研究一次急性力竭运动对大鼠骨骼肌红肌、白肌中脂质过氧化水平的影响及血清酶的变化情况。发现运动组大鼠红肌中丙二醛(MDA)含量和碱性磷酸酶(AKP)活性比安静组显著升高;白肌中MDA含量、超氧化物歧化酶(SOD)活性和AKP活性有升高的趋势;血清肌酸激酶(CK)活性运动组比安静组有显著升高;但肌肉中MDA含量与血清CK活性之间无显著相关。因此,MDA增多可能并不是血清CK活性升高的主要原因。  相似文献

目的:观察伊普异黄酮(IP)对雄性动物生长、骨骼肌肉发育以及相关内分泌的影响。方法:24只1月龄雄性大鼠分为IP组和对照组,IP组于基础日粮中添加伊普异黄酮3mg·kg^-1,实验持续30d。结果:与对照组相比,IP组大鼠日增重和采食量分别提高12.4%(P〈0.01)和17.8(P〈0.01);胴体重增高10.70%(P〈0.05),骨骼肌重有所增加,而腰胁部脂肪重则显著降低;股骨重和骨密度均有增加;血液睾酮含量IP组大鼠超过对照组约42%,而雌二醇含量略有降低。结论:伊普异黄酮能促进雄性大鼠生长,内源睾酮在参与这一生理过程中起重要作用。  相似文献

自由基代谢普遍存在于机体各组织。正常情况下,人体自由基的产生与清除处于平衡状态。而人体自由基产生过多或机体清除自由基能力下降将给机体带来损伤。本文采用文献资料法,阐述了自由基的类型、功能、自由基产生的机制以及抗氧化系统的种类等基本理论。通过对力竭运动、耐力运动和无氧运动3种不同形式的运动对自由基代谢及抗氧化酶活性的研究得出结论:不同的运动形式及负荷方式对体内自由基代谢及抗氧化酶活性将产生不同影响;运动对自由基代谢影响具有器官与组织差异性。这些研究成果为科学指导运动训练和健身活动、快速有效的消除运动性疲劳并增强运动能力提供重要的科学理论依据。  相似文献

本文以大强度耐力训练大鼠为模型,对红景天对运动训练大鼠睾酮含量、物质代谢及抗运动疲劳能力的影响进行了研究。试验中分别以0.58、1.16、4.48 g.kg-1/d的剂量给大鼠灌胃42 d,并进行负重游泳实验、血清睾酮等生化指标测定。结果显示,红景天各剂组力竭游泳时间长于运动对照组(T组)(P<0.01);血清睾酮高于T组(P<0.01),血清皮质酮低于T组(P<0.05);各组间血清睾酮与皮质酮比值变化与睾酮变化较为一致;肝糖原(P<0.05)、肌糖原(P<0.01)高于T组;血清尿素氮低于T组(P<0.05);血红蛋白高于T组(P<0.05)。从而表明补充红景天可以减轻大鼠血睾酮、皮质酮受高强度运动量的影响,维持在正常生理水平;促进蛋白质合成,抑制氨基酸和蛋白质分解,提高血红蛋白含量和糖原的储备,增强抗疲劳能力,具有多靶点、多途径的显著特点。  相似文献

目的:探讨茶多酚对营养性肥胖大鼠肝脏自由基代谢的影响。方法:采用高脂饲料喂养,体重(200±20)g的雄性SD大鼠32只,随机分为4组(n=8),测定各组大鼠肝脏细胞O自由基和N自由基。结果:高脂饲料组大鼠肝脏超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性显著提高,茶多酚补充组丙二醛(MDA)含量比对照组及高脂饲料组显著下降;高脂饲料组大鼠肝脏TNOS、iNOS活性及NO含量显著升高,茶多酚降低了总-氧化氮合酶(TNOS)、诱导型一氧化氮合酶(iNOS)活性及NO含量。结论:高脂饲料诱导了大鼠肝脏细胞的氧化应激状态,茶多酚提高了营养性肥胖大鼠肝脏的抗氧化能力,对营养性肥胖大鼠肝脏有一定的保护作用。  相似文献

