沙棘共生固氮根瘤及其内生弗兰克氏菌   总被引:1,自引:0,他引:1  
用透射电镜研究了中国沙棘(Hippophae rhamnoydes L.)根瘤的超微结构。它的侵梁细胞位于皮层中部,非侵染细胞与之间排列,富含多酚和淀粉粒。在根瘤的发育过程中,具有以下特征:(1)早期侵染细胞中具有核仁联合体;(2)由内生菌丝趋核生长而形成的核膜内陷处,有许多核孔出现;(3)在中期侵染细胞的内生菌丝和泡囊的荚膜附近,有成束的微管存在;(4)在4、5月瘤样的维管束细胞、非侵染细胞及早  相似文献

非侵染细胞是豆科根瘤中一种非常重要的细胞,它不仅参与了根瘤共生固氮,而且还在其中起了极为重要的作用,这些作用的发挥与其具有与此相适应的形态结构和分布有关。非侵染细胞体积小、数量少,分散于侵染细胞之间。在大豆根瘤中,它们彼此相连接,形成一条条的线和一个个的面,由根瘤中心向四周辐射,直至与皮层细胞相接,其作用可能与根瘤中心组织和皮层细胞之间的物质运输,特别是固氮产物的输出密切有关。非侵染细胞与侵染细胞之间的主要差异在于它不含细菌,中央有一个大液泡,细胞质中含有大量的过氧化物酶体和许多膨大的管状内质网,而且细胞壁上还有丰富的胞间连丝和纹孔,但主要存在于非侵染细胞与非侵染细胞之间的连接壁上。  相似文献

用透射电镜观察红豆草根瘤侵染细胞核在细胞凋亡过程中的超微结构,以探讨红豆草根瘤侵染细胞核在发育过程中的超微结构变化及其与细胞凋亡的关系.结果表明,红豆草根瘤侵染细胞核的超微结构随细胞发育程度不同而不同.在幼龄侵染细胞中,细胞核体积较大,近似圆形.在即将成熟和成熟的侵染细胞中,细胞核膜有内陷现象,其核仁常具有核仁泡和核仁联合体.在早期凋亡的侵染细胞中,细胞核体积减小,形状变得不规则,核膜出现大量内陷,在其表面形成许多大的突起和深的沟槽,有时还有内质网、线粒体、小液泡和细菌等位于核膜的内陷处,而且核仁也开始裂解.在后期凋亡的侵染细胞中,除细菌解体外,还出现核仁消失,核膜破裂,核质外流,并在细胞质中形成一些电子密度很高,无一定形状的团块状物质.  相似文献

在大豆根瘤中有一种非常特殊的细胞质内含物,只存在于侵染细胞中,一个细胞通常只有一个,一般位于细胞的外周部分,常常靠近胞间隙。这种内含物通常为圆形,其直径在1-2μm之间,主要由颗粒状物质,管状和泡状成分组成。它经甲苯胺蓝O染色后呈深蓝色,苏丹黑B染色后呈深黑色,考马斯蓝染色后呈蓝色,因此它不是类聚核糖核蛋白体,也不是一般的蛋白体和脂滴或脂质体,而可能是一种蛋白质和脂的复合物。其作用可能与共生固氮中  相似文献

用透射电镜对红豆草根瘤侵入线的超微结构进行了观察研究.结果表明,(1)红豆草根瘤侵入线由胞间隙和胞间层细胞壁内陷形成,它们的体积较小,多为管状,基质丰富,含菌很少,常有分叉和1个以上的基质区,而且不同基质区的电子密度、细菌数量和侵入线壁厚度都不相同.(2)红豆草根瘤的侵入线十分丰富,它们不仅大量存在于根瘤分生细胞和幼龄侵染细胞中,也经常出现在发育成熟的侵染细胞内.(3)红豆草根瘤中有一种近似圆形的特殊结构,表面由一层膜包围,其内电子密度较低且无固定结构,且只位于侵染细胞的细胞质中,常在侵入线附近,从不出现在侵染细胞的液泡内和非侵染细胞里面.  相似文献

