  • 1.1. The adrenocortical responsiveness to an induced Stressor was monitored in free-living spotted hyenas belonging to a number of social and reproductive categories.
  • 2.2. No significant differences between sexes, or changes in mean cortisol concentrations during serial sampling within the sexes, could be demonstrated.
  • 3.3. The extreme individual variance in temporal cortisol profiles recorded in this study is inexplicable, as it was not related to differences in immobilization procedure, sex, age, reproductive or social category.
  • 4.4. Females, which are the dominant sex in this species, generally showed larger percentage increases in cortisol concentrations during serial sampling.

  • 1.1. A method is described for the accurate and rapid measurement of protein- and non-protein-bound cortisol by miniature gel filtration in small volumes of plasma, e.g. of rodents.
  • 2.2. Binding of cortisol by guinea pig plasma proteins is strongly reduced at elevated temperature (4°C: 102 ± 12ng/ml; 40°C: 5 ± 2 ng/ml).
  • 3.3. Incubation of guinea pig plasma with 1–5000 ng cortisol resulted in a dose-dependent increase in cortisol bound to proteins (specific binding by corticosteroid binding globulin: 230 ± 12 ng/ml).
  • 4.4. Administration of 20 IU (1–24)ACTH induced a significant increase of total protein-bound and non-protein-bound cortisol.
  • 5.5. Values reported in this study agree well with those of previous investigations, in which bound and non-bound glucocorticosteroids were separated by gel filtration on large Sephadex® columns.

  • 1.1. Freshwater nonanadromous rainbow trout, Salmo gairdneri, were injected three times a week with either saline, 10μg cortisol/g, 1.0μg thyroxine/g or 10μg cortisol/g + 1.0μg thyroxine/g during a period of 28 days (12 injections). A separate group was derived as a subgroup from the thyroxine group on day 14 and received Cortisol + thyroxine from day 14 until day 28 (six injections).
  • 2.2. Gill chloride cell number and Na+/K+-ATPase activity increased by cortisol treatment, the changes being significant on days 7 and 14, respectively.
  • 3.3. Thyroxine treatment did not affect gill Na+/K+-ATPase activity or chloride cell number directly. Neither did it modify the stimulatory effect of cortisol on these parameters.
  • 4.4. Muscle water decreased in cortisol-treated fish and increased in thyroxine-treated fish, while no changes were observed in the combined hormone groups.
  • 5.5. No changes were observed in plasma chloride in any group during the experiment.
  • 6.6. The results demonstrate a putative role of cortisol in stimulating hypo-osmoregulatory mechanisms and suggest that thyroxine is without a direct or a supportive effect for cortisol action.

  • 1.1. A mechanical tissue chopper was used to obtain liver explants (35–75 mg) from 2- to 3-week-old chickens to determine both tissue sensitivity and metabolic effects of isoproterenol, avian insulin and glucagon.
  • 2.2. Avian insulin had no effect on lipogenesis; however, lipogenesis was decreased by dibutyryl cyclic AMP. Insulin did not overcome a decrease in lipogenesis caused by catecholamines. Therefore, this control mechanisms is not modulated by insulin.
  • 3.3. Preincubation in the presence of glucagon decreased in vitro lipogenesis. Preincubation in the presence of a 19–29 amino acid construct that approximated the radioimmune site for glucagon did not result in a similar effect. Therefore, this site does not relate to the biopotency of the hormone.
  • 4.4. A previously noted catecholamine induced decrease in in vitro lipogenesis was verified, showing that points of in vitro regulation are under phosphorylation-dephosphorylation control.
  • 5.5. Preincubation of slices (1 hr) with propranolol blocked the inhibition of lipogenesis caused by α and β adrenergic agonists (arterenol or isoproterenol) during a subsequent 2-hr incubation.
  • 6.6. Preincubation of slices with either of these agonists decreased lipogenesis even following an extensive washout.
  • 7.7. Inhibition could be overcome with propranolol, a β adrenergic antagonist.

