卵形鲳鯵对刺激隐核虫的免疫应答和免疫保护研究   总被引:1,自引:0,他引:1  
用刺激隐核虫(Cryptocaryon irritans)的幼虫对卵形鲳鯵(Trachinotus ovatus)进行腹腔注射和体表感染,然后每隔一周用阻动试验(Immobilization assay)检测免疫鱼的抗血清和皮肤培养液对刺激隐核虫幼虫的阻动效价,在第14周中,分别用亚致死剂量和致死剂量的刺激隐核虫幼虫对免疫鱼攻毒以检测所产生的免疫保护力。实验结果显示:两种免疫方法都能让卵形鲳鯵的血清和皮肤生成阻动刺激隐核虫幼虫的特异性抗体,并能使被免疫鱼获得明显的免疫保护,但是体表感染免疫组的血清和皮肤培养液的阻动效价都要比腹腔注射免疫组高,所获得的免疫保护力也更强。同时还发现,免疫鱼血清和皮肤培养液中的抗体存在明显的差异:两者的最初生成时间、达到峰值的时间、变化规律以及阻动效价等都不一致。因此,我们推测鱼类的系统免疫应答和皮肤黏膜免疫应答有可能是相互独立的,或者是不同步的。鱼类的体液免疫应答,特别是黏膜免疫应答对抵御刺激隐核虫的感染起了重要的作用,采用刺激隐核虫虫体疫苗可能成为预防海水鱼类白点病的一种选择。    相似文献

作者从卵圆鲳鲹Trachinotus blochii分离了一株刺激隐核虫Cryptocaryon irritans,再经人工感染的方法收集各期虫体,制成电镜样品,对虫体的胞口超微结构进行了观察.同时,用银染和免疫荧光染色以及共聚焦显微镜对虫体的胞口周围及内部的纤维和微管进行了观察.结果表明刺激隐核虫的胞口结构与前口类纤毛虫(Prostome)的胞口结构有诸多相似之处,而与归属于膜口类Ophryoglenina小瓜虫差异较大,因此,作者认为刺激隐核虫的分类更适合归属于前口类Prostome,而不是膜口类Ophryoglenina.  相似文献

刺激隐核虫感染对褐菖鲉的胁迫及鱼体的免疫应答   总被引:2,自引:0,他引:2  
为探明刺激隐核虫感染对褐菖鲉生理机能的影响,研究分别用2500、5000、7500和10000幼虫/鱼的刺激隐核虫感染褐菖鲉,并分别检测感染后24h、48h、72h和96h各时间点血清中皮质醇(COR)、血糖(GLU)、总蛋白(TP)含量;肝脏中丙二醛(MDA)和维生素C(VC)含量,超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活力;鳃和皮肤中溶菌酶(LZM)活力。结果显示,随着感染浓度的增加,血液中COR和GLU含量均出现不同程度的升高,其中2500、5000和7500幼虫/鱼组COR含量的最高值均出现在感染后第3天;而TP含量总体呈现逐步下降的趋势,尤其当感染浓度达到5000幼虫/鱼后,TP含量的下降程度明显增加;肝脏中MDA含量呈先降后升的变化趋势,其中24h、48h和72h各点MDA含量的最大值均出现在10000幼虫/鱼组,最小值则集中出现于在2500和5000幼虫/鱼组;而VC含量则与MDA含量的趋势相反;SOD和CAT活力均出现不同程度的升高;鳃和皮肤LZM活力总体呈先上升后回落的变化趋势。综上可知,刺激隐核虫感染会对鱼体造成氧化胁迫和脂质过氧化反应,其严重程度与感染的虫细胞浓度相关。低浓度感染组的鱼所受胁迫较轻,在滋养体脱落后仍具有一定的自我修复能力;而高浓度感染组鱼免疫因子的释放受到抑制或出现紊乱,即便在虫体脱落后,其体质也很难恢复。  相似文献

