报告了钙流通抑制剂钌红对缺氧条件下拟南芥中ADH基因表达的诱导和植株存活的影响。结果表明 ,缺氧早期ADH基因的激活和表达需要钙离子 ,钌红处理可以延长缺氧条件下拟南芥植株的存活期。据此推测 :拟南芥中缺氧诱导的细胞死亡是一个钙离子介导的主动过程 ,钌红通过阻止细胞内钙离子浓度的增加而抑制这一过程。延长缺氧处理的时间会导致拟南芥叶片细胞内发生核凝聚和染色体断裂的现象 ,也进一步验证了这种构想。表明缺氧处理引起的叶片细胞损伤直至植株死亡是一个程序化的过程  相似文献

The Arabidopsis thaliana CONSTANS (CO) gene which promotes flowering in long days was recently isolated by chromosome walking. The mapping of QTLs controlling flowering time in Brassica species has identified genomic regions that contain homologues of the CO gene. Four genes homologous to the Arabidopsis CO gene were isolated from a pair of homoeologous loci in each of two doubled-haploid Brassica napus lines displaying different flowering times, N-o-1 and N-o-9. The four genes, BnCOa1, BnCOa9, BnCOb1 and BnCOb9, are located on linkage groups N10 and N19, and are highly similar to each other and to the Arabidopsis CO gene. Two regions of the proteins are particularly well conserved, a N-terminal region with two putative zinc fingers and a C-terminal region which may contain a nuclear localization signal. All four genes appear to be expressed in B. napus. The BnCOa1 allele was shown to complement the co-2 mutation in Arabidopsis in a dosage-dependent manner causing earlier flowering than in wild type under both long- and short-day conditions.  相似文献

We characterized the response of root hair density to phosphorus (P) availability in Arabidopsis thaliana. Arabidopsis plants were grown aseptically in growth media with varied phosphorus concentrations, ranging from 1 mmol m3 to 2000 mmol m3 phosphorus. Root hair density (number of root hairs per mm of root length) was analysed starting at 7 d of growth. Root hair density was highly regulated by phosphorus availability, increasing significantly in roots exposed to low-phosphorus availability. The initial root hairs produced by the radicle were not sensitive to phosphorus availability, but began to respond after 9 d of growth. Root hair density was about five times greater in low phosphorus (1 mmol m3) than in high phosphorus (1000 mmol m3) media. Root hair density decreased logarithmically in response to increasing phosphorus concentrations within that range. Root hair density also increased in response to deficiencies of several other nutrients, but not as strongly as to low phosphorus. Indoleacetic acid (IAA), the auxin transport inhibitor 2-(p-chlorophenoxy)-2-methylpropionic acid (CMPA), the ethylene precursor 1-aminocyclopropane-1-carboxylic acid (ACC), and the ethylene synthesis inhibitor amino-oxyacetic acid (AOA) all increased root hair density under high phosphorus but had very little effect under low phosphorus. Low phosphorus significantly changed root anatomy, causing a 9% increase in root diameter, a 31% decrease in the cross-sectional area of individual trichoblasts, a 40% decrease in the cross-sectional area of individual atrichoblasts, and 45% more cortical cells in cross-section. The larger number of cortical cells and smaller epidermal cell size in low phosphorus roots increased the number of trichoblast files from eight to 12. Two-thirds of increased root hair density in low phosphorus roots was caused by increased likelihood of trichoblasts to form hairs, and 33% of the increase was accounted for by changes in low phosphorus root anatomy resulting in an increased number of trichoblast files. These results show that phosphorus availability can fundamentally alter root anatomy, leading to changes in root hair density, which are presumably important for phosphorus acquisition.  相似文献

We analyzed the complete genome sequence of Arabidopsis thaliana and sequence data from 83 genes in the outcrossing A. lyrata, to better understand the role of gene expression on the strength of natural selection on synonymous and replacement sites in Arabidopsis. From data on tRNA gene abundance, we find a good concordance between codon preferences and the relative abundance of isoaccepting tRNAs in the complete A. thaliana genome, consistent with models of translational selection. Both EST-based and new quantitative measures of gene expression (MPSS) suggest that codon preferences derived from information on tRNA abundance are more strongly associated with gene expression than those obtained from multivariate analysis, which provides further support for the hypothesis that codon bias in Arabidopsis is under selection mediated by tRNA abundance. Consistent with previous results, analysis of protein evolution reveals a significant correlation between gene expression level and amino acid substitution rate. Analysis by MPSS estimates of gene expression suggests that this effect is primarily the result of a correlation between the number of tissues in which a gene is expressed and the rate of amino acid substitution, which indicates that the degree of tissue specialization may be an important determinant of the rate of protein evolution in Arabidopsis.  相似文献

