In this study, hydrolysis and methanolysis of glycerol trioleate (TG) by lipase Candida sp. 99–125 were investigated under different water conditions. Both the reaction rates were relatively low without water, while increasing water content to 5 wt.% (or more, from 10–20%) based on the TG amount caused remarkable higher TG conversion for both reactions. Moreover, comparing the time course curves of the hydrolysis and methanolysis, it could be concluded that the methanolysis reaction catalyzed by this Candida sp. 99–125 appeared to accord with the successive reaction mechanism. TG was first hydrolyzed to partial glycerides and oleic acid (OA), then oleic acid methyl ester (OAME) was produced by esterification of the OA with methanol. This water effect was also confirmed by the experiments that water substitutions such as t-butanol and some surfactants added into the system did not get such high yields as that of the water included system. So these results showed that water took part in the methanolysis reaction, and successive hydrolysis–esterification process might be the catalytic mechanism of this lipase.  相似文献

对假丝酵母Candidasp.99—125发酵生产脂肪酶的过程中油脂代谢情况进行了研究。分析了发酵过程中甘油酯、油酸、棕榈酸等物质浓度随发酵时间的变化趋势,以及它们与菌体生长和产酶之间相互影响关系,结果发现油酸的消耗能够显著地促进脂肪酶的合成(油酸质量浓度从30g/L降低到10g/L),并且细胞对油酸和棕榈酸的利用没有选择性,最终发酵脂肪酶活力可达8000U/mL。  相似文献

A novel enzymatic method for preparing water-soluble derivatives of salicylic acid catalyzed by immobilized lipase was described. This study is the first to describe the enzymatic transesterification of methyl salicylate in organic solvents with different hydroxyl donors. The acyl-transfer between methyl salicylate and sorbitol was best supported by solvents of log P values –0.33 to 1.4. With Candida antarctica lipase in tert-amyl alcohol, a sorbitol conversion yield of 98% can be obtained by transesterification with sorbitol and methyl salicylate in one step.  相似文献

A novel immobilized lipase (from Candida rugosa) on hydrophobic and superparamagnetic microspheres was prepared and used as a biocatalyst to catalyze esterification reactions in diverse solvents and reaction systems. The results showed that the immobilized lipase had over 2-fold higher activities in higher log P value solvents. An exponential increase of lipase activity against log P of two miscible solvent mixtures was observed for the first time. Both free and immobilized lipase achieved its maximum activity at the range of water activity (a(w)) 0.5-0.8 or higher. At a(w) 0.6, the immobilized lipase exhibited markedly higher activities in heptane and a solvent-free system than did the native lipase. In multicompetitive reactions, the alcohol specificity of the lipase showed a strong chain-length dependency, and the immobilized enzyme exhibited more preference for a longer-chain alcohol, which is different from previous reports. The immobilized lipase showed higher specificities for butyric acid and the medium-chain-length fatty acids (C(8)-C(12)). Then, the immobilized lipase was extended to solvent-free synthesis of glycerides from glycerol and fatty acids. Recovered by magnetic separation, the immobilized lipase exhibited good reusability in repeated batch reaction, indicating its promising feature for biotechnology application.  相似文献

Thermolysin-catalyzed peptide synthesis using N-benzyloxycarbonyl)-l-phenylalanine (Z-Phe) and l-phenylalanine methyl ester (Phe-OMe) as substrates was done mainly in a water-organic one phase solvent system. The organic solvent content used was less than the saturation concentration in buffer. With organic solvents with high log P values, the enzymatic activity increased as the organic solvent content increased; but further increases in the organic solvent content decreased the enzymatic activity, showing an “organic activity” profile. On the other hand, with organic solvents of low log P values, the enzymatic reaction was inhibited even by the initial addition of organic solvents. When a correlation between maximum activities and logP values or Hildebrand solubility parameters was investigated, a linear correlation was obtained among the same category of organic solvents, but not between categories. This suggests that the direct effect of organic solvents on the microenvironment of the enzyme largely depends on the molecular structure of the solvents.  相似文献

Lipase activity was measured in homogenates of rat cerebral hemispheres using radioactive glycerol trioleate emulsified with Triton X-100 as substrate. The labeled oleic acid was separated from the ester with a methanol-chloroform-heptane mixture. Under these assay conditions, the activity showed pH optima at about 5.5 and 7.5. The final products of these lipase activities were suggested to be free fatty acid and glycerol.  相似文献

Oleic acid esters were shown to be the best carbon source for both cell growth and lipase production by Candida rugosa. Use of a cosolvent, dodecane, in fermentations improved the solubility of solid substrates and increased oxygen solubility. This resulted in the highest lipase activity in batch fermentation with glycerol trioleate and dodecane. Lipase activity reached 77.1 units ml–1.  相似文献