 我们用超声法制备了内部包裹阿霉素,表面有抗人胃癌细胞M85的单克隆抗体3Hll或非相关抗体IgG的阿霉素靶向脂质体和阿霉素非靶向脂质体,研究了这些脂质体和游离阿霉素抑制靶细胞M85和非靶细胞HeLa细胞生长的能力。结果表明,一方面,与游离阿霉素或阿霉素非靶向脂质体相比,阿霉素靶向脂质体抑制M85细胞生长的能力提高了2—4倍,而在抑制非靶细胞HeLa细胞的生长方面,阿霉素靶向脂质体的效力还不如游离阿霉素或阿霉素非靶向脂质体。过量的游离单克隆抗体3Hll可以有效地抑制阿霉素靶向脂质体对M85细胞的细胞毒作用,而另一种单克隆抗体3G9则不能。空靶向和空非靶向脂质体对M85细胞生长均无影响。因此,靶向脂质体可以把包裹的抗癌药物专一地送给胃癌细胞,井杀伤它们。  相似文献

箭舌豌豆根瘤幼龄侵染细胞的壁和质膜比较光滑,成熟侵染细胞与此不同,不仅细胞壁厚薄均,有较多的胞间连丝,而且质膜常常内陷形成各种突起,然后离质膜形成小泡。这些位于质膜附近的小泡体积较小,多呈圆形,既可单独存在,也可多个聚在一起。在向细胞中央移动中,有的小泡靠近细胞质膜,甚至与细菌周期融合,有的小泡不民附近的小液泡融合变为较大液泡,并常用降解程度不同的细菌位于其中,在衰老侵染细胞中,细胞壁附近有小泡,  相似文献

用扫描电镜观察了豌豆根瘤的侵染细胞.结果表明,在这些细胞中有大量的细菌,它们主要是杆状细菌,其次是球形、Y形和T形细菌,其它形状的细菌很少.除了细菌形状不同外,还有一些细菌比较特殊,如有的细菌较长,菌体出现部分收缩并形成一个或一个以上的收缩环,其形状类似一条莲根;有的细菌很大,它的体积是普通细菌的2倍或2倍以上;有的细菌粗细不均匀,端部膨大,呈棒槌状.侵染细胞中有许多小泡,它们大小不同,呈球形.它们存于细菌之间,其中一些小泡还位于细菌的表面上,而且附近细菌的表面有时还有各种隆起.  相似文献

在中国丰收11号大豆根瘤侵染细胞中,我们发现了一种电子密度很高,体积很大,形状为圆形或近似圆形,外面没有界膜,常位于胞间隙附近的特殊的细胞质内含物。高尔基体及其小泡,丰富的粗糙型内质网和核糖体常在它的附近,其中一些核糖体正沉积在它的表面。它主要是由核糖体凝聚而成,高尔基体和内质网在它的形成中也起了一定作用。它的内部含有颗粒状,纤维状,泡状和管状物质。它的出现似乎与侵染细胞固氮有关。  相似文献

Intracellular hyphae and vesicles in mycorrhizal roots of yellow poplar were examined by electron microscopy. An investing layer of host wall material and cytoplasm enclosed the endophyte within the cells. Young developing hyphae contained abundant cytoplasm and few vacuoles. As hyphae matured, they became highly vacuolated and accumulated carbohydrate (glycogen) and lipid reserves. Mature vesicles were engorged with lipid droplets, possessed a trilaminate wall and were also enclosed by host wall material and cytoplasm. Compared with uninfected cells, infected cortical cells showed an increase in cytoplasmic volume, enlarged nuclei, and a reduction of starch reserves. Host nuclei were always proximal to the hyphae during hyphal development and deterioration. While other cytoplasmic components of infected and uninfected cells were comparable large electron-dense bodies occurred in vacuoles of most cells containing hyphae. Deterioration of intracellular hyphae occurred throughout the samples examined. Septa separated functional and degenerating portions of the hyphae. Hyphal deterioration involved degeneration and ultimate disappearance of fungal cytoplasm as well as collapse of hyphal walls. Based on these observations, the authors hypothesize that deterioration of the endophyte may release significant quantities of mineral nutrients, via hyphal contents, which are absorbed by the host.  相似文献

