Potato is commonly affected by various pathogens of viral, bacterial, and fungal origin; therefore, simple and accurate diagnostic and identification techniques are of key importance both for the production of virus free planting material and for the monitoring of the phytosanitary state of planting areas. The newly developed test systems based on qualitative fluorescent amplification-based specific hybridization (FLASH-PCR) enable fast and accurate diagnostics of the major potato pathogens, i.e. potato viruses A, Y, X, M, and S, potato leaf roll virus, potato mop top virus, as well as potato spindle tuber viroid, while minimizing the risk of the working zone contamination.  相似文献

The present report describes fossil evidence of insect pathogens, heretofore, almost non-existent, from six samples of amber ranging in age from 15 to 100 million years. They include a cytoplasmic polyhedrosis virus and trypanosomatid infection in an adult biting midge (Diptera: Ceratopogonidae), and a nuclear polyhedrosis virus in an adult sand fly (Diptera: Phlebotomidae), both from Early Cretaceous Burmese amber, several types of fungal thalli on the cuticle of an adult mosquito (Culicidae: Diptera), as well as a fungal growth on the prothorax of a fungus gnat (Mycetophilidae: Diptera) in Dominican amber and large tumors in the body cavity of a caterpillar (Lepidoptera) in Mexican amber. These discoveries suggest that insect polyhedrosis viruses were present 100 million years ago and present the possibility that vertebrate arboviruses (especially those in the family Reoviridae) could have evolved from cytoplasmic polyhedrosis viruses infecting biting insects. The flagellates in the Early Cretaceous biting midge represent the first fossil record of monogenetic trypanosomatid infections of arthropods.  相似文献

Fungi are frequently found within insect galls. However, the origin of these fungi, whether they are acting as pathogens, saprophytes invading already dead galls, or fungal inquilines which invade the gall but kill the gall maker by indirect means, is rarely investigated. A pathogenic role for these fungi is usually inferred but never tested. I chose the following leaf-galling-insect/host-plant pairs (1) a cynipid which forms two-chambered galls on the veins of Oregon white oak, (2) a cynipid which forms single-chambered galls on California coast live oak, and (3) an aphid which forms galls on narrowleaf cottonwood leaves. All pairs were reported to have fungi associated with dead insects inside the gall. These fungi were cultured and identified. For the two cynipids, all fungi found inside the galls were also present in the leaves as fungal endophytes. The cottonwood leaves examined did not harbor fungal endophytes. For the cynipid on Oregon white oak, the fungal endophyte grows from the leaf into the gall and infects all gall tissue but does not directly kill the gall maker. The insect dies as a result of the gall tissue dying from fungal infection. Therefore, the fungus acts as an inquiline. Approximately 12.5% of these galls die as a result of invasion by the fungal endophyte.  相似文献

The fungal metabolite kojic acid, which is produced by Aspergillus and Penicillium species fungi that may be pathogens of both insects and plants, was a significant inhibitor of phenoloxidase of different representative beetle and caterpillar insect species. Fusaric acid and picolinic acid, produced by Fusarium spp., were also significant inhibitors of phenoloxidase, while dipicolinic acid and beauvericin were ineffective at concentrations tested. Previous reports of the ability of kojic and fusaric acid to inhibit defensive enzymes of plants suggest that these compounds may be important in allowing the producing fungi to be pathogens of both insects and plants.  相似文献

The Anticarsia gemmatalis nucleopolyhedrovirus (AgMNPV) is the most successful viral biopesticide in use worldwide. We have demonstrated that despite widespread apoptosis and no protein synthesis at 48 h p.i., UFL-AG-286 cells infected with a mutant of AgMNPV (vApAg), produced significant amounts of budded virus (BVs) and viral DNA late in infection. However, a different susceptible cell line (BTI-Tn5B1-4) showed no signs of apoptosis and produced 3.5 times more budded virus when infected with vApAg. A comparison of DNA from AgMNPV and vApAg digested with the same restriction enzymes showed differences in the restriction pattern, indicating that the vApAg phenotype might be due to a mutation in a gene or genes responsible for directly or indirectly inhibiting apoptosis in UFL-AG-286 cells.  相似文献

