Larvae of the nudibranch Phestilla sibogae were used to study whether a natural dissolved settlement cue (from their prey, Porites compressa, an abundant coral on Hawaiian reefs) induces behavioral responses that can affect larval transport to suitable settlement sites. As cue and larvae are mixed in the turbulent flow over a reef, cue is distributed in fine-scale filaments that the larva experiences as rapid (seconds) on/off encounters. To examine larval responses in this setting, individual larvae were tethered in a small flume with flow simulating water velocity relative to a freely swimming larva, and their responses to realistic temporal patterns of cue encounter were videotaped. Competent larvae quickly ceased swimming in cue filaments and resumed swimming after exiting filaments. The threshold cue concentration eliciting a response was 3%-17% of concentrations within heads of P. compressa in nature. When moving freely in filtered seawater, competent larvae swam along straight paths in all directions at approximately 0.2 cm s(-1), whereas in water conditioned by P. compressa, most ceased swimming and sank at approximately 0.1 cm s(-1). The ability of larvae to rapidly respond (by sinking) to brief encounters with dissolved settlement cues can enhance their rapid transport to the substratum, even in wave-driven turbulent flow.  相似文献

Unlike other venomous predators, the parasitoid wasp Ampulex compressa incapacitates its prey, the cockroach Periplaneta americana, to provide a fresh food supply for its offspring. We first established that the wasp larval development, from egg laying to pupation, lasts about 8 days during which the cockroach must remain alive but immobile. To this end, the wasp injects a cocktail of neurotoxins to manipulate the behavior of the cockroach. The cocktail is injected directly into the head ganglia using biosensors located on the stinger. The head sting induces first 30 min of intense grooming followed by hypokinesia during which the cockroach is unable to generate an escape response. In addition, stung cockroaches survive longer, lose less water, and consume less oxygen. Dopamine contained in the venom appears to be responsible for inducing grooming behavior. For the hypokinesia, our hypothesis is that the injected venom affects neurons located in the head ganglia, which send descending tonic input to bioaminergic neurons. These, in turn, control the thoracic premotor circuitry for locomotion. We show that the activity of identified octopaminergic neurons from the thoracic ganglia is altered in stung animals. The alteration in the octopaminergic neurons' activity could be one of the mechanisms by which the venom modulates the escape circuit in the cockroach's central nervous system and metabolism in the peripheral system.  相似文献

Larvae of the nudibranch Phestilla sibogae are induced to metamorphose by a water-borne chemical cue released by the adult nudibranch's prey, the coral Porites compressa. In competent larvae, the apical sensory organ (ASO) includes five serotonergic parampullary neurons; five ampullary neurons, the ampullae of which are filled with sensory cilia; and a basal neuropil. After sensing the coral cue, the ASO undergoes radical morphological changes: a deterioration of sensory elements in the ASO and serotonergic axons originating from them to innervate the velum. Three hours after metamorphic induction, the velar lobes are lost, the serotonergic axons begin to break apart, the five parampullary neurons begin to degenerate, and the five ampullary neurons retract away from the epidermal surface. The extent of deterioration evident by this time suggests that the parampullary and ampullary components of the ASO are no longer functional. By 10 h after metamorphic induction, labeling of the ciliary bundles in the ampullary neurons has disappeared, and it is likely that these cells have degenerated. The results presented here provide evidence that the sensory neurons of the ASO and probably the entire organ are solely larval structures that do not persist into the adult sensory-nervous system in P. sibogae.  相似文献

Coral tumors are atypical skeletal forms found on coral reefs worldwide. Here we present an analysis of the microbial communities associated with skeletal tumors on the coral Porites compressa. Microbial growth rates on both healthy and tumorous P. compressa were decoupled from the surrounding water column. Microbial communities associated with tumorous colonies had a significantly faster growth rate than those associated with healthy P. compressa. The microbial community associated with the tumors contained more culturable Vibrio spp. and could utilize more carbon sources than the microbes associated with healthy colonies. Presence of tumors affected the composition and dynamics of the microbial population associated with the entire colony.  相似文献

