Two carotenoids, neurosporene and spheroidene, have been successfully added to chromatophores from the carotenoidless mutant of Rhodopseudomonas sphaeroides R26. Carotenoids reconstituted in this way into the B-850 light-harvesting pigment-protein complex both sensitise bacteriochlorophyll fluorescence and protect the complex from the photodynamic reaction. 相似文献

The light-harvesting complex of the marine diatom Phaeodactylum tricornutum was fractionated into two large pigment-protein particles. One pigment-protein particle, which was contained in a yellow fraction, has a molecular weight, determined by gel filtration, of approx. 230 000 and can be dissociated in sodium dodecyl sulfate/mercaptoethanol solution to apopolypeptides of approx. 15 000. Characterization of particles with regard to molecular weights, subunits, protein and pigments suggests approx. 12 subunits per particle. The other pigment-protein particle, which was found in a green fraction, of approx. 95 000 molecular weight also reduces to apopolypeptide subunits of approx. 15 kDa. The relative molar proportions of chlorophyll a, chlorophyll c, fucoxanthin and total other accessory pigments in the former fraction are 3:1.3:6:2, whereas the proportions in the latter fraction are 5:1:3:1. 相似文献

We have used picosecond absorption spectroscopy with low intensity (5 · 10 11–5 · 10 12 photons · pulse −1 · cm −2) continuously tunable infrared (800–900 nm) pulses to study the energy transfer dynamics in the isolated B800–850 pigment-protein complex of Rhodobacter sphaeroides. Our results suggest the following picture of the energy transfer dynamics: (i) a fast transfer, within approx. 1 ps, from BChl 800 to BChl 850; (ii) transfer among different BChl 800's with a rate which is at the most of the same order of magnitude as that of BChl 800 → BChl 850 transfer; (iii) very fast transfer ( k > 1 · 10 12 s −1) between BChl 850 molecules. Assuming Förster type of energy transfer maximum distances of about 22 and 15 Å are obtained for the BChl 800–BChl 850 and BChl 850–BChl 850 separations, respectively. 相似文献

A role for the peripheral (B800-850) light-harvesting complex in vesicularization of the Rhodobacter sphaeroides intracytoplasmic membrane (ICM), suggested from studies in mutant strains lacking one or more of the pigment-protein complexes,
was examined further in the wild-type strain NCIB 8253 grown at high (∼1000 W m –2), moderate (∼300 W m –2), and low (∼100 W m –2) light intensities. The resulting ICM vesicles (chromatophores) had B800-850 levels related inversely to irradiance and banded
in rate-zone sedimentation at ∼1.10, 1.09, and 1.07 g ml –1, respectively. Equilibrium centrifugation on iso-osmotic gradients indicated that this distinct sedimentation behavior resulted
solely from differences in hydrodynamic radii. These size differences were confirmed by gel-exclusion chromatography and in
electron micrographs of thin-sectioned cells. A pulse-chase study of ICM growth following a tenfold reduction in light intensity
showed a relatively slow equilibration of membrane proteins during adaptation, and that new protein was incorporated largely
into additional ICM formed at the lowered illumination level, giving rise to chromatophores of reduced size and elevated B800-850
content. These results provide further evidence for a model in which the B800-850 complex both drives development of vesicular
ICM in Rba. sphaeroides and determines the size of resulting vesicles.

