The primary objective of this research project is explore a possible adipogenic effect of iron and/or copper in albino Wistar rats kept on standard (STD) and high-fat (HFD) diets. The female Wistar rats in the study were divided into eight experimental groups (n = 6). Rats maintained on STD and HFD received 3 mg/l FeSO4·7H2O, 4.88 mg/l CuSO4 and a combination of 1.5 mg/l FeSO4·7H2O and 2.44 mg/l CuSO4 with drinking water. Control groups were kept on STD and HFD and received pure water without metal salts. Consumption of iron and copper in the groups of rats maintained on an STD did not produce a significant increase in weight, adipose tissue content or body mass index. However, the adipocyte size and infiltration were increased in the adipose tissue of STD-fed rats receiving a mixture of iron and copper with drinking water. The rats fed iron and copper and, especially, their combination on a HFD background had a significantly higher weight gain, adipose tissue content, morphometric parameters values and adipocyte size compared to STD- and HFD-fed controls. Iron and copper consumption produced their accumulation in the rats’ adipose tissue. Moreover, the studied metals reduced adipose tissue concentration of chromium and vanadium. The lipoprotein profile and serum oxidative stress biomarkers were affected in the rats receiving the metals and STD. Hyperglycemia was observed in the rats receiving the studied metals on HFD-background. Based on the analysis of the test subjects, the study suggests that iron and copper administration, especially combined, may potentiate adipogenic effect of HFD.  相似文献

The fatty acid composition, the effect of different concentrations of nitrogen (16.5-344 mg ?L?1), phosphorus (9–45 mg? L?1), iron (9–45 mg? L?1) and salinity levels (0–20 psu) on lipid production in the green microalga Scenedesmus dimorphus KMITL, a new strain isolated from a tropical country, Thailand, were studied. The alga was isolated from a freshwater fish pond, and cultured in Chlorella medium by varying one parameter at a time. The main fatty acid composition of this strain was C16–C18 (97.52 %) fatty acids. A high lipid content was observed in conditions of 16.5 mg? L?1-N, or 22 mg ?L?1-P, or 45 mg ?L?1-Fe, or 5 psu salinity, which accumulated lipids to 20.3?±?0.4, 19.4?±?0.2, 24.7?±?0.5, and 14.3?±?0.2 % of algal biomass, respectively. Increasing lipid content and lipid productivity was noted when the alga was cultured under high iron concentration and high salinity, as well as under reduced phosphorus conditions, whereas nitrogen limitation only resulted in an increased lipid content.  相似文献

Iris pseudacorus L. (yellow flag) is a wide-use wetland plant for constructed wetlands for removing metals from wastewater. This study aims to understand effects of root iron plaque on sequestration and translocation of Cr and Ni in yellow flag seedlings using a hydroponic experiment. Yellow flag seedlings (4-week-old seedlings with 4–6 leaves) with or without iron plaque induction (at 50 mg Fe2+ L?1 for 72 hours) were spiked for 6 days in the Hoagland solution with Cr or Ni at 0.5, 5, and 50 mg L?1, equivalent to 1, 10, 100 times of thresholds of surface water quality, respectively. Results indicated that root iron plaque significantly reduced translocation of Cr and Ni to root but increased from root to shoot. Root iron plaque formation counteracted Cr toxicity to yellow flag seedlings while the control showed Cr toxicity to root at 5 mg L?1and to shoot at 50 mg L?1 with significant biomass loss. Neither Ni exposures caused significant biomass loss nor root iron plaque formation significantly changed Ni distribution among plant parts. Our study suggests that root iron plaque effects on metal sequestration and translocation in yellow flag seedlings were metal-dependent.  相似文献

