Growth of dinoflagellates representing three orders, the Gymnodiniales,Peridiniales, and the Prorocentrales was examined following treatment withbarley straw extract. Selected dinoflagellate taxa showed growth responsessimilar to those reported for freshwater algae including: inhibition(Gyrodinium galatheanum, Gymnodiniumsanguineum, Heterocapsa triquetra andH. pygmaea); stimulation (Gyrodinium instriatum,Prorocentrum minimum and P. micans); and no effect(Gyrodinium estuariale, G. uncatenum,Ceratium furca, Peridinium sp.).Although barley straw extracts do not appear to have value as a universalmanagement tool for dinoflagellates, they may have potential in management ofspecific taxa and possibly taxonomic groups.  相似文献

Bioassays were conducted to determine the efficacy of barley straw liquor in controlling algal growth of 12 freshwater species of algae representing three divisions. Barley straw liquor inhibited the growth of three nuisance algae common in freshwater: Synura petersenii, Dinobyron sp., and Microcystis aeruginosa. However, Selenastrum capricornutum, Spirogyra sp., Oscillatoria lutea var. contorta, and Navicula sp. had significantly increased growth in the presence of straw liquor. The growth of the remainder, Ulothrix fimbriata, Scenedesmus quadricauda, Chlorella vulgaris, Anabaena flos-aquae, and Synedra sp. showed no significant difference from controls. In a related field study, we treated four of six ponds with barley straw and monitored their chlorophyll a levels for one growing season. While phytoplankton populations in all ponds decreased in midsummer, the phytoplankton biomass in treated ponds did not differ significantly from that of control ponds, suggesting that the application of barley straw had no effect on algal growth in these systems.  相似文献

The least ambiguous genetic markers are those based on completely characterized DNA sequence polymorphisms. Unfortunately, assaying allele states by allele sequencing is slow and cumbersome. The most desirable type of genetic marker would be unambiguous, inexpensive to assay and would be assayable singly or in parallel with hundreds of other markers (multiplexable). In this report we sequenced alleles at 54 barley (Hordeum vulgare ssp. vulgare) loci, 38 of which contained single-nucleotide polymorphisms (SNPs). Many of these 38 loci contained multiple polymorphisms, and a total of 112 polymorphisms were scored in five barley genotypes. The polymorphism data set was analyzed both by using the individual mutations as cladistic characters and by reducing data for each locus to haplotypes. We compared the informativeness of these two approaches by consensus tree construction and bootstrap analysis. Both approaches provided similar results. Since some of the loci sequenced contained insertion/deletion events and multiple point mutations, we thought that these multiple-mutated loci might represent old alleles that predated the divergence of barley from H. spontaneum. We evaluated sequences from a sample of H. spontaneum accessions from the Eastern Mediterranean, and observed similar alleles present in both cultivated barley and H. spontaneum, suggesting either multiple domestication events or multiple transfers of genes between barley and its wild ancestor.  相似文献

Isolated chloroplasts of barley seedlings ( Hordeum vulgare L.) when kept in light, released a soluble, thermostable factor that inhibited phenylalanine ammonia-lyase (PAL) in vitro . Highest inhibition was found when chloroplasts were suspended in PAL-containing extract and kept in light. Efficiency of PAL activity inhibition did not depend greatly on the type of medium used for chloroplast isolation, nor on the composition of buffer in which the enzymatic activity was measured. It is proposed that in green tissues chloroplasts may participate in regulation of cytoplasmic PAL activity.  相似文献

The algistatic properties of aqueous barley straw (Hordeum vulgare) extracts have been observed in laboratory studies and in situ. This reported algistatic property has been used by farmers and horticulturists to control algal blooms in various systems and has become standard practice in some areas. However, both inhibition and stimulation of algal growth in freshwater and marine species have been demonstrated. While the number of taxa known to be inhibited by barley straw has increased, comparatively little has been done to isolate and classify the compound(s) responsible for this algistatic effect. A microplate assay system using Microcystis aeruginosa was developed to isolate and identify the inhibitory components of barley straw extract. M. aeruginosa was selected for the bioassay because it is consistently inhibited by barley straw extract in studies conducted by our laboratory and others. The 24-well plate assay utilizes in vivo fluorescence monitoring with a TECAN GENios plate reader to determine chlorophyll-a levels in each culture. Fractionation and partial chemical characterization of inhibitory extracts suggests that the inhibitors are polyphenolics with molecular weights (MW) between 1,000 and 3,000 Da. Percolation of the aqueous extract through a Polyamide CC6 resin or through various MW cutoff filters resulted in the loss of algistatic activity, which confirms this assertion, while hydrolysis resulted in little change in the activity profile. Fractionation by HPLC methods yielded a highly potent multi-compound fraction, showing toxicity at 353 mg L−1 and algistatic activity between 11.1 and 3.53 mg L−1.  相似文献

