The somatic recombinogenic activity of the phenylpyrrole fungicide fludioxonil, in diploid Aspergillus nidulans was found similar to that caused by aromatic hydrocarbon and dicarboximide fungicides (AHDFs), such as iprodione, chlozolinate and tolclofos–methyl. All these fungicides not only increased the number of mitotic recombinants but also provided similar appearance, small sectors, of white and yellow mitotic recombination products. Fludioxonil highly resistant strains (resistant factor approximately 5000) of Botrytis cinerea were isolated at high frequency (1.08 × 10−5). Study of cross-resistance patterns of mutant strains to other fungicides, revealed cross-resistance of fludioxonil with dicarboximides (iprodione, procymidone, and chlozolinate) and aromatic hydrocarbons, such as tolclofos–methyl, pentachloronitrobenzene (PCNB), tecnazene and chloroneb. The positive cross-resistance relationships found between phenylpyrroles and members of the AHDFs and their ability to increase mitotic instability in diploid A. nidulans , indicate that phenylpyrroles should be included with AHDFs. A study of fitness parameters in wild-type and representative fludioxonil-resistant mutants of B. cinerea , showed that the mutation(s) leading to fludioxonil resistance may or may not affect some fitness-determining characteristics, such as sensitivity to high osmolarity, growth rate, conidial germination and germ-tube elongation. Pathogenicity tests on cucumber seedlings showed that an osmosensitive representative strain of B. cinerea , resistant to fludioxonil, was as virulent as the wild-type strain. The phenylpyrrole fungicide was ineffective, even in high concentrations, to control grey mould caused by this isolate.  相似文献

Dicarboximide fungicides have been used for the control of grey mould in protected crops in Crete since 1977. During the 1980 growing season a decline of their efficacy was observed. In successive surveys carried out in May 1980, February 1981 and May 1981 in 28, 10 and 13 plastic houses repectively, a considerable proportion of resistant strains was found. From each of the plastic houses sampled mostly either only resistant or only sensitive strains were isolated. In three of the plastic houses with resistant strains there was an acute disease control problem. The ED50 of 15 resistant strains studied was in the area of 3·5 μg/ml vinclozolin as compared with 0·2 μg/ml for the wild type strains. The vinclozolin-resistant strains were also resistant to procymidone, iprodione, and dicloran. In most of the cases strains resistant to vinclozolin were also resistant to benomyl and strains sensitive to vinclozolin were also sensitive to benomyl. In the absence of fungicide, resistant strains grew more slowly on PDA than sensitive ones, but spores germinated equally well. Vinclozolin (0·75 mg a.i./ml) did not protect eggplant seedlings against resistant strains but gave satisfactory control of sensitive ones.  相似文献

A wild-type isolate of Epicoccum purpurascens was exposed to shortwave ultraviolet light. One of the resulting cultures (M-20-A) was grown on media amended with the fungicides iprodione or vinclozolin and fungicide-tolerant strains were obtained. Several comparisons were made between new strains and the wild type. Sporulation was improved compared with the wild type. Strains varied in their tolerance to iprodione and vinclozolin but were not tolerant to the fungicide benomyl. Strains R4000, 16-B, and 7-A inhibited Sclerotinia sclerotiorum in vitro more than either the wild type or M-20-A, and exhibited improved control of white mold of bean in the greenhouse compared with M-20-A. Cultures of B. squamosa when grown in concentrations from 4–2500 μg/ml of iprodione in agar, produced iprodione-insensitive isolates distributed at random over the concentration range. Thirty of the 37 isolates when subsequently transferred to fungicide-free agar were slower growing than the parent cultures. Similar results were obtained with single spore and multispore isolates. Iprodione insensitivity occurred in cultures derived from benomyl tolerant and sensitive field isolates. Insensitive isolates grown on agar, containing 500 μg/ml iprodione, did not revert to sensitivity after several cultural cycles on fungicide-free agar. Spores of iprodione insensitive isolates generally proved less infective than spores from sensitives isolates when sprayed onto untreated onion leaves. In general, protectant sprays of iprodione did not enhance the infectivity of insensitive isolates. Inoculation of glasshouse and field raised plants indicated that although selected aggressive iprodione-insensitive isolates, from treatments with low and high concentrations of the fungicide, readily infected onion leaves, they were less capable of sporulation than the sensitive parent forms of the fungus.
Iprodione-insensitive isolates were cross-tolerant to the chloronitrobenzene fungicides quintozene and tecnazene and to the dichloroaniline fungicide dicloran.  相似文献

