Saposins: structure, function, distribution, and molecular genetics.   总被引:11,自引:0,他引:11  
Saposins A, B, C, and D are small heat-stable glycoproteins derived from a common precursor protein, prosaposin. These mature saposins, as well as prosaposin, activate several lysosomal hydrolases involved in the metabolism of various sphingolipids. All four saposins are structurally similar to one another including placement of six cysteines, a glycosylation site, and conserved prolines in identical positions. In spite of the structural similarities, the specificity and mode of activation of sphingolipid hydrolases differs among individual saposins. Saposins appear to be lysosomal proteins, exerting their action upon lysosomal hydrolases. Prosaposin is a 70 kDa glycoprotein containing four domains, one for each saposin, placed in tandem. Prosaposin is proteolytically processed to saposins A, B, C and D, apparently within lysosomes. However, prosaposin also exists as an integral membrane protein not destined for lysosomal entry and exists uncleaved in many biological fluids such as seminal plasma, human milk, and cerebrospinal fluid, where it appears to have a different function. The physiological significance of saposins is underlined by their accumulation in tissues of lysosomal storage disease patients and the occurrence of sphingolipidosis due to mutations in the prosaposin gene. This review presents an overview of the occurrence, structure and function of these saposin proteins.  相似文献

The Pantanal is one of the faunistic provinces considered as a priority area for invertebrate conservation. However, it is one of the areas in Brazil where the local fauna is less assessed, thus needing more scientific information that could allow political decisions to be made regarding conservation. The continuous pressure for new pasture areas leads to improper habitat occupation and destruction, like fragmentation of forest areas in the region. Such alterations can cause different impacts on the local fauna, including the soil arthropods. The main objective of this work was to compare the morphospecies composition, diversity and density of the soil arthropod fauna between a secondary single species forest (Cambarazal) and a cultivated pasture with exotic and native grass species, using only pitfall traps as sampling method. We found a great variation on the vegetal cover among environments. A higher humidity in the forest soil was observed, as well as a greater compaction of the soil in the cultivated pasture. A total of 3635 individuals were collected, belonging to 214 different morphospecies. 139 morphospecies were collected in the forest (37% exclusive to this environment), while 134 morphospecies were collected in the cultivated pasture (35% exclusive). The diversity was higher in the forest (H′ = 1.634) than in the cultivated pasture (H′ = 1.253). However, considering the area as a whole (forest and pasture) the global diversity was increased. In this paper we discuss about the effects of environmental changes on soil arthropod diversity and propose a hypothetical model for invertebrate management in mosaic ecosystems.  相似文献

The Invertebrate section of the Museum of Zoology QCAZ at the Pontifical Catholic University of Ecuador in Quito maintains nearly two million curated specimens, and comprises Ecuador’s largest collection of native taxa. We review 1902 type specimens from 6 subspecies and 320 species in 121 genera and 42 families, currently kept in the Museum. The list includes 116 holotypes, 10 allotypes, 1774 paratypes and 2 neoparatypes. The collection of type specimens is particularly strong in the Coleoptera (family Carabidae and Staphylinidae) and Hymenoptera. However, other insect orders such as Diptera and Lepidoptera and non-insect arthropods such as Acari, Aranea and Scorpiones, are moderately represented in the collection. This report provides original data from labels of every type specimen record. An analysis of the geographic distribution of type localities showed that collection sites are clustered geographically with most of them found towards the northern region of Ecuador, in Pichincha, Cotopaxi and Napo provinces. Sites are mainly located in highly accessible areas near highways and towns. Localities with a high number of type species include the cloud forest reserve Bosque Integral Otonga and Parque Nacional Yasuní in the Amazon rainforest near PUCE’s Yasuní Scientific Station. Type localities are not well represented in the Ecuadorian National System of Protected Areas. Future fieldwork should include localities in the southern region of Ecuador but also target less accessible areas not located near highways or towns. We discuss the value of the collection as a source of information for conservation and biodiversity policies in Ecuador.  相似文献