The potential of arsenic-resistant bacteria in association with Pteris vittata to reduce the level of arsenic from soil was studied. The physicochemical characteristics of contaminated paddy soil were analyzed, and 3 bacterial isolates amongst 11 were screened and were selected for further study. These three isolates were characterized by 16S rDNA sequencing and identified as Bacillus altitudinis Strain SS8 (KJ432582), Bacillus megaterium Strain SS9 (KJ432583) and Lysinibacillus sp. Strain SS11 (KJ432584). Of these, Lysinibacillus sp. Strain SS11 displayed arsenic tolerance of 3256 mg L?1 for arsenate and 1136 mg L?1 for arsenite. Additionally, it showed bioaccumulation capacity of 23.43 mg L?1 for arsenate and 5.65 mg L?1 for arsenite. It also showed resistance to other heavy metals, especially towards iron, copper and chromium. It was also observed that Pteris vittata was able to take up more arsenic and iron from soil in the presence of these bacterial strains than in their absence, leading to contaminant-free soil. Thus, this system appears to be an effective bioremediating process to remove arsenic from contaminated soil.  相似文献

Antioxidative responses of Elodea nuttallii (Planch.) H. St. John to short-term iron exposure were investigated in the study. Results showed that iron accumulation in E. nuttallii was concentration dependent. Growth of E. nuttallii was promoted by low iron concentration (1–10 mg L?1 [Fe3+]), but growth inhibition was observed when iron concentration beyond 10 mg L?1. The synthesis of protein and pigments increased within 1–10 mg L?1 [Fe3+] range. The activities of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD) and glutathione-S-transferase (GST) were up to maximal values at 10 mg L?1 [Fe3+]. High iron concentration inhibited the synthesis of protein and pigments as well as activities of antioxidative enzymes, and accelerated degradation of pigment and production of ROS. Low iron concentration had no significant influences on PSII maximal quantum yield, activity of PSII and relative electron transport rate though PSII. Malondialdehyde (MDA) and proline concentrations were highest at 100 and 1 mg L?1 [Fe3+], respectively.  相似文献

This article reports the health risk associated with chronic intake of metals in the municipal water supplies of Eleyele and neighboring towns in the Ibadan metropolitan area of Nigeria. A total of 42 composite samples, consisting of treated water from the water treatment facility and residential areas receiving personal-use water (i.e., tap water) directly from the facility, as well as raw water from the treatment facility's water supply dam were sampled twice every month for 7 months. Concentrations of the metals were determined by atomic absorption spectrophotometry. Among the metals studied, Cd, Co, Cr, and Pb were detected at concentrations higher than maximum regulatory limits. Cd, Co, Cr, and Pb concentrations in treated water at the treatment facility ranged from 0.08–0.10, 0.14–0.16, 0.04–0.22 and 0.07–0.36 mg L?1, respectively, while personal-use water ranged from 0.08–0.11, 0.15–0.29, 0.02–0.29, and 0.12–0.65 mg L1, respectively. Likewise, concentrations of the metals at the dam ranged from 0.06–0.08, 0.17–0.20, 0.13–0.37, and 0.03–0.15 mg L1, respectively. It is estimated that exposure to the metals in the water supply results in oncological and non-oncological systemic health risks higher than is generally acceptable for drinking water.  相似文献

Abstract

This study evaluates the biosorption of copper by aerobic biomass that was selected from surface waters of the San Pedro River in Sonora, Mexico. Using a batch system, 73% biosorption of copper was obtained in 75 minutes. Continuous biosorption assays were carried out for 133 days in an ascending flow aerobic reactor packed with zeolite (AFAR-PZ) that was inoculated with a bacterial consortium. Strains were grown until 1g L?1 of biomass was obtained. Tests using continuous biosorption were performed as follows: (i) the addition of 50 mg Cu2+ L?1 without recirculation of biomass; (ii) the addition of 20 mg Cu2+ L?1without recirculation of biomass; and (iii) the biomass were recirculated with the addition of 20 mg Cu2+ L?1 to pH 3 to 4. The fourth and fifth assays varied pH between 4 and 5, with 20 mg Cu2+ L?1and the biomass recirculated. Biosorption capacity of the first and second assays was 96% on the first day of experimentation. During the third trial 97% of biosorption was obtained during 6 days and the process was improved by varying the pH. Copper biosorption equilibrium was investigated under the same operating conditions. Langmuir adsorption isotherms were used to fit experimental data. The biosorption capacity of aerobic biomass was 3.08 mmol g?1. It was demonstrated that this biomass is capable of biosorbing copper and this method has potential for the treatment of industrial effluents contaminated with heavy metals.  相似文献

