The effects of an extract of the brown seaweed Ascophyllum nodosum (AN) in grower–finisher pigs are reported. In Experiment 1, the effect of dietary supplementation with increasing levels of AN extract (ANE) on growth performance, carcass characteristics and gastrointestinal microflora was investigated. A total of 360 pigs were randomly allocated, based on initial live-weight and sex, to one of four experimental treatments as follows; control diet (no ANE), control diet plus 3 g ANE/kg, control diet plus 6 g ANE/kg and control diet plus 9 g ANE/kg. These diets were fed ad libitum up to slaughter. In Experiment 2, eight male pigs were allocated to a control diet (no ANE) or the control diet plus 2.5 g ANE/kg to determine effects of ANE on coefficient of total tract apparent digestibility (CTTAD) of nutrients and nitrogen (N) balance. Supplementation with increasing levels of ANE in Experiment 1 resulted in reduced daily gain, carcass weight and kill-out yield during the combined grower–finisher period (P<0.05); however, there were no effects of treatment on feed intake, feed conversion ratio or carcass characteristics. Increasing levels of dietary ANE resulted in decreased ileal coliform counts (P<0.05). Increasing dietary ANE also tended to increase adherent lactobacilli in the colon (P=0.080) but caecal bifidobacteria declined (P<0.05). There were trends towards a linear reduction in colonic bifidobacteria (P=0.077) and towards a quadratic effect on rectal lactobacilli (P=0.077). Intestinal pH was unaffected by ANE supplementation (P>0.05). In Experiment 2, the CTTAD was unaffected by the inclusion of ANE (P>0.05). Overall, the intestinal coliform reductions obtained suggest that ANE may provide a dietary means to improve gut health and potentially reduce pathogen carriage in finishing pigs. However, the negative effects on growth performance observed in healthy animals will most likely limit the commercial use of dietary ANE as a feed additive.  相似文献

Two experiments were conducted to evaluate the effects of dietary supplementation with Astragalus polysaccharide (APS) on growth performance, apparent ileal digestibilities (AID) of amino acids (AA), and their serum concentrations in early weaned piglets. In Exp. 1, 60 pigs were weaned at 21 days of age (BW 7.35 ± 0.23 kg) and allocated to three treatments (20 pigs/treatment), representing supplementing 0.0% (control), 0.02% colistin (antibiotic), or 0.1% APS to a corn- and soybean meal-based diet. Average daily gain (ADG), average daily feed intake (ADFI), and feed/gain ratio (F/G) were measured weekly. Blood samples were obtained from five pigs selected randomly from each treatment for the measurement of serum free AA concentrations on days 7, 14, and 28. In Exp. 2, 12 pigs were weaned at 21 day of age (BW 7.64 ± 0.71 kg), assigned to three treatment groups as in Exp. 1, and surgically fitted with a simple T-cannula at the terminal ileum. Ileal digesta samples were obtained for the measurement of AID of AA on days 7, 14 and 28. Dietary APS did not affect ADFI, but enhanced (P < 0.05) ADG by 11 and 4.4%, and improved F/G by 5.6 and 8.4%, respectively, compared with the control and antibiotic groups. Addition of APS to the diet increased AID and serum concentrations of most nutritionally essential and non-essential AA (including arginine, proline, glutamate, lysine, methionine, tryptophan, and threonine) on days 14 and 28. Circulating levels of total AA were affected by the age of pigs and treatment × time interaction. Collectively, these findings indicate that APS may ameliorate the digestive and absorptive function and regulate AA metabolism to beneficially increase the entry of dietary AA into the systemic circulation, which provide a mechanism to explain the growth-promoting effect of APS in early weaned piglets.  相似文献

