Summary The contribution of soil respiration to the photosynthesis of the shade flora in the Amazon forest was evaluated by measuring the 13C values of leaves collected at different levels in two forest communities. Canopy leaves have an average 13C of-30.5 in the podsol forest and-28.7 in the laterite forest. Recent developments in (13) C NMR spectrometry have allowed the determination of intramolecular (13) C/(12) C ratios with high precision. However, the analysis of carbohydrates requires their derivatization to constrain the anomeric carbon. Fructose has proved to be particularly problematic because of a byproduct occurring during derivatization and the complexity of the NMR spectrum of the derivative. Here, we describe a method to determine the intramolecular (13) C/(12) C ratios in fructose by (13) C NMR analysis of the acetyl-isopropylidene derivative. We have applied this method to measure the intramolecular (13) C/(12) C distribution in the fructosyl moiety of sucrose and have compared this with that in the glucosyl moiety. Three prominent features stand out. First, in sucrose from both C(3) and C(4) plants, the C-1 and C-2 positions of the glucosyl and fructosyl moieties are markedly different. Second, these positions in C(3) and C(4) plants show a similar profile. Third, the glucosyl and fructosyl moieties of sucrose from Crassulacean acid metabolism (CAM) metabolism have a different profile. These contrasting values can be interpreted as a result of the isotopic selectivity of enzymes that break or make covalent bonds in glucose metabolism, whereas the distinctive (13) C pattern in CAM sucrose probably indicates a substantial contribution of gluconeogenesis to glucose synthesis.  相似文献

Intracerebroventricularly (i.c.v.)-administered [d-Ala2]deltorphin II (20 μg) produced a marked locomotor hyperactivity in male ICR mice. The locomotor hyperactivity induced in response to i.c.v. [d-Ala2]deltorphin II (20 μg) was suppressed by pretreatment with naltriben (NTB, 10 μg) but not 7-benzylidene naltrexone (BNTX, 1 μg) and d-Phe-Cys-Tyr-d-Try-Orn-Thr-Phe-Thr-NH2 (CTOP, 100 ng). The influence of antisense oligodeoxynucleotide to δ-opioid receptor mRNA (δ-AS oligo) or a mismatch oligodeoxynucleotide (MM oligo) on the locomotor hyperactivity induced by [d-Ala2]deltorphin II was determined. Groups of mice pretreated i.c.v. with δ-AS oligo (1 μg), MM oligo (1 μg) or saline (4 μl) once a day for 3 days, were injected i.c.v. [d-Ala2]deltorphin II (10 or 20 μg) and the locomotor response to [d-Ala2]deltorphin II was measured. The locomotor hyperactivity of i.c.v. [d-Ala2]deltorphin II (10 or 20 μg) were significantly suppressed by i.c.v. pretreatment with δ-AS oligo but not MM oligo. The present results indicate that pretreatment with δ-AS oligo suppresses mouse locomotor hyperactivity produced by stimulation of δ2-opioid receptors in the brain.  相似文献

The Golan Heights borders the Upper Jordan Valley on its eastern side and likely served as a prime foraging area for hominin groups that inhabited the Upper Jordan Valley during the Mid-Pleistocene. This study tests the hypothesis that Mid-Pleistocene climate in the Golan region was similar to that of the present day. Carbon isotope composition of present day plant communities and soil organic carbon from the Golan were compared to those of paleosols from Nahal Orvim to reconstruct Mid-Pleistocene paleoclimatic conditions. After correcting the paleosol values for recent changes in atmospheric carbon isotope values and potential biodegradation, the isotopic results show a strong similarity to those of present day local plants and soils. These results indicate that during the Mid-Pleistocene, the Golan was dominated by C3 vegetation, shared similar climatic conditions with the present day, and displayed long-term environmental stability. The span of time of paleosol formation is unknown and might cover multiple climatic episodes; thus, although short climatic fluctuations may have occurred, their impact was not substantial enough to be detected in the Nahal Orvim paleosols. This study concludes that the Golan slopes provided hominins and large grazers with a reliable and highly nutritious foraging area that complemented the Jordan Valley riparian ecosystem.  相似文献

