place in the starch granule. The model proposed to explain these results - a starch granule with a solid amylose/amylopectin core but with a number of patches of protruding amylopectin clusters - may have important implications in an industrial context.  相似文献

为探讨常压室温等离子体诱变的3株高产多糖猴头菌和出发菌株的多糖组分差异,通过液体发酵获得的菌丝体经水提、分级醇沉获得8个胞内多糖组分,对它们的理化性质、结构特征及体外免疫活性进行了研究。结果表明,3株ARTP诱变菌株414、321、236菌丝体多糖含量较出发菌株有较明显提升;ARTP诱变的猴头菌20%醇沉多糖组分较出发菌株分子量大,所占比例增加;诱变菌株60%醇沉多糖组分的分子量略大于出发菌株,所占比例相近。20%醇沉多糖主要由半乳糖、葡萄糖、甘露糖构成,诱变菌株该多糖组分中葡萄糖和甘露糖的比例较出发菌株均有明显提升,60%醇沉多糖组分单糖组成无明显差异;8个多糖组分均具有体外刺激巨噬细胞释放NO的活性,其中20%醇沉多糖的活性优于60%醇沉多糖,诱变菌株的生物活性优于出发菌株。本研究探讨了ARTP诱变对猴头菌胞内多糖结构及活性的影响,为猴头菌相关产品的开发提供了优质资源。  相似文献

Enzymes are widely applied in various industrial applications and processes, including the food and beverage, animal feed, textile, detergent and medical industries. Enzymes screened from natural origins are often engineered before entering the market place because their native forms do not meet the requirements for industrial application. Protein engineering is concerned with the design and construction of novel enzymes with tailored functional properties, including stability, catalytic activity, reaction product inhibition and substrate specificity. Two broad approaches have been used for enzyme engineering, namely, rational design and directed evolution. The powerful and revolutionary techniques so far developed for protein engineering provide excellent opportunities for the design of industrial enzymes with specific properties and production of high-value products at lower production costs. The present review seeks to highlight the major fields of enzyme application and to provide an updated overview on previous protein engineering studies wherein natural enzymes were modified to meet the operational conditions required for industrial application.  相似文献

Chemical modifications of galactomannans are applied to improve and/or modify their solubility, rheological and functional properties, but have limited specificity and are often difficult to control. Enzymatic reactions, catalyzed under mild process conditions, such as depolymerization, debranching and oxidation, represent a viable and eco-friendly alternative. In this study, we describe oxidation of guar galactomannan primary hydroxyl groups by a fungal laccase using the stable radical TEMPO as mediator. Four fungal laccases were investigated from: Trametes versicolor, Myceliophthora thermophila, Thielavia arenaria, Cerrena unicolor. The laccase from T. versicolor was found to efficiently oxidize TEMPO and to be free of mannanase side activity. Oxidation of galactomannan with this enzyme plus TEMPO brought about a ten-fold increase in viscosity of a guar galactomannan solution and altered its rheological profile, by converting a viscous polysaccharide solution into an elastic gel. This structural modification is presumably due to formation of inter-chain hemiacetalic bonds between newly generated carbonyl groups and free OH groups, yielding a cross-linked gel. These findings could be of practical importance, considering that polysaccharides with high viscosity, gelling and elastic properties can find interesting and novel applications as thickeners, viscosifiers and emulsion stabilizers in several industrial applications such as: personal care, oil operations, paper coating, paints, construction and mining.  相似文献

Over the past few decades, high-throughput screening (HTS) has made great contributions to new drug discovery. HTS technology is equipped with higher throughput, minimized platforms, more automated and computerized operating systems, more efficient and sensitive detection devices, and rapid data processing systems. At the same time, in vitro neurogenesis is gradually becoming important in establishing models to investigate the mechanisms of neural disease or developmental processes. However, challenges remain in generating more mature and functional neurons with specific subtypes and in establishing robust and standardized three-dimensional (3D) in vitro models with neural cells cultured in 3D matrices or organoids representing specific brain regions. Here, we review the applications of HTS technologies on in vitro neurogenesis, especially aiming at identifying the essential genes, chemical small molecules and adaptive microenvironments that hold great prospects for generating functional neurons or more reproductive and homogeneous 3D organoids. We also discuss the developmental tendency of HTS technology, e.g., so-called next-generation screening, which utilizes 3D organoid-based screening combined with microfluidic devices to narrow the gap between in vitro models and in vivo situations both physiologically and pathologically.  相似文献

