The relationship between progesterone (P4) synthesis by luteal tissue and prostaglandin F (PGF) synthesis by endometrium and luteal tissue from two stages of the cycle, Days 7 to 8 and 15 to 16, was determined. Luteal and endometrial tissues were collected from pigs in three experimental groups at two stages of the cycle: (A) 6 pigs on Days 7 to 8 with spontaneous, 5 to 6 day old corpora lutea (CL); (B) 5 pigs on Days 15 to 16 with spontaneous, 13 to 14 day old CL; and (C) 6 pigs on Days 15 to 16 with spontaneous, 13 to 14 day old CL and 5 to 6 day old CL induced by pregnant mares serum gonadotropin (PMSG) and human chorionic gonadotropin (HCG) injections. Pigs with spontaneous, 13 to 14 day old CL of the cycle and PMSG-HCG induced accessory, 5 to 6 day old CL were used so that P4 and PGF synthesis in tissue from old and new CL could be compared in the same pig on Day 15 to 16 of the cycle. Tissues (100 mg minces) were incubated in 5 ml of Krebs Ringer solution in an atmosphere of 95% 02:5% CO2 for 2 hours at 0° C, 37° C, or 37° C with 1.3 x 10−4M indomethacin (IND). An aliquot of the incubation medium and an aliquot of the supernatant after homogenization of the tissue in the remaining medium of each flask was quantified for P4 and PGF by radioimmunoassay. P4 and PGF release into the medium and total accumulation of P4 and PGF in the flasks indicated that synthesis had occured at 37° C. Compared to tissue from 13 to 14 day old CL, tissue from 5 to 6 day old CL synthesized more P4 per flask (53.9 25.0 ng/mg tissue, P<.001) and released more P4 into the medium (20.8 8.8 ng/mg, P<.001). P4 synthesis by luteal tissue from 5 to 6 day old and 13 to 14 day old CL from pigs in group C was similar to P4 synthesis by luteal tissue from pigs in group A and group B, respectively. Luteul PGF synthesis was not affected significantly by either the age of the CL or by PMSG-HCG treatment. For endometrial samples, the synthesis of PGF was not significantly different among pigs in groups A, B and C. If uterine PGF is involved in luteal regression in the pig, the sensitivity of the CL to PGF may be more important than an increase in PGF secretion during the late luteal phase of the estrous cycle.  相似文献

The possibility of PGF(2)alpha production and presence of prostaglandin F synthase (PGFS; PGD(2) 11-ketoreductase) was studied in control and oxytocin (OT)-stimulated myometrial slices isolated from cyclic (Days 14-16) and early pregnant (Days 14-16) sows. Oxytocin (10(-7) M) stimulated (p<0.01) PGF(2)alpha production in both cycling and early pregnant myometrial slices. Prostaglandin F(2)alpha release was higher (p<0.01) in control as well as OT-treated myometrium of early pregnant sows in comparison to cycling myometrium. Prostaglandin F synthase expression at protein level was evident in myometrial slices of cyclic as well as early pregnant sows. The signals of PGFS was stronger (p<0.05) in cycling myometrium exposed to OT compared to that of control. There were no significant differences (p>0.05) in PGFS protein expression between control and OT-stimulated myometrial tissue of early-pregnant sows. The results of this study indicate the local PGF(2)alpha synthesis and the presence of PGFS in porcine cycling and early pregnant myometrial tissue. In addition, OT increased PGD(2) 11-ketoreductase protein expression in myometrium harvested during the porcine estrous cycle. However, the OT-stimulated PGF(2)alpha myometrial secretion was observed in both, cycling and pregnant gilts.  相似文献

Luteul PGF synthesis was not affected significantly by either the age of the CL or by PMSG-HCG treatment. For endometrial samples, the synthesis of PGF was not significantly different among pigs in groups A, B and C. If uterine PGF is involved in luteal regression in the pig, the sensitivity of the CL to PGF may be more important than an increase in PGF secretion during the late luteal phase of the estrous cycle.  相似文献