Much of the primatology literature on stable isotope ratios of carbon (δ13C) and nitrogen (δ15N) has focused on African and New World species, with comparatively little research published on Asian primates. Here we present hair δ13C and δ15N isotope values for a sample of 33 long-tailed macaques from Singapore. We evaluate the suggestion by a previous researcher that forest degradation and biodiversity loss in Singapore have led to a decline in macaque trophic level. The results of our analysis indicated significant spatial variability in δ13C but not δ15N. The range of variation in δ13C was consistent with a diet based on C3 resources, with one group exhibiting low values consistent with a closed canopy environment. Relative to other macaque species from Europe and Asia, the macaques from Singapore exhibited a low mean δ13C value but mid-range mean δ15N value. Previous research suggesting a decline in macaque trophic level is not supported by the results of our study.  相似文献

 The origin of carbon in the spores of arbuscular mycorrhizal (AM) fungi was quantified based on their obligate symbiosis with C3 and C4 plants showing clearly different δ13C values. The δ13C values of individual spores of the AM fungus Gigaspora margarita were analyzed. In monoculture pots of a C3 or a C4 plant species, spore δ13C values were ca. 3.5‰ lower than those of host roots. In coculture pots of a C3 and a C4 plant species, spore δ13C values varied between those of the roots of C3 and C4 plants, and increased linearly from the C3 to the proximity of the C4 plant (P<0.01). This reflects the higher δ13C values in C4 plants than in C3 plants. Thus the carbon origin of G. margarita spores changed with growth state and combination of host plants. In the presence of fresh plant residue instead of living host plants, spore δ13C values did not vary with distance from the residue. This finding supports the current view that AM fungi are obligate symbionts. Accepted: 12 February 1999  相似文献

In 2005 and 2006, air samples were collected at the base of a Douglas-fir watershed to monitor seasonal changes in the delta13CO2 of ecosystem respiration (delta13C(ER)). The goals of this study were to determine whether variations in delta13C(ER) correlated with environmental variables and could be used to predict expected variations in canopy-average stomatal conductance (Gs). Changes in delta13C(ER) correlated weakly with changes in vapor pressure deficit (VPD) measured 0 and 3-7 days earlier and significantly with soil matric potential (psi(m)) (P value <0.02) measured on the same day. Midday G (s) was estimated using sapflow measurements (heat-dissipation method) at four plots located at different elevations within the watershed. Values of midday Gs from 0 and 3-7 days earlier were correlated with delta13C(ER), with the 5-day lag being significant (P value <0.05). To examine direct relationships between delta13C(ER) and recent Gs, we used models relating isotope discrimination to stomatal conductance and photosynthetic capacity at the leaf level to estimate values of stomatal conductance ("Gs-I") that would be expected if respired CO2 were derived entirely from recent photosynthate. We compared these values with estimates of Gs using direct measurement of transpiration at multiple locations in the watershed. Considering that the approach based on isotopes considers only the effect of photosynthetic discrimination on delta13C(ER), the magnitude and range in the two values were surprisingly similar. We conclude that: (1) delta13C(ER) is sensitive to variations in weather, and (2) delta13C(ER) potentially could be used to directly monitor average, basin-wide variations in Gs in complex terrain if further research improves understanding of how delta13C(ER) is influenced by post-assimilation fractionation processes.  相似文献

