  • 1.1. Unidirectional Na+ influx in lamprey red blood cells was determined using 22Na as a tracer.
  • 2.2. Total Na+ uptake and amiloride-inhibitable Na+ influx increased in a saturable fashion as a function of external Na+ concentration (Nae).
  • 3.3. At 141 mM Nae, the average value of net Na+ influx was 13 ± 1.1 and the amiloride-sensitive Na+ influx was 5.3±1.1 mmol/l cells per hr (±SE).
  • 4.4. The amiloride-sensitive component of Na+ influx was significantly activated by 10−5 M isoproterenol, by 2 × 10−5 M DNP, and by cell shrinkage.
  • 5.5. Furosemide (1 mM) had no effect on the Na+ transport in red cells.
  • 6.6. The residual amiloride-insensitive component of Na+ transport was a linear function of Nae in the range of 5–141 mM. This transport seems to be accounted for by simple diffusion.

  • 1.1. The effects of extracellular pH on Na+ and Cl absorption were studied in vitro in the small intestine of the winter flounder, Pseudopleuronectes americanus.
  • 2.2. Reductions in bathing solution pH inhibited Jmsna (mucosal-to-serosal flux) and Jnetna (net flux) (r = 0.90) and JnetCl (r = 0.92) [due to an increase in JsmCl, (serosal-to-mucosal)] and decreased short circuit current (Isc).
  • 3.3. Luminal bumetanide (0.1 mM) and amiloride (1 mM) inhibited Na+ and Cl absorption by reducing Jms.
  • 4.4. Luminal barium (5mM) and luminal copper (100 μM) decreased JmsCl and increased JsmCl.
  • 5.5. We conclude that reductions in extracellular pH inhibit a luminal membrane NaCl absorptive process (Na+-K+-2Cl) and stimulate an electrogenic Cl secretory process.

  • 1.1. In sea-water, adult salmon (S. salar) exchange an average of 12.6% of total body sodium/hr.
  • 2.2. Following transfer to fresh water sodium uptake follows Michaelis-Menton kinetics. Fmax = 2.40 mmol Na/1 ECF/hr, Km = 0.26 mmol Na/1. The uptake system is fully activated immediately following transfer to fresh water.
  • 3.3. Post smolts adapted to sea-water for 3 months take up sodium at only one third of the rate of adult fish following return to fresh water.
  • 4.4. The concentration of prolactin in the plasma is low in sea-water adapted fish and does not rise during the first 8 hr in fresh water.
  • 5.5. At pH 5 sodium uptake is reduced by almost 90%, even in the absence of aluminium, but recovers immediately on return to neutral water.
  • 6.6. At pH 5 and 20 μmol Al/1 there is little further effect on sodium uptake but after 6 hr in aluminium the inhibition of sodium uptake continues after return to neutral aluminium fresh water and uptake is only 50% of normal 24 hr later.

  • 1.1. Acute (AT) or chronic (CT) administration of exogenous aldosterone brought about a considerable increase in transmural potential difference (PD), short-circuit current (Isc) and net sodium flux (JnetNa).
  • 2.2. A good relationship between Isc and Jm-sNa was observed in CT but not in AT or UC (untreated controls).
  • 3.3. Addition of mucosal amiloride reduced Isc in AT and CT colons, but did not cause any change in UC colons. The relationship between Isc and Jm-sNa, observed in CT was totally suppressed by the diuretic.
  • 4.4. JnetNa was reduced in AT tissues and abolished in CT colons by amiloride.
  • 5.5. The present results strongly suggest that aldosterone levels quantitatively and qualitatively modify sodium absorption across colonie mucosa in a dose-dependent fashion and that lower levels of the hormone are required to induce the electrogenic Na absorption than to suppress the electroneutral transport.

  • 1.1. 14C-dichlorofarnesoate permeated rapidly into Haemonchus contortus (infective juveniles) and Panagrellus redivivus (mixed cultures) and was strongly bound by hydrophobic association (Ks > 10−4M).
  • 2.2. Uptake rose linearly with increases in temperature (5–38°C) and external concentration (C0; 0.07–2.15 × 10−4 M). Within 1 hr the internal concentration, C1 was >C C0.

