Using morphometric methods, five pathologists analyzed the positive staining for carcinoembryonic antigen (CEA) in sections from 17 ovarian tumors, with the intraclass correlation coefficient (ICC) and the mean values of the coefficients of variation (CV) used to assess reproducibility and variation. First, field and point scores for epithelium and mucin were estimated using 25 randomly selected square fields in sections from each of the tumors. The ICC range in the whole sample field was 0.53 to 0.81 (slight to substantial reproducibility) while the mean values of CV were 0.50 to 0.75. Second, the results of using random and standardized individual fields for the measurements were studied in three tumors. In random fields, the ICC was 0.57 to 0.71 (slight to moderate reproducibility) and the mean values of CV were 0.53 to 0.65. The corresponding values in standardized fields were 0.71 to 0.73 (moderate reproducibility) and 0.41 to 0.57, respectively. The results show that the variation is smaller and the degree of reproducibility higher in standardized fields. Considerable variation remains, however, revealing human factors as an important source of variation in practical morphometry.  相似文献

The aim of this study was to examine the intraobserver reliability of a posturographic assessment in patients with low back pain. We investigated 24 symptomatic subjects with defined low back pain (mean: 57.9?years) and a pair-matched control group including 24 asymptomatic persons (mean: 58.1?years). Each participant underwent two measurements on a posturographic device (32?Hz sampling rate) based on the Interactive Balance System (time interval: 7?d). Test procedure consisted of tests on solid ground with eyes open (1) and eyes closed (2). Data analysis included parameters of motor output and a frequency band analysis. Reliability tests were realized using by intraclass correlations (ICC). Coefficients of ICC ranged from 0.36 (95% CI: 0.01–0.73) to 0.94 (95% CI: 0.86–0.97) in both test positions. For 69% (11/16) of the investigated parameters a high level (ICC?>?0.75) of intraobserver reliability was reached. Based on the results, the posturographic measurement system used in this study seems to be appropriate for use in longitudinal study designs in an orthopaedic setting.  相似文献

Determination of the thermal thresholds is used clinically for evaluation of peripheral nervous system function. The aim of this study was to evaluate reliability of the method of levels performed with a new, low cost device for determining cutaneous temperature sensitivity. Nineteen male subjects were included in the study. Thermal thresholds were tested on the right side at the volar surface of mid-forearm, lateral surface of mid-upper arm and front area of mid-thigh. Thermal testing was carried out by the method of levels with an initial temperature step of 2°C. Variability of thermal thresholds was expressed by means of the ratio between the second and the first testing, coefficient of variation (CV), coefficient of repeatability (CR), intraclass correlation coefficient (ICC), mean difference between sessions (S1-S2diff), standard error of measurement (SEM) and minimally detectable change (MDC). There were no statistically significant changes between sessions for warm or cold thresholds, or between warm and cold thresholds. Within-subject CVs were acceptable. The CR estimates for warm thresholds ranged from 0.74°C to 1.06°C and from 0.67°C to 1.07°C for cold thresholds. The ICC values for intra-rater reliability ranged from 0.41 to 0.72 for warm thresholds and from 0.67 to 0.84 for cold thresholds. S1-S2diff ranged from -0.15°C to 0.07°C for warm thresholds, and from -0.08°C to 0.07°C for cold thresholds. SEM ranged from 0.26°C to 0.38°C for warm thresholds, and from 0.23°C to 0.38°C for cold thresholds. Estimated MDC values were between 0.60°C and 0.88°C for warm thresholds, and 0.53°C and 0.88°C for cold thresholds. The method of levels for determining cutaneous temperature sensitivity has acceptable reliability.  相似文献

The purpose of this study was to determine the intratester reliability of surface electromyography (EMG) assessment of the gluteus medius muscle in healthy people and people with chronic nonspecific low back pain (CNLBP) during barefoot walking. Gluteus medius muscle activity was measured twice in 40 people without and 30 people with CNLBP approximately 7 days apart. Walking gluteus medius muscle activity was normalised to maximal voluntary isometric contractions during side-lying hip abduction with manual resistance. Good intratester reliability (ICC > 0.75) was found for mean, peak, and peak to peak amplitude for healthy people. Only mean amplitude demonstrated good intratester reliability in those with CNLBP. Peak amplitude and peak to peak amplitude of the gluteus medius muscle of those with CNLBP, and the time of peak amplitude in both groups, demonstrated moderate reliability (ICC ranged from 0.50 to 0.58). Moderate to large standard error of measurement and minimal detectable change values were reported for outcome measurements. These results suggest that potentially large levels of random error can occur between sessions. Future research can build on this study for those with pathology and attempt to establish change values for EMG that are clinically meaningful.  相似文献