Most metals disperse easily in environments and can be bioconcentrated in tissues of many organisms causing risks to the health and stability of aquatic ecosystems even at low concentrations. The use of plants to phytoremediation has been evaluated to mitigate the environmental contamination by metals since they have large capacity to adsorb or accumulate these elements. In this study we evaluate Salvinia minima growth and its ability to accumulate metals. The plants were cultivated for about 60 days in different concentrations of Cd, Ni, Pb and Zn (tested alone) in controlled environmental conditions and availability of nutrients. The results indicated that S. minima was able to grow in low concentrations of selected metals (0.03 mg L?1 Cd, 0.40 mg L?1 Ni, 1.00 mg L?1 Pb and 1.00 mg L?1 Zn) and still able to adsorb or accumulate metals in their tissues when cultivated in higher concentrations of selected metals without necessarily grow. The maximum values of removal metal rates (mg m2 day?1) for each metal (Cd = 0.0045, Ni = 0.0595, Pb = 0.1423 e Zn = 0.4046) are listed. We concluded that S. minima may be used as an additional tool for metals removal from effluent.  相似文献

In South China, high manganese content in the drinking water source influenced by upstream manganese mine drainage has become a major concern. To investigate the extent of metal pollution and environmental risk in upstream sediments and native aquatic macrophytes, a study was conducted on a manganese mining-impacted river named the Heishui River. The results indicated that streambed sediments collected were polluted by Mn and other metals with the highest contents of Mn 43349.4 mg kg?1, Pb 128.6 mg kg?1, Zn 502.9 mg kg?1, and Cu 107.2 mg kg?1. The level of Mn in all sediments was higher than the consensus-based Probable Effect Concentration, indicating that adverse effects on sediment-dwelling organisms were likely to occur frequently. Among the studied metals, Mn had the highest bioavailability and ecological risk, followed by Zn. Native aquatic macrophytes accumulate large amounts of the studied metals. A significantly positive correlation was found between exchangeable fractions of the studied metals in sediments and in aquatic macrophytes. The risk assessment code showed the following risk levels of metals in sediments in descending order: Mn > Zn > Cu > Pb. In conclusion, the river impacted by manganese mining drainage poses a high risk to both the local ecosystem and downstream drinking water.  相似文献

Diatoms are a group of highly abundant and diverse aquatic algae species. They contain high lipid content along with many bioactive compounds that can be exploited for biotechnological applications. Despite these attractive attributes, diatoms are underrepresented in production projects due to difficulties in their cultivation. To optimize the growth of three freshwater diatom isolates, Cyclotella sp., Synedra sp. and Navicula sp., an orthogonal assay on N, P, Si and Fe, as well as temperature and pH, was designed using traditional single‐factor tests. We also studied the effect of using nanosilica as an alternate Si source on growth and found that the diatom isolates studied achieved their highest growth rates under different combinations of nutrient and environmental conditions. Silica had the greatest influence on growth, followed by phosphate and iron. The optimized growth conditions for Synedra sp. were N: 30 mg L?1, P: 3 mg L?1, Si: 14.8 mg L?1, Fe: 0.448 mg L?1, temperature 25°C and pH 8. For Navicula sp.: N: 20 mg L?1, P: 2.5 mg L?1, Si: 19.7 mg L?1, Fe: 0.112 mg L?1, temperature 30°C and pH 7.5–8. For Cyclotella sp.: N: 20 mg L?1, P: 2.5 mg L?1, Si: 19.7 mg L?1, Fe: 0.448 mg L?1, temperature 30°C and pH 7.5–8. Nano silica negatively affected growth in Navicula sp. and Cyclotella sp., but no such effect was observed in Synedra sp. Fatty acid profiling showed C16:0, C16:1(n ? 7), C18:0 and C20:5(n ? 3) as major fatty acids, with no significant differences in fatty acid methyl ester profiles between traditional and modified media. This work gives us a new insight into the growth requirements of freshwater diatom species, which are less studied than marine species.  相似文献

Abstract

Water quality characteristics affecting toxicity of metals to aquatic life include pH, inorganic and organic ligands (negatively charged ions and molecules), and water hardness. Ligands control the ability of natural waters to bind metals which could adversely affect aquatic life. Bioavailability of metals in natural waters is primarily controlled by alkalinity. Hardness does not affect metal complexation but can reduce acute toxicity through antagonistic mechanisms. In most natural waters, concentrations of alkalinity and hardness are similar, but they may be very different in some waters.

