Vitamin analysis was carried out on five microalgae used in aquaculture:Tetraselmis suecica, Isochrysis galbana, Pavlova lutheri, Skeletonema costatum andChaetoceros calcitrans and one macroalga,Sargassum muticum, which is invasive on the Atlantic shores of France. Both liposoluble (provitamin A, E, K) and hydrosoluble (B1, B2, B6, B12, C, PP) vitamins were quantified. For most of them, greater amounts were obtained in the algal products than in the usual sources. On a dry weight basis,Tetraselmis suecica contained 4280 μg g?1 provitamin A and 6323 μg g?1 vitamin E,Pavlova lutheri 1162 μg g?1 vitamin B12 and 837 μg g?1 vitamin C,Isochrysis galbana 2690 μg g?1 vitamin PP and 183 μg g?1 vitamin B6, andSkeletonema costatum 710 μg g?1 vitamin B1.  相似文献

Culture conditions for the extracellular production of menaquinone (MK) by a mutant strain, K3-15, of Flavobacterium sp. 238-7 were investigated. A detergent-supplemented culture medium consisted of 60 g of glycerol, 23 g of peptone, 3 g of yeast extract, 7 g of K2HPO4, 5 g of NaCl, 0.8 g of MgSO4 · 7H2O, and 0.5 g of Rikanon UA 5012 in 1 l of tap water, pH 7.0, was constructed. Amounts of MK-4, MK-6, and total MK in 1 l of the medium were 101 mg, 39 mg, and 140 mg, respectively, after 7 d of cultivation at 28°C. Further studies with some additives showed that the addition of cedar wood oil increased the productivity of MK, especially MK-4. In the presence of 0.1% Rikanon UA 5012 and 0.1% cedar wood oil, mutant strain K3-15 produced 155 mg/l intracellularly and 105 mg/l extracellularly) of MK-4 and 27 mg/l (16 mg/l intracellularly and 11 mg/l extracellularly) of MK-6, and the total amount of MK reached 182 mg/l.  相似文献

This study was conducted to evaluate the importance of aeration in free and immobilized cell systems in an aerated bioreactor for xylitol production from an oat hull hemicellulosic hydrolysate using an integrated process. The aeration rate (AR) or oxygen mass transfer coefficient (kLa) demonstrated a significant role in controlling cell (Candida guilliermondii FTI 20037) regeneration and bioconversion performance in free and immobilized cell systems. In the free cell system, an aeration rate of 1.25 vvm corresponding to kLa of 15.8 1/h resulted in maximum values of product yield (Yp/s: 0.87 g/g), productivity (Qp: 0.57 g/l/h), and final xylitol concentration (Pf: 55 g/l) from the hydrolysate with a 74.5 g/l xylose concentration. However, in the aerated immobilized cell system, maximum and almost similar results (almost Pf: 54 g/l, Qp: 0.57 g/l/h and Yp/s: 0.84 g/g) were obtained with aeration rates from 1.25 to 1.5 vvm using composites based on polypropylene (PP) and partially delignified fiber (PDF). Composites based on acid treated fiber (ATF) containing a high amount of lignin showed some inhibitory impact on xylose uptake and xylitol formation (Pf: 47 g/l and Qp < 0.49 g/l/h) with the optimal aeration rate of 1.5 vvm in the initial cycle of the bioconversion; this inhibition impact could be resolved in the next consecutive cycles. The surface modifier polyethyleneimine (PEI) slightly enhanced cell retention in the immobilized form on the ATF-based cell support. This investigation helps fill in the knowledge gaps existing on the integrated processing of the lignocellulosic biomass for xylitol bioproduction and biorefinery industry; however, more scale-up studies are recommended for commercialization.  相似文献

