目的:构建小鼠RelA 基因的RNA 干扰慢病毒载体,转染小鼠成骨样细胞并鉴定。方法:针对小鼠RelA 基因序列,设计特异 性的shRNA 序列,应用基因重组技术插入慢病毒载体GV-248。得到的重组质粒转化感受态大肠杆菌DH5-alpha,筛选得到阳性克隆 并扩大培养。所得质粒进行测序分析确定载体构建成功。重组质粒载体及包装辅助质粒转染293T 细胞,得到目的病毒并测定相 应病毒滴度。慢病毒转染MC3T3-E1 细胞后,Real-time PCR 及Western blot 检测MC3T3-E1 细胞RelA 基因及成骨相关基因 ALP、OCN、RANKL的表达。结果:成功构建小鼠RelA 基因的RNA干扰慢病毒载体,感染MC3T3-E1 细胞后,RelA 基因的表达 明显受到抑制,同时RANKL基因表达水平明显下降,ALP、OCN基因表达水平明显上升。结论:成功构建了小鼠RelA 基因的 RNA 干扰慢病毒载体。当小鼠成骨细胞RelA基因表达被干扰,NF-资B 通路被抑制后,小鼠成骨细胞成骨相关基因ALP、OCN的 表达明显上升,成骨功能增强;同时RANKL 的表达明显下降,其介导的破骨细胞骨吸收功能减弱。  相似文献

Abstract : Studies on iron uptake into the brain have traditionally focused on transport by transferrin. However, transferrin receptors are not found in all brain regions and are especially low in white matter tracts where high iron concentrations have been reported. Several lines of research suggest that a receptor for ferritin, the intracellular storage protein for iron, may exist. We present, herein, evidence for ferritin binding sites in the brains of adult mice. Autoradiographic studies using 125I-recombinant human ferritin demonstrate that ferritin binding sites in brain are predominantly in white matter. Saturation binding analyses revealed a single class of binding sites with a dissociation constant ( K D) of 4.65 × 10-9 M and a binding site density ( B max) of 17.9 fmol bound/μg of protein. Binding of radiolabeled ferritin can be competitively displaced by an excess of ferritin but not transferrin. Ferritin has previously been shown to affect cellular proliferation, protect cells from oxidative damage, and deliver iron. The significance of a cellular ferritin receptor is that ferritin is capable of delivering 2,000 times more iron per mole of protein than transferrin. 丝氨酸/苏氨酸蛋白激酶2(serine/threonine protein kinase2,Akt 2)是胰岛素信 号通路的关键基因, 与细胞生存和癌症的发生密切相关. 但Akt2在前体脂肪细胞分化中 的作用仍然不是十分清楚. 本研究构建了慢病毒干扰载体pLentiH1-Akt2-shRNA 1, 2, 3及scrambled, 经酶切和测序鉴定均正确; 这4种干扰载体分别转染HEK293T细胞后, 均 获得有感染性的病毒颗粒并感染猪前体脂肪细胞. 转染HEK293T细胞48 h 后real-time PCR分析和转染72 h 后Western 印迹分析表明, Akt2-shRNA2 和shRNA3 介导的Akt2 mRNA和蛋白表达被显著下调 (P <0.05), 其中pLentiH1-Akt2-shRNA3 介导的对Akt2 mRNA和蛋白的干扰效率均达到70%以上 (P <0.01). 进一步研究发现, 猪前体脂肪细胞 中Akt2被有效干扰后, 细胞中脂滴明显减少并变小, 且成脂标志基因PPARγ和aP2蛋白 水平被显著下调. 本研究结果表明, Akt2 knockdown可显著抑制猪前体脂肪细胞分化.  相似文献