Two experiments were conducted to investigate the effects of dietary supplementation of bacteriophage cocktail, probiotics and a combination of these two supplements on performance and gut health of weanling pigs. In Experiment 1, 150 weaned piglets were randomly allotted to three treatments on the basis of BW. The dietary treatments included a basal diet supplemented with 0 (control), 1.0 and 1.5 g/kg bacteriophage cocktail. Pigs fed 1.0 and 1.5 g/kg bacteriophage product had greater (P<0.05) average daily gain (ADG), apparent total tract digestibility of dry matter from day 22 to 35, ileal Lactobacillus spp., villus height (duodenum and jejunum), and fewer coliforms (ileum) and Clostridium spp. (ileum). In Experiment 2, 200 weaned piglets were randomly allotted to four treatments. Dietary treatments included basal diet, basal diet supplemented with 3.0 g/kg fermented probiotic product (P), 1.0 g/kg bacteriophage cocktail (B) and combination of 1.0 g/kg bacteriophage cocktail and 3.0 g/kg fermented probiotic product. Pigs fed bacteriophage cocktail diets had greater (P<0.05) overall ADG, gain to feed ratio (G:F), fecal score from day 8 to day 21, and pigs fed bacteriophage cocktail diets had fewer coliforms (ileum) Clostridium spp. (ileum and cecum). Probiotics significantly increased G:F, colonization of Lactobacillus spp. in ileum. At day 35, bacteriophage treatment group showed greater (P<0.05) villus height of the duodenum, but a deeper crypt in duodenum. The present results indicate that the bacteriophage cocktail had a potential to enhance the performance and gut health of weanling pigs, however their combination with probiotics did not show an interaction.  相似文献

Thirty-six 2.5-year-old wether Inner Mongolian White Cashmere Goats (IMWG) (BW = 42.7 ± 3.44 kg) were used to determine the effects of dietary copper (Cu) concentration on growth performance, nutrient digestibility and fiber characteristics during the cashmere slow-growing period. Wethers were stratified by weight and randomly assigned to four dietary treatments, which included a control diet containing 5.60 mg Cu/kg DM, the control diet supplied, respectively, with 10, 20 and 30 mg Cu/kg DM (total dietary Cu level of 5.60, 15.6, 25.6 and 35.6 mg/kg DM). The experiment lasted 50 days including a 10-day preliminary trial and 10-day metabolism trial. Average daily feed intake (ADFI) did not differ among treatment groups (P > 0.05), except that the supplement providing 30 mg Cu/kg DM decreased average daily gain and gain efficiency (P < 0.05). Copper supplementation had no influence on digestibility of DM, CP and ADF (P > 0.05), however, NDF digestibility of the treatment group supplemented with 30 mg Cu/kg DM was lower compared with that of other groups (P < 0.05). Length and growth rate of cashmere fiber were higher in the treatment group supplemented with 20 mg Cu/kg DM compared with other groups (P < 0.05), but cashmere diameter was not affected by Cu supplementation (P > 0.05). In conclusion, supplementation of Cu at the levels of 10, 20 and 30 mg/kg DM to the basal diet containing 5.60 mg Cu/kg DM had no influence on ADFI or nutrient digestibility of DM, CP and ADF in cashmere goats, while 30 mg Cu/kg DM supplementation had a negative effect on growth performance and NDF digestibility. However, 20 mg Cu/kg DM supplementation of the basal diet enhanced cashmere growth. Hence, the appropriate supplemental level during the cashmere slow-growing period is deemed to be 20 mg Cu/kg DM (total dietary Cu level of 25.6 mg/kg DM).  相似文献

Two experiments in a 2?×?2 factorial arrangement were conducted to evaluate the effect of crude protein (CP) (130 vs. 200 g/kg) and phosphorus (P) (4.0 vs. 6.0 g total P/kg) level in a phytase supplemented diet (500 FTU [phytase units]/kg) in grower-finisher pigs. Owing to the design of the experiment, as dietary P level increased, there was also an increase in dietary calcium (Ca) level in order to maintain a dietary Ca to P ratio of 1.6:1. In Experiment 1, four diets were fed to 56 pigs (n?=?14, initial body weight [BW] 36.7?±?4.2 kg) to investigate the interaction between CP and P on growth performance, bone mineralisation and digesta pH. Experiment 2 consisted of 16 entire male pigs (n?=?4; offered identical diets to that offered in Experiment 1) for the determination of total tract apparent digestibility and nitrogen (N), P and Ca utilisation. There was an interaction between CP and P level on bone ash, bone P and bone Ca concentrations (p?<?0.05). Pigs offered low CP–low P diets had a higher bone ash, P and Ca concentrations than pigs offered high CP–low P diets. However, there was no effect of CP level at high P levels on bone ash, P and Ca concentrations. Pigs offered low P diets had a lower ileal pH compared with pigs offered high P diets (p?<?0.05). In conclusion, offering pigs a high CP–low P, phytase-supplemented diet resulted in a decrease in bone mineralisation.  相似文献