  • 1.1. A comparison was made of the mechanical performance of heart muscle from mouse, an atricial mammal, with corticosterone as glucocorticoid and spiny mouse (Acomys cahirinus), a precocial mammal, with cortisol as glucocorticoid.
  • 2.2. Force-frequency responses were negative in mouse and positive in spiny mouse.
  • 3.3. During recovery, there was a gradual increase and an overshoot in the mouse, while in the spiny mouse there was an initial enhanced response, diminishing gradually with time.
  • 4.4. High calcium concentration inhibited contractile tension in mouse heart, while it was positively inotropic in spiny mouse heart. Changes in the concentration of calcium did not change the patterns of force-frequency response.
  • 5.5. Lowering the experimental temperature increased the time course and amplitude of the tension curve. However, various parameters exhibited different temperature sensitivity.
  • 6.6. There was a significant difference in the levels of circulating cortisol between male and female spiny mice.
  • 7.7. It is proposed that the differences in the mechanical responses of mouse and spiny mouse hearts may be explained in terms of the effects of the specific glucocorticoid hormone on the development of the sodium-calcium exchanger.

  • 1.1. As previously shown, 14 mM d-glucose, a non-insulinotropic concentration in isolated chicken pancreas, permits an insulin release in response to d-glyceraldehyde, (d-GA; a glycolytic fuel) and l-leucine or α-ketoisocaproic acid (α-KIC) (non-glycolytic fuels), which alone are not initiators of insulin release in this species.
  • 2.2. The “permissive” effect of d-glucose was also observed in the presence of d-mannose (which, as shown herein, is not insulinotropic alone).
  • 3.3. The specificity of glucose for this “permissive” effect was, therefore, subsequently questioned in the presence of 10mM α-KIC by substituting various glycolytic and non-glycolytic fuels to glucose.
  • 4.4. d-GA (at 5 and 15mM), d-mannose (30 and 50 mM), or the association of l-glutamine + l-asparagine permitted an insulin release in response to α-KIC.
  • 5.5. The response was, however, delayed with d-GA, only occasionally with 50 mM d-mannose, and required high concentrations and was delayed in the presence of l-glutamine + l-asparagine as compared to that obtained with 14mM d-glucose + α-KIC.
  • 6.6. In conclusion, the threshold of fuel-induced insulin release is much higher in the chicken than in mammals and this threshold is most efficiently lowered by glucose.

  • 1.1. The adrenal cortex is necessary for survival of echidnas at low ambient temperatures. In this study, adrenal gland activity was investigated in echidnas exposed to 2 days of cold (14°C) and fasting, alternating with 2 days at room temperature and feeding ad lib.
  • 2.2. In the cold. 2.75 ± 0.29% (SD) of the initial body weight was lost daily. Plasma amino acid concentration did not change while glucose concentration decreased from 2.6 ± 0.3 to 1.5 ± 0.3 mmol/l with consecutive sessions of cold.
  • 3.Plasma concentrations of corticosterone (7.2 ± 1.4 nmol/l) and cortisol (4.4 ± 1.9 nmol/l) were unchanged by repeated cold exposure. However, the adrenal response to ACTH stimulation decreased and the clearance of corticosteroids increased after cold exposure.
  • 4.It was concluded that exposure to cold increases the utilization of glucocorticoids and decreases the capacity for their biosynthesis.

  • 1.1. The effect of regular handling on fear reactions was investigated in mallard (Anas platyrhynchos) by exposing six hand-reared and four wild ducks to an approaching human being and recording heart rates with an external ECG device.
  • 2.2. All ducks reacted to the approach with tachycardia, but the response was significantly less in tame birds.
  • 3.3. Hand-reared females showed less response than males. No sex-linked differences were apparent in the wild ducks.
  • 4.4. Decreasing responses throughout the experiments were only found in tame birds.
  • 5.5. Fear or stress reactions can apparently be diminished through habituation induced by regular handling.

