Anthers of two rice (Oryza sativa L.) varieties, Lunhui 422 (P1) and Jinzao 5 (P2), their F1, F2 and first backcross generation-F1 x Lunhui 422 (B1), and F1 x Jinzao 5 (B2)-were cultured in L8 medium to study the inheritance of rice anther culturability using generation mean analysis. Significant effects of generation were observed for the four traits measured: anther response (%), frequency of callus induction (%), frequency of callus differentiation (%) and culture efficiency (%). Variation among the generations was similar for all traits: significant differences were found among six generations and the means of the P2 and B2 were significantly lower than those of the other generations. The frequency of callus differentiation showed a nonsignificant difference among the P1, F1, F2 and B1 generations which had slightly high values than the P2 and B2. Additive genetic variance (VA) was higher than non-additive genetic variance (VD) for anther response and frequency of callus induction. However, AV was lower than VD for frequence of callus differentiation and culture efficiency, VA was significant for all traits except for the culture efficiency, and VD was nonsignificant for all traits except for the frequency of callus differentiation. On the other hand, environmental variation (VE) was significant for the 4 traits. Summary Plastid DNA (ptDNA) in albino rice plants regenerated from pollen by anther culture was investigated by Southern blotting. Of the 20 albino plants investigated, 7 contained ptDNA that had suffered large-scale deletion. The size and location of the deletions differed among the plants. In all cases about 30 kbp of the region containing the PstI-2 fragment (15.7 kbp) had been retained. Summary Eleven primary trisomics of rice, variety Nipponbare, were subjected to anther culture. The 12th trisomic did not produce normal anthers. A total of 3,734 plants were obtained, which were examined morphologically at the seedling stage in the greenhouse. A number of plants appeared in the progenies of ten trisomics which had unique morphological features. The frequency of these variant types differed among different progenies. Cytological observations revealed that 43 variant plants in the progenies of nine trisomics had 13 chromosomes (n + 1), and 56 were tetrasomics (2n = 26). The tetrasomic plants in the progeny of a trisomic were morphologically identical. Similarly, n + 1 plants in the progeny of a trisomic were also identical. Plants with 23, 25, 36, 39, and 73 chromosomes were also obtained. Results show that valuable aneuploids such as n + 1 and 2n + 2 can be obtained in the anther-culture-derived progenies of trisomics. 相似文献

The results of anther culture of F2 pollen issued from 23 single crosses are presented. A relation between the morphology of the panicle and the microspore stage was established. After cold-pretreatment (8 days at 4°C), the anthers were cultured on the callus-induction medium N6 supplemented with 1 mg l–1 naphthaleneacetic acid. The calli were transferred to MS plant regeneration medium supplemented with 3 mg l–1 kinetin + 0.5 mg l–1 naphthaleneacetic acid. The induction frequency varied from 0.22% to 29% and the regeneration frequency from 0% to 144.4%, dependent upon the crosses used. On average, 27% of the plants obtained were albinos and 59% of the green plants underwent spontaneous chromosome doubling. Thirtynine doubled haploid lines were evaluated and multiplied in the field. Lines with an excellent behaviour in upland culture conditions were selected from two crosses. 相似文献

Summary Anthers of two maize (Zea mays L.) inbred lines, DBTS (P1) and B73 (P2), their F1, F2 and first backcross generations — F1 x DBTS (B1), and F1 x B73 (B2) — were float cultured in YP medium to study the inheritance of corn anther culturability using generation mean analysis. Significant effects of generation were observed for the three traits measured: anther response (%), frequency of embryos (%) and anther productivity. Variation among the generations was similar for anther response and frequency of embryos: no significant differences were found among the P1, F1, F2 and B1 means, but the means of P2 and B2 were significantly lower than those of the other generations. For anther productivity, the F2 generation tended to have a slightly higher tendency for multiple embryo formation. A simple additive-dominance model was adequate in explaining the inheritance of anther response and frequency of embryos, but digenic epistasis (additive x dominance) was involved in the inheritance of anther productivity. Additive genetic variance was higher than non-additive genetic variance for all the traits; however, only environmental variance was significant. Narrow-sense heritability estimates were 65% and 75% for anther response and frequency of embryos, respectively. Significant inter-plant variation was observed within generations, even for the inbred line DBTS, but isozymic analysis involving five enzyme loci did not reveal any genotypic variability within the inbred lines DBTS and B73. 相似文献