The effects of conditions pre-dating germination on growth rate of impeded barley cv. Harrington roots were measured using an agar-capillary tube technique. Seedling root tips were directed into glass capillary tubes twothirds filled with agar at eight concentrations ranging from 1.6 to 9.6%, equivalent to penetrometer resistances of 25 to 1240 kPa. The rate of unrestricted root elongation (growth in air) of seed stored for 13 months (old seed), and of seed grown for a second generation without subsequent storage (new seed) was compared with growth in agar over a 24-hour interval. Root elongation rate of old and new seed was identical in the absence of resistance. At low to intermediate agar concentrations, elongation was significantly slower in roots from old, compared with new seed. At high agar concentrations root growth of old and new seed was the same. In both old and new seed, root growth through agar was greater in seed that germinated after 24, compared with 48 h. Differences in impeded root growth between old and new seed were lost in progeny of the test seed. Effects of UV-B radiation on the developing chloroplast of barley (Hordeum vulgare L.) seedling during greening were determined by Chl contents, Fo, Fv and fluorescence quenching coefficients. In greening of etiolated barley seedling, the value of Fo was greatly increased after the initiation of greening. However Fv and Fv/Fo were gradually increased. In greening with the additional irradiation of UV-B radiation, the value of Fo was strikingly decreased than that of the control after the initiation of greening, but Fv was gradually decreased from than that of the control during the greening period. These results suggest that the function of light-harvesting Chl are more sensitive than photosynthetic electron transport system by UV-B. Chl contents, Fv/Fo, qP and qNP, were decreased from than that of the control during the 72 h greening, especially, qR was strikingly decreased, but qE was slightly decreased by UV-B. These suggest that the sites of inhibition by UV-B are PSII and all sites of photosynthetic electron transport system. But PQ pool seems to be slightly inhibited by UV-B.  相似文献

The effects of competition from volunteer barley (Hordeum vulgare) on the growth and yield of oilseed rape (Brassica napus) were investigated in four experiments over three seasons. The growth of rape in the autumn was reduced by 50 - 91 % by competition from 400 barley plants m-2. A lower barley density of 200 plants m-2 had less effect but still reduced growth of rape by 65 - 81% in two of the experiments and 25 - 40% in the other two. During winter and spring the barley decreased in vigour and in the spring the rape started to recover, especially on the early drilled (23 - 30 August) plots. The rape sown in mid-September recovered less quickly. In Experiment 3, herbicides applied in November to control barley did not result in increased growth of rape in winter but led to greater recovery in spring. The barley died during the winter in Experiments 2 and 4, even in the absence of herbicides. Despite the marked effects of barley on the growth of rape in the autumn, yields on plots that had previously contained 200 barley plants m-2 were reduced by a maximum of only 16% in three of the experiments. In Experiment 3, where the barley was most competitive, this density and 400 plants m-2 lowered yields by 39% and 78%, respectively. Where a herbicide was used in November to control the barley these yield losses were reduced to 5%. In many rape crops the cost of herbicide treatment would be greater than the financial returns from the expected increase in yield resulting from the control of weeds. Possible reasons for the small loss in yield of rape from barley densities that had substantial effects on the growth of rape in the autumn are discussed.  相似文献

A cathepsin D-like aspartic proteinase (EC 3.4.23) is abundant in ungerminated barley ( Hordeum vulgare ) seed while a 30 kDa cysteine endoproteinase (EC 3.4.22) is one of the proteinases synthesized de novo in the germinating seed. In this work, the localization of these two acid proteinases was studied at both the tissue and subcellular levels by immunomicroscopy. The results confirm that they have completely different functions. The aspartic proteinase was present in the ungerminated seed and, during germination, it appeared in all the living tissues of the grain, including the shoot and root. Contrary to previous suggestions, it was not observed in the starchy endosperm. By immunoblotting, the high molecular mass form of the enzyme (32 + 16 kDa) was found in all the living tissues, whereas the low molecular mass form (29 + 11 kDa) was not present in the shoot or root, indicating that the two enzyme forms have different physiological roles. The aspartic proteinase was localized first in the scutellar protein bodies of germinating seed, and later in the vacuoles which are formed by fusion of the protein bodies. In contrast to the aspartic proteinase, the expression of the 30 kDa cysteine proteinase began during the first germination day, and it was secreted into the starchy endosperm; first from the scutellum and later from the aleurone layer. It was not found in either shoots or roots. The 30 kDa cysteine proteinase was detected in the Golgi apparatus and in the putative secretory vesicles of the scutellar epithelium. These results suggest that the aspartic proteinase functions only in the living tissues of the grain, as opposed to the 30 kDa cysteine proteinase which is apparently one of the proteases initiating the hydrolysis of storage proteins in the starchy endosperm.  相似文献