Summary Young rats were treated with high doses of parathyroid hormone (PTH). Osteoclasts from these animals revealed characteristic alterations in comparison to control cells: a) The cytoplasm contained large vacuoles with phagocytosed cells, some of which resembled osteoblasts or osteocytes. The vacuoles were interpreted as lysosomes because the engulfed cells often appeared partly digested and the vacuoles contained acid phosphatase as demonstrated histochemically, b) lipid droplets were present in the cytoplasm and usually located close to the endoplasmic reticulum and/or in regions with many free ribosomes, c) the Golgi complex was more frequently separated from the nuclei than in control cells, d) small coated cytoplasmic bodies were numerous in the peripheral cytoplasm, e) the membranes of the endoplasmic reticulum were fused in some places, f) cytoplasmic regions with numerous free ribosomes were frequent, g) large ring-shaped granules occurred in some mitochondria. Energy dispersive X-ray analysis of these granules provided evidence that they contained calcium and probably phosphorus, h) in some osteoclasts the mitochondria were enlarged. — The findings are consistent with an increased activity of osteoclasts and in particular a stimulation of the lysosomal system in these cells.This research was supported by grants no. 512-819, 512-1545 and 512-1912 from the Danish Research Council. 电镜观察结果表明,幼龄箭舌豌豆根瘤侵染细胞的细胞质较少,中央是一些体积较大的液泡。细胞质中侵入线经常可见,由侵入线释放出来的细菌均有细菌周膜。这些细菌只位于细胞质中,不出现在液泡里面。成熟根瘤中的侵染细胞与此不同,它们中有大量的成熟侵染细胞,细胞质丰富,里面充满大量细菌,中央常有一个大液泡。当中央液泡发育到一定程度时,位于其附近的细菌可通过液泡膜内吞、液泡膜与细菌周膜融合及液泡膜破裂3种途径进入液泡,后一种途径常伴有寄主细胞质。液泡中的细菌绝大部分裸露在外,只有个别细菌具有细菌周膜且多位于液泡膜的破损处附近,因此细菌周膜可能是原来就有的。  相似文献

Icosahedral viral particles were found in the cytoplasm of erythrocytes and splenic reticular cells of a marine toad (Bufo marinus) collected from Costa Rica. Capsids had a maximum diameter of 312 nm and a spherical core with biphasic electron density. Viruses in erythrocytes were associated with cytoplasmic assembly areas and vacuoles in cytoplasm. Nuclei had finely granular material of decreased electron density located centrally, but contained no viral particles. A group of unenveloped viral particles was seen extracellularly in a splenic vessel. The virus was consistent with an iridovirus. In a blood smear stained with Giemsa round basophilic bodies with average diameters of 1.70 microns and morphologically similar to Pirhemocyton sp. were seen in the cytoplasm of erythrocytes and occasionally in the cytoplasm of monocytes or extracellularly. Erythrocytes containing these bodies had vacuoles and irregular pale-staining areas in the cytoplasm and pale-staining areas in the nucleus. These changes corresponded to the viral particles, assembly areas, vacuoles and nuclear changes at the ultrastructural level.  相似文献