The olfactory nervous systems of insects and mammals exhibit many similarities, suggesting that the mechanisms for olfactory learning may be shared. Neural correlates of olfactory memory are distributed among many neurons within the olfactory nervous system. Perceptual olfactory learning may be mediated by alterations in the odorant receptive fields of second and/or third order olfactory neurons, and by increases in the coherency of activity among ensembles of second order neurons. Operant olfactory conditioning is associated with an increase in the coherent population activity of these neurons. Olfactory classical conditioning increases the odor responsiveness and synaptic activity of second and perhaps third order neurons. Operant and classical conditioning both produce an increased responsiveness to conditioned odors in neurons of the basolateral amygdala. Molecular genetic studies of olfactory learning in Drosophila have revealed numerous molecules that function within the third order olfactory neurons for normal olfactory learning.  相似文献

Assimilation of inorganic nitrogen from nutrient-poor tropical seas is an essential challenge for the endosymbiosis between reef-building corals and dinoflagellates. Despite the clear evidence that reef-building corals can use ammonium as inorganic nitrogen source, the dynamics and precise roles of host and symbionts in this fundamental process remain unclear. Here, we combine high spatial resolution ion microprobe imaging (NanoSIMS) and pulse-chase isotopic labeling in order to track the dynamics of ammonium incorporation within the intact symbiosis between the reef-building coral Acropora aspera and its dinoflagellate symbionts. We demonstrate that both dinoflagellate and animal cells have the capacity to rapidly fix nitrogen from seawater enriched in ammonium (in less than one hour). Further, by establishing the relative strengths of the capability to assimilate nitrogen for each cell compartment, we infer that dinoflagellate symbionts can fix 14 to 23 times more nitrogen than their coral host cells in response to a sudden pulse of ammonium-enriched seawater. Given the importance of nitrogen in cell maintenance, growth and functioning, the capability to fix ammonium from seawater into the symbiotic system may be a key component of coral nutrition. Interestingly, this metabolic response appears to be triggered rapidly by episodic nitrogen availability. The methods and results presented in this study open up for the exploration of dynamics and spatial patterns associated with metabolic activities and nutritional interactions in a multitude of organisms that live in symbiotic relationships.  相似文献

Culture medium conditioned by C-6 glioma cells (previously shown to support the survival of chick sensory neurons in vitro [7]) was tested on the PC12 clone of pheochromocytoma cells. It is demonstrated that fibre outgrowth can be induced and the activity of the enzyme choline acetyltransferase stimulated by this medium. The activity of the conditioned medium is protease sensitive and the peak activity has an apparent molecular weight (on gel filtration) of approx. 50000. Similar effects can be obtained by conditioned medium from another rat glioma, D-74, and by extracts from rat central and peripheral nervous system. In no case is the effect blocked by specific antibodies against nerve growth factor (NGF). Media conditioned by other cell lines, together with other tissue extracts (at the same concentration as those taken from nervous tissue) have no detectable effect on the PC12 cells. It is suggested that PC12 cells will be useful as a tool for the further characterisation of factors with NGF-like activities.  相似文献

We have studied the effects of dopamine on the gill withdrawal reflex evoked by tactile siphon stimulation in the margine mollusc Aplysia. Physiological concentrations of dopamine (diluted in seawater) were perfused through the gill during siphon stimulation series. The amplitude of the reflex was potentiated by dopamine and habituation of the reflex was prevented. This occurred with no change in the activity evoked in central motor neurons. These results lead us to conclude that the dopaminergic motor neuron L9 is modulating habituation in the periphery and that the central nervous system facilitatory control of the peripheral nervous system may act via a dopaminergic pathway.  相似文献

To explore how microbial community composition and function varies within a coral reef ecosystem, we performed metagenomic sequencing of seawater from four niches across Heron Island Reef, within the Great Barrier Reef. Metagenomes were sequenced from seawater samples associated with (1) the surface of the coral species Acropora palifera, (2) the surface of the coral species Acropora aspera, (3) the sandy substrate within the reef lagoon and (4) open water, outside of the reef crest. Microbial composition and metabolic function differed substantially between the four niches. The taxonomic profile showed a clear shift from an oligotroph-dominated community (e.g. SAR11, Prochlorococcus, Synechococcus) in the open water and sandy substrate niches, to a community characterised by an increased frequency of copiotrophic bacteria (e.g. Vibrio, Pseudoalteromonas, Alteromonas) in the coral seawater niches. The metabolic potential of the four microbial assemblages also displayed significant differences, with the open water and sandy substrate niches dominated by genes associated with core house-keeping processes such as amino acid, carbohydrate and protein metabolism as well as DNA and RNA synthesis and metabolism. In contrast, the coral surface seawater metagenomes had an enhanced frequency of genes associated with dynamic processes including motility and chemotaxis, regulation and cell signalling. These findings demonstrate that the composition and function of microbial communities are highly variable between niches within coral reef ecosystems and that coral reefs host heterogeneous microbial communities that are likely shaped by habitat structure, presence of animal hosts and local biogeochemical conditions.  相似文献