Optical and structural properties of the B875 light-harvesting complex of purple bacteria were examined by measurements of low-temperature circular dichroism (CD) and excitation spectra of fluorescence polarization. In the B875 complex isolated from wild-type Rhodopseudomonas sphaeroides, fluorescence polarization increased steeply across the long-wavelength Q y bacteriochlorophyll a (BChl) absorption band at both 4 and approx. 300 K. With the native complex in the photosynthetic membranes of Rhodospirillum rubrum and Rps. sphaeroides wild-type and R26-carotenoidless strains, this significant increase in polarization from 0.12 to 0.40 was only observed at low temperature. A polarization of ?0.2 was observed upon excitation in the Q x BChl band. The results indicate that about 15% of the BChl molecules in the complex absorb at wavelengths about 12 nm longer than the other BChls. All BChls have approximately the same orientation with their Q y transition dipoles essentially parallel and their Q x transitions perpendicular to the plane of the membrane. At low temperature, energy transfer to the long-wavelength BChls is irreversible, yielding a high degree of polarization upon direct excitation, whereas at room temperature a partial depolarization of fluorescence by energy transfer between different subunits occurs in the membrane, but not in the isolated complex. CD spectra appear to reflect the two spectral forms of B875 BChl in Rps. sphaeroides membranes. They also reveal structural differences between the complexes of Rps. sphaeroides and Rhs. rubrum, in both BChl and carotenoid regions. The CD spectrum of isolated B875 indicates that the interactions between the BChls but not the carotenoids are altered upon isolation. 相似文献

Low-temperature heterogeneous absorption and circular dichroism spectra of the Rb. sphaeroides LH2 complexes are calculated within the framework of the mini-exciton theory and diagonal static random disorder for the pure electronic transitions of the monomeric Bchl molecules. The coupling of Bchl molecules with the surrounding amino acid residues has been shown to change both the exciton distribution between the pigment molecules in each of the exciton states. The value of the delocalization index depends on the excitation wavelength and varies between 2-6 Bchl molecules. The optical transitions occurring at 780-790 and 820 nm have been found to be strongly mixed so that all Bchl molecules of the LH2 complex predetermine absorption in these spectral regions. On the other hand, absorption at 800 and 850 nm is mainly determined by the cycles of 9 and 18 Bchl molecules, respectively. Thus, the light energy absorbed by the B800 molecules at 800 nm is transferred to the B850 molecules by the interlevel exciton relaxation processes due to the population of the heavily mixed 820-nm exciton levels. The width of the heterogeneous absorption band for the cyclic monomeric aggregate has been shown to decrease as compared with the monomeric absorption band by square root(Ndel) time, where Ndel is the mean number of pigments over which the exciton is delocalized within the excited absorption band. 相似文献

Antenna and reaction centre complexes purified from photosynthetically-grown cells of Rhodopseudomonas sphaeroides have been mixed with cytoplasmic membranes prepared from an aerobically-grown bacteriochlorophyll-less mutant of Rp. sphaeroides (designated 01) in the presence of 1% sodium cholate. After removal of the cholate by dialysis, the dialysate was subjected to isopycnic centrifugation. Reconstituted cytochrome c2 photooxidation and cytochrome b photoreduction were demonstrated in a pigmented fraction recovered from the sucrose gradient, suggesting that the pigment-proteins were incorporated into the 01 membrane. The fluorescence properties of the system were examined. The appearance of a variable component after the initial fast fluorescence rise indicated that energy transfer occurred between the antenna and reaction centre proteins in the presence of 01 membrane. The order in which the system was assembled was important. Reconstituted energy transfer with a pre-dialysed reaction centre-antenna complex was more effective than when all the components were mixed at once. Energy transfer was also reconstituted between added reaction centre protein and the endogenous antenna present in membranes from the pigmented, but aerobically-grown reaction centre-less mutant PM8dp of Rp. sphaeroides. Preparations of 01 membranes reconstituted with reaction centre exhibited a light intensity dependent cytochrome c2 photooxidation. At low exciting light intensities, preparations containing reconstituted antenna protein in addition to reaction centres showed greater membrane cytochrome c2 photooxidation than preparations with the antenna omitted; this improvement was maximal when a pre-dialysed antenna-reaction centre complex was used. 相似文献