The relative levels of microtubule-associated protein 2(MAP2) were determined during postnatal development of the mouse in six different discrete brain regions: cerebellum, cortex, hippocampus, olfactory bulb, brainstem, and hypothalamus. Brain homogenates were electrophoresed on sodium dodecyl sulfate-containing gels and analyzed by immunoblotting with MAP2-specific antibodies. The levels of MAP2 in each region were determined using radiolabeled secondary antibodies and densitometric quantification of the autoradiograms over a range that was determined to have a linear response. The results indicated that in all regions and at all ages there was only one high-molecular-weight polypeptide of MAP2, which did not change in electrophoretic mobility after dephosphorylation. In most regions, the levels of MAP2 increased during the first 2 postnatal weeks. However, there were differences in the time course and relative levels of MAP2 between regions. In addition, all regions of the brain expressed the low-molecular-weight form of MAP2 (MAP2c) that was present at birth as a heterogeneous group of polypeptides with an apparent molecular weight of 70K. Most of the heterogeneity of MAP2c, however, was eliminated after dephosphorylation. The levels of MAP2c decreased dramatically after 2 weeks postnatally, except for the olfactory bulb, where the levels of MAP2c remained relatively high even in adults.  相似文献

目的:应用RNA干扰(RNAi)技术调节大鼠c-jun基因在Cos-7细胞中的表达.方法:分别构建大鼠c-jun基因的RNA干扰载体和真核表达GFP(绿色荧光蛋白)载体,将两者共转染Cos-7细胞,镜下观察大鼠C-Jun-GFP融合蛋白的表达,应用Western blot方法检测抑制效率.结果:酶切和测序结果表明,大鼠c-jun基因的RNA干扰载体和真核表达GFP载体构建成功,镜下及Western blot共转染结果均显示随着RNA干扰载体浓度的增加C-Jun-GFP融合蛋白表达量逐渐减少.结论:在Cos-7细胞中应用RNAi技术成功调节大鼠c-jun基因的表达.  相似文献

从不同年龄(20天,30天,1年)的小白鼠全脑制得细胞质混合氨酰tRNA合成酶。用异源体系(即用酵母tRNA和小白鼠全脑氨酰tRNA合成酶)测定了氨酰tRNA合成酶分别载运~3H标记的Asp、Gly、Glu、Lys和Ala的活力。结果表明除未检出tRNA~(Glu)的合成酶活力外,对其余四种氨基酸都有明显的活力,特别是年龄20天小白鼠的氨酰tRNA合成酶对~3H-Gly具有高达35%的载运活力。对~3H-Gly、~3H-Lys和~3H-Ala的载运活力有随增龄而下降的趋势,但对~3H-Asp的载运活力则随年龄增长而增高。  相似文献

目的:探讨特异性抑制NRP2基因表达对人胃癌细胞SGC7901裸鼠移植瘤生长及淋巴管新生的影响。方法:采用小RNA干扰方法,构建shNRP2质粒,稳定转染入SGC7901细胞株,Westen blot检测转染前后NRP2蛋白的表达。建立人胃癌细胞SGC7901裸鼠移植瘤模型,随机分为shNRP2组(实验组)、shCon组(HK阴性对照组)和正常对照组,观察移植瘤的生长情况。6周后处死裸鼠,免疫组化检测NRP2蛋白的表达及微淋巴管密度(Micro-vessel density,MLD)。结果:成功构建shNRP2质粒,与另两组比较,shNRP2组细胞NRP2蛋白表达明显降低,且移植瘤组织生长、NRP2表达及MLD明显受抑制。结论:抑制NRP2基因的表达,可以抑制胃癌裸鼠移植瘤的生长及淋巴管的形成,NRP2基因有可能成为一个潜在的胃癌生物治疗靶点。  相似文献