Most toxicity studies have not distinguished between reduced toxicity resulting from effects of hardness and that resulting from complexation of metals by ligands. A series of acute and long-term experiments were conducted to assess these relationships while exposing rainbow trout (Oncorhynchus mykiss) to cadmium (Cd) in waters of low alkalinity (30 mg L?1) and hardnesses of 400, 200, and 50 mg L?1 adjusted with magnesium sulfate (MgSO4). These tests did not show a strong antagonistic influence of Mg hardness on Cd toxicity. At Mg hardnesses of 50, 200, and 400 mg L?1, 96-h LC50s were 3.02, 6.12, and 5.70 μg Cd L?1, differing by a factor of only 1.8. Similarly, chronic values derived from 100-day experiments in waters with the same range of hardness were 1.47, 3.57, and 3.64 μg L?1, respectively. With an eight-fold difference in Mg hardness, chronic values differed by a factor of only 2.5. Antagonistic properties of hardness are primarily controlled by Ca with Mg playing a minor role. The long-term role of Ca in reducing metal toxicity will require further investigation.  相似文献

Abstract

The concentration, mobility, bioavailability, distribution and associations of two essential micronutrient elements (copper and zinc) to the oil palm in wetland soils of the Niger delta region of Nigeria was assessed by means of chemical fractionation analysis. The water soluble and plant available fractions were introduced into the sequential extraction scheme. Also assessed were the bioaccumulation (concentration and distribution) of these metals in the leaves and fruits of the oil palm of various ages found around the soil profiles. Copper was obtained more in the amorphous Fe-oxide fraction (151.05 mg kg?1), but evenly distributed in the exchangeable component (16.16%) with a maximum value of 126.6 mg kg?1. The water soluble and plant available fractions had 16.15 and 7.54% distribution of Cu respectively. Zinc had 2.35 and 30.42% distribution in the water soluble and plant available fractions respectively. The lowest mean amount of Cu (1.33 mg kg?1) was determined in the leaves of palms of ages 15–32 years. Palms greater than 60 years had the highest mean concentration of copper (3.91 mg kg?1) in the leaves while the endosperm (kernel) of palms between 2 and 10 years had concentration of 9.07 mg kg?1. The fibrous oily mesocarp had the highest amount (16.78 mg kg?1) of copper in the older palms (>60 years). Similarly, Zn was dominant in the older palms with a mean concentration of 187.14mg kg?1 obtained in the endosperm (kernel). The bioaccumulation pattern of both metals by the palms irrespective of age and the fractionation analysis revealed sufficient bioavailable and reserved amounts of Zn and Cu in the wetland soils.  相似文献

A deltamethrin containing insecticide formulation (Decis®) was evaluated for its toxic potential in developing chick embryos. For the present study, three water emulsified concentrations of Decis® (12.5 mg L?1, 25 mg L?1, and 50 mg L?1) were used. Fertilized eggs of Gallus domesticus were immersed in these three concentrations of the insecticide for 60 min at 37°C on day 0 of incubation and kept for incubation till embryonic day 7. Recovered embryos were evaluated for teratogenic and biochemical changes. The results revealed that administration of Decis® at its lower concentrations (12.5 mg L?1 and 25 mg L?1) did not show any significant teratological changes but the significant number of abnormal survivors was observed at 50 mg L?1 of dose concentration when compared with vehicle-treated control. Among biochemical changes, total glycogen and RNA contents of embryos was significantly decreased at 25 mg L?1 and 50 mg L?1 of Decis® concentrations. Similarly, significant alteration (p ≤ .05) was observed in alanine transaminase activity at 50 mg L?1 concentration of Decis®. Thus, the present study concluded that the no-effect-level for developmental toxicity for Decis® is below the concentration of 25 mg L?1 under standard laboratory conditions.  相似文献

