Effect of abscisic acid on the transport of assimilates in barley   总被引:15,自引:0,他引:15  
The effect of abscisic acid (ABA) on assimilate transport in barley was investigated in two parallel experiments. First, the effect upon [14C]sucrose transport from the flag leaf to the ear of a single ABA application made at different stages of growth of the fruits was investigated; the effect was measured 24 h after treatment. Second, the effect of a single application of ABA made at the same stages of growth as above on grain weight of the mature plant was investigated. In both types of experiments ABA was applied once to the ear of different plants as an aqueous solution (10-3–10-5 M), one to five weeks after anthesis. [14C] sucrose was applied by means of agar blocks. Parallel to these experiments, the endogenous content of ABA was investigated in the developing grains. When ears were treated with ABA two or four weeks after anthesis, an increase of up to 70% in the 14C-transport from the flag leaf to the ear was observed within a 24-h period after treatment (short duration experiments). At these growth stages the endogenous concentrations of ABA were low. In sharp contrast, ABA, especially in a concentration of 10-3 M, decreased 14C-import from the flag leaf when applied three weeks after anthesis. At this stage the endogenous ABA content had reached its maximum. Long duration experiments with a single application of ABA to the car two weeks after anthesis resulted in a marked increase of weight per thousand kernels. ABA applications made earlier or later than two weeks after anthesis either reduced the grain weight or had no effect. It is concluded that ABA is involved in the regulation of assimilate transport from the leaves to the grains, possibly by influencing the unloading of sieve tubes in the ears. Promotion or inhibition of assimilate import by exogenously applied ABA may depend on the developmental stage of the grains and on the endogenous ABA level.Abbreviations ABA abscisic acid - TKW weight per thousand kernels  相似文献

Background and Aims

Previous studies have reported effects of pollen source on the oil concentration of maize (Zea mays) kernels through modifications to both the embryo/kernel ratio and embryo oil concentration. The present study expands upon previous analyses by addressing pollen source effects on the growth of kernel structures (i.e. pericarp, endosperm and embryo), allocation of embryo chemical constituents (i.e. oil, protein, starch and soluble sugars), and the anatomy and histology of the embryos.

Methods

Maize kernels with different oil concentration were obtained from pollinations with two parental genotypes of contrasting oil concentration. The dynamics of the growth of kernel structures and allocation of embryo chemical constituents were analysed during the post-flowering period. Mature kernels were dissected to study the anatomy (embryonic axis and scutellum) and histology [cell number and cell size of the scutellums, presence of sub-cellular structures in scutellum tissue (starch granules, oil and protein bodies)] of the embryos.

Key Results

Plants of all crosses exhibited a similar kernel number and kernel weight. Pollen source modified neither the growth period of kernel structures, nor pericarp growth rate. By contrast, pollen source determined a trade-off between embryo and endosperm growth rates, which impacted on the embryo/kernel ratio of mature kernels. Modifications to the embryo size were mediated by scutellum cell number. Pollen source also affected (P < 0·01) allocation of embryo chemical compounds. Negative correlations among embryo oil concentration and those of starch (r = 0·98, P < 0·01) and soluble sugars (r = 0·95, P < 0·05) were found. Coincidently, embryos with low oil concentration had an increased (P < 0·05–0·10) scutellum cell area occupied by starch granules and fewer oil bodies.

Conclusions

The effects of pollen source on both embryo/kernel ratio and allocation of embryo chemicals seems to be related to the early established sink strength (i.e. sink size and sink activity) of the embryos.Key words: Zea mays, maize, pollen, kernel, embryo, endosperm, oil, protein, starch, soluble sugars  相似文献

The reported inheritance pattern of the seg1 shrunken endosperm mutant of barley (Hordeum vulgare L. cv Betzes) suggests that some defective process in the maternal plant tissues, and not in the endosperm, prevents normal grain filling in the mutant. To identify the physiological mechanism of the mutation, we compared growth, carbon exchange, and assimilate transport of Betzes and seg1 plants. Betzes and seg1 plants did not differ in mean relative growth rate, mean net assimilation rate, or carbon exchange rate. The rate and duration of grain growth of seg1 was lower than Betzes on intact plants and on detached, cultured spikes. Increasing the supply of sucrose in culture media up to 300 mm sucrose did not eliminate differences between normal and mutant grain growth. Translocation of 14C-labeled assimilates into seg1 grains ceased by 21 days after anthesis, and assimilates were diverted to lower plant parts. In contrast, assimilates were still entering Betzes grains at 29 days after anthesis. Evidence suggests that some maternal spike or grain tissue is affected by the mutation after the onset of grain filling. Identification of the specific seg1 defect may provide information about the cessation of normal grain filling.  相似文献

