The red alga, Porphyridium cruentum, which is one of the potential sources of arachidonic acid, was cultured in batch and continuous vessels. The growth rates in batch cultures were correlated to the mean light intensity in the vessels, and the cell concentrations in continuous cultures were estimated by those results. The yield of arachidonic acid was about 1.2 g per 1012 cell at cell concentrations ranging from 0.5 to 1.5 × 1010 cell/l and independent of the mean light intensity.  相似文献

A protocol has been standardized for establishment and characterization of cell suspension cultures of Stevia rebaudiana in shake flasks, as a strategy to obtain an in vitro stevioside producing cell line. The effect of growth regulators, inoculum density and various concentrations of macro salts have been analyzed, to optimize the biomass growth. Dynamics of stevioside production has been investigated with culture growth in liquid suspensions. The callus used for this purpose was obtained from leaves of 15-day-old in vitro propagated plantlets, on MS medium fortified with benzyl aminopurine (8.9 μM) and naphthalene acetic acid (10.7 μM). The optimal conditions for biomass growth in suspension cultures were found to be 10 g l?1 of inoculum density on fresh weight basis in full strength MS liquid basal medium of initial pH 5.8, augmented with 2,4-dichlorophenoxy acetic acid (0.27 μM), benzyl aminopurine (0.27 μM) and ascorbic acid (0.06 μM), 1.0× NH4NO3 (24.7 mM), 3.0× KNO3 (56.4 mM), 3.0× MgSO4 (4.5 mM) and 3.0× KH2PO4 (3.75 mM), in 150 ml Erlenmeyer flask with 50 ml media and incubated in dark at 110 rpm. The growth kinetics of the cell suspension culture has shown a maximum specific cell growth rate of 3.26 day?1, doubling time of 26.35 h and cell viability of 75 %, respectively. Stevioside content in cell suspension was high during exponential growth phase and decreased subsequently at the stationary phase. The results of present study are useful to scale-up process and augment the S. rebaudiana biological research.  相似文献

The continuous production of nisin, an antibiotic polypeptide, by Lactococcus lactis in a bioreactor system coupled to a microfiltration module is described. Nisin productivity with respect to both cultivation time (ND) and the quantity of glucose consumed (ND/Sf) in continuous production was enhanced by maintaining a low concentration of lactic acid in the broth. A maximum ND of 7.80 × 104l−1·h−1 and ND/Sf of 5.20 × 103 U·g−1·h−1 were obtained when the glucose concentration in the feed medium was 15 g/l. These values represent about 4.1- and 4.5-fold increases, respectively, over those obtained in batch culture.  相似文献

To avoid cells adhering to the ion-exchange membrane and damage of the cells in electrodialysis fermentation (ED-F), ED-F with a microfiltration (MF) module, was used in lactate fermentation. In this system, after 38 h of cultivation, the dry cell weight (2.89 g/l) and the number of viable cells (4.8 × 109/ml) were about 1.8 and 1.6 times more than those of the control fermentation, respectively. Lactate dehydrogenase activity in this system was maintained at a high level, resulting in reduction of fermentation time to complete batch culture within 38 h. From these results, ED-F with an MF module enabled high speed batch culture.  相似文献

Summary Deficiency of inorganic phosphate caused the hyper production of invertase and the derepression of acid phosphatase in a continuous culture ofSaccharomyces carlsbergensis. The specific invertase activity was 40,000 enzyme units per g dry cell weight at a dilution rate lower than 0.05 h–1 with a synthetic glucose medium of which the molecular ratio of KH2PO4 to glucose was less than 0.006. This activity is eight fold higher than in a batch growth and 1.5 fold as much as the highest enzyme activity observed so far in a glucose-limited continuous culture.For the hyper production of invertase, it is necessary to culture the yeast continuously by keeping the Nyholm's conservative inorganic phosphate concentration at less than 0.2 m mole per g dry weight cell. Direct conversion of gaseous H2 and CO2 to CH4 was achieved with Methanobacterium thermoautotrophicum ΔH (DSM 1053) cells fixed either on a cellulose acetate membrane or inside a porous silica-alumina ceramic support.In a membrane bioreactor with cellulose acetate (5 μmø), methane production rate increased in proportion to the contact area between the gases and the methanogen cells, giving a methane production rate of 0.75 ml CH4/cm2 contact area/h. The initial fixed-cell mass of 0.2 mg dry cell/cm2 of contact area increased to 1 mg/cm2 after 12 h of cultivation (steady state).In the ceramic bioreactor (cylindrical, 30 mmø × 70; av. pore size 100 μ, and porosity 79.7%), the methane production rate at steady state was 6 l CH4/l ceramic/l. The methanogen cells grew homogeneously inside the ceramic up to 7 cm depth, and the cell density ranged from 20 to 30 mg dry cell/cm3 ceramic.  相似文献

