In woody plants, photosynthetic capacity is closely linked to rates at which the plant hydraulic system can supply water to the leaf surface. Drought‐induced embolism can cause sharp declines in xylem hydraulic conductivity that coincide with stomatal closure and reduced photosynthesis. Recovery of photosynthetic capacity after drought is dependent on restored xylem function, although few data exist to elucidate this coordination. We examined the dynamics of leaf gas exchange and xylem function in Eucalyptus pauciflora seedlings exposed to a cycle of severe water stress and recovery after re‐watering. Stomatal closure and leaf turgor loss occurred at water potentials that delayed the extensive spread of embolism through the stem xylem. Stem hydraulic conductance recovered to control levels within 6 h after re‐watering despite a severe drought treatment, suggesting an active mechanism embolism repair. However, stomatal conductance did not recover after 10 d of re‐watering, effecting tighter control of transpiration post drought. The dynamics of recovery suggest that a combination of hydraulic and non‐hydraulic factors influenced stomatal behaviour post drought.  相似文献

To maintain long-distance water transport in woody plants is critical for their survival, growth and development. Water under tension is in a metastable state and prone to cavitation and embolism, which leads to loss of hydraulic conductance, reduced productivity, and eventually plant death. In face to water stress-induced cavitation, plants either reduce frequency of embolism occurrence through cavitation resistance with specialized anatomical struc- ture, or/and form a metabolically active embolism repair mechanism. For the xylem embolism and repair, however, there are controversies regarding the occurring frequency, conditions and underlying mechanisms. In this review paper, we first examined the process, temporal dynamics and frequency of xylem embolism and repair. Then, we summarized hypotheses for the mechanisms of the novel refilling in xylem embolism repair, including the osmotic hypothesis, the reverse osmotic hypothesis, the phloem-driven refilling hypothesis, and the phloem unloading hypothesis. We further compared differences in xylem embolism and repair between conifers and angiosperms tree species, and examined the trade-offs between cavitation resistance and xylem recovery performance. Finally, we proposed four priorities in future research in this field: (1) to improve measuring technology of xylem embolism; (2) to test hypotheses for the mechanisms of the novel refilling in xylem embolism repair and the signal triggering xylem refilling; (3) to explore species-specific trait differences related to xylem embolism and repair and their underlying trade-off relationships; and (4) to enhance studies on the relationship between the involvement of carbon metabolism and aquaporins expression in xylem embolism and repair.  相似文献

Diel variation in specific hydraulic conductivity (ks) was recorded in petioles of two savanna tree species, Schefflera macrocarpa and Caryocar brasiliense, from central Brazil. These two species have compound leaves with long petioles (10–30 cm). In both species, petiole ks decreased sharply with increasing transpiration rates and declining leaf water potentials (ψL) during the morning. Petiole ks increased during the afternoon while the plants were still transpiring and the water in the non‐embolized vessels was still under tension. Dye experiments confirmed that in both species diel variation in ks was associated with embolism formation and repair. When transpiration was prevented in individual leaves, their petiole ks and water potential remained close to their maximum values during the day. When minimum daily ψL on selected branches was experimentally lowered by 0.2–0.6 MPa, the rate of ks recovery during the afternoon was slower in comparison with control branches. Several field manipulations were performed to identify potential mechanisms involved in the refilling of embolized petiole vessels. Removal of the cortex or longitudinal incisions in the cortex prevented afternoon recovery of ks and refilling of embolized vessels. When distilled water was added to petiole surfaces that had been abraded to partially remove the cuticle, ks increased sharply during the morning and early afternoon. Evidence of starch to sugar conversion in the starch sheath cells surrounding the vascular bundles of the petioles was observed during periods of rapid transpiration when the abundance of starch granules in the starch sheath cells surrounding the vascular bundles decreased. Consistent with this, petiole sugar content was highest in the early afternoon. The most parsimonious explanation of the field observations and the experimental results was that an increase in osmotically active solutes in cells outside the vascular bundles at around midday leads to water uptake by these cells. However, the concurrent increase in tissue volume is partially constrained by the cortex, resulting in a transient pressure imbalance that may drive radial water movement in the direction of the embolized vessels, thereby refilling them and restoring water flow. This study thus presents evidence that embolism formation and repair are two distinct phenomena controlled by different variables. The degree of embolism is a function of tension, and the rate of refilling a function of internal pressure imbalances.  相似文献

