  • 1.1. Subcellular distribution of (NA+, K+-ATPase and ouabain-insensitive ATPase (Mg2+-ATPase) are compared in branchial tissues of the euryhaline crab, Eriocheir sinensis, acclimated to fresh water.
  • 2.2. Both the anterior and posterior gills contain cAMP-dependent protein kinase and endogenous protein substrate for phosphorylation.
  • 3.3. Phosphorylation occurs in both “particulate” and “soluble” subcellular fractions but its stimulation by cAMP is restricted to the “soluble” fraction.
  • 4.4. serotonin (5-HT) and dopamine receptors are present only in the “light particulate” fraction isolated from the posterior gills.

  • 1.1. Elastase has been purified from the hepatopancreas of the king crab (Paralithodes camtschatica). Specific activity of the enzyme measured toward Suc-(Ala)3-pNA and Boc-(Ala)3-pNA was 926 and 3700 mUnits per mg of protein, respectively.
  • 2.2. The enzyme is an anion protein (pI 4.5) with an approximate mol.wt of 28.5 kDa.
  • 3.3. The enzyme exhibited a bell-shaped pH-dependence for the hydrolysis of Suc-(Ala)3-pNA with a maximum at 8–8.5. Under these conditions the values of Km and kcat of the crab elastase are 4 mM and 4.75 s−1, respectively.
  • 4.4. The serine elastase is effectively inhibited by elastinal and diisopropylfluorophosphate.
  • 5.5. It is shown that some salts except HgCl2 activate the protease. In the presence of HgCl2 with concentrations of 10 mM and higher, the crab elastase is inactive. SDS and Triton X-100 have no any effect on the activity of crab elastase.

  • 1.1. A rapid method for the isolation of a plasma membrane-rich fraction from crab leg muscle, with high purity and yield recovery was developed.
  • 2.2. The method is based on sodium iodide extraction of the crude homogenate, followed by centrifugation on Percoll self-creating gradient.
  • 3.3. (Na+K+)ATPase and alkaline phosphodiesterase I were used as marker enzymes for the plasma membrane and revealed levels of purification of approximately 13-fold and yields recovery of the total activity in the crude muscle homogenate of approximately 18%, for both species of crab studied.

  • 1.1. Egg yolk lipoproteins from four species of Crustacea were isolated by differential density gradient ultracentrifugation.
  • 2.2. Egg yolk proteins from freshwater prawn, striped stone crab and mitten crab consissted of high-density lipoprotein (HDL) and lipid-free protein, while low-density lipoprotein (LDL) was present in the egg yolk protein of sand crayfish as well as HDL and lipid-free protein.
  • 3.3. HDL was a major component in the egg yolk proteins from four species of Crustacea. HDL was identical to egg yolk lipovitellin.
  • 4.4. Both HDL and LDL possessed phospholipid as a major lipid.
  • 5.5. HDL, but not LDL, contained carotenoids. The color of HDL from mitten crab showed a reddish purple and was distinct from other Crustacea whose color was orange. The reddish purple color was characterized by an absorption flexion at 600–650 nm.

  • 1.1. A half platelet preparation from Chinese crab (Eriocheir sinensis) gill is described which allows electrophysiological investigations of ion transport by gill epithelial monolayer when mounted in a modified Ussing chamber.
  • 2.2. The resistance of these preparations equals half that of complete gill platelets (containing the gill epithelium and cuticle twice) indicating that cell damage during preparation of half platelets is negligible.
  • 3.3. The transepithelial resistance (resistance of cuticle subtracted previously) was determined to be about 140 Ω cm2 when both sides are bathed with identical salines.
  • 4.4. Similarities to the results obtained with perfused complete gills demonstrates the reliability of this preparation.
  • 5.5. When identical salines are applied on both sides of the epithelium an outside positive transepithelial potential difference (PDte) up to 40 mV was measured.
  • 6.6. The occurrence of such a high PDte under symmetric conditions and its sensitivity to CN suggests the PDte to be generated by active transport processes.