The effects of Cloprostenol administration on porcine luteal lipid metabolism, progesterone production, and prostaglandin F production were examined in 32 pigs at day 12 of the estrous cycle. Pigs were killed between 0 and 18 hours after treatment. Recovered luteal tissue was incubated at 0 C and at 37 C in the absence and presence of dibutyryl cyclic AMP and indomethacin. Net release of progesterone from luteal tissue was depressed within 1 hour after Cloprostenol treatment whereas net release of prostaglandin F was accelerated 4 hours after Cloprostenol treatment. Inclusion of dibutyryl cyclic AMP in the incubation media did not alter progesterone production by did enhance prostaglandin F production at 0 and 1 hour after Cloprostenol treatment. Inclusion of indomethacin in the incubation media completely inhibited the Cloprostenol-induced acceleration of luteal PGF production. Cloprostenol treatment increased luteal triglycerides and decreased luteal free cholesterol and cholesterol esters within 1 hr after treatment. Arachidonic acid percentages in free fatty acids and triglycerides were also increased within 1 hr after treatment. When 37 C and 0 C incubations were compared, luteal accumulation of free fatty acids was maximum at 1 hr after Cloprostenol treatment. accumulation of triglycerides in luteal tissie was comparatively uniform at all times examined during the first 18 hr after Cloprostenol treatment. Comparison of 37 C and 0 C incubations further revealed that luteal triglycerides were active in accumulation of arachidonic acid. Inclusion of dibutyryl cyclic AMP and/or indomethacin in the incubation media did not alter luteal lipid contents or fatty acid compositions. Blood plasma progesterone was depressed at 4 hours after Cloprostenol whereas 13,14-dihydro-15-keto-prostaglandin F2a was elevated at 18 hours after treatment. Blood plasma free fatty acids increased 330 percetn at 4 hours and free fatty acid compositions also changed at this time. In both luteal tissue and blood plasma, changes in steroid and fatty acid metabolism occurred prior to changes in prostaglandin metabolism, suggesting that Cloprostenol induced functional luteal regression prior to altering prostaglandin metabolism.  相似文献

Five normal estrous cycling multiparous non-lactating Brahman cows were utilized to determine if pregnancy-specific protein B (PSPB) would alter prostaglandin F2 alpha (PGF) and prostaglandin E2 (PGE) synthesis/release by endometrial tissue. The uterine horn ipsilateral to the corpus luteum was excised on Day 16 of the estrous cycle. Endometrial tissue (200 mg wet wt) was cultured in Nutrient Mixture F-10 medium in a perifusion system. The tissue and medium were aerated with 95% O2: 5% CO2 and temperature was maintained at 39 degrees C. The medium flow rate was 100 microliters/min and fractions were collected at 20 min intervals. After a 120 min settling period, tissue culture continued with: 1) control (medium only); 2) 2 micrograms [Asu1,6]-oxytocin/ml medium for 1 h; 3) 4 or 8 micrograms PSPB/ml medium for 2 h; or 4) 4 or 8 micrograms PSPB/ml medium for 2 h plus 2 micrograms oxytocin/ml medium during the second h. Differences in PGF and PGE secretion rate were not found between 4 and 8 micrograms PSPB. Therefore, groups were combined and data were analyzed according to tissue not receiving PSPB (control); receiving PSPB and receiving PSPB plus oxytocin. A nonsignificant rise (p greater than 0.10) in PGF secretion was observed in response to PSPB and PSPB plus oxytocin above the control by the end of the perifusion period (263.7 +/- 41.7, 220.0 +/- 41.7 and 166.1 +/- 41.7 pg/(100 mg tissue/min), respectively). Treatment with PSPB alone elevated (p less than 0.05) PGE secretion rate above control by 100 and 160 min post-removal of PSPB treatment. Treatment with PSPB plus oxytocin elevated (p less than 0.05) PGE release above control by 20 min after starting oxytocin treatment and continued throughout the duration of the perifusion. Pregnancy-specific protein B plus oxytocin-induced PGE release was greater (p less than 0.05) than PSPB alone after initiating the oxytocin treatment until 20 min after removal of the treatments. However, no further differences between PSPB alone and PSPB plus oxytocin treatments were detected throughout the remainder of the perifusion period. It appears that PSPB tends to elevate PGF release and significantly elevates PGE release from Day 16 endometrial tissue.  相似文献