  • 1.1. Halobacterium halobium has two chromatographically distinct forms of glutamate dehydrogenase which differ in their thermolability and other properties. One glutamate dehydrogenase utilizes NAD, the other NADP as a coenzyme.
  • 2.2. The NADP-specific glutamate dehydrogenase (EC 1.4.1.4) was purified 65-fold from crude extracts of H. halobium.
  • 3.3. The Michaelis constants for 2-oxoglutarate (13.3 mM), ammonium (3.1 mM) and NADPH (0.077 mM) indicate that the enzyme catalyzes in vivo the formation of glutamate from ammonium and 2-oxoglutarate.
  • 4.4. The amination of 2-oxoglutarate by NADP-specific glutamate dehydrogenase is optimal at the pH value of 8.0–8.5. The optimal NaCl or KCl concentration for the reaction is 1.6 M.
  • 5.5. None of the several metabolites tested for a possible role in the regulation of glutamate dehydrogenase activity appeared to exert an appreciable influence on the enzyme.
  • 6.6. NAD- and NADP-dependent glutamate dehydrogenases from H. halobium showed apparent molecular weights of 148,000 and 215,000 respectively.

  • 1.1. Partially purified rat liver ornithine decarboxylase is inhibited by several diamines including putrescine, 1,3-diaminopropane, cadaverine and p-phenylenediamine.
  • 2.2. The inhibition is dependent on pH, being strong at pH above 8 and negligible below pH 6.5.
  • 3.3. The kinetic study of the inhibition showed that while the aromatic diamine behaved as a simple competitive inhibitor, the aliphatic diamines presented a more complex pattern of inhibition in which two molecules of inhibitor might bind to the enzyme active site.
  • 4.4. The KI values for the different inhibitors were calculated and the degree of affinity for the enzyme was p-phenylenediamine > putrescine > cadaverine > 1,3-diaminopropane.
  • 5.5. A molecular mechanism explaining how one or two molecules of inhibitor can bind to the enzyme is proposed.

  • 1.1. Porcine adipose tissue was incubated with radiolabeled glucose, acetate or lactate. Saturation curves indicated that lactate > glucose > acetate in providing two-carbon units for fatty-acid synthesis.
  • 2.2. Competition between individual substrates indicated that lactate was the best lipogenic substrate.
  • 3.3. Incubation of all three substrates at concentrations observable in serum indicated that at 5.56mM, glucose was the preferred lipogenic substrate in the presence of 0.1 mM acetate and 1.0 mM lactate.
  • 4.4. At elevated concentrations (18.52mM glucose, 1.0 mM acetate and 10.0 mM lactate), acetate and lactate were preferred to glucose as lipogenic substrates.

The change of the enzyme activation kinetics by Mg2+, ATP, Na+ and K+ from simple saturation in FW to cooperativity in BW and other habitat-dependent variations including the pH alkaline shift in BW are hypothetically related to an adaptive significance to the different environmental salinity.
  • 4.4. Gill total lipids and phospholipids are 30% lower in BW than in FW while their ratio is constant; some differences in gill total lipid fatty acid composition between FW and BW do not significantly affect the unsaturation parameters.

The (Na+ + K+)- and Na+-ATPases, both present in kidney microsomes of Sparus auratus L., have different activities and optimal assay conditions as, in the first of the two stocks of fish used (A), the spec. act. of the former is 51.7 μmol Pi mg prot−1 hr−1 at pH 7.5, 100 mM Na+, 10 mM K+, 17.5 mM Mg2+, 7.5 mM ATP and that of the latter is 6.5 μmol Pi mg prot−1 hr−1 at pH 6.5, 40 mM Na+, 4.0 mM Mg2+, 2.5 mM ATP.
  • 2.2. Ouabain and vanadate specifically inhibit the (Na+ + K+)-ATPase but not the Na+-ATPase that is preferentially inhibited by ethacrynic acid.
  • 3.3. While the (Na+ + K+)-ATPase is strictly specific for ATP and Na+, Na+-ATPase can be activated by various monovalent cations and, apart from ATP, hydrolyses CTP, though less efficiently.
  • 4.4. The second stock B, subjected to higher salinity than A, shows an acidic shifted Na+-ATPase optimal pH, opposed to the stability of that of the (Na+ + K+)-ATPase, a decreased (Na+ + K+)-ATPase and a strikingly depressed Na+-ATPase.
  • 5.5. The results are compared with literature data and discussed on the basis of the presumptive different roles as well as functional prevalence in various salinities of the two ATPases.