Minisatellites, or DNA fingerprinting sequences, have been utilized in animal linkage studies for several years but have not
been used as markers for plant genome mapping. In animal genome mapping they have resulted in limited success because they
are evenly dispersed in some species but are often clustered near telomeric regions, as observed on human chromosomes. The
purpose of the present study was to generate DNA fingerprints utilizing several rice-derived minisatellites containing different
core sequences and numbers of repeat units, followed by assessing their potential for use as genetic markers when mapped to
a rice recombinant inbred line (RIL) population. Sites of segregating minisatellite loci were mapped onto 11 of the 12 rice
RIL linkage maps. The implications for the use of rice minisatellite core sequences as genetic markers on linkage maps in
rice are discussed.

Summary Inheritance of ability to form callus in rice anther culture was studied using the diallel technique. Anthers containing uninucleate microspores from two japonica cultivais (Minehikari and Taipei 309), two indica cultivars (Mingolo and Suweon 290), and 12 F1's of the diallel crosses involving these four parents were cultured on Chaleffs R2 medium and evaluated for callus induction. The parents showed significant differences in anther callus formation, from 41.9% (Taipei 309) to 0% (Suweon 290). Callus induction ability was inherited as a recessive character conditioned by a single block of genes. Additive gene effects were predominant. The japonica types seemed to be good combiners for callus induction. The order of dominance among the four parents was Suweon 290, Mingolo, Minehikari and Taipei 309. 相似文献

Summary Inheritance of the ability to respond in wheat anther culture was studied from 6×2 reciprocal crosses between six varieties with high and two varieties with low capacity for green plant formation and their parents, replicated in two environments. Effects of genotypes dominated embryo formation and percentages of green plants, accounting for 78.4% and 85.4% of total variation, respectively, while smaller genetic effects were indicated for regeneration. Nuclear genes could explain almost all the genotype effects in this material. Embryo formation showed heterosis over high parent for 5 of the 12 hybrids, while percentages of green plants from the hybrids were intermediate to the parents. General Combining Ability (GCA) could explain 78.8% of the variation for embryo formation among the hybrids, whereas differences in percentage of green plants were dominated by Specific Combining Ability (SCA), accounting for 67.9% of hybrid variation. A positive correlation (r=0.81**) was observed between the genetic capacity for regeneration and green plant formation. Analysis of covariance indicated that effects causing GCA for green plant formation were mainly responsible for this correlation. A regression model with two parallel lines divided the six parent lines with high green plant formation into three groups with respect to their reactions with the two testers. The results are discussed with regard to possible involvement of two sets of nuclear genes affecting the percentage of green plants obtained in wheat anther culture: one set consisting of mainly additive effects affecting green plant percentage through an initial effect on regeneration ability, and another set of two or a few more major genes with dominance or epistatic effects uncorrelated with regeneration. 相似文献