Aluminium/silicon interactions in barley (Hordeum vulgare L.) seedlings   总被引:3,自引:0,他引:3  
The response of seedlings of the monocot Hordeum vulgare L. cv. Bronze to 0,25 and 50 M aluminium in factorial combination with 0, 1.4, 2.0 and 2.8 mM Si was tested in hydroponic culture at pH 4.5. Nutrient solution (500 M calcium nitrate) and Al/Si treatments were designed to avoid the precipitation of Al from solution. Silicon treatments gave significant amelioration of the toxic effects of Al on root and shoot growth and restored calcium levels in roots and shoots at harvest to levels approaching those of control plants. Aluminium uptake by roots was also significantly diminished in the presence of Si. Silicon alone gave a slight stimulation of growth, insufficient to explain its ameliorative effect on Al toxicity. Spring barley ( Hordeum vulgare L. cv. Golf) was grown at different nitrate supply rates, controlled by using the relative addition rate technique, in order to elucidate the relationship between nitrate-N supply and root and shoot levels of abscisic acid (ABA). The plants were maintained as (1) standard cultures where nitrate was supplied at relative addition rates (RAs) of 0.03, 0.09 and 0.18 day−1, and (2) split-root cultures at RA 0.09 day−1 but with the nitrate distributed between the two root parts in ratios of 100:0, 80:20 and 60:40. Time-dependent changes in root and shoot concentrations of ABA (determined by radioimmunoassay using a monoclonal antibody) were observed in both standard and split-root cultures during 12 days of acclimation to the different nitrate regimes. However, the ABA responses were similar at all nitrate supply rates. Further experiments were performed with split-root cultures where the distribution of nitrate between the two root parts was reversed from 80:20 to 20:80 so that short-term effects to local perturbations of nitrate supply could be studied without altering whole-plant N absorption. Transient increases in ABA concentrations (maximum of 25 to 40% after 3 to 4 h) were observed in both subroot parts, as well as in xylem sap and shoot tissue. By pruning the root system it was demonstrated that the change in ABA had its origin in the subroot part receiving the increased nitrate supply (i.e. switched from 20 to 80% of the total nitrate supply). The data indicate that ABA responses are easily transmitted between different organs, including transmission from one set of seminal roots to another via the shoot. The data do not provide any indication that long-term nitrate supplies or general nitrogen status of barley plants affect, or are otherwise related to, the average tissue ABA concentrations of roots and shoots.  相似文献

Polymorphism at the Hor 1 locus of barley (Hordeum vulgare L.)   总被引:1,自引:0,他引:1  
The Hor 1 locus of barley encodes a group of seed storage polypeptides called C hordein. Two-dimensional electrophoretic analysis of C-hordein fractions from six cultivars with different alleles at the Hor 1 locus showed extensive polymorphism. A total of 34 major polypeptides was mapped, with between 4 and 18 present in each cultivar. There was less variation among the same cultivars in the numbers (6 to 10) of restriction fragments of genomic DNA which hybridized to a cDNA clone related to C hordein. The total number of restriction fragments was also lower (22), and most pairs of cultivars had more restriction fragments than polypeptides in common. A total number of about 20–30 C-hordein genes per haploid genome was estimated. The results indicate that cultivars differ mainly in the extent of gene and polypeptide divergence, rather than in the degree of gene reiteration. They are consistent with the proposed origin of the multiple structural genes at the Hor 1 locus by the duplication and divergence of a single ancestral gene.NACB was supported by a grant from the Home Grown Cereals Authority.  相似文献

Ambient temperature plays an important role in plant development. In cereals, little is known about the exact effects of ambient temperature in the range between it being a vernalising agent and an abiotic stress factor; thus the genetic determinants involved in the registering and response to ambient temperature, and their natural variation has not been dissected either. Principally, we wished to establish the level of natural variation in response to ambient temperature in barley via studying plant phenological development. The responses to temperature of 168 barley genotypes of different provenances and seasonal growth habit groups were observed in controlled environments. The effects of four temperature regimes (13°C, 16.5°C, 18°C and 23°C) on the duration of plant phenophases were examined. The plant development was characterised in a series of consecutive phenophases that span the plant life cycle from germination through flowering to attainment of maximum plant height. Ambient temperature affected significantly plant development, with substantial variation in responses among the genotypes. Six major types of responses were identified, which depended strongly on seasonal growth habit, with only a small degree of overlap. Although the differences in the timing of development among clusters were significant under each temperature regime, the 23°C treatment resulted in the largest diversity of responses, with significant changes in the ranking of the six clusters compared to other treatments. Two clusters showed particularly unusual responses to 23°C: the development of one winter barley cluster was extremely accelerated by the 23°C treatment, whereas the development of one spring barley cluster was significantly delayed. Ambient temperature assumes importance as a regulatory cue in the intricate and complex temporal and spatial regulation network of plant development in cereals and acts mostly through its regulatory effect on certain developmental phases such as the onset and duration of the intensive stem elongation.  相似文献

