The germination of Empress tree (Paulownia tomentosa Steud.) seeds is phytochrome-controlled. Liquid smoke could not induce germination in darkness but red light irradiation of liquid smoke imbibed seeds induced a high percentage of germination. Maximum germination was achieved at liquid smoke concentration of 0.1% (v/v) when present during the imbibition phase or during the phase of phytochrome activity. The light requirement of these seeds could be completely substituted by exogenously applied gibberellins. In the presence of liquid smoke, optimal concentrations of GA3, GA4, and GA9 necessary for inducing germination were several times lower than in the controls, while that of GA7 was equally active when applied at a concentration one order of magnitude lower. The inhibitory effect of the applied growth retardants was strongly reduced and liquid smoke, in the presence of retardants, allowed light-induced germination, if applied simultaneously or after retardants treatment.  相似文献

Abstract

The spores of Woodwardia radicans can germinate indifferently either in water or in culture media containing mineral salts at temperatures (15-24°C) falling within a range believed optimal for many other ferns (15-30 C).

The spores are photosensitive, will not germinate in the dark and the addition of gibberellic acid is ineffective in substituting a light requirement. Spore germination was induced by white and red light and phytochrome seems to be implicated in the control of germination since far-red light (and not the blue irradiation) can reverse the stimulating effect of the red light.

Spore morphology and spore germination pattern was studied using light and scanning electron microscopes.

It was concluded that the progressive disappearance of W. radicans from the Italian localities is not due to difficulties in spore germination but is related to problems that arise during the subsequent stages.  相似文献

Germination occurs usually in response to multiple environmental cues. Seeds with the ecophysiological ability to simultaneously sense the previous presence of fire and appropriate levels of temperature, light and soil nitrate could restrict germination to postfire, winter and competition-free microhabitats, where the potential for seedling survival is enhanced. Germination responses of 16 species with a range of life forms, fire responses and seed weights were determined under controlled conditions of 15°C temperature, a 12 h light cycle, exposure to 1 g L?1 nitrate solution, and six conditions of light quality (white, blue, yellow, red, far-red light and darkness). Germination in Oenothera stricta, a weedy naturalized ephemeral, and two small-seeded indigenous Asteraceae species of mulga woodlands, Leucochrysum fitzgibbonii and Craspedia sp., were enhanced by white, yellow or red light compared with germination achieved in the dark, or under far-red or blue light. In red light, KNO3 further enhanced germination of these positively photoblastic species. The germination response of Trachyandra divaricata, a naturalized herb of sandy, seaside locations, and several native jarrah forest legumes (four Acacia species, Bossiaea aquifolium, Gompholobium marginatum and Sphaerolobium vimineum) proved to be negatively photoblastic. Of these seven negatively photoblastic herb and shrub species, exposure to KNO3 overcame the inhibition of light only in the resprouter species, Acacia lateriticola. In the serotinous, negatively photoblastic tree species, Corymbia calophylla and Eucalyptus marginata, KNO3 seemed to be required before the negative response to light exposure was recorded. A dose–curve experiment on two positively photoblastic and three negatively photoblastic species indicated that although KNO3 exposure affected germination in all species, different concentrations of KNO3 (0, 0.5, 1, 2, and 5 g L?1) produced different levels of response. Detailed studies with additions of KNO3 (1 g L?1) and the growth hormone, gibberellic acid (GA3; 50 mg L?1), showed that increased germination percentages of the positively photoblastic species, Oenothera stricta, occurred in the light, but blocking endogenous gibberellic synthesis with paclobutrazol, or adding exogenous GA3 or KNO3 had no effect on the light-induced germination levels. In the negatively photoblastic species Trachyandra divaricata, additions of KNO3 and GA3 had no influence on the germination inhibition induced by exposure to light nor did blocking endogenous GA synthesis. The 16 species growing naturally in Western Australia, Australia show a range of germination responses to environmental conditions, but depending on their natural habitat, the ecophysiology of each species appears to be optimized for subsequent seedling survival.  相似文献