Dicarboximide fungicides are widely used to control various fungal species. Their primary action is not known, due to a lack of knowledge concerning the mechanism of action of the dicarboximide group. The cytotoxicities of vinclozolin and iprodione in rat hepatoma-derived Fa32 cells were investigated. Cytotoxicity was measured by neutral red uptake inhibition after treatment for 24 hours. Iprodione was more toxic than vinclozolin. Vinclozolin was less toxic in glutathione-depleted cells than in control cells. This was also true for iprodione at lower concentrations, but iprodione became more toxic at higher concentrations. Both the fungicides increased the endogenous glutathione content by 20% after 1 hour. After 24 hours, the glutathione content was doubled by vinclozolin, but was not affected by iprodione. No effect on glutathione S-transferase activity or reactive oxygen species formation could be observed. Cytochrome P450-dependent ethoxyresorufin-O-deethylase and pentoxyresorufin-O-depentylase activities were moderately activated by iprodione and strongly activated by vinclozolin. A glutathione-related cytochrome P450-dependent metabolic attack of vinclozolin and iprodione could be responsible for their cytotoxicity in Fa32 cells. Further research is needed to fully elucidate these (or other) mechanisms.  相似文献

Smoulder, caused by Botrytis narcissicola, is the most widespread foliar disease of narcissus (Narcissus cultivars) in the UK and causes significant losses in bulb and flower yield. A range of current and novel fungicide treatments was examined to determine if control of the disease could be improved and to measure the effect of treatments on bulb yield. In assays on pot-grown plants, carbendazim+flusilazole, epoxiconazole, iprodione + thiophanate-methyl, tebuconazole and vinclozolin reduced lesion size by more than 90%, compared with untreated plants, when applied 1 day before inoculation with the fungus; vinclozolin and tebuconazole were also effective when applied 2 days after inoculation. In field experiments in Cambridgeshire and Lincolnshire, significant reductions were observed in secondary smoulder symptoms (leaf lesions and stem rot) following fungicide sprays. Spray programmes of the novel fungicides azoxystrobin, cyprodinil, kresoxim-methyl, tebuconazole and pyrimethanil were as effective, or more so, than the standard treatments (chlorothalonil, vinclozolin) used by growers. Mixtures of carbendazim + tebuconazole and azoxystrobin + tebuconazole also gave effective control of smoulder. Programmes of four to six sprays, using two or three fungicides with different modes of action, applied alternately, reduced smoulder by 35–69% and increased bulb yields by 7–59%. Although treatment around and after flowering resulted in the greatest control of secondary smoulder, treatment before flowering was required for the highest bulb yields. The choice of fungicides for use in spray programmes to provide effective and reliable control of smoulder, and the possibility of control using fewer sprays, are discussed.  相似文献

Fludioxonil is a new non-systemic phenylpyrrole fungicide. It is a derivative of the antibiotic pyrrolnitrin and is highly active against Ascomycetes. Basidiomycetes and Deuteromycetes. In the present study we determined the baseline sensitivity of B. fuckeliana for fludioxonil in comparison to the dicarboximide vinclozolin. Subsequently these baselines were compared with the sensitivity of dicarboximide-resistant field isolates and with resistant laboratory strains selected on media amendedwith vinclozotin or fludioxonil, respectively. Additionally, sensitivities of genetically homogeneous ascospore progenies of sexual crosses of sensitive isolates with dicarboximide-resistant field isolates or resistant laboratory strains were compared to, the baselines. Baseline ECst, values for the inhibition of germination ranged from 0.07 to 0.20 μg ml-1 of vinclozolin and for mycelial growth from 0.003 to 0.016 μg ml’ of fludioxonil and from 1.7 to 2.0 μg ml-1 of vinclozolin and for mycelial growth from 0.003 to 0.016 μg ml-1 of fludioxonil and from 0.13 to 0.27 μg ml-1 of vinclozolin. Sensitivity to fludioxonil of all dicarboximide-resistant field isolates did not differ from their baseline. However, resistant laboratory strains that were selected either on media amended with fludioxonil or vinclozolin showed decreased sensitivities to both active ingredients. All sexual progenies of the crosses described above were of the parental phenotypes. We found no independent segregation of the resistance markers.  相似文献