The effect of exogenous abscisic acid (ABA) on the rate of sucrose uptake by soybean (Glycine max L. Merr.) embryos was evaluated in an in vitro system. In addition, the concentrations of endogenous ABA in seeds of three soybean Plant Introduction (PI) lines, differing in seed size, were commpared to their seed growth rates. ABA (10−7 molar) stimulated in vitro sucrose uptake in soybean (cv `Clay') embryos removed from plants grown in a controlled environment chamber, but not in embryos removed from field-grown plants of the three PI lines. However, the concentration of ABA in seeds of the three field-grown PI lines correlated well with their in situ seed growth rates and in vitro [14C] sucrose uptake rates.

Across genotypes, the concentration of ABA in seeds peaked at 8.5 micrograms per gram fresh weight, corresponding to the time of most rapid seed growth rate, and declined to 1.2 micrograms per gram at physiological maturity. Seeds of the large-seeded genotype maintained an ABA concentration at least 50% greater than that of the small-seeded genotype throughout the latter half of seed filling. A higher concentration of ABA was found in seed coats and cotyledons than in embryonic axes. Seed coats of the large-seeded genotype always had a higher concentration of ABA than seed coats of the small-seeded line. It is suggested that this higher concentration of ABA in seed coats of the large-seeded genotype stimulates sucrose unloading into the seed coat apoplast and that ABA in cotyledons may enhance sucrose uptake by the cotyledons.

Abscisic acid (ABA; free form) is a naturally occurring physiological growth hormone of higher plants. A detailed study involving the time course growth of developing seed tissues associated with endogenous levels of free ABA were investigated using a novel enzyme-linked immunosorbent assay. Seed filling in castor (Ricinuc communis L.) endosperm, embryo, and pod is marked with a rapid increase in fresh weight during the mid-developmental stages [21–42 days after pollination (DAP)], followed by a steady decline at the maturation stages (42–63 DAP) accompanied with a rapid lipid synthesis (in endosperm and embryo) during the same period, except for in pod. Endogenous ABA levels in endosperm (0.001–0.32 μg/g) and embryo (0.003–0.13 μg/g) followed a concurrent pattern with seed reserve filling, showing a rapid increase during the mid-developmental stages 21–42 DAP, whereas ABA levels in seed pod (0.2–22.9 μg/g) showed a different accumulation pattern with rapid increase and decline during the early-mid developmental stages, preceded by the maximal increase during the maturation stage (63 DAP). Together, our results provide evidence for the association of endogenous ABA in seed filling as well as in reserve deposition and provides clue for the effective usage of exogenous ABA concentrations in developing seeds with a focus, on improving seed reserve complex in castor.  相似文献

Factors regulating assimilate transport into developing maize (Zea mays L.) kernels have been difficult to determine because of the structural complexity of basal kernel tissues and the damage that results from tissue dissection. The sensitivity of maize kernels to experimental manipulation is such that substantial maternal tissue is required to support kernel growth in vitro. Consequently, sugar transport experiments with isolated seed tissues or detached kernels have not unequivocally demonstrated how sugar transport occurs. In the present study, Tassel-seed Tunicate (Ts-5 Tu) maize kernels were investigated as a model system for introducing test solutions into the pedicel apoplast with minimal wounding. Transpiration in leafy glumes drew 14C-sugar solutions up the 8- to 10-millimeter-long pedicel stalks into the basal endosperm transfer cell region. 14C from fructose was incorporated into starch for 8 days. Sugar uptake into endosperm and embryo tissue showed specificity and inhibitor sensitivity. In particular, p-chloromercuribenzene sulfonate partially inhibited fructose uptake into the endosperm but had no effect on the metabolic conversion of that fructose that entered the endosperm. These results are consistent with active, carrier-mediated sugar transport, but a definitive determination would require more detailed tissue analysis. We propose that further refinement of the incubation solution may allow long-term kernel growth without cob tissue and thus provide a more precise determination of which maternal factors influence seed development.  相似文献