Responses to oligogalacturonic acid (OGA) were determined in transgenic Arabidopsis thaliana seedlings express-ing the calcium reporter protein aequorin. OGA stimulated a rapid, substantial and transient increase in the concentration of cytosolic calcium ([Ca^2 ]cyt) that peaked after ca. 15 s. This increase was dose-dependent, saturating at ca. 50 μg Gal equiv/ml of OGA. OGA also stimulated a rapid generation of H202. A small, rapid increase in H2O2 content was followed by a much larger oxidative burst, with H2O2 content peaking after ca. 60 min and declining thereafter. Induction of the oxidative burst by OGA was also dose-dependent, with a maximum response again being achieved at ca. 50 μg Gal equiv/mL. Inhibitors of calcium fluxes inhibited both increases in [Ca^2 ]cyt and [H2O2], whereas inhibitors of NADPH oxidase blocked only the oxidative burst. OGA increased strongly the expression of the defence-related genes CHS,GST, PAL and PR-1. This induction was suppressed by inhibitors of calcium flux or NADPH oxidase, indicating that increases in both cytosolic calcium and H2O2 are required for OGA-induced gene expression.  相似文献

EMS30是拟南芥经甲基磺酸乙酯(EMS)诱变得到的白化突变体。该突变体的叶绿体结构存在严重缺陷,同时伴随叶绿素缺失。遗传分析显示EMS30突变体的突变表型受隐性单基因控制。采用图位克隆的方法对EMS30突变基因进行定位的结果显示,该基因位于拟南芥第一条染色体的分子标记F21M12和F14N23之间的96kb区间内,该区间包含25个基因。通过生物信息学分析发现,该区间内有3个基因定位在叶绿体或与叶绿体发育相关。这些结果有助于该基因的克隆,为阐释叶绿体发育提供线索。  相似文献

利用Peri,C 语言编写了鉴定和分析简单重复序列的一系列程序,在全基因组水平上分析了拟南芥(Arabidopsisthaliana L.)简单重复序列的分布及简单重复序列和基因的关系.共发现5 652个简单重复序列(≥20bp),大约每20.6kb有1个简单重复序列.拟南芥各染色体之间简单重复序列的密度基本一致.拟南芥的27 480条编码序列中,只有677条编码序列含有725个简单重复序列,其中的3碱基简单重复序列多数对应的是小的亲水性的氨基酸.在拟南芥和水稻(Oryza sativa L.)第4号染色体的高度保守的基因中,简单重复序列却并不保守.通过比较拟南芥和水稻之间简单重复序列的差异,推论出:水稻的全基因组和基因中简单重复序列的密度都比拟南芥大,这可能是水稻基因组序列比拟南芥大的原因之一,水稻基因组中0.21%来自简单重复序列,而拟南芥中只有0.13%;不但不同物种的基因组对简单重复序列的偏好性不同,而且不同物种的基因对简单重复序列的偏好性也不同.在水稻和拟南芥中部发现了一些嵌套性的卫星序列.  相似文献

DEGP家族蛋白酶广泛分布于原核生物和真核生物细胞中。在拟南芥中有16个DEGP类似的蛋白酶,根据蛋白质组学数据,其中有4个定位于叶绿体中,分别命名为DEG1、DEG2、DEG5和DEG8。结合生物化学和分子生物学等研究手段对拟南芥叶绿体中的DEGP蛋白酶进行了分析,现有的研究初步证明了这些蛋白酶参与光系统II(PSII)复合物反应中心D1蛋白的降解,从而在PSII复合物的修复循环和功能维护中起重要作用。该文概述了拟南芥叶绿体中DEG蛋白酶的结构和功能的最新研究进展。  相似文献

Arabidopsis seed coat development using light and transmission electron microscopy revealed major morphological changes associated with the transition of the integuments into the mature seed coat. By the use of a metachromatic staining procedure, cytological events such as the production of phenolic compounds and acidic polysaccharides were followed. Immediately after fertilization, the cells of the inner epidermis of the inner integument became vacuolated and subsequently accumulated pigment within them. This pigment started to disappear from the cytoplasm at the torpedo stage of the embryo, as it became green. During the torpedo stage, mucilage began to accumulate in the cells of the external epidermis of the outer integument. Furthermore, starch grains accumulated against the central part of the inner periclinal wall of these cells, resulting in the formation of small pyramidal domes that persisted until seed maturity. At the maturation stage, when the embryo became dormant and colourless, a new pigment accumulation was observed in an amorphous layer derived from remnants of crushed integument layers. This second pigment layer was responsible for the brown seed colour. These results show that seed coat formation may proceed in a coordinated way with the developmental phases of embryogenesis. Blue light regulates many physiological and developmental processes in higher plants through the action of multiple photosensory systems. The analysis of photomorphogenic mutants is leading to a better understanding of how the different photosensory systems mediate the wide range of responses observed in blue light. A review of the current literature on photomorphogenic mutants makes it apparent that redundancies exist in photoreceptor function. For example, many blue-light responses that have been shown to be regulated by a blue-light photosensory system are also under phytochrome control. The study of various light-response mutants suggests that a complex sensory network regulates light-mediated responses. This article attempts to piece together information regarding the sensory systems responsible for blue-light-regulated responses.  相似文献