We investigated the influence of solvent on the positional selectivity of Novozym 435 which was the immobilized Candida antarctica lipase B (CALB) during the esterification of oleic acid with glycerol for 1,3-diolein preparation previously. Herein, molecular modeling was used to elucidate the underlying mechanism of the solvent effect on the positional selectivity of the enzyme. The results showed that the binding energy of sn-1 hydroxyl of glycerol molecular with CALB became higher, and the binding energy of sn-2 hydroxyl of glycerol molecular with CALB became lower along with the increase of the solvent log P. It was demonstrated that, increasing log P of the solvent, the enzyme selectivity to sn-1 hydroxyl of glycerol molecular grew weaker, and the selectivity to sn-2 hydroxyl of glycerol molecular grew stronger.  相似文献

本研究开发了一种用石油醚提取毛油的工艺,研究了以提取的毛油和甲醇为原料,用固定化Candida sp.99-125脂肪酶催化合成脂肪酸甲酯(FAMEs)的可行性。同时考察了磷脂对固定化酶活性、反应起始速率、固定化酶使用批次的影响以及毛油和精炼油对固定化酶使用批次等的影响。研究结果表明,用磷脂质量分数为1%的石油醚悬液浸泡过的脂肪酶比仅用石油醚浸泡过的脂肪酶初始转酯化速率显著下降。当大豆油中无磷脂时,15min时FAMEs的产率为26.2%;磷脂质量分数为5%时,FAMEs降为12.4%。当大豆油中磷脂质量分数小于1%时,固定化酶使用10个批次,FAMEs产率无明显变化。固定化脂肪酶催化石油醚浸提得到的大豆和小桐子毛油,经过10个批次反应FAMEs产率都保持在70%以上,该固定化酶直接催化毛油生产生物柴油具有良好的工业化前景。  相似文献

In this study, we report the enzymatic production of glycerol acetate from glycerol and methyl acetate. Lipases are essential for the catalysis of this reaction. To find the optimum conditions for glycerol acetate production, sequential experiments were designed. Type of lipase, lipase concentration, molar ratio of reactants, reaction temperature and solvents were investigated for the optimum conversion of glycerol to glycerol acetate. As the result of lipase screening, Novozym 435 (Immobilized Candida antarctica lipase B) was turned out to be the optimal lipase for the reaction. Under the optimal conditions (2.5 g/L of Novozym 435, 1:40 molar ratio of glycerol to methyl acetate, 40 °C and tert-butanol as the solvent), glycerol acetate production was achieved in 95.00% conversion.  相似文献

The Candida rugosa lipase catalyzed Dynamic Kinetic Resolution of racemic ibuprofen methyl ester produced (S)-ibuprofen in over 90% yield within 72 h at pH 7.6. The best concentration of various buffers for these reactions ranged from 0.2 to 0.5 M. The commercial lipase was found to be acidic altering the final pH of the reaction mixtures. Dimethylformamide co-solvent maintained the reaction pH better than dimethylsulfoxide. Lower concentrations of ibuprofen methyl ester and higher stirring rates led to faster conversions. The minimal amount of lipase needed was 20 mg/mL buffer. Reaction of (R)-ibuprofen methyl ester under the optimized conditions excluding the lipase led to no racemization, indicating that the conversion of (R)-ibuprofen methyl ester to (S)-ibuprofen is catalyzed by the enzyme, thus, indicating Candida rugosa lipase possess Isomerase activity.  相似文献

Feruloylated mono- and di-acylglycerols were synthesized in a two step reaction: ethyl ferulate was first transesterified with glycerol and then this was esterified with oleic acid. The yield of the combined feruloylated mono- and di-acylglycerols in the second reaction reached 96% when esterification of 0.3 g transesterification products with 2.22 g oleic acid was catalyzed with 0.25 g Candida antarctica lipase at 60°C under a vaccum of 10 mmHg for 1.33 h.  相似文献

In this paper, immobilized lipase catalyzed biodiesel production from lard was studied. Using Candida sp. 99-125, the effect of temperature, water content, enzyme amount, solvent and three-step methanolysis were investigated. The optimal conditions for processing 1 g of lard were: 0.2 g immobilized lipase, 8 ml n-hexane as solvent, 20% water based on the fat weight, temperature 40 °C, and three-step addition of methanol. As a result, the fatty acid methyl esters (FAMEs) yield was 87.4%. The lipase was proved to be stable when used repeatedly for 180 h.  相似文献

以1株分解麻风树油的脂肪酶产生菌Pseudomonas sp. LP-1为出发菌株, 通过麻疯树油定向驯化筛选获得1株酶活较高且产酶稳定的菌株P. sp. X-2-45, 其水解酶活为29.79 U/mL, 比原始菌株提高了288%。对P. sp. X-2-45生长与产酶特征、对植物油脂水解能力及在有机相中催化脂肪酸和有机醇间的酯化反应研究发现, 该菌株生长速率和产酶速率明显加快, 培养30 h时生物量和酶活达到最大, 稳定期延长, 培养过程中脂肪酶在培养基中的稳定性提高。以麻疯树油诱导合成的P. sp. X-2-45脂肪酶对麻疯树油的水解能力比原始菌株提高了378%, 说明采用麻风树油定向驯化可提高脂肪酶对相应底物的水解能力。X-2-45脂肪酶可以催化月桂酸与正丁醇、正辛醇、月桂醇和丙三醇之间, 棕榈酸、硬脂酸与甲醇、正辛醇、月桂醇和丙三醇之间, 油酸与甲醇、正丁醇、正辛醇、月桂醇和丙三醇之间发生酯化反应。  相似文献