An isolated light-harvesting pigment-protein complex contains polypeptides which bind chlorophyll a and b. The individual complexes can be purified from detergent-solubilized membranes. The isolated light-harvesting complex, when dialyzed to remove detergents, was examined by freeze-fracture electron microscopy. The material consisted of planar sheets of 80-Å subunits which interacted via an edge-to-edge contact. Addition of cations caused the planar light-harvesting complex sheets to become tightly appressed in multilamellar stacks, with distinct subunits still visible within each lamellar sheet. A transition of particle organization from random to crystalline occurred in parallel with the cation-induced lamellar association. Treatment of the dialyzed light-harvesting complex subunits with low levels of the proteolytic enzyme trypsin removed a 2000 molecular weight segment of the major polypeptide of the light-harvesting complex and blocked all subsequent cation-induced changes in structural organization of the isolated light-harvesting complex lamellar sheets.To gain further evidence for mechanisms of cation effects upon the organization of the light-harvesting complex in native membranes, the light-harvesting complex was incorporated into uncharged (phosphatidylcholine) lipid vesicles. The protein complexes spanned the lipid bilayer and were arranged in either a random pattern or in hexagonal crystalline lattices. Addition of either monovalent or divalent cations to ‘low-salt’ (20 mM monovalent cation) vesicles containing light-harvesting complex caused extensive regions of membrane appression to appear. It is concluded that this cation-induced membrane appression is mediated by surface-exposed segments of the light-harvesting complex since (a) phosphatidylcholine vesicles themselves did not undergo cation-induced aggregation, and (b) mild trypsin digestion of the surface-exposed regions of the light-harvesting complex blocked cation-induced lamellar appression. The particles in the appressed vesicle membranes tended to form long, linear arrays of particles, with occasional mixed quasi-crystalline arrays with an angular displacement near 72°. Surface-mediated interactions among light-harvesting complex subunits of different membranes are, therefore, related to changes in structural organization and interaction of the particles within the lipid phase of the membrane.Numerous previous studies have implicated the involvement of the light-harvesting complex in mediating grana stocking in intact chloroplast membranes. The data presented herein provide a simulation of the membrane appression phenomena using a single class of chloroplast-derived membrane subunits. The data demonstrate that specific surface-localized regions of the light-harvesting complex are involved in membrane-membrane interactions. 相似文献

【目的】探求不产氧光合细菌(APB)外周捕光复合体(LH2)中类胡萝卜素(Car)结构和能量传递效率的关系和规律。【方法】通过二苯胺(DPA)抑制Car合成的方法从固氮红细菌134K20中获得部分缺失Car的LH2 (LC-LH2);采用TLC和HPLC法从3种APB中制备球形烯(SE)、玫红品(RP)和奥氏酮(OK) 3种Car;在含0.1%十二烷基二甲基胺氧化物(LDAO)的10 mmol/L Tris-HCl (pH 8.0)缓冲液中采用超声孵育法分别将这3种Car与LC-LH2体外组装,采用吸收光谱法、拉曼光谱法和荧光光谱法对组装LH2进行结构与功能分析。【结果】制备的部分缺失Car的LH2中,SE缺失率约为64.7%。这3种共轭长度、取代基的极性不同的Car均能与这种部分缺失SE的LH2自组装,Car组装率约在24.0%?29.4%之间,其中SE和OK的组装率高于RP。与部分缺失Car LH2中原有SE构象一致,重组的Car在LH2中也呈现较为伸展的平面构象。LH2中重组Car到细菌叶绿素(BChl)的能量传递效率由高到低的顺序依次为SE-LH2>RP-LH2>OK-LH2,与Car共轭体系大小的关系一致,而与Car极性大小没有明显的关系。【结论】在组装的LH2中Car采用平面构象与脱辅基蛋白结合,Car共轭长度仍是决定和影响LH2中Car-BChl能量传递效率的主要因素,而Car的取代基和极性影响较小。 相似文献

