Xylem recovery from embolism was studied in Laurus nobilis L. stems that were induced to cavitate by combining negative xylem pressure potentials (PX = ?1.1 MPa) with positive air pressures (PC) applied using a pressure collar. Xylem refilling was measured by recording the percentage loss of hydraulic conductance (PLC) with respect to the maximum 2 min, 20 min and 15 h after pressure release. Sodium orthovanadate (an inhibitor of many ATP‐ases) strongly inhibited xylem refilling while fusicoccin (a stimulator of the plasma membrane H+‐ATPase) promoted complete embolism reversal. So, the refilling process was interpreted to result from energy‐dependent mechanisms. Stem girdling induced progressively larger inhibition to refilling the nearer to the embolized stem segment phloem was removed. The starch content of wood parenchyma was estimated as percentages of ray and vasicentric cells with high starch content with respect to the total, before and after stem embolism was induced. A closely linear positive relationship was found to exist between recovery from PLC and starch hydrolysis. This, was especially evident in vasicentric cells. A mechanism for xylem refilling based upon starch to sugar conversion and transport into embolized conduits, assisted by phloem pressure‐driven radial mass flow is proposed.  相似文献

The ‘hydraulic vulnerability segmentation’ hypothesis predicts that expendable distal organs are more susceptible to water stress‐induced embolism than the main stem of the plant. In the current work, we present the first in vivo visualization of this phenomenon. In two separate experiments, using magnetic resonance imaging or synchrotron‐based microcomputed tomography, grapevines (Vitis vinifera) were dehydrated while simultaneously scanning the main stems and petioles for the occurrence of emboli at different xylem pressures (Ψx). Magnetic resonance imaging revealed that 50% of the conductive xylem area of the petioles was embolized at a Ψx of ?1.54 MPa, whereas the stems did not reach similar losses until ?1.9 MPa. Microcomputed tomography confirmed these findings, showing that approximately half the vessels in the petioles were embolized at a Ψx of ?1.6 MPa, whereas only few were embolized in the stems. Petioles were shown to be more resistant to water stress‐induced embolism than previously measured with invasive hydraulic methods. The results provide the first direct evidence for the hydraulic vulnerability segmentation hypothesis and highlight its importance in grapevine responses to severe water stress. Additionally, these data suggest that air entry through the petiole into the stem is unlikely in grapevines during drought.  相似文献

Quercus ilex L. growing in the southern Mediterranean Basin region is exposed to xylem embolism induced by both winter freezing and summer drought. The distribution of the species in Sicily could be explained in terms of the different vulnerability to embolism of its xylem conduits. Naturally occurring climatic conditions were simulated by: (1) maintaining plants for 3h at ambient temperatures of 0, -1.5, -2.5, -5.0 and -11°C; and (2) allowing plants to dry out to ratios of their minimum diurnal leaf water potentials (Ψ1) to that at the turgor loss point (Ψtlp) of 0.6, 0.9, 1.05, 1.20 and 1.33. The loss of hydraulic conductivity of one-year-old twigs reached 40% at -1.5°C and at Ψ1tlP= 1.05. Recovery from these strains was almost complete 24 h after the release of thermal stress or after one irrigation, respectively. More severe stresses reduced recovery consistently. The percentages of xylem conduits embolized following application of the two stresses, were positively related to xylem conduit diameter. The capability of the xylem conduits to recover from stress was positively related to the conduit diameter in plants subjected to summer drought, but not in the plants subjected to winter freezing stress. The ecological significance of the different vulnerabilities to embolism of xylem conduits under naturally occurring climatic conditions is discussed.  相似文献

Drought and freezing are both known to limit desert plant distributions, but the interaction of these stressors is poorly understood. Drought may increase freezing tolerance in leaves while decreasing it in the xylem, potentially creating a mismatch between water supply and demand. To test this hypothesis, we subjected Larrea tridentata juveniles grown in a greenhouse under well‐watered or drought conditions to minimum temperatures ranging from ?8 to ?24 °C. We measured survival, leaf retention, gas exchange, cell death, freezing point depression and leaf‐specific xylem hydraulic conductance (kl). Drought‐exposed plants exhibited smaller decreases in gas exchange after exposure to ?8 °C compared to well‐watered plants. Drought also conferred a significant positive effect on leaf, xylem and whole‐plant function following exposure to ?15 °C; drought‐exposed plants exhibited less cell death, greater leaf retention, higher kl and higher rates of gas exchange than well‐watered plants. Both drought‐exposed and well‐watered plants experienced 100% mortality following exposure to ?24 °C. By documenting the combined effects of drought and freezing stress, our data provide insight into the mechanisms determining plant survival and performance following freezing and the potential for shifts in L. tridentata abundance and range in the face of changing temperature and precipitation regimes.  相似文献

