The extent of inhibition of stem elongation and the reducedflowering induced by growth retardants daminozide and chlormequatchloride depends upon the K content of the plants in S. sisymbrifolium.In plants with a low content of K, the decrease in length ofthe internodes and the enhanced development of the lateral shootsinduced by the two growth retardants is greater. In the reproductivephase, a decreased production of inflorescences and a diminutionin the number of flowers borne are accompanied by a slight increasein the number of fully developed flowers. The number of femalesterile flowers, characteristic of the low-K plants, drops significantly.Daminozide and chlormequat chloride not only reverse the morphogeneticeffects of a low level of K but they produce additive effectsin plants with a high K content. The chlorflurecol-methyl-induced elongation of the main axisis greater in the high-K plants but that of the lateral shootsis considerably more in the low-K plants. In the reproductivephase, flowering is slightly reduced but the development ofthe sterile flowers borne by plants with a low content of Kis not affected. Indeed, the effects of chlorflurecol-methylare inconsistent. The significance of these observations and of the results communicatedin Sections I and II of this report is discussed.  相似文献

Aspects of the carbohydrate and nitrogen metabolism of Acerpseudcplatanus, L cell suspension cultures grown on a syntheticmedium containing 2 per cent glucose and 1.0 mg/l 2,4-dichlorophenoxyaceticacid and kinetin either at 0.25 mg/l (low kinetin) or at 2.5mg/l (high kinetin) are described. High kinetin inhibits growthas measured by increase in cell number, packed-cell volume,and cell dry weight. Although not inhibitory to glucose utdization,high kinetin inhibits the O2 uptake of the cells. Such cellscontain only a trace amount of fructose and their rate of O2uptake can be raised to that of the low kinetin cells by a periodof fructose feeding. The O2 uptake of both kinds of cell issensitive to malonate but the stimulation of O2 uptake inducedby bis(hexafiuoroacetonyl)-acetone (‘1799’) at 0.2mM is much less with the high-kinetin than the low-kinetin cells.The enzymes phosphoglucoseiseomerase and glucose-6-phosphatedehydrogenase are much less active in the high-kinetin cells.Mitochondria isolated from both kinds of cells show good respiratorycontrol although slightly lower values for QO2(N), ADP/O ratioand control ratio are recorded with mitochondria from the highkinetin cells. Kinetin at 2.5 mg/l slightly reduces the ADP/Oratio of isolated mitochondria but at 4.0 mg/l their responseto ADP is completely suppressed. Extracellular hemicelluloseformed in presence of high kinetin has a reduced content ofgalactose and xylose and an increased content of glucose. Theseobservations indicate that the inhibition of respiration byhigh kinetin is mainly due to suppression of glucose conversionto other sugars rather than to inhibition of glycolysis or terminalrespiration. High kinetin decreases the rate of protein but not of amino-acidsynthesis. Suppression of the synthesis of particular proteinsmay be an important factor responsible for the reduced cellyield of the cultures in presence of high kinetin. The significanceof these observations to our understanding of the critical metaboliceffects of cytokinina is discussed. Acer pseudoplatanus cells release amino acids into their culturemedium early in the period of batch culture and largely reabsorbthem as incubation proceeds.  相似文献

As compared to a high level of foliar potassium, a low but nota deficient level promotes shoot elongation and flowering inS.sisymbrifolium.This is accompanied by formation of flowers with rudimentaryovaries in which the megagametophyte in the ovules aborts atthe two- or the four-nucleate stage. Plants having a higherK content do not bear the female sterile flowers. Inflorescencesof the high-K plants with 20 per cent fewer buds have 23 percent higher dry weight than inflorescences of low-K plants.The ratio of organic nitrogen to dry weight is not affected,indicating that the inflorescences of the high-K plants arebetter supplied with metabolites in general.  相似文献