Modulation of inflammasome pathways by bacterial and viral pathogens   总被引:1,自引:0,他引:1  
Inflammasomes are emerging as key regulators of the host response against microbial pathogens. These cytosolic multiprotein complexes recruit and activate the cysteine protease caspase-1 when microbes invade sterile tissues or elicit cellular damage. Inflammasome-activated caspase-1 induces inflammation by cleaving the proinflammatory cytokines IL-1β and IL-18 into their biologically active forms and by releasing the alarmin HMGB1 into the extracellular milieu. Additionally, inflammasomes counter bacterial replication and clear infected immune cells through an inflammatory cell death program termed pyroptosis. As a countermeasure, bacterial and viral pathogens evolved virulence factors to antagonize inflammasome pathways. In this review, we discuss recent progress on how inflammasomes contribute to host defense against bacterial and viral pathogens, and we review how viruses and bacteria modulate inflammasome function to their benefit.  相似文献

The European Pharmacopoeia (Ph. Eur.) requires avian viral vaccines to be free of adventitious agents. Purity testing is an essential quality requirement of immunological veterinary medicinal products (IVMPs) and testing for extraneous agents includes monitoring for many different viruses. Conventional virus detection methods include serology or virus culture, however, molecular tests have become a valid alternative testing method.Nucleic acid testing (NAT) is fast, highly sensitive and has a higher degree of discrimination than conventional approaches. These advantages have led to the development and standardization of polymerase chain reaction (PCR) assays for the detection of avian leucosis virus, avian orthoreovirus, infectious bursal disease virus, infectious bronchitis virus, Newcastle disease virus, infectious laryngotracheitis virus, influenza A virus, Marek's disease virus, turkey rhinotracheitis virus, egg drop syndrome virus, chicken anaemia virus, avian adenovirus and avian encephalomyelitis virus. This paper reviews the development, standardization and assessment of PCR for extraneous agent testing in IVMPs with examples from an Official Medicines Control Laboratory (OMCL).  相似文献

The production of viral vectors or virus-like particles for gene therapy or vaccinations using the baculovirus expression system is gaining in popularity. Recently, reports of a viral vector based on adeno-associated virus (AAV) produced in insect cells using the baculovirus expression vector system have been published. This system requires the triple infection of cells with baculovirus vectors containing the AAV gene for replication proteins (BacRep), the AAV gene for structural proteins (BacCap), and the AAV vector genome (BacITR). A statistical approach was used to investigate the multiplicities of infection of the three baculoviruses and the results were extended to the production of AAVs containing various transgenes. Highest AAV yields were obtained when BacRep and BacCap, the baculovirus vectors containing genes that code for proteins necessary for the formation of the AAV vector, were added in equal amounts at high multiplicities of infection. These combinations also resulted in the closest ratios of infectious to total AAV particles produced. Overexpression of the AAV structural proteins led to the production of empty AAV capsids, which is believed to overload the cellular machinery, preventing proper encapsidation of the AAV vector transgene, and decreased the viability of the insect cells. Delaying the input of BacCap, to reduce the amount of capsids produced, resulted in lower infectious AAV titers then when all three baculoviruses were put into the system at the same time. The amount of BacITR added to the system can be less than the other two without loss of AAV yield.  相似文献

Insect pathogens play significant roles in the biocontrol of medical and agricultural pests. Cui et al. demonstrated that genetically modified (GM) fungi expressing host mosquito miRNAs could enhance the efficacy of the fungus by suppressing the host immune response. Sera from 210 pronghorns (Antilocapra americana) ranging in southeastern Alberta were tested for antibodies to disease agents present in indigenous cattle. No antibodies to Brucella abortus, Leptospira interrogans serovars pomona, hardjo, or grippotyphosa, or infectious bovine rhinotracheitis virus were found. Antibodies at prevalences of 43.8% and 49.2% were detected to bovine virus diarrhea (BVD) and parainfluenza type 3 (PI-3) viruses, respectively. The much higher prevalence of BVD virus antibodies in cattle than in pronghorns, and the occurrence of clinical bovine PI-3 infection in the study area, suggest that cattle may be a source of infection to the pronghorns.  相似文献