The effects of various substances upon germination and tube growth of pollen of Setaria sphacelata were investigated in hanging-drop culture. Both sucrose (0.6–0.7 M) and boron (1–5 ppm, as borate) are essential for germination. Comparable results were obtained with boric acid, sodium tetraborate and tri-n-butyl borate as boron sources, but sodium tetraphenylboron was inhibitory. Good germination and growth were obtained with raffinose and cellobiose, alone or in combination with sucrose (total 0.5 M); 0.25 M rhamnose, lactose and glycerol were without effect, and xylose, galactose and glucose were slightly inhibitory in the presence of 0.25 M sucrose; 0.25 M arabinose, fructose, mannose, sorbose, maltose, mannitol and sorbitol completely inhibited germination, even in the presence of 0.25 M sucrose. IAA and GA (0.01–10 ppm) could not replace or supplement the effects of borate on germination and growth, indicating the pollen to be self-sufficient in this respect. Riboflavin (0.1–10 ppm) and calcium pantothenate (1–100 ppm) stimulated germination and growth, whereas 0.01–10 ppm of thiamine, pyridoxine, nicotinic acid and ascorbic acid were generally without effect. Although copper sulphate, manganese sulphate, zinc sulphate and ammonium molybdate could not replace boric acid, 1.0 ppm of copper, manganese and zinc stimulated germination and growth in the presence of boron. In no instance were tubes found comparable in length to those required for fertilisation in vivo. It was concluded that the pollen probably requires a complex mixture of substances, including sucrose and borate, before this can be achieved in vitro.  相似文献

In order to understand the biological activity of humic substances (HS), the effect of four humic acids (HA) and one fulvic acid (FA) on seed germination and early growth of cosmopolitan weed Chenopodium album agg. were tested. Humic substances of diverse origin were used, namely purified commercial HA, HA isolated from lignite, cambisol and podzol, and FA from mountain spruce forest soil. Data processing by two-way ANOVA has shown that type of the tested substances was a more important factor on seed development than used concentration. The major differences in germination and length of shoots were found in the first days of the experiment. Commercial and lignite HA stimulated the seed germination and growth, while podzol HA inhibited them. After this initial phase, all humic substances stimulated the seed development, but these stimulations were not significant (P < 0.05). At the end of germination test, the greatest stimulation effect (up to 20%) was achieved with the lignite HA.  相似文献

Passage rate through the digestive tracts of zebu cattle and sheep, and subsequent germination of egested seeds of four woody species from the Sudanian savanna, Acacia dudgeoni, Acacia seyal, Burkea africana and Prosopis africana, were studied. The result indicates large differences in passage rate among woody species, as well as between animals. The values ranged from 46% to 87% for seeds ingested by cattle while the lowest passage rate was 2.3% and the highest being 74% for seeds ingested by sheep. Among plant species, seeds of Prosopis africana had the highest passage rate through the digestive tract of both cattle and sheep. Seed passage through the gut showed a significant positive correlation with seed mass and thickness for cattle and sheep, respectively. The gut treatment and the retention time in the gut did not improve germination capacity and the speed of germination of dormant seeds. For non-dormant seeds of Acacia dudgeoni, the germination capacity was higher for seeds ingested by cattle than sheep. The speed of germination was also significantly higher for egested seeds than the control. It can be concluded that large herbivores could play an essential role in long distance dispersal of seeds. Gut treatment alone was not effective in breaking seed coat-imposed dormancy, although it enhanced the rate of germination of non-dormant seeds. To get a complete picture of the effect of frugivore on the release of seed dormancy, the combined effect of initial mastication and subsequent gut treatment needs to be investigated.  相似文献