  • Positive sap pressures are produced in the xylem of birch trees in boreal conditions during the time between the thawing of the soil and bud break. During this period, xylem embolisms accumulated during wintertime are refilled with water. The mechanism for xylem sap pressurization and its environmental drivers are not well known.
  • We measured xylem sap flow, xylem sap pressure, xylem sap osmotic concentration, xylem and whole stem diameter changes, and stem and root non‐structural carbohydrate concentrations, along with meteorological conditions at two sites in Finland during and after the sap pressurisation period.
  • The diurnal dynamics of xylem sap pressure and sap flow during the sap pressurisation period varied, but were more often opposite to the diurnal pattern after bud burst, i.e. sap pressure increased and sap flow rate mostly decreased when temperature increased. Net conversion of soluble sugars to starch in the stem and roots occurred during the sap pressurisation period. Xylem sap osmotic pressure was small in comparison to total sap pressure, and it did not follow changes in environmental conditions or tree water relations.
  • Based on these findings, we suggest that xylem sap pressurisation and embolism refilling occur gradually over a few weeks through water transfer from parenchyma cells to xylem vessels during daytime, and then the parenchyma are refilled mostly during nighttime by water uptake from soil. Possible drivers for water transfer from parenchyma cells to vessels are discussed. Also the functioning of thermal dissipation probes in conditions of changing stem water content is discussed.

It is well known that xylem embolism can be repaired by bark water uptake and that the sugar required for embolism refilling can be provided by corticular photosynthesis. However, the relationship between corticular photosynthesis and embolism repair by bark water uptake is still poorly understood. In this study, the role of corticular photosynthesis in embolism repair was assessed using Salix matsudana branch segments dehydrated to ?1.9 MPa (P50, water potential at 50% loss of conductivity). The results indicated that corticular photosynthesis significantly promoted water uptake and nonstructural carbohydrate (NSC) accumulation in the bark and xylem during soaking, thereby effectively enhancing the refilling of the embolized vessels and the recovery of hydraulic conductivity. Furthermore, the influence of the extent of dehydration on the embolism refilling enhanced by corticular photosynthesis was investigated. The enhanced refilling effects were much higher in the mildly dehydrated (?1.5 MPa) and moderately dehydrated (?1.9 MPa) branch segments than in the severely dehydrated (?2.2 MPa) branch segments. This study provides evidence that corticular photosynthesis plays a crucial role in xylem embolism repair by bark water uptake for mildly and moderately dehydrated branches.  相似文献

Root pressure and plasma membrane intrinsic protein (PIP) availability in the xylem have been recognized to participate in the refilling of embolized conduits, yet integration of the two mechanisms has not been reported in the same plant. In this study, 4‐month‐old seedlings of a hybrid poplar (Populus alba × Populus glandulosa) clone 84K were subjected to two contrasting soil‐water treatments, with the drought treatment involving withholding of water for 17 days to reduce the soil‐water content to 10% of the saturated field capacity, followed by a re‐watering cycle. The percentage loss of stem hydraulic conductance (PLC) sharply increased, and stomatal conductance and photosynthesis declined in response to drought stress; these processes were gradually restored following the subsequent re‐watering. Embolism was most severe in the middle portions of the stem, followed by the basal and top portions of the stems of seedlings subjected to drought stress and subsequent re‐watering. Although drought stress eliminated root pressure, re‐watering partially restored it in a short period of time. The expression of PIP genes in the xylem was activated by drought stress, and some PIP genes were further stimulated in the top portion after re‐watering. The dynamics of root pressure and differential expression of PIP genes along the stem coincided with changes in PLC, suggesting that root pressure and PIPs work together to refill the embolized vessels. On the basis of the recovery dynamics in PLC and gsmax (maximum stomatal conductance) after re‐watering, the stomatal closure and xylem cavitation exhibited fatigue due to drought stress.  相似文献