自网箱养殖的卵圆鲳鲹(Trachinotus blochii)分离了一株刺激隐核虫(Cryptocaryon irritans),再经人工感染的方法收集各期虫体,制成电镜样品,对虫体进行超微结构研究。研究表明:刺激隐核虫的毛基体具有典型的9套三联微管系统。幼虫体部单毛基单元有如下特征:单毛基单元结构,具有稍分支的纤毛后微管束(Postciliary microtubular ribbon),由3根微管组成,起源于第3三联微管,向后延伸至细胞表面。滋养体体部单毛基单元特征是具有分支的纤毛后微管束,由3-6根微管组成,起源于第9三联微管。毛基体的第4-7组三联微管处发出一条长而无分支重叠的动纤丝(KD),KD向右前方伸延到质膜脊的顶端。在横微管(T)左侧的第3三联微管处有一个横行支持物即横带,由3根微管组成,与来源于第3-4三联微管的横纤维(Tf)相连。刺激隐核虫的体部毛基单元具有一个独特特征:即具有体部线带(N),是一束平行排列的微管,它们起源于邻接横微管和横纤维的致密板,滋养体的体部线带相互重叠交错排列,以此支撑细胞的横轴。滋养体期还有厚的表质层(Epiplasmic layer)。体部双毛基单元即为邻近口区的双毛基单元,每一排动基列的前几个毛基单元都是双毛基单元,极似口部毛基单元,口部双毛基单元的后面有3-4个体部双毛基单元。幼虫和滋养体的体部双毛基单元均具有2个毛基体,其结构模式相似。体部毛基单元的2个毛基体各长出1根纤毛,毛基体与一个旁体囊(PS)相连,其前面有动纤丝。文中分析了刺激隐核虫在形态上与淡水小瓜虫以及相关种类的毛基单元和皮层结构差异,认为刺激隐核虫的分类更适合归于前口类(Prostomatea),而不是膜口类(Hymenostomatida)。  相似文献

卵形鲳鲹胚胎及早期仔鱼耗氧量的研究   总被引:2,自引:0,他引:2  
运用SKW-3微量呼吸仪对卵形鲳鲹(Trachinotus ovatus)胚胎和早期仔鱼的耗氧量进行测定,研究温度、盐度及pH的变化对胚胎耗氧量和Cu2+、Cd2+等重金属离子对早期仔鱼的耗氧量的影响。结果表明:在(25±0.5)℃条件下,卵形鲳鲹胚胎和早期仔鱼的耗氧量均随着发育时间的延长总体上呈上升的趋势,胚胎耗氧量在出膜前达到最大值,其中以原肠期和出膜期耗氧量变化最为显著。胚胎耗氧量随着温度、盐度、pH的增大而逐渐升高,在水温25℃、盐度35和pH为8时耗氧量到达最大值,随后逐渐减小。随着Cu2+浓度的升高,早期仔鱼耗氧量呈先增大后减小,在0.01mg·L-1时耗氧率最大:胚胎耗氧量随着Cd2+浓度的升高而逐渐降低。  相似文献

