Behavioral analysis of near-isogenic dicofol-resistant and dicofol-susceptible spider mites did not uncover behavior patterns, present in resistant individuals yet absent in susceptible individuals, that would have reduced exposure to dicofol. On the contrary, homozygous resistant individuals were more likely than homozygous susceptible individuals to remain in contact with treated leaf areas. In the case of dicofol resistance in twospotted spider mites, resistance appears to have resulted in the loss, rather than the acquisition, of behavioral traits that lessen exposure to pesticide. Therefore, we conclude that behavioral resistance does not play a major role in twospotted spider mite resistance to dicofol.

The carmine spider mite, Tetranychus cinnabarinus (Boisduval) and the twospotted spider mite, Tetranychus urticae Koch, are serious pests of strawberries and many other horticultural crops. Control of these pests has been heavily dependent upon chemical acaricides. Objectives of this study were to determine the resistance status of these two pest species to commonly used acaricides on strawberries in a year‐round intensive horticultural production region. LC90 of abamectin for adult carmine spider mites was 4% whereas that for adult twospotted spider mites was 24% of the top label rate. LC90s of spiromesifen, etoxazole, hexythiazox and bifenazate were 0.5%, 0.5%, 1.4% and 83% of their respective highest label rates for carmine spider mite eggs, 0.7%, 2.7%, 12.1% and 347% of their respective highest label rates for the nymphs. LC90s of spiromesifen, etoxazole, hexythiazox and bifenazate were 4.6%, 11.1%, 310% and 62% of their respective highest label rates for twospotted spider mite eggs, 3%, 13%, 432,214% and 15% of their respective highest label rates for the nymphs. Our results suggest that T. cinnabarinus have developed resistance to bifenazate and that the T. urticae have developed resistance to hexythiazox. These results strongly emphasize the need to develop resistance management strategies in the region.

Wolbachia-induced 'hybrid breakdown' in the two-spotted spider mite Tetranychus urticae Koch

The most common post-zygotic isolation mechanism between populations of the phytophagous mite Tetranychus urticae is 'hybrid breakdown', i.e. when individuals from two different populations are crossed, F1 hybrid females are produced, but F2 recombinant male offspring suffer increased mortality. Two-spotted spider mites collected from two populations, one on rose and the other on cucumber plants, were infected with Wolbachia bacteria. These bacteria may induce cytoplasmic incompatibility in their hosts: uninfected (U) females become reproductively incompatible with infected (W) males. We report on the effect of Wolbachia infections in intra- and interstrain crosses on (i) F1 mortality and sex ratios (a test for cytoplasmic incompatibility), and (ii) the number of haploid offspring and mortality in clutches of F1 virgins (a test for hybrid breakdown). U x W crosses within the rose strain exhibited partial cvtoplasmic incompatibility. More interestingly, F2 males suffered increased mortality, a result identical to the hybrid breakdown phenomenon. The experiments were repeated using females from the cucumber strain. In interstrain U x W and U x U crosses, hybrid breakdown was much stronger in the former (80 versus 26%). This is the first report of a Wolbachia infection causing a hybrid breakdown phenotype. Our results show that Wolbhachia infections can contribute to reproductive incompatibility between populations of T. urticae.

Starting from the Sambucus strain of the two-spotted spider miteTetranychus urticae Koch, the strains ND1 and ND2 were selected for lack of diapause under a short-day regimen (LD 1014, 18°C). The genetic basis of the suppression of diapause in ND1 and ND2 was analysed by means of mendelian crosses and backcrosses, using the photoperiodic cycle LD 1014 as diagnostic for the distinction of diapausing and non-diapausing phenotypes. Suppression of diapause is inherited as a recessive trait. From backcross analysis it was concluded that the suppression of diapause in ND1 and ND2 is under monogenic control. The major genes for suppression of diapause in ND1 and ND2 are alleles at a locus designated asd. A mutation for albinism of the locusa-p is pleiotropic for suppression of diapause. It was shown from crosses between ND1 and the albino strain, thatd anda-p are distinct loci.

Genetic analysis of the QRPH strain of two spotted spider mite Tetranychus urticae Koch indicates that hexythiazox resistance in that strain is not sex linked and has a dominance factor of 0.152 (incomplete dominance). Hexythiazox resistance appears to be controlled by a single gene. This work contributes in part for the fulfilment of the requirements of the degree of PhD at the University of Sydney.