Summary The 13C values of whole body samples of the beetle Tribolium castaneum are closely correlated with the 13C values of the plant carbon in its diet. The d13C values of deciduous and evergreen tree leaves were compared in open and closed- canopy environments throughout a rainy season in Panamá. Newly emerging leaves had higher d13C values than older leaves of all seedlings and trees at all dates sampled. This was apparently not caused by a decline in water use efficiency as leaves develop because instantaneous ci/ca was significantly higher in newly emerging than in expanded leaves on the same twigs of trees in the field as well as on seedlings growing in a controlled, unchanging environment. Higher d13C values in newly emerging leaves occurred across diverse environmental comparisons. For example, leaves emerging during the rainy season had higher d13C values than corresponding mature leaves that had emerged both during the dry season and when water was abundant. The early enrichment in 13C may thus reflect the translocation of carbon to initiate a new leaf. Furthermore, the lack of sensitivity of this enrichment to a microclimate suggests that it might be the result of processes that occur after carbon fixation by Rubisco. Other changes in d13C values as leaves developed may also have resulted from carbon translocation processes. Foliar d13C decreased significantly after most of the leaf biomass of the deciduous Apeiba membranacea had developed. The d13C values of the evergreen Cecropia insignis were lower in the open canopy than in closed-canopy forests at the end of the rainy season. These findings suggest that the d13C values of leaves can yield ecological information about the allocation of carbon within trees.  相似文献

The foliar δ13 C values of 226 plant species sampled from 7 transects belonging to 2 rainforests (named Chengzi and Bubeng, respectively) in Xishuangbanna of Yunnan Province, China were analyzed. The characteristics of foliar δ13 C values and water use efficiency (WUE) of plants indicated by δ13 C values in this region were studied. In the mean time, the average foliar δ13 C values of plants in different transects and with different growth forms (i.e., tree, shrub, herb and liana) were compared. The results showed that: 1. the foliarδ13 C value in Xishuangbanna, ranging from -3.865%--2.760%, was similar to those of other typical rainforests in the world (Amazon and Puerto Rico, for instance), which indicated that the rainforests in this region had the typical characteristics of rainforests of the world; 2. there were significant differences between Chengzi and Bubeng in average foliarδ13 C values, with the latter being lower than that of the former; significant differences also existed between different transects in the same forest, which indicated that water conditions and orientation of transects might have resulted in these differences; 3. the average foliarδ13 C value of evergreen plants was significantly lower than that of deciduous plants, and there also existed significant differences between different growth forms, implying that the WUE of these plant species was also quite different.   相似文献

Stable isotope analysis is frequently used to infer resource use in natural populations of fishes. Studies have examined factors, other than diet, that influence δ15N and δ13C including tissue-specific rates of equilibration and starvation. Most such studies completed under laboratory conditions tightly control food quantity and its isotopic composition, but it is also necessary to evaluate the influence of these factors under more natural conditions. Using pumpkinseed sunfish (Lepomis gibbosus) we evaluated whether restricted rations below minimum daily requirements affects tissue equilibration to a change in diet by holding fish on two treatments that often reflect divergent resource use in natural populations (pelagic zooplankton or littoral macroinvertebrates). Over 42 days, δ15N values increased while δ13C values did not change, additionally neither were related to diet treatment. Increased δ15N values were negatively related to body condition while δ13C values were not, indicating that stable isotope values were more affected by decreasing body condition than by diet. Additionally, δ15N values changed more in the blood and liver tissues than in white muscle tissue, indicating that restricting food availability had greater effects on tissues with greater metabolic activity. We hypothesize that stable isotope values of consumers are subject to a tissue-specific trade-off between sensitivity to changes in resource use and resistance to the effects of low resource availability. This trade-off may require consideration in stable isotope studies of wild populations facing periodic limitations of food availability.  相似文献

Paleoecological records suggest that growing season length and/or cloudiness may affect peatland carbon accumulation and testate amoeba-based environmental reconstructions, highlighting a need to understand how light intensity affects microbial communities. We shaded plots on two peatlands for two years to examine effects on testate amoeba communities, the relative abundance of mixotrophic and heterotrophic testate amoebae, transfer-function performance, and δ13C values of two species of mixotrophic testate amoebae. Surprisingly, relative abundance of mixotrophic species increased in shade, although compositional changes did not affect transfer-function performance. Shading did not affect δ13C values of Hyalosphenia papilio and Heleopera sphagni, which ranged from −23.5 to −19.6‰ and −23.2 to −19.2‰, respectively. These δ13C values were higher than those of potential food sources and lower than literature-derived values for Chlorella, the zoochlorellae inhabiting mixotrophic testate amoebae. δ13C values thus suggest that these mixotrophic species obtain some carbon from Chlorella, although coupled dietary and isotope studies are needed to quantify this contribution. More research is needed to assess impacts of light variability on peatland microbial communities; however, carbon sources are recorded by δ13C values of testate amoebae, indicating potential for studies of carbon cycling and how mixotrophy varies temporally and spatially.  相似文献