Conversion of lactose into ethyl acetate by Kluyveromyces marxianus allows economic reuse of whey-borne sugar. The high volatility of ethyl acetate enables its process-integrated recovery by stripping. This stripping is governed by both the aeration rate and the partition coefficient, K EA,L/G. Cultivation at elevated temperatures should decrease the K EA,L/G value and thus favor stripping. K. marxianus DSM 5422 as a potent producer of ethyl acetate was cultivated aerobically in whey-borne media for studying temperature-dependent growth and ester formation. Shake flask cultivation proved thermal tolerance of this yeast growing from 7 to 47 °C with a maximum rate of 0.75 h?1 at 40 °C. The biomass yield was 0.41 g/g at moderate temperatures while low and high temperatures caused distinct drops. The observed μ-T and Y X/S-T dependencies were described by mathematical models. Further cultivations were done in an 1-L stirred reactor for exploring the effect of temperature on ester synthesis. Cultivation at 32 °C caused significant ester formation (Y EA/S?=?0.197 g/g) while cultivation at 42 °C suppressed ester synthesis (Y EA/S?=?0.002 g/g). The high temperature affected metal dissolution from the bioreactor delivering iron for yeast growth and preventing ester synthesis. Cultivation at 32 °C with a switch to 42 °C at the onset of ester synthesis allowed quick and efficient ester production (Y EA/S?=?0.289 g/g). The high temperature lowered the K EA,L/G value from 78 to 44 L/L which heightened the gas-phase ester concentration (favoring ester recovery) without increasing the liquid-phase concentration (avoiding product inhibition).  相似文献

Field-collected populations of mayflies, Ephemera orientalis were tested for susceptibility to 10 different insecticides using a direct-contact mortality bioassay. Ephemera orientalis subimagoes were susceptible to the insecticides chlorpyrifos, fenitrothion and chlorfenapyr with LD50 values of 69.7, 78.8 and 81.9 μg/♀, and adults had LD50 values of 71.9, 78.8 and 85.4 μg/♀, respectively. Susceptibility ratios (SRs) of subimagoes and adults of E. orientalis to the 10 insecticides were 1.0 to1.2 folds. The mayflies showed higher susceptibility to organophosphates than to pyrethroids. The SRs of Anopheles sinensis to E. orientalis were 514 to 1438 folds higher for organophosphates (LD50 values of 0.05 to 0.23 μg/♀) and 62 to 1155 folds higher for pyrethroids (LD50 values of 0.13 to 2.41 μg/♀). The SRs of Culex pipiens to E. orientalis were 606 to 3595 folds higher for organophosphates with LD50 values of 0.02–0.17 μg/♀ and 81 to 1365 folds higher for pyrethroids with LD50 values of 0.11–1.83 μg/♀. These results indicate that the use of ineffective insecticides will result in unsatisfactory control against field populations of the subimagoes and adults of E. orientalis.  相似文献

The fundamental characteristics of an automatic glucose analyzer which consists of sampling, sensor, and operation units were examined. The glucose sensor is a dual cathode type which has an immobilized glucose oxidase membrane coupled with an oxygen sensor. Using this glucose sensor combined with an automatic sampling device, on-off control of the glucose concentration in fed-batch cultures of Saccharomyces cerevisiae was carried out. When the glucose concentration to be controlled was set at 0.3 and 10 g/l, the concentration was well maintained within the range of 0.08−0.54 g/l in the former, and within 9.2–11.1 g/l in the latter. In the former experiment, 1.67 g/l of ethanol was produced at the end of cultivation (OD570=34). On the other hand, 12.9 g/l of ethanol was accumulated at the end of cultivation (OD570=43) in the latter experiment. Fed-batch cultures of Micrococcus ruteus were also carried out. The glucose concentration was set at 2.5 g/l. The microorganism grew up to OD610=264 and the glucose concentration was maintained within 2.0 and 3.1 g/l.  相似文献