Appropriate reliability is a necessary condition for the use of surface EMG for evaluation of hamstring muscle function in cases of knee joint pathologies or ligament injuries. The aim of the study was to investigate the test-retest reliability of power spectrum and amplitude of surface electromyographic (EMG) measurements of semitendinosus (ST) and biceps femoris (BF) during ramp isometric contractions. Eleven males performed maximum isometric contractions (MVC) of the knee flexors in two sessions, a week apart with simultaneous recording of surface EMG of the BF and ST. Intra class correlation (ICC) and standard error measurements (SEM) were applied to assess test-retest reliability of the averaged EMG (aEMG) and the median frequency (MF) over 10 levels of force, from 0% to 100% of the maximum. The ICC values ranged from 0.38 to 0.96 for the aEMG with SEM values reaching 11.37% of MVC. For the MF, the ICCs ranged from 0.44 to 0.98 (SEM range 4.49–18.19 Hz). In our set up, ramp contractions can be used to examine hamstring EMG patterns with acceptable reliability.  相似文献

The purpose of the present study was to determine the day-to-day reliability in stride characteristics in rats during treadmill walking obtained with two-dimensional (2D) motion capture. Kinematics were recorded from 26 adult rats during walking at 8 m/min, 12 m/min and 16 m/min on two separate days. Stride length, stride time, contact time, swing time and hip, knee and ankle joint range of motion were extracted from 15 strides. The relative reliability was assessed using intra-class correlation coefficients (ICC(1,1)) and (ICC(3,1)). The absolute reliability was determined using measurement error (ME). Across walking speeds, the relative reliability ranged from fair to good (ICCs between 0.4 and 0.75). The ME was below 91 mm for strides lengths, below 55 ms for the temporal stride variables and below 6.4° for the joint angle range of motion. In general, the results indicated an acceptable day-to-day reliability of the gait pattern parameters observed in rats during treadmill walking. The results of the present study may serve as a reference material that can help future intervention studies on rat gait characteristics both with respect to the selection of outcome measures and in the interpretation of the results.  相似文献

Use of a shank shell has been shown to estimate tibial transverse rotations better than skin-mounted markers. However, the day-to-day reliability of the transverse tibial rotations using an individually molded shank shell has not been previously investigated. This study examined the between-tests and trials reliability of an individually molded shank shell for measuring peak tibial internal and external rotations, time of peak values, and tibia range of motion during 5 walking trials. The trial-to-trial reliability of tibial transverse rotations was measured in 14 healthy individuals while the test-retest reliability was measured in 10 persons on two occasions. Trial-to-trial reliability for peak transverse rotations, time of peak values, and tibia range of motion ranged from ICC (3,1) 0.59-0.95. The PCA between trials showed that 88-99 % of values were within 3 degrees of agreement. Test-retest reliability for peak rotations, tibia range of motion, and time of peak values ranged from ICC (3,1) 0.70-0.89 with SEM 1.6-2.21 degrees , 0.021 %, and 0.034 %, respectively. The PCA between tests showed that 70-100 % of values were within 3 degrees of agreement. The use of an individually molded shell and the close attachment of the shank shell to the individual's shank resulted in reliable test-retest and trial-to-trial data.  相似文献

Purpose

To assess wavefront derived refraction and full eye biometry including ciliary muscle dimension and full eye axial geometry in pseudophakic eyes using spectral domain OCT equipped with a Shack-Hartmann wavefront sensor.