Inheritance and linkage relationships of several morphological and isozyme loci are described in chickpea (Cicer arietinum L.). Segregation data obtained from several F2 families confirmed the previously observed mode of inheritance for most of the morphological loci. Additional morphological markers in chickpea are also described. Most of the isozyme loci studied showed codominant expression and fit expected Mendelian segregation ratios. However, distorted ratios were also observed for some loci. Linkage was found betweenPgd-c, the locus encoding the cytosolic form of 6-phosphogluconate dehydrogenase, andHg, the locus controlling plant growth habit. These 2 loci were separated by approximately 18 recombinational map units. A similar linkage between comparable loci was previously reported in pea (Pisum sativum L.) (Weeden and Wolko 1990). Linkage was also detected among 3 isozyme loci; the cytosolic form of phosphoglucomutase (Pgm-c), glucose-1-phosphate transferase (Gpt1), and the plastid specific form of 6-phosphogluconate dehydrogenase (Pgd-p). The linkage of 2 loci (Pgm-c andPgd-p) in this cluster is also conserved in pea and lentil (Lens Miller). The linkage between an acid phosphatase locus (Acp3) and the locus specifying the cytosolic form of glucosephosphate isomerase (Gpi-c) in chickpea suggested another linkage group in common with pea. Additionally, other linkages that were not previously observed in chickpea or related genera included the linkage of the cytosolic form of aconitase (Aco-c) with adenylate kinase (Adk1) and fructokinase (Fk3), and the linkage of a locus encoding the mitochondrial specific aconitase (Aco-m) with a seed protein locus (Spr1). The loci determining flower color (P), epicotyl color (Gst), seed coat color (T
3), and seed surface (Rs) were associated with the locus encoding glucose-1-phosphate transferase (Gpt2). These results, along with previous studies, suggest that pea, lentil and chickpea have several common linkage groups consisting of homologous genes. Genetic organization of isozyme variation in rice (Oryza sativa L.) was investigated based on 17 polymorphic isozyme loci using a sample of 511 accessions of worldwide origin. The genetic
diversity within the species was very high (H=0.36 with 4.82 alleles per locus), as compared with most selfing plant species.
Three diversity centers were detected for isozyme variation including South Asia, China and Southeast Asia. The accessions
were classified into three well-differentiated cultivar groups corresponding to the indica and japonica subspecies, and a new unnamed group. Variation within the cultivar groups accounted for 80% of the total isozyme variation.
Within-country variation accounted for 58% of the total variation while among-region and among-country variation within the
cultivar groups accounted for only 14% and 8% of the total variation. Analyses using log-linear models revealed that pronounced
non-random associations between and among alleles at many unlinked isozyme loci were organized in a non-hierarchical pattern,
and subspecific and macro-geographic differentiation was much more pronounced in multilocus phenotype frequencies than in
allelic frequencies at individual loci. These results suggest that selection on multilocus gene complexes was largely responsible
for the maintenance of the extensive isozyme variation within the species and the indica-japonica differentiation. Our results further suggest the independent domestication of indica and japonica, the dual origins of the indica rice from China and South Asia (India), and the differentiation of the ecotypes ’javanica’ and the ’temperate japonica’ within the japonica subspecies.

Summary The genetics behind response in barley anther culture was studied with 22 reciprocal and one single: cross between three varieties with high and four varieties with low capacity for green plant formation. Effects of genotypes dominated embryo formation and percentages of green plants, accounting for 62 and 76% of total variation, respectively, with almost no genetic effect on the ability to regenerate plants from pollen embryos. Nuclear genes could explain all genotype effects in this plant material, since no reciprocal effects were indicated. The three parents with high and the four parents with low capacity for green plant formation formed two phenotypically homogeneous groups, producing 27–52% and 0–7% green plants, respectively. Genetic variation within hybrids for both embryo and green plant formation could be explained completely by general combining ability (GCA). The results are discussed with respect to a previous similar study in hexaploid wheat and the reported existence of DNA deletions in the plastid genomes in albino plants from anther culture of wheat and barley. 相似文献