The fermentation kinetics of both fibre choice and maize particle size were studied in vitro from the ileal contents of cannulated pigs given five different experimental diets. Additionally, in vitro batch systems were used to study the quantitative effect of fibre choice and maize particle size on Salmonella Typhimurium growth. Freeze-dried ileal effluents obtained from five cannulated pigs given the five experimental diets in a latin square were used as incubation substrates. The experimental diets consisted of: (1) a standard maize-based diet (ST) where all the ingredients were milled to pass through a 2.5-ml screen, (2) the same diet with the maize ground using a 4.0-ml screen (CG), (3 and 4) two diets in which the maize was partially replaced by (3) sugar beet pulp (80 g/kg; BP) or (4) wheat bran (100 g/kg; WB), or (5) a combination of 80 g sugar beet pulp and 100 g wheat bran/kg (diet HF). Results showed that substrate from BP and HF generated more gas than substrate from ST (P<0.001). In addition, short chain fatty acids production during the entire incubation period was higher (P=0.001) for all the substrates compared to ST substrate (P<0.001) where lower acetate and higher propionate, valerate and branched-chain fatty acids molar ratios were measured. The molar ratio of acetate was highest and the branched-chain fatty acids was lowest for substrates from diets that included sugar beet pulp. The butyrate molar ratio was lower for all substrates compared to the ST diet except for the WB diet. After 12 h of incubation, HF substrate presented the lowest ammonia (91.7 mg/l vs. 125.3 mg/l, P=0.014) and the highest purine bases concentration (0.28 μmol/ml vs. 0.22 μmol/ml, P=0.009). None of the substrates showed any inhibitory effect on Salmonella growth when the batch systems were inoculated with S. Typhimurium. In summary, under the in vitro conditions used, the changes in hindgut fermentation promoted by the inclusion of different fibrous ingredients or by different maize particle size did not produce an inhibitory effect on the growth of S. The effect of feeding Lactobacillus fermentum I5007 on the immune system of weaned pigs with or without E. coli challenge was determined. Twenty-four weaned barrows (6.07 ± 0.63 kg BW) were randomly assigned to one of four treatments (N = 6) in a factorial design experiment. The first two treatments consisted of healthy piglets with half of the pigs receiving no treatment while the other half was orally administered with L. fermentum I5007 (108 CFU/ml) at a daily dose of 20 ml. Pigs in the second two treatments were challenged on the first day with 20 ml of E. coli K88ac (108 CFU/ml). Half of these pigs were not treated while the remaining pigs were treated with 20 ml of L. fermentum I5007 (108 CFU/ml). Peripheral blood lymphocytes subsets were determined using flow cytometry. The intestinal mucosal immunity of the pigs was monitored by real time polymerase chain reaction. The cytokine content of the pig’s serum was also analyzed. Oral administration of L. fermentum I5007 increased blood CD4+ lymphocyte subset percentage as well as tumor necrosis factor-α and interferon-γ expression in the ileum. Pigs challenged with E. coli had elevated jejunal tumor necrosis factor-α while interferon-γ expression was increased throughout the small intestine. There was no difference in the concentration of the cytokines interleukin-2, interleukin-6, tumor necrosis factor-α and interferon-γ in the serum. CD8+ and CD4+/CD8+ in peripheral blood were not affected by treatment. In conclusion, L. fermentum I5007 can enhance T cell differentiation and induce ileum cytokine expression suggesting that this probiotic strain could modulate immune function in piglets.  相似文献