黄斑蓝子鱼(Siganus oramin)具有天然抗刺激隐核虫(Cryptocaryon irritans)能力, 并从其血清中分离纯化了一种抗虫蛋白。以黄斑蓝子鱼皮肤黏液为对象, 研究了其对刺激隐核虫(C. irritans)、多子小瓜虫(Ichthyophthirius multifiliis)、布氏锥虫(Trypanosoma brucei brucei)及一些病原菌的杀灭和抑制效果。结果显示: 黄斑蓝子鱼黏液对刺激隐核虫、多子小瓜虫和布氏锥虫均具有明显的杀虫效果, 黏液蛋白对3种寄生虫的最低杀寄生虫浓度(Minimum Parasiticidal Concentration, MPC)分别为4.0、5.0和3.0 mg/mL。显微镜观察发现, 刺激隐核虫和多子小瓜虫的幼虫在经过黄斑蓝子鱼黏液作用后, 均出现纤毛脱落、虫体肿胀、外膜破裂和内容物泄漏等现象。黏液抑菌活性实验结果表明, 除大肠杆菌(Escherichia coli)外, 黄斑蓝子鱼皮肤黏液对金黄色葡萄球菌(Staphylococcus aureus)、霍乱弧菌(Vibrio cholerae)、海豚链球菌(Streptococcus iniae)、温和气单胞菌(Aeromonas sobria)、溶藻弧菌(Vibrio alginolyticus)、副溶血弧菌(Vibrio Parahaemolyticus)、星形诺卡氏菌(Nocadia asteroides)和美人鱼发光杆菌杀鱼亚种(Photobacterium damselae subsp.)皆有明显的抑菌效果。在最低抑菌浓度(Minimum Inhibitory Concentration, MIC)测定中, 黄斑蓝子鱼黏液对金黄色葡萄球菌、霍乱弧菌、海豚链球菌、溶藻弧菌、温和气单胞菌和副溶血弧菌的MIC 分别为0.16、0.16、0.08、0.63、0.63和0.63 mg/mL。黄斑蓝子鱼皮肤黏液对小麦赤霉菌(Gibberella saubinetii)和黑曲霉菌(Aspergillus niger)没有明显抑菌作用。    相似文献

自然海域和养殖水体环境频现低氧,本研究针对卵形鲳鲹(Trachinotus ovatus)不耐低氧的特性,将(31.59±3.01)g(n=30)的卵形鲳鲹在(23±0.7)℃下进行3、6、12和24 h的急性和14 d的慢性低氧[溶解氧为(1.55±0.20)mg/L]胁迫。运用光学和电子显微技术,比较急、慢性低氧胁迫对卵形鲳鲹肝组织显微和超微结构的影响。通过测定肝组织中的丙二醛(MDA)含量及过氧化氢酶(CAT)、超氧化物歧化酶(SOD)和谷胱甘肽还原酶(GSH)的活性,分析低氧胁迫对卵形鲳鲹肝是否造成氧化损伤。急性低氧胁迫下,卵形鲳鲹肝组织间出现空泡、小叶结构破坏,细胞内线粒体数量减少,出现过氧化物酶体,肝细胞间血窦剧烈扩张。这些病理损伤随胁迫时间延长更趋严重,24 h时甚至出现局部肝细胞融合、坏死,慢性低氧胁迫14 d时,肝细胞局部坏死,细胞膜溶解,细胞核破裂分解,胞质内细胞器不明显,只可分辨粗面内质网,细胞内空泡体积大,细胞结构松散,血窦扩张。急性低氧胁迫下丙二醛(MDA)含量随时间先上升后下降,慢性低氧胁迫14 d时丙二醛(MDA)则显著增加。急性低氧胁迫下超氧化物歧化酶(SOD)和谷胱甘肽还原酶(GSH)上升后恢复,过氧化氢酶(CAT)则持续上调,慢性低氧胁迫下超氧化物歧化酶(SOD)和谷胱甘肽还原酶(GSH)显著上调,过氧化氢酶(CAT)活性下降。结果表明,卵形鲳鲹幼鱼肝组织在低氧胁迫下病理变化明显,氧化损伤严重,且慢性低氧胁迫比急性更甚。  相似文献

通过盐度渐变和温度骤变的方法,分别研究了不同盐度(10、20、30、40)处理和不同温度(18.0℃、21.0℃、24.6℃、29.0℃、32.0℃)处理对卵形鲳鲹(Trachinotus ovatus)选育群体肝超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GPX)活力的影响.在实验结束时,盐度10组酶活力与对照组无显著差异(P>0.05),盐度20组SOD活力极显著低于对照组(P<0.01);盐度40组120 h时,SOD和GPX活力极显著低于对照组(P<0.01),CAT活力与对照组差异不显著(P>0.05).18.0℃和21.0℃ SOD活力在1、3、6、12、24 h这5个取样时间点均高于对照组,CAT活力在实验结束时(24 h)极显著高于对照组(P <0.01);29.0℃SOD和CAT活力在实验结束时(24 h)显著高于对照组(P<0.05);32.0℃ SOD和CAT活力在5个取样时间点均显著低于对照组(P<0.05).结果表明,适当的盐度或温度变化可以改变卵形鲳鲹肝抗氧化酶活力,达到机体耐受极限时酶活力下降.  相似文献