Embolism and the refilling of xylem vessels are intrinsic to the ability of plants to handle the transport of water under tension. Although the formation of an embolized vessel is an abiotic process, refilling against the pressure gradient requires biological activity to provide both the energy and the water needed to restore xylem transport capacity. Here, we present an analysis of the dynamics of embolism and refilling in Populus trichocarpa and follow temporal dynamics of co‐occurring changes in expression level of aquaporins. Under mesic conditions, we found that the percent loss of conductance (PLC) varied diurnally by as much as 20%, suggesting a continuous embolism/refilling cycle. An increase in water stress tilted the balance between the two processes and increased the PLC to as much as 80%. Subsequent re‐watering resulted in the reversal of water stress and recovery of PLC to pre‐stress levels. Stem parenchyma cells responded to drought stress with considerable up‐regulation of the PIP1 subfamily of water channels but not the PIP2 subfamily. Even more significant was the finding that PoptrPIP1.1 and PoptrPIP1.3 genes were up‐regulated in response to embolism, but not to water stress, and were down‐regulated after embolism removal, suggesting a local ability of plants to sense an embolism presence.  相似文献

The consequences of manipulating abscisic acid (ABA) biosynthesis rates on stomatal response to drought were analysed in wild‐type, a full‐deficient mutant and four under‐producing transgenic lines of N. plumbaginifolia. The roles of ABA, xylem sap pH and leaf water potential were investigated under four experimental conditions: feeding detached leaves with varying ABA concentration; injecting exogenous ABA into well‐watered plants; and withholding irrigation on pot‐grown plants, either intact or grafted onto tobacco. Changes in ABA synthesis abilities among lines did not affect stomatal sensitivity to ABA concentration in the leaf xylem sap ([ABA]xyl), as evidenced with exogenous ABA supplies and natural increases of [ABA]xyl in grafted plants subjected to drought. The ABA‐deficient mutant, which is uncultivable under normal evaporative demand, was grafted onto tobacco stock and then presented the same stomatal response to [ABA]xyl as wild‐type and other lines. This reinforces the dominant role of ABA in controlling stomatal response to drought in N. plumbaginifolia whereas roles of leaf water potential and xylem sap pH were excluded under all studied conditions. However, when plants were submitted to soil drying onto their own roots, stomatal response to [ABA]xyl slightly differed among lines. It is suggested, consistently with all the results, that an additional root signal of soil drying modulates stomatal response to [ABA]xyl.  相似文献

Lipids have been observed attached to lumen-facing surfaces of mature xylem conduits of several plant species, but there has been little research on their functions or effects on water transport, and only one lipidomic study of the xylem apoplast. Therefore, we conducted lipidomic analyses of xylem sap from woody stems of seven plants representing six major angiosperm clades, including basal magnoliids, monocots and eudicots, to characterize and quantify phospholipids, galactolipids and sulfolipids in sap using mass spectrometry. Locations of lipids in vessels of Laurus nobilis were imaged using transmission electron microscopy and confocal microscopy. Xylem sap contained the galactolipids di- and monogalactosyldiacylglycerol, as well as all common plant phospholipids, but only traces of sulfolipids, with total lipid concentrations in extracted sap ranging from 0.18 to 0.63 nmol ml−1 across all seven species. Contamination of extracted sap from lipids in cut living cells was found to be negligible. Lipid composition of sap was compared with wood in two species and was largely similar, suggesting that sap lipids, including galactolipids, originate from cell content of living vessels. Seasonal changes in lipid composition of sap were observed for one species. Lipid layers coated all lumen-facing vessel surfaces of L. nobilis, and lipids were highly concentrated in inter-vessel pits. The findings suggest that apoplastic, amphiphilic xylem lipids are a universal feature of angiosperms. The findings require a reinterpretation of the cohesion-tension theory of water transport to account for the effects of apoplastic lipids on dynamic surface tension and hydraulic conductance in xylem.  相似文献