  • 1.1. A bioassay for octopus saliva, based on detachment of crab dactylopodite flexor muscle under standard conditions, has been developed.
  • 2.2. There is a direct relationship between increasing caseinolytic activity of saliva from Eledone cirrhosa and decreasing muscle detachment time.
  • 3.3. Fractionation of saliva, using preparative isoelectric focusing, shows that muscle releasing activity is restricted to fractions containing proteins with high isoelectric points and maximum caseinase activity.

  • 1.1. Oxygen consumption in six crustacean species from the continental shelf of the NW Gulf of Mexico was related to the ash free weight per dry weight (afdw) gram of the tissue excluding the exoskeleton, suggesting a close relationship to their life strategies.
  • 2.2. Dry weight exoskeleton proportion varied among species, with highest values recorded in callapid brachyuran crab species (46.8–52.5%) against the values recorded in portunid crab (45.8–48%) and the shrimp species (26.7–35.7%).
  • 3.3. Oxygen consumption was related to afdw in each species according to the expression y = axb, with b values ranging from 0.68 to 2.92.
  • 4.4. Rates of oxygen consumption per afdw/dw were larger in shrimps than in portunids and callapids, and was related to the morphophysiology and lifestyle of six species described; the former as the morphological design of the exoskeleton versus the muscle content in the species and the latter as the activity rate in the environment.
  • 5.5. The oxygen extraction efficiency, calculated from oxygen consumption, was higher in the eurytopic species Penaeus aztecus and Callinecies similis than in species restricted to the marine environment, hence considered as a response to environmental fluctuations.

The lipid components of three animals, the rock crab Nectocarcinus integrifons, the rock flathead Platycephalus laevigatus and the southern garfish Hyporhamphus melanochir, feeding in the seagrass beds at Corner Inlet, Victoria, Australia have been examined in detail in order to provide further information on seagrass community structure.
  • 2.2. Biological marker compounds detected within animal gut content material were used to recognize dietary sources and then utilized by community members.
  • 3.3. Both H. melanochir and N. integrifons have been shown to ingest and to varying degrees incorporate seagrass lipid material, thus further confirming the importance of seagrass carbon in the Corner Inlet environment.
  • 4.4. The southern sea garfish H. melanochir is observed to remove C18 PUFAs (polyunsaturated fatty acids) from ingested seagrass material.
  • 5.5. Seagrass sterols are altered during incorporation into the lipids of this fish.
  • 6.6. Lipid-rich digestive juices play a role in the digestive processes of all three animals.
  • 7.7. Components tentatively identified as (NMI) (non-methylene interrupted) fatty acids have been detected in the lipids of the garfish H. melanochir and the crab N. integrifons.
  • 8.8. The fecal material of all three animals represent possible sources of these lipids (NMI acids) in Corner Inlet sediments.
  • 9.9. Based on lipid compositional data, N. integrifons feeds on Posidonia australis detritus and associated epiphyte material.
  • 10.10. The removal of both plant and epibiota cellular lipids along the digestive tract of the crab was observed, although structural components such as long chain mono- and α,ω-dicarboxylic acids, which have been previously recognized as seagrass marker lipids are not directly absorbed.