A rapid method for establishing the constants in the Mark–Houwink–Sakurada equation, relating intrinsic viscosity and molecular weight (MW), of guar galactomannan is described. Following partial acid hydrolysis, the galactomannan was analyzed using high performance size exclusion chromatography employing viscosimetry and right angle light scattering detectors. In this way, a large number of samples of polysaccharides with a wide range of MW distributions were prepared, without need for isolation, and intrinsic viscosity and MW rapidly determined. The a and K values found for guar galactomannan were 0.72 and 5.13×10−4 ([η] in dl/g) respectively, in good agreement with previously published values.  相似文献

The saccharification of the polysaccharides of barley, oat, and wheat straws and Solka Floc was studied using the extracellular enzyme system synthesized by mutant strain NTG III/6 of the fungus Penicillium pinophilum 87160iii. The enzymes obtained in cultures containing Solka Floc or barley straw as the carbon source were compared. Solka Floc at 10% (w/v) concentration was hydrolyzed to the extent of 70% in 72 h at 50 degrees C using a reaction mixture containing 7 filter paper units/mL of cellulase induced on Solka Floc, but hydrolysis was increased to 90% when the enzyme induced on barley straw was used. Under the same conditions, the polysaccharides in barley, oat, and wheat straws were hydrolyzed, respectively, in 72 h, to the extent of 42-48%, 62%, and 52%, but hydrolysis was increased to 93%, 100%, and 92%, respectively, after treatment of the substrates with alkaline-H(2)O(2) reagent at room temperature.  相似文献

The activities of superoxide dismutase, catalase and glutathione reductase were not affected by in vitro incubation with the intracellular proteinase calpain, suggesting that these enzymes are not in vivo substrates of calpain. In contrast, the activity of another important antioxidant enzyme, glutathione peroxidase, is stimulated in vitro by calpain. This may explain the correlation between elevations in glutathione peroxidase activity and calpain activity which occur in aging, exercised and dystrophic muscle. Calpain treatment in vitro caused a large decrease in the activity of carnosine synthetase which is involved in the synthesis of the putative antioxidant carnosine. This may be the reason for the in vivo correlation between elevated calpain and diminished carnosine levels in aging, hypertensive, denervated and dystrophic muscles.  相似文献

猴头菌不同发育阶段产生的多糖结构特征及免疫活性   总被引:1,自引:0,他引:1  
采收猴头菌Hericium erinaceus 7个不同发育阶段的子实体,经热水浸提后分别采用20%、50%、70%的乙醇终浓度进行分级沉淀,获得21个多糖组分,对它们的结构特征及体外免疫活性进行了研究。结果表明,在猴头菌发育过程中,多糖总得率呈现先增大后减小的趋势,在第5阶段达到最大值0.92%。20%醇沉的多糖含量也呈先增大后减小的趋势,在第5阶段达到最大值42.87%,其大分子量多糖(1 000-5 000kDa)所占比例最高。50%、70%醇沉多糖均为小分子量多糖,约为10-40kDa。获得的猴头菌多糖组分中,单糖组成多以岩藻糖、半乳糖、葡萄糖和甘露糖为主(相对比例存在一定差异);另外,第1阶段20%、70%醇沉多糖还含有少量的核糖,第7阶段20%醇沉多糖含有一定量的鼠李糖;7个时期50%醇沉多糖均含有一定量的葡萄糖醛酸。所得多糖样品均具有刺激巨噬细胞释放NO的活性,其中20%醇沉多糖的活性优于50% 和70% 醇沉多糖,在50μg/mL时就表现出显著的体外免疫活性。此外,在第5阶段即中菌刺期产生的多糖活性最优,说明在此阶段采摘可以获得最佳的猴头菌多糖原材料。本研究为猴头菌生长发育过程中活性多糖的动态形成研究提供了一定的理论基础。  相似文献