The dry weight of harvested grain legume seeds is strongly related to their growth rate during the period of storage accumulation in the cotyledons, which begins approximately at the end of embryo cell division. Depodding, defoliation, shading or changes in air CO2 concentration were applied during seed filling (i.e. during the decrease in seed water concentration) to field and glasshouse-grown plants, in order to affect the source-sink ratio. The experiments involved three legume species, namely pea (Pisum sativum L.), soybean (Glycine max L. Merr.) and white lupin (Lupinus albus L.). Some treatments affected the number of abortions of less developed seeds from younger pods, but they did not significantly affect the number or the growth rate of filling seeds, demonstrating the priority of carbohydrate partitioning to filling seeds. The maximum growth rate of seeds was achieved regardless of the intra-plant competition level, and the duration of seed growth was shortened if the photosynthetic activity was not sufficient to fulfil the assimilate demand of filling seeds.  相似文献

The maize (Zea mays L.) kernel undergoes large changes in water content during its development. Whether such changes regulate the pattern of kernel development or are simply a consequence of it has not yet been established because other factors, such as assimilate supply, can also affect the rate and duration of kernel growth. This study was conducted to determine whether variation in kernel weight (KW) in response to source-sink treatments is mediated by a change in kernel water relations. Two hybrids were sown at three stand densities (one, eight and 18 plants m-2), and kernel numbers were restricted to control the post-flowering source-sink ratio within each stand density. Kernel development and water relations [water content, water potential (psiw), osmotic potential (psis) and turgor] were monitored throughout grain filling. Final KW varied from 253 to 372 mg per kernel in response to source-sink treatments. For both genotypes, variation in KW was a result of a change in kernel growth rate (r2 = 0.91; P < 0.001) and not in the duration of kernel filling. Final KW was closely correlated with maximum kernel water content (r2 = 0.94; P < 0.001) achieved during rapid dry matter accumulation. However, variation in KW was not reflected in kernel water status parameters (psiw, psis or turgor), which remained fairly stable across treatments. These results indicate that maximum water content provides an easily quantifiable measure of kernel sink capacity in maize. Kernel water status parameters may affect the duration of grain filling, but have no discernible impact on kernel growth rate.  相似文献

This study tests the hypotheses that in vivo oxygen levels inside developing maize grains locally affect assimilate partitioning and ATP distribution within the kernel. These questions were addressed through combined topographical analysis (O2- and ATP-mapping), metabolite profiling, and isotope flux analysis. Internal and external oxygen levels were also experimentally altered. Under ambient conditions, mean O2 concentration immediately inside starchy endosperm dropped to only 1.4% of atmospheric saturation (approximately 3.8 microm), but was 10-fold higher in the oil-storing embryo. Increasing the O2 supply to intact kernels stimulated their O2 demand, shifted ATP localization within the kernel, and elevated their ATP/ADP ratio. Enhanced O2 availability also increased steady-state levels of glycolytic intermediates and those of the citric acid cycle, as well as some related pools of free amino acids. Subsequent analyses indicated that starch formation within endosperm, but not lipid biosynthesis within embryo, was adapted to the endogenous low oxygen. Increasing the O2 supply did not change ADP-glucose levels, activity of ADP-glucose pyrophosphorylase, 13C-labeling of ADP-glucose, or flux of 14C-sucrose into starch. In contrast, enhanced O2 availability increased 14C-label uptake into the embryo, 13C-labeling of acetyl-coenzyme A, and finally 14C-incorporation into lipids. Lipid accumulation in embryo appeared highest in regions with higher ATP. Consistent with labeling data, a decrease in O2 supply most strongly affected the embryo, whereas rising O2 levels expanded ATP-rich zones toward the starch-storing endosperm and the scutellar part of embryo. The latter might be responsible for higher 14C-label uptake into the embryo and flux toward lipid. Collectively, data indicate that the in vivo oxygen distribution in maize kernels markedly affects ATP gradients, metabolite levels, and favors assimilate partitioning toward starch within the O2-depleted endosperm. Clear advantages are thus evident for peripheral localization of the protein and lipid storing structures in maize kernels.  相似文献