  • 2.2. Between 159 and 40 tort, blood pH decreases slightly from 7.77 ± 0.03 to 7.65 ± .04, and at 15 torr, blood pH drops to 7.36 ± 0.06.

After 2 and 5 hr exposure to 15 tort, blood lactate levels increase to 3.29 ± 0.47 and 8.91 ± 0.14 mM/l blood, respectively. Upon return to normoxia, blood lactate levels decrease and are comparable to normoxic controls after 13 hr.
  • 4.4. During mild hypoxia, P. elarki maintains adequate oxygen transport by utilizing a high O2 affinity hemocyanin in conjunction with a low metabolic demand by its tissues.

  • 1.1. This work represents the first approach to characterize the transport system of haem pathway precursors, such as δ-aminolevulinic acid (ALA), in two strains of Saccharomyces cerevisiae, a wild type, D27, and a HEM R+ mutant.
  • 2.2. ALA transport occurs unidirectionally by a sole active system with an apparent KM of 0.10 mM, at the optimum pH of 5.0. ALA uptake is influenced by both the carbon and nitrogen source; this suggests a rather complex regulation mechanism.
  • 3.3. This transport is not mediated by the general amino acid permease (GAP).
  • 4.4. ALA uptake is strongly inhibited by compounds harboring a methyl-amine terminus suggesting that this group is essential for ALA transport; however, the electric environment of the carboxylic group may be also important for the interaction between ALA and its transporter active site.
  • 5.5. We have found differences in ALA transport which would indicate a different regulation mechanism for this system in both strain cells.

  • 3.3. The chemically stable, tritium-labeled nucleosides are eluted from the mini-columns (capacity ≈ 1.8 ml) with 10mM sodium phosphate, pH 6.0; subsequently, the nucleotides are eluted completely with 0.5 M HCl/0.5 M NaCl.
  • 4.4. Quantitation is based on liquid scintillation counting of aliquots of the eluates.

  • 1.1. The properties of Na+/K+-transporting ATPase in microsomal fractions from the nervous tissue of the grasshopper, Poekilocerus bufonius were investigated.
  • 2.2. Two components of ATPase activity are present.
  • 3.3. Inclusion of 1 mM ouabain in the incubation media reduced the activity of total and Na+/K+-ATPase by 57 and 79%, respectively.
  • 4.4. The maximum velocity (Vmax) was decreased by the addition of 1 mM ouabain, whereas the apparent Km value was not affected indicating a non-competitive type of inhibition.
  • 5.5. The calculated value of the pI50 was 6.4 (I50 = 3.98 × 10−7M) for ouabain inhibition of the enzyme showing great sensitivity to the cardiac glycoside ouabain.
  • 6.6. The present results show that the physicochemical properties of Na+/K+-transporting ATPase from the brain of P. bufonius are essentially the same as for the enzyme prepared from the excretory system of the insect which has been previously investigated.
  • 7.7. Dissimilarities were also observed between these tissues in the way that the enzyme from the brain was sensitive to ouabain inhibition with a non-competitive type rather than a ouabain-resistance and a competitive type of inhibition for the enzyme from the excretory system.
  • 8.8. These dissimilarities are probably due to different isoenzyme patterns available in the same insect.

  • 1.1. Inorganic phosphate (Pi) was absorbed rapidly by suspension-cultured cells of Catharanthus roseus which had previously been cultured in Pi-free Murashige Skoog medium.
  • 2.2. The intracellular levels of ATP, ADP and 5-phosphoribosyl-l-pyrophosphate (PRPP) increased markedly during the 24 hr which followed the addition of Pi (1.25mM).
  • 3.3. Availability of PRPP in vivo, estimated by the measurement of nucleotide synthesis from [8-14C]adenine, was also increased by addition of Pi.
  • 4.4. Only a 20% increase in the maximum catalytic activity of PRPP synthetase was observed in extracts of cells, prepared 24 hr after addition of Pi.
  • 5.5. In contrast to results for mammalian PRPP synthetase, the activity of PRPP synthetase, partially purified from Catharanthus roseus, was inhibited by concentration of Pi greater than 5mM.
  • 6.6. The mechanisms involved in the increased availability of PRPP and the synthesis of adenine nucleotides in the plant cells cultured in Pi-containing medium are discussed.