Abstract. The hypothesis that ray parenchyma cells are actively involved in the refilling of embolized xylem of Pinus sylvestris L. was tested by killing the ray parenchyma and comparing rehydration of killed stems with that of control material. Killing of ray parenchyma was achieved using hot water or sodium azide. In most experiments, the available water for refilling was at negative water potential. Experiments were done on three kinds of plant material: small branch segments, potted seedlings and small potted trees. In all experiments, there was no indication that the azide-killed xylem was slower to refill than the control material and it was concluded that the parenchyma has no role in the refilling process, which therefore must be purely physical or physicochemical. Stems treated with hot water did not refill; we suggest that this may be caused by high temperatures decreasing the water permeability of the tracheid wall. The refilling of small branch segments may be explained by surface tension forces (capillary action), which inside the tracheid lumen may lower the water potential down to -9.7kPa; this may be enough to draw in water from the available water that in experiment one was at -2kPa. In the case of seedlings or saplings, capillary action cannot explain refilling, because the xylem water potentials were always lower than those estimated from tracheid radii. Condensation of water during diurnal cycles of warming and cooling is also unlikely to contribute to refilling significantly. To account for refilling in these cases, it is supposed that the tracheid wall may be chemically active and able to lower the water potential below the value expected by capillarity.  相似文献

Vulnerability curves using the 'Cavitron' centrifuge rotor yield anomalous results when vessels extend from the end of the stem segment to the centre ('open-to-centre' vessels). Curves showing a decline in conductivity at modest xylem pressures ('r' shaped) have been attributed to this artefact. We determined whether the original centrifugal method with its different rotor is influenced by open-to-centre vessels. Increasing the proportion of open-to-centre vessels by shortening stems had no substantial effect in four species. Nor was there more embolism at the segment end versus centre as seen in the Cavitron. The dehydration method yielded an 'r' shaped curve in Quercus gambelii that was similar to centrifuged stems with 86% open-to-centre vessels. Both 'r' and 's' (sigmoidal) curves from Cercocarpus intricatus were consistent with each other, differing only in whether native embolism had been removed. An 'r' shaped centrifuge curve in Olea europaea was indistinguishable from the loss of conductivity caused by forcing air directly across vessel end-walls. We conclude that centrifuge curves on long-vesselled material are not always prone to the open vessel artefact when the original rotor design is used, and 'r' shaped curves are not necessarily artefacts. Nevertheless, confirming curves with native embolism and dehydration data is recommended.  相似文献

Diurnal variation in petiole specific hydraulic conductivity and simultaneous measurements of leaf water potential were recorded in red maple, tulip tree and fox grape. Petiole specific conductivity was determined from in situ measurements of water flow into the distal (leaf‐bearing) end of an attached petiole as a function of applied hydrostatic pressure and petiole dimensions. The hydraulic properties of the petiole dominated the measurements, indicating that this technique can be used for rapid estimates of petiole hydraulic conductivity. There was a significant decrease in petiole specific conductivity associated with increasingly more negative leaf water potentials in maple and tulip tree, but not in grape. Petiole specific conductivity increased during the afternoon while the plant was actively transpiring and the xylem sap was under tension. The recovery of petiole conductivity during the afternoon suggests that hydraulic conductivity reflects a dynamic balance between a loss of hydraulic conductivity with increasing water stress, and its restoration as tension within the xylem decreases. Three experimental manipulations were applied to red maple and tulip tree to examine the sensitivity of diurnal changes in petiole conductivity to various physiological perturbations. Both phloem girdling and application of HgCl2 to the transpiration stream resulted in a marked decrease in the degree to which petiole specific conductivity recovered as xylem tension relaxed during the afternoon. Delivery of a surfactant to the xylem, however, did not significantly alter the relation between leaf water potential and petiole hydraulic conductivity.  相似文献