Carotenoids in the peripheral light-harvesting complexes (LH2) of the green mutant (GM309) of Rhodobacter sphaeroides were identified as containing neurosporenes, which lack the polar CH(3)O group, compared to spheroidenes in native-LH2 of R. sphaeroides 601. After LH2 complexes were treated with 1-anilino-8-naphthalene sulfonate (ANS), new energy transfer pathways from ANS or tryptophan to carotenoids were discovered in both native- and GM309-LH2. The carotenoid fluorescence intensity of GM309-LH2 was greater than that of native-LH2 when bound with ANS, suggesting that the elimination of polarity in the neurosporene increases the energy transfer from ANS to carotenoid. The fact that two alpha-tyrosines (alpha-Tyr 44, 45, B850-binding sites) in each alpha-apoprotein of GM309-LH2 were more easily modified than those of native-LH2 by N-acetylimidazole (NAI) indicates that the elimination of polarity in the neurosporene terminus increases the exposure of these sites to solution. 相似文献

Fluorescence quenching and resonance energy transfer have been studied by steady-state fluorescence spectroscopy. The experimental and theoretical values for the rate constants of the electronic energy transfer (kET) and critical radius (R0) were determined for prodan and laurdan as donors and octadecyl rhodamine B as acceptor. The spectroscopic data show, that prodan and laurdan in solution create an inhomogeneous spectroscopic medium in which multi-channel luminescence phenomena take place. This finding indicated that the modified form of the Stern-Volmer relation should be used for analyzing fluorescence quenching data. Results of performed studies point out, that dipole-dipole interaction is responsible for the resonance energy transfer from prodan and laurdan to octadecyl rhodamine B. The relative quenching efficiencies of both dyes depend on polarity of the medium and are higher for more polar solvent (AcN). 相似文献

The light-harvesting accessory pigment-protein complex (LHC) with a chlorophyll (Chl) ratio of 1.2 was isolated by treating pea chloroplasts with Triton X-100. The LHC was used to investigate the action of ionic (sodium dodecyl sulfate) and non-ionic (Triton X-100) detergents. By optical methods (absorption and fluorescence spectra, measurements of fluorescence yield, ?, and lifetime, τ) two successive stages of the process were demonstrated, namely (1) interaction between detergent monomers and proteins and (2) solubilization of pigments into detergent micelles, which is facilitated by the presence of salts. The concentration ranges, characteristic of these stages, differ by 1.5–2 orders of magnitude for SDS, but slightly overlap for Triton X-100. At the second stage, certain changes occur in LHC absorption and fluorescence spectra. Several stable states of the LHC were established: (1) an aggregated state formed in the presence of 10 mM MgSO 4 with τ ≈ 0.6 ns; (2) the dialyzed LHC with τ ≈ 0.9 ns; (3) the states of the LHC in detergent solution with τ ≈ 2.3, 2.9, 3.4 ns; (4) a 30 kilodalton monomer obtained by SDS-polyacrylamide gel electrophoresis with τ ≈ 4.1 ns. The fluorescence parameters of the LHC states were compared with those of Chl a in detergent micelles (for the micelles τ = 5.6–6.0 ns. The ratio (the criterion for emission heterogeneity) for the LHC in the absence of a detergent was shown to be higher at least by a factor of 3.5 than that for Chl a in the presence of a detergent. Successive additions of the detergent to the LHC cause gradual decrease in the ratio, and for the LHC monomer it reaches practically the same value as for Chl a in detergent micelles. The results are discussed on the basis of the data obtained previously. It is suggested that in vivo LHCs do not form such aggregates as in water solution without a detergent. 相似文献