Seed dormancy characteristics of Spartina alterniflora were delineated previously by Plyler and Carrick (American Journal of Botany, vol. 80, pp. 752–756, 1993). This study was undertaken in order to determine whether or not the dormancy characteristics of S. patens are similar. As in the previous study, the site of a dormancy mechanism was determined by assessing the germinability of surgically altered dormant seeds. Likewise, the effects of three growth-regulating substances (abscisic acid, fusicoccin, and gibberellic acid), prechilling, and light were studied by assessing the germinability of appropriately treated dormant seeds. Surgical injury to the endosperm, and to a lesser extent the scutellum, produced significant germination in S. patens, whereas only injury to the scutellum produced germination in S. alterniflora. Exogenously applied abscisic acid was highly effective in maintaining dormancy in injured seeds of both species. Gibberellic acid and fusicoccin were ineffective in promoting germination in S. patens, but fusicoccin was highly effective in breaking dormancy in 5. alterniflora. Prechilling was effective in breaking dormancy in S. patens but not in S. alterniflora. In S. patens, treatments that broke dormancy were only successful when seeds were exposed to light during the germination period. It is concluded that the dissimilarities in the dormancy mechanisms may be manifestations of the different environmental adaptations these closely related species display.  相似文献

The effect of the application of gibberellins to Tagetes minuta L. achenes (seeds) was determined at both 25°C, the optimal germination temperature, and 35°C, at which temperature the achenes are thermoinhibited. Both GA3 and GA4+7 accelerated germination at 25°C. Seed germination at 25°C was inhibited by paclobutrazol, but on subsequent application of GA4+7 rapid germination was induced. Following application of GA3 or GA4+7 to thermoinhibited seeds, a significantly higher final germination percentage was observed than in the distilled water control. However, endogenous gibberellin levels in germinating and thermoinhibited seeds did not differ significantly.  相似文献

Both dark and red light germination of lettuce seeds (cv. “Maikönig”) as well as their root and hypocotol elongation were inhibited when the seeds were sown in petri dishes together with a few seeds of Heracleum laciniatum Horn. This inhibition was not significantly counteracted by the presence of gibberellic acid (GA3) or/and 6-benzylaminopurine (BA). However, a large proportion of the lettuce seeds germinated abnormally (only cotyledons emerged) when treated with BA in the presence of Heracleum seeds. GA3 had alone no significant effect on abnormal germination, but it counteracted the effect of BA to some extent. The inhibitory effect of Heracleum seeds gradually disappeared during a moist incubation period of one to seven days in darkness at 25°C. When lettuce seeds were pre-incubated together with Heracleum seeds for one to five days the remaining, non-germinated lettuce seeds had lost their ability for subsequent germination in darkness in distilled water. This induced dark dormancy was to a great extent broken by red light, but not by GA3 or/and BA. H. laciniatum seeds inhibited the germination of Salix pentandra seeds and to some extent also the germination of radish but had no effect on the germination of spruce.  相似文献

Summary Effect of cumin seed exudates on spore germination of Curvularia lunata, Alternaria solani and Colletotrichum capsici was studied in the laboratory. Three-hour exudate obtained for first 3-hour favoured initial protrusion but inhibited subsequent growth of the germ tubes. Two-hour exudate obtained between 5 and 7 hours after soaking was suggested to have no effect on initial protrusion but was found to have promotive effect on subsequent growth of germ tube. Short treatment with inhibitor for 45 minutes followed by a treatment with promotor ensured high germination and uniform germ tube growth. The effects of solutions of sodium hypochlorite and calcium hypochlorite (bleach) on dormancy and germination of Alectra vogelii seeds were investigated. Dry (non-pretreated) and 10-day water-pretreated seeds were exposed to various bleach concentrations, as well as to the host (Vigna unguiculata) root exudate. The 15-month-old Alectra seeds used were partially dormant in that only 40% or less of the 10-day pretreated seeds could be stimulated to germinate by the standard host root exudate. Comparable percentage germination of nonpretreated seeds was achieved with certain concentrations of bleach and the halogens, chlorine and bromine. Bleach and the halogens served not only as germination stimulant for pretreated Alectra seeds but appropriate concentrations induced also high percentage (70–90%) germination indicating a breakage of seed dormancy as well. The activity of the bleach in stimulating high percentage germination could be significantly reduced when the optimally bleach-treated seeds were rinsed daily with water during the germination period. Vigna root exudate, but not bleach, was shown to act as root stimulant for Alectra seedlings. The possible mechanism of the bleach effect on germination and dormancy of Alectra seeds is discussed.  相似文献