Abstract Isolates of R. leguminosarum bv. viciae from pea and lentil nodules taken at one field site in France were tested in the laboratory for their ability to donate and receive plasmids by conjugation. Five isolates of 20 tested as donors were found to be capable of donating a plasmid which restored the ability to nodulate V. sativa to an isolate which had spontaneously lost this ability. Of 16 isolates tested as recipients all were found to be competent to receive one or more Tn5-labelled test plasmids at a frequency that varied widely (10−9− 10−3 per recipient) dependent upon both the recipient and the plasmid transferred. Three distinct plasmids carrying genes essential for symbiotic functions (pSym) were consistently shown to be transferred at a lower frequency than a cryptic plasmid. Collectively, these results indicate a significant potential for plasmid transfer within the natural soil population. During this work, several independent derivatives were obtained which contained two bv. viciae pSym. These plasmids usually appeared to be compatible together in cells ex planta, but the one acquired in matings was apparently frequently lost (10−2 per cell) in nodules of V. sativa . Hybrid derivatives containing bv. viciae and bv. phaseoli pSym, apparently retained both plasmids in nodules when P. vulgaris was the host plant but lost the bv. phaseoli pSym at high frequency (4 × 10−1 per cell) in nodules of V. sativa . Structural rearrangements among the plasmids of these transconjugants were also detected in cells recovered from nodules.  相似文献

An outbreak of grey leaf spot caused by Stemphylium solani was observed on tomato in Shandong Province of China in recent years and brought huge economical losses. Fludioxonil is a phenylpyrrole fungicide with strong antifungal activity against S. solani. To evaluate the risk of S. solani developing fludioxonil resistance, a total of 145 field isolates were examined for sensitivity to fludioxonil by measuring mycelial growth. The baseline sensitivity was distributed as a unimodal curve with a mean EC50 value of 0.0659 (±0.0170) µg mL?1. Five mutants with high resistance to fludioxonil (RF > 1000) were obtained by successively selecting on fludioxonil‐amended plates in the laboratory. All the resistant mutants associated with strongly reduced fitness in mycelial growth, sporulation and pathogenicity. Fludioxonil had positive cross‐resistance with procymidone and iprodione, but there was no cross‐resistance with other fungicides including boscalid, fluazinam, azoxystrobin and flusilazole. Based on the current results, resistance risk of S. solani to fludioxonil could be moderate. This is the first report of baseline sensitivity of S. solani to fludioxonil and its risk assessment. In order to delay the resistance development, it is recommended that fludioxonil can be used as one component of the mixture or fungicides with different modes of action should be alternatively used for this disease management.  相似文献

A method based on solid-phase extraction (SPE) and high-performance liquid chromatography (HPLC) was developed for the simultaneous determination of 3-(3,5-diclorophenyl)-5-ethenyl-5-methyl-2,4-oxazolidinedione (vinclozolin) and 3-(3,5-diclorophenyl)-N-(1-methylethyl)-2,4-dioxo-1-imidazolidinecarboxamide (iprodione) in human urine. Urine samples containing vinclozolin and iprodione were collected by solid phase extraction using C(18) cartridges. The chromatographic separation was achieved on a Spherisorb ODS2 (250 mm x 4.6 mm, 5 microm) column with an isocratic mobile phase of acetonitrile-water (60:40, v/v). Detection was UV absorbance at 220 nm. The calibration graphs were linear from 30 to 1000 ng/mL for the two fungicides. Intra- and inter-day R.S.D. did not exceed 2.9%. The quantitation limit was 50 ng/mL for vinclozolin and 30 ng/mL for iprodione, respectively.  相似文献