There has been considerable interest in cultivation of green microalgae (Chlorophyta) as a source of lipid that can alternatively be converted to biodiesel. However, almost all mass cultures of algae are carbon-limited. Therefore, to reach a high biomass and oil productivities, the ideal selected microalgae will most likely need a source of inorganic carbon. Here, growth and lipid productivities of Tetraselmis suecica CS-187 and Chlorella sp were tested under various ranges of pH and different sources of inorganic carbon (untreated flue gas from coal-fired power plant, pure industrial CO2, pH-adjusted using HCl and sodium bicarbonate). Biomass and lipid productivities were highest at pH 7.5 (320?±?29.9 mg biomass L?1 day?1and 92?±?13.1 mg lipid L?1 day?1) and pH 7 (407?±?5.5 mg biomass L?1 day?1 and 99?±?17.2 mg lipid L?1 day?1) for T. suecica CS-187 and Chlorella sp, respectively. In general, biomass and lipid productivities were pH 7.5?>?pH 7?>?pH 8?>?pH 6.5 and pH 7?>?pH 7.5?=?pH 8?>?pH 6.5?>?pH 6?>?pH 5.5 for T. suecica CS-187 and Chlorella sp, respectively. The effect of various inorganic carbon on growth and productivities of T. suecica (regulated at pH?=?7.5) and Chlorella sp (regulated at pH?=?7) grown in bag photobioreactors was also examined outdoor at the International Power Hazelwood, Gippsland, Victoria, Australia. The highest biomass and lipid productivities of T. suecica (51.45?±?2.67 mg biomass L?1 day?1 and 14.8?±?2.46 mg lipid L?1 day?1) and Chlorella sp (60.00?±?2.4 mg biomass L?1 day?1 and 13.70?±?1.35 mg lipid L?1 day?1) were achieved when grown using CO2 as inorganic carbon source. No significant differences were found between CO2 and flue gas biomass and lipid productivities. While grown using CO2 and flue gas, biomass productivities were 10, 13 and 18 %, and 7, 14 and 19 % higher than NaHCO3, HCl and unregulated pH for T. suecica and Chlorella sp, respectively. Addition of inorganic carbon increased specific growth rate and lipid content but reduced biomass yield and cell weight of T. suecica. Addition of inorganic carbon increased yield but did not change specific growth rate, cell weight or content of the cell weight of Chlorella sp. Both strains showed significantly higher maximum quantum yield (Fv/Fm) when grown under optimum pH.  相似文献

Higher lipid production and nutrient removal rates are the pursuing goals for synchronous biodiesel production and wastewater treatment technology. An oleaginous alga Chlorella sp. HQ was tested in five different synthetic water, and it was found to achieve the maximum biomass (0.27 g L?1) and lipid yield (41.3 mg L?1) in the synthetic secondary effluent. Next, the effects of the stationary phase elongation and initial nitrogen (N) and phosphorus (P) concentrations were investigated. The results show that the algal characteristics were affected apparently under different N concentrations but not P, which were verified by Logistic and Monod models. At the early stationary phase, the algal biomass, lipid and triacylglycerols (TAGs) yields, and P removal efficiency increased and reached up to 0.19 g L?1, 46.7 mg L?1, 14.3 mg L?1, and 94.3 %, respectively, but N removal efficiency decreased from 86.2 to 26.8 % under different N concentrations. And the largest TAGs yield was only 6.4 mg L?1 and N removal efficiency was above 71.1 % under different P concentrations. At the late stationary phase, the maximal biomass, lipid and TAGs yields, and P removal efficiencies primarily increased as the initial N and P concentrations increase and climbed up to 0.49 g L?1, 99.2 mg L?1, 54.0 mg L?1, and 100.0 %, respectively. It is concluded that stationary phase elongation is of great importance and the optimal initial N/P ratio should be controlled between 8/1 and 20/1 to serve Chlorella sp. HQ for better biodiesel production and secondary effluent purification.  相似文献