The objective of this study was to quantify the genetic associations between a range of carcass-related traits including wholesale cut weights predicted from video image analysis (VIA) technology, and a range of pre-slaughter performance traits in commercial Irish cattle. Predicted carcass cut weights comprised of cut weights based on retail value: lower value cuts (LVC), medium value cuts (MVC), high value cuts (HVC) and very high value cuts (VHVC), as well as total meat, fat and bone weights. Four main sources of data were used in the genetic analyses: price data of live animals collected from livestock auctions, live-weight data and linear type collected from both commercial and pedigree farms as well as from livestock auctions and weanling quality recorded on-farm. Heritability of carcass cut weights ranged from 0.21 to 0.39. Genetic correlations between the cut traits and the other performance traits were estimated using a series of bivariate sire linear mixed models where carcass cut weights were phenotypically adjusted to a constant carcass weight. Strongest positive genetic correlations were obtained between predicted carcass cut weights and carcass value (min rg(MVC) = 0.35; max rg(VHVC) = 0.69), and animal price at both weaning (min rg(MVC) = 0.37; max rg(VHVC) = 0.66) and post weaning (min rg(MVC) = 0.50; max rg(VHVC) = 0.67). Moderate genetic correlations were obtained between carcass cut weights and calf price (min rg(HVC) = 0.34; max rg(LVC) = 0.45), weanling quality (min rg(MVC) = 0.12; max rg(VHVC) = 0.49), linear scores for muscularity at both weaning (hindquarter development: min rg(MVC) = −0.06; max rg(VHVC) = 0.46), post weaning (hindquarter development: min rg(MVC) = 0.23; max rg(VHVC) = 0.44). The genetic correlations between total meat weight were consistent with those observed with the predicted wholesale cut weights. Total fat and total bone weights were generally negatively correlated with carcass value, auction prices and weanling quality. Total bone weight was, however, positively correlated with skeletal scores at weaning and post weaning. These results indicate that some traits collected early in life are moderate-to-strongly correlated with carcass cut weights predicted from VIA technology. This information can be used to improve the accuracy of selection for carcass cut weights in national genetic evaluations.  相似文献

Bacillus cereus ATCC 14579 can respond to nutrient changes by adopting different forms of surface translocation. The B. cereus ATCC 14579 ΔplcR mutant, but not the wild type, formed dendritic (branched) patterns on EPS [a low-nutrient medium that contains 7.0 g K2HPO4, 3.0 g KH2PO4, 0.1 g MgSO4·7H2O, 0.1 g (NH4)2SO4, 0.01 g CaCl2, 0.001 g FeSO4, 0.1 g NaCl, 1.0 g glucose, and 125 mg yeast extract per liter] containing 0.7% agar. The dendritic patterns formed by sliding translocation of nonflagellated cells are enhanced under low-nutrient conditions and require sufficient production of a biosurfactant, which appears to be repressed by PlcR. The wild-type and complemented strains failed to slide on the surface of EPS agar because of the production of low levels of biosurfactant. Precoating EPS agar surfaces with surfactin (a biosurfactant produced by Bacillus subtilis) or biosurfactant purified from the ΔplcR mutant rescued the ability of the wild-type and complemented strains to slide. When grown on a nutrient-rich medium like Luria-Bertani agar, both the wild-type and ΔplcR mutant strains produced flagella. The wild type was hyperflagellated and elongated and exhibited swarming behavior, while the ΔplcR mutant was multiflagellated and the cells often formed long chains but did not swarm. Thin-layer chromatography and mass spectrometry analyses suggested that the biosurfactant purified from the ΔplcR mutant was a lipopeptide and had a mass of 1,278.1722 (m/z). This biosurfactant has hemolytic activity and inhibited the growth of several gram-positive bacteria.  相似文献

The electron paramagnetic resonance (epr) properties of cytochrome c oxidase have been examined with special attention to the effect of added ligands and of interactions between the redox components. The fully oxidized preparations have a very small g6 signal which increases greatly as the redox potential is made more negative, a process exactly paralleling the disappearance of the g3 signal. The potential for half appearance or disappearance (Em), respectively, is 380 mV at pH 7.0 and 300 mV at pH 8.5. This identifies the changes as accompanying reduction of cytochrome a3 because the Em of the “invisible copper” is 340 mV and pH independent. Nitric oxide (NO) binds reduced cytochrome a3 to form a paramagnetic species. This resulting epr signal is strongly dependent on the redox state of cytochrome a, another expression of heme-heme interaction in cytochrome oxidase. The NO compound is also unique in that under the appropriate conditions three of the four redox components (cytochrome a3, cytochrome a, and the “visible” copper) are epr active. In potentiometric titrations in the presence of azide the formation of the azide compound responsible for the g2.9 signal appears to require reduction of both cytochrome a3 and the “invisible copper.” An internal sulfur compound is present which, at alkaline pH values, can bind the heme responsible for the g6 signal and change it to a low-spin sulfur compound with a signal at approximately g2.6. Evidence is also presented for the cytochrome c oxidase in situ being an equilibrium mixture of two different conformational states.  相似文献