The diversity and distribution of modern benthic foraminifera has been extensively studied in order to aid the paleoecological interpretation of their fossil record. Traditionally, foraminiferal species are identified based on morphological characters of their organic, agglutinated or calcareous tests. Recently, however, new molecular techniques based on analysis of DNA sequences have been introduced to study the genetic variation in foraminifera. Although the number of species for which DNA sequence data exist is still very limited, it appears that morphology-based studies largely underestimated foraminiferal diversity. Here, we present two examples of the use of DNA sequences to examine the diversity of benthic foraminifera. The first case deals with molecular and morphological variations in the well-known and common calcareous genus Ammonia. The second case presents molecular diversity in the poorly documented group of monothalamous (single-chambered) foraminifera. Both examples perfectly illustrate high cryptic diversity revealed in almost all molecular studies. Molecular results also confirm that the majority of foraminiferal species have a restricted geographic distribution and that globally distributed species are rare. This is in opposition to the theory that biogeography has no impact on the diversity of small-sized eukaryotes. At least in the case of foraminifera, size does not seem to have a main impact on dispersal capacities. However, the factors responsible for the dispersal of foraminiferal species and the extension of their geographic ranges remain largely unknown. 碱性螺旋-环-螺旋(Basic helix-loop-helix protein,bHLH)家族成员Twist2对间质细胞系的发生和发育起转录调节作用,经直接或间接机制发挥分子开关功能,从而激活或抑制靶基因。Twist2能直接结合DNA上 E-box保守序列,招募共激活物或抑制剂;能与E蛋白调节因子发生蛋白-蛋白相互作用,干扰激活或抑制功能。Twist2无义突变导致Setleis综合征。对Twist2的早期研究多集中在骨骼发育,随后在多种肿瘤中发现其有表达差异,研究表明Twist2在肿瘤的上皮-间质转化(Epithelial-mesenchymal transition,EMT)中发挥着重要作用。Twist2参与了多条通路的调控,其调控作用的发挥受到时空表达、磷酸化、二聚化和细胞定位的调节,在机体的正常发育、体内平衡和疾病发生机制中研究Twist2的作用显得尤为重要。文章对Twist2在成骨分化、肿瘤形成和EMT中的作用及其分子机制进行综述,以便帮助了解Twist2的生物学功能,为进一步在疾病的诊断、发展、以及治疗等方面的转化应用研究提供依据。  相似文献

Originally identified as transporters for inorganic phosphate, solute carrier 17 (SLC17) family proteins subserve diverse physiological roles. The vesicular glutamate transporters (VGLUTs) package the principal excitatory neurotransmitter glutamate into synaptic vesicles (SVs). In contrast, the closely related sialic acid transporter sialin mediates the flux of sialic acid in the opposite direction, from lysosomes to the cytoplasm. The two proteins couple in different ways to the H+ electrochemical gradient driving force, and high-resolution structures of the Escherichia coli homolog d-galactonate transporter (DgoT) and more recently rat VGLUT2 now begin to suggest the mechanisms involved as well as the basis for substrate specificity.  相似文献

Petals, a characteristic feature of eudicots, have evolved elaborations in various ways and across diverse clades. In this survey of petal and staminode elaborations throughout the eudicots, based on both new studies and a review of the literature, the diversity of such structures and their functions is discussed. Petal elaborations are primarily present as marginal lobes and ventral lobes of various shapes. Lobation patterns can be loosely classified as pinnate, binate, or ternate. One of these patterns may be dominant within a family (e.g. pinnate in Anisophylleaceae, binate in Caryophyllaceae, ternate in Elaeocarpaceae); transitional forms also occur (e.g. between binate and ternate in Onagraceae). Coronas between the corolla and androecium are found in several groups, for example in several families of Malpighiales or in Apocynaceae. In some clades, petal elaborations are especially prominent and can be used as approximate systematic markers (Anisophylleaceae, Elaeocarpaceae, Rhizophoraceae). Petal elaborations are especially diverse in rosids. In asterids, which are characterized by sympetaly, elaborations are more conspicuous at the level of the architecture of the entire corolla, rather than at the level of individual petals. Evolutionary trends in petal elaboration in certain larger clades are shown and their involvement in floral biological functions is discussed.  相似文献