Two genetically related wheat lines growing in cabinets were given different temperatures during grain filling, and abscisic acid (ABA) was measured in whole grains by gas chromatography with an electron-capture detector. Three genetically related barley lines grown in the field were assayed for ABA content in endosperm and embryo fractions separately by radiommunoassay.Maximum grain growth rate and final weight per grain of the two wheat lines differed by 50–60% at low temperature and 30–40% at high temperature. During grain development two peaks in ABA level were observed at low temperature but only one at high temperature. At times when differences in grain growth rate between genotypes and between temperature treatments were large, the corresponding differences in ABA concentration were small. In barley, one line (Iabo 14) had 30% heavier grains than the other two (Onice and Opale). Endosperm ABA concentrations showed no clear differences between genotypes until grain filling was nearly complete. Embryo ABA levels were up to 10-times greater than those in the endosperm, with Opale having significantly less ABA in the embryo than the other two cultivars.Our experiments did not provide evidence for a causal relationship between ABA levels during grain filling and grain growth rate or final weight.Abbreviations ABA Abscisic acid - DAA days after anthesis - DW dry weight - FW fresh weight  相似文献

The maize cob presents an excellent opportunity to screen visually for mutations affecting assimilate partitioning in the developing kernel. We have identified a defective kernel mutant termed rgf1, reduced grain filling, with a final grain weight 30% of the wild type. In contrast with most defective endosperm mutants, rgf1 shows gene dosage-dependent expression in the endosperm. rgf1 kernels possess a small endosperm incompletely filling the papery pericarp, but embryo development is unaffected and the seeds are viable. The mutation conditions defective pedicel development and greatly reduces expression of endosperm transfer layer-specific markers. rgf1 exhibits striking morphological similarities to the mn1 mutant, but maps to a locus approximately 4 cM away from mn1 on chromosome 2 of maize. Despite reduced starch accumulation in the mutant, no obvious lesion in starch biosynthesis has been detected. Free sugar levels are unaltered in rgf1 endosperm. Rates of sugar uptake, measured over short (8 h) periods in cultured kernels, are increased in rgf1 compared to the wild type. rgf1 and wild-type kernels, excised at 5 DAP and cultured in vitro also develop differently in response to variations in sugar regime: glucose concentrations above 1% arrest placentochalazal development of rgf1 kernels, but have no effect on cultured wild-type kernels. These findings suggest that either uptake or perception of sugar(s) in endosperm cells at 5-10 DAP determines the rgf1 kernel phenotype.  相似文献

Water status parameters, flag leaf photosynthetic activity, abscisic acid (ABA) levels, grain yield, and storage protein contents were investigated in two drought-tolerant (Triticum aestivum L. cv. MV Emese and cv. Plainsman V) and two drought-sensitive (cvs. GK élet and Cappelle Desprez) wheat genotypes subjected to soil water deficit during grain filling to characterize physiological traits related to yield. The leaf water potential decreased earlier and at a higher rate in the sensitive than in the tolerant cultivars. The net CO2 assimilation rate (P N) in flag leaves during water deficit did not display a strict correlation with the drought sensitivity of the genotypes. The photosynthetic activity terminated earliest in the tolerant cv. Emese, and the senescence of flag leaves lasted 7 days longer in the sensitive Cappelle Desprez. Soil drought did not induce characteristic differences between sensitive and tolerant cultivars in chlorophyll a fluorescence parameters of flag leaves during post-anthesis. Changes in the effective quantum yield of PSII (ΦPSII) and the photochemical quenching (qP) depended on the genotypes and not on the sensitivity of cultivars. In contrast, the levels of ABA in the kernels displayed typical fluctuations in the tolerant and in the sensitive cultivars. Tolerant genotypes exhibited an early maximum in the grain ABA content during drought and the sensitive cultivars maintained high ABA levels in the later stages of grain filling. In contrast with other genotypes, the grain number per ear did not decrease in Plainsman and the gliadin/glutenin ratio was higher than in the control in Emese during drought stress. A possible causal relationship between high ABA levels in the kernels during late stages of grain filling and a decreased grain yield was found in the sensitive cultivars during drought stress.  相似文献