Feeding pharmacological zinc (Zn) to weaned pigs improves growth, and dietary phytase improves P and Zn availability. Metallothionein (MT) increases in the duodenum, kidney, and liver of pigs fed 1000 mg Zn/kg with phytase or 2000 mg Zn/kg with or without phytase when fed for 14 d postweaning. The goal of this study was to determine the effects of feeding pharmacological Zn and phytase on tissue minerals, MT, mineral excretion, and apparent retention. Twenty-four newly weaned pigs (20 d; 7.2 kg) were individually fed twice daily, a basal diet supplemented with 0, 1000, or 4000 mg Zn/kg as Zn oxide, without or with phytase (500 phytase units [FTU]/kg) for 14 d, followed by a basal diet (100 mg Zn/kg) without phytase for 7 d. Pigs fed 4000 mg Zn/kg without phytase had higher (p=0.01) plasma, hepatic, renal Zn, renal Cu, and hepatic, renal, and jejunal MT than pigs fed the basal diet or 1000 mg Zn/kg. Duodenal MT was higher (p=0.0001) in pigs fed 1000 and 4000 mg Zn/kg than in pigs fed the basal diet. In pigs fed 1000 and 4000 mg Zn/kg, Zn loading occurred during the first 11 d of supplementation; by d 14, excess Zn was being excreted in the feces.  相似文献

In vitro regeneration and morphogenesis studies in common bean   总被引:1,自引:0,他引:1  
An efficient protocol for high frequency in vitro regeneration of multiple shoots and somatic embryos from the embryonic axis of common bean (Phaseolus vulgaris) was developed. Ten common bean cultivars representing a wide range of diversity among current commercial market classes were used for in vitro regeneration evaluation in our study. These cultivars were tested on 63 different media formulations consisting of combinations of cytokinins, namely benzyladenine (BA) and thidiazuron (TDZ) at concentration levels of 0.0, 1.0, 2.5, 5.0 and 10.0 mg l−1 and auxin, namely naphthalene acetic acid (NAA) and indole-3-acetic acid (IAA) at concentration levels of 0.0, 0.05, 0.1 and 1.0 mg l−1. P. vulgaris cv. Olathe pinto bean performed the best producing over 20 multiple shoots per explant while cv. Condor black bean was the poorest with nine multiple shoots per explant. The optimum media for regeneration of multiple shoots was 4.4 mg l−1 Murashige and Skoog (MS) containing 2.5 mg l−1 BA and 0.1 mg l−1 IAA supplemented with 30 mg l−1 silver nitrate. Adventitious shoots and somatic embryos were regenerated on 4.4 mg l−1 MS medium containing 1 mg l−1 TDZ and 0.05 mg l−1 NAA supplemented with 30 mg l−1 silver nitrate or activated charcoal. Efficient and effective rooting of plantlets was achieved by dipping the cut end base of in vitro regenerated shoots in 1.0 mg l−1 indole-3-butyric acid (IBA) solution and culturing on media containing 4.4 mg l−1 MS supplemented by 0.1 mg l−1 IAA, NAA or IBA.  相似文献

The objective of the study was to determine the coefficients of ileal apparent digestibility (CIAD) of sorghum protein and amino acids (AA) in weaned piglets and growing pigs. Digestibility coefficients were estimated using the regression and difference methods for the weaned piglets; and the direct and difference methods for the growing pigs. To test the hypothesis that CP and AA digestibility of sorghum is lower in weaned piglets than in growing pigs, two experiments were conducted. In experiment one, 20 weaned piglets were fitted with a 'T' cannula at 21 days of age and were fed for 2 weeks one of five dietary treatments: a reference or control diet providing 200 g of CP/kg from casein (C) as the sole protein source, and four casein-sorghum (C-S) diets kept isoproteic to C by the appropriate adjustment of C and maize starch proportions; the amount of sorghum (S) in these diets was 135, 307, 460 and 614 g/kg. In experiment 2, fifteen castrated pigs weighing 57.8 ± 2.8 kg were used and randomly allotted to one of three dietary treatments: a reference casein-maize starch diet containing C as the sole protein source, a C-S diet, both diets containing 160 g of CP/kg, and a fortified S diet containing 68 g of CP/kg. In piglets the CIAD for CP and AA decreased linearly (P < 0.05) as the amount of S in the diet increased. The average ileal digestibility of AA from C was 0.858 ± 0.111, and decreased to 0.663 ± 0.191 at the higher S level. The CIAD estimated using the regression or difference methods were similar for leucine, cysteine, glutamic acid, serine, alanine and tyrosine, and different for the other AA. In growing pigs the CIAD of protein and AA (except alanine and cysteine) were similar (P > 0.05) for the C and the C-S diets, but higher (P < 0.05) than those for the S diet. The CIAD for S obtained by the difference method were higher (P < 0.05) than those obtained using the direct method, except for lysine, isoleucine, valine, methionine, threonine and cysteine. The results indicate that except for lysine and cysteine, growing pigs' ability to digest AA and protein is superior than weaned piglets.  相似文献