Repeated-batch fermentation by a flocculating fusant, Saccharomyces cerevisiae HA 2, was done in a molasses medium that contained 20% (w/v) total sugar, at 30°C in an automatically controlled fermentor, and the effects of ethanol concentration on the specific growth rate and the specific production rate of ethanol were studied. Both the specific growth rate and the specific production rate of ethanol fell with increase of ethanol concentration, and there was a linear correlation between each rate and the concentration of thanol. The maximum specific growth rate (μmax) and the maximum specific production rate of ethanol (qmax) were 0.12 h−1 and 0.1 g ethanol/109 cells·h, respectively. The specific growth rate and the specific production rate of ethanol fell to zero at ethanol concentration of 89 g/l and 95 g/l, respectively. The number of viable cells, calculated from the linear inhibition equation, was 1.3 × 109 cells/ml for production of 85 g/l ethanol at a dilution rate (D1) of 0.2 h−1. Based on this estimation, a laboratory-scale continuous fermentation, using two fermentors in series, was done. In the second fermentor, 85 g/l ethanol was produced at a dilution rate (D1) of 0.2 h−1 by the active feedig of the fermented mash from the first fermentor into the second fermentor by pumping (hereafter called active feeding). To maintain the number of viable cells above 109 cells/ml in the second fermentor, a active feeding ratio of more than 23% was required. Under these conditions, 81 g/l ethanol was produced in the second fermentor at a dilution rate (Dt) of 0.25 h−1, and the high ethanol productivity of 20.3 g/l·h could be achieved. A bench-scale continuous fermentation, using two fermentors in series, with a active feeding ratio of 25% was done. An ethanol concentration of 84 g/l in the second fermentor at a dilution rate (Dt) of 0.25 h−1 was achieved, just as it was in the laboratory-scale fermentation test.  相似文献

Summary The growth of Clostridium acetobutylicum was studied by three ways. 1. In batch fermentation, referred to as the control. 2. Fermentation in dialysis which permits elimination of all the products of metabolism: acids, solvents and gases. In order to test the toxic effect of acids, cultures were dialysed against 2 g l-1 acetic acid or 2 g l-1 butyric acid. 3. To test the toxic effect of gases only, batch fermentations were carried out under vacuum or with a continuous bubbling of nitrogen. The first method resulted in a productivity of 1.2 g l-1 dry cell weight and a maximal specific growth rate of 0.2 h-1; the second, 20 g l-1 dry cell weight and a constant maximum specific growth rate (μ=0.39 h-1) between 14 and 20 h. The toxic effect of acetic and butyric acids, starts at low concentrations and about 4 g l-1 of both acids results in a decrease of 50% of maximal specific growth rate. The third series of experiments showed that gases produced by the bacteria have a high toxic effect, comparable to that of 5 g l-1 of acid.  相似文献

Gmelina arborea L. seedling growth and diurnal stomatal opening (as measured by stomatal resistance) were studied at soil matric potential 0, -0.1 and -0.72 × 10?5 Pa. Leaf area, leaf number, plant height and dry weights of the vegetative parts were significantly reduced as soil matric potential decreased from 0 to -0.72 × 10?5 Pa. The growth responses followed the same trend as net assimilation rate and relative growth rate. The highest moisture stress induced leaf senescence and leaf fall. Leaf water potential decreased from - 2 × 10?5 Pa to - 20 × 10?5 Pa with increasing soil moisture stress. Results indicate that the diurnal stomatal opening is controlled by photon flux density when this species is grown at soil matrio potential 0 Pa. However, with decreasing soil matrio potential (- 0.10 and -0.72 × 10?5 Pa) the internal plant water deficit appears to oontrol the stomatal opening.  相似文献