Mature embryos of einkorn (Triticum monococcum ssp. monococcum) and bread (Triticum aestivum L.) wheat were used for callus induction on media containing four different doses (0, 1, 2 and 4 mg L?1) of 2,4-D and dicamba supplemented with five different boron concentrations (0, 6.2, 12.4, 24.8, and 37.2 mg L?1). The obtained callus was transferred to culture media with three (0, 0.5, and 2 mg L?1) different BAP doses with five boron concentrations for further regeneration. The maximum callus weight in einkorn wheat was in culture media with 1 mg L?1 dicamba and 6.2 mg L?1 (3.71?±?0.13 g). Bread wheat had the maximum callus weight on culture media with 4 mg L?1 dicamba and 12.4 mg L?1 (3.46?±?0.40 g). The highest plantlet numbers were in only 2 mg L?1 BAP (2.92?±?0.88) for einkorn wheat and 0.5 mg L?1 BAP supplemented with 6.2 mg L?1 boron (3.71?±?1.12) for bread wheat. This indirect regeneration protocol using mature embryos of einkorn and bread wheat under boron stresses expected to be useful for future wheat breeding studies.

The effects of five (5 000, 10 000, 15 000, 20 000, 24 000 kg ha?1 year?1) different doses of organic fertilizer (cow dung) were studied on pond productivity in terms of plankton production and fish biomass in freshwater fish ponds. The grow out period was 60 days. Physico-chemical factors of pond waters were also monitored. With an increase in the fertilizer dose, biochemical oxygen demand (BOD) (1.7 ± 0.1 – 10.35 ± 0.05 mg L?1), O-PO4 (0.04 ± 0.0 – 0.77 ± 0.02 mg L?1) and NH4-N (0.03 ± 0.02 – 0.32 ± 0.02 mg L?1) increased significantly (P < 0.05). Alkalinity (79.0 ± 1.6 – 164.0 ± 3.8 mg L?1) also increased with the increase in fertilizer dose, declining after 60 and 75 days (48.8 ± 1.13 – 67.9 ± 2.1 mg L?1). NO3-N was maximum (1.66 ± 0.2 mg L?1) in the ponds which received cow dung at 15 000 kg ha?1 year?1, and declined (0.94 ± 0.5 mg L?1) at higher doses. Dissolved oxygen (DO) remained significantly high (4.7 mg L?1) up to the third (15 000 kg ha?1 year?1) treatment. Highest plankton population (phytoplankton 17 350.0 ± 1 250.0 no L?1), zooplankton (373.0 ± 22.0 no L?1), species diversity (phytoplankton 3.0, zooplankton 2.3), fish biomass (4.45 kg) and specific growth rate (SGR) (2.36 % body weight (BW) d?1) were also observed in ponds which were treated with fertilizer at 15 000 kg ha?1 year?1. However, at higher doses, a decline in these parameters (phytoplankton, 0.0 – 8 810.0 ± 690.0 no L?1; zooplankton, 0.0 – 205.0 ± 25.0 no L?1; fish biomass, 2.3 kg; SGR, 1.25 % body weight (BW) d?1) was observed. Furthermore, with a decrease in the water temperature from 24 °C (on day 60) to 21 °C (on day 75), a decline in nutrient release, plankton population L?1 and species diversity was observed. Sediment analysis indicated that with an increase in the fertilizer dosage, a significant and progressive increase in the accumulation of organic carbon (0.787 ± 0.006 – 0.935 ± 0.01), total nitrogen (0.877 ± 0.071 – 1.231 ± 0.03), NH4-N (54.4 ± 0.57 – 68.95 ± 0.81), NO3-N (78.5 ± 1.21 – 98.5 ± 0.35), total P (140.0 ± 0.50 – 151.0 ± 1.27) and soluble P (7.15 ± 0.18 – 10.1 ± 0.56) took place; similarly, electrical conductivity (EC) values of sediment also increased progressively (from 200.0 ± 7.1–300.0 ± 10.63 μ mhos cm?1).  相似文献