Field experiments using two soybean (Glycine max L. Merrill)cultivars (‘Elgin 87’ and ‘Essex’) wereconducted for 2 years near Lexington, KY, USA to evaluate theeffect of source-sink alterations on seed carbohydrate statusand growth. Sucrose concentrations in developing cotyledonsof control plants were consistently low (<50 m M) early inseed development, but they increased to 100–150 m M byphysiological maturity. The concentrations increased in bothyears by 47 to 59% when 90% of the pods were removed from ‘Elgin87’, but the increase had no effect on individual seedgrowth rate (SGR). Shading (80%) reduced cotyledon sucrose levelsand SGR in both years. The critical cotyledon sucrose concentration(the concentration providing 80% of the maximum cotyledon growthrate) was estimated fromin vitro cotyledon growth at sucroseconcentrations of 0–200 m M. These critical concentrationsvaried from 72–124 m M;in planta control cotyledon sucroseconcentrations were below this critical level during the firsthalf of seed growth but exceeded it in the later stages of growthin all experiments. The estimated critical concentration wasconsistent with the failure of in planta SGR to respond to anincrease in assimilate supply and with the reduction in SGRassociated with a decrease in assimilate supply. The resultssuggest that soybean SGR is generally sink limited if photosynthesisincreases during seed filling, but source limited if photosynthesisis reduced. Growth and development of plants are known to be affected by exposure to red and blue light. Mechanisms by which light quality influences gene expression in maize (Zea mays L.) embryos have not been explored. Maize kernels can be cultured in vitro allowing experimental manipulation of environmental factors during seed development. We used the in vitro kernel culture system to investigate the response of developing maize seeds, which normally develop without exposure to light, to controlled light quality. Kernels grown under red light accumulated more dry weight than those grown in darkness, whereas kernels grown under blue light accumulated less. Reciprocal color shift experiments showed that light quality during the first week in culture had more influence on kernel weight than during the subsequent three weeks in culture. Soluble sugars were higher in both light treatments than in darkness. Blue-grown kernels had higher amino acid and lower lipid levels than red-or dark-grown kernels. Embryo morphology was markedly affected by red light, under which the upper shoot axis was longer than under blue light or in darkness. Embryo morphology was influenced by light quality during the later stages of development rather than the first week. We suggest, based on these results, that gene expression in the embryo and endosperm of developing maize seeds is sensitive to light quality, and the mechanism and time dependence of this effect warrant further study. Accumulation of cations in roots and shoots and influx of K+(86Rb+) and Ca2+(45Ca2+) into the roots were investigated in spring wheat (Triticum aestivum L. cv. Svenno). Plants were sampled at four main developmental stages: tillering, shooting, heading and grain filling. The effects of switches between a high and a low supply of nutrients were characterized. Growth of the shoots and roots was affected by the switches. A high supply of nutrients at the seedling stage and towards tillering supported a high growth rate, while a further high supply of nutrients increased vegetative growth and delayed grain filling. An early high supply of nutrients followed by a low supply at shooting, heading and grain filling accelerated root growth and growth of the main culm. Switches of the mineral supply gave only small changes in concentration of Mg in the plants. Generally, the K+(86Rb+) influx into the roots decreased during ontogenesis, while Ca2+(45Ca2+) influx increased more or less independent of the switches between nutrients levels.  相似文献

Maize (Zea mays L. cv. Pioneer 3925) subjected to post-anthesiswater stress during the first 2 weeks of kernel developmenthad lower leaf-water potentials and higher leaf-ABA concentrationsthan well-watered controls. There was a concomitant rise inABA concentration in kernel tissues 3 and 7 d after pollination(DAP), after which the concentration decreased to control levelsby 13 DAP. Kernel water potential, however, remained unchangedby the water stress. Radiolabelled ABA, fed to a leaf, was translocatedto kernels, where free ABA as well as several ABA metaboliteswere the major labelled fractions. This suggested that the stress-inducedkernel ABA was of maternal origin. Since ABA plays a putativerole in seed maturation of several crop species, and appliedABA or water stress often hastens seed development, we expectedthat a water-stress-induced rise in kernel ABA concentrationearly in grain development may serve to prematurely induce storage-productaccumulation. Development of Brassica napus L. cv Tower embryos of different ages cultured in vitro with and without abscisic acid (ABA) was compared with normal development in situ to investigate the role of ABA in embryo maturation. Endogenous ABA levels were measured by radioimmunoassay, and sensitivity to ABA was assayed in terms of its ability to suppress precocious germination and stimulate accumulation of storage protein and storage protein mRNA. During development in situ, the levels of endogenous ABA and 12S storage protein mRNA both reach their peaks just before the embryos begin to desiccate. The ABA levels during this phase of development also correlate with the time required in culture before germination is evident. Following these peaks, increasing concentrations of exogenous ABA are required to both suppress germination and continue storage protein accumulation in vitro. Thus, both endogenous ABA and ABA sensitivity decline during maturation. The concentrations of exogenous ABA required to suppress germination at these later stages result in abnormally high levels of endogenous ABA and appear to be toxic. These results are consistent with the hypothesis that in maturing rapeseeds, low water content rather than ABA prevents germination during the later stages of development.  相似文献