Abscisic acid (ABA) is involved in regulating diverse biological processes, but its signal transduction genes and roles in hemp seed germination are not well known. Here, the ABA signaling pathway members, PYL, PP2C and SnRK2 gene families, were identified from the hemp reference genome, including 7 CsPYL (pyrab-actin resistance1-like, ABA receptor), 8 CsPP2CA (group A protein phosphatase 2c), and 7 CsSnRK2 (sucrose nonfermenting1-related protein kinase 2). The content of ABA in hemp seeds in germination stage is lower than that in non-germination stage. Exogenous ABA (1 or 10 μM) treatment had a significant regulatory effect on the selected PYL, PP2C, SnRK2 gene families. CsAHG3 and CsHAI1 were most significantly affected by exogenous ABA treatment. Yeast two-hybrid experiments were performed to reveal that CsPYL5, CsSnRK2.2, and CsSnRK2.3 could interact with CsPP2CA7 and demonstrate that this interaction was ABA-independent. Our results indicated that CsPYL5, CsSnRK2.2, CsSnRK2.3 and CsPP2CA7 might involve in the ABA signaling transduction pathway of hemp seeds during the hemp seed germination stages. This study suggested that novel genetic views can be brought into investigation of ABA signaling pathway in hemp seeds and lay the foundation for further exploration of the mechanism of hemp seed germination.  相似文献

We investigated seasonal variations in allelopathic potential ofArtemisia princeps var.orientalis. Aqueous and meth-anol extracts and volatile substances were prepared in the laboratory from samples collected monthly (April through October). Their impacts were then assessed on the germination and seedling growth ofLactuca sativa andAchyranthes japonica. The allelopathic potential varied with the time of sample collection and the concentration tested. For example, germination ofL. sativa was not inhibited by the aqueous extract but seedling growth (shoots and roots) was, with its seasonal effect being significant. ForA. japonica, seed germination was not inhibited at lower concentrations (except for August samples). However, at higher concentrations and in certain months (especially July), germination was more negatively affected. The degree of seedling growth inhibition also differed by month and by extract concentration, with roots being impacted more than shoots. Volatile substances also had a time-dependent influence on the germination and seedling elongation ofA. japonica. In a separate experiment, the ethyl-acetate and water fractions of a crude methanol extract were prepared monthly fromA. princeps var.orientalis. Here, we examined their antimicrobial activities against three gram-positive bacteria (Bacillus cereus, Bacillus subtilis, andStaphylococcus aureus), two gramnegative bacteria (Escherichia coli andPseudomonas fluorescens), and one lactic acid bacterium,Lactobacillus plantar urn. The ethyl-acetate fraction that was sampled in September was remarkably potent againstB. cereus andB. subtilis, whereas the water fraction collected in August and September showed great antimicrobial activity against the grampositive and -negative bacteria. In contrast,L. plantarum was not inhibited by the water fraction, regardless of the sampling month. Likewise, the ethyl-acetate and water fractions collected in April and October had the lowest levels of antimicrobial activity.  相似文献