The parasitoid wasp Ampulex compressa induces behavioral changes in the cockroach prey by injecting venom into its central nervous system. In contrast to most other venomous predators, the wasp's sting does not induce paralysis. Rather, the two consecutive stings in the thoracic and head ganglia induce three stereotypic behavioral effects. The prey behavior is manipulated in a way beneficial to the wasp and its offspring by providing a living meal for its newborn larva. The first sting in the thorax causes a transient front leg paralysis lasting a few minutes. This paralysis prevents the cockroach from fighting with its front legs, thereby facilitating the second sting in the head. A postsynaptic block of central synaptic transmission mediates this leg paralysis. Following the head sting, dopamine identified in the venom induces 30 minutes of intense grooming that appears to prevent the cockroach from straying until the last and third behavioral effect of hypokinesia commences. In this lethargic state that lasts about three weeks, the cockroach does not respond to various stimuli nor does it initiates movement. However, other specific behaviors of the prey are unaffected. We propose that the venom represses the activity of head ganglia neurons thereby removing the descending excitatory drive to specific thoracic neurons.  相似文献

The Caribbean sponge Mycale laevis is often found growing in close proximity to living scleractinian corals. This commonly observed sponge–coral association has been considered a mutualism, with the coral providing substratum for the sponge, and the sponge protecting the coral skeleton from boring organisms. We examined the specificity of sponge recruitment to live corals, expecting a positive and specific settlement response if a mutualism exists. Benthic surveys conducted off Key Largo, Florida, and Bocas del Toro, Panama, revealed that individuals of M. laevis grew on substrata that included dead coral and other species of sponges. Selectivity analysis indicated that at three of the four survey sites, M. laevis was not randomly distributed, but associated with live corals more frequently than expected from proportional coral cover. However, settlement assays demonstrated that larvae of M. laevis did not preferentially respond to the presence of live coral. We have previously demonstrated that adults of M. laevis are chemically undefended and readily eaten by spongivorous fishes unless protected by adjacent substrata such as live corals. In overfished areas, where spongivore density is low, the sponge is not selectively distributed near corals. Initial results of settlement experiments with different substrata suggested that larvae of M. laevis responded positively to the presence of the chemically defended sponge Amphimedon compressa, perhaps indicating an associational defense. Further experiments revealed that larvae were reacting to artificially high concentrations of exudates from cut surfaces of Am. compressa; settlement was not enhanced in response to healed pieces of Am. compressa. In addition, the larvae of M. laevis did not selectively respond to live coral or to chemically defended heterospecifics. These results indicate that the commonly observed proximity of M. laevis to live corals is not driven by larval settlement behavior, but instead by post‐settlement mortality due to predation.  相似文献

The objectives of this study were to investigate the presence and annual cycle of sex steroids in scleractinian coral, Euphyllia ancora. The free and conjugated forms of sex steroids in coral and spawning seawater were investigated, and aromatase activity in the coral tissue was identified. Polyps collected from corals and seawater were extracted with diethyl ether, and purified by alumina column and reversed-phase HPLC; testosterone and estradiol-17beta (E2) was measured by a validated RIA. E2 and testosterone in their free and glucuronide forms were consistently detected in coral tissue throughout the year. Peak concentrations of free E2, E2 glucuronide, and testosterone glucuronide were obtained in the coral tissue just prior to spawning. The presence of specific aromatase activity was demonstrated in coral tissue. Free E2 and E2 glucuronide concentrations were higher than androgen (testosterone and testosterone glucuronide) in coral tissue and spawning seawater. Higher concentrations of free E2 than E2 glucuronide were detected in coral tissues throughout the year. In contrast, higher concentrations of E2 glucuronide than free E2 and testosterone glucuronide were found in seawater during mass coral spawning. No steroid sulfate could be detected in the coral tissue and seawater. We suggest that the release of E2 glucuronide may play an important role in coral mass spawning.  相似文献