The structural complexity of habitats and the distance from the dispersion source are considered important determinants of species diversity. The fractal dimensions of substrates are a strong alternative measure for quantifying the structural complexity. In this paper, we tested the hypothesis that structural complexity and distance from a dispersion source affect the density and diversity of macro‐invertebrates on submersed macrophytes. To test this hypothesis, we used artificial substrates with different fractal dimensions, which were incubated at different distances from a dispersion source (a stand of macrophytes) in a Neotropical floodplain lake. The relationship between the structural complexity of macrophytes and the density and diversity of invertebrates attached to artificial substrates was positive and significant. On the other hand, distance from the dispersion source negatively affected both the density and diversity of the attached invertebrates. For Oligochaeta, however, there was a significant effect of structural complexity for the artificial plants positioned further away, but it was not significant in the plants positioned closer to the dispersion source. Thus, the effects of complexity depended on the distance from the dispersion source, at least for this specific group of macro‐invertebrates. These results suggest that both structural complexity and distance from the dispersion source should be considered together to explain the diversity of invertebrates in river–floodplain systems. In addition, our results show that there is great potential for using fractal dimensions as a tool to quantify structural complexity in aquatic habitats, especially at the small spatial scales perceived by macro‐invertebrates. The parasitoid assemblages associated with grass-feeding chalcid wasps in Great Britain were used to examine the relationships between diversity (species richness), community function (total parasitism rate) and stability (variability in parasitism rate over time). Species-rich communities did not generate higher parasitism rates than species-poor communities, nor was temporal variation of parasitism rates related to parasitoid community richness. The mechanisms underlying hypotheses linking species richness and community function and stability are discussed in the light of these results. Because all parasitoid species represent a single functional group, a lack of complementarity in the ways they use their resources may explain why diversity is not linked to function or community stability. A second likely reason is that these parasitoid communities are under bottom-up control, thus exerting little or no influence on total system function and variability. This is likely to be common in parasitoid communities.  相似文献

The genetic diversity of haematozoan parasites in island avifauna has only recently begun to be explored, despite the potential insight that these data can provide into the history of association between hosts and parasites and the possible threat posed to island endemics. We used mitochondrial DNA sequencing to characterize the diversity of 2 genera of vector-mediated parasites (Plasmodium and Haemoproteus) in avian blood samples from the western Indian Ocean region and explored their relationship with parasites from continental Africa. We detected infections in 68 out of 150 (45·3%) individuals and cytochrome b sequences identified 9 genetically distinct lineages of Plasmodium spp. and 7 lineages of Haemoproteus spp. We found considerable heterogeneity in parasite lineage composition across islands, although limited sampling may, in part, be responsible for perceived differences. Two lineages of Plasmodium spp. and 2 lineages of Haemoproteus spp. were shared by hosts in the Indian Ocean and also on mainland Africa, suggesting that these lineages may have arrived relatively recently. Polyphyly of island parasites indicated that these parasites were unlikely to constitute an endemic radiation and instead probably represent multiple colonization events. This study represents the first molecular survey of vector-mediated parasites in the western Indian Ocean, and has uncovered a diversity of parasites. Full understanding of parasite community composition and possible threats to endemic avian hosts will require comprehensive surveys across the avifauna of this region.  相似文献

Genetic analysis of nociceptive behaviors in the nematode Caenorhabditis elegans and the fruit fly Drosophila melanogaster has led to the discovery of conserved sensory transduction channels and signaling molecules. These are embedded in neurons and circuits that generate responses to noxious signals. This article reviews the neurons and molecular mechanisms that underlie invertebrate nociception. We begin with the neurobiology of invertebrate nociception, and then focus on molecules with conserved functions in vertebrate nociception and sensory biology.  相似文献

The superfamily of serine proteinase inhibitors (serpins) are involved in a number of fundamental biological processes such as blood coagulation, complement activation, fibrinolysis, angiogenesis, inflammation and tumor suppression and are expressed in a cell-specific manner. The average protein size of a serpin family member is 350-400 amino acids, but gene structure varies in terms of number and size of exons and introns. Previous studies of all known serpins identified 16 clades and 10 orphan sequences. Vertebrate serpins can be conveniently classified into six sub-groups. We provide additional data that updates the phylogenetic analysis in the context of structural and functional properties of the proteins. From these, we can conclude that the functional classification of serpins relies on their protein structure and not on sequence similarity.  相似文献

In the last 20 years interest in fish granulocyte identification, distribution, and function has burgeoned, as evidenced by the increasing number of reports in scientific literature. Neutrophils, eosinophils, and basophils are present in peripheral blood and certain organs of fish. Some species of fish have all three cell types, while other species may only possess one cell type. Granulopoiesis in fish occurs in the spleen, kidneys, epigonal organ, organ of Leydig, and other specialized tissue, with the specific locations depending on the species. In some fish, neutrophils are actively phagocytic. In vitro assays indicate phagocytosis, mobilization in response to chemotaxins, detectable chemiluminescence, and an active respiratory burst when appropriately stimulated. In fish species without neutrophils, the eosinophil may be responsible for phagocytosis. Eosinophils also function in antiparasite immunity in certain species. Basophil function has not been investigated. Responses of fish neutrophils and eosinophils can be altered by season of the year, environmental pollutants, disease, and others stressors. Differences among fish species in type, cellular distribution, and function of granulocytes are the focus of this review.  相似文献

MicroRNAs (miRNAs) are endogenous approximately 22 nt RNAs that can play important regulatory roles in animals and plants by targeting mRNAs for cleavage or translational repression.