Among woody plants, grapevines are often described as highly vulnerable to water‐stress induced cavitation with emboli forming at slight tensions. However, we found native embolism never exceeded 30% despite low xylem water potentials (Ψx) for stems of field grown vines. The discrepancy between native embolism measurements and those of previous reports led us to assess vulnerability curve generation using four separate methods and alterations (i.e. segment length and with/without flushing to remove embolism prior to measurement) of each. Centrifuge, dehydration and air‐injection methods, which rely on measurement of percentage loss of hydraulic conductivity (PLC) in detached stems, were compared against non‐invasive monitoring of xylem cavitation with nuclear magnetic resonance (NMR) imaging. Short segment air‐injection and flushed centrifuge stems reached >90 PLC at Ψx of‐0.5 and ?1.5 MPa, respectively, whereas dehydration and long‐segment air‐injection measurements indicated no significant embolism at Ψx > ?2.0 MPa. Observations from NMR agreed with the dehydration and long segment air‐injection methods, showing the majority of vessels were still water‐filled at Ψx > ?1.5 MPa. Our findings show V. vinifera stems are far less vulnerable to water stress‐induced cavitation than previously reported, and dehydration and long segment air‐injection techniques are more appropriate for long‐vesseled species and organs.  相似文献

In vivo magnetic resonance imaging of xylem vessel contents in woody lianas   总被引:1,自引:0,他引:1  
Previous reports suggest that in some plant species the refilling of embolized xylem vessels can occur while negative pressure exists in the xylem. The aim of this experiment was to use non‐destructive nuclear magnetic resonance imaging (MRI) to study the dynamics of xylem cavitation and embolism repair in‐vivo. Serial 1H‐MRI was used to monitor the contents of xylem vessels in stems of two dicotyledonous (Actinidia deliciosa and Actinidia chinensis, kiwifruit) and one monocotyledonous (Ripogonum scandens, supplejack) species of woody liana. The configuration of the horizontal wide bore magnet and probe allowed the imaging of woody stems up to 20 mm in diameter. Tests using excised stems confirmed that the image resolution of 78 µm and digital image subtraction could be used to detect the emptying and refilling of individual vessels. Imaging was conducted on both intact plants and excised shoots connected to a water supply. In the case of Ripogonum the excised shoots were long enough to allow the distal end of the shoot, including all leaves, to be exposed to ambient conditions outside the building while the proximal end was inside the MRI magnet. In total, six stems were monitored for 240 h while the shoots were subjected to treatments that included light and dark periods, water stress followed by re‐watering, and the covering of all leaves to prevent transpiration. The sudden emptying of water‐filled vessels occurred frequently while xylem water potential was low (below ?0.5 MPa for Actinidia, ?1.0 MPa for Ripogonum), and less frequently after xylem water potential approached zero at the end of water‐stress treatments. No refilling of empty vessels was observed at any time in any of the species examined. It is concluded that embolism repair under negative pressure does not occur in the species examined here. Embolism repair may be more likely in species with narrower xylem vessels, but further experiments are required with other species before it can be concluded that repair during transpiration is a widespread phenomenon.  相似文献

Drought‐induced xylem embolism is a key process closely related to plant mortality during extreme drought events. However, this process has been poorly investigated in crop species to date, despite the observed decline of crop productivity under extreme drought conditions. Interspecific variation in hydraulic traits has frequently been reported, but less is known about intraspecific variation in crops. We assessed the intraspecific variability of embolism resistance in four sunflower (Helianthus annuus L.) accessions grown in well‐watered conditions. Vulnerability to embolism was determined by the in situ flow‐centrifuge method (cavitron), and possible trade‐offs between xylem safety, xylem efficiency and growth were assessed. The relationship between stem anatomy and hydraulic traits was also investigated. Mean P 50 was ?3 MPa, but significant variation was observed between accessions, with values ranging between ?2.67 and ?3.22 MPa. Embolism resistance was negatively related to growth and positively related to xylem‐specific hydraulic conductivity. There is, therefore, a trade‐off between hydraulic safety and growth but not between hydraulic safety and efficiency. Finally, we found that a few anatomical traits, such as vessel density and the area of the vessel lumen relative to that of the secondary xylem, were related to embolism resistance, whereas stem tissue lignification was not. Further investigations are now required to investigate the link between the observed variability of embolism resistance and yield, to facilitate the identification of breeding strategies to improve yields in an increasingly arid world.  相似文献