Summary A thermophilic methanogenic bacterium, Methanococcus thermolithotrophicus, was grown on H2 and CO2 in both batch and continuous culture, in a fermentor equipped with either a straight blade impeller or a Rushton impeller. In a study of the control of metabolite formation, prodigiosin production by Serratia marcescens was used as a model. Specific production rates of prodigiosin formation were determined using batch culture technique. Sucrose as carbon source and NH4NO3 as nitrogen source resulted in a specific production rate of 0.476 mg prodigiosin (g cell dry weight)−1 h−1. Prodigiosin formation and productivity was inversely correlated to growth rate when the bacterium was grown under carbon limitation on a defined medium in a chemostat culture. The maximum specific growth rate (μmax) was 0.54 h−1 and prodigiosin was formed in amounts over 1 mg l−1 up to a growth rate (μ) of 0.3 h−1 at steady state conditions. At a dilution rate of 0.1 h−1 growth at steady state with carbon and phosphate limitation supported prodigiosin formation giving a similar specific yield [1.17 mg prodigiosin (g cell dry weight)−1 and 0.94 mg g−1, respectively], however, cells grown with nitrogen limitation [(NH4)2SO4] did not form prodigiosin. Productivity in batch culture was 1.33 mg l−1 h−1 as compared to 0.57 mg l−1 h−1 in the chemostat.  相似文献

The non-pollutant plant support material of the dwarf duckweed Wolffia arrhiza (Fam. Lemnaceae) was used for the entrapment of living yeast cells (Kluyveromyces fragilis) which hydrolyse lactose with the subsequent fermentation of glucose and galactose at high cell densities (up to 7.0 × 108/ml support). The stabile yeast-plant cell immobilizates are able to produce ethanol from lactose-containing media (e.g. whey) by batch fermentation (on a rotary shaker) or continuous fermentation (in a turbulence reactor) for several days (at a pH below 4.2 and a temperature of 30°C). The removal of whey proteins by a preceding heat denaturation of whey, high dilution rates, CSo values of 50 to 60 g lactose per litre whey and the preferential use of the K. fragilis strain DSM 7238 were determined as the prerequisites for an optimum continuous fermentation. Economically interesting productivities (Pmax ? 15 g ethanol/1 · h, D = 0.72 h?1) with an actual lactose turnover of 90% were obtained by using these parameters.  相似文献

Nitrosomonas europaea is a chemolithoautotrophic nitrifier, a gram-negative bacterium that can obtain all energy required for growth from the oxidation of ammonia to nitrite, and this may be beneficial for various biotechnological and environmental applications. However, compared to other bacteria, growth of ammonia oxidizing bacteria is very slow. A prerequisite to produce high cell density N. europaea cultures is to minimize the concentrations of inhibitory metabolic by-products. During growth on ammonia nitrite accumulates, as a consequence, N. europaea cannot grow to high cell concentrations under conventional batch conditions. Here, we show that single-vessel dialysis membrane bioreactors can be used to obtain substantially increased N. europaea biomasses and substantially reduced nitrite levels in media initially containing high amounts of the substrate. Dialysis membrane bioreactor fermentations were run in batch as well as in continuous mode. Growth was monitored with cell concentration determinations, by assessing dry cell mass and by monitoring ammonium consumption as well as nitrite formation. In addition, metabolic activity was probed with in vivo acridine orange staining. Under continuous substrate feed, the maximal cell concentration (2.79?×?1012/L) and maximal dry cell mass (0.895 g/L) achieved more than doubled the highest values reported for N. europaea cultivations to date.  相似文献

