Plasma immunoreactive estrogen was significantly lower on days 9 and 8 before menstruation in a group of 15 women with premenstrual tension (PMT) syndrome, than in a control group of 17 women. Estrogen thereafter started to increase and was significantly higher in the PMT-group than in the controls on day 5 before menstruation. This increase was significant until day 1 before menstruation. Progesterone levels were constantly lower in the PMT-group than in the controls.The mean FSH level in plasma in PMT patients was significantly increased above controls on day 9, 8, 7 and 6 before menstruation. No difference between groups was seen in LH levels nor in serum albumin and binding capacity of testosterone-estradiol binding globulin (TeBG), neither could changes in LH, albumin or TeBG capacity explain the increase seen in estrogen concentration. The possible role of FSH in the PMT syndrome is discussed in order to explain the increased estrogen secretion.  相似文献

The hypothalamic-pituitary-ovarian axis of female rats was disrupted at the site of LHRH stimulation by active immunization against LHRH or at the site of LH action by active immunization against LH. Active immunization against LH was associated with an increase in pituitary LHRH receptors to levels comparable to control values at pro-oestrus whereas immunization against LHRH led to a marked reduction in receptor numbers. Ovarian LHRH receptor concentrations were increased by both treatments. It is concluded, therefore, that (1) LHRH receptors in the pituitary and ovary are not concomitantly controlled, and (2) pituitary receptor numbers are primarily under positive autoregulatory control by LHRH and that ovarian LHRH receptor concentrations may be under long-term influence of LH.  相似文献

The uptake of 125I-labelled LH by equal numbers of granulosa cells from small, medium or large follicles was greater by cells from large follicles. In contrast, granulosa cells obtained from small follicles bound much more 125I-labelled FSH per cell than did cells obtained from medium and large follicles. Competition studies with unlabelled hormones indicated that porcine granulosa cells have specific receptors for LH and FSH. The addition of diethylstilboestrol enhanced the binding of 125I-labelled LH and inhibited the binding of 125I-labelled FSH to granulosa cells harvested from small and medium-sized follicles, but had no effect on those from large follicles.  相似文献

We investigated morphometrically and densitometrically LH, FSH, PRL, and ACTH cells in pituitary glands in cows with follicular cysts and normal animals. Three groups were considered: cows in diestrus, cows in periestrus and cows with follicular cysts. The periestrous group was obtained by merging pro- and metaestrous groups, since the two did not differ when evaluated separately. The attribution to groups was confirmed by plasma progesterone measurements and by postmortem examination of the ovaries. After immunocytochemical labeling the pituitary cells were counted, and their area and total immunoreactivity were measured. The results suggest hypofunction of the LH cells (P < 0.05) and hyperfunction of the ACTH cells (P < 0.05) from the cows of the follicular cyst group. The FSH and PRL cells were similar in the three groups. These changes are consistent with the reduced LH release reported by most authors in cows with follicular cysts and should be relevant to the pathogenesis of the bovine follicular cysts. The subnormal activity of LH cells might be secondary to the activation of the ACTH cells.  相似文献

Adult Soay rams were infused for 21 days with 50 micrograms buserelin/day, using s.c. implanted osmotic mini-pumps. The continuous treatment with this LHRH agonist induced a supraphysiological increase in the blood concentrations of LH (15-fold) and testosterone (5-fold) followed by a decrease below pre-treatment values after 10 days. The blood concentrations of FSH showed only a minimal initial increase but the subsequent decrease was dramatic, occurring within 1 day. By Day 10 of treatment, the blood concentrations of all 3 hormones were low or declining, LH pulses were absent in the serial profiles based on 20-min blood samples and the administration of LHRH antiserum failed to affect the secretion of LH or testosterone. By Day 21, the secretion of FSH, LH and testosterone was maximally suppressed. The i.v. injection of 400 ng LHRH was totally ineffective at stimulating an increase in the blood concentrations of LH while the i.v. injection of 50 micrograms ovine LH induced a normal increase in the concentrations of testosterone; this confirmed that the chronic treatment with the LHRH agonist had desensitized the pituitary gonadotrophs without markedly affecting the responsiveness of the testicular Leydig cells. The ratio of bioactive: radioimmunoactive LH did not change during the treatment. The long-term effect of the infusion was fully reversible as shown by the increase in the blood concentrations of FSH, LH and testosterone and the return of normal pulsatile fluctuations in LH and testosterone within 7 days of the end of treatment.  相似文献