Dormancy was induced during storage of Liatris spicata cormsgrown in Israeli summer conditions, but plants left in soilcontinued vegetative growth. Corms of winter-grown plants sproutedfreely. Treatment with GA3 restored both sprouting and floweringin summer-grown corms, but in winter corms GA2 was effectiveonly after corms were stored at low temperature. All the plantsflowered after 4 weeks at 2 °C and GA3 treatment. The content of gibberellins in the main bud of freshly excavatedcorms decreased during the first 18 d of storage but increasedto the initial level after 4 months of cold storage. The numberof flowering stems increased to 2.5 per corm when corms werecold-stored up to 75 d, but decreased with a longer storage. In the stem of Phaseolus vulgaris L. the specific activity ofacid invertase was highest in the most rapidly elongating internode.Activity of the enzyme was very low in internodes which hadcompleted their elongation, in young internodes before the onsetof rapid elongation, and in the apical bud. From shortly afterits emergence from the apical bud the elongation of internode3 was attributable mainly to cell expansion. Total and specificactivities of acid invertase in this internode rose to a maximumat the time of most rapid elongation and then declined. Transferof plants to complete darkness, or treatment of plants withgibberellic acid (GA3), increased the rate of internode elongationand final internode length by stimulating cell expansion. Bothtreatments rapidly increased the total and specific activitiesof acid invertase in the responding internodes; peak activitiesof the enzyme occurred at the time of most rapid cell expansion. In light-grown plants, including those treated with GA3, rapidcell and internode elongation and high specific activities ofacid invertase were associated with high concentrations of hexosesugar and low concentrations of sucrose. As cell growth ratesand invertase activities declined, the concentration of hexosefell and that of sucrose rose. In plants transferred to darkness,stimulated cell elongation was accompanied by a rapid decreasein hexose concentration and the disappearance of sucrose, indicatingrapid utilization of hexose. No sucrose was detected in theapical tissues of light-grown plants. The results are discussed in relation to the role of acid invertasein the provision of carbon substrates for cell growth. Seedlings of Pharbitis nil, strain Kidachi, were grown undercontinuous light at 20°C in vessels containing 5,000-mlnutrient solution, 24 plants per vessel. NAA (0.005–0.5µM), GA3 (0.1–0.5 µM), kinetin (0.5–5µM), benzyladenine (0.05–5 µM) or abscisicacid (4 µM) added to the nutrient solution induced long-dayflowering, and the flowering was always accompanied by suppressionof root elongation. 3,4-Dichlorobenzoic acid (0.05–10µM) and some other benzoic acid derivatives which arehighly effective for the induction of flowering in Lemna paucicostataalso showed similar effects. Neither NAA, kinetin nor 3,4-dichlorobenzoicacid applied via the apical part of the hypocotyl could causeflowering or suppression of root elongation. Thus, the flower-inducingeffect of the above substances was presumed to be secondaryto the suppression of root elongation. Phaseolus multiflorus plants at three stages of developmentwere decapitated either immediately below the apical bud orlower down at a point 1 cm above the insertion of the primaryleaves. Growth regulators in lanolin were applied to the cutstem surface. IAA always inhibited axillary bud elongation anddry-matter accumulation, and enhanced internode dry weight butnot elongation. GA3 applied below the apical bud greatly increasedinternode elongation and dry weight, but simultaneously reducedbud elongation and dry-weight increase. Application of GA3 1cm above the buds had no effect on bud elongation in the youngestplants, but enhanced their elongation in the two older groups.IAA always antagonized GA3-enhancement of internode extensiongrowth, whereas its effects on GA3-enhanced dry-matter accumulationdepended on the stage of internode development. Bud elongationwas greater in plants treated with GA3+IAA than in plants treatedonly with IAA, except in the youngest plants decapitated immediatelybelow the apical bud, where GA3 caused a slight increase inIAA-induced bud inhibition. GA3 increased inhibition of buddry weight by IAA in the two youngest groups of plants, butslightly reduced it in the oldest plants. No simple compensatorygrowth relationship existed between internode and buds. It wasconcluded that, (1) auxin appears to be the principal growthhormone concerned in correlative inhibition, and (2) availabilityof gibberellin to internode and buds is of importance as a modifyingfactor in auxin-regulated apical dominance by virtue of itslocal effects on growth in the internode and in the buds.  相似文献