In this study, (S)-3-hydroxy-3-phenylpropionate was prepared continuously by coupling microbial transformation and membrane separation. The effect of several factors on membrane flux, reactor capacity, and reaction conversion were investigated. A kinetic model of the continuous reduction process was also developed. The appropriate molecular weight cut-off of the ultrafiltration membrane was 30 kDa. The reactor capacity reached a maximum of 0.136/h at a biomass concentration and membrane flux of 86 g/L (dry weight/reaction volume) and 20 mL/h, respectively. The (S)-3-hydroxy-3-phenylpropionate yield was 3.68 mmol/L/day after continuous reduction over seven days. The enantiometric excess of (S)-3-hydroxy-3-phenylpropionate reached above 99.5%. The kinetic constants of continuous reduction were as follows: r m = 3.00 × 10−3 mol/L/h, k cat = 3.49 × 10−4 mol/L/h, k 1 = 3.09 × 10−2 mol/L, and k 2 = 5.00 × 10−7 mol/L. The kinetic model was in good agreement with the experimental data obtained during continuous reduction. Compared with batch reduction, continuous reduction can significantly improve the catalytic efficiency of microbial cells and increase the reactor capacity.  相似文献

Batch and continuous production of high fructose syrup from Jerusalem artichoke tubers has been studied using yeast cells immobilized in open pore gelatin matrix. In a batch reactor, the hydrolysis was 93% (d-fructose/d-glucose = 90/10) and 42 mg d-fructose per ml was produced from the artichoke tuber extract by immobilized cells in 3 h. The same immobilized cells were recycled and used repeatedly for 10 batch cycles starting with fresh juice at the beginning of each cycle. It was found that immobilized cells were extremely stable and the percent hydrolysis was almost constant for all 10 batch cycles. In a continuous reactor using an immobilized cell concentration of 65.7 g (dry wt) l?1 of total working bioreactor volume, the percent hydrolysis was found to remain constant at ~100% at dilution rates <1.26 h?1, but beyond that it decreased. Volumetric productivity attained its maximum value at D = 2.08 h?1 and was found to be 100 g l?1 h?1. This was achieved at a feed sugar conversion of 80%. At 90% conversion and D = 1.66 h?1, the productivity was found to be 90 g l?1 h?1. Continuous operation of the immobilized cell bioreactor at a constant dilution rate of 1.65 h?1 for 240 h resulted in only 2% loss of original activity.  相似文献

Glucaric acid (GlucA) is a valuable glucose-derived chemical with promising applications as a biodegradable and biocompatible chemical in the manufacturing of plastics, detergents and drugs. Recently, there has been a significant focus on producing GlucA microbially (in vivo) from renewable materials such as glucose, sucrose and myo-inositol. However, these in vivo GlucA production processes generally lack efficiency due to toxicity problems, metabolite competition and suboptimal enzyme ratios. Synthetic biology and accompanying cell-free biocatalysis have been proposed as a viable approach to overcome many of these limitations. However, cell-free biocatalysis faces its own limitations for industrial applications due to high enzyme costs and cofactor consumption. We have constructed a cell-free GlucA pathway and demonstrated a novel framework to overcome limitations of cell-free biocatalysis by i) the combination of both thermostable and mesophilic enzymes, ii) incorporation of a cofactor regeneration system and iii) immobilisation and recycling of the pathway enzymes. The cell-free production of GlucA was achieved from glucose-1-phosphate with a titre of 14.1 ± 0.9 mM (3.0 ± 0.2 g l−1) and a molar yield of 35.2 ± 2.3% using non-immobilised enzymes, and a titre of 8.1 ± 0.2 mM (1.70 ± 0.04 g l−1) and a molar yield of 20.2 ± 0.5% using immobilised enzymes with a total reaction time of 10 h. The resulting productivities (0.30 ± 0.02 g/h/l for free enzymes and 0.170 ± 0.004 g/h/l for immobilised enzymes) are the highest productivities so far reported for glucaric acid production using a synthetic enzyme pathway.  相似文献

l-Methionine-enriched cells production of an ethionine-resistant mutant of Candida boidinii no. 2201 was greatly improved by the control of pH and by feeding of methanol and other medium components during cultivation in a jar fermentor. Under the optimal conditions, 38.5 g (as dry weight)_of cells abd 282 mg of pool methionine (intracellular pool of free l-methionine) per l of culture broth were obtained after 11 d of cultivation.The culture conditions for production of l-methionine-enriched cells in continuous culture were investigated. With limited methanol in continuous cultivation, pool methionine productivity reached a maximum value of 1.14 mg·l−1·h−1 at a dilution rate of 0.05·h−1. During methanol-limited growth in continuous cultivation, the pool methionine content of the mutant was about 20–35% higher than that in batch cultivation.  相似文献