The hydrolysis of triglycerides by grossly normal male human aortas has been studied in vitro. The tissue contains an acid lipase (pH optimum, 5.4) and an alkaline lipase (pH optimum, 8.8). Both lipases catalyze the hydrolysis of saturated triglycerides; the rate decreases with increasing fatty acyl chain from C(10) to C(18). Glycerol trioleate, trilinoleate, and trilinolenate are hydrolyzed at similar rates. Alkaline lipase is inhibited about 50% at 7.2 mm glycerol trioleate, while acid lipase is unaffected at this concentration. Both lipases are activated by Ca(++) ions. The acid lipase is easily inactivated by deionized water used either as a homogenizing or dialyzing medium. Acid lipase is strongly inhibited by BSA, sodium deoxycholate, and sodium taurocholate; alkaline lipase is unaffected by BSA and is activated about twofold by bile salts. The products of hydrolysis of glycerol trioleate by aortic lipases are predominantly oleic acid and glycerol 1,2-dioleate with a small accumulation of glycerol monooleate. The aortic preparations appear to contain inhibitors for both the acid and alkaline lipase. The substance which inhibits alkaline lipase also inhibits pancreatic lipase; it is heat-stable and dialyzable. The inhibitor of the acid lipase is also heat-stable but is nondialyzable.  相似文献

The rates of hydrolysis by b-D-glucosidase, b-D-xylosidase and a-L-arabinofuranosidase isolated from Sclerotium rolfsii were increased 17 to 220 fold in organic solvents as compared to aqueous system with the highest rates occurring in chlorinated hydrocarbons. The molecular weight, log P, Hildebrand solubility parameter and dielectric constant of the solvents correlated with the activities of glycosidases.  相似文献

We studied the resolution of racemic glycidol through esterification with butyric acid catalyzed by porcine pancreatic lipase in organic media. A screening of seven solvents (log P values between 0.49 and 3.0, P being the n-octanol-water partition coefficient of the solvent) showed that neither log P nor the logarithm of the molar solubility of water in the solvent provides good correlations between enantioselectivity and the properties of the organic media. Chloroform was one of the best solvents as regards the enantiomeric purity (e. p.) of the ester produced. In this solvent, the optimum temperature for the reaction was determined to be 35 degrees C. The enzyme exhibited maximum activity at a water content of 13 +/- 2% (w/w). The enantiomeric purity obtained was 83 +/- 2% of (S)-glycidyl butyrate and did not depend on the alcohol concentration or the enzyme water content for values of these parameters up to 200 mM and 25% (w/w), respectively. The log P value of pressurized CO(2) at 50 degrees C was determined from the solubility of 1-octanol in CO(2) and compared with other solvent parameters such as permittivity, epsilon, and polarity parameter, E(T)(30). The log P indicated that pressurized CO(2) is rather hydrophilic although it seems hydrophobic being judged from epsilon(r) and E(T)(30). With a change in pressure from 3 to 11.8 MPa, the log P changed from 0.9 to 2.0 while epsilon(r) and E(T)(30) changed only slightly. The log P was linearly correlated to the logarithm of the solubility of water among organic solvents. Pressurized CO(2) was located close to the linear correlation line among the solvents at high pressure (>11 MPa) but its location deviated to the hydrophilic side with a decrease in pressure. Lipase-catalyzed esterification of stearic acid with ethanol and hydrolysis of ethyl stearate were carried out in pressurized CO(2), benzene (log P = 2.0), and n-hexane (log P = 3.5). In spite of the lowest log P value for CO(2), the reaction rate in CO(2) was the highest among solvents tested in pressure range over 10 MPa. 以化学改性后的壳聚糖为载体固定假丝酵母99-125脂肪酶,研究了不同的活化剂对壳聚糖表面羟基基团的活化程度,及以活化后壳聚糖为载体采用不同固定化方法对假丝酵母脂肪酶固定效果的影响。结果表明1-乙基-3-(3-甲基氨基)丙基碳二亚胺可有效的活化壳聚糖表面羟基,活化后的壳聚糖表面氨基与戊二醛偶联后形成的壳聚糖为良好的脂肪酶固定化载体,其固定脂肪酶的水解活力可高达86.8U/g。此外,还对影响固定化进程中的各种因素进行了研究,确定最优条件,比较了固定化前后酶的热稳定性、有机溶剂稳定性及最适反应温度。并考察了该固定化脂肪酶催化合成棕榈酸十六酯的操作稳定性,结果表明,连续反应16批之后棕榈酸十六酯的转化率仍能达到85%以上。  相似文献
