Relaxation is the process by which, after contraction, the muscle actively returns to its initial conditions of length and load. In rhythmically active muscles such as diaphragm, relaxation is of physiological importance because diaphragm must return to a relatively constant resting position at the end of each contraction-relaxation cycle. Rapid and complete relaxation of the diaphragm is likely to play an important role in adaptation to changes in respiratory load and breathing frequency. Regulation of diaphragm relaxation at the molecular and cellular levels involves Ca(2+) removal from the myofilaments, active Ca(2+) pumping by the sarcoplasmic reticulum (SR), and decrease in the number of working cross bridges. The relative contribution of these mechanisms mainly depends on sarcomere length, muscle tension, and the intrinsic contractile function. Increased capacity of SR to take up Ca(2+) can arise from increased density of active SR pumping sites or in slow-twitch fibers from phosphorylation of phospholamban, whereas impaired coupling between ATP hydrolysis and Ca(2+) transport into the SR or intracellular acidosis reduces SR Ca(2+) pump activity. In experimental conditions of decreased contractile performance, slowed, enhanced, or unchanged relaxation rates have been reported in vitro. In vivo, a slowing in the rate of decline of the respiratory pressure is generally considered an early reliable index of respiratory muscle fatigue. Impaired relaxation rate may, in turn, favor mismatch between blood flow and metabolic demand, especially at high breathing frequencies.  相似文献

A primary mediastinal endodermal sinus tumor in a young man was diagnosed by cytologic examination of a pleural effusion. Subsequent evaluation revealed a greatly elevated serum alphafetoprotein (AFP); computed tomographic scan of the chest showed a large anterior mediastinal mass. Routine examination of the smears and cell block preparations revealed clusters of tumor cells with a few intracytoplasmic hyaline droplets. Immunohistochemical stains for AFP, alpha-1-antitrypsin and cytokeratin were positive in the tumor cells while stains for carcinoembryonic antigen and the beta subunit of human chorionic gonadotropin were negative. This supported the diagnosis of endodermal sinus tumor, a rare primary tumor within the mediastinum.  相似文献

PurposeTo develop an on-lattice agent-based model describing the growth of multicellular tumor spheroids using simple Monte Carlo tools.MethodsCells are situated on the vertices of a cubic grid. Different cell states (proliferative, hypoxic or dead) and cell evolution rules, driven by 10 parameters, and the effects of the culture medium are included. About twenty spheroids of MCF-7 human breast cancer were cultivated and the experimental data were used for tuning the model parameters.ResultsSimulated spheroids showed adequate sizes of the necrotic nuclei and of the hypoxic and proliferative cell phases as a function of the growth time, mimicking the overall characteristics of the experimental spheroids. The relation between the radii of the necrotic nucleus and the whole spheroid obtained in the simulations was similar to the experimental one and the number of cells, as a function of the spheroid volume, was well reproduced. The statistical variability of the Monte Carlo model described the whole volume range observed for the experimental spheroids. Assuming that the model parameters vary within Gaussian distributions it was obtained a sample of spheroids that reproduced much better the experimental findings.ConclusionsThe model developed allows describing the growth of in vitro multicellular spheroids and the experimental variability can be well reproduced. Its flexibility permits to vary both the agents involved and the rules that govern the spheroid growth. More general situations, such as, e. g., tumor vascularization, radiotherapy effects on solid tumors, or the validity of the tumor growth mathematical models can be studied.  相似文献

Summary The bovine testis has a central mediastinum consisting of longitudinally oriented rete channels and spacious lymph vessels, embedded in the mediastinal stroma. The latter represents a contractile-elastic unit and is composed of myofibroblasts, collagen bundles and accumulations of elastin, connecting the myofibroblasts. The dimension of the mediastinum varies in cross sections at different levels between 3.5 and 31.8 mm2. In one cross section 30 rete channels and 30 openings of straight testicular tubules are encountered. Nearly 25% of the area is occupied by thinwalled, valveless lymph vessels. Arterial convolutes, interpolated between straight centripetal and straight centrifugal branches of the testicular artery flank the rete on all sides. It is concluded that the pulsation within these convolutes together with the contractile-elastic stroma promotes lymph and rete content in a caudo-cranial direction. Chordae retis as described by Roosen-Runge and Holstein (1978) for the human testis are a common feature in the bovine mediastinum testis. The rete channels are lined by a simple cuboidal or columnar epithelium. Short intraepithelial crypts are present and function as epithelial reserve for dilatation and expansion of the rate. The inventory of organelles is rather inconspicuous in the rete epithelium. The apical border bears short microvilli and gives a strong reaction for alkaline phosphatase. The basal cytoplasm contains many small to medium-sized electron-dense bodies and is site of a strong acid phosphatase reaction. The rete epithelium as a whole reacts strongly with leucine aminopeptidase, the marker enzyme of the testicular excurrent duct system. Many free mononuclear cells, mostly macrophages, are observed in the basal half of the rete epithelium.  相似文献