Interactions between three growth retardants and gibberellinA9 (GA9) in Chrysanthemum morifolium Ramat. cv. Bright GoldenAnne grown in short days have revealed that the particular retardantemployed modifies the response to this gibberellin. When plants were given a foliar spray of daminozide (a substitutedsuccinamic acid), the effects of a subsequent application ofGA9 to each lateral shoot, on both stem length and the rateof inflorescence development, were relatively small. However,GA9 was highly active on plants previously sprayed with piproctanylbromide (a quaternary ammonium, piperidinium compound) or onthose given a compost drench of chlorphonium chloride (a quaternaryphosphonium retardant). In the absence of GA9 all three retardantsreduced stem length and delayed flowering. Irrespective of the extent of the responses to various dosesof GA9 (1–50 µg per shoot), a close relationshipwas maintained between internode extension and earliness offlowering, as in previous studies with retardants and othergibberellins. On plants which had received chlorphonium chlorideGA3 and GA13 were highly active and poorly active, respectively. The data suggest that daminozide could act as a retardant notonly by blocking the biosynthesis of gibberellins but also,wholly or partly, by restricting the metabolism or action ofendogenous gibberellins.  相似文献

Lines representing a range of internode length and floweringgenotypes in Pisum sativum L. were grown in 8 h of daylightfollowed by either 16 h of darkness or incandescent light. Thestem elongation response index (RI = length in 24 h ÷length in 8 h) was least in the very short internode nana types,which are grossly deficient in gibberellins (GAs), and the verylong internode slender types, which behave as if saturated withGAs. The common tall (genotype Le) and dwarf (le) types (lepartially blocks conversion of GA20 to the active form, GA1)were all markedly responsive but the peak RI (based on the mostresponsive internode) was less in tall lines (1.79 to 2.78)than in dwarf lines (2.32 to 5.01) and the peak RI tended tooccur about three to four internodes earlier in tall than indwarf lines. The cry8 mutation reduced the RI. (Duplicate lengthloci La and Cry are probably concerned with GA reception.) Amongle dwarf lines, genotype La cry8, was generally less responsivethan La Cry, La cryc and la Cry. Data from crosses showed thaton either an le La or le la background cry8 segregates had alower RI than cry8 segregates. On an le la background, cry8plants were shorter than cryc plants, cry8 was partially dominantto cry8 and segregation was clear only in long days. On an lela background, cryc plants were shorter than cryc plants, cry8was partially dominant to cry8 and segregation was clear inlong or short days. The very high peak RI (5.0) of the microcryptodwarfline, L57, appeared to result, in part, from a marked foreshorteningof internodes 4 to 10 in the 8 h regime. In the 24 h regimeL57 (lm) had a fairly similar growth pattern to normal (Lm)cryptodwarf types. The peak RI tended to occur at a lower internode in early thanlate flowering lines, especially among dwarf types, and genotypeswith a day neutral flowering habit (genotype sn or dne) wereless responsive than their photoperiodic counterparts (Sn Dne). Abscisic acid (ABA) at 1 x 10–4 M or 3 x 10–4 Mwas applied to the apical buds of Chenopodium rubrum plantsexposed to different photoperiodic treatments and showing differentpatterns of floral differentiation. Stimulation of growth inwidth of the apical meristem of the shoot and/or inhibitionof growth in length was obtained under all photoperiodic treatments.This change of growth pattern was followed by different effectson flowering. In non-induced plants grown under continuous light ABA stimulatedpericlinal divisions in the peripheral zone and the initiationof leaves as well as the growth in width of bud primordia. Inplants induced by two short days reduced growth of the meristemcoincided with ABA application. Longitudinal growth of the meristemwas inhibited in this case and only a temporary stimulationof inflorescence formation took place. In plants induced ata very early stage, ABA exerted a strong inhibitory effect onflowering. A permanent and reproducible stimulatory effect onflowering was obtained in plants induced by three sub-criticalphotoperiodic cycles if ABA was applied to apices released fromapical dominance. In this case formation of lateral organs andinternodes was promoted by ABA and was followed by stimulatedinflorescence formation. Gibberellic acid (GA2) at 1x 10–4M or 3 x 10–4 M brought about a similar effect on floweringas ABA, although the primary growth effect was different, i.e.GA2 stimulated longitudinal growth. The effects of ABA and GA2 on floral differentiation have beencompared with earlier results obtained from auxin and kinetinapplications. These growth hormones are believed to regulateflowering by changing cellular growth within the shoot apex.Depending on the actual state of the meristem identical growthresponses may result in different patterns of organogenesisand even in opposite effects on flowering. Exogenous gibberellin A3(GA3) reduced the number of leaf nodesat flowering and time to flowering and increased the stem heightat flowering in three genotypes of spring rape (Brassica napusvar.annua L.). The responses to GA3were similar to those forlong days (LD) and low-temperature treatments, suggesting thatthe effect of photoperiod and the vernalization response areprobably mediated through gibberellins. The response to exogenousGA3was greatest in non-cold-treated plants in short days (SD)suggesting that endogenous GAs are limiting in these conditions.CCC, an inhibitor of gibberellin biosynthesis, caused a smallincrease in the number of leaf nodes at flowering and time toflowering and a small decrease in the stem height at flowering,but unexpectedly, its effect was hardly influenced by the applicationof exogenous GA3. Genotypes that showed the clearest responsesto the treatments with regard to the number of leaf nodes atflowering and time to flowering did not show the clearest responseswith regard to the stem height at flowering; the pattern ofresponses of the number of leaf nodes at flowering and timeto flowering was distinct from that of stem height at flowering.This indicates that flower formation and stem elongation areseparable developmental processes which may be controlled bydifferent endogenous gibberellins, different levels of a specificendogenous gibberellin, or different responses to gibberellin.Copyright 1999 Annals of Botany Company Brassica napus var. annua, gibberellin, photoperiod, spring rape, vernalization.  相似文献