Abstract Bacillus anthracis can be identified on the basis of the detection of virulence factor genes located on two plasmids, pXO1 and pXO2. Thus isolates lacking both pXO1 and pXO2 are indistinguishable from closely related B. cereus group bacteria. We developed a multiplex PCR assay for characterization of B. anthracis isolates, and simultaneous confirmation of the species identity independent of plasmid content. The assay amplifies lef, cya, pag (pXO1) and cap (pXO2) genes, and a B. anthracis specific chromosomal marker, giving an easy-to-read profile. This system unambiguously identified virulent (pXO1+/2+) and avirulent (pXO1+/2, pXO1/2+ and pXO1/2) strains of B. anthracis and distinguished 'anthrax-like' strains from other B. cereus group bacteria.  相似文献

The virucidal properties of iodophor, chlorine (sodium hypochlorite), formalin, thimerosal (organic mercurial compound), malachite green, and acriflavine were tested on infectious pancreatic necrosis virus (IPNV). Iodine and chlorine showed good activity, but efficacy depended on the concentration of virus, the presence of organic matter (calf serum), and water pH. Water hardness (0-300 mg 1−1 as CaCO3) did not affect virucidal activity. In a 5 min exposure, 4 mg 1−1 available iodine inactivated 103.9 TCID50 m1−1 IPNV but 16 mg 1−1 iodine were needed for inactivation of 106.3TCID50m1−1. The addition of 0-5% calf serum significantly reduced the iodine concentration and the virucidal activity. In comparison, 4 mg 1−1 chlorine were needed to inactivate 1046 TCID50 m1−1 IPNV in 5 min. However, the addition of 0-07 % serum greatly reduced the chlorine concentration and extended the virucidal contact time to 30 min or more. IPNV at 106.3 TCID60 m1−1 was not inactivated by exposures for 60 min to 0-2% formalin, 10 min to 0-2% thimerosal, 60 min to 5 mg 1−1 malachite green, or 20 min to 500 mg 1−1 acriflavine. However, acriflavine at 0-5 mg 1−1 in cell culture media prevented the development of cytopathology caused by IPNV and may be useful in the treatment of the disease.  相似文献

The cAMP signal transduction pathway mediates the switch between yeast-like and filamentous growth and influences both sexual development and pathogenicity in the smut fungus Ustilago maydis. Signaling via cAMP may also play a role in fungicide resistance in U. maydis. In particular, the adr1 gene, which encodes the catalytic subunit of the U. maydis cAMP-dependent protein kinase (PKA), is implicated in resistance to the dicarboximide and aromatic hydrocarbon fungicides. In this study, we examined the sensitivity of PKA to vinclozolin and could not demonstrate direct inhibition of protein kinase activity. However, we did find that mutants with disruptions in the ubc1 gene, which encodes the regulatory subunit of PKA, were resistant to both vinclozolin and chloroneb. We also found that these fungicides altered the morphology of both wild-type and ubc1 mutant cells. In addition, strains that are defective in ubc1 display osmotic sensitivity, a property often associated with vinclozolin and chloroneb resistance in other fungi.  相似文献

The new fungicides iprodione, vinclozolin and prochloraz, and also a mixture containing carbendazim and maneb, were compared with the established protectant fungicides dichlofluanid and chlorothalonil for effectiveness against grey mould ( Botrytis cinerea ) in an unheated tomato crop. Iprodione and vinclozolin gave the best control of lesions on leaves and stems but dichlofluanid was the most effective in controlling ghost-spotting of fruit. The carbendazim/maneb mixture was effective against a carbendazim-sensitive isolate but not against an insensitive isolate of B. cinerea . When applied 1 or 2 days after inoculation, all five protectant fungicides controlled the insensitive isolate better than the sensitive isolate.  相似文献