The microbial community associated with the reef building coral Pocillopora damicornis located on the Great Barrier Reef was investigated using culture-independent molecular microbial techniques. The microbial communities of three separate coral colonies were assessed using clone library construction alongside restriction fragment length polymorphism and phylogenetic analysis. Diversity was also investigated spatially across six replicate samples within each single coral colony using 16S rDNA and rpoB-DGGE analysis. Clone libraries demonstrated that the majority of retrieved sequences from coral tissue slurry libraries affiliated with gamma-Proteobacteria. This contrasted with clone libraries of seawater and coral mucus, which were dominated by alpha-Proteobacteria. A number of retrieved clone sequences were conserved between coral colonies; a result consistent with previous studies suggesting a specific microbe-coral association. rpoB-DGGE patterns of replicate tissue slurry samples underestimated microbial diversity, but demonstrated that fingerprints were identical within the same coral. These fingerprints were also conserved across coral colonies. The 16S rDNA-DGGE patterns of replicate tissue slurry samples were more complex, although non-metric multidimensional scaling (nMDS) analysis showed groupings of these banding patterns indicating that some bacterial diversity was uniform within a coral colony. Sequence data retrieved from DGGE analysis support clone library data in that the majority of affiliations were within the gamma-Proteobacteria. Many sequences retrieved also affiliated closely with sequences derived from previous studies of microbial diversity of healthy corals in the Caribbean. Clones showing high 16S rDNA sequence identity to both Vibrio shiloi and Vibrio coralliilyticus were retrieved, suggesting that these may be opportunist pathogens. Comparisons of retrieved microbial diversity between two different sampling methods, a syringe extracted coral mucus sample and an airbrushed coral tissue slurry sample were also investigated. Non-metric multidimensional scaling of clone library data highlighted that clone diversity retrieved from a coral mucus library more closely reflected the diversity of surrounding seawater than a corresponding coral tissue clone library.  相似文献

We report on a novel sponge disease, hereafter termed 'sponge white patch' (SWP), affecting the Caribbean sponge species Amphimedon compressa. SWP is characterized by distinctive white patches of variable size that are found irregularly on the branches of diseased sponges. Nearly 20% of the population of A. compressa at Dry Rocks Reef, Florida, USA, showed symptoms of SWP at the time of investigation (November 2007-July 2010). Approximately 21% of the biomass of SWP individuals was bleached, as determined by volume displacement. Scanning electron microscopy analysis showed severe degradation of bleached tissues. Transmission electron microscopy of the same tissues revealed the presence of a spongin-boring bacterial morphotype that had previously been implicated in sponge disease (Webster et al. 2002; Mar Ecol Prog Ser 232:305-309). This particular morphotype was identified in 8 of 9 diseased A. compressa individuals investigated in this study. A close relative of the aforementioned disease-causing alphaproteobacterium was also isolated from bleached tissues of A. compressa. However, whether the spongin-boring bacteria are true pathogens or merely opportunistic colonizers remains to be investigated. Molecular fingerprinting by denaturing gradient gel electrophoresis (DGGE) demonstrated a distinct shift from the microbiota of healthy A. compressa to a heterogeneous mixture of environmental bacteria, including several phylotypes previously implicated in sponge stress or coral disease. Nevertheless, tissue transplantation experiments conducted in the field failed to demonstrate infectivity from diseased to healthy sponges, leaving the cause of SWP in A. compressa to be identified.  相似文献

We previously reported that noradrenergic (NA) neurons in the nucleus of the solitary tract (NST) are necessary for exogenous CCK octapeptide to inhibit food intake in rats. To determine whether NST NA neurons also are necessary for lithium chloride (LiCl) to inhibit food intake and/or to support conditioned avoidance behavior, saporin toxin conjugated to an antibody against dopamine beta hydroxylase (DSAP) was microinjected bilaterally into the NST to ablate resident NA neurons. DSAP and sham control rats subsequently were tested for the ability of LiCl (0.15M, 2% body wt) to inhibit food intake and to support conditioned flavor avoidance (CFA). LiCl-induced hypophagia was significantly blunted in DSAP rats, and those with the most extensive loss of NST NA neurons demonstrated the most attenuated LiCl-induced hypophagia. Conversely, LiCl supported a robust CFA that was of similar magnitude in sham control and DSAP rats, including rats with the most extensive NA lesions. A terminal c-Fos study revealed intact LiCl-induced c-Fos expression in the lateral parabrachial nucleus and central amygdala in DSAP rats, despite significant loss of NST NA neurons and attenuated c-Fos activation of corticotropin-releasing hormone-positive neurons in the paraventricular nucleus of the hypothalamus (PVN). Thus, NST NA neurons contribute significantly to LiCl-induced hypophagia and recruitment of stress-responsive PVN neurons but appear to be unnecessary for CFA learning and expression. These findings support the view that distinct central nervous system circuits underlie LiCl-induced inhibition of food intake and conditioned avoidance behavior in rats.  相似文献