Emission and absorption spectra in the temperature range 4–300 K have been obtained for bacteriochlorophyll light-harvesting complexes (B800–850 complexes) from several mutants of Rhodopseudomonas sphaeroides and a nonphotosynthetic mutant of Rhodopseudomonas capsulata. The energy-transfer properties of these complexes were remarkably similar despite differences in carotenoid composition. Between 300 and 200 K the excitation densities in B800 and B850 are in thermal equilibrium, indicating rapid energy transfer from B800 to B850 and vice versa. The temperature dependence of the ratio of the B800 and B850 emission yields allows the determination of the ratio of the number of B800 and B850 molecules in the complex which is close to 0.5. Below 200 K thermal equilibrium no longer exists. At 4–100 K the B800 emission yield increases with decreasing temperature and becomes dependent on the wavelength of excitation. From the B800 emission yield at 4 K the B800–850 dipole-dipole distance was calculated to be equal to or smaller than 21 Å for all B800–850 complexes. Excitation spectra for B800 and B850 emission show that the overall energy-transfer efficiencies from carotenoid and B800 to B850 are greater than 90% at all temperatures. At 4 K the carotenoid transfers its excitation energy preferentially to B850. Experiments with chromatophores indicated that the energy-transfer properties of the B800–850 complexes were not modified by the isolation procedures. 相似文献

The activity of the protein kinase that phosphorylates the light-harvesting chlorophyll-protein of Photosystem II (LHCP) has been investigated in intact chloroplasts isolated from maize mesophyll cells. Measurements of 32P incorporation into LHCP, ATP concentration, ratio, ΔpH, chlorophyll fluorescence and oxygen evolution were made in the presence of different metabolic substrates. Without added substrate a high level of LHCP phosphorylation was observed which was suppressed by addition of oxaloacetate or phosphoglycerate but stimulated by pyruvate. Whereas no correlation was observed between LHCP phosphorylation and adenylate status, a clear effect of redox state on protein kinase activity was observed. A correlation between a highly reduced electron-transfer chain (produced under conditions which favour cyclic electron flow) and the maximum level of protein phosphorylation was observed. The regulation of kinase activity and its dependence on electron transfer and carbon assimilation are discussed. 相似文献

The non-bilayer lipid monogalactosyldiacylglycerol (MGDG) is the most abundant type of lipid in the thylakoid membrane and
plays an important role in regulating the structure and function of photosynthetic membrane proteins. In this study, we have
reconstituted the isolated major light-harvesting complexes of photosystem II (PSII) (LHCIIb) and a preparation consisting
of PSII core complexes and minor LHCII of PSII (PSIICC) into liposomes that consisted of digalactosyldiacylglycerol (DGDG),
sulfoquinovosyldiacylglycerol (SQDG) and phosphatidylglycerol (PG), with or without MGDG. Transmission electron microscopy
and freeze-fracture studies showed unilamellar proteoliposomes, and demonstrated that most of the MGDG is incorporated into
bilayer structures. The impact of MGDG on the functional interaction between LHCIIb and PSIICC was investigated by low temperature
(77 K) fluorescence emission spectra and the photochemical activity of PSII. The additional incorporation of LHCIIb into liposomes
containing PSIICC markedly increased oxygen evolution of PSIICC. Excitation at 480 nm of chlorophyll (Chl) b in LHCIIb stimulated a characteristic fluorescence emission of the Chl a in PSII (684.2 nm), rather than that of the Chl a in LHCIIb (680 nm) in the LHCIIb–PSIICC proteoliposomes, which indicated that the energy was transferred from LHCIIb to PSIICC
in liposome membranes. Increasing the percentage of MGDG in the PSIICC–LHCIIb proteoliposomes enhanced the photochemical activity
of PSII, due to a more efficient energy transfer from LHCIIb to PSIICC and, thus, an enlarged antenna cross section of PSII. 相似文献

We present an optically detected magnetic resonance (ODMR) and electron paramagnetic resonance (EPR) spectroscopic study on the quenching of photo-induced chlorophyll triplet states by carotenoids, in the intrinsic light-harvesting complex (LHC) from the dinoflagellate Amphidinium carterae.Two carotenoid triplet states, differing in terms of optical and magnetic spectroscopic properties, have been identified and assigned to peridinins located in different protein environment. The results reveal a parallelism with the triplet-triplet energy transfer (TTET) process involving chlorophyll a and luteins observed in the LHC-II complex of higher plants. Starting from the hypothesis of a conserved alignment of the amino acid sequences at the cores of the LHC and LHC-II proteins, the spin-polarized time-resolved EPR spectra of the carotenoid triplet states of LHC have been calculated by a method which exploits the conservation of the spin momentum during the TTET process. The analysis of the spectra shows that the data are compatible with a structural model of the core of LHC which assigns the photo-protective function to two central carotenoids surrounded by the majority of Chl a molecules present in the protein, as found in LHC-II. However, the lack of structural data, and the uncertainty in the pigment composition of LHC, leaves open the possibility that this complex posses a different arrangement of the pigments with specific centers of Chl triplet quenching. 相似文献