  • 1.1. The activity of brush border enzymes (alkaline phosphatase, maltase, sucrase, trehalase, leucine amino peptidase) was higher in purified membranes prepared with calcium. The contamination of these membranes with basolateral membranes was also lower (1.27 for Na-K-ATPase activity ratio).
  • 2.2. The extraction of brush border lipids was carried out according to Folch adapted method. Two dimensional thin layer chromatography was used to separate the phospholipidic fractions. Fatty acids of phospholipids were analysed using gas chromatography after acid transmethylation (column SP 2330).
  • 3.3. Phospholipids are composed of phosphatidylcholine (PC: 33%), phosphatidylethanolamine (PE: 30%), sphingomyeline (SM: 21%), phosphatidylserine (PS: 14%) and phosphatidylinositol (PI: 2%). 4. PC, PE and PS are characterized by high levels of unsaturated fatty acids (monounsaturated MUFA: 21.5% and polyunsaturated PUFA: 34.9%). The most abundant PUFA belong to the (n-3) family [18:3 (n-3), 20:5 (n-3) and 22:6 (n-3)].
  • 4.5. Fatty acids from sphingomyelin of purified membranes have low proportions of PUFA (13.5%) but higher proportions of MUFA (39.5%).
  • 5.6. No specific differences were found between calcium and magnesium prepared membranes.
  • 6.7. The low content in LPC and the absence of LPE confirmed the absence of major structural lipids transformation during the membrane purification with calcium or magnesium.
  • 7.8. Glycine transport was measured during 10 sec at different temperatures using the rapid filtration technique. Glycine transport was higher with Na+ than with K+. In the presence of Na+, this transport increases with temperature.
  • 8.9. Arrhenius curves were mono phasic without obvious breakpoint and indicated no phase transition in the lipid bilayer.
  • 9.10. A significant Na+ dependent glycine transport has been characterized at low temperatures (0°C) which suggests a possible role of membrane polyunsaturated fatty acids in the control of glycine transport.

  • 4.4. A plea is made for the investigation of temporal changes in the [Mg2+] of the haemolymph of a number of crustaceans and for more studies of Mg2+ homoeostasis in general.

  • 1.1. Lipid, glucose and glycogen concentrations were measured in different tissues of the crab Chasmagnathus granulata during emersion.
  • 2.2. After 6 hr of emersion no reduction in the total amount of carbohydrates was found to occur, suggesting that a general metabolic arrest was taking place.
  • 3.3. A transitory increase in haemolymphatic glucose and lipid levels was observed. Possible causes are therefore discussed in relation to changes in the flux of substrates for energy production.

  • 1.1. The carnitine-responsive mutant yeast, Candida pintolopesii ATCC 26014 and the wild type strain (ATCC 22987) were used to investigate the role of carnitine and the carnitine acetyltransferase system.
  • 2.2. [3H]l-Carnitine, supplied to the cells, was incorporated into acetylcamitine and [14C]pantothenate was incorporated into CoA and its derivatives.
  • 3.3. Both bioautography and quantitative assays indicated that the relative amounts of CoA and acetylCoA were very different in the mutant and wild type cells.
  • 4.4. The wild type yeast maintained an acetylCoA/CoA ratio of 0.33 ± 0.09 indicating that most of the CoA in the cell is in the free CoA form. Carnitine was not required to establish this ratio nor did its presence lower it further.
  • 5.5. In contrast, the mutant cells contained a high acetylCoA/CoA ratio (12.8 ± 3.0).
  • 6.6. In the mutant cells, carnitine lowered the ratio by decreasing the intracellular acetylCoA concentration and releasing free CoA.
  • 7.7. These data indicated that wild type yeast possess an effective mechanism that is not related to the CAT system for regulating the acetylCoA/CoA ratio.
  • 8.8. This mechanism appears to be lacking in the mutant. The CAT system decreased the acetylCoA/CoA ratio in the mutant cells but not to the value which is found in the wild type strain.
  • 9.9. In both stains of Candida pintolopesii, in the presence of carnitine, an acetylcamitine pool can be created whose concentration exceeds that of acetylCoA.
  • 10.10. The intracellular apparent equilibrium constant (Kapp) for carnitine acetyltransferase for wild type Candida pintolopesii ATCC 22987 was 0.73 ± 0.12, close to the established value of 0.6, indicating that the CAT system ran close to equilibrium.
  • 11.11. The Kapp for the CAT system of the carnitine-responsive mutant yeast was 7.7 ± 1.7 indicating that this reaction was not at equilibrium.
