探究西藏不同地区昆诺阿藜种子内生真菌的物种组成、多样性等群落结构特征,丰富昆诺阿藜种子相关微生物的可利用资源,对于微生物资源利用和昆诺阿藜病害的生物防控具有重要意义。本研究收集西藏不同海拔3个地区(日喀则、拉萨和林芝)的昆诺阿藜种子,利用传统分离方式进行了真菌的培养。从46份昆诺阿藜种子中共分离出947株真菌,经ITS序列分析结合形态观察鉴定为1门9目12科26属77种。所有的真菌都归类为子囊菌门Ascomycota,菌株数量相对丰度最高的3个属为链格孢属Alternaria (40.2%)、镰刀菌属Fusarium (17.4%)和茎点霉属Phoma (13.9%)。按照多样性指数,日喀则地区的昆诺阿藜内生可培养真菌的多样性明显高于拉萨和林芝两个地区。  相似文献

  • Fungi have essential functions in plant health and performance. However, the plant-associated functions of many cultured fungi have not been established in detail.
  • Here, the fungal species diversity in Salvia miltiorrhiza roots and rhizosphere was assessed for the first time using culturomics and high-throughput sequencing. We present a comprehensive functional metagenomic analysis of these fungi and verified activity of cellulase and chitinase predicted in the metagenomic analysis.
  • We first collected and cultured fungi from the root and rhizosphere of S. miltiorrhiza. We found 92 species across 37 families and five phyla, with Ascomycota being dominant. Many rDNA internal transcribed spacer sequences could not be assigned to lower taxonomic levels. There were 19 genera of endophytic fungi and 37 genera of rhizosphere fungi. The culturomics approach had lower taxonomic diversity than high-throughput sequencing, but some fungi were only found in cultures. Structural analyses indicated that the dominant species differed in cultured and non-cultured samples at other levels, apart from the phylum level. Functional analysis mapped 223 carbohydrate enzyme families and 393 pathways in the CAZy and KEGG databases, respectively. The most abundant families were glycoside hydrolases and those involved in carbohydrate metabolism. As predicted by metagenomics, we experimentally verified cellulase and chitinase activity for 29 and 74 fungi, respectively.
  • We provide the first evidence of biomass recycling by fungi that are associated with plants. Culturing is essential to reveal the hidden microbial community and critical functions in plant–microbe interactions.

Samples of microorganisms from the surface of constructions of Mir Space Station (Mir SS) were taken and examined after 13 years of operation. The following microorganisms were isolated and identified: 12 fungal species belonging to the genera Penicillium, Aspergillus, Cladosporium, and Aureobasidium; 3 yeast species belonging to the genera Debaryomyces, Candida, and Rhodotorula; and 4 bacterial species belonging to the genera Bacillus, Myxococcus, and Rhodococcus. The predominant species in all samples was Penicillium chrisogenum. It was shown that the fungi isolated could damage polymers and induce corrosion of aluminum-magnesium alloys. We commenced a study of microbial degraders on constructions of the Russian section of the International Space Station (RS ISS). Twenty-six species of fungi, bacteria, yeasts, and actinomycetes, known as active biodegraders, were identified in three sample sets taken at intervals. We founded a collection of microorganisms surviving throughout space flights. This collection can be used to test spacecraft production materials, in order to determine their resistance to biodegradation.  相似文献

The phylum Neocallimastigomycota contains eight genera (about 20 species) of strictly anaerobic fungi. The evolutionary relationships of these genera are uncertain due to insufficient sequence data to infer their phylogenies. Based on morphology and molecular phylogeny, thirteen isolates obtained from yak faeces and rumen digesta in China were assigned to Neocallimastix frontalis (nine isolates), Orpinomyces joyonii (two isolates) and Caecomyces sp. (two isolates), respectively. The phylogenetic relationships of the eight genera were evaluated using complete ITS and partial LSU sequences, compared to the ITS1 region which has been widely used in this phylum in the past. Five monophyletic lineages corresponding to six of the eight genera were statistically supported. Isolates of Caecomyces and Cyllamyces were present in a single lineage and could not be separated properly. Members of Neocallimastigomycota with uniflagellate zoospores represented by Piromyces were polyphyletic. The Piromyces-like genus Oontomyces was consistently closely related to the traditional Anaeromyces, and separated the latter genus into two clades. The phylogenetic position of the Piromyces-like genus Buwchfawromyces remained unresolved. Orpinomyces and Neocallimastix, sharing polyflagellate zoospores, were supported as sister genera in the LSU phylogeny. Apparently ITS, specifically ITS1 alone, is not a good marker to resolve the generic affinities of the studied fungi. The LSU sequences are easier to align and appear to work well to resolve generic relationships. This study provides a comparative phylogenetic revision of Neocallimastigomycota isolates known from culture and sequence data.  相似文献