Abstract: Using immunocytochemical localization, the distribution of the glycine transporters GLYT1 and GLYT2 in the developing mouse brain was studied. GLYT1 and GLYT2 immunoreactivity begins during the period of fiber outgrow and synaptogenesis. GLYT2 is first expressed in spinal and spinothalamic white matter and is followed by the expression of synaptophysin. In the postnatal stages, GLYT2 staining in the white matter disappears, and a punctuated pattern in the gray matter emerges. In contrast, in the fetal brain GLYT1 immunoreactivity coincides with gray matter neuropil and processes of radial glia. GLYT1 is distributed over a much wider area of the brain than GLYT2. However, the distribution of these two GLYTs implies that GLYT1 and GLYT2 operate in concert within the area where both are present. At the day 12 embryo stage, GLYT1 antibodies stain the liver, and later they also react with the pancreas and the gastroduodenal junction. No other organs exhibit significant GLYT1 immunoreactivity. We additionally observed the presence of GLYT1 in rat fetal cerebral cortex and hippocampus, which was not detected in fetal mouse brain. Moreover, GLYT1 immunoreactivity was found in the mouse floor plate and the ventral commissure but was not present in the same regions in rats. These findings suggest possible differences in the expression of GLYT1 between these two species.  相似文献

We isolated a mouse genomic clone that hybridized with small RNA present in the cytoplasm of the brain. The RNA was about 150 nucleotides long. This RNA seemed to be specific to the brain, since it was not found in the liver or kidney. The clone DNA contained a sequence homologous to 82-nucleotide "identifier" core sequence of cDNA clones of rat. The sequence contained a split promoter for RNA polymerase III and was flanked by a 12-nucleotide direct repeat (ATAAATAATTTA).  相似文献

为了研究慢病毒介导的shRNA(Short hairpin RNA,shRNA)在柯萨奇B组3型病毒(Coxsackievirus B3,CVB3)导致的心肌炎小鼠模型中的抗病毒作用,合成针对CVB3基因组3753~3771区域的慢病毒Lenti-sh3753,感染HeLa细胞后感染CVB3病毒,通过荧光显微镜观测shRNA的表达和病毒致细胞病变效应,并测定培养上清中的病毒滴度,将慢病毒Lenti-sh3753感染BALB/c小鼠后感染CVB3病毒,观察小鼠的存活率,心脏组织中的病毒滴度和病理变化。结果发现Lenti-sh3753能在HeLa细胞中表达shRNA,并能有效抑制细胞中病毒RNA的复制。在小鼠模型上,Lenti-sh3753能提高小鼠的存活率,降低心脏中的病毒含量,从而减轻病理反应。这些结果提示,Lenti-sh3753在细胞和动物模型中能针对性地降解CVB3病毒RNA,明显降低病毒滴度,有效控制病毒感染。  相似文献

据世界卫生组织(WHO)报道,全世界约有20亿人曾感染过乙型肝炎病毒(HBV),其中3.5亿人为慢性HBV感染者.我国现有1.3亿乙肝病毒携带者和3 000多万乙肝患者,其中约有20%~40%的患者经过多年慢性炎症的反复发作可发展为肝硬化和肝癌.然而,至今人们仍没有找到一种能够彻底治愈慢性乙肝的特效药物.自从RNA干扰(RNA interference,RNAi)技术建立以来,人们致力于将其应用于抗病毒药物的研究与开发.研究结果表明,RNAi可有效地抑制乙肝病毒的复制,但靶向目的基因的不同RNA干扰片段所沉默的效率不同.关于将RNAi抗病毒药物应用于人体治疗的安全性和有效性还有待进一步研究,RNAi发生"脱靶"的现象是临床应用的难点之一.  相似文献

Brain Tubulin Microheterogeneity in the Mouse During Development and Aging   总被引:5,自引:3,他引:2  
Abstract: Mouse brain tubulin was analyzed on isoelectric focusing gels. High-resolution gels utilizing Bio-Rad ampholytes (pH 4-6) revealed 5-6 bands in the region corresponding to the α-subunit of tubulin and 10 or more for the β-subunit. The same general banding pattern was observed regardless of the method of preparation of the tubulin. Two species prominent in the brains of immature mice, α6 and β2, virtually disappeared during maturation, while species β6 to β10 appeared. No significant changes from the mature pattern were seen during aging (examined at 12, 23, and 30 months of age).  相似文献