We tested the reliability of herbivore faecal δ13C and δ15N values for reconstructing diet through review of an extensive database derived from a 3-year study of ungulates in South Africa's Kruger National Park. Faeces are a useful material for stable isotope studies of diet because they record dietary turnover at very short time scales, and because sampling is non-invasive. However, the validity of faecal isotope proxies may be questioned because they represent only undigested food remains. Results from Kruger Park confirm that free-ranging browsers have faecal δ13C consistent with C3 feeding, grazer faeces are C4, and mixed-feeder faeces intermediate. Although the respective ranges do not overlap, there is significant variation in faecal δ13C of browsers and grazers (~2.0–4.0‰) across space and through time. We demonstrate that most (~70%) of this variation can be ascribed to corresponding patterns of variation in the δ13C of C3 and C4 plants, respectively, re-enforcing the fidelity of faecal isotope proxies for diet but highlighting a need for mixing models that control for variations in plant δ13C in order to achieve accurate diet reconstructions. Predictions for the effects of climate (rainfall) and ecophysiology on 15N-abundance variations in mammals do not persist in faeces. Rather, faecal δ15N tracks changes in plant δ15N, with further fractionation occurring primarily due to variations in dietary protein (reflected by %N). Controlling for these effects, we show that a dual-isotope multiple source mixing model (Isosource) can extend diet reconstructions for African savanna herbivores beyond simplified C3/C4 distinctions, although further understanding of variations in mammal δ15N are needed for greater confidence in this approach.  相似文献

 Skeletal δ13C levels in symbiotic reef corals are believed to be predominantly influenced by metabolic fractionation. Therefore, environmental variables influencing coral metabolism should also affect skeletal δ13C levels. To test this hypothesis, we measured the effects of light (which drives photosynthesis) and relative zooplankton levels (heterotrophy) on skeletal δ13C values in the corals Pavona clavus and P. gigantea at two depths (1 m and 7 m). For both species, decreases in light or increases in zooplankton resulted in significant decreases in skeletal δ13C levels. A significant decrease in δ13C values with depth was observed in Pavona gigantea only. Thus, light and zooplankton directly affect coral skeletal δ13C values, supporting the hypothesis that metabolic fractionation significantly contributes to skeletal δ13C levels. Simultaneous decreases in both light and zooplankton resulted in decreases in skeletal δ13C values, reflecting decreases in light. In Pavona clavus, intra-annual variation in skeletal δ13C values over one year correlated with seasonal changes in irradiance. Further study is needed to resolve how skeletal δ13C values vary at intermediate levels of irradiance and zooplankton, and in other coral species. Accepted: 14 July 1998  相似文献