The hormonal interactions required for the generation of a secondary surge of FSH on the evening of proestrus have not been clearly defined. The role of GnRH in driving a surge of FSH has been questioned by findings in previous studies. In the current study, gonadotropin secretion was measured from pituitary fragments obtained from rats at 0900 and 2400 h on each day of the estrous cycle. Pituitary fragments were perifused in basal (unstimulated) conditions or in the presence of GnRH pulses to determine whether a selective increase in basal release of FSH and/or an increase in the responsiveness to GnRH occurs during the secondary FSH surge. Each anterior pituitary was cut into eighths and placed into a microchamber for perifusion. Seven pulses of GnRH (peak amplitude = 50 ng/ml; duration = approximately 2 min) were administered at a rate of one per hour starting at 30 min. Fractions of perfusate were collected every 5 min and frozen until RIA for LH and FSH. The mean total amount of LH or FSH secreted during the hour interval following each of the last six pulses of GnRH (or the corresponding basal hour) was calculated. Analysis of variance with repeated measures indicated that the evening secretion of LH on proestrus (2400 h) dropped significantly (p less than 0.05) from a maximum on the morning of proestrus (0900 h), whereas the FSH secretion remained elevated at this time. Therefore, the ratio of FSH to LH secreted in response to GnRH pulses was highest during the secondary FSH surge and lowest on the morning of proestrus.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A 'double isotope' technique has been used to describe the temporal relationship between plasma and follicular concentrations of LH after injection of 51Cr and 125I-rat LH into immature rats. Radiolabelled LH was detectable in all follicles 1 min after injection. Concentrations in small antral and large preovulatory follicles were not significantly different at any time and reached a maximum of 34.2 +/- 3.0% of plasma concentrations at 40 min. Concentrations of LH in preovulatory follicles exposed to an ovulatory dose of hCG 4 h previously were significantly greater (P less than 0.05) than those in small antral and preovulatory follicles at all times, and reached a maximum of 46.2 +/- 1.7% of plasma concentrations after 1 h. Polyacrylamide gel electrophoresis and immunoprecipitation with an antibody specific for rat LH indicated that radioactivity in plasma and follicular fluid represented radio-iodinated LH. Steroidogenic activities, light microscopy and measurements of follicular volume of each class of follicle confirmed that small antral, preovulatory follicles and preovulatory follicles exposed to an ovulatory dose of hCG in vivo could be isolated specifically. Based on these findings it is possible to calculate that, during an endogenous pulse of LH secretion, follicular concentrations of LH never exceed 20% of peak plasma concentrations. Pronounced increases in functional activities during antral growth were not correlated with increased follicular permeability. Only after acute exposure to an ovulatory dose of hCG in vivo was permeability significantly increased. We conclude that entry of LH into antral follicles is restricted and that exposure to an ovulatory dose of hCG results in greater amounts of LH entering preovulatory follicles.  相似文献

To study the plasma gonadotrophin profiles of 9 cows after parturition, blood samples were obtained every 20 min for 12 hrs on three occasions between 5 and 50 days postpartum and analysed by RIA techniques. The time of the first ovulation, as judged by plasma progesterone levels, varied from 30 to more than 60 days postpartum. Variations in mean levels of FSH and LH were not significantly correlated with the postpartum interval. However, the mean levels of plasma FSH and number of LH pulses were lower in females which had not ovulated than in those which had. The cows could be classified into four groups: group 1 with less than 4 LH pulses in 12 hrs and a mean plasma FSH level less than 138 ng/ml; group 2 with more than 4 LH pulses in 12 hrs and varying plasma FSH levels; group 3 with less than 4 LH pulses in 12 hrs and a mean plasma FSH level greater than 138 ng/ml; group 4 which had ovulated. This classification indicated that the LH and FSH levels progressed significantly (2.46 to 3.56 ng/ml, P less than 0.05; 120 to 159 ng/ml, P less than 0.01, respectively) from groups 1 to 3, and that they decreased in the females which had ovulated (group 4). Since the time of the first ovulation after parturition varied, it was not possible to demonstrate any relationship between that interval and the mean plasma gonadotrophin profiles. However, when ovulation was considered as time zero there was a clear increase in plasma gonadotrophin before ovulation.  相似文献