An experiment was conducted to evaluate the effects of cereal carbohydrate form (isolated v. cereal matrix) and level, especially mixed-linked β-glucan (hereafter referred to as β-glucan) and starch amylase/amylopectin ratio on nutrient digestibility and fermentation parameters in the intestines of weaned pigs. Four hulless barley cultivars containing varying β-glucan levels (41 to 84 g/kg) were compared with hulled barley, supplemented or not with a β-glucan concentrate (BBG; 270 g/kg β-glucan) and two oat cultivars for digestibility and fermentation metabolites. Seventy-two weaned piglets (BW = 12.8 ± 1.9 kg) were assigned to one of nine diets composed of 815 g/kg cereal, 60 g/kg whey, 90 g/kg soy protein isolate and 35 g/kg minerals. After 15 days, the pigs were killed, and digesta collected from ileum and colon were analyzed for proximate nutrients, short-chain fatty acids (SCFAs), lactic acid (LA) and ammonia. Ileal and total tract digestibility of proximate nutrients and non-starch polysaccharides (NSPs) were determined using HCl-insoluble ash as a marker. Organic matter (OM) ileal digestibility was greater (P < 0.05) for diets based on hulless barley (77% ± 1.1% on average), as compared with hulled barley (64% ± 1.4%) and oat (58% ± 1.5%). Similar trends were found for total tract OM digestibility, varying from 90% ± 0.3% for hulless barley to 67% ± 0.4% for oat, on average. NSP digestibility differed (P < 0.05) within and between cereal types, ranging from 20% (hulled barley plus 163 g/kg BBG or 40 g/kg β-glucan) to 51% (SB94893 hulless barley cultivar with high β-glucan and high amylose ratio) at the ileum and from 44% (hulled barley) to 84% (SB94893 cultivar) at the total tract level. No dietary effect (P > 0.05) was found for SCFA concentration in ileal contents, whereas in colonic contents, SCFA was lower in pigs fed oat (P < 0.001). LA concentration was greater (P < 0.001) in the colon of pigs fed hulless barley than in pigs fed hulled barley and oat. Expressed per kg carbohydrate (NSP + starch) fermented, the ammonia concentration at the colon was lowest for hulled barley diets (supplemented with β-glucan) and the highest for oat diets. In conclusion, the interaction of both form and level of β-glucan impacted nutrient digestibility and fermentation. Hulless barleys with high soluble NSP such as β-glucan and resistant starch yielded, in general higher SCFA and LA and lower ammonia. Hulless barleys may, therefore, have potential for use in feeding strategies designed to improve gut health in pigs.  相似文献

Studies were conducted on the enumeration of 7 groups of fecal microflora including total aerobes, total anaerobes, coliforms, lactobacilli, staphylococci, streptococci and yeasts and molds of 18-day old piglets. These pigs were early weaned (21 days) on different modifications of an early-weaning ration. The above mentioned microflora were enumerated again when some of the pigs in a replicate started scouring. The occurrence of diarrhea was always associated with significant increases in the numbers of coliforms and corresponding decreases in the lactobacilli counts. No other single group of fecal microflora differed in the scouring and non-scouring animals. The composition of the early-weaning ration offered to the animals did not, in itself, influence the fecal microflora to any appreciable extent. In another series of experiments, enumeration of coliforms and lactobacilli was conducted on samples obtained from different segments of the intestinal tracts of scouring and non-scouring pigs. Increased numbers of coliforms and decreased numbers of lactobacilli were observed at all levels of the intestinal tracts of the scouring animals. However, these changes were more marked in the duodenal samples than in those obtained from other parts of the intestine.  相似文献