The coastal marine ecosystems of Vietnam are one of the global biodiversity hotspots, but the biodiversity of marine fungi is not well known. To fill this major gap of knowledge, we assessed the genetic diversity (ITS sequence) of 75 fungal strains isolated from 11 surface coastal marine and deeper waters in Nha Trang Bay and Van Phong Bay using a culture-dependent approach and 5 OTUs (Operational Taxonomic Units) of fungi in three representative sampling sites using next-generation sequencing. The results from both approaches shared similar fungal taxonomy to the most abundant phylum (Ascomycota), genera (Candida and Aspergillus) and species (Candida blankii) but were different at less common taxa. Culturable fungal strains in this study belong to 3 phyla, 5 subdivisions, 7 classes, 12 orders, 17 families, 22 genera and at least 40 species, of which 29 species have been identified and several species are likely novel. Among identified species, 12 and 28 are new records in global and Vietnamese marine areas, respectively. The analysis of enzyme activity and the checklist of trophic mode and guild assignment provided valuable additional biological information and suggested the ecological function of planktonic fungi in the marine food web. This is the largest dataset of marine fungal biodiversity on morphology, phylogeny and enzyme activity in the tropical coastal ecosystems of Vietnam and Southeast Asia. Biogeographic aspects, ecological factors and human impact may structure mycoplankton communities in such aquatic habitats.  相似文献

Molecular studies on the evolution and systematics of fungi have been established primarily based on the neutral theory by analyzing neutral mutations in some defined segments of housekeeping genes as genetic markers. Such an approach is, however, hardly applicable for analyzing ancient evolutionary radiations. In the present study, we looked for DNA sequences characterizing higher taxa, and discovered a unique macroevolutionary genomic marker, megB1, that specifies the phylum Basidiomycota. megB1 is an approximately 500-bp DNA element, which is defined by terminal sequences and five internal segments conserved throughout the phylum. megB1 resides on the rDNA intergenic spacer 1 (IGS1) from 27 species of 10 Basidiomycota genera examined. While megB1 was not found in IGS1 from the other 92 species of the 27 Basidiomycota genera, several genera representing them carry megB1 in some other genomic regions. No known taxonomic criteria fit into the classification on the basis of whether megB1 resides on rDNA. Neighbor-joining analysis of the megB1 sequence, however, properly assigned species to their respective genera. Thus far, megB1 has not been found in any genomic or genetic databases currently available for other phyla. These results suggest that megB1 may have emerged upon the occurrence of Basidiomycota, and that this phylum evolved thereafter leaving this element conserved throughout their further differentiation. megB1 may be a novel genomic marker useful in the analysis of ancient through the latest evolutionary radiation in Basidiomycota.  相似文献

The potential of fungal pretreatment to improve fermentable sugar yields from wheat straw or Miscanthus was investigated. We assessed 63 fungal strains including 53 white-rot and 10 brown-rot fungi belonging to the Basidiomycota phylum in an original 12 day small-scale solid-state fermentation (SSF) experiment using 24-well plates. This method offers the convenience of one-pot processing of samples from SSF to enzymatic hydrolysis. The comparison of the lignocellulolytic activity profiles of white-rot fungi and brown-rot fungi showed different behaviours. The hierarchical clustering according to glucose and reducing sugars released from each biomass after 72 h enzymatic hydrolysis splits the set of fungal strains into three groups: efficient, no-effect and detrimental-effect species. The efficient group contained 17 species belonging to seven white-rot genera and one brown-rot genus. The yield of sugar released increased significantly (max. 62%) compared with non-inoculated controls for both substrates.  相似文献

Vegetatively colonized agar cores of 69 basidiomycete fungus isolates (48 species in 30 genera and 17 families) were stored at 5 degrees C in tubes of sterile distilled water without manipulation for 20 years. These were represented by 34 isolates of saprotrophic fungi (29 species in 19 genera) and 35 isolates of mycorrhizal fungi (19 species in 11 genera). Viability was evaluated based on revived growth on agar media at room temperature. Fifty-seven of the 69 isolates (82.6%) grew vigorously when revived after storage for 20 years; of the 34 saprotrophic fungus isolates, 30 revived (88.2%); of the 35 mycorrhizal fungus isolates, 27 revived (77.1%). Thirteen isolates of Laccaria were all viable after 20 years, indicating cold storage in sterile water to be a good method for maintaining this important genus of mycorrhizal fungi. In general, however, mycorrhizal fungus species demonstrated lower viability than saprotrophic fungi.  相似文献