Methods

Twenty-eight adult subjects (32 pseudophakic eyes) having recently undergone cataract surgery were enrolled in this study. A custom system combining two optical coherence tomography systems with a Shack-Hartmann wavefront sensor was constructed to image and monitor changes in whole eye biometry, the ciliary muscle and ocular aberration in the pseudophakic eye. A Badal optical channel and a visual target aligning with the wavefront sensor were incorporated into the system for measuring the wavefront-derived refraction. The imaging acquisition was performed twice. The coefficients of repeatability (CoR) and intraclass correlation coefficient (ICC) were calculated.

Results

Images were acquired and processed successfully in all patients. No significant difference was detected between repeated measurements of ciliary muscle dimension, full-eye biometry or defocus aberration. The CoR of full-eye biometry ranged from 0.36% to 3.04% and the ICC ranged from 0.981 to 0.999. The CoR for ciliary muscle dimensions ranged from 12.2% to 41.6% and the ICC ranged from 0.767 to 0.919. The defocus aberrations of the two measurements were 0.443 ± 0.534 D and 0.447 ± 0.586 D and the ICC was 0.951.

Conclusions

The combined system is capable of measuring full eye biometry and refraction with good repeatability. The system is suitable for future investigation of pseudoaccommodation in the pseudophakic eye.  相似文献

The purpose of the present study was to evaluate the intra and interday reliability of surface electromyographic amplitude values of the scapular girdle muscles and upper limbs during 3 isometric closed kinetic chain exercises, involving upper limbs with the fixed distal segment extremity on stable base of support and on a Swiss ball (relatively unstable). Twenty healthy adults performed the exercises push-up, bench-press and wall-press with different effort levels (80% and 100% maximal load). Subjects performed three maximal voluntary contractions (MVC) in muscular testing position of each muscle to obtain a reference value for root mean square (RMS) normalization. Individuals were instructed to randomly perform three isometric contraction series, in which each exercise lasted 6 s with a 2-min resting-period between series and exercises. Intra and interday reliabilities were calculated through the intraclass correlation coefficient (ICC 2.1), standard error of the measurement (SEM). Results indicated an excellent intraday reliability of electromyographic amplitude values (ICC ? 0.75). The interday reliability of normalized RMS values ranged between good and excellent (ICC 0.52–0.98). Finally, it is suggested that the reliability of normalized electromyographic amplitude values of the analyzed muscles present better values during exercises on a stable surface. However, load levels used during the exercises do not seem to have any influence on variability levels, possibly because the loads were quite similar.  相似文献

The aim of this systematic review was to explore the test-retest reliability of isometric mid-thigh pull maximum strength assessment. We searched through five databases to find studies that examined the test-retest reliability of peak force in the isometric mid-thigh pull exercise. From each included study, we extracted intra-class correlation coefficients (ICC) and/or coefficient of variation (CV). The methodological quality of the included studies was evaluated using the COSMIN checklist. A total of 16 good-to-excellent quality studies were included in the review. When considering results from all included studies, ICCs ranged from 0.73 to 0.99 (median ICC = 0.96), where 78% of ICCs were ≥ 0.90, and 98% of ICCs were ≥ 0.75. The range of reported CVs was from 0.7% to 11.1% (median CV = 4.9%), where 58% of CVs were ≤ 5%. Reliability was also good-to-excellent for both relative and absolute peak force and for both bilateral and unilateral isometric mid-thigh pull tests. The majority of studies did not find significant differences between testing sessions. It can be concluded that the isometric mid-thigh pull maximum strength assessment has good-to-excellent test-retest reliability. The isometric mid-thigh pull maximum strength assessment can be used as a reliable test in sports practice and for research purposes.  相似文献

The purpose of this study was to investigate the consistency of commonly reported rapid force characteristics utilizing both automated and manual muscle contraction onset detection methods. Twenty-four healthy volunteers performed isometric strength testing of the plantar flexor muscle group on two nonconsecutive days. Test–retest reliability was evaluated using intraclass correlation coefficients (ICCs), standard errors of measurement (SEM), and the SEM as a percentage of the mean (SEM%) for rate of force development (RFD), relative RFD, contractile impulse, and absolute force–time values at various epoch durations using automated and manual onset detection methods. For all rapid force variables, ICC and SEM% values ranged from 0.52 to 0.96 and 7.56% to 37.56%, respectively. For the majority of these variables (20 of 23), the automated onset detection method resulted in higher ICC and lower SEM% values compared to the manual onset detection method. Regardless of onset detection methodology, the consistency of relative RFD values declined following 50% of MVC. Collectively, these findings indicated that commonly evaluated rapid muscle force variables demonstrated acceptable relative and absolute consistency values. However, these values were generally superior for the automated onset detection methodology. Additionally, the consistency of relative RFD values declines following 50% MVC and therefore should be evaluated with caution.  相似文献