The microspore origin of anther-culture-derived plants of flax was determined using inter-simple sequence repeat (ISSR) and randomly amplified polymorphic DNA (RAPD) markers. Polymorphic fragments between the two parents of the F1 donor plants were identified and their segregation patterns in anther-culture-derived plants were used to elucidate the origin of those plants and to determine the degree of independence of plants regenerated from the same callus. Using one ISSR primer (UBC 889) and two RAPD primers (UBC 556 and 561), 12 out of 16 plants were unequivocally identified as being derived from microspores. Plants derived from the same callus had identical PCR patterns at five polymorphic loci and thus were likely derived from the same microspore. Therefore, it is proposed that the number of calli forming shoots be used to describe the anther culture efficiency in flax. Summary Iron tolerance of rice (Oryza sativa L.) was investigated using an oxygen depleted hydroculture system. Treatment with high concentrations of Fe2+ induced yellowing and bronzing symptoms as well as iron coatings at the root surface. Root and shoot growth were inhibited by increasing iron concentration in the medium. All symptoms were more pronounced in an iron sensitive cultivar (IR 64) compared to an iron tolerant one (IR 9764-45-2). Superoxide dismutase and peroxidase activity of root extracts of IR 97 were about twice that of IR 64 in untreated control plants. No significant increase of peroxidase activity was detected with increasing iron concentration in the medium. Catalase activity of IR 64 was slightly higher than that of IR 97, independent of iron concentration.Abbreviations SOD

Inheritance of three anther and culture characters, callus induction, green plant regeneration and culture efficiency was studied using incomplete diallel crosses with a gamete model. It was suggested that callus induction was mainly controlled by gametic additive effects and with less effect of the maternal effects. Green plant regeneration was mainly determined by maternal effects with less influence of gametic additive effects. Culture efficiency was controlled by gametic additive, maternal and cytoplasmic effects. Cultivar Lunhui 422 showed positive genetic effects for all three traits and was a very good parent for rice anther culture breeding. Significant positive heterosis was observed for callus induction. Both gametic additive and maternal correlations contributed to the significant genotypic and phenotypic correlations between callus induction and green plant regeneration suggesting these two traits to be linked.Abbreviations 2,4-d

This is the first report on the beneficial effect of microalgal and cyanobacterial biomass on anther cultures of maize (Zea mays L.). Investigations were made on the cytokinin- and auxin-like activity and content of terrestrial and fresh-water living microalgal and cyanobacterial strains. The influence of media supplemented with biomass from four selected strains on the anther induction, the frequency of microspore-derived embryo-like structures, and regeneration capacity in anther cultures of maize was also studied. The addition of cyanobacterial and microalgal biomass to the induction and regeneration media in concentrations of 1 or 2 g/L improved the androgenic response, and was able to reduce the quantity of the synthetic auxin 2,4-dichlorophenoxy-acetic acid (2,4-D) required, or replace it completely. 相似文献