Erica andevalensis Cabezudo & Rivera is a threatened edaphic endemic species of Andalusia (SW Spain). Under natural conditions, the plants produce a very large number of small seeds (0.3–0.4 mm) but very few seedlings survive. Different treatments (high temperature, cold pre-treatment, nitrogen salts, and gibberellic acid applications) were tested to assess germination patterns in different populations and to determinate the most favorable conditions for germination. Gibberellic acid was provided in five different concentrations from 0 to 400 ppm GA3, while nitrogen was applied as 10 mM of either KNO3 or NH4NO3. The effect of pH on germination was also tested. The species always showed a low germination rate (6.50–22%) that was not stimulated either by 1 or 4 months in dry cold pre-treatment, nitrogen application, acid pH medium, or by high temperature (80°C for 10 min); although gibberellic acid application (100–400 ppm) significantly enhanced germination. The highest percentage of germination (41.6%) was achieved with a mean germination time to start germination (t 0) of 7.6 ± 0.54 days when the seeds were subjected to 400 ppm gibberellic acid treatment. The population origin did not have a significant effect on germination percentage.  相似文献

Divalent cations were found to cause a specific release of peptidase from mechanically encysted zoospores of the fungusAphanomyces astaci, whereas germination could be induced only by CaCl2, SrCl2, and MgCl2 with CaCl2 being most efficient. It is concluded that peptidase secretion is a prerequisite to enable the cysts to become capable of germination, but that germination is not a consequence of peptidase release. Following this conclusion it is suggested that CaCl2, SrCl2, and MgCl2 in themselves are unable to elicit germination and that other unknown processes in the cell are triggered which allow germination to take place. These processes are short-lived and can be elicited only in young encysted spores, whereas 15-min-old mechanically encysted zoospores did respond to CaCl2 by the release of peptidase but did not germinate. The mechanism by which divalent cations caused the release of peptidases could not be elucidated, since drugs known to affect exocytosis in animal cells lysed the cysts even at very low concentrations.  相似文献

Seeds were collected from plants of Medicago sativa var. local inoculated with Glomus macrocarpum and G. fasciculatum separately in pot experiments. These seeds were sown in garden soil and the percentage germination, general health and yield of subsequent plants (the F1 generation) were studied. The percentage germination was highest in seeds of G. macrocarpum-inoculated parents followed by those inoculated with G. fasciculatum; seeds of uninoculated parent plants showed the lowest germination. Vegetative yield of the progeny decreased in the order of plants inoculated with G. fasciculatum, with G. macrocarpum, and uninoculated. On the other hand, reproductive yield was highest for plants whose parents were inoculated with G. macrocarpum, followed by G. fascicullatum, and lowest for seeds of uninoculated parent plants.  相似文献

Seed dormancy induction and alleviation in the winter‐flowering, moist temperate woodland species Galanthus nivalis and Narcissus pseudonarcissus are complex and poorly understood. Temperature, light and desiccation were investigated to elucidate their role in the germination ecophysiology of these species. The effect of different seasonal temperatures, seasonal durations, temperature fluctuations, the presence of light during different seasons and intermittent drying (during the summer period) over several ‘years’ on seed germination was investigated with outdoor and laboratory experiments. Warm summer‐like temperatures (20 °C) were necessary for germination at subsequent cooler autumn‐like temperatures (greatest at 15 °C in G. nivalis and 10 °C in N. pseudonarcissus). As the warm temperature duration increased, so did germination at subsequent cooler temperatures; further germination occurred in subsequent ‘years’ at cooler temperatures following a second, and also third, warm period. Germination was significantly greater in darkness, particularly in G. nivalis. Dormancy increased with seed maturation period in G. nivalis, because seeds extracted from green capsules germinated more readily than those from yellow capsules. Desiccation increased dormancy in an increasing proportion of N. pseudonarcissus seeds the later they were dried in ‘summer’. Seed viability was only slightly reduced by desiccation in N. pseudonarcissus, but was poor and variable in G. nivalis. Shoot formation occurred both at the temperature at which germination was greatest and also if 5 °C cooler. In summary, continuous hydration of seeds of both species during warm summer‐like temperatures results in the gradual release of seed dormancy; thereafter, darkness and cooler temperatures promote germination. The effect of iron on root elongation and seed germination of Lactuca sativa was investigated within the limits of the plant's sensitivity to an acidic environment and iron solubility under experimental conditions. Because ionic strength was found to have phytotoxic effects, the same pH and ionic strength conditions were kept constant over the entire iron concentration range, so as to evaluate solely the effect of iron. 0.14 mM Fe3+ was found to be the lowest effective concentration inhibiting root elongation. The low solubility of iron at a pH of 3 (the lowest value which could be applied for root elongation) excluded the testing of higher iron concentrations. Due to the lower sensitivity of seed germination to acidity, a pH of 2.6 could be applied in this case. The lowest effective iron concentration (the lowest applied concentration causing a significant decrease in germination as compared to the control, P < 0.05), EC50 (the concentration at which germination was 50% of that of the control specimens) and the completely effective iron concentration (the concentration that completely inhibited seed germination) were 0.6, 0.92 and 2.0 mM, respectively.  相似文献