Host specificity of the marine ciliate Cryptocaryon irritans was investigated using seven parasite isolates and six experimental fish species. The low degree of host specificity of C. irritans to teleost fish has been confirmed. In order to elucidate the immune-protective mechanisms of inactivated Cryptocaryon irritans vaccine, different doses of C. irritans theronts were used to immunize orange-spotted grouper (Epinephelus coioides). We measured serum immobilization titer, blood leukocyte respiratory burst activity, serum alternative complement activity, and serum lysozyme activity weekly. In addition, the expression levels of immune-related genes such as interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), major histocompatibility complexes I and II (MHC I and II), and transforming growth factor-β1 (TGF-β1) were determined in spleen and gills. The results showed that the immobilization titer, respiratory burst activity, and alternative complement activity of immunized fish were significantly increased, and the levels of the last two immune parameters in the high-dose vaccine group were significantly higher than in the low-dose vaccine group. Serum lysozyme activity in the high-dose vaccine group was significantly higher than in the PBS control group. Vaccination also regulated host immune-related gene expression. For example, at 2- and 3- weeks post immunization, IL-1β expression in the high-dose vaccine group spleen was significantly increased. At 4-weeks post immunization, the fish were challenged with a lethal dose of parasite, and the survival rates of high-dose vaccine group, low-dose vaccine group, PBS control group, and adjuvant control group were 80%, 40%, 0%, and 10% respectively. These results demonstrate that inactivated C. irritans vaccination improves specific and nonspecific immune responses in fish, enhancing their anti-parasite ability. These effects are vaccine antigen dose-dependent.  相似文献

The objective of this study was to determine whether immunization of Mozambique tilapia with different Cryptocaryon irritans i-antigen serotypes elicited cross-protection against challenge infection by both serotypes. Fish were directly exposed to live theronts of isolate W1 or isolate K1, that express different surface i-antigens. There was no significant difference in the number of trophonts infecting the fish between the two isolates, W1 and K1, following primary exposure. Serum from immunized fish exposed to live theronts showed higher immobilization titres and ELISA values against homologous isolates than to heterologous isolates after the primary exposure. However, mucus antibody did not immobilize theronts although the ELISA results clearly indicated that mucus antibodies recognizing C. irritans were generated. In a study with Western blot analyses, serum antibodies recognized only an antigen of the corresponding serotype and no proteins common to both serotypes were identified. Sequence analyses of 754 bases of rDNA nucleotide sequence including complete nuclear ribosomal ITS-1–5.8S rDNA–ITS-2 region were conducted and found to be identical for W1- and K1-isolates. These findings confirmed that both isolates were members of the species, C. irritans, and that rDNA analysis would not distinguish the two isolates. In conclusion, despite the fact that the immobilization assays and ELISA detected two serotypes in vitro, challenge assays provided evidence for only one type of C. irritans.  相似文献

The protective immunity of grouper (Epinephelus coioides) against Cryptocaryon irritans was determined after immunisation by surface exposure or intraperitoneal injection. Specific antibody titres of immunised fish serum and skin culture supernatant were determined by enzyme-linked immunosorbent assay and immobilisation assays. Specific antibody can be detected in some immunised fish at Week 1 and in all immunised fish at Week 2, and the peaks were between Weeks 4-6. Specific antibody was still evident in the serum and skin of immunised fish at Week 8, and provided good protection against challenge with C. irritans. These findings indicated that humoral and skin mucosal immunity play important roles in fish against C. irritans infection.  相似文献