They consisted of the gregarines (subclass Gregarinasida) (1624 named species and 231 named genera), the hemogregarines (family Haemogregarinidae) (399 species and 4 genera), the eimeriorins (order Eimeriorida) (1771 species and 43 genera), the hemospororids (order Haemospororida) (444 species and 9 genera), the piroplasmids (order Piroplasmorida) (173 species and 20 genera), and a few others (105 species and 32 genera). The first apicomplexan protozoon was seen by Antony van Leeuwenhoek; in 1674 he saw oocysts of Eimeria stiedai in the gall bladder of a rabbit. The first member of the phylum to be named (by Dufour in 1828) was Gregarina ovata in earwigs. During the quarter century 1826–1850, 41 species and 6 genera of Apicomplexa were named. These numbers increased progressively. In the quarter century 1951–1975, 1873 new species and 83 new genera were named. Data are given for the numbers of named species and genera of apicomplexan protozoa of each group known in 1850, 1875, 1900, 1925, 1950, 1975, and 1987.  相似文献

以1、2、3年生的楸树实生苗和嫁接苗(梓树砧木)根系为研究对象,通过对ITS rDNA区域标记扩增子的Illumina MiSeq测序,分析不同苗龄楸树实生苗和嫁接苗根相关真菌的结构组成和多样性。获得根相关真菌OTU共842个,分属4门、24纲、70目、134科、233属、347种;丛枝菌根真菌(arbuscular mycorrhizal fungi)AMF-OTU共42个,分属1门、1纲、3目、3科、3属、13种。根相关真菌和AMF的OTU数量、丰度和多样性在实生苗中均随苗龄的增加而降低,而在嫁接苗中则随着苗龄的增加而增加。门水平上,实生苗与嫁接苗根相关真菌的优势菌都是子囊菌门Ascomycota、担子菌门Basidiomycota和接合菌门Zygomycota,但它们的相对丰度有所差异;属水平上,实生苗和嫁接苗根相关真菌的优势菌种在组成和数量上都具有一定的差异性。楸树根相关真菌拥有3种营养模式和12个生态功能群,其中实生苗根系中病理营养型真菌的比例大于嫁接苗,腐生营养型则差异不大,而共生营养型则小于嫁接苗。生态功能群分析显示大多数楸树根系真菌表现出多种生存策略,部分真菌可以在植物-真菌-动物中跨界侵染。该研究可为楸树根相关真菌的利用提供一定的理论依据和基础。  相似文献

玉米秸秆还田对土壤丛枝菌根真菌群落的影响   总被引:4,自引:0,他引:4  
为揭示农业管理活动对土壤丛枝菌根(AM)真菌的影响机制,基于Illumina Miseq高通量测序平台以及脂肪酸指纹图谱方法,研究了连续4年玉米秸秆还田后,AM真菌群落组成、AM真菌生物量及其与土壤环境因子间的相互关系.结果表明:所获得的2430个AM真菌OTUs从门到种依次分类,共分为1门、3纲、4目、8科、10属、143种,但不同处理间AM真菌群落丰富度(Chao1指数和ACE指数)、多样性(Shannon、Simpson多样性指数)没有显著差异.AM真菌中类球囊霉属、球囊霉属为优势属.随秸秆还田量的增加,球囊霉属丰度降低;3000、9000 kg·hm^-2秸秆还田量下,类球囊霉属、无梗囊霉属的丰度与对照(0 kg·hm^-2)间差异达极显著水平;原囊霉属、类球囊霉属、球囊霉属在3000 kg·hm^-2秸秆还田量下与对照间差异显著,非度量多维尺度(NMDS)分析表明,9000、12000 kg·hm^-2的秸秆还田量下土壤AM真菌β多样性与对照间聚集度较其他处理相差较远,秸秆还田量对AM真菌β多样性的影响显著.多元分析结果能在累积变量82.8%上揭示土壤主要理化性状与AM真菌丰富度、多样性的空间变化关系.土壤全氮、碱解氮是影响以磷脂脂肪酸表征的土壤主要微生物类群生物量以及以中性脂肪酸表征的AM真菌生物量的主要因子.持续玉米秸秆还田改变了AM真菌属水平上的分类学组成;随秸秆还田量的增加,AM真菌特有的微生物种类减少,AM真菌群落组成间的相似度下降;秸秆还田增加了土壤AM真菌生物量及其占土壤微生物总生物量的比例.  相似文献