目的:研究沙棘油对6周递增负荷运动训练大鼠心肌及肝脏自由基的影响。方法:通过对运动大鼠按一定方式分组实验,选取心脏及肝脏的超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-PX)四个抗氧化指标进行测试。结果:运动训练后灌胃沙棘油能显著提高大鼠心肌及肝脏中超氧化物歧化酶,谷胱甘肽过氧化物酶以及过氧化氢酶的活性,并能显著降低丙二醛的含量。结论:证实了沙棘油具有增强抗氧化酶活性和提高大鼠运动能力的作用。  相似文献

本文以大强度耐力训练大鼠为模型,对菟丝子对运动训练大鼠睾酮含量、物质代谢及抗运动疲劳能力的影响进行了研究.试验中分别以1.16、2.32、6.96 g.kg-1.d-1的剂量给大鼠灌胃42d,并进行负重游泳实验、血清睾酮等生化指标测定.结果显示,菟丝子各剂量组力竭游泳时间长于运动对照组(T组)(P<0.01);血清睾酮高于T组(P<0.01),血清皮质酮低于T组(P<0.05);各组间血清睾酮与皮质酮比值变化与睾酮变化较为一致;肝糖原(P<0.05)、肌糖原(P<0.01)高于T组;血清尿素氮低于T组(P<0.05);血红蛋白高于T组(P<0.05).从而表明补充菟丝子可以减轻大鼠血睾酮、皮质酮受高强度运动量的影响,维持在正常生理水平;促进蛋白质合成,抑制氨基酸和蛋白质分解,提高血红蛋白含量和糖原的储备,增强抗疲劳能力,具有多靶点、多途径的显著特点.  相似文献

The age-dependent peculiarities of stimulation of free radical processes in subcellular fractions of skeletal muscle of rats subjected to long-term immobilization stress were studied in order to improve knowledge about changes of muscular tissue during ontogenesis. It is found that adult animals do not show accumulation of proteins carbonyls, TBA-reactive substances, and Schiff bases in subcellular fractions of the thigh muscle when immobUized. Long-term immobilization causes apparent manifestation of oxidative stress only in mitochondrial fraction in pubertal rats. Mitochondrial oxidative stress defense systems are sufficiently effective, however, direction of pathways of free radical oxidation carbonyl products catabolism alters in the cytoplasm of myocytes in old rats under long-term immobilization conditions.  相似文献

目的:探讨复方中药提取物对大鼠脑组织自由基代谢和抗氧化系统能力的影响机制。方法:选取70只健康Wistar大鼠,随机分为2组(n=35):对照组(N)和服药组(M)。适应性喂养1周,服药组大鼠连续服用8周的复方中药提取物,9周后将2组大鼠分别于安静状态、定量负荷、力竭运动即刻、力竭恢复12 h、力竭恢复24 h状态下处死。分别测定上述2组大鼠在不同功能状态下脑组织中丙二醛(MDA)含量,谷胱甘肽过氧化物酶(GSH-PX)、还原性谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)活性。结果:五种状态下,服药组MDA含量均显著低于对照组,GSH-PX、GSH、SOD、T-AOC活性均不同程度的高于对照组。结论:复方中药提取物可降低不同功能状态下大鼠脑组织中的MDA含量,提高其脑组织GSH-PX、GSH、SOD、T-AOC活性。  相似文献

目的:为探讨"复方中药提取物"对训练大鼠脑组织自由基代谢和抗氧化系统能力的影响及机制。方法:选取105只健康Wistar大鼠,随机分为3组(n=35):对照组(N)、训练组(T)和训练服药组(TM)。适应性喂养1周后,训练组和训练服药组递增时间游泳训练8周。9周后将3组大鼠分别于安静状态、定量负荷、力竭即刻、力竭恢复12 h、力竭恢复24 h状态下处死。测定上述3组大鼠脑组织中丙二醛(MDA)含量,谷胱甘肽过氧化物酶(GSH-Px)、还原性谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)活性。结果:五种状态下,训练组和训练服药组MDA含量均不同程度低于对照组,训练服药组低于训练组,训练组和训练服药组GSH-Px、GSH、SOD、T-AOC活性不同程度的高于对照组,训练服药不同程度高于训练组。结论:"复方中药提取物"可降低不同功能状态下训练大鼠脑组织中的MDA含量,提高其脑组织GSH-PX、GSH、SOD、T~AOC活性。  相似文献