The nutrient removal potential of filamentous Oscillatoria sp. was quantitatively studied. Oscillatoria sp. showed satisfactory growth on secondary activated sludge effluent supplemented with 1% NaHCO3. Continuous open culture of Oscillatoria sp. was kept stable using a continuous stirred tank equipped with a filter-separator. Kinetic constants YN and YP were 10.6 g cells/g NO3-N and 37.7 g cells/g PO43− respectively through the analysis of the results of continuous culture experiments. Monoalgal continuous culture of more than 90% purity could be maintained for 6 months without contamination. The harvested Oscillatoria cells were proven to have an excellent filterability. They also have excellent autoflotation. The amino acid composition of the Oscillatoria algal protein compares favorably with the tentative standard for ideal protein defined by FAO/WHO.  相似文献

The effects of five alternative nitrogen sources, namely, malt sprout (MS), corn steep liquor (CSL), NH4Cl, NH4NO3 and diamine citrate (DC) were investigated on the l-(+)-lactic acid (LA) production by thermophile Lactobacillus plantarum As.1.3. Through the statistical analysis of the results by three steps of response surface methodology (RSM) design, MS and CSL were found to have significant effects on the LA production and their optimal concentrations in the medium should be 16.0 g/L and 12.0 g/L, respectively. The verification of the optimized medium showed that the maximum specific growth rate (μm) was 1.09 h−1, the cell yield coefficient (YX/S) and the l-(+)-lactic acid yield coefficient (YP/S) were 0.233 (OD620/g) and 0.98 (g/g), and the maximum volumetric productivity and the average volumetric productivity were 13.0 g/L h and 3.20 g/L h, respectively. The results indicate that the LA production can also be enhanced with the inexpensive nitrogen source alternatives.  相似文献

Rice yellow mottle virus (RYMV) is the most harmful virus that affects irrigated and lowland rice in Africa. The RBe24 isolate of the virus is the most pathogenic strain in Benin. A total of 79 genotypes including susceptible IR64 (Oryza sativa) and the resistant TOG5681 (O. glaberrima) as checks were screened for their reactions to RBe24 isolate of RYMV and the effects of silicon on the response of host plants to the virus investigated. The experiment was a three-factor factorial consisting of genotypes, inoculation level (inoculated vs. non-inoculated), and silicon dose (0, 5, and 10 g/plant) applied as CaSiO3 with two replications and carried out twice in the screen house. Significant differences were observed among the rice genotypes. Fifteen highly resistant and eight resistant genotypes were identified, and these were mainly O. glaberrima. Silicon application did not affect disease incidence and severity at 21 and 42 days after inoculation (DAI); it, however, significantly increased plant height of inoculated (3.6% for 5 g CaSiO3/plant and 6.3% for 10 g CaSiO3/plant) and non-inoculated (1.9% for 5 g CaSiO3/plant and 4.9% for 10 g CaSiO3/plant) plants at 42 DAI, with a reduction in the number of tillers (12.3% for both 5 and 10 g CaSiO3/plant) and leaves (26.8% for 5 g CaSiO3/plant and 28% for 10 g CaSiO3/plant) under both inoculation treatments. Our results confirm O. glaberrima germplasm as an important source of resistance to RYMV, and critical in developing a comprehensive strategy for the control of RYMV in West Africa.  相似文献

Background

Personality traits are associated with health outcomes including non-communicable diseases. This could be partly explained by lifestyle related factors including diet. The personality traits neuroticism, extraversion, openness, agreeableness, and conscientiousness are linked with resilience, meaning adaptability in challenging situations. Resilient people usually comply with favorable health behaviors.

Objective

Our objective was to explore the associations between food and nutrient intake, personality traits and resilience.

Design

A validated semi-quantitative food frequency questionnaire was used to measure diet and the NEO-personality inventory to assess personality in 1681 subjects. Linear regression analysis was used to explore diet-personality associations and cluster analysis to define resilient and non-resilient personality profiles.