Aneuhaploids and tetrasomics in rice (Oryza sativa L.) derived from anther culture of trisomics. 总被引:1,自引:0,他引:1
Eight types of aneuhaploids (Aneuhaplo 4, 5, 6, 8, 9, 10, 11, and 12) and eight types of tetrasomics (Tetraplo 4, 5, 6, 7, 8, 9, 10, and 12) of rice have been obtained from anther culture of trisomics. This paper reports the plant morphology of these aneuploids and their chromosome behavior at metaphase I. Aneuhaploids for different chromosomes are distinguishable from each other and are morphologically similar to the parental trisomics, suggesting that the extra chromosome has similar genetic effects on plant morphology at the haploid level as at the diploid level. Similarly, tetrasomics with different extra chromosomes are distinguishable from each other and are similar morphologically to the parental trisomic. However, stronger changes in morphological characters were observed in tetrasomics compared with trisomics having the same extra chromosome, as a result of a dosage effect of the extra chromosomes. Comparing plant size between aneuhaploid, tetrasomic, and trisomic with the same extra chromosome, it was shown that the trisomic was the largest, the tetrasomic was of medium size, and the aneuhaploid was the smallest, except for those plants with an extra chromosome 8 in which plant size is dramatically decreased in both the aneuhaploid and the tetrasomic. At metaphase I, aneuhaploids showed chromosome configurations of 1 II + 11 I and 13 I. The frequency of the 1 II + 11 I configuration is higher than 70%, indicating that homologous chromosomes in aneuhaploids tend to stay associated in meiosis. Intragenome chromosome pairing (2 II + 9 I), so called secondary association, was observed in the aneuhaploid for chromosome 5. Tetrasomic plants showed 5 kinds of chromosome configurations: 1 IV + 11 II, 1 III + 11 II + 1 I, 13 II, 12 II + 2 I, and 11 II + 4 I. A chromosome configuration of 13 II was often observed in tetrasomics with shorter extra chromosomes and a chromosome configuration of 1 IV + 11 II was often observed in tetrasomics with longer extra chromosomes. Aneuhaploids had complete seed sterility. Tetrasomics showed very poor pollen fertility and complete seed sterility, except for a few shriveled seeds that were observed in Tetraplo 6 and 9. This is the first report in rice where many aneuhaploids and tetrasomics have been characterized. This information will help to further unravel rice aneuploidy and cytogenetics. In order to enhance the resolution of an existing genetic map of rice, and to obtain a comprehensive picture of marker utility
and genomic distribution of microsatellites in this important grain species, rice DNA sequences containing simple sequence
repeats (SSRs) were extracted from several small-insert genomic libraries and from the database. One hundred and eighty eight
new microsatellite markers were developed and evaluated for allelic diversity. The new simple sequence length polymorphisms
(SSLPs) were incorporated into the existing map previously containing 124 SSR loci. The 312 microsatellite markers reported
here provide whole-genome coverage with an average density of one SSLP per 6 cM. In this study, 26 SSLP markers were identified
in published sequences of known genes, 65 were developed based on partial cDNA sequences available in GenBank, and 97 were
isolated from genomic libraries. Microsatellite markers with different SSR motifs are relatively uniformly distributed along
rice chromosomes regardless of whether they were derived from genomic clones or cDNA sequences. However, the distribution
of polymorphism detected by these markers varies between different regions of the genome.

The thermo-sensititve genic male-sterile (TGMS) gene in rice can alter fertility in response to temperature and is useful in the two-line system of hybrid rice production. However, little is known about the TGMS gene at the molecular level. The objective of this study was to identify molecular markers tightly linked with the TGMS gene and to map the gene onto a specific rice chromosome. Bulked segregant analysis of an F2 population from 5460s (a TGMS mutant line) x Hong Wan 52 was used to identify RAPD markers linked to the rice TGMS gene. Four hundred RAPD primers were screened for polymorphisms between the parents and between two bulks representing fertile and sterile plants; of these, 4 primers produced polymorphic products. Most of the polymorphic fragments contained repetitive sequences. Only one singlecopy sequence fragment was found, a 1.2-kb fragment amplified by primer OPB-19 and subsequently named TGMS1.2. TGMS1.2 was mapped on chromosome 8 with a RIL population and confirmed by remapping with a DHL population. Segregation analysis using TGMS1.2 as a probe indicated that TGMS1.2 both consegregated and was lined with the TGMS gene in this population. It is located about 6.7 cM from the TGMS gene. As TGMS1.2 is linked to the TGMS gene, the TGMS gene must be located on chromosome 8.This research was supported by the Rockefeller Foundation and China National High-Tech Research and Development Program. Summary Comparison of different post-plating temperature regimes with a control treatment (27° C) revealed that a short-term cold (8/14°C:2/2 days or 14°C:4 days) as well as a heat treatment (30°C:14 days) increased the production of embryro-like-structures (ELS) from cultured maize anthers. The beneficial effects of short-term cold treatments were magnified 2–3 times when L-proline (PROL) was added to the induction medium (125–500 mg/L). In the best treatment (14°C:4 days, 125 mg/L L-proline) one genotype produced 143.5 ELS/100 anthers. Anthers subjected to high temperature (30°C:4 days, 30°C:7 days, 30°C:14 days) generally showed a lower response than did cold treated anthers, although genotypic differences were observed. Regeneration frequency did not appear to be affected by the presence of L-proline in the induction medium.Abbreviations ELS