用常规电镜方法观察了锦橙[Citrussinensis (L.) Osb.]汁囊从原始细胞到发育为一个具柄的成熟汁囊的过程中,汁囊构成细胞超微结构的变化。锦橙汁囊原始细胞及发育为球状体时的构成细胞以及柱状结构顶端的细胞都是一种典型的分生组织细胞。在细胞质中有包括线粒体、质体、内质网、核糖体等丰富的细胞器,但没有观察到高尔基体。这些分生细胞分裂一段时期后就停止活动,逐渐分化为适应贮藏功能的液泡化薄壁细胞。分生细胞开始分化时,在细胞中出现许多小液泡和高尔基体。这些小液泡逐渐地融合,同时细胞质变少,最后形成一个有中央大液泡的薄壁细胞,在紧贴细胞膜的薄薄的一层细胞质中有线粒体、质体、高尔基体以及含有许多脂滴的杂色体。但成熟果实中汁囊的薄壁细胞中几乎没有任何细胞器。  相似文献

Ultrastructure of Jincheng juice sac of Citrus sinensis (L.) Osb. was continuously investigated from the initial cell to the stalk-bearing sac. The initial cell and cells formed globularstructure, as well as the uper cells of the column-structure were typical meristem cells with mitochondria, plastids, rough endoplasmic reticulum, rich ribosome without Golgi body in their dense cytoplasm. These meristem cells would differentiate into parenchyma ceils pro2 viding storage function. At the beginning of differentiation of the meristem cells, the number of small vacuoles increased and some Golgi bodies appeared. Small vacuoles gradually fused into a central vacuole. During the fusion of small vacuoles, the cytoplasm became thinned, but still contained mitochondria, plastids, Golgi bodies, end0plasmic reticulum and some chromplasts with lipid drops. Almost no organelle were ever observed in the parenchyma cells of juice sac from mature fruit.  相似文献

Scarified seeds of Medicago sativa L. ''Ranger'' and ''Lahontan'' alfalfa were allowed to imbibe water for 36 hr and then were inoculated with stem nematodes, Ditylenchus dipsaci Kühn. Seedlings were grown in sterilized Provo sand at 20 C and hypocotyl sections harvested at 1, 3 and 7 days. Evidence from electron micrographs indicated that cells of noninfected control plants contained normally developing chloroplasts bearing stroma, thylakoids, starch grains and plastoglobuli. The cytoplasm contained a nucleus, endoplasmic reticulum, vacuoles, mitochondria, ribosomes and dictyosomes. No morphological symptoms of nematode infection were observed in infected plants of either Ranger of Lahontan alfalfa 1 day after inoculation. Electron micrographs of tissue from the infected plants, however, indicated more osmiophilic bodies (lipid bodies) per cell than did the noninfected control, with more lipid bodies present in Ranger than in Lahontan. Three and 7 days after planting, swollen hypocotyls could be seen; the degree of swelling was greater in Ranger than in Lahontan. Electron micrographs of infected tissues indicated that both cultivars were undergoing the same kind of damage. Injured organelles were endoplasmic reticulum, chloroplasts and the nucleus. Histochemical staining indicated no changes in the middle lamellae.  相似文献

Colletotrichum graminicola, a pathogen of sorghum and corn, was investigated prior and during germination as to certain aspects of acid phosphatase activity and lipid mobilization. Ungerminated conidia cytoplasm was filled with lipid deposits, which were mobilized during the germination process. Cytochemical ultrastructural examination showed that conidia vacuoles exhibit acid phosphatase activity, which is suggestive of lytic activity. Lipid bodies, stored in the ungerminated conidia cytoplasm, were internalized by vacuoles in a process analogous to microautophagy and were apparently digested inside them. The lipid bodies disappeared and vacuoles became enlarged in conidial cells during germination. Appressoria also showed acid phosphatase activity in multiple heterogeneous vesicles which were, in most cases, juxtaposed with lipid bodies. These results suggest that the vacuolar system plays an important role during C. graminicola germination and that the initial stages of lipid metabolization are taking place inside the vacuoles.  相似文献