对卵形鲳鲹(Trachinotus ovatus)肝脏、脾脏、头肾和体肾4种组织器官内各种血细胞的发生和外周血液的血细胞分类组成进行了观察。结果表明:卵形鲳鲹的红细胞主要在脾脏、体肾和头肾中发生;淋巴细胞主要在体肾、头肾和脾脏中发生;粒细胞主要在头肾和体肾中发生;单核细胞在肝脏、头肾和脾脏中均有发生;而血栓细胞的发生在这4种组织中均未观察到。卵形鲳鲹在外周血液中的白细胞包括淋巴细胞、血栓细胞、嗜中性粒细胞和单核细胞。在这几种白细胞中,单核细胞数量最少,很难观察到,数量少于1%;嗜中性粒细胞稍多,有(9.25±5.64)%;血栓细胞占(38.23±21.86)%;淋巴细胞最多,达(52.52±21.65)%。  相似文献

Cryptocaryon irritans causes Cyptocaryonosis or white spot disease in a wide range of marine fish including Lates calcarifer (Asian seabass). However, the immune response of this fish to the parasite is still poorly understood. In this study, quantitative polymerase chain reaction (qPCR) was performed to assess the expression profile of immune-related genes in L. calcarifer infected by C. irritans. A total of 21 immune-related genes encoding various functions in the fish immune system were utilized for the qPCR analysis. The experiment was initiated with the infection of juvenile fish by exposure to theronts from 200 C. irritans cysts, and non-infected juvenile fish were used as controls. Spleen, liver, gills and kidney tissues were harvested at three days post-infection from control and infected fish. In addition, organs were also harvested on day-10 post-infection from fish that had been allowed to recover from day-4 up to day-10 post-infection. L. calcarifer exhibited pathological changes on day-3 post-infection with the characteristic presence of white spots on the entire fish body, excessive mucus production and formation of a flap over the fish eye. High quality total RNA was extracted from all tissues and qPCR was performed. The qPCR analysis on the cohort of 21 immune-related genes of the various organs harvested on day-3 post-infection demonstrated that most genes were induced significantly (p < 0.05) in all tissues, particularly liver (11/21 genes) and kidney (11/21). The expression profile demonstrated that induction of the MHC Class IIα gene was the highest compared to the other genes followed by serum amyloid A, CC chemokine and hepcidin-2 precursor genes. In fish that were allowed to recover from the C. irritans infection (10 days post-infection), expression of the immune-related genes was down-regulated to levels similar to the control fish. These results provide insights into the interaction between C. irritans and L. calcarifer and suggest that the innate immune system plays an important role in early defence against parasite infection allowing the fish to eventually recover from the infection.  相似文献

The present study was conducted to investigate the effects of dietary chitooligosaccharides (COS) supplementation on the innate immune response and protection against Vibrio harveyi infection in Trachinotus ovatus. A basal diet was supplemented with 0.0 (control), 2.0, 4.0 and 6.0 g COS kg?1 to formulate four experimental diets. Each diet was randomly allocated to triplicate groups of ?sh in ?oating sea cages (1.5 × 1.0 × 2.0 m), and each cage was stocked with 80 ?sh (initial average weight 10.8 ± 0.05 g). After 8 weeks of feeding trial, Both the final weight and specific growth rate (SGR) signi?cantly increased with increasing dietary COS levels up to 4.0 g kg?1, whereas there were no signi?cant differences for COS levels from 4.0 to 6.0 g kg?1. A decreased feed conversion ratio (FCR) was observed with increasing dietary COS levels. The total leukocyte counts (WBC), differential leukocyte counts, respiratory burst activity, lysozyme and superoxide dismutase (SOD) activity were signi?cantly increased with the increased levels of dietary COS (P < 0.05), and reached a maximum at level of 4.0 g kg?1 COS. There were no signi?cant differences in those immunological parameters between 4.0 and 6.0 g kg?1 COS. Moreover, the dietary COS supplementation groups also exhibited a decrease in the cumulative symptom rates compared to the controls when challenged with V. harveyi. These results indicated that dietary intake containing COS could enhance the immune responses of fish and improve its resistance to infection by V. harveyi. Especially supplementation with 4.0 g kg?1 COS to the fish for 56 days showed considerable improvement in the growth, survival and immune response of the fish.  相似文献