Grey mould, caused by Botrytis cinerea Pers ex Fr., is one of the most common diseases of tomato worldwide. Fludioxonil belongs to the phenylpyrrole fungicides, which have high activity against B. cinerea. The sensitivity of fludioxonil was evaluated on the basis of the level of inhibition of mycelium growth in 274 B. cinerea isolates collected from different locations (untreated with this fungicide) in Henan Province, China. The EC50 values for fludioxonil ranged from 0.0033 to 0.0415 mg/l, and the average EC50 values were 0.0156 ± 0.0078 mg/l. Three fludioxonil‐resistant mutants were obtained by subculturing fludioxonil‐sensitive wild‐type isolates on continuously increasing fludioxonil concentrations. For the cross‐resistance assay, fludioxonil revealed positive cross‐resistance with procymidone but did not reveal cross‐resistance with pyrimethanil, boscalid and trifloxystrobin. Mycelial growth, conidial production, hyphal dry weight and pathogenicity were diminished significantly between the fludioxonil‐resistant mutants and their sensitive wild‐type parental isolates. This study shows for the first time that fludioxonil‐resistant isolates of B. cinerea are still not present in Henan Province because this fungicide is an attractive and effective fungicide for chemical control. Recommendations can be made to growers to use fludioxonil to control grey mould and to consider the potential moderate resistance risk of using this fungicide.  相似文献

In this study, sensitivities of 156 Sclerotinia sclerotiorum isolates collected from sunflower fields of West Azarbaijan province, Iran, were assessed to carbendazim and iprodione, and the baseline sensitivities were established for azoxystrobin and tebuconazole. Resistance to carbendazim and iprodione was observed in 53.85% and 4.49% of the isolates, respectively. The 50% effective concentration (EC50) values of azoxystrobin for the isolates ranged from 0.017 to 3.515 μg/ml with a mean of 0.330 μg/ml, and 8.97% of the strains showed low levels of resistance to the fungicide. However, in the presence of salicylhydroxamic acid, all isolates were sensitive to azoxystrobin and EC50 values ranged from 0.015 to 0.263 μg/ml with a mean of 0.086 μg/ml. All isolates were found to be sensitive to tebuconazole, and EC50 values ranged from 0.003 to 0.177 μg/ml with a mean of 0.036 μg/ml. Among the multiple-resistant isolates, the strains exhibiting resistance to both carbendazim and iprodione were detected in the highest frequency (4.49%). No correlation was observed between mycelial growth and aggressiveness with fungicide sensitivity of the isolates suggesting the absence of fitness cost associated with resistance to the studied fungicides. The results indicated that iprodione, azoxystrobin and tebuconazole could be effectively used in rotation or mixture in spray programmes to manage S. sclerotiorum in the region. The baselines established for azoxystrobin and tebuconazole would be useful in monitoring the fungal populations in the province to assess possible shifts in fungicide sensitivity of S. sclerotiorum isolates in the future.  相似文献

Sclerotinia stem rot caused by Sclerotinia sclerotiorum is one of the most important diseases in oilseed rape‐growing areas of China. To determine the frequency of resistance of field isolates of S. sclerotiorum to carbendazim and dimethachlone, a total of 556 isolates from 10 different regions of Henan Province were obtained between 2015 and 2016. The frequency of isolates with a high‐resistance phenotype and a moderate‐resistance phenotype to carbendazim was 69.2% and 10.8%, respectively. However, S. sclerotiorum isolates resistant to dimethachlone were not detected. The baseline sensitivity of S. sclerotiorum to dimethachlone was distributed as a unimodal curve with a mean EC50 value of 0.39 ± 0.09 μg ml?1 for the inhibition of mycelial growth. Four dimethachlone‐resistant mutants were obtained from 20 wild‐type isolates induced by exposure to increasing concentrations of the fungicide in vitro. The mutants showed high levels of resistance to dimethachlone, with resistance factors that ranged from 179 to 323. Positive cross‐resistance occurred between dimethachlone and procymidone, iprodione, and fludioxonil; however, no cross‐resistance was observed for carbendazim and boscalid. The fitness of the dimethachlone‐resistant mutants was significantly lower than that of the wild‐type isolates, as measured by mycelial growth, hyphal dry weight, sclerotium number and dry weight, and pathogenicity. Additionally, based on osmotic tests, the inhibition of mycelial growth caused by NaCl applied at different concentrations was significantly higher for the dimethachlone‐resistant mutants than for their wild‐type parents.  相似文献