The binding of high specific activity 3H-prostaglandin F2 alpha to membranes derived from human prostate tissue is described. Binding is specific for PGF2 alpha and is readily displaced by cold PGF2 alpha. The influence of testosterone and lactogen on the binding of the prostaglandin is investigated. Testosterone appears to enhance binding while the binding observed in the presence of lactogen is difficult to interpret.  相似文献

Human endometrial cells were dispersed with collagenase and maintained in culture overnight. The synthesis of PGF by the dispersed cells incubated at 37 degrees C in serum-free medium was stimulated by estradiol (10(-7)M - 10(-5)M), histamine (5X10(-7)M - 5X10(-5)M), bradykinin (10(-6)M), phorbol myristate (PMA, 3X10(-8)M) and arachidonate (5X10(-6)M). Preincubation of the cells for 3 h with cortisol (5X10(-7)M - 5X10(-5)M), progesterone (10(-6)M) or mepacrine (10(-6)M - 2X10(-4)M) inhibited the response to histamine, bradykinin and PMA but not to arachidonate. Perfusion of the cultured cells in filtration chambers yielded similar results to those obtained in the incubation system but differences in the onset and duration of the responses to stimuli were found. In the perifusion system the responses to histamine and bradykinin were rapid and of short duration (peak response in less than 60 min) while the responses to PMA and arachidonate were of longer duration with a slower onset. We conclude that these observations using dispersed endometrial cells are consistent with previous work showing that histamine, bradykinin and PMA act by stimulating acylhydrolase activity, thereby liberating precursors such as arachidonic acid which are converted to prostaglandins by the cyclo-oxygenase complex.  相似文献

In order to promote better understanding of the physiological roles of prostaglandin F in the mouse testis, we investigated the protein expression and the cellular localization of the enzymes cyclooxygenase and prostaglandin F synthase that are essential for the production of prostaglandin F, and the binding site, which is the prostaglandin Freceptor (FP). Western blot exhibited the expression of FP protein in wild type mouse testis, and that of prostaglandin F synthase and cyclooxygenase-1 proteins in the both of wild type mouse and FP-deficient mouse testes. The expression of prostaglandin F synthase and cyclooxygenase-1 were detected intensely in Leydig cell-rich fraction, and that of FP was detected equally in Leydig cell-rich fraction and the other fraction. Immunohistochemistry for cyclooxygenase-1 and prostaglandin F synthase demonstrated their co-localization in mouse Leydig cells. Histochemistry for FP demonstrated the localization in Leydig cells and in spermatids of seminiferous tubules. Double histochemical staining confirmed the co-localization of cyclooxygenase-1, prostaglandin F synthase and FP in the Leydig cells. These findings indicate that prostaglandin F may have an effect on the functions of Leyding cells in an autocrine fashion. It implies that prostaglandin F synthase and FP are involved in the control of testosterone release from Leydig cells and in spermatogenesis via the local pathway and the hypothalamo-hypophysial-testis pathway, and affect the testicular function.  相似文献

Separation and identification of prostaglandins in canine bile was performed by extraction and thin layer chromatography. The system provided tentative identification of the prostaglandin F compounds as the major prostaglandin subgroup present in bile. The prostaglandin was subsequently purified on silicic acid columns and quantitated by radioimmunoassay with tritiated PGF and anti PGF antibody employing the double antibody technique. Basal levels in hepatic bile were found to be 1028 ± 98 pg/ml.  相似文献

PGF2 alpha was administered intrauterine in 115 patients during the 11th to 20th week of pregnancy for abortion induction. An intra-amniotic method was used in 61 cases, an extra-amniotic one in 54 cases. Average total dose administered was 35.1 (range 5 to 65 mg) in the amniotic group and 6358 mcg (range 1500 to 14000 mcg) in the extra-amniotic group. The intra-amniotic group had an abortion rate of 92% and a 74% rate of side effects, mainly gastrointestinal irritation. Corresponding figures for the extra-amniotic group were 72% and 54% respectively. In the extra-amniotic group, doses of 4750 mcg or more increased the abortion rate up to 80% and side effects up to 64%. There were no serious complications. The intra-amniotic approach of prostaglandin induction is suitable for second trimester therapeutic abortions. The extra-amniotic approach is useful in cases of fetus mortuus and hydatiform mole.  相似文献