Bordered pits are cavities in the lignified cell walls of xylem conduits (vessels and tracheids) that are essential components in the water-transport system of higher plants. The pit membrane, which lies in the center of each pit, allows water to pass between xylem conduits but limits the spread of embolism and vascular pathogens in the xylem. Averaged across a wide range of species, pits account for > 50% of total xylem hydraulic resistance, indicating that they are an important factor in the overall hydraulic efficiency of plants. The structure of pits varies dramatically across species, with large differences evident in the porosity and thickness of pit membranes. Because greater porosity reduces hydraulic resistance but increases vulnerability to embolism, differences in pit structure are expected to correlate with trade-offs between efficiency and safety of water transport. However, trade-offs in hydraulic function are influenced both by pit-level differences in structure (e.g. average porosity of pit membranes) and by tissue-level changes in conduit allometry (average length, diameter) and the total surface area of pit membranes that connects vessels. In this review we address the impact of variation in pit structure on water transport in plants from the level of individual pits to the whole plant.  相似文献

It is well known that xylem embolism can be repaired by bark water uptake and that the sugar required for embolism refilling can be provided by corticular photosynthesis. However, the relationship between corticular photosynthesis and embolism repair by bark water uptake is still poorly understood. In this study, the role of corticular photosynthesis in embolism repair was assessed using Salix matsudana branch segments dehydrated to ?1.9 MPa (P50, water potential at 50% loss of conductivity). The results indicated that corticular photosynthesis significantly promoted water uptake and nonstructural carbohydrate (NSC) accumulation in the bark and xylem during soaking, thereby effectively enhancing the refilling of the embolized vessels and the recovery of hydraulic conductivity. Furthermore, the influence of the extent of dehydration on the embolism refilling enhanced by corticular photosynthesis was investigated. The enhanced refilling effects were much higher in the mildly dehydrated (?1.5 MPa) and moderately dehydrated (?1.9 MPa) branch segments than in the severely dehydrated (?2.2 MPa) branch segments. This study provides evidence that corticular photosynthesis plays a crucial role in xylem embolism repair by bark water uptake for mildly and moderately dehydrated branches.  相似文献

We investigated the common assumption that severing stems and petioles under water preserves the hydraulic continuity in the xylem conduits opened by the cut when the xylem is under tension. In red maple and white ash, higher percent loss of conductivity (PLC) in the afternoon occurred when the measurement segment was excised under water at native xylem tensions, but not when xylem tensions were relaxed prior to sample excision. Bench drying vulnerability curves in which measurement samples were excised at native versus relaxed tensions showed a dramatic effect of cutting under tension in red maple, a moderate effect in sugar maple, and no effect in paper birch. We also found that air injection of cut branches (red and sugar maple) at pressures of 0.1 and 1.0 MPa resulted in PLC greater than predicted from vulnerability curves for samples cut 2 min after depressurization, with PLC returning to expected levels for samples cut after 75 min. These results suggest that sampling methods can generate PLC patterns indicative of repair under tension by inducing a degree of embolism that is itself a function of xylem tensions or supersaturation of dissolved gases (air injection) at the moment of sample excision. Implications for assessing vulnerability to cavitation and levels of embolism under field conditions are discussed.  相似文献

Abstract Hydraulic conductivity of the xylem is computed as the quotient of mass flow rate and pressure gradient. Measurements on excised plant stems can be difficult to interpret because of time-dependent reductions in flow rate, and because of variable degrees of embolism. Using Acer saccharum Marsh. stems, we found that certain perfusing solutions including dilute fixatives (e.g. 0.05% formaldehyde) and acids with pH below 3 (e.g. 10 mol m?3 oxalic) prevent long-term decline in conductivity. Xylem embolism can be quantified by expressing the initial conductivity as a percentage of the maximum obtained after flow-impeding air emboli have been removed by repeated high-pressure (175 kPa) flushes. Correlation between microbial contamination and declining conductivity suggests that long-term (> 4h) declines are caused by microbial growth within the vessels. Unpredictable trends in short-term (< 4h) measurements may be caused by movements of air emboli in vessels and/or participate matter.  相似文献