Rates of oxygen consumption were measured in the geothermal, hot spring fish, Oreochromis alcalicus grahami by stopped flow respirometry. At 37° C, routine oxygen consumption followed the allometric relationship: V o2=0.738 M 0.75, where V o2 is ml O2 h −1 and M is body mass (g). This represents a routine metabolic rate for a 10 g fish at 37° C of 0.415 ml O2 g−1 h −1 (16.4 μmol O2 g −1 h −1). Acutely increasing the temperature from 37 to 42° C significantly elevated the rate of O2 consumption from 0.739 to 0.970 ml O2 g −1 h −1 ( Q 10=l.72). In the field, O. a. grahami was observed to be 'gulping' air from the surface of the water especially in hot springs that exceeded 40° C. O. a. grahami may utilize aerial respiration when O2 requirements are high.  相似文献

This study identified the main pathogens causing fruit rots of mature peaches in northern Greece, the major peach producing area of Greece. The brown rot pathogen Monilinia laxa was responsible for approximately 70% and 78% of rotted peaches in 2005 and 2006 respectively. Serious damage (up to 5%) was also caused with the fungus Phomopsis amygdali. Other pathogens isolated from rotted peaches at a low percentage were Alternaria alternata, Aspergillus niger, Aspergillus flavus, Botrytis cinerea, Sclerotinia sclerotiorum, Fusarium spp., Colletotrichum gloeosporioides, Rhizopus stolonifer and Gilbertella persicaria. Most fungal isolates originated from the rotted peaches were tested for their sensitivity to the fungicides iprodione, carbendazim, thiophanate methyl and tebuconazole at label recommended concentrations. All fungicides inhibited the growth of M. laxa, A. niger, A. flavus, S. sclerotiorum, P. amygdali and B. cinerea on poisoned agar. Apart from iprodione, all other fungicides inhibited the mycelium growth of the pathogen Fusarium sp. The mycelium growth of Fusarium sp. was significantly less with iprodione than control. Only iprodione and tebuconazole were effective against A. alternata and R. stolonifer. Tebuconazole inhibited the mycelium growth of R. stolonifer, while iprodione reduced significantly in comparison to control. The mycelium growth of the fungus C. gloeosporioides was inhibited by tebuconazole and reduced significantly by the fungicides thiophanate methyl, carbendazim and iprodione. Among all the fungi tested, only M. laxa and B. cinerea isolates were found resistant to benzimidazoles [the EC50 (50% effective concentration) value was 100–200 mg/l and 200–300 mg/l for the largest number of thiophanate methyl‐ and carbendazim‐resistant M. laxa isolates respectively, while the biggest number of B. cinerea thiophanate methyl‐ and carbendazim‐resistant isolates showed EC50 value 200–300 mg/l and 300–400 mg/l, respectively]. However, these strains were sensitive to tebuconazole and iprodione. Therefore, these fungicides can be used as an alternative method to control benzimidazole‐resistant Monilinia and Botrytis isolates.  相似文献

Sclerotia of Botrytis cinerea were found to be a source of primary inoculum for bunch rot of grapes in the Hunter Valley of New South Wales. Conidia-bearing sclerotia were found on canes on grapevines. Resporulation of sclerotia was observed and sporogenic germination led to infection of flowers and berries. Sclerotia survived for eight months in untreated soil in the laboratory. Application of dicarboximides (procymidone, iprodione and vinclozolin) reduced infection of flowers and bunch rotcaused by the sporogenic germination of sclerotia.  相似文献