BACKGROUND: It is important to evaluate the reliability of common used methods of examining muscle fatigue from the lower back since the methods are used in patient evaluation. METHODS: To establish between-days reliability, ten subjects without lower-back pain performed a Sorensen test, a prone test for back extensor muscles against gravity, on three separate days. EMG was recorded from the L1 and L5 of the back extensor muscles. Fatigue was subjectively rated using a Borg CR-10 scale. Intraclass correlation coefficient, standard error of measurement and coefficient of variation were calculated from a one-way ANOVA. Percent agreement was also calculated. RESULTS: The study revealed good reliability for the slope for the total time (ICC 0.65-0.90), the initial and end median frequency (ICC 0.75-0.89), median frequencies at Borg ratings of three (ICC 0.63-0.88), five (ICC 0.62-0.84) and seven (ICC 0.67-0. 87), endurance time (ICC 0.89). The Borg ratings of the first minute agreed better than those of the second and the third. The Borg ratings at the second and the third test agreed to 40-80%, indicating a need for a practice session. CONCLUSION: The protocol used for assessing fatigue in the back extensor muscles proved to be reliable and is recommended for further use.  相似文献

Background

GBLUP (genomic best linear unbiased prediction) uses high-density single nucleotide polymorphism (SNP) markers to construct genomic identity-by-state (IBS) relationship matrices. However, identity-by-descent (IBD) relationships can be accurately calculated for extremely sparse markers. Here, we compare the accuracy of prediction of genome-wide breeding values (GW-BV) for a sib-evaluated trait in a typical aquaculture population, assuming either IBS or IBD genomic relationship matrices, and by varying marker density and size of the training dataset.

Methods

A simulation study was performed, assuming a population with strong family structure over three subsequent generations. Traditional and genomic BLUP were used to estimate breeding values, the latter using either IBS or IBD genomic relationship matrices, with marker densities ranging from 10 to ~1200 SNPs/Morgan (M). Heritability ranged from 0.1 to 0.8, and phenotypes were recorded on 25 to 45 sibs per full-sib family (50 full-sib families). Models were compared based on their predictive ability (accuracy) with respect to true breeding values of unphenotyped (albeit genotyped) sibs in the last generation.

Results

As expected, genomic prediction had greater accuracy compared to pedigree-based prediction. At the highest marker density, genomic prediction based on IBS information (IBS-GS) was slightly superior to that based on IBD information (IBD-GS), while at lower densities (≤100 SNPs/M), IBD-GS was more accurate. At the lowest densities (10 to 20 SNPs/M), IBS-GS was even outperformed by the pedigree-based model. Accuracy of IBD-GS was stable across marker densities performing well even down to 10 SNPs/M (2.5 to 6.1% reduction in accuracy compared to ~1200 SNPs/M). Loss of accuracy due to reduction in the size of training datasets was moderate and similar for both genomic prediction models. The relative superiority of (high-density) IBS-GS over IBD-GS was more pronounced for traits with a low heritability.

Conclusions

Using dense markers, GBLUP based on either IBD or IBS relationship matrices proved to perform better than a pedigree-based model. However, accuracy of IBS-GS declined rapidly with decreasing marker densities, and was even outperformed by a traditional pedigree-based model at the lowest densities. In contrast, the accuracy of IBD-GS was very stable across marker densities.  相似文献