植物体内长距离水分运输是植物生理生态学研究中的一个重要问题,长期为植物生理学家和生理生态学家所关注。木质部探针技术的问世,掀起了近年来植物生理学界最为激烈的一场争论。提出了已经有100多年,风行40年的内聚力-张力(Cohesion-Tension, C-T)学说受到质疑。随后维护派和质疑派围绕木质部探针技术、压力室技术(C-T理论的主要支撑实验技术)的可靠性展开辩论。进一步从物理学原理和各种实验上就C-T理论的3个支柱(木质部导管或管胞中巨大的张力、沿树高的压力梯度、连续水柱)进行争论。这场争论似暂告一段落,C-T理论没有被推翻,但仍留有问题期待以后的研究。  相似文献

A new method is described for measuring dissolved gas concentrations in small volumes of xylem sap using membrane inlet mass spectrometry. The technique can be used to determine concentrations of atmospheric gases, such as argon, as reported here, or for any dissolved gases and their isotopes for a variety of applications, such as rapid detection of trace gases from groundwater only hours after they were taken up by trees and rooting depth estimation. Atmospheric gas content in xylem sap directly affects the conditions and mechanisms that allow for gas removal from xylem embolisms, because gas can dissolve into saturated or supersaturated sap only under gas pressure that is above atmospheric pressure. The method was tested for red trumpet vine, Distictis buccinatoria (Bignoniaceae), by measuring atmospheric gas concentrations in sap collected at times of minimum and maximum daily temperature and during temperature increase and decline. Mean argon concentration in xylem sap did not differ significantly from saturation levels for the temperature and pressure conditions at any time of collection, but more than 40% of all samples were supersaturated, especially during the warm parts of day. There was no significant diurnal pattern, due to high variability between samples.  相似文献

The annual course of xylem embolism in twigs of adult beech trees was monitored, and compared to concurrent changes of tree water status and hydraulic resistances. Xylem embolism was quantified in 1-year-old apical twigs by the hydraulic conductivity as a percentage of the maximum measured after removal of air emboli. Tree and root hydraulic resistances were estimated from water potential differences and sap flux measurements. The considerable degree of twig embolism found in winter (up to 90% loss of hydraulic conductivity) may be attributed to the effect of freeze-thaw cycles in the xylem. A partial recovery from winter embolism occurred in spring, probably because of the production of new functional xylem. Xylem embolism fluctuated around 50% throughout the summer, without significant changes. Almost complete refilling of apical twigs was observed early in autumn. A significant negative correlation was found between xylem embolism and precipitation; thus, an active role of rainfall in embolism reversion is hypothesized. Tree and root hydraulic resistances were found to change throughout the growing period. A marked decrease of hydraulic resistance preceded the refilling of apical twigs in the autumn. Most of the decrease in total tree resistance was estimated to be located in the root compartment.  相似文献

Pressure probe measurements have been interpreted as showing that xylem pressures below c. –0.4 MPa do not exist and that pressure chamber measurements of lower negative pressures are invalid. We present new evidence supporting the pressure chamber technique and the existence of xylem pressures well below –0.4 MPa. We deduced xylem pressures in water-stressed stem xylem from the following experiment: (1) loss of hydraulic conductivity in hydrated stem xylem (xylem pressure = atmospheric pressure) was induced by forcing compressed air into intact xylem conduits; (2) loss of hydraulic conductivity from cavitation and embolism in dehydrating stems was measured, and (3) the xylem pressure in dehydrated stems was deduced as being equal and opposite to the air pressure causing the same loss of hydraulic conductivity in hydrated stems. Pressures determined in this way are only valid if cavitation was caused by air entering the xylem conduits (air-seeding). Deduced xylem pressure showed a one-to-one correspondence with pressure chamber measurements for 12 species (woody angiosperms and gymnosperms); data extended to c. –10 MPa. The same correspondence was obtained under field conditions in Betula occidentalis Hook., where pressure differences between air- and water-filled conduits were induced by a combination of in situ xylem water pressure and applied positive air pressure. It is difficult to explain these results if xylem pressures were above –0.4 MPa, if the pressure chamber was inaccurate, and if cavitation occurred by some mechanism other than air-seeding. A probable reason why the pressure probe does not register large negative pressures is that, just as cavitation within the probe limits its calibration to pressures above c. –0.5 MPa, cavitation limits its measurement range in situ.  相似文献