Chlorophyll fluorescence, 9-aminoacridine fluorescence and O 2 evolution have been measured in a chloroplast system reconstituted to simulate the induction kinetics observed in leaves. Transients in redox state and energy state, both of which control the yield of fluorescence, were seen to depend upon (a) light intensity, (b) electron-transfer rate as controlled by ferredoxin level, (c) the initial levels of ADP and phosphate and (d) the initial level of NADP. These factors were shown to interact to produce a range of fluorescence patterns. It is suggested that in vivo fluorescence transients in part are due to reduction and phoshorylation of the finite NADP and ADP pools that exist in the chloroplast prior to illumination. 相似文献

Typical purple bacterial photosynthetic units consist of supra-molecular arrays of peripheral (LH2) and core (LH1-RC) antenna
complexes. Recent atomic force microscopy pictures of photosynthetic units in intact membranes have revealed that the architecture
of these units is variable (Scheuring et al. ( 2005) Biochim Bhiophys Acta 1712:109–127). In this study, we describe methods for the construction of heterologous photosynthetic
units in lipid-bilayers from mixtures of purified LH2 (from Rhodopseudomonas acidophila) and LH1-RC (from Rhodopseudomonas viridis) core complexes. The architecture of these reconstituted photosynthetic units can be varied by controlling ratio of added
LH2 to core complexes. The arrangement of the complexes was visualized by electron-microscopy in combination with Fourier
analysis. The regular trigonal array of the core complexes seen in the native photosynthetic membrane could be regenerated
in the reconstituted membranes by temperature cycling. In the presence of added LH2 complexes, this trigonal symmetry was
replaced with orthorhombic symmetry. The small lattice lengths for the latter suggest that the constituent unit of the orthorhombic
lattice is the LH2. Fluorescence and fluorescence-excitation spectroscopy was applied to the set of the reconstituted membranes
prepared with various proportions of LH2 to core complexes. Remarkably, even though the LH2 complexes contain bacteriochlorophyll
a, and the core complexes contain bacteriochlorophyll b, it was possible to demonstrate energy transfer from LH2 to the core complexes. These experiments provide a first step along
the path toward investigating how changing the architecture of purple bacterial photosynthetic units affects the overall efficiency

The yield of P-700 photooxidation has been studied in isolated chloroplast membranes by measuring the extent of the flash-induced absorption increase at 820 nm ( ΔA820) in the microsecond time range. The extent of ΔA820 induced by non-saturating laser flashes was increased by the following treatments. (1) Suspension of chloroplast membranes in Mg 2+ free medium (plus 15 mM K +) which leads to unstacking of grana (as detected by a decrease in chlorophyll fluorescence). (2) Reduction of Q, the primary acceptor of Photosystem II, in the presence of 20 μM 3-(3,4 dichlorophenyl)-1,1-dimethylurea by a saturating xenon flash, fired 300 ms before the laser flash. (3) Phosphorylation of light harvesting chlorophyll complex, which occurs in the presence of ATP after activation of protein kinase in the dark with NADPH and ferredoxin. We conclude that the Mg 2+ concentration, the redox state of Q and the protein-phosphorylation all can control the photochemical efficiency of P-700 photooxidation in isolated chloroplasts, and we discuss these results in relation to control of excitation energy distribution between the two photosystems. We also discuss the significance of these results in relation to the regulation of photosynthetic electron transport in vivo. 相似文献