The addition of 40 mM putrescine (Put) to Murashige and Skoog's (MS) medium resulted in increased shoot multiplication and shoot growth in untransformed plants relative to transformed plants of Cichorium intybus L. Put at a concentration of 40 mM also resulted in flowering in both systems on the 28th day, with elevated titers of endogenous conjugated Put and spermine (Spm) in both untransformed and transformed plants. The addition of 40 µM AgNO3 to untransformed axillary buds of C. intybus L. cultured on MS media resulted in increased shoot multiplication (36.9DŽ.63 shoots per culture) and increased shoot growth (7.82ǂ.76 cm) as compared to transformed ones (11.6ǂ.89 shoots per culture; 3.20ǂ.24 cm). Moreover, cultures treated with 40 µM AgNO3 showed in vitro flowering on the 28th day in both systems, with the endogenous levels of conjugated spermine being higher in untransformed plants than in transformed ones. The morphogenetic response and the endogenous conjugated pool of polyamines were lower following !-DL-difluromethylarginine and !-DL-difluromethylornithine treatments; the addition of put (40 mM) and AgNO3 (40 µM) restored these to normal levels. Under exogenous put feeding, ethylene production was lower in both the untransformed and transformed cultures. We believe that an interplay between polyamine and ethylene biosynthesis is involved in regulating the morphogenetic response in both transformed and untransformed shoots of C. intybus. Illus. 1963.—Dry matter accumulation of aerial parts, and heights of tomato, cucumber, and pea plants were markedly reduced when kinetin was incorporated into the culture solution root medium in concentrations ranging from 10–5 to 10–7 M. Concentrations which suppressed top growth (height, dry weight) generally had lesser effects on root growth and, in some instances, enhanced it. Thus top/root ratios were greatly reduced and approached unity in kinetin-treated peas and tomatoes. Flowering was inhibited in tomatoes and accelerated in peas. There were marked changes in root morphology, including the formation of pseudonodules. Kinetin had an effect which was opposite to that of gibberellin on internode elongation, root extension, top/root ratios and flowering of dwarf peas. N6-benzyladenine was more active than kinetin in suppressing the growth of intact green plants. The data show that kinetin can markedly alter the behavior of intact plants when absorbed by the roots from culture solutions in which the concentrations are comparable to those which are biologically active on explants.  相似文献