Progesterone and estradiol interact to regulate secretion of prostaglandin (PG) F2 alpha from the ovine endometrium in response to oxytocin. Two experiments were conducted to determine if these effects were due to changes in activity of phospholipase C or in the second messenger responsive pathways that regulate production of PGF2 alpha. In both experiments, ovariectomized ewes were assigned to one of four treatment groups (control, estradiol, progesterone, progesterone and estradiol). Steroids were administered, in vivo, to mimic the changes that occur during the estrous cycle. On Day 16 of steroid treatment, endometrial tissue was collected and incubated, in vitro, to measure activity of phospholipase C and release of PGF2 alpha. Treatment with progesterone, in vivo, enhanced basal and oxytocin-induced activity of phospholipase C and release of PGF2 alpha, in vitro. Estradiol suppressed oxytocin-induced activity of phospholipase C, both in the presence and absence of progesterone. In contrast to its effects on phospholipase C, estradiol inhibited basal and oxytocin-induced release of PGF2 alpha when administered alone, but not when administered with progesterone. Steroids had similar effects on the release of PGF2 alpha induced by phorbol 12-myristate 13-acetate and A23187. It was concluded that progesterone and estradiol regulate endometrial release of PGF2 alpha by affecting both the activity of phospholipase C and its associated second messenger responsive pathways that may regulate production of PGF2 alpha.  相似文献

In humans eicosapentaenoic acid can be converted to 3-series prostaglandins (PGF3 alpha, PGI3, and PGE3). Whether 3-series prostaglandins can protect the gastric mucosa from injury as effectively as their 2-series analogs is unknown. Therefore, we compared the protective effects of PGF3 alpha and PGF2 alpha against gross and microscopic gastric mucosal injury in rats. Animals received a subcutaneous injection of either PGF3 alpha or PGF2 alpha in doses ranging from 0 (vehicle) to 16.8 mumol/kg and 30 min later they received intragastric administration of 1 ml of absolute ethanol. Whether mucosal injury was assessed 60 min or 5 min after ethanol, PGF3 alpha was significantly less protective against ethanol-induced damage than PGF2 alpha. These findings indicate that the presence of a third double bond in the prostaglandin F molecule between carbons 17 and 18 markedly reduces the protective effects of this prostaglandin on the gastric mucosa.  相似文献

Chain-shortening of prostaglandin F2 alpha by rat liver peroxisomes   总被引:5,自引:0,他引:5  
Liver peroxisomes were isolated from di(2-ethylhexyl)phthalate treated rats by isopycnic sucrose gradient centrifugation of a light mitochondrial fraction. Incubation of prostaglandin F2 alpha with purified peroxisomes resulted in conversion into a more polar product(s). In contrast, incubation with mitochondrial fractions and microsomal fractions under the same conditions did not result in any detectable conversion. The polar material obtained from a preparative incubation was purified by high performance liquid chromatography and characterized by radio-gas chromatography and gas chromatography-mass spectrometry. The structure of the polar compound was shown to be 5,7,11-trihydroxy-tetranorprost-9-enoic acid (tetranor-prostaglandin F1 alpha). Prostaglandin F2 alpha was thus chain-shortened by four carbon atoms.  相似文献

Two mature heifers were slaughtered on days 3, 6-7, 10-11, 16, 18-19 or on day 21 of the oesterus cycle. Endometrium was incubated in quadruplicates with medium-199 at 37 C and a water saturated gas phase of 95% O2 + 5% CO2. Half ml medium samples were taken after 6, 12 and 24 h of incubation for determination of PGF2 alpha and PGFM. PGF2 alpha was secreted by endometrium at each stage of the oestrous cycle. Maximal secretion was measured around oestrus (p less than 0.01) compared with days 6-16 of the cycle. Concentration of PGFM in medium had a similar trend. Highest ability of endometrium for PGF2 alpha metabolism (indicated by the ratio PGF2 alpha:PGFM) was on days 6-16 of the cycle. Data suggest that PGF2 alpha metabolism by the endometrium may depend on ovarian steroids and that this metabolism may also protect the corpus luteum from the luteolytic action of PGF2 alpha besides reduced production of this prostaglandin during the luteal phase.  相似文献