喀斯特是我国南方广泛分布的地貌类型,土壤真菌对喀斯特植被演替恢复具有重要调节功能,不同石漠化程度的喀斯特区植被演替受到土壤微生物影响,因此研究不同石漠化区域植被演替阶段的土壤真菌组成及多样性,探索土壤真菌在喀斯特植被演替过程中的作用机制具有重要意义。本文采用时空替代法采集了不同石漠化程度(潜在、中度和强度)的喀斯特区植被演替乔木、灌木和草本演替阶段土壤样品,通过Illumina HiSeq第二代高通量测序分析了土壤真菌组成及多样性。结果表明,试验共获得3 871个OTUs,分属4门17纲116科174属;潜在和中度石漠化区各演替阶段土壤真菌优势门均为担子菌门,强度石漠化区各演替阶段土壤真菌无相同优势门;土壤真菌组成及多样性在潜在石漠化区表现为乔木>草本>灌木,中度石漠化区为灌木>乔木>草本,强度石漠化区为灌木>草本>乔木,且石漠化程度对真菌组成及多样性的影响大于植被演替的影响;土壤理化性质随石漠化程度及演替阶段发生变化,且显著影响真菌多样性指数,以碱解氮为主导因子显著影响土壤真菌群落。  相似文献

Arbuscular mycorrhizal (AM) fungi in coalmine spoil, island forest and saline soils were enriched in pot culture with maize (Zea mays L.), tobacco (Nicotiana tabacum L.), white clover (Trifolium repens Linn.) and silverweed cinquefoil (Potentilla anserina L.). Based on spores, there were more species of AM fungi in the coalmine spoil (15 species, 3 genera), than in the forest soil (11 species, 4 genera) and the saline soil (5 species, 2 genera). In the trap cultures, the total of 28 species in Acaulospora, Gigaspora, Glomus, and Sclerocystis detected in the original soils were all recovered with at least one of the four trap plants. The highest spore and species numbers were recovered in trap cultures of T. repens inoculated with coalmine spoil. Glomus constrictum and Glomus multicaule were the dominant species associated with N. tabacum grown in saline soil and forest soil. The dominant species of AM fungi on the four hosts was Acaulospora mellea, which had over 90% of the spore incidence in pot trap culture in coalmine spoil. It is suggested that there be selectivity between host plants and AM fungi. The number of species of AM fungi detected was influenced by host plants under certain conditions and white clover was generally the optimal host plant to detect diversity of AM fungi.  相似文献

【背景】高通量测序分析作为深入了解环境微生物群落组成的重要方法,已成为植物内生真菌多样性研究的有效手段,然而由于引物的扩增差异,采用不同引物可对实验结果分析造成影响。同时,盐角草作为世界上最耐盐的植物之一,存在着多种功能性的内生真菌,而较为全面介绍其内生真菌组成和多样性的报道鲜见。【目的】为了揭示盐角草内生真菌的多样性,解析不同扩增引物对内生菌多样性分析的影响。【方法】分别采用真菌高通量测序常用引物对ITS1-5F、ITS1-1F、ITS2对采自乌鲁木齐达坂城盐湖的盐角草内生真菌进行扩增,开展其内生真菌OTU的分析。【结果】通过不同引物对扩增并测序共获得102个盐角草内生真菌OTU,涉及真菌界8个门和未分类菌群,其中子囊菌门(Ascomycota)占绝对优势,其次为担子菌门(Basidiomycota);在属层次上,盐角草内生真菌共涉及64个属及20个未分类属,其中Alternaria、Cladosporium、Podospora等3个属为盐角草内生真菌优势菌群。对不同引物对扩增测序结果分析表明,不同引物对扩增对分析内生真菌OTU数量和种类具有明显的影响,在全部所得的102个OTU中,ITS1-5F引物对获得44个OTU、ITS1-1F引物对获得55个OTU、ITS2引物对获得25个OTU,但以上3对引物扩增均检测到的OTU数仅为5个。物种组成和多样性分析表明,内生真菌多样性分析中采用以ITS1-1F为主,ITS1-5F为辅的分析策略,可较为全面地展现内生真菌的多样性。【结论】盐角草存在较为丰富的内生真菌资源,不同扩增引物对高通量分析盐角草内生真菌组成和分布具有明显的影响。  相似文献