Tetranychus urticae is a phytophagous mite that forms colonies of several thousand individuals. These mites construct a common web to protect the colony. When plants become overcrowded and food resources become scarce, individuals gather at the plant apex to form a ball composed of mites and their silk threads. This ball is a structure facilitating group dispersal by wind or animal transport. Until now, no quantitative study had been done on this collective form of migration. This is the first attempt to understand the mechanisms that underlie the emergence and growth of the ball. We studied this collective behaviour under laboratory conditions on standardized infested plants. Our results show that the collective displacement and the formation of balls result from a recruitment process: by depositing silk threads on their way up to the plant apex, mites favour and amplify the recruitment toward the balls. A critical threshold (quorum response) in the cumulative flow of mites must be reached to observe the emergence of a ball. At the beginning of the balls formation, mites form an aggregate. After 24 hours, the aggregated mites are trapped inside the silk balls by the complex network of silk threads and finally die, except for recently arrived individuals. The balls are mainly composed of immature stages. Our study reconstructs the key events that lead to the formation of silk balls. They suggest that the interplay between mites' density, plant morphology and plant density lead to different modes of dispersions (individual or collective) and under what conditions populations might adopt a collective strategy rather than one that is individually oriented. Moreover, our results lead to discuss two aspects of the cooperation and altruism: the importance of Allee effects during colonization of new plants and the importance of the size of a founding group.

The relative toxicity of someacaricides to the predatory mite, Phytoseiulus persimilis and the twospottedspider mite, Tetranychus urticae (Acari: Phytoseiidae, Tetranychidae) wasevaluated in laboratory. Five of theacaricides tested, including bifenazate, acequinocyl, chlorfenapyr, flufenoxuron andfenbutatin oxide, were much less toxic to adultfemales and immatures of P. persimilisthan to those of T. urticae, and adultfemale predators treated with these fiveacaricides produced 84±96% as many eggs as didcontrol females. Etoxazole did not seriouslyaffect the survival and reproduction of adultfemale predators but caused high mortalityrates in eggs and larvae of P.persimilis. Milbemectin and fenazaquin werevery toxic to adult females and immatures ofP. persimilis. Adult female predatorssurvived on a diet of spider mites treated withbifenazate, acequinocyl, chlorfenapyr, flufenoxuron and fenbutatin oxide, and theirfecundity, prey consumption and the sex ratioof the progeny were not substantially affected. Based on the results, bifenazate, acequinocyl, chlorfenapyr, flufenoxuron and fenbutatin oxideappeared to be the promising candidates for usein integrated mite management programs whereP. persimilis is the major naturalenemy.

Differences among plant species in acceptance by the spider mite Tetranychus urticae Koch
Abstract: The spider mite Tetranychus urticae Koch has a broad range of host plants. However, the spider mite does not accept all plants to the same degree because of differences in nutritive and toxic constituents. Other factors, such as the induction of secondary metabolites, the morphology of a leaf surface and the presence of natural enemies, also play an important role in plant acceptance. We compared plants from various families in their degree of acceptance by the spider mite, to get an indication of the plant's direct defence. Glycine max (soybean), Humulus lupulus (hop), Laburnum anagyroides (golden chain) and Nicotiana tabacum (tobacco) were highly accepted by the spider mites. Different glandular hair densities among tobacco cultivars did not affect their suitability towards spider mites significantly. Solanum melalonga (eggplant), Robinia pseudo-acacia (black locust), Vigna unguiculata (cowpea) and Datura stramonium (thorn apple) were accepted by the spider mites to a lesser degree. Vitis vinifera (grapevine) was poorly accepted by the spider mite. It might be that the food quality of the leaves was not high enough to arrest the spider mites. Also, Capsicum annuum (sweet pepper) and especially Ginkgo biloba (ginkgo) were poorly accepted by the spider mite, probably because of the presence and concentration of certain of the secondary metabolites in the leaves. The spider mites accepted all the plants belonging to the Fabaceae for feeding, but those belonging to the Solanaceae showed a larger variance in spider mite acceptance varying from well accepted (tobacco) to poorly accepted (sweet pepper).