以普洱地区14种常见植物种子为材料,在实验室条件下研究了其在白光、黑暗、红光和蓝光条件下的萌发特性,并分析了种子大小与萌发率、萌发速率、萌发开始时间的关系。结果表明:光质对四方蒿、沙针、尖子木、藿香蓟种子萌发率和萌发速率均有显著影响(P0.05)。光质对大叶斑鸠菊、云南山枇花、臭灵丹、车桑子、光萼猪屎豆、葫芦茶、云南地桃花、西南宿苞豆、岗柃、中国宿苞豆10个物种的种子萌发率和萌发速率均没有显著影响(P0.05),以上物种中除中国宿苞豆外,其他物种种子萌发率均在20%以下,处于休眠状态。四方蒿种子在白光(89.9%)和红光(84.7%)下萌发率最高,红光下种子萌发最快(4.93),蓝光下种子萌发开始时间最晚(11.3 d);沙针种子在白光下萌发率最高(80.4%)、萌发速率最快(2.71),在黑暗和蓝光下萌发率较低(43.9%和38%)、萌发速率最慢(0.73和0.85),白光、红光下萌发开始最早(11 d),黑暗条件下萌发开始最晚(21.7 d);尖子木种子萌发率在白光、黑暗、蓝光下均在86%以上,而红光下仅32%且萌发速率最慢(1.29),在蓝光下萌发开始时间最晚(13 d);藿香蓟种子萌发率和萌发速率在红光下最高(分别为71.3%和6.46),黑暗条件下最低(分别为42.5%和2.62);大叶斑鸠菊萌发开始时间在黑暗条件下最早(6 d),其次是白光下(7 d),蓝光和红光下较晚,分别为8 d和7.7 d。14个物种种子的萌发率与种子大小间均有显著负相关关系;种子萌发速率、萌发开始时间与种子大小间也有负相关关系,但不显著;种子大小与萌发率、萌发速率和萌发开始时间的关系不会随着光质的变化而发生变化。  相似文献

Summary Eluates of stigmas of Brassica oleracea that were known to contain S locus-specific glycoproteins (SLSG) discriminated between self and cross pollen in vitro in three different media. Discrimination was equally evident in experiments that were the in vitro equivalents of reciprocal pollinations. In a TAPS-buffered medium, self eluates depressed pollen germination in a dose-dependent manner. TAPS medium allowed a bioassay of the effects of SLSG in eluates because it optimized germination in a way that eliminated the complicating features of the stimulatory substances in the eluates. Stigma eluates affected percentage pollen germination and optimum calcium concentrations in vitro whether or not SLSG were present in the eluates, but differently in different media, and depending on whether the eluates were cross or self with respect to the pollen tested. Thus, the effect of stigma eluates on the in vitro germination of pollen in Brassica depends on the balance of stimulatory versus inhibitory substances in the eluates.  相似文献