Influence of seawater Sr content on coral Sr/Ca and Sr thermometry   总被引:3,自引:0,他引:3  
The Ca content of a Porites coral from Xisha, South China Sea is quite uniform along its 18-year growth axis. A comparison with previously published data shows that the Ca content of corals from different sites varies by only 0.4%. This is much smaller than the variation of Ca in seawater (2.2%), indicating that Ca variations in seawater do not significantly affect the Ca compositions of coral skeletons. The variation in skeletal Ca contents results in only ±0.6°C of uncertainty in SST calculations, which is much smaller than the large disparities observed for previously established coral Sr/Ca thermometers. In contrast, Sr in tropical seawater varies spatially by as much as 2.4%, corresponding to ~4°C offset for coral Sr/Ca calibrations. The effect of seawater Sr variations on coral Sr/Ca thermometers is evaluated and we demonstrate that the content of seawater Sr is the major factor responsible for disparities in these coral Sr/Ca thermometers. The disparities can be significantly reduced when seawater Sr contents are included in the Sr/Ca thermometers.  相似文献

The response of the central nervous system to stress is often critical to the adaptation of an organism to its environment. However, in humans the response to stress also can be maladaptive, resulting in the expression or exacerbation of many neurological and psychiatric disorders. In this review, we examine the impact of stress on the synthesis and release of dopamine within mesocortical, mesoaccumbens, and nigrostriatal dopamine projections. We note that whereas stress increases the neurochemical activity of each of these populations of dopamine neurons, heterogeneities do exist. Specifically, acute stress evokes a greater increase in dopamine metabolism and release within the prefrontal cortex than the subcortical sites. Furthermore, whereas prior exposure to chronic stress enhances the response of mesocortical dopamine neurons to an acute novel stressor, this does not occur in the subcortical sites. In addition to these regional heterogeneities, we also note that even within a single dopamine projection there can be heterogeneous regulation of dopamine synthesis and release. Specifically, whereas stress-induced dopamine release in the neostriatum is mediated by an action of glutamate on the dopamine cell body, stress-induced dopamine synthesis in the neostriatum is mediated by an action of glutamate on the dopamine nerve terminal. Finally, we propose that regional heterogeneities in the responsiveness of central dopamine neurons to stress may ultimately play a role in the expression and exacerbation of symptoms associated with schizophrenia.  相似文献

Axon regeneration in the adult central nervous system (CNS) is limited by several factors including a lack of neurotrophic support. Recent studies have shown that glia from the adult rat CNS, specifically retinal astrocytes and Müller glia, can promote regeneration of retinal ganglion cell axons. In the present study we investigated whether retinal glia also exert a growth promoting effect outside the visual system. We found that retinal glial conditioned medium significantly enhanced neurite growth and branching of adult rat dorsal root ganglion neurons (DRG) in culture. Furthermore, transplantation of retinal glia significantly enhanced regeneration of DRG axons past the dorsal root entry zone after root crush in adult rats. To identify the factors that mediate the growth promoting effects of retinal glia, mass spectrometric analysis of retinal glial conditioned medium was performed. Apolipoprotein E and secreted protein acidic and rich in cysteine (SPARC) were found to be present in high abundance, a finding further confirmed by western blotting. Inhibition of Apolipoprotein E and SPARC significantly reduced the neuritogenic effects of retinal glial conditioned medium on DRG in culture, suggesting that Apolipoprotein E and SPARC are the major mediators of this regenerative response.  相似文献

The olfactory placode gives rise to both olfactory receptor neurons, which remain as a component of the peripheral nervous system, and to luteinizing hormone-releasing hormone (LHRH) neurons, which migrate to the central nervous system. In this study, we used chick olfactory placode explants to ask several questions regarding LHRH neuronal differentiation. We found that explants of ectoderm from the fronto-nasal region of embryos as early as Hamilton & Hamburger (HH) stage 12 gave rise to LHRH neurons, that explants from all regions of the olfactory placode were able to generate LHRH neurons, that both brain conditioned medium and disruption of the olfactory placode increase the number of LHRH neurons observed in explants, and that the combination of these two manipulations results in the production of more LHRH neurons than either treatment alone. We conclude that LHRH neurons originate in the olfactory epithelium and that some of the same factors which influence olfactory receptor neuron development also affect LHRH neuronal development.  相似文献