The twospotted spider mite, Tetranychus urticae Koch, is an important pest of ivy geranium and other ornamental plants. As a part of our long-term goal to develop an integrated crop management program for ivy geraniums, the focus of this study was to produce a reliable sampling method for T. urticae on this bedding plant. Within-plant mite distribution data from a greenhouse experiment were used to identify the young-fully-opened leaf as the sampling unit. We found that 53% of the mites on a plant are on the young-fully-opened leaves. On average 22, 37, and 41% of the leaves belonged to the young, young-fully-opened, and old leaf categories, respectively. We then developed a presence-absence sampling method for T. urticae in ivy geranium using generic Taylor's coefficients for this pest. We found the optimal binomial sample sizes for estimating populations of T. urticae at densities of between 0 and 3 mites/leaf to be quite large; therefore, we recommend the use of numerical sampling within this range of T. urticae densities. We also suggest that population estimates of T. urticae on ivy geranium be done based on mite density/unit area of greenhouse space, both for conventional greenhouse pest management, and for determining how many phytoseid predators to release when using biological control.

Populations of the two-spotted spider mite, Tetranychus urticae Koch collected from various localities and from various host plants in Japan showed wide variations in diapause attribute. Diapause percentages at 18°C/9L15D varied from nearly 100% in the north to 0% in the south-west. At intermediate latitudes the mites showed wide inter-population variations. Populations on herbaceous hosts in vinyl- or glass-houses gave significantly lower incidence of diapause than those on roses and deciduous fruit trees. Presence of winter hosts and better host quality under protected environments seemed to favour non-diapausing mites. The temperature threshold for diapause expression also varied widely among local populations. Northern populations consistently had higher and less variable thresholds than populations at intermediate latitudes with thresholds between 15 and 18°C. Inbred lines derived from a population in Kyoto exhibited a wide variation in diapause percentage at 18°C. These results show that diapause in T. urticae is a quantitative threshold trait and that populations in central Japan consist of a variety of genotypes with different diapause traits. This might provide a genetic source for adaptation to local and temporal variations in environmental conditions.

The life cycle of a non-diapausing T. urticae population was studied in a pear orchard in the southern part of Okayama Prefecture, western Honshû, Japan. The mites overwintered only on biennial weeds in the orchard without entering diapause, started to increase in number in early spring on them and then moved to other weeds as they emerged. The occurrence of the mites on pear began in early May several weeks after the unfolding of pear leaves and the population on pear was initiated by the migrants from the weeds around the base of pear trees. The population on pear showed two distinct peak occurrences, one in June and the other in mid-autumn. After pear leaves dropped in late autumn, the mites returned to biennial weeds for overwintering.

Background
The two-spotted spider mite, Tetranychus urticae, is infected with Wolbachia, which have the ability to manipulate host reproduction and fitness. MicroRNAs (miRNAs) are small non-coding RNAs that are involved in many biological processes such as development, reproduction and host-pathogen interactions. Although miRNA was observed to involve in Wolbachia-host interactions in the other insect systems, its roles have not been fully deciphered in the two-spotted spider mite. Results
Small RNA libraries of infected and uninfected T. urticae for both sexes (in total four libraries) were constructed. By integrating the mRNA data originated from the same samples, the target genes of the differentially expressed miRNAs were predicted. Then, GO and pathway analyses were performed for the target genes. Comparison of libraries showed that Wolbachia infection significantly regulated 91 miRNAs in females and 20 miRNAs in males, with an overall suppression of miRNAs in Wolbachia-infected libraries. A comparison of the miRNA and mRNA data predicted that the differentially expressed miRNAs negatively regulated 90 mRNAs in females and 9 mRNAs in males. An analysis of target genes showed that Wolbachia-responsive miRNAs regulated genes with function in sphingolipid metabolism, lysosome function, apoptosis and lipid transporting in both sexes, as well as reproduction in females. Conclusion
Comparisons of the miRNA and mRNA data can help to identify miRNAs and miRNA target genes involving in Wolbachia-host interactions. The molecular targets identified in this study should be useful in further functional studies.Electronic supplementary material