Experimental thyrotoxicosis in rats is accompanied by the increase of serum alanine aminotransferase (AlAT), aspartate aminotransferase (AsAT), creatine phosphokinase-MB (CPK-MB) activities and the content of primary products of lipid peroxidation, conjugated dienes, in liver, heart and blood. This suggests impairments in functioning of these organs, which accompany intensification of free radical processes. Melatonin administration resulted in the decrease of AlAT, AsAT, CPK-MB and conjugated dienes; this indicates positive effect of melatonin in this pathology. Thyrotoxicosis is accompanied by the increase of catalase activity in rat liver, heart and serum. Exogenous melatonin decreased specific activity of serum and heart catalase by 22 and 43%, respectively, compared with rats subjected to hyperthyroidism. However, there was insignificant increase in specific activity of liver catalase (by ~15%). Melatonin administration caused a decrease of α-tocopherol content increased in rat tissues under conditions of hyperthyroidism. Thus, exogenous melatonin is capable to reduce intensity of lipid peroxidation in hyperthyroidism and to act as an adaptogen, regulating free radical homeostasis in response to action of pathogenic factors on organism that is associated by concomitant reduction of mobilization of components of the antioxidant system.  相似文献

Currently, considerable research activities are focussing on biochemical, physiological and pathological aspects of the creatine kinase (CK) — phosphorylcreatine (PCr) — creatine (Cr) system (for reviews see [1, 2]), but only little effort is directed towards a thorough investigation of Cr metabolism as a whole. However, a detailed knowledge of Cr metabolism is essential for a deeper understanding of bioenergetics in general and, for example, of the effects of muscular dystrophies, atrophies, CK deficiencies (e.g. in transgenic animals) or Cr analogues on the energy metabolism of the tissues involved. Therefore, the present article provides a short overview on the reactions and enzymes involved in Cr biosynthesis and degradation, on the organization and regulation of Cr metabolism within the body, as well as on the metabolic consequences of 3-guanidinopropionate (GPA) feeding which is known to induce a Cr deficiency in muscle. 目的:探讨有氧运动对衰老大鼠骨骼肌线粒体能量代谢的影响。方法:将20只12月龄的雌性Wistar大鼠随机分为老年安静组(AC,n=10)及老年运动组(AE,n=10),另取10只2月龄的雌性Wistar大鼠为青年安静组(YC,n=10);安静组大鼠进行正常饲养,运动组大鼠进行坡度为5°,速度为15.2 m/min,第1天运动15 min、第2天运动30 min、从第3天开始每天运动45 min,每周6 d,共12周。12周后所有大鼠断头处死,取腓肠肌样本,差速离心法提取线粒体,测定SOD和GSH-Px活性、MDA含量、三羧酸循环限速酶(CS、ICD和α-KGDHC)活性及呼吸链酶复合体(RCCⅠ~Ⅳ)活性。结果:①与YC组相比,AC组骨骼肌线粒体SOD活性和MDA含量显著增加(P<0.05),CS和α-KGDHC活性均显著降低(P<0.05),RCCⅠ、RCCⅡ和RCCⅣ活性均显著下降(P<0.05),RCCⅢ活性显著升高(P<0.05);AE组骨骼肌线粒体SOD、GSH-Px活性和MDA含量均显著增加(P<0.01),CS、ICD和α-KGDHC活性均显著升高(P<0.01),RCCⅠ~Ⅳ活性均显著升高(P<0.01)。②与AC组相比,AE组骨骼肌线粒体SOD、GSH-Px活性均显著升高(P<0.05),MDA含量显著下降(P<0.05),CS、ICD、α-KGDHC和RCCⅠ~Ⅳ活性均显著升高(P<0.01)。结论:有氧运动可以提高老年大鼠骨骼肌线粒体抗氧化能力,降低脂质过氧化水平,提高三羧酸循环及呼吸链功能,促进线粒体能量代谢,延缓衰老过程中线粒体的退行性变化。  相似文献