Bicycle seat pressure is often examined by researchers exploring solutions for reducing seat injuries as it is thought a critical determinant; however, a reliable and valid methodology for this undertaking has not been reported. The current study was designed to address this shortcoming and to establish baseline interface pressure measurements for females and males. Participants completed two separate identical bicycle ergometer trials at 118 W in the tops and drops and at 300+/-82.4 W in top handlebar positions. Seat pressures were quantified from a pressure-sensing mat and the validity of the pressure system was examined through the relationship between known seat weights and the sum of seat pressures. Within trial intraclass correlation coefficients (ICCs) ranged between 0.90 and 0.99 and the between trial ICC values ranged between 0.02 and 0.96. The relationship (Pearson correlation coefficient) between seat weight and the sum of pressures was 0.97. Significant peak pressures were 39% greater in the 118 W top than 300 W top handlebar condition and 29% greater than the 118 W drop handlebar condition (p < 0.05). The gender comparison revealed that significant male peak pressure values were greater (24%) and that female values were less influenced by the hand position factor. Select within trial pressure measurements were found to be reliable during pedaling and valid statically. Additionally, the results indicated that workrate and hand position were factors influencing seat pressure and that males and females responded differently to adjustments of these factors.  相似文献

The Michaelis-Menten kinetics of blood-brain barrier transport of fourteen amino acids was investigated with a tissue-sampling, single-injection technique in the anesthetized rat. Tracer quantities of 14C-labelled amino acids and 3H2O, used as a freely diffusible internal reference, were mixed in 0.2 ml of buffered Ringer's solution and injected rapidly into a common carotid artery. Circulation was terminated by decapitation at 15s following injection. A brain uptake index (Ib) was determined from the ratio of 14C dpm in the brain tissue and the injection mixture divided by the same ratio for the 3H2O reference. Brain clearance of tracer concentration of amino acid was saturable when various concentrations of unlabeled amino acid were added to the injection solution. Double reciprocal plots of the saturation data yielded Km (mM) values that ranged from a low of 0.09 mM for arginine to a high of 0.75 mM for cycloleucine. Transport V values were determined from the relationship P = V/Km where P is the blood-brain barrier permeability constant (ml/g per min): P was calculated from the Ib for each amino acid based on a cerebral blood flow of 0.56 ml/g per min and a fractional extraction of 0.75 for the 3H2O reference 15s following carotid injection. The V values ranged from a low of 6.2 nmol/g per min for lysine to a high of 64 nmol/g per min for l-DOPA. Efflux of the tracer amino acid during the 15-s period after injection was assumed to be slow, since the rate constant of cycloleucine from brain to blood was low, 0.11 min-1.  相似文献

The aim of the study was to determine prevalence of the signs and symptoms related to esophageal dysfunction in irritable bowel syndrome (IBS) patients, and to investigate sensorimotor function impairment based on the esophageal manometry study, thus to determine the correlation between them. The study included 30 patients with IBS, 14 of them with diarrhea (IBSd) and 16 with constipation (IBSc) as a predominant discomfort. Control group consisted of 30 healthy subjects. The patients were included in the study on the basis of the Rome criteria for IBS. In addition to thorough history and physical examination patient underwent esophagogastroduodenoscopy and esophageal manometry. The values of esophageal manometry obtained in healthy subjects served as controls in manometry studies. The patients with IBS suffered a great number of both colonic and extracolonic signs and symptoms, however, there was no statistically significant difference in the prevalence of particular symptoms between the two patient subgroups. In comparison with healthy subjects, the patients suffering from IBS showed pathologically altered values in the majority of parameters of esophageal motility. Comparison of the two subgroups of IBS patients according to esophageal motility characteristics yielded differences in only few of them. The results obtained in the study could explain why the patients with IBS quite commonly complain of the symptoms related to upper gastrointestinal tract, such as heartburn and chest pain of noncardiac genesis. The results also suggest that the IBS might be associated with considerably more extensive smooth muscle or innervation changes than presumed before.  相似文献