It is not clear if B vitamins supplied to the small intestine of dairy cows from dietary and rumen microbial sources are provided in sufficient quantity to maximize animal performance. Our objective was to determine effects of adding a ruminally protected B vitamin blend supplement, containing biotin, folic acid, pantothenic acid and pyridoxine, to the diet of high producing dairy cows on their productivity. Two dairy facilities located in California (USA) were used, one with mid lactation Holstein cows (Experiment 1) and the other with early lactation Holstein cows (Experiment 2). In each Experiment, cows were randomly assigned to treatment in a 2 × 2 crossover design with 28 d (Experiment 1) or 35 d (Experiment 2) experimental periods. In Experiment 1, milk and milk fat yield were unaffected by treatment, although milk fat proportion was lower (37.1 versus 36.3 g/kg; P<0.01), but milk protein yield was higher (1.21 versus 1.24 kg/d; P=0.02) in cows fed B vitamins. In Experiment 2, milk (39.60 versus 40.46 kg/d; P=0.02), milk fat (1.40 versus 1.47 kg/d; P<0.01) and milk protein yield (1.10 versus 1.16 kg/d; P<0.01), as well as milk energy output (113.2 versus 117.8 MJ/d; P<0.01) were all higher with B vitamin feeding. Body condition score (BCS) increased more with B vitamin feeding in Experiment 2, but was unaffected in Experiment 1. Body locomotion score (BLS) increased with B vitamin feeding in both experiments (P=0.01 and < 0.01, respectively), possibly an indication of reduced locomotory ability. Overall, productivity of high producing lactating dairy cows responded positively to feeding a mixture of ruminally protected B vitamins, although differences in the extent of the positive responses between experiments perhaps suggests that early lactation cows, with lower DM intake to milk yield ratios, may be more responsive to ruminally protected B vitamins than mid lactation cows, with higher DM intake to milk yield ratios.  相似文献

This study was designed to determine the effect of ultra-fine Chinese herbal powder as a dietary additive on serum concentrations and apparent ileal digestibilities (AID) of amino acids (AA) in young pigs. In Experiment 1, 60 Duroc x Landrace x Yorkshire piglets weaned at 21 days of age were randomly assigned to one of three treatments, representing supplementation with 0 or 2 g/kg of the powder, or 0.2 g/kg of colistin (an antibiotic) to corn- and soybean meal-based diets (n = 20 per group). Blood samples from five piglets per group were collected on days 7, 14, and 28 to determine serum AA concentrations. In Experiment 2, 12 barrows with an average initial body weight of 7.64 kg were randomly assigned to one of the three dietary treatments, followed by surgical placement of a simple T-cannula at the terminal ileum. All of the diets contained 0.1% titanium oxide as a digestibility marker. The samples of terminal ileal digesta were collected on day 7 for determining AID of AA. Results show that dietary supplementation with the herbal powder increased (P < 0.05) serum concentrations and AID of most AA by 10-50% and 10-16%, respectively. As an indicator of improved intestinal function, AID values of calcium were also enhanced in piglets supplemented with the herbal powder. Dietary supplementation of colistin increased serum concentrations and AID values of some AA by 8-44% and 10-15%, respectively, in comparison with the non-supplemented group. These novel findings demonstrate that the herbal powder can enhance the digestibility of dietary protein and the intestinal absorption of AA into the systemic circulation in post-weaning pigs, therefore providing a new mechanism for its growth- and immunity-promoting efficacy.  相似文献

Two experiments were conducted to evaluate the digestible (DE) and metabolisable energy (ME), apparent (AID) and standardised ileal digestibility (SID) of amino acids (AA) in yeast extract (YE) and spray-dried porcine plasma (SDPP). In Experiment 1, 18 barrows (25.1 ± 1.2 kg body weight [BW]) were randomly allotted to three treatments with six replicates per treatment. The DE and ME of YE was 20.64 and 19.31 MJ/kg, respectively, which were not significantly different with the DE and ME of SDPP (18.74 and 18.05 MJ/kg, respectively). In Experiment 2, six barrows (20.6 ± 2.6 kg BW) fitted with ileal T-cannulas were fed three diets in a repeated 3-period Latin square design. For Met and Glu, the AID tended to be, while the SID was significantly higher (< 0.05), in YE than in SDPP. The AID of Cys tended to be lower in YE (p = 0.07), while the SID of Phe tended to be higher in YE than in SDPP (p = 0.06). Accordingly, YE could be a potential substitute for SDPP as a superior protein ingredient in diets for pigs in terms of the available energy and AA digestibility.  相似文献