We examined the relationships between xylem resistance to cavitation and 16 structural and functional traits across eight unrelated Populus deltoides×Populus nigra genotypes grown under two contrasting water regimes. The xylem water potential inducing 50% loss of hydraulic conductance (Ψ50) varied from ?1.60 to ?2.40 MPa. Drought‐acclimated trees displayed a safer xylem, although the extent of the response was largely genotype dependant, with Ψ50 being decreased by as far as 0.60 MPa. At the tissue level, there was no clear relationship between xylem safety and either xylem water transport efficiency or xylem biomechanics; the only structural trait to be strongly associated with Ψ50 was the double vessel wall thickness, genotypes exhibiting a thicker double wall being more resistant. At the leaf level, increased cavitation resistance was associated with decreased stomatal conductance, while no relationship could be identified with traits associated with carbon uptake or bulk leaf carbon isotope discrimination, a surrogate of intrinsic water‐use efficiency. At the whole‐plant level, increased safety was associated with higher shoot growth potential under well‐irrigated regime only. We conclude that common trade‐offs between xylem resistance to cavitation and other physiological traits that are observed across species may not necessarily hold true at narrower scales.  相似文献

Recovery of hydraulic conductivity after the induction of embolisms was studied in woody stems of laurel (Laurus nobilis). Previous experiments confirming the recovery of hydraulic conductivity when xylem pressure potential was less than −1 MPa were repeated, and new experiments were done to investigate the changes in solute composition in xylem vessels during refilling. Xylem sap collected by perfusion of excised stem segments showed elevated levels of several ions during refilling. Stem segments were frozen in liquid N2 to view refilling vessels using cryoscanning electron microscopy. Vessels could be found in all three states of presumed refilling: (a) mostly water with a little air, (b) mostly air with a little water, or (c) water droplets extruding from vessel pits adjacent to living cells. Radiographic probe microanalysis of refilling vessels revealed nondetectable levels of dissolved solutes. Results are discussed in terms of proposed mechanisms of refilling in vessels while surrounding vessels were at a xylem pressure potential of less than −1 MPa. We have concluded that none of the existing paradigms explains the results.  相似文献

Embolism reversal in rice plants was studied by testing the plant's ability to refill embolized conduits while xylem pressures were substantially negative. Intact, potted plants were water-stressed to a xylem pressure of -1.88 ± 0.1 MPa and a 66.3 ± 3.8% loss of xylem conductivity (PLC) by cavitation. Stressed plants were carefully rewatered, allowing xylem pressure to rise, but not above the theoretical threshold of c. -0.15 MPa for embolism collapse. Despite xylem pressures being more negative than this threshold, the PLC fell significantly (28.5 ± 5.6%), indicating the refilling of vessels. Above c. -1.0 MPa, almost all plants regained their maximum hydraulic conductivity. Dye uptake experiments showed the same pattern of embolism refilling despite negative pressure. Refilling was prevented in plants that were light-starved for 5 d, suggesting the unknown mechanism is dependent on metabolic energy. Results are among the first showing that herbaceous plants can reverse embolism without bulk xylem pressures rising near or above atmospheric.  相似文献

Pre-dawn xylem pressures were measured with bubble manometers attached near the stem bases of 32 species of vines on Barro Colorado Island, Panama, to determine if pressures were sufficient to allow for possible refilling of embolized vessels. Of 29 dicotyledonous species 26 exhibited only negative xylem pressures, even pre-dawn during the wet season. In contrast, three members of the Dilleniaceae exhibited positive pre-dawn xylem pressures, with a maximum of 64 kPa in Doliocarpusmajor. A pressure of 64 kPa is sufficient to push water to a height of 6.4 m against gravity, but the specimens reached heights of 18 m. Thus, in all 29 dicotyledons examined, the xylem pressures were not sufficient to refill embolized vessels in the upper stems. In contrast, two of the smaller, non-dicotyledonous vines, the climbing fern Lygodiumvenustrum and the viny bamboo Rhipidocladumracemiflorum, had xylem pressures sufficient to push water to the apex of the plants. Therefore, a root pressure mechanism to reverse embolisms in stem xylem could apply to some but not to most of the climbing plants that were studied.