Limited data exist on the performance of high-throughput proteomics profiling in epidemiological settings, including the impact of specimen collection and within-person variability over time. Thus, the Olink (972 proteins) and SOMAscan7Kv4.1 (7322 proteoforms of 6596 proteins) assays were utilized to measure protein concentrations in archived plasma samples from the Nurses’ Health Studies and Health Professionals Follow-Up Study. Spearman's correlation coefficients (r) and intraclass correlation coefficients (ICCs) were used to assess agreement between (1) 42 triplicate samples processed immediately, 24-h or 48-h after blood collection from 14 participants; and (2) 80 plasma samples from 40 participants collected 1-year apart. When comparing samples processed immediately, 24-h, and 48-h later, 55% of assays had an ICC/r ≥ 0.75 and 87% had an ICC/r ≥ 0.40 in Olink compared to 44% with an ICC/r ≥ 0.75 and 72% with an ICC/r ≥ 0.40 in SOMAscan7K. For both platforms, >90% of the assays were stable (ICC/r ≥ 0.40) in samples collected 1-year apart. Among 817 proteins measured with both platforms, Spearman's correlations were high (r > 0.75) for 14.7% and poor (r < 0.40) for 44.8% of proteins. High-throughput proteomics profiling demonstrated reproducibility in archived plasma samples and stability after delayed processing in epidemiological studies, yet correlations between proteins measured with the Olink and SOMAscan7K platforms were highly variable.  相似文献

Tricyclic antidepressants have been widely used for the treatment of depression and as a therapeutic agent for the altered gastrointestinal (GI) motility of irritable bowel syndrome (IBS). The aim of this study was to clarify whether antidepressants directly modulate pacemaker currents in cultured interstitial cells of Cajal (ICC). We used the whole-cell patch-clamp techniques at 30 degrees C in cultured ICC from the mouse small intestine. Treatment of pinacidil, an ATP-sensitive K(+) channel opener, in the ICC using the current clamping mode, produced hyperpolarization of the membrane potential and decreased the amplitude of the pacemaker potentials. With the voltage clamp mode, we observed a decrease in the frequency and amplitude of pacemaker currents and increases in the resting outward currents. These effects of pinacidil on pacemaker potentials and currents were completely suppressed by glibenclamide, an ATP-sensitive K(+) channel blocker. Also, with the current clamp mode, imipramine blocked the affect of pinacidil on the pacemaker potentials. Observations of the voltage clamp mode with imipramine, desipramine and amitryptyline suppressed the action of pinacidil in the ICC. Next, we examined whether protein kinase C (PKC) and the G protein are involved in the action of imipramine on pinacidil induced pacemaker current inhibition. We used chelerythrine, a potent PKC inhibitor and GDPbetaS, a nonhydrolyzable guanosine 5-diphosphate (GDP) analogue that permanently inactivates GTP-binding proteins. We found that pretreatment with chelerythrine and intracellular application of GDPbetaS had no influence on the blocking action of imipramine on inhibited pacemaker currents by pinacidil. We conclude that imipramine inhibited the activated ATP-sensitive K(+) channels in ICC. This action does not appear to be mediated through the G protein and protein kinase C. Furthermore, this study may suggest another possible mechanism for tricyclic antidepressants related modulation of GI motility.  相似文献

The 5-repetition sit-to-stand test (FRSTST) is a widely used measure of functional strength, particularly among older adults. The purpose of this review was to summarize the findings of research using the intraclass correlation coefficient (ICC) to describe the test-retest reliability of the FRSTST. A search of 3 electronic databases and hand searches were used to identify relevant articles. Information on the subjects, test sessions and the ICCs reported was abstracted from the articles. The searches identified 10 relevant articles. The ICCs reported in the articles ranged from 0.64 to 0.96. The adjusted mean ICC calculated from the reported ICCs was 0.81. The test-retest reliability of the FRSTST can be interpreted as good to high in most populations and settings.  相似文献

Irritable bowel syndrome (IBS) is a functional disorder of the gastrointestinal system characterized by abdominal pain related to bowel emptying, defecation impairment and abdominal distention. The aim of the study was to objectify lower gastrointestinal system disturbances in IBS patients. Thirty IBS patients and 30 healthy subjects were included in the study. IBS patients were divided into two subgroups: IBS with predominant diarrhea (IBSd) and IBS with predominant constipation (IBSc). All study subjects underwent physical examination (including digitorectal examination), standard laboratory testing and anorectal manometry. Endoscopy was performed only in group of IBS patients. A statistically significant difference was recorded in most manometric parameters between healthy subjects and IBS patients, which was even more pronounced in IBSd patients. Study results showed that the intestinal motility disorder underlying IBS could be objectified by use of anorectal manometry.  相似文献