The foliar stable carbon isotope compositions (δ13C) of nine dominant species in seven sites, Yangling, Yongshou, Tongchuan, Fuxian, Ansai, Mizhi, and Shenmu, standing from the south to the north in the Loess Plateau of China were studied. The results showed that foliar δ13C values ranged from −22.61 to −30.73 ‰ with an average of −27.23 ‰ in 141 C3 plant samples collected from the Loess Plateau. Foliar δ13C values varied significantly (p<0.001) among the nine C3 species, which were Pinus tabulaeformis Carr., Robinia pseudoacacia L., Zizyphus jujuba Mill. var. spinosus Hu., Rubus parvifolius L., Hippophae rhamnoides L., Caragana korshinskii Kom., Lespedeza davurica (Laxm.) Schindl., Artemisia sacrorum Ledeb. var. incana Mattf., and Agropyron cristatum Gaertn. Comparatively, R. pseudoacacia, H. rhamnoides, and C. korshinskii had much higher δ13C values than the other six species, while A. sacrorum had the lowest δ13C value. There was no significant difference in foliar δ13C value among five species, P. tabulaeformis, Z. jujuba, R. parvifolius, L. davurica, and A. cristatum. Considering the life forms categorized from nine C3 species, trees and shrubs had significantly higher δ13C values than herbs (p<0.001). The deciduous tree R. pseudoacacia had much higher δ13C value than the evergreen tree P. tabulaeformis (p<0.01). Among the four shrubs, foliar δ13C values in H. rhamnoides and C. korshinskii were markedly higher (p<0.01) than those in Z. jujuba and R. parvifolius. Among the three herbs, L. davurica and A. cristatum had significantly higher δ13C values than A. sacrorum (p<0.01). Leguminous species such as R. pseudoacacia, C. korshinskii, and L. davurica as well as a non-leguminous species with nitrogen-fixation capacity, H. rhamnoides, had higher δ13C values than other non-leguminous species with same life-form. The mean δ13C value increased by about 7 % from Yangling in the south to Shenmu in the north as climatic drought increased, and foliar δ13C values differed much (p<0.001) among the seven sites. For nine species in the Loess Plateau, foliar δ13C values were significantly and negatively (p<0.001) correlated with the mean annual precipitation, moreover, an increase of 100 mm in annual precipitation would result in a decrease of 1.2 ‰ in δ13C value.  相似文献

The effects of anticoagulant EDTA and sodium heparin (SH) on stable carbon δ13C and nitrogen δ15N isotopic values of red blood cells (RBC) and blood plasma in juvenile blacktip reef sharks Carcharhinus melanopterus were analysed. Plasma preserved with anticoagulants was not isotopically distinct from plasma stored in no-additive control tubes but RBC δ15N values exhibited small enrichments when preserved with EDTA and SH. Results suggest EDTA and SH are viable anticoagulants for stable isotopic analyses of blood fractions but further studies are advised to validate results.  相似文献

Summary 13C values were measured for 45 Poaceae species collected in the northern Sahara desert, at the foot of the Saharan Atlas. The results indicate a clear relationship between carbon isotope discrimination and phytogeographical distribution of the grasses. Mediterranean species predominantly had 13C values indicating the C3 pathway of photosynthesis. By contrast, nearly all species belonging to the Saharo-Arabian and /or Sudanian group showed a C4 like carbon isotope composition. Leaf material of two species, Lygeum spartum and Stipa tenacissima, had 13C values in the region of-20, i.e. intermediate between the mean 13C values of C3 and C4 plants. However, additional speciments of both these grasses obtained from a different source (herbarium of the Hebrew University, Jerusalem) yielded a C3 like carbon isotope composition.  相似文献

Summary The 13C values of submerged aquatic plants from contrasting but relatively defined habitats, and the 13C values of emergent, floating and submerged leaves of dimorphic aquatic plants, were measured. In many instances the 13C values of dissolved inorganic carbon in the water were also measured. Plant 13C values in the vicinity of-40 to-50 were found in rapidly flowing spring waters with carbonate 13C values of-16 to-21, consistent with the notion that species such as Fontinalis antipyretica almost exclusively assimilate free CO2 via RuP2 carboxylase. Plant 13C values in the vicinity of-10 to-15 in sluggish water with carbonate 13C values of about-5 were observed, consistent with the notion that boundary layer diffusion and/or HCO3 - uptake may determine the 13C value of submerged aquatic plants in these circumstances. Comparisons of 13C values of the same or related species growing in waters of similar carbonate 13C value but different flow rates confirmed this view; more negative 13C values were frequently associated with plants in fast moving water. In Britain, but not in Finland, the 13C values of submerged leaves of dimorphic plants were almost invariably more negative than in aerial leaves. The 13C value of carbonate from chalk streams and in acid springs indicate substantial inputs of respiratory CO2, as opposed to atmospheric carbon. The contributions of these variations in 13C of the carbon source, and of isotope fractionation in diffusion, to the 13C value of submerged parts of dimorphic plants is discussed.  相似文献