The carbon balance and changes in leaf structure in Clusia minorL., were investigated in controlled conditions with regardto nitrogen supply and responses to low and high photosyntheticallyactive radiation (PAR). Nitrogen deficiency and high PAR ledto the production of smaller leaves with higher specific leafdry weight (SLDW) and higher leaf water content, but with lowerchlorophyll content. Nitrogen and PAR levels at growth alsoaffected CO2 exchange and leaf area. In – N conditions,total daily net CO2 uptake and leaf area accumulation were slightlyless for high-PAR-grown plants. In contrast, high-PAR-grownplants supplied with nitrogen showed about a 4-fold higher totaldaily CO2 uptake and about twice the total leaf area of low-PAR-grownplants. Although total daily net CO2 uptake of +N plants wasonly slightly higher than –N plants under the low PARlevel, –N plants produced almost three times more leafarea but with lower SLDW. Under well-watered conditions, low-PAR-grownplants showed only CO2 evolution during the night and malicacid levels decreased. However, there was considerable night-timeaccumulation of titratable protons due to day/night changesin citric acid levels. High-PAR-grown plants showed net CO2uptake, malate and citrate accumulation during the dark period.However, most of the CO2 fixed at night probably came from respiratoryCO2. Positive night-time CO2 exchange was readily observed forlow-PAR-grown plants when they were transferred to high PARconditions or when they were submitted to water stress. In plantsgrown in high and low PAR, CAM leads to a substantial increasein daily water use efficiency for water-stressed plants, althoughtotal net CO2 uptake decreased.  相似文献

The growth response of Tapinanthus bangwensis (Engler and K.Krause) Danser to exogenous growth substances, indole-3yl-aceticacid (IAA), kinetin, and gibberellic acid (GA3), not only varieswith the concentrations of each of these substances in the mediumbut follows a similar trend in each case. Kinetin, IAA, andGA3 were found to enhance growth but not by very appreciableamounts, kinetin being the most effective growth promoter. Highconcentrations of these substances (5.0 and 10.0 ppm) exceptGA3 were toxic to the parasite. Gibberellic acid induced theproduction of accessory leaves at all the concentrations used.Kinetin was also effective in inducing accessory leaves butonly at a concentration of 1.0 ppm. Some of the lower concentrationsof the growth substances affected significantly some aspectsof the parasite's growth and development. Nevertheless, thegrowth in vitro was still slower than that in vivo.  相似文献

Kinetin at a concentration from 3.10-6 M to 1.10-3 M was applied to the plumule ofChenopodium rubrum plants during photoperiodic induction. Different levels of induction were compared (one and three short days). The higher concentrations of kinetin applied to induced plants inhibited flower formation. The rate of leaf initiation was increased under these treatments. Lower concentrations of kinetin (from 3.10-6 M to 1.10-5 M) usually promoted lateral bud formation and flowering. The step-wise application of kinetin revealed that the inhibitory effect on flowering had been restricted to the inductive period. The effects of kinetin, benzyladenine and trans-zeatin were compared in plants partially induced by two short days. High concentrations always inhibited flowering. Benzyladenine was the most effective in this respect. Root removal diminished the inhibitory effects of cytokinins on flowering as was stated with benzyladenine. It is assumed that endogenous cytokinins play a role in the regulation of organogenetic activity of the stem apical meristem. Depending on the photoperiodic conditions, they presumably exert their activity by maintaining the vegetative functions of the apex.  相似文献