Laboratory studies were conducted to determine the effects of the mycoinsecticide Naturalis-L (Beauveria bassiana conidial formulation) on the twospotted spider mite Tetranychus urticae. Compatibility of B. bassiana with triflumuron (benzoylphenyl urea typically used as an insecticide, but also with acaricide effects), was also investigated in order to incorporate both in the control of this pest. For each juvenile stage, 180–22,800 viable conidia/mL on deutonymphs, 380–12,160 viable conidia/mL on protonymphs, and 712–7480 viable conidia/mL on larvae were evaluated. For the adult stage, the concentrations ranged from 213 to 54,720 viable conidia/mL. The mortality data used in the analysis were those accumulated after 5 days of treatment for deutonymphs and protonymphs, 7 days for larvae and 9 days for adults. The lethal concentration to kill 50% (LC50) for the juvenile stages was 3184 viable conidia/mL (their probit-log concentration regression lines were the same), and 1949 viable conidia/mL for adults. No significant differences in mortality were observed among egg age classes (24-, 48-, 72-, and 96-h-old eggs) at the tested concentrations (1400–22,800 viable conidia/mL). When B. bassiana was combined with 0.25 g Alsystin (25% triflumuron as a wettable powder)/L, mite egg mortality decreased significantly. Triflumuron reduced mycelial growth but not conidial germination of B. bassiana. This fungus is a possible candidate to be included in integrated pest management programs with triflumuron of T. urticae. In such programs, the possible antagonist effects of triflumuron should be considered.

Greenhouse experiments were conducted to determine whether different ivy geranium cultivars and different irrigation frequencies influenced population growth of the twospotted spider mite (TSM), Tetranychus urticae. Results showed that mite numbers at harvest (about 4 weeks after plants were artificially infested) were very low on 'Sybil Holmes', but significantly higher on 'Amethyst 96'. Irrigation frequency of ivy geraniums had no measurable effect on TSM populations over a range in which water was applied when pot weight dropped 15, 30 or 45% from container capacity. However, the most frequently irrigated plants (15% treatment) produced as much or more growth than plants that received less water. These plants also had the lowest incidence of oedema. There was no significant interaction between water management and host plant resistance, implying the difference in host plant resistance to TSM between the two cultivars should remain constant, at least within the range of the irrigation frequencies studied. Our findings suggest that growers can take advantage of plant resistance to TSM on ivy geranium cultivars of current commercial importance, and that watering practices can be adjusted to optimize plant production without affecting pest populations.

In field plots in which populations of predaceous species were suppressed using carbaryl, populations of the twospotted spider mite, Tetranychus urticae Koch, on field corn declined in association with aerial dispersal of the mites or with epizootics of the entomogenous fungus, Neozygites floridana Weiser & Muma. The relative importance of these two factors varied from year to year depending on environmental conditions. When moist weather conditions induced epizootics of N. floridana before corn plants became heavily infested with mites, the pathogen appeared to be the major factor causing mite population declines. When dry weather conditions allowed mite populations to expand unchecked until corn plants became entirely infested, mite aerial dispersal was the major factor associated with mite population declines. In every field in which there was an epizootic of N. floridana, routine applications of the fungicide maneb delayed epizootics and reduced or delayed the impact of the pathogen on mite populations. The total number of Neozygites-infected mites produced in maneb-treated plots throughout the test period was significantly greater than that in untreated plots when epizootics were initiated during periods of mite population growth. Fewer total infected mites were produced in maneb-treated plots compared to untreated plots when epizootics were initiated during periods of mite population decline.

Distribution of spider mites over cucumber leaves is determined by the topical trophic specialization of the pest manifested in preference for the central leaf zone. After artificial infestation of leaves with female mites, the modified aggregated distribution of the pest depended on the number of mites on the leaf, the morpho-physiological state and stage of development of the leaf and the whole plant, and the cucumber cultivar.

Alcohol extract, saponins and flavones fractionated from Dodonaea viscosa leaves were tested for their acaricidal activity against the adult females of Tetranychus urticae Koch. It was found that crude alcohol extract was more potent than the isolates obtained. At LC50 and LC90 levels, the crude extract ranked the first. All the biotic factors are significantly affected. The fecundity was seriously reduced however, the hatchability of eggs was not affected. Two flavonoids were identified as Querectin 3–0-glucose 7-0-rhamnoside and the other as Querectin 3-0 arabinose 7-0 rhammoside from the ethyl acetate fraction of the alcohol extract.