We have examined the short-term effects of leptin on protein metabolism in the rat. Indeed, an intravenous leptin administration (100 microg/kg body weight), which resulted in no changes in circulating insulin in the time interval studied, induced a decrease in the incorporation of (14)C-leucine to (14)C-skeletal muscle protein. No changes were observed in relation to muscle protein degradation (either measured in vivo following isotope preloading or in vitro as tyrosine released into the incubation medium) and gene expression associated with the different proteolytic systems (cathepsin B, m-calpain and ubiquitin-proteasome system). The effects of leptin on amino acid incorporation into muscle protein do not seem to be direct because incubation of isolated EDL muscles in the presence of 10 microg/ml of leptin did not modify either the protein incorporation or the oxidation of (14)C-leucine. It may, therefore, be suggested that leptin is able to influence protein synthesis in skeletal muscle through the action of an unknown mediator.  相似文献

The roles of creatine kinase (CK) and myoglobin (Mb) on steady-state facilitated diffusion and temporal buffering of ATP and oxygen, respectively, are assessed within the context of a reaction-diffusion model of muscle energetics. Comparison of the reaction-diffusion model with experimental data from a wide range of muscle fibers shows that the experimentally observed skeletal muscle fibers are generally not limited by diffusion, and the model further indicates that while some muscle fibers operate near the edge of diffusion limitation, no detectable effects of Mb and CK on the effectiveness factor, a measure of diffusion constraints, are observed under steady-state conditions. However, CK had a significant effect on average ATP concentration over a wide range of rates and length scales within the reaction limited regime. The facilitated diffusion functions of Mb and CK become observable in the model for larger size cells with low mitochondrial volume fraction and for low boundary O(2) concentration and high ATP demand, where the fibers may be limited by diffusion. From the transient analysis it may be concluded that CK primarily functions to temporally buffer ATP as opposed to facilitating diffusion while Mb has a small temporal buffering effect on oxygen but does not play any significant role in steady-state facilitated diffusion in skeletal muscle fibers under most physiologically relevant regions.  相似文献

[Purpose]

The purpose of the study was to verify the effects of Pilates exercise by observing the impact of 8 weeks of Pilates exercise on lipid metabolism and inflammatory cytokine mRNA expression in female undergraduates in their 20s who had no prior experience in Pilates exercise and had not exercised in the previous 6 months.

[Methods]

There were 18 subjects with no prior experience in Pilates exercise. The subjects were separated into the Pilates exercise group (n = 9) and the non-exercise control group (n = 9). The former performed Pilates exercise for 60-70 minutes over 8 weeks with a gradual strength increase of 9-16 in the Rating of Perceived Exercise (RPE). The body composition, creatine kinase in the bloodstream and lipid metabolism (TC, LDL-C, HDL-C, TG) were measured before and after the experiment and Real-Time PCR was used to investigate the mRNA expression of the inflammatory cytokines IL-6 and TNF-⍺.

[Results]

The creatine kinase (CK) in the blood had significant differences between the groups. The test group showed significant increase compared to the control group after 8 weeks of Pilates exercise (p = 0.007). Lipid analysis showed that the level of high-density lipoprotein cholesterol (HDL-C) was significantly different in the two groups (p = 0.049), with the Pilates exercise group exhibiting significantly higher levels compared to the control group. No significant differences were observed in the levels of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and triglyceride (TG). IL-6 mRNA expression did not show significant differences between the groups either. Timing and TNF-α mRNA expression showed significant effect in both the exercise and the control groups (p = 0.013) but no correlation.

[Conclusion]

It was found from the study that Pilates exercise for 8 weeks affected CK expression (the muscle damage marker) and induced positive changes in the levels of high-density lipoprotein.  相似文献