The effect of fed-batch operation (FBO) strategy was investigated using pretreated-beet molasses, containing galactose that induces the lac promoter, on benzaldehyde lyase (BAL) production by recombinant Escherichia coli BL21(DE3)pLySs. After batch cultivation with 30 g l?1 pretreated-beet molasses consisting of 7.5 g l?1 glucose and 7.5 g l?1 fructose, three FBO strategies were applied at dissolved oxygen (=40%) cascade to air-flow rate. In FBO1 when air-flow rate decreased considerably, feed was given to the system in pulses in such a way that pretreated-beet molasses concentration increased by 10 kg m?3 (containing 2.5 g l?1 glucose+2.5 g l?1 fructose); however, decrease in air-flow rate demonstrated only the absence of glucose but not fructose. Thus, in FBO2 when fructose and glucose were completely utilized, pretreated-beet molasses was pulse-fed and its concentration increased by 10 g l?1. In FBO3 with the decreased amount of pretreated-beet molasses (6 g l?1), shift response time from glucose to fructose consumption was avoided, and glucose and fructose consumptions were well correlated with air-flow rate, and the highest C X (8.04 g l?1) and BAL (2,315 U ml?1) production were obtained (t?=?24 h) with the highest substrate yield on cell and product formation.  相似文献

The paper re-evaluates Verhulst and Monod models. It has been claimed that standard logistic equation cannot describe the decline phase of mammalian cells in batch and fed-batch cultures and in some cases it fails to fit somatic growth data. In the present work Verhulst, population-based mechanistic growth model was revisited to describe successfully viable cell density (VCD) in exponential and decline phases of batch and fed-batch cultures of three different CHO cell lines. Verhulst model constants, K, carrying capacity (VCD/ml or μg/ml) and r, intrinsic growth factor (h−1) have physical meaning and they are of biological significance. These two parameters together define the course of growth and productivity and therefore, they are valuable in optimisation of culture media, developing feeding strategies and selection of cell lines for productivity. The Verhulst growth model approach was extended to develop productivity models for batch and fed-batch cultures. All Verhulst models were validated against blind data (R2 > 0.95). Critical examination of theoretical approaches concluded that Monod parameters have no physical meaning. Monod-hybrid (pseudo-mechanistic) batch models were validated against specific growth rates of respective bolus and continuous fed-batch cultures (R2 ≈ 0.90). The reduced form of Monod-hybrid model CL/(KL + CL) describes specific growth rate during metabolic shift (R2 ≈ 0.95). Verhulst substrate-based growth models compared favourably with Monod-hybrid models. Thus, experimental evidence implies that the constants in the Monod-hybrid model may not have physical meaning but they behave similarly to the biological constants in Michaelis–Menten enzyme kinetics, the basis of the Monod growth model.  相似文献

Fed-batch culture of Alcaligenes latus, ATCC 29713, was investigated for producing the intracellular bioplastic poly(β–hydroxybutyric acid), PHB. Constant rate feeding, exponentially increasing feeding rate, and pH-stat fed batch methods were evaluated. pH-stat fed batch culture reduced or delayed accumulation of the substrate in the broth and led to significantly enhanced PHB productivity relative to the other modes of feeding. Presence of excessive substrate appeared to inhibit PHB synthesis, but not the production of cells. In fed-batch culture, the maximum specific growth rate (0.265?h?1) greatly exceeded the value (0.075?h?1) previously observed in batch culture of the same strain. Similarly, the maximum PHB production rate (up to 1.15?g?·?l?1?·?h?1) was nearly 8-fold greater than values observed in batch operations. Fed-batch operation was clearly superior to batch fermentation for producing PHB. A low growth rate was not a prerequisite for PHB accumulation, but a reduced or delayed accumulation of substrate appeared to enhance PHB accumulation. Under the best conditions, PHB constituted up to 63% of dry cell mass after 12?h of culture. The average biomass yield coefficient on sucrose was about 0.35, or a little less than in batch fermentations. The highest PHB concentrations attained were about 18?g?·?l?1.  相似文献