Results

Adjusting for age, education and energy intake, and applying Bonferroni corrections, openness in men was associated with higher vegetable (14.9 g/d for 1 SD increase in the personality score, PBonf <0.01) and lower confectionery and chocolate (−2.8 g/d, PBonf <0.01) intakes. In women, neuroticism was associated with lower fish (−4.9 g/d, PBonf <0.001) and vegetable (−18.9 g/d, PBonf <0.01) and higher soft drink (19.9 g/d, PBonf <0.001) intakes. Extraversion, in women, associated with higher meat (5.9 g/d, PBonf <0.05) and vegetable (24.8 g/d, PBonf<0.001) intakes, openness with higher vegetable (23.4 g/d, PBonf <0.001) and fruit (29.5 g/d, PBonf <0.01) intakes. Agreeableness was associated with a lower soft drink (−16.2 g/d, PBonf <0.01) and conscientiousness with a higher fruit (32.9 g/d, PBonf<0.01) intake in women. Comparing resilient and non-resilient subjects, we found resilience in women to be associated with higher intakes of vegetables (52.0 g/d, P<0.001), fruits (58.3 g/d, P<0.01), fish (8.6 g/d, P<0.01) and dietary fiber (1.6 g/d, P<0.01).

Conclusion

Personality traits are associated with dietary intake and especially subjects with resilient personality profiles had healthier dietary intakes. These associations were stronger in women than in men.  相似文献

We wished to examine the effects of diabetes on muscle glutamine kinetics. Accordingly, female Wistar rats (200 g) were made diabetic by a single injection of streptozotocin (85 mg/kg) and studied 4 days later; control rats received saline. In diabetic rats, glutamine concentration of gastrocnemius muscle was 33% less than in control rats: 2.60 ± 0.06 μmol/g vs. 3.84 ± 0.13 μmol/g (P < 0.001). In gastrocnemius muscle, glutamine synthetase activity (Vmax) was unaltered by diabetes (approx. 235 nmol/min per g) but glutaminase Vmax increased from 146 ± 29 to 401 ± 94 nmol/min per g; substrate Km values of neither enzyme were affected by diabetes. Net glutamine efflux (AZ concentration difference × blood flow) from hindlimbs of diabetic rats in vivo was greater than control values (?30.0 ± 3.2 vs. ?1.9 ± 2.6 nmol/min per g (P < 0.001) and hindlimb NH3 uptake was concomitantly greater (about 27 nmol/min per g). The glutamine transport capacity (Vmax) of the Na-dependent System Nm in perfused hindlimb muscle was 29% lower in diabetic rats than in controls (820 ± 50 vs. 1160 ± 80 nmol/min per g (P < 0.01)), but transporter Km was the same in both groups (9.2 ± 0.5 nM). The difference between inward and net glutamine fluxes indicated that glutamine efflux in perfused hindlimbs was stimulated in diabetes at physiological perfusate glutamine (0.5 mM); ammonia (1 mM in perfusate) had little effect on net glutamine flux in control and diabetic muscles. In Intramuscular Na+ was 26% greater in diabetic (13.2 μmol/g) than control muscle, but muscle K+ (100 μmol/g) was similar. The accelerated rate of glutamine release from skeletal muscle and the lower muscle free glutamine concentration observed in diabetes may result from a combination of; (i), a diminished Na+ electrochemical gradient (i.e., the net driving force for glutamine accrual in muscle falls); (ii), a faster turnover of glutamine in muscle and (iii), an increased Vmax/Km for sarcolemmal glutamine efflux.  相似文献

Avermectins are macrocyclic lactones produced by Streptomyces avermitilis. Abamectin is a blend of B1a and B1b avermectins that is being used as a seed treatment to control plant-parasitic nematodes on cotton and some vegetable crops. No LD50 values, data on nematode recovery following brief exposure, or effects of sublethal concentrations on infectivity of the plant-parasitic nematodes Meloidogyne incognita or Rotylenchulus reniformis are available. Using an assay of nematode mobility, LD50 values of 1.56 μg/ml and 32.9 μg/ml were calculated based on 2 hr exposure for M. incognita and R. reniformis, respectively. There was no recovery of either nematode after exposure for 1 hr. Mortality of M. incognita continued to increase following a 1 hr exposure, whereas R. reniformis mortality remained unchanged at 24 hr after the nematodes were removed from the abamectin solution. Sublethal concentrations of 1.56 to 0.39 μg/ml for M. incognita and 32.9 to 8.2 μg/ml for R. reniformis reduced infectivity of each nematode on tomato roots. The toxicity of abamectin to these nematodes was comparable to that of aldicarb.  相似文献