【背景】对于饮用水中的微生物污染,绝大多数研究集中在细菌、病毒及原虫和蠕虫。由于真菌中包含多种潜在致病菌,应当引起人们的关注。【目的】研究城市供水系统中真菌数量、群落组成变化及可能存在的潜在致病真菌。【方法】利用MEA和RB两种培养基,对供水系统中的原水和两个水厂净水工艺过程及不同供水模式用户龙头水样进行培养法计数统计。提取上述样本总DNA,并应用Illumina MiSeq平台进行ITS1区高通量测序。【结果】从15个样本中共获得有效序列579 470条,1 260个OTU,包含8个门26个纲67个目228个属的真菌。门水平上子囊菌门真菌为供水系统优势真菌,属水平上不同样本存在差异,但曲霉属和枝顶孢属真菌在所有样本中均存在;培养法和高通量测序结果共同显示,活性炭过滤出水真菌数量和物种丰富度均较前一工艺有所上升;氯化消毒对真菌数量、物种多样性及物种组成影响显著;经过供水管网输配和二次供水设施后,用户龙头水样本真菌数量和物种丰富度明显高于出厂水。【结论】供水系统中的优势真菌为子囊菌门(Ascomycota),该门真菌可穿透净水工艺过程的多级屏障;水厂净水工艺能有效去除水中可培养真菌,生物活性炭过滤工艺能够影响真菌数目及物种多样性;供水管网及二次供水设施是末端饮用水中真菌污染的重要来源;供水系统中存在潜在致病真菌。  相似文献

Tardigrades constitute a phylum of miniaturized metazoans with ca. 1030 living species, a fossil record that probably dates back to the Cambrian, and physiological properties that allow them to live in almost any environment known to host life on Earth—they can also survive in space. Despite broad consensus regarding their membership of the superclade Ecdysozoa, the exact position of the phylum remains contested (some analyses suggest onychophorans and arthropods as their closest relatives, while others suggest a relationship to nematodes and nematomorphs) and the internal relationships of the phylum are still poorly understood. In the present study, we present a hypothesis of tardigrade relationships by examining more taxa and more markers than any previously published phylogeny of the group. We generated novel data for three markers (18S rRNA, 28S rRNA, COI) for 42 individuals of 16 carefully identified species, comprising 12 genera and five families from the classes Heterotardigrada and Eutardigrada, and analysed them in conjunction with nearly all data available from GenBank. Our results show certain disagreement with current taxonomy both at higher ranks (families, orders, classes) and at low (generic) taxonomic levels. When studying the sensitivity to outgroup choice, the class Eutardigrada was monophyletic under only one combination of outgroups; all other combinations placed the eutardigrade order Apochela as sister to the class Heterotardigrada. Phylogenetic relationships within the other eutardigrade order, Parachela, were stable to outgroup choice. Eutardigrade superfamilies recently proposed by Sands and collaborators in the order Parachela were tested with the introduction of new sequences from additional genera, and the possible morphological synapomorphies supporting those superfamilies are discussed. © The Will Henning Society 2011.  相似文献

本研究根据沙冬青现有的生长状况,将内蒙古西鄂尔多斯保护区珍稀濒危植物蒙古沙冬青群落分为轻度衰退、中度衰退和重度衰退3个衰退等级,并选取一个未衰退的沙冬青群落作为对照,应用Illumina Hiseq测序平台,对这4个沙冬青群落的植株根内真菌进行高通量测序分析。测序共获得215个真菌的OTU,不同衰退等级沙冬青根内真菌群落结构在门、科、属水平上都有显著差异。Top 10属中的AgaricusTomentellaTricholomaFusariumInocybe以及Tuber这6个属真菌在所有衰退等级的沙冬青都有分布。不同衰退等级的沙冬青根内都有腐生或寄生真菌和“共生”真菌分布,不同衰退等级两类群的真菌占比不同且呈现出动态变化,随着衰退等级的增加沙冬青根内腐生真菌或寄生真菌与“共生”真菌比例显著增高。土壤有机质与土壤容重具有协同作用,且对AgaricusInocybeFusariumPenicilliumAmphinema具有正相关影响,对TricholomaTomentellaTuber具有负相关影响。  相似文献