The in vitro plant regeneration frequencies for immature scutella, leaf-bases/apical meristems (LB/AM) and mature embryos of four commercially important barley genotypes were compared. Production of shoots from mature embryos or calluses of LB/AM incubated on media containing 1.0 or 2.0 mg l–1 6-benzylaminopurine (BA) were comparable to regeneration frequencies obtained for scutella-derived calluses of the same genotypes. Incubation of excised mature embryos and LB/AM on media containing the plant growth regulator, thidiazuron (TDZ), resulted in an increased shoot production. However, TDZ treatment did not stimulate plant regeneration from calluses derived from scutella or LB/AM. Shoots formed from TDZ-treated mature embryos and LB/AM were induced without a callus interphase and the in vitro culture system gave a three- to eight-fold higher regeneration frequency than recorded for scutella-derived calluses on BA medium. The simplicity and rapid development of shoots using the mature embryo system could potentially be used for the regeneration and genetic transformation of barley over alternative regeneration systems.  相似文献

The enzyme sucrose: sucrose 1-fructosyltransferase was partially purified from barley leaf growth zones. Four steps (ammonium sulphate precipitation and polyethylene glycol precipitation, followed by chromatography on Concanavalin A-sepharose and hydroxylapatite) yielded a 35-fold purification. The resulting preparation of 1-SST which still contained a number of different activities related to fructan metabolism, was subjected to preparative isoelectric focusing, and sections of the gel were analysed individually for 1-SST and related activities, using sucrose and 1-kestose as substrates. This procedure yielded a 196-fold purification and revealed the presence of two isozymes of 1-SST with pI values of 4.93 and 4.99, as determined by analytical isoelectric focusing of the corresponding fractions. Both isozymes produced glucose and 1-kestose when incubated with sucrose. In addition, small amounts of 6-kestose and tetrasaccharides were formed. In particular, one of the two 1-SST isozymes yielded fructose when incubated with 1-kestose, indicating that it also acts as a fructan exohydrolase. The other isozyme exhibited less fructan exohydrolase activity. Nystose was also degraded by the fructan exohydrolase activity but less than 1-kestose, whereas 6-kestose was not a substrate for the enzyme. Incubation of both 1-SSTs with different concentrations of sucrose showed that the enzyme was not saturated even at 500 mM. As for the barley sucrose: fructan 6-fructosyltransferase, both isozymes of 1-SST yielded two polypeptide bands of molecular weight 50 and 22 kDa upon sodium dodecylsulphate polyacrylamide gel electrophoresis, suggesting their close relationship to invertase (composed of two subunits of similar size), as previously reported for other plants.  相似文献

Boron (B) is an essential micronutrient in crop growth but its sufficiency range is narrow. Boron toxicity is a widespread problem in arid and semi-arid areas with cold weather. We investigated the effect of soil temperature (5, 10 and 15°C) on development of symptoms of B toxicity, plant growth and plant development, and on content and concentration of B in tissue of seedlings of four barley lines grown in soil with high level of available B (12 mg kg–1). Visual symptoms of toxicity were first observed in the high B soil concentration treatment at 5 °C at 12 days after emergence. Concentration of B in tissue decreased with increasing soil-temperatures. There was no effect of soil temperature on B content or B concentration in plant tissue at the final sample (17 days after emergence). High soil B reduced seedling and leaf emergence rates, although the final seedling emergence and number of leaves were unaffected. Barley lines differed in concentration of B in tissues and visual toxicity symptom development. Adaptation to high B was either through maintaining low tissue B concentration or through tolerance to high tissue B concentration. While the investigated range of temperature does influence B toxicity in barley seedlings, it remains to be determined whether it affects crop yield.  相似文献

Barley anthers from cold pretreated spikes produced no or few calluses when plated with both loculi in contact with the medium (flat). When anthers were plated with only one loculus in contact with the medium (up), a high proportion of the anthers produced calluses. The top loculus of the up anthers was most productive. Flat anthers, when compared with up anthers, were not only slower to produce multicellular pollen grains (MCPs) and microcalluses, but also produced fewer of them and ceased production earlier. The MCPs and microcalluses in flat anthers grew more slowly and few developed beyond the 30 cell stage. These results establish the importance of anther orientation for barley anther culture.  相似文献