  • 1.1. The effect of adenosine separately or in combination with alpha-1 adrenergic antagonist prazosin and alpha-2 adrenergic antagonist yohimbine as well as adenosine antagonists 8-phenyltheophylline and xanthine amine conjugate on glucose-induced insulin secretion from isolated rat pancreatic islets was studied.
  • 2.2. Their in vivo effects on serum glucose and insulin levels were also investigated. Adenosine at 10 and 100 μM inhibited significantly, insulin secretion from the isolated islets whereas at 10 mM slightly increased the secretion of insulin.
  • 3.3. Prazosin used at 100 μM inhibited insulin secretion. When it combined with adenosine (10 μM) it augmented the inhibitory effect of adenosine.
  • 4.4. In vivo prazosin (21 mg/kg bodywt) caused a hyperglycaemia which was accompanied by hypoinsulinaemia.
  • 5.5. Concurrent administration of this drug with adenosine neither affect the hyperglycaemic nor the hypoinsulinaemic effects of adenosine.
  • 6.6. On the other hand, yohimbine (100 μM) has no effect neither separately nor in combination with adenosine (10 μM) in modulating the inhibitory effect of adenosine on insulin secretion.
  • 7.7. When Yohimbine administered at 19.5 mg/kg body wt it did not alter serum glucose but it markedly increased the serum insulin level. Its combined administration with adenosine reduced the hyperglycaemic effect of adenosine with a remarkable increase in serum insulin.
  • 8.8. Both adenosine-antagonists were ineffective in alteration of insulin secretion.
  • 9.9. However, combination of 8-phenyltheophylline with adenosine (10 μM) totally blocked the inhibitory effect of adenosine on insulin secretion while xanthine amine conjugate failed to prevent this effect of adenosine.
  • 10.10. These results indicate that the inhibitory effect of adenosine on insulin secretion is neither mediated via alpha-1 nor alpha-2 adrenoceptors. It might be via activation of specific adenosine receptors on rat islets which are sensitive to blockade by 8-phenyltheophylline.

  • 1.1. The mechanism of action of glyburide (a sulfonylurea) on muscle has been investigated by measuring glucose uptake and glucose transporter (GLUT4) protein levels after chronic glyburide treatment.
  • 2.2. A dietary induced insulin resistant rat model (4 wk of high-fat, high-sucrose feeding) was given glyburide (2mg/kg/day) for 10 days and glucose uptake was measured in a perfused hindquarter preparation.
  • 3.3. Protein levels of the GLUT4 glucose transporter were determined by Western analysis.
  • 4.4. After 7 days of treatment, rats fed glyburide had lower blood glucose concentrations 2 hr (72 ± 5 vs 103 ± 12 mg/dl) and 24 hr (97 ± 7 vs 123 ± 7 mg/dl) after glyburide administration with no difference in serum insulin levels compared to vehicle treated animals.
  • 5.5. Glucose uptake was approx doubled in basal state (0 insulin) in response to glyburide (2.8 + 0.4 vs 1.7 ± 0.2μ mol/g per hr).
  • 6.6. Maximal insulin (100 nM) stimulated glucose uptake tended to be higher in the glyburide treated group, but did not reach statistical significance (8.0 ± 0.7 vs 7.0 ± 0.6 μmol/g per hr).
  • 7.7. Western analysis revealed no significant effect of glyburide on the GLUT4 protein level in skeletal muscle.
  • 8.8. These results suggest that glyburide alters glucose uptake through some mechanism other than alterations in the level of the GLUT4 glucose transporter protein.

  • 1.1. Rainbow trout were acclimated to salt water (1.5, 2.0 or 3.0%, which means 40, 60 or 85% concentrated sea-water) and the electrolyte, glucose and cortisol concentrations of the plasma as well as the extra- and intracellular muscle space, the muscle electrolyte concentrations and the ATPase activity were analysed.
  • 2.2. Plasma osmolality, Na+, Ca2+ and Mg2+ concentrations of the plasma had a maximum at 24 hr after the start of acclimation when acclimated to 3.0% salt water. Plasma osmolality, Na+ and Mg2+ concentrations were significantly higher during the whole acclimation time when exposed to 3.0% salt water.
  • 3.3. Variations and regulations of ECS and ICS were clearly demonstrated. The intracellular electrolyte concentrations were also maximal at 24 hr.
  • 4.4. The plasma glucose level was just slightly elevated, but the cortisol level clearly indicated a stress response at 24 hr.
  • 5.5. The activity of gill Na-K-ATPase increased during the acclimation time.
  • 6.6. The regulatory processes in trout during acclimation to salt water are compared with those occurring in tilapia and carp.