Abstract

In order to determine ileal losses of nitrogen (N) and amino acids (AA) and the coefficients of apparent and true ileal digestibility (AID, TID) of N and AA from casein in piglets and pigs, two experiments were conducted. In Experiment 1, 24 piglets were used. The piglets were weaned at 17 days of age, weighing 6.4 kg and cannulated at terminal ileum. Ileal digesta was collected at 28 – 29 and 35 – 36 days of age in period 1 and 2, respectively. Feed intake was 150 and 300 g · d?1 during the first and second period. In Experiment 2, 16 castrates weighing 52.5 kg and cannulated at terminal ileum were used. The intake level of digestible energy was 2.5 times their maintenance requirement. The experiment lasted 7 days and ileal digesta was collected on day 6 – 7. Treatments consisted of four levels of N from casein: 8, 16, 24 and 32 g N · kg?1 feed, respectively. Results showed that N level did not increase N or AA ileal losses. In piglets, N and AA ileal losses were similar between periods, except for period 2, where losses per kg DMI were about 47 and 64% higher for glycine and proline, respectively (p < 0.05). When ileal losses from pigs and piglets were compared, piglets had higher (p < 0.05) ileal losses of N and AA (excepted glutamic acid and alanine). A lower (p < 0.05) AID was observed in piglets in period 2 for N, methionine, glutamic acid, glycine and proline. With exception of glycine in pigs, all values for TID of N and AA of casein were superior to 0.90. Piglets had higher TID of N, leucine, isoleucine, valine and phenylalanine. These results showed that piglets have higher ileal losses than pigs.  相似文献

Two experiments were conducted to evaluate the effects of vitamin C supplementation on performance, iron status and immune function of pigs during the 21-day post-weaning period. In experiment one, 48 crossbred pigs (Chester White ‐ Large White ‐ Yorkshire), weaned at 30 days of age and weighing 7.7 ± 0.9kg, were allotted to diets containing either 0 or 300 mg/kg vitamin C. In experiment two, 96 crossbred pigs (Chester White ‐ Large White ‐ Yorkshire), weaned at 20 ± 2 days and weighing 7.1 ± 0.5kg, were allotted to diets containing 0,75 or 300 mg/kg vitamin C. Six replicate pens were assigned to each treatment in experiment one while experiment two had eight replicates. All pens housed two barrows and two gilts. In both experiments, no improvement (P > 0.05) in growth rate, feed intake or feed conversion was observed as a result of vitamin C supplementation. Plasma iron concentration increased (P < 0.10) with increased vitamin C in the diet while free and total iron binding capacity were unaffected by treatment. There were no differences in the intradermal response to the mitogen phytohemaggutinin used as an indicator of cellular immunity (P > 0.05). In trial 2, the plasma levels of the immunoglobulin IgG showed a linear (P = 0.07) increase with increasing levels of vitamin C and the same trend was noted in trial 1. Antibody titers to bovine serum albumin also tended to increase in both trials but the increases were not statistically significant. In conclusion, the overall results of these experiments indicate that weanling pig performance is not improved as a result of vitamin C supplementation. Whether or not vitamin C plays a role in stimulating humoral immune function in pigs requires further study since the results of our experiments do not completely rule out the possibility that such a role exists.  相似文献

Here, we report for the first time the synthesis of bismuth-coated silver nanoparticles in dichroic bismuth glass nanocomposites by a novel and simple one-step melt quench technique without using any external reducing agent. The metallic silver nanoparticles (Ag NPs) were generated first, and subsequently, metallic bismuth was deposited on the Ag NPs and formed a thick layer. The reduction of Bi3+ to Bio and subsequently its deposition on the Ag NPs (which were formed earlier than Bio) in the K2O–Bi2O3–B2O3 (KBB) glass system have been explained by their standard reduction potentials. The UV–vis absorption spectra show a prominent surface plasmon resonance (SPR) absorption band at 575 nm at lower concentrations (up to 0.01 wt%); three bands at 569, 624 and 780 nm at medium concentration (0.02–0.03 wt%); and two weak bands at 619 and 817 nm at highest concentration (0.06 wt%) of silver. They have been explained by the electrodynamics theories. TEM images reveal the conversion of spheroidal (5–15 nm) to hexagonal (10–35 nm) shaped Ag NPs with the increase in concentration of silver (up to 0.06 wt%). SAED pattern confirms the crystalline planes of rhombohedral bismuth and cubic silver. Thermal treatment at 360 °C, which is the glass transformation temperature (T g) of the sample containing lower concentration of silver (0.007 wt%), shows red-shifted SPR band due to increase in size of NPs. Whereas the sample containing higher concentration (0.06 wt%) of silver under similar treatment exhibited changes in SPR spectral profile happened due to conversion to spherical NPs from hexagonal shape and reduction in size (10–20 nm) of NPs after heat treatment for 65 h. HRTEM images corroborate the different orientations of the NPs. FESEM images reveal hexagonal disk like structure having different orientations. Dichroic nature of the nanocomposites has been explained with the size and shape of Ag nanoparticles. We believe that this work will create new avenues in the area of nanometal–glass hybrid nanocomposites and the materials have significant applications in the field of optoelectronics and nanophotonics.  相似文献