本研究旨在探讨奶油栓孔菌子实体多糖(TLFPS)的化学性质和酒精性肝损伤的保护作用。首先用水提醇沉法提取得到多糖,通过化学组成分析、紫外吸收光谱法、傅里叶红外光谱法和刚果红染色法对多糖结构进行初步表征,以DPPH、ABTS、超氧阴离子自由基的清除能力和铁离子还原能力为指标评价了多糖体外抗氧化能力,在小鼠急性肝损伤之前连续灌喂多糖8d,比较各组小鼠血清和肝的相关生化指标以及组织病理切片来确定多糖对酒精性肝损伤小鼠的保护作用。结果显示:多糖具有β-吡喃糖环结构,含有较高含量的糖醛酸和硫酸根基团,空间构型不具备三股螺旋结构。在体外抗氧化活性方面,多糖对DPPH、ABTS和超氧阴离子自由基均有良好的清除活性,铁离子还原能力也呈良好的剂量依赖性。在小鼠保肝实验中,与模型组相比,多糖能够显著延长小鼠的醉酒时间和缩短醒酒时间,降低了酒精性肝损伤小鼠的肝指数,并且显著降低血清中谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)、甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)和肝脏中丙二醛(malondialdehyde,MDA)的含量,同时明显提高了肝脏中超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase,CAT)的活性。结果证实奶油栓孔菌子实体多糖具有良好的抗氧化能力,可以减轻由酒精引起的急性肝细胞损伤,而且对机体的毒害作用很小。肝组织病理切片也进一步证实了奶油栓孔菌多糖的保肝活性。研究结果不仅丰富了奶油栓孔菌的药用价值,而且对多糖在功能食品领域的应用提供了药效基础。  相似文献

Mounting evidence has emphasized the potential of cell therapies in treating various diseases by restoring damaged tissues or replacing defective cells in the body. Cell therapies have become a strong therapeutic modality by applying noninvasive in vivo molecular imaging for examining complex cellular processes, understanding pathophysiological mechanisms of diseases, and evaluating the kinetics/dynamics of cell therapies. In particular, mesenchymal stem cells (MSCs) have shown promise in recent years as drug carriers for cancer treatment. They can also be labeled with different probes and tracked in vivo to assess the in vivo effect of administered cells, and to optimize therapy. The exact role of MSCs in oncologic diseases is not clear as MSCs have been shown to be involved in tumor progression and inhibition, and the exact interactions between MSCs and specific cancer microenvironments are not clear. In this review, a multitude of labeling approaches, imaging modalities, and the merits/demerits of each strategy are outlined. In addition, specific examples of the use of MSCs and in vivo imaging in cancer therapy are provided. Finally, present limitations and future outlooks in terms of the translation of different imaging approaches in clinics are discussed.  相似文献

We investigated the protective effects of Glycyrrhizae Radix extract against peroxynitrite (ONOO-)-induced oxidative stress under in vivo as well as in vitro conditions. The extract showed strong ONOO- and nitric oxide (NO) scavenging effects under in vitro system, in particular higher activity against ONOO-. Furthermore, elevations of plasma 3-nitrotyrosine levels, indicative of in vivo ONOO- generation and NO production, were shown using a rat in vivo ONOO--generation model of lipopolysaccharide injection plus ischemia-reperfusion. The administration of Glycyrrhizae Radix extract at doses of 30 and 60 mg/kg body weight/day for 30 days significantly reduced the concentrations of 3-nitrotyrosine and NO and decreased inducible NO synthase activity. In addition, the nitrated tyrosine protein level and myeloperoxidase activity in the kidney were significantly lower in rats given Glycyrrhizae Radix extract than in control rats. However, the administration of Glycyrrhizae Radix extract did not result in either significant elevation of glutathione levels or reduction of lipid peroxidation in renal mitochondria. Moreover, the in vivo ONOO- generation system resulted in renal functional impairment, reflected by increased plasma levels of urea nitrogen and creatinine, whereas the administration of Glycyrrhizae Radix extract reduced these levels significantly, implying that the renal dysfunction induced by ONOO- was ameliorated. The present study suggests that Glycyrrhizae Radix extract could protect the kidneys against ONOO- through scavenging ONOO- and/or its precursor NO, inhibiting protein nitration and improving renal dysfunction caused by ONOO-.  相似文献

The lichenised ascomycete, Collema leptosporum Malme, was extracted with aqueous methanol to give traces of mannitol and 3-O-β- -glucopyranosyl- -mannitol (2.7% yield). The residue was consecutively extracted with hot water to give a complex uronic acid-containing polysaccharide, and then with hot aqueous alkali which provided a mixture of polysaccharides. This was fractionated with Cetavlon to give a branched galactomannan, which had the lowest content of galactose yet reported for such a lichen polysaccharide. It has a main chain of (1→6)-linked -Manp units partly substituted at O-2,4 by non-reducing end-units of Manp and Galp, shown by NMR spectroscopy to have - and β-configurations, respectively. The other polysaccharide component was unexpectedly a branched (1→3), (1→6)-linked β-glucan, which is typical of a basidiomycete, whereas those of ascomycetes contain similar linkages but in linear glucans.  相似文献