  • 1.1. Changes in glomerular nitration rate (GFR), urine and blood properties and plasma catecholamines of carp were investigated during and following hypoxia.

  • 6.7.No differences in the vascular smooth muscle responsiveness to acetylcholine or sodium nitrate was observed between groups or within different vascular beds.
  • 7.8.The increased sensitivity of males to adrenergic challenge could explain in part some of the existing sex differences in cardiovascular disease and hypertension.

  • 1.1. Hormonal regulation of apolipoprotein E (apoE) gene expression by insulin and thyroid hormone was studied in a human hepatoma cell line, HepG2.
  • 2.2. Changes at the mRNA level, mRNA translation, in vivo synthesis and secretion were monitored.
  • 3.3. Both insulin and triiodothyronine were found to have no significant effect on apoE mRNA levels.
  • 4.4. Insulin treatment caused an inhibition of: (a) the in vitro translation of endogenous apoE mRNA in a HepG2 cell-free system (25%), and (b) the incorporation of radioactivity into newly-synthesized apoE in an in vivo pulse-chase labeling experiment (32%).
  • 5.5. Interestingly, apoE secretion rate was found to be significantly reduced with insulin (84%) suggesting that a major portion of newly-synthesized apoE may be shunted into a degradative pathway.
  • 6.6. Using a similar experimental approach, triiodothyronine showed no significant effect on the rate of apoE synthesis or translation (6–15% decrease), however a slight reduction (20%) in secretion rate was shown.
  • 7.7. Overall, apoE gene expression does not appear to be influenced by triiodothyronine significantly but is modulated by insulin at the translational and post-translational level.

  • 1.1. The intestinal absorption of insulin in carps was assessed examining the transepithelial passage of ingested gold-labeled hormone by electron microscopy. Insulin transfer occurred mainly through the intercellular spaces between the enterocytes.
  • 2.2. When reaching the lamina propria, the gold-labeled hormone gathered predominantly around the granules of the granular cells, and therefore can enter the circulatory system via the blood capillaries which are found in close contact with these cells.
  • 3.3. Winter-acclimatized carp were also capable of internalizing the hormone when fed with insulin.
  • 4.4. Furthermore, the absorbed hormone revealed full activity in regard to the observed changes in the ultrastructure of the liver cells of the treated cold-adapted fish.
  • 5.5. The fish ingesting the hormone underwent the same type of hepatic ultrastructure reprogramming observed when winter-acclimatized carps are injected intraperitoneally with insulin, i.e. conversion to a phenotype corresponding to hepatocytes from summer-adapted carp.
  • 6.6. The oral absorption of insulin by winter-acclimatized fish and its effect in reversing the cold-adaptive state might be useful for the fish culturing industry.

  • 1.1. The aim of this study was to find out whether the anaerobic threshold (AT) can be estimated in rats running at increasing speed and if so what is the reproducibility of the measurements.
  • 2.2. Lactate (LA) concentrations in blood taken from 11 rats were determined during a discontinued, multistage treadmill exercise test repeated four times in each animal.
  • 3.3. It was found that blood LA changes vs speed have an exponential pattern with a distinct, rapid rise at the speed above 25 m/min which corresponds to blood LA of approx. 4 mmol/1.
  • 4.4. The variation coefficient of the speed at which AT occurred in individual animals ranged between 10 and 20%.
  • 5.5. These results offer a potential application of AT determination in the animal studies concerning mechanisms controlling exercise metabolism.