The role of photosynthesis in flower induction in the short-dayplants Kalanchoe blossfeldiana and Xanthium pensylvanicum wasinvestigated by chemical suppression of photosynthesis and preventionof chlorophyll formation in the induced leaf. ‘Bleaching’leaves with streptomycin completely prevented flowering in X.pensylvanicum at concentrations shown to reduce the chlorophylland carotenoid content of the leaf significantly. Such leaveswere unable to induce flowering even when supplied with sugarsand other photosynthetic products. Photosystem II inhibitors,DCMU and cadmium ion, inhibited induction in both species aswell as suppressing photosynthesis (as tested by O2 evolutionand starch production) whereas the photosystem I inhibitor,metronidazole, had no effect. Antimycin A inhibited floweringin K. blossfeldiana and may have a similar site of action toDCMU. Neither ammonium ion nor DBMIB, which acts upon plastoquinone(i.e. between PS I and PS II in the ‘Z scheme’),had any effect on floral induction and it is argued that theinductive process is independent of photosynthetic phosphorylationbut a step in the electron transport pathway between the sitesof action of DCMU and DBMIB may be crucial. DSPD and its hydrolysisproduct, salicylaldehyde, suppressed flowering in K. blossfeldianabut the uncertainty regarding their chemistry precludes anyfirm conclusions regarding the nature of their action.  相似文献

Flowering in Lemna gibba, a long-day duckweed, can be inducedunder a short-day condition when the photoperiodic regimes areR7FR3 (7 hr red followed by 3 hr far-red), R5FR5 and R3FR7.This indicates the necessity of a proper balance between redand far-red effects for flowering. The flowering induced bythese regimes is inhibited by a brief exposure to red givenat the start of darkness and this inhibition is reversed bysubsequent exposure to far-red. Thus, the red/far-red reversibleeffect is found only at the beginning of darkness for floweringof L. gibba. However, flowering of L. gibba is promoted by a red light breakgiven near the middle of a 14 hr dark period. The promotiveeffect is not reversed by subsequent exposure to far-red, i.e.,the effect of the red break converts from inhibition to promotionas when given later in the dark period, which suggests the involvementof a timing mechanism. The effect of two culture media, potato-2 and N6 supplementedwith kinetin and either 2,4-dichlorophenoxyacetic acid (2,4-D)or -naphthalene acetic acid (NAA) on anther culture responseof two interspecific rice hybrids was studied. While calluscould be successfully induced and plants regenerated from theF1 of O. saliva x O. rufipogon, the other hybrid, O. salivax O. longistaminala did not respond to the anther culture. Nevertheless,some success in callus induction was achieved when anthers froma few selected F2 plants were cultured from the latter cross.No interaction effects between the media (potato-2, N6 and growthhormones (2,4-D and NAA) for anther response to callusing wereobserved. Kinetin retarded the decrease in chlorophyll content in leafdiscs from 5 species of plants with amphistomatous leaves, wherethe upper surface was exposed to air, but not in Rumex acetosera.When leaf discs were floated so that the lower surface was exposed,the effect of kinetin was less evident. Kinetin also stimulatedtranspiration in leaf discs from Nicotiana tabacum (amphistomatous),but not in leaf discs from Paederia chinensis (hypostomatous).Nor kinetin did retard chlorophyll breakdown in this specieswhen leaf discs were floated so that the stomatal surface wasin contact with the solution. The ineffectiveness of cytokininsin chlorophyll retention in leaf discs from hypostomatous leaveswas not due to reduced uptake of benzylaminopurine-14C. Chlorophyll retention was severely inhibited by coating theleaf surface with vaseline either in the presence or absenceof kinetin. Leaf discs floated on a solution exposed to CO2-lessair retained more chlorophyll than those in normal air. Thereis thus a close relationship between stomatal opening (as measuredby stimulation of transpiration) and chlorophyll retention,as influenced by cytokinins. It is suggested that cytokinin-induced chlorophyll retentionand odier effects on leaf tissues could be mediated throughits effects on stomatal opening. (Received January 22, 1976; )  相似文献