Ascidians are marine animals with a great ability to synthesize bioactive substances. This study examined the cytotoxic potential of 10 ascidians found in the coastal waters of Northeast Brazil. Samples of the species Eudistoma vannamei Millar, 1977, Eudistoma sp., Didemnum ligulum Monniot F., 1983, Didemnum psammatodes (Sluiter, 1895), Didemnum sp., Polysyncraton sp., Trididemnum sp., Cystodytes dellechiajei (Della Valle, 1877), Euherdmania sp., and an unidentified species belonging to the Holozoidae family were extracted in methanol 5:1 (v/w). The extracts were tested for cytotoxicity using the brine shrimp lethality assay, sea urchin egg development assay, hemolysis assay, and MTT assay using tumor cell lines. The extract of E. vannamei showed the highest toxicity in brine shrimp (LD50=34.7 μg/ml) and in all tumor cell lines tested, with an IC50 of <2 μg/ml for CEM, 11.2 μg/ml for HL-60, 23.8 μg/ml for B16, and 14.3 μg/ml for HCT-8. In sea urchin eggs, it inhibited the cell cycle progression mainly at the blastula stage (IC50=74.8 μg/ml). The extract of Euherdmania sp. also exhibited some toxicity in these assays, but at a lower potency than that of E. vannamei. The extracts of D. psammatodes and Polysyncraton sp. showed a strong inhibition of the sea urchin egg cell cycle during both phases examined, first cleavage and blastula, with a possible action on the cell microfilament apparatus. The extract of D. ligulum showed selective toxicity toward HCT-8 cells (IC50=35.3 μg/ml). The extract from the Holozoidae was the only one that possessed a hemolytic effect, with an IC50 of 175.2 μg/ml. Further studies are necessary for a better characterization of the active principles of these extracts and a possible elucidation of the mechanisms of action.  相似文献

Ecological relationships between sulfate-reducing and methane-producing bacteria in mud of Lake Vechten have been studied by continuous culture studies using the chemostat technique. The maximum specific growth rate (μ max) and saturation constant (K s) were, respectively, 0.36 hr−1 and 0.047 mM for lactate-limited growth ofDesulfovibrio desulfuricans and 0,011 hr−1 and 0.17 mM for acetate-limited growth ofMethanobacterium sp. Calculated values for the true molar growth yieldsY G) and maintenance coefficients (m) were 30.6 g bacterial mass/mole of lactate and 0.53 g substrate/g dry wt hr forD. desulfuricans and 37.8 g bacterial mass/mole of acetate and 0.54 g substrate/g dry wt hr forMethanobacterium. No growth ofMethanobacterium was observed at apS2− value (the hydrogen sulfide potential) of more than 11 and there was no effect on the growth atpS2− values above 13. In mixed continuous culture experiments the concentration of acetate decreased in the secondstage growth vessel, whereas that of methane increased stoichiometrically. If the substrate concentration in the reservoirs (S r) was increased from 0.1 to 0.5 mg/ml, the population ofDesulfovibrio increased and that ofMethanobacterium was washed out of the culture vessel, since the concentration of hydrogen sulfide reached apS2− value of 10.5. From the mixed continuous culture experiments a commensalism between the two species can be described, i.e., the acetate-fermentingMethanobacterium benefits from the acetate released byDesulfovibrio which is, in turn, not affected in the presence of the former.  相似文献

The effect of dextran from Leuconostoc mesenteroides (DEX500), added to milk prior to acidification with glucono-δ-lactone (GDL) or Streptococcus thermophilus DSM20259, was studied with respect to polysaccharide concentration. The incorporation of 5–30 g/kg DEX500 significantly affected gelation behavior. Increasing DEX500 concentrations resulted in a linear increase of gel stiffness (GDL gels: R2 = 0.96; microbial acidification: R2 = 0.94; P < 0.05) and 30 g/kg DEX500 resulted in a 2-fold higher stiffness compared to gels without polysaccharide. The respective stirred gels depicted a significant reduction in syneresis, which decreased from 30.4% (0 g/kg DEX500) to 22.0% (30 g/kg DEX500) for chemically acidified gels after 1 d of storage. Physical characteristics of DEX500 in aqueous solution were helpful to explain its behavior in the complex system milk.  相似文献