The microalgae Chlorella vulgaris produce lipids that after extraction from cells can be converted into biodiesel. However, these lipids cannot be efficiently extracted from cells due to the presence of the microalgae cell wall, which acts as a barrier for lipid removal when traditional extraction methods are employed. Therefore, a microalgae system with high lipid productivity and thinner cell walls could be more suitable for lipid production from microalgae. This study addresses the effect of culture conditions, specifically carbon dioxide and sodium nitrate concentrations, on biomass concentration and the ratio of lipid productivity/cellulose content. Optimization of culture conditions was done by response surface methodology. The empirical model for biomass concentration (R2 = 96.0%) led to a predicted maximum of 1123.2 mg dw L?1 when carbon dioxide and sodium nitrate concentrations were 2.33% (v/v) and 5.77 mM, respectively. For lipid productivity/cellulose content ratio (R2 = 95.2%) the maximum predicted value was 0.46 (mg lipid L?1 day?1)(mg cellulose mg biomass?1)?1 when carbon dioxide concentration was 4.02% (v/v) and sodium nitrate concentration was 3.21 mM. A common optimum point for both variables (biomass concentration and lipid productivity/cellulose content ratio) was also found, predicting a biomass concentration of 1119.7 mg dw L?1 and lipid productivity/cellulose content ratio of 0.44 (mg lipid L?1 day?1)(mg cellulose mg biomass?1)?1 for culture conditions of 3.77% (v/v) carbon dioxide and 4.01 mM sodium nitrate. The models were experimentally validated and results supported their accuracy. This study shows that it is possible to improve lipid productivity/cellulose content by manipulation of culture conditions, which may be applicable to any scale of bioreactors. Biotechnol. The microalga Nannochloropsis sp. was cultured under different initial population densities (IPDs) ranging from 0.11 to 9.09 g L?1. The IPD affected the biomass and lipid accumulation significantly. The algal cultured with higher IPD resulted higher biomass concentration (up to 13.07 g L?1) in 10 days growth. The biomass productivity with 0.98 g L?1 IPD was 0.75 g L?1 d?1 which was higher than that of other IPDs. For IPDs ranging from 0.11 to 0.98 g L?1, with the increase of IPD, the biomass productivity increased, while for IPD over 0.98 g L?1, the biomass productivity decreased. Lipid content of the algal culture started with 0.11 g L?1 IPD reached to 42 % of dry weight. But with the increase of IPD, the lipid content decreased. Lipid composition was analyzed using thin layer chromatography coupled with flame ionization detection (TLC/FID). Seven lipid classes were identified and quantified. The main reserve lipid, triacylglyceride (TAG), accumulated under all different IPD conditions. However, with the increasing IPD values, TAG content decreased from 59.1 to 23.5 % of the total lipids. Based on these results, to obtain the maximal biomass productivity and lipid productivity of Nannochloropsis sp. in mass cultivation systems, it is necessary to select an appropriate IPD.  相似文献

Abstract

The aim of this study was to determine by static bioassay whether water hardness affects the toxicity of Zn and Cu to a fish, Gambusia holbrooki Girard, 1859. The acute toxicity of selected heavy metals to G. holbrooki was determined in soft, hard and very hard water (25, 125 and 350 mg L?1 as CaCO3). Results showed that water hardness had a significant effect on Cu and on Zn toxicity on fish. Toxicity of Cu and of Zn increased with decreasing water hardness. The 96 hours LC50 values for G. holbrooki were higher in the hard and very hard water compared with soft water. Water hardness had a much smaller effect upon the acute toxicity of Cu than that of Zn. It was observed that the 96 hours LC50 for Cu at the soft, hard and very hard water was found to be 0.017, 0.17 and 0.65 mg L?1, respectively, while the 96 hours LC50 for Zn at the soft, hard and very hard water was found to be 0.46, 48.1 and 121.6 mg L?1, respectively.  相似文献