Plant growth-promoting rhizobacteria (PGPR) colonize plant roots and exert beneficial effects on plant health and development. We are investigating the mechanisms by which PGPR elicit plant growth promotion from the viewpoint of signal transduction pathways within plants. We report here our first study to determine if well-characterized PGPR strains, which previously demonstrated growth promotion of various other plants, also enhance plant growth in Arabidopsis thaliana. Eight different PGPR strains, including Bacillus subtilis GB03, B. amyloliquefaciens IN937a, B. pumilus SE-34, B. pumilus T4, B. pasteurii C9, Paenibacillus polymyxa E681, Pseudomonas fluorescens 89B-61, and Serratia marcescens 90-166, were evaluated for elicitation of growth promotion of wild-type and mutant Arabidopsis in vitro and in vivo. In vitro testing on MS medium indicated that all eight PGPR strains increased foliar fresh weight of Arabidopsis at distances of 2, 4, and 6 cm from the site of bacterial inoculation. Among the eight strains, IN937a and GB03 inhibited growth of Arabidopsis plants when the bacteria were inoculated 2 cm from the plants, while they significantly increased plant growth when inoculated 6 cm from the plants, suggesting that a bacterial metabolite that diffused into the agar accounted for growth promotion with this strain. In vivo, eight PGPR strains promoted foliar fresh weight under greenhouse conditions 4 weeks after sowing. To define signal transduction pathways associated with growth promotion elicited by PGPR, various plant-hormone mutants of Arabidopsis were evaluated in vitro and in vivo. Elicitation of growth promotion by PGPR strains in vitro involved signaling of brassinosteroid, IAA, salicylic acid, and gibberellins. In vivo testing indicated that ethylene signaling was involved in growth promotion. Results suggest that elicitation of growth promotion by PGPR in Arabidopsis is associated with several different signal transduction pathways and that such signaling may be different for plants grown in vitro vs. in vivo.  相似文献

ABA不同浓度和不同时间处理拟南芥野生型7天龄幼苗,采用实时荧光定量PCR方法,分析小G蛋白ROP10和ROP乌苷酸交换因子RopGEFs基因的表达水平差异。结果表明,RopGEF7-10,12及33没有检测到表达,而RopGEFI-6,11,14ROP10在ABA处理下表现出不同的应答趋势,其中RopGEF5基因的表达变化与ROP10的变化相似。推测RopGEF5有可能在ROP10介导的ABA信号转导途经中起调控作用。  相似文献

用生物信息学方法对拟南芥叶状子叶(LEC)基因的核苷酸序列以及推导氨基酸序列的组成、功能结构域、亲水性等进行了分析。结果表明,拟南芥的LEC蛋白为位于细胞核中的亲水性不稳定蛋白;通过对LEC蛋白的功能结构域的分析发现,LEC1和L1L蛋白中高度保守的区域即为CBF-NFYB-HMF结构域,LEC2和FUSCA3蛋白中高度保守的区域为B3结构域。  相似文献

Weakly bound cell wall proteins of Arabidopsis thaliana were identified using a proteomic and bioinformatic approach. An efficient protocol of extraction based on vacuum-infiltration of the tissues was developed. Several salts and a chelating agent were compared for their ability to extract cell wall proteins without releasing cytoplasmic contaminants. Of the 93 proteins that were identified, a large proportion (60%) was released by calcium chloride. From bioinformatics analysis, it may be predicted that most of them (87 out of 93) had a signal peptide, whereas only six originated from the cytoplasm. Among the putative apoplastic proteins, a high proportion (67 out of 87) had a basic pI. Numerous glycoside hydrolases and proteins with interacting domains were identified, in agreement with the expected role of the extracellular matrix in polysaccharide metabolism and recognition phenomena. Ten proteinases were also found as well as six proteins with unknown functions. Comparison of the cell wall proteome of rosettes with the previously published cell wall proteome of cell suspension cultures showed a high level of cell specificity, especially for the different members of several large multigenic families.  相似文献