Changes in systemic FSH and LH pulsatility in temporal association with follicular-wave emergence and follicle deviation were studied in cattle. Wave emergence was defined as occurring when the future dominant follicle first reached 4 mm, as established retrospectively. Follicle deviation was defined as the beginning of a change in growth rates between the 2 largest follicles and occurred 60.4 +/- 4.2 h after wave emergence. Follicles were tracked by transrectal ultrasound scanning every 8 h, and blood samples for pulse characterization were collected every 20 min from before emergence until after deviation. Pulses were characterized by the Pulsar program applied to each 8-h increment, centered on the hour of follicle scanning in each heifer (n = 6). Pulsatility of FSH was not detected for any of the 8-h increments. The mean FSH concentrations for the 24 samples per 8 h increased (P < 0.05) between 8 h before and 8 h after wave emergence, followed by a decrease 40 to 16 h before deviation. The low mean values continued for 24 h after deviation. Pulses of LH were detected for all 8-h increments. The LH mean of all concentrations per 8 h and pulse frequency increased (P < 0.05) between the hour of wave emergence and 32 h after emergence and then pulse frequency plateaued at a mean interpeak interval of 1.3 h. Increased LH means for all concentrations per 8 h and basal concentration were reached 32 h before deviation. The results indicated that elevated concentrations of LH and reduced concentrations of FSH were present 32 to 16 h before to at least 24 h after the beginning of follicle deviation. However, an abrupt, short-term change in FSH concentrations or in LH pulsatility in close temporal association with follicle deviation that could act as an acute stimulus to initiate deviation was not found.  相似文献

Progesterone (P4) can participate in the development of female mammalian antral follicles through nuclear receptor (PGR). In this experiment, the differences of P4 synthesis and PGR expression in different developmental stages of sheep antral follicles (large > 5mm, medium 2-5mm, small < 2mm) were detected by enzyme-linked immunosorbent assay, immunohistochemistry, qRT-PCR and Western blotting. Secondly, sheep follicular granulosa cells were cultured in vitro. The effects of different concentrations of FSH and LH on P4 synthesis and PGR expression were studied. The results showed that acute steroid regulatory protein (StAR), cholesterol side chain lyase (P450scc) and 3β Hydroxysteroid dehydrogenase (3β-HSD) and PGR were expressed in antral follicles, and with the development of antral follicles in sheep, StAR, P450scc and the expression of 3β-HSD and PGR increased significantly. In vitro experiments showed that FSH and LH alone or together treatment could regulate P4 secretion and PGR expression in sheep follicular granulosa cells to varying degrees, hint P4 and PGR by FSH and LH, and LH was the main factor. Our results supplement the effects of FSH and LH on the regulation of P4 synthesis during follicular development, which provides new data for further study of steroid synthesis and function in follicular development.  相似文献

We studied the plasma GH profiles in 6 patients with Turner's syndrome and 6 normal girls of short stature by sampling every 20 min for 24 hours. We observed episodic secretion of GH in these subjects. The mean plasma 24 h GH level in patients with Turner's syndrome was 3.6 +/- 1.4 (SD) ng/ml which was significantly lower than that of normal short girls (7.1 +/- 2.2 ng/ml, p less than 0.01). The GH secretion during both nighttime and daytime was decreased in the patients with Turner's syndrome, however the number of pulses did not differ significantly. There were no correlations between the mean plasma 24 h GH level on one hand and peak GH level obtained after GH provocative test and plasma somatomedin C on the other. Plasma FSH and LH levels were also measured in 4 patients with Turner's syndrome. Both levels were elevated and there observed no clear pulsatile secretion of FSH, but, some pulsatile secretion of LH was observed in two patients. These data indicate that patients with Turner's syndrome have decreased endogenous GH secretion, even though they show normal GH responses to GH provocative tests.  相似文献