Neonates are at increased risk for inflammatory bowel disease, but effective prevention and treatments are currently limited. This study was conducted with the lipopolysaccharide (LPS)-challenged piglet model to determine the effects of dietary supplementation with α-ketoglutarate (AKG) on the intestinal morphology and function. Eighteen 24-day-old pigs (weaned at 21 days of age) were assigned randomly to control, LPS, and LPS + AKG groups. The piglets in the control and LPS groups were fed a corn- and soybean meal-based diet, whereas the LPS + AKG group was fed the basal diet supplemented with 1% AKG. On days 10, 12, 14, and 16, piglets in the LPS and LPS + AKG groups received intraperitoneal administration of LPS (80 μg/kg BW), whereas piglets in the control group received the same volume of saline. On day 16, d-xylose was orally administrated to all pigs at the dose of 0.1 g/kg BW, 2 h after LPS or saline injection, and blood samples were collected 3 h thereafter. Twenty-four hours post-administration of LPS or saline, pigs were killed to obtain intestinal mucosae for analysis. Compared with the control group, LPS challenge reduced (P < 0.05) protein levels, the ratio of villus height to crypt depth, and the ratio of phosphorylated mTOR to total mTOR in duodenal, jejunal, and ileal mucosa. These adverse effects of LPS were attenuated (P < 0.05) by AKG supplementation. Moreover, AKG prevented the LPS-induced increase in intestinal HSP70 expression. Collectively, these novel results indicate that dietary supplementation with 1% AKG activates the mTOR signaling, alleviates the mucosal damage, and improves the absorptive function of the small intestine in LPS-challenged piglets. The findings not only help understand the mode of AKGs actions in the neonatal gut but also have important implications for infant nutrition under inflammatory conditions.  相似文献

Background and Aims

The clinical onset and severity of intestinal disorders in humans and animals can be profoundly impacted by early life stress. Here we investigated the impact of early weaning stress in pigs on intestinal physiology, clinical disease, and immune response to subsequent challenge with enterotoxigenic F18 E. coli (ETEC).

Methodology

Pigs weaned from their dam at 16 d, 18 d, and 20 d of age were given a direct oral challenge of F18 ETEC at 26 d of age. Pigs were monitored from days 0 to 4 post-infection for clinical signs of disease. On Day 4 post-ETEC challenge, ileal barrier function, histopathologic and inflammatory cytokine analysis were performed on ileal mucosa.

Results

Early weaned pigs (16 d and 18 d weaning age) exhibited a more rapid onset and severity of diarrhea and reductions in weight gain in response to ETEC challenge compared with late weaned pigs (20 d weaning age). ETEC challenge induced intestinal barrier injury in early weaned pigs, indicated by reductions in ileal transepithelial electrical resistance (TER) and elevated FD4 flux rates, in early weaned pig ileum but not in late weaned pigs. ETEC-induced marked elevations in IL-6 and IL-8, neutrophil recruitment, and mast cell activation in late-weaned pigs; these responses were attenuated in early weaned pigs. TNF levels elevated in ETEC challenged ileal mucosa from early weaned pigs but not in other weaning age groups.

Conclusions

These data demonstrate the early weaning stress can profoundly alter subsequent immune and physiology responses and clinical outcomes to subsequent infectious pathogen challenge. Given the link between early life stress and gastrointestinal diseases of animals and humans, a more fundamental understanding of the mechanisms by which early life stress impacts subsequent pathophysiologic intestinal responses has implications for the prevention and management of important GI disorders in humans and animals.  相似文献
